An oligopeptidase from the probiotic nonstarter strain Lactobacillus rhamnosus HN001 (DR20) cleaves alpha(sub s1)-casein fragment 1-23 at Pro-5-Ile-6, Lys-7-His-8, His-8-Gln-9, and Gln-9-Gly-10. Peptidases from lactic acid bacteria play an important role in cheese flavor.

A study has revealed that the low-water activity and high-salt-concentration characteristic of cheese imparts lactocepin III characteristics to lactocepin I. The effects of these parameters on the specificity of lactocepin III on the stability of lactocepin I and III were accurately determined. Findings indicate that proteolysis can elucidate certain key factors in cheese flavor development, since proteolytic enzymes are instrumental in the ripening process of most cheeses.

The specificity of hydrolysis of alpha-s1-, beta-, and kappa-caseins by the cell envelope-associated proteinase lactocepin from Lactococus lactis subspecie lactis BN1 was examined at water activities [a(sub w)] and salt concentrations reflecting those in cheddar cheese by using various humectant systems. Findings showed that the specificity of lactocepin was dependent on a (sub w) and medium hydrophobicity. It was clear that the characteristic differences of the lactocepin types were not fully reflected in the cheese environment.

Cells of Lactococcus lactis subsp. cremoris H2 were subjected to lysozyme treatment in the presence of 50 mM calcium chloride. The treatment resulted in the release of three forms of proteinases, having molecular weights of 137-, 145- and 180-kDa, respectively. The former two proteinases were similar to those produced in the absence of calcium ions, and exist as dimers in their native forms. The 180-kDa form, meanwhile, is probably a larger aggregate of the native form. The isolation of these enzymes in the presence of calcium is an important step in establishing correlations between in vitro activity and the in situ process.

- Abstract
Provision of suitable habitat for waterbirds is a major challenge for environmental managers in arid and semiarid regions with high spatial and temporal variability in rainfall. It is understood in broad terms that to survive waterbirds must move according to phases of wet-dry cycles, with coastal habitats providing drought refugia and inland wetlands used during the wet phase. However, both inland and coastal wetlands are subject to major anthropogenic pressures, and the various species of waterbird may have particular habitat requirements and respond individualistically to spatiotemporal variations in resource distribution. A better understanding of the relationships between occurrence of waterbirds and habitat condition under changing climatic conditions and anthropogenic pressures will help clarify patterns of habitat use and the targeting of investments in conservation. We provide the first predictive models of habitat availability between wet and dry phases for six widely distributed waterbird species at a large spatial scale. We first test the broad hypothesis that waterbirds are largely confined to coastal regions during a dry phase. We then examine the contrasting results among the six species, which support other hypotheses erected on the basis of their ecological characteristics. There were large increases in area of suitable habitat in inland regions in the wet year compared with the dry year for all species, ranging from 4.14% for Australian White Ibis to 31.73% for Eurasian Coot. With over half of the suitable habitat for three of the six species was located in coastal zones during drought, our study highlights the need to identify and conserve coastal drought refuges. Monitoring of changes in extent and condition of wetlands, combined with distribution modeling of waterbirds, will help support improvements in the conservation and management of waterbirds into the future.

The action of the cell-wall-associated proteinases from Lactococcus lactis subsp. cremoris strains H2 and SK112 on bovine ß-casein was compared. The proteinase from the H2 strain was characterised as a P
I -type proteinase since it did not hydrolyse as1 -casein and the initial trifluoroacetic acid-soluble products of ß-casein hydrolysis were identical to those previously identified as hydrolysis products of PI -type lactococcal proteinase action. The time-course of product formation by the proteinase from the H2 strain indicated that the bonds Tyr193 -Gln194 and Gln182 -Arg183 were the first to be hydrolysed. Cleavage of the bonds Gln175 -Lys176 , Ser168 -Lys169 , Ser166 -Gln167 and Leu163 -Ser164 was also very rapid. Four of the five bonds in ß-casein most susceptible to hydrolysis by the PIII -type proteinase from strain SK112 were different from those cleaved by the PI -type proteinase, initial hydrolysis being at the sites Tyr193 -Gln194 , Leu192 -Tyr193 , Asp43 -Glu44 , Gln46 -Asp47 and Phe52 -Ala53 . Early hydrolysis at the three sites in the N-terminal region of ß-casein, leading to cleavage of the N-terminal phosphopeptide and rapid precipitation of the residual fragment, represents a marked contrast to the action of PI -type proteinases where cleavage at sites in the N-terminal region occurs only very slowly.- Published

The cell wall-associated proteinase from Lactococcus lactis subsp. cremoris SK11 was partially purified and incubated with a
s1 -casein for various times up to 48 h. Sixteen trifluoroacetic acid-soluble oligopeptide hydrolysis products were identified by determination of the aminp acid sequence. Eleven of these oligopeptides originated from the 78-residue sequence comprising the C-terminal region of as1 -casein and were present among the products after the first 60 min of digestion. Three oligopeptides from the N-terminal region and two others from the central region of the as1 -casein sequence were also present among the early digestion products although in smaller amounts than most of the oligopeptides from the C-terminal region. No cleat consensus sequence of amino acid residues surrounding the cleavage sites could be identified.- Published
