Delineating the spectrum of cellular heterogeneity will provide new insights into normal cellular properties within the breast tissue that might become dysregulated in the initial stages of cancer. Investigation of surface marker expression provides a valuable approach to resolve complex cell populations. However, the majority of cell surface maker expression of primary breast cells have not been investigated. Methods: To determine the differences in expression of a range of uninvestigated cell surface markers between the normal breast cell subpopulations, primary human breast cells were analysed using high-throughput flow cytometry for the expression of 242 cell surface proteins in conjunction with EpCAM/CD49f staining. Results: We identified 35 surface marker proteins expressed on normal breast epithelial and/or stromal subpopulations that were previously unreported. We also show multiple markers were equally expressed in all cell populations (e.g. CD9, CD59, CD164) while other surface markers were confirmed to be enriched in different cell lineages: CD24, CD227 and CD340 in the luminal compartment, CD10 and CD90 in the basal population, and CD34 and CD140b on stromal cells. Conclusions: Our dataset of CD marker expression in the normal breast provides better definition for breast cellular heterogeneity.

The latest flow cytometry techniques and applications are also described, featuring examples of the data that can be generated and, importantly, how the data can be analysed. Life is spatially partitioned within lipid membranes to allow the isolated formation of distinct molecular states inside cells and organelles. Cell fusion is the merger of two or more cells to form a single cell. Here we provide a protocol for cell fusion of two different cell types. Fused hybrid cells are enriched by flow cytometry-based sorting, followed by fluorescence microscopy of hybrid cell structure and function. Fluorescently tagged proteins generated by genome editing are imaged inside fused cells, allowing cellular structures to be identified based on fluorescence emission and referenced back to the cell type of origin. This robust and general method can be applied to different cell types or organelles of interest, to understand cellular structure and function across a range of fundamental biological questions.

We report on the latest iteration of the baseline opto-mechanical architecture of the LISA instru- ment, which has been developed within the current LISA Mission Formulation study under ESA con- tract. The collective features of the current architec- ture have been consolidated in an extensive trade of various alternative payload configurations, including variants with only one active proof mass per space- craft and the application of “In-Field Pointing” for accommodation of constellation breathing. With respect to the original configuration [1], the newly established architecture most notably distin- guishes itself by the use of an off-axis telescope and a “non-frequency-swap” science interferometer for stray light mitigation, as well as the implementa- tion of ancillary pathlength metrology in terms of an “Optical Truss” and Point Ahead Angle sensing.

We review possible alternatives for the realization of the LISA laser assembly against the requirements and present a concept for a tailored master oscillator fiber power amplifier solution. An all-solid-state, all- fiber coupled approach with a Nd:YAG NPRO seed appears particularly attractive due to the possibility to combine excellent spectral properties and high output powers with superior environmental robustness.

We present a detailed concept for the realization of the LISA optical metrology subsystem, which employs heterodyne interferometry in a so-called “strap-down” architecture to accomplish a highly sensitive detection of gravity-wave induced displacements of dedicated mass references within the payload of the three LISA satellites. A frequency swap between transmitted and local reference beam is introduced to minimize the impact of stray light on the measurement sensitivity. The performance of the system is demonstrated by first optical modeling.

