The severe life-threatening effects of systemic chemotherapy are well documented. Doxorubicin, DOX is the most effective antineoplastic agent but with the least specific action that is responsible for severe cardiotoxicity and myelosuppression that necessitates careful monitoring while administering. Stimuli-sensitive/intelligent drug delivery systems, specifically those utilizing temperature as an external stimulus to activate the release of encapsulated drugs, have become a subject of recent research. Thus, it would be ideal to have a nanocarrier comprising safe excipients and controllable drug release capacity to deliver the drug at a particular site to minimize unwanted and toxic effects of chemotherapeutics. We have developed a simple temperature-responsive nanocarrier based on eutectic mixture of fatty acids. This study aimed to develop, physicochemically characterize and investigate the biological safety of eutectic mixture of fatty acids as a novel construct for temperature-responsive drug release potential.Methods: We have developed phase change material, PCM, based on a series of eutectic mixtures of fatty acids due to their unique and attractive physicochemical characteristics such as safety, stability, cost-effectiveness, and ease of availability. The reversible solid-liquid phase transition of PCM is responsible to hold firm or actively release the encapsulated drug. The eutectic mixtures of fatty acids (stearic acid and myristic acid) along with liquid lipid (oleic acid) were prepared to exhibit a tunable thermoresponsive platform. Doxorubicin-loaded lipid nanocarriers were successfully developed with combined hot melt encapsulation (HME) and sonication method and characterized to achieve enhanced permeability and retention (EPR) effect-based solid tumor targeting in response to exogenous temperature stimulus. The cytotoxicity against melanoma cell lines and in vivo safety studies in albino rats was also carried out.Results: Doxorubicin-loaded lipid nanocarriers have a narrow size distribution (94.59– 219.3 nm), and a PDI (0.160– 0.479) as demonstrated by photon correlation microscopy and excellent colloidal stability (Z.P value: − 22.7 to − 32.0) was developed. Transmission electron microscopy revealed their spherical morphology and characteristics of a monodispersed system. A biphasic drug release pattern with a triggered drug release at 41°C and 43°C and a sustained drug release was observed at 37°C. The thermoresponsive cytotoxic potential was demonstrated in B16F10 cancer cell lines. Hemolysis assay and acute toxicity studies with drug-free and doxorubicin lipid nanocarrier formulations provided evidence for their non-toxic nature.Conclusion: We have successfully developed a temperature-responsive tunable platform with excellent biocompatibility and intelligent drug release potential. The formulation components being from natural sources present superior characteristics in terms of cost, compatibility with normal body cells, and adaptability to preparation methods. The reported preparation method is adapted to avoid complex chemical processes and the use of organic solvents. 3. Interplay between $M_W$, $\Omega_{\rm CDM} h^2$, and $(g-2)_{\mu}$ in Flavor Symmetry-Based Supersymmetric Models

Abstract

We study the phenomenological implications of the minimal supersymmetric standard model (MSSM) augmented by a non-abelian flavor symmetry labeled as sMSSM. Incorporating this flavor symmetry allows for a significant reduction in the original plethora of free parameters present in the MSSM, ultimately reducing them down to just seven in sMSSM. This reduction of free parameters is not achieved through ad hoc assumptions like in the constrained MSSM (CMSSM); rather, it is grounded in theoretical considerations. Our work focuses on exploring the interplay between the $W$ boson mass ($M_W$) predictions, the cold dark matter (CDM) relic abundance ($\Omega_{\rm CDM} h^2$), and the $(g-2)_{\mu}$ anomaly. We identified correlations among the theoretical parameters arising from this interplay, which can be complemented by experimental constraints such as the Higgs boson mass, B-physics observables, and charge and color breaking minima. Additionally, our investigations show that the $(g-2)_{\mu}$ discrepancy and the Planck bounds on $\Omega_{\rm CDM} h^2$ can be addressed within the sMSSM, but only in a very narrow region of the parameter space.