Objective Arcuate proopiomelanocortin (POMC) neurons are critical nodes in the control of body weight. Often characterized simply as direct targets for leptin, recent data suggest a more complex architecture. Methods Using single cell RNA sequencing, we have generated an atlas of gene expression in murine POMC neurons. Results Of 163 neurons, 118 expressed high levels of Pomc with little/no Agrp expression and were considered “canonical” POMC neurons (P+). The other 45/163 expressed low levels of Pomc and high levels of Agrp (A+P+). Unbiased clustering analysis of P+ neurons revealed four different classes, each with distinct cell surface receptor gene expression profiles. Further, only 12% (14/118) of P+ neurons expressed the leptin receptor (Lepr) compared with 58% (26/45) of A+P+ neurons. In contrast, the insulin receptor (Insr) was expressed at similar frequency on P+ and A+P+ neurons (64% and 55%, respectively). Conclusion These data reveal arcuate POMC neurons to be a highly heterogeneous population. Induction of long-lasting immunity against viral respiratory tract infections remains an elusive goal. Using a nonhuman primate model of human respiratory syncytial virus (hRSV) infection, we compared mucosal and systemic immune responses induced by different DNA delivery approaches to a novel parenteral DNA prime-tonsillar adenoviral vector booster immunization regimen. Intramuscular (i.m.) electroporation (EP) of a DNA vaccine encoding the fusion protein of hRSV induced stronger systemic immune responses than intradermal EP, tattoo immunization, and conventional i.m. DNA injection. A single EP i.m., followed by two atraumatic tonsillar immunizations with the adenoviral vector, elicited strong systemic immune responses, an unique persistent CD4 + and CD8 + T cell response in the lower respiratory tract and protection from intranasal hRSV challenge. Thus, parenteral DNA priming followed by booster immunization targeted to a mucosal inductive site constitutes an effective vaccine regimen for eliciting protective immune responses at mucosal effector sites. IMPORTANCE The human respiratory syncytial virus (hRSV) is the most common cause of severe respiratory tract disease in infancy and leads to substantial morbidity and morality in the elderly. In this study, we compared the immunogenicity and efficacy of several gene-based immunization protocols in rhesus macaques. Thereby, we found that the combination of an initially parenterally delivered DNA vaccine with a subsequent atraumatic tonsillar adenoviral vector immunization results in a strong systemic immune response accompanied by an exceptional high T-cell response in the mucosa. Strikingly, these animals were protected against a RSV challenge infection controlling the viral replication indicated by a 1,000-fold-lower viral load in the lower respiratory tract. Since mucosal cellular responses of this strength had not been described in earlier RSV vaccine studies, this heterologous DNA prime-tonsillar boost vaccine strategy is very promising and should be pursued for further preclinical and clinical testing.

The C/EBPα transcription factor plays a pivotal role in myeloid differentiation and E2F-mediated cell cycle regulation. Although CEBPA mutations are common in acute myeloid leukaemia (AML), little is known regarding pre-leukemic alterations caused by mutated CEBPA. Here, we investigated early events involved in pre-leukemic transformation driven by CEBPA N321D in the LMPP-like cell line Hoxb8-FL (Redecke et al., Nat Methods 2013), which can be maintained in vitro as a self-renewing LMPP population using Flt3L and estradiol, as well as differentiated both in vitro and in vivo into myeloid and lymphoid cell types. Hoxb8-FL cells were retrovirally transduced with Empty Vector (EV), wild-type CEBPA (CEBPA WT) or its N321D mutant form (CEBPA N321D). CEBPA WT-transduced cells showed increased expression of cd11b and SIRPα and downregulation of c-kit, suggesting that wild-type CEBPA was sufficient to promote differentiation even under LMPP growth conditions. Interestingly, we did not observe the same phenotype in CEBPA N321D-transduced cells. Upon withdrawal of estradiol, both EV and CEBPA WT-transduced cells differentiated rapidly into a conventional dendritic cell (cDC) phenotype by day 7 and died within 12 days. By contrast, CEBPA N321D-transduced cells continued to grow for in excess of 56 days, with an initial cDC phenotype but by day 30 demonstrating a plasmacytoid dendritic cell precursor phenotype. CEBPA N321D-transduced cells were morphologically distinct from EV-transduced cells. To test leukemogenic potential in vivo, we performed transplantation experiments in lethally irradiated mice. Serial monitoring of peripheral blood demonstrated that Hoxb8-FL derived cells had disappeared by 4 weeks, and did not reappear. However, at 6 months CEBPA N321D-transduced cells could still be detected in bone marrow in contrast to EV-transduced cells but without any leukemic phenotype. To identify early events involved in pre-leukemic transformation, the differentiation profiles of EV, CEBPA WT and CEBPA N321D-transduced cells were examined with single cell RNA-seq (scRNA-seq). 576 single cells were taken from 3 biological replicates at days 0 and 5 post-differentiation, and analysed using the Automated Single-Cell Analysis Pipeline (Gardeux et al., Bioinformatics 2017). Visualisation by t-SNE (Fig 1) demonstrated: (i) CEBPA WT-transduced cells formed a distinct cluster at day 0 before withdrawal of estradiol; (ii) CEBPA N321D-transduced cells separated from EV and CEBPA WT-transduced cells after 5 days of differentiation, (iii) two subpopulations could be identified within the CEBPA N321D-transduced cells at day 5, with a cluster of five CEBPA N321D-transduced single cells distributed amongst or very close to the day 0 non-differentiated cells. Differential expression analysis identified 224 genes upregulated and 633 genes downregulated specifically in the CEBPA N321D-transduced cells when compared to EV cells after 5 days of differentiation. This gene expression signature revealed that CEBPA N321D-transduced cells switched on a HSC/MEP/CMP transcriptional program and switched off a myeloid dendritic cell program. Finally, in order to further dissect the effect of the N321D mutation, the binding profile of endogenous and CEBPA N321D was compared by ChIP-seq before and after 5 days of differentiation. Integration with scRNA-seq data identified 160 genes specifically downregulated in CEBPA N321D-transduced cells which were associated with the binding of the mutant protein. This list of genes included genes previously implicated in dendritic cell differentiation (such as NOTCH2, JAK2), as well as a number of genes not previously implicated in the evolution of AML, representing potentially novel therapeutic targets. Because of their high antigen specificity and metabolic stability, genetically engineered human monoclonal antibodies are on the way to becoming one of the most promising medical diagnostics and therapeutics. In order to establish an in vitro system capable of producing such biosimilar antibodies, we used human constant chain sequences to design the novel human antibody expressing vector cassette pMAB-ABX. A bidirectional tetracycline (tet)-controllable promotor was used for harmonized expression of immunoglobulin type G (IgG) heavy and light chains. As an example we used anti-prion protein (anti-PrP) IgGs. Therefore, the variable heavy (V(H)) and light chain (V(L)) sequences of anti-PrP antibodies, previously generated in our laboratory by DNA immunization of prion protein knock-out mice, were isolated from murine hybridoma cell lines and inserted into pMAB-ABX vector. After transfection of Chinese hamster ovary (CHO) cells, a number of stable antibody producing cell clones were selected. One cell line (pMAB-ABX-13F10/3B5) stably expressing the recombinant humanized antibody (rechuAb) 13F10/3B5 was selected for detailed characterization by Western blot, immunofluorescence, and flow cytometric analyses. The full-length recombinant humanized IgG antibody showed a high level of expression in the cytoplasm. In conclusion, the new cell system described here is a suitable tool to produce functional intact full-length humanized IgG antibodies.

The LISA Mission (Laser Interferometer Space Antenna) is currently under mission formulation with a launch date planned in 2020. The purpose of the mission is the observation of gravitational waves at frequencies between 0.1 mHz and 1 Hz by measuring distance fluctuations between inertial reference points, represented by cubic proof masses. In order to provide a sufficient sensitivity of the instrument, distance fluctuations between two inertial reference points must be measured with a strain accuracy of around 10−20 Hz−1/2. This is achieved by setting up a laser interferometer with a base-length of 5⋅106 km and a path-length measurement noise in the order of 10 pm Hz−1/2. For a correct evaluation of the data on the ground, it is essential that the science data telemetry preserves all required frequency domain information. That is, any on-board data-processing and down-sampling must be done with great care in order not to introduce aliasing or other artifacts into the data stream. As an additional complication, most of the optical metrology data is dominated by laser phase noise which is about eight orders of magnitude larger than the required instrument sensitivity. However, by applying a method called “time-delayed interferometry” during the ground data processing, this laser phase noise can be eliminated from the data. This method has already been demonstrated in a detailed simulation environment, but it requires a very careful filtering, synchronization, and interpolation of the individual data streams. Last but not least, a calibration of system parameters is necessary in many areas of the LISA measurement system. The system design must therefore ensure that all data required for these calibrations is available on-ground in a quality that allows a successful computation of the calibration coefficients within a reasonable time-frame. The data streams do not only include data from the optical metrology system, but also from the drag-free and attitude control system which are used to derive other information, such as the charge state of the proof mass. This yields a strong coupling between the different disciplines since data that is only used for housekeeping purposes in other missions becomes an essential part of the science data stream for the LISA mission. This paper gives an overview of the LISA measurement and data-processing chain. It highlights the most challenging areas that have been identified so far and describes the intended solution methods.