The data was collected from T2DM out-patient clinics. The participants were recruited by using a simple random sampling method. To assess the association of demographics with the level of adherence, binary logistics regression analysis was applied. Moreover, the Mann–Whitney U-test was used to evaluate the impact of adherence on QoL.Results: A total of 384 patients participated in this study. Amongst them, 60.2% were male and 39.8% were female. Low adherence was seen in illiterate patients and patients older than 40 years. The results showed that good QoL in T2DM patients was significantly associated with treatment adherence (p= 0.004).Conclusion: The finding of the current study showed that the non-adherence prevailed in illiterate strata of the study population and the medication adherence significantly affects the QoL in T2DM patients. These findings suggest that health regulatory agencies should focus on implementing disease-education interventions for improving the adherence to medications in patients with long-term conditions.Keywords: type 2 diabetes, drug adherence, quality of life

Docetaxel is relatively less hydrophobic and twice as potent as paclitaxel. Like other taxane chemotherapeutic agents, docetaxel is not well tolerated and shows toxicity in the patients. Nanoencapsulation of potent chemotherapeutic agents has been shown to improve tolerability and therapeutic outcome. Therefore, the present study was designed to fabricate chitosan and sodium tripolyphosphate (STPP) based on ionically cross-linked nanoparticles for sustained release of docetaxel.Methods: Nanoparticles were prepared by the ionic-gelation method by dropwise addition of the STPP solution into the chitosan solution in different ratios. CNPs were characterized for post-formulation parameters like size, zeta potential, scanning electron microscope (SEM), FTIR, DSC/TGA, pXRD, and in-vitro drug release, as well as for acute oral toxicity studies in Wistar rats.Results and discussion: The optimized docetaxel loaded polymeric nanoparticles were in the size range (172.6nm–479.65 nm), and zeta potential (30.45–35.95 mV) required to achieve enhanced permeation and retention effect. In addition, scanning electron microscopy revealed rough and porous surface, whereas, FTIR revealed the compatible polymeric nanoparticles. Likewise, the thermal stability was ensured through DSC and TG analysis, and powder X-ray diffraction analysis exhibited solid-state stability of the docetaxel loaded nanoparticles. The in-vitro drug release evaluation in phosphate buffer saline (pH 7.4) showed sustained release pattern, i.e. 51.57–69.93% within 24 hrs. The data were fitted to different release kinetic models which showed Fickian diffusion as a predominant release mechanism (R2= 0.9734–0.9786, n= 0.264–0.340). Acceptable tolerability was exhibited by acute oral toxicity in rabbits and no abnormality was noted in growth, behavior, blood biochemistry or histology and function of vital organs.Conclusion: Ionically cross-linked chitosan nanoparticles are non-toxic and biocompatible drug delivery systems for sustained release of chemotherapeutic agents, such as docetaxel.Keywords: tripolyphosphate, ionic gelation, sustained release, targeting, biocompatible, biodegradable