Recent advances in the cellular and molecular biology of single stem cells have uncovered significant heterogeneity in the functional properties of stem cell populations. This has prompted the development of approaches to study single cells in isolation, often performed using multiparameter flow cytometry. However, many stem cell populations are too rare to test all possible cell surface marker combinations, and virtually nothing is known about functional differences associated with varying intensities of such markers. Here we describe the use of index sorting for further resolution of the flow cytometric isolation of single murine hematopoietic stem cells (HSCs). Specifically, we associate single-cell functional assay outcomes with distinct cell surface marker expression intensities. High levels of both CD150 and EPCR associate with delayed kinetics of cell division and low levels of differentiation. Moreover, cells that do not form single HSC-derived clones appear in the 7AAD(dim) fraction, suggesting that even low levels of 7AAD staining are indicative of less healthy cell populations. These data indicate that when used in combination with single-cell functional assays, index sorting is a powerful tool for refining cell isolation strategies. This approach can be broadly applied to other single-cell systems, both to improve isolation and to acquire additional cell surface marker information.

Contractile myoepithelial cells dominate the basal layer of the mammary epithelium and are considered to be differentiated cells. However, we observe that up to 54% of single basal cells can form colonies when seeded into adherent culture in the presence of agents that disrupt actin-myosin interactions, and on average, 65% of the single-cell-derived basal colonies can repopulate a mammary gland when transplanted in vivo. This indicates that a high proportion of basal myoepithelial cells can give rise to a mammary repopulating unit (MRU). We demonstrate that myoepithelial cells, flow-sorted using two independent myoepithelial-specific reporter strategies, have MRU capacity. Using an inducible lineage-tracing approach we follow the progeny of myoepithelial cells that express α-smooth muscle actin and show that they function as long-lived lineage-restricted stem cells in the virgin state and during pregnancy.

Toll-like receptor (TLR) ligands are being considered as adjuvants for the induction of antigen-specific immune responses, as in the design of vaccines. Polyriboinosinic-polyribocytoidylic acid (poly I:C), a synthetic double-stranded RNA (dsRNA), is recognized by TLR3 and other intracellular receptors. Poly ICLC is a poly I:C analogue, which has been stabilized against the serum nucleases that are present in the plasma of primates. Poly I:C12U, another analogue, is less toxic but also less stable in vivo than poly I:C, and TLR3 is essential for its recognition. To study the effects of these compounds on the induction of protein-specific immune responses in an animal model relevant to humans, rhesus macaques were immunized subcutaneously (s.c.) with keyhole limpet hemocyanin (KLH) or human papillomavirus (HPV)16 capsomeres with or without dsRNA or a control adjuvant, the TLR9 ligand CpG-C. All dsRNA compounds served as adjuvants for KLH-specific cellular immune responses, with the highest proliferative responses being observed with 2 mg/animal poly ICLC (p = 0.002) or 6 mg/animal poly I:C12U (p = 0.001) when compared with immunization with KLH alone. Notably, poly ICLC—but not CpG-C given at the same dose—also helped to induce HPV16-specific Th1 immune responses while both adjuvants supported the induction of strong anti-HPV16 L1 antibody responses as determined by ELISA and neutralization assay. In contrast, control animals injected with HPV16 capsomeres alone did not develop substantial HPV16-specific immune responses. Injection of dsRNA led to increased numbers of cells producing the T cell–activating chemokines CXCL9 and CXCL10 as detected by in situ hybridization in draining lymph nodes 18 hours after injections, and to increased serum levels of CXCL10 (p = 0.01). This was paralleled by the reduced production of the homeostatic T cell–attracting chemokine CCL21. Thus, synthetic dsRNAs induce an innate chemokine response and act as adjuvants for virus-specific Th1 and humoral immune responses in nonhuman primates., Author Summary Novel adjuvants that facilitate the induction of strong cellular immunity could be of help in the design of vaccine strategies to combat infections such as HIV or tuberculosis. Our immune cells possess archaic receptors recognizing structures of infectious pathogens, and the interaction of these receptors with their ligands results in an activation of the immune system. Here we exploited synthetic forms of one of these ligands, i.e., dsRNA, to define an adjuvant for the induction of cellular immune responses in primates. We injected model and viral proteins together with three different forms of dsRNA subcutaneously (s.c.) in rhesus macaques, and all compounds served as adjuvants for the induction of cellular immunity without the incidence of major side effects. These adjuvant effects depended on the adjuvant dose and coincided with profound alterations in the chemokine production in the draining lymph nodes. dsRNA also helped to induce cellular and humoral immune responses against capsomeres of low immunogenicity derived from the human papillomavirus 16, the causative agent in about 50% of all cases of cervical cancer worldwide. Therefore, formulations involving synthetic dsRNA are promising candidates for development of novel vaccines.

We investigated the immunogenicity and efficacy of a bimodal prime/boost vaccine regimen given by various routes in the Simian immunodeficiency virus (SIV) rhesus monkey model for AIDS. Twelve animals were immunized with SIV DNA-vectors followed by the application of a recombinant adenovirus (rAd5) expressing the same genes either intramuscularly (i.m.) or by oropharyngeal spray. The second rAd5-application was given i.m. All vaccinees plus six controls were challenged orally with SIVmac239 12 weeks post-final immunization. Both immunization strategies induced strong SIV Gag-specific IFN-γ and T-cell proliferation responses and mediated a conservation of CD4+ memory T-cells and a reduction of viral load during peak viremia following infection. Interestingly, the mucosal group was superior to the systemic group regarding breadth and strength of SIV-specific T-cell responses and exhibited lower vector specific immune responses. Therefore, our data warrant the inclusion of mucosal vector application in a vaccination regimen which makes it less invasive and easier to apply.

To evaluate the efficacy of a multigenic vaccine and its protective immunity in the SIVmac239 challenge model, 12 rhesus macaques were divided into two groups. The vaccine group was intramuscularly immunized with multigenic DNA and recombinant adenovirus vaccine, while the control group received buffers. At 16 weeks after the last immunization, all macaques were challenged orally with pathogenic SIVmac239. The mean plasma SIV RNA loads of the vaccine group were significantly lower than those of the placebo control group up to 16 weeks post-challenge. The vaccine-induced Gag-specific IFN-gamma ELISPOT T cell responses inversely correlated with the viral loads before the chronic phase. Two out of six vaccinated macaques with strong and sustained Gag-specific T cell responses showed viremia control and maintained CD4+ T cell percentage. However, the other four vaccinated macaques showed high viral loads and reduced level of CD4+ T cell percentages during the chronic phase, comparable to those in control macaques. Five out of six vaccinated macaques survived for more than 72 weeks, while five out of six controls died of an AIDS-related disease. Therefore, the vaccination conferred not only reduction of viral loads in a portion of vaccinated macaques (2/6), but also prolonged survival of all vaccinated macaques regardless of viremia control. Our results further suggest that new experimental approaches may be needed to assess protective effects from AIDS-associated disease in the immunized macaques after oral SIV challenge.

Background Due to an ever increasing shortage of rhesus macaques ofIndian origin (InR) that have been generally used for preclinical AIDS vac-cine trials in non-human primates, demand is rising for Chinese rhesusmacaques (ChR). However, the immunogenicity of an AIDS vaccine candi-date has not been compared in parallel in both rhesus macaque subspecies.Methods ChR and InR were immunized with SIV/HIV DNA andadenovirus vaccine and their immune responses to SIV and HIV evaluated.Results SIV Gag- and Env-specific T-cell responses and SIV-specificlymphoproliferative responses measured in ChR were significantly weakerthan those in InR (P < 0.05). By contrast, antibody responses to SIV Env,Tat, and Nef in ChR were stronger than those in InR (P < 0.05).Conclusions Immunogenicity of an AIDS vaccine can vary significantlydepending on the geographic origin implying genetic differences of maca-ques. The Laser Interferometer Space Antenna (LISA) Mission will enable scientists to detect and characterize gravitational waves emitted by different sources and events in our universe. LISA consists of three spacecraft flying in a triangular constellation with an arm-length of 5 mio. km in a heliocentric orbit approximately 20 degrees behind or in front of the Earth. One core technology required in order to achieve the mission requirements is the establishment of excellent free-fall conditions for the two proof-masses on board of each of the three spacecraft. The proof-masses must be isolated from external disturbances such that the residual acceleration remains below 3×10 m/s/√Hz between 0.1 and 1 mHz. In addition to the free-fall conditions, the relative jitter between the proof-masses and the spacecraft must be controlled to 2 nm/√Hz between 1 mHz and 1 Hz in order to ensure that the optical metrology achieves its required performance. Last but not least, the instrument pointing must be controlled to 8 nrad/√Hz between 1 mHz and 1 Hz in order to ensure that the inter-spacecraft laser-link achieves its required metrology performance. In this paper, a methodology is developed such that requirements for all control loops can be derived from system level requirements. Based on the requirements for each individual control loop, the feasibility of a controller design can be ensured even before an actual design has been performed. A reference design is then tested on a fully coupled linear model and verified in the framework of a non-linear time-domain simulation..

LISA, the Laser Interferometer Space Antenna, will be launched by NASA and ESA in the 2014 timeframe and will be the first gravitational wave observatory for the many fascinating sources at low frequencies (down to 10−5 Hz). One of the challenges is to perform precision laser interferometry over a distance of 5 million km. This paper describes recent developments in the interferometer design.

In order to achieve the required measurement performance on LISA, the laser frequency must be stabilized to approximately 30 Hz/Hz×[1+(1mHz/f)4]1/2 in the LISA measurement bandwidth from 0.03 mHz up to 1 Hz for the master laser in the constellation. All other lasers are offset locked to the master laser such that the Doppler shifts are taken into account and beat signals between 3 MHz and 18 MHz are produced on all detectors in the constellation. Ensuring sufficient frequency stability can be established by different methods. A straight forward approach is to use an optical cavity. It turns out that a cavity alone significantly drives the thermal stability requirements at low frequencies. Therefore, different versions of arm‐locking are considered in order to provide both, frequency stabilization at low frequencies as well as at high frequencies. An overview of the LISA measurement chain is given and the design of the spacecraft with the main focus on the LISA payload is described. The main highlights of the current payload design baseline are the introduction of the strap‐down system in connection with a dedicated actuator for the point‐ahead angle compensation, and the laser frequency swap. The strap‐down system separates the problems of the 5 mio. km interferometry from the proof‐mass metrology. This gives more design freedom in the mechanical configuration, a clear separation of the different subsystems, and it also improves the testability on ground. The laser frequency swap significantly reduces the problems of straylight and also yields a clear separation between the incoming and outgoing laser beam which introduces the possibility of an optimal adjustment of the local oscillator independently from the outgoing beam. Both design changes are presented and their impacts on the overall system design and the measurement performance is discussed. Compared to the last study in 2000, the mechanical design has changed in an effort of reducing the overall system mass while keeping a reliable and technologically feasible design. The design baseline is supported by the corresponding structural, thermal, drag‐free and attitude control, vacuum, and optical analyses such that the revised error budgets are now supported by results from more detailed models instead of rough estimates. Finally, an overview of the latest performance estimates is given and the derived requirements on the main subsystems are discussed. The paper presents the current status of the LISA system design as it is established in the framework of the European Space Agency’s Mission Formulation Study at EADS Astrium.

As part of the current LISA Mission Formulation Study, and based on prior internal investigations, Astrium Germany has defined and preliminary assessed novel payload architectures, potentially reducing overall complexity and improving budgets and costs. A promising concept is characterized by a single active inertial sensor attached to a single optical bench and serving both adjacent interferometer arms via two rigidly connected off‐axis telescopes. The in‐plane triangular constellation “breathing angle” compensation is accomplished by common telescope in‐field of view pointing actuation of the transmit/received beams line of sight. A dedicated actuation mechanism located on the optical bench is required in addition to the on bench actuators for differential pointing of the transmit and receive direction perpendicular to the constellation plane. Both actuators operate in a sinusoidal yearly period. A technical challenge is the actuation mechanism pointing jitter and the monitoring and calibration of the laser phase walk which occurs while changing the optical path inside the optical assembly during re‐pointing. Calibration or monitoring of instrument internal phase effects e.g. by a laser metrology truss derived from the existing interferometry is required. The architecture exploits in full the two‐step interferometry (strap down) concept, separating functionally inter spacecraft and intra‐spacecraft interferometry (reference mass laser metrology degrees of freedom sensing). The single test mass is maintained as cubic, but in free‐fall in the lateral degrees of freedom within the constellation plane. Also the option of a completely free spherical test mass with full laser interferometer readout has been conceptually investigated. The spherical test mass would rotate slowly, and would be allowed to tumble. Imperfections in roundness and density would be calibrated from differential wave front sensing in a tetrahedral arrangement, supported by added attitude information via a grid of tick marks etched onto the surface and monitored by the laser readout.

The primary mission of a wind scatterometer is to determine wind speed and direction over the ocean. This is achieved by performing a set of radar cross-section measurements at different azimuth view-angles over the resolution cell, and inverting the backscatter model, a so-called geophysical model function (GMF), to extract the wind information using the azimuth anisotropy of the radar backscatter by sea-surface in presence of wind. A new concept of rotating fanbeam radar was introduced which operates in C-band. The present paper describes an analysis of the new concept by means of wind retrieval simulations and an investigation of advanced features such as multi-beam, dual-polarisation, dual-frequency and polarimetric capabilities in improving the wind retrieval accuracy. End-to-end simulations of the complete system are performed starting from wind-fields which are sampled by the scatterometer model. The simulated radar echos are then converted to sets of backscattering coefficients (sigma-naught) which are inverted to obtain again the wind-fields containing measurement errors and noise. The performance of the system is assessed by analysing the quality of retrieved wind as functions of the instrument configuration and characteristics (parameters).

As part of the on-going LISA Mission Formulation study under ESA contract, EADS Astrium has recently suggested and investigated a variety of novel LISA payload architectures utilizing so-called "In-Field Pointing" for accommodation of seasonal constellation dynamics. Here, the annual variation in the angle between the interferometer arms of roughly ±1° is compensated by steering the lines of sight of the individual telescopes with a small actuated mirror located in an intermediate pupil plane inside the telescopes. This introduces a certain flexibility for the overall payload configuration and allows for very compact designs. In particular, it enables a "single active proof mass" mode with a true cold redundancy between a nominal and a backup GRS system on board each spacecraft, and thus enhances mission robustness.

In the context of the LISA Mission Formulation Study, the LISA System was studied in detail and a new baseline architecture for the whole mission was established. This new baseline is the result of trade-offs on both, mission and system level. The paper gives an overview of the different mission scenarios and configurations that were studied in connection with their corresponding advantages and disadvantages as well as performance estimates. Differences in the required technologies and their influence on the overall performance budgets are highlighted for all configurations. For the selected baseline concept, a more detailed description of the configuration is given and open issues in the technologies involved are discussed.

Background Since there is still no protective HIV vaccine available, better insights into immune mechanism of persons effectively controlling HIV replication in the absence of any therapy should contribute to improve further vaccine designs. However, little is known about the mucosal immune response of this small unique group of patients. Using the SIV-macaque-model for AIDS, we had the rare opportunity to analyze 14 SIV-infected rhesus macaques durably controlling viral replication (controllers). We investigated the virological and immunological profile of blood and three different mucosal tissues and compared their data to those of uninfected and animals progressing to AIDS-like disease (progressors). Results Lymphocytes from blood, bronchoalveolar lavage (BAL), and duodenal and colonic biopsies were phenotypically characterized by polychromatic flow cytometry. In controllers, we observed higher levels of CD4+, CD4+CCR5+ and Gag-specific CD8+ T-cells as well as lower immune activation in blood and all mucosal sites compared to progressors. However, we could also demonstrate that immunological changes are distinct between these three mucosal sites. Intracellular cytokine staining demonstrated a significantly higher systemic and mucosal CD8+ Gag-specific cellular immune response in controllers than in progressors. Most remarkable was the polyfunctional cytokine profile of CD8+ lymphocytes in BAL of controllers, which significantly dominated over their blood response. The overall suppression of viral replication in the controllers was confirmed by almost no detectable viral RNA in blood and all mucosal tissues investigated. Conclusion A strong and complex virus-specific CD8+ T-cell response in blood and especially in mucosal tissue of SIV-infected macaques was associated with low immune activation and an efficient suppression of viral replication. This likely afforded a repopulation of CD4+ T-cells in different mucosal compartments to almost normal levels. We conclude, that a robust SIV-specific mucosal immune response seems to be essential for establishing and maintaining the controller status and consequently for long-term survival.