Thus, chemotherapeutic drugs, including Docetaxel, are administered via an intravenous route, which poses many disadvantages of its own. Recent advances in pharmaceutical research have focused on designing new and efficient drug delivery systems for site-specific targeting, thus leading to improved bioavailability and pharmacokinetics. A decent number of studies have been reported for the safe and effective oral delivery of Docetaxel. These nanocarriers, including liposomes, polymeric nanoparticles, metallic nanoparticles, hybrid nanoparticles, dendrimers and so on, have shown promising results in research papers and clinical trials. The present article comprehensively reviews the research efforts made so far in designing various advancements in the oral delivery of Docetaxel. Different strategies to improve oral bioavailability, prevent first-pass metabolism and inhibition of efflux pumping leading to improved pharmacokinetics and anticancer activity are discussed. The final portion of this review article presents key issues such as safety of nanomaterials, regulatory approval and future trends in nanomedicine research. These systems exploit hyperthermic tempera­tures (>39°C), which may be applied by some external means or due to an encountered symptom in inflammatory diseases such as cancer and arthritis. The objective of this paper was to provide some solid evidence in support of the hypothesis that solid lipid nanoparticles (SLNs) can be used for thermoresponsive targeting by undergoing solid–liquid phase transition at their melting point (MP). Thermoresponsive lipid mixtures were prepared by mixing solid and liquid natural fatty acids, and their MP was measured by differential scanning calorimetry (DSC). SLNs (MP 39°C) containing 5-fluorouracil (5-FU) were synthesized by hot melt encapsulation method, and were found to have spherical shape (transmission electron microscopy studies), desirable size (90% drug was released at 39°C after 5 hours, suggesting that the SLNs show thermoresponsive drug release, thus confirming our hypothesis. Drug release from SLNs at 39°C was similar to oleic acid and linoleic acid nanoemulsions used in this study, which further confirmed that thermoresponsive drug release is due to solid–liquid phase transition. Next, a differential pulse voltammetry-based electrochemical chemical detection method was developed for quick and real-time analysis of 5-FU release, which also confirmed thermoresponsive drug release behavior of SLNs. Blank SLNs were found to be biocompatible with human gingival fibroblast cells, although 5-FU-loaded SLNs showed some cytotoxicity after 24 hours. 5-FU-loaded SLNs showed thermoresponsive cytotoxicity to breast cancer cells (MDA-MB-231) as cytotoxicity was higher at 39°C (cell viability 72%–78%) compared to 37°C (cell viability >90%) within 1 hour. In conclusion, this study presents SLNs as a safe, simple, and effective platform for thermoresponsive targeting. Abstract

Sixth-generation (6G) networks pose substantial security risks because confidential information is transmitted over wireless channels with a broadcast nature, and various attack vectors emerge. Physical layer security (PLS) exploits the dynamic characteristics of wireless environments to provide secure communications, while reconfigurable intelligent surfaces (RISs) can facilitate PLS by controlling wireless transmissions. With RIS-aided PLS, a lightweight security solution can be designed for low-end Internet of Things (IoT) devices, depending on the design scenario and communication objective. This article discusses RIS-aided PLS designs for 6G-IoT networks against eavesdropping and jamming attacks. The theoretical background and literature review of RIS-aided PLS are discussed, and design solutions related to resource allocation, beamforming, artificial noise, and cooperative communication are presented. We provide simulation results to show the effectiveness of RIS in terms of PLS. In addition, we examine the research issues and possible solutions for RIS modeling, channel modeling and estimation, optimization, and machine learning. In order to optimize SLNs for temperature-responsive release, lipid mixtures were prepared using different ratios of solid (stearic acid or lauric acid) and liquid (oleic acid) fatty acids. SLNs were prepared by microemulsification (53 nm), hot melt encapsulation (10.4 nm), and a solvent emulsification-evaporation technique (7.8 nm). The physicochemical characteristics of SLNs were studied by Zetasizer, Fourier transform infrared, and X-ray diffraction analysis. High encapsulation of diacerein was achieved with diacerein-loaded and simultaneously GNP-diacerein-loaded SLNs. In vitro dissolution studies revealed a sustained release pattern for diacerein over 72 hours for diacerein-loaded SLNs and 12 hours for GNP-diacerein-loaded SLNs. An increase in diacerein payload increased the release time of diacerein while GNPs decreased it. In addition, rapid release of diacerein over 4 hours was observed at 40°C (melting point of optimized fatty acid mixture), demonstrating that these binary SLNs could be used for thermoresponsive drug delivery. Energy requirement of the world is increasing day by day, and using conventional methods to fulfil this requirement might cause devastating effects on atmosphere due to an emission of different harmful by-products released. 26. Mapping Temporary Slums from Satellite Imagery using a Semi-Supervised Approach

Abstract

One billion people worldwide are estimated to be living in slums, and documenting and analyzing these regions is a challenging task. As compared to regular slums; the small, scattered and temporary nature of temporary slums makes data collection and labeling tedious and time-consuming. To tackle this challenging problem of temporary slums detection, we present a semi-supervised deep learning segmentation-based approach; with the strategy to detect initial seed images in the zero-labeled data settings. A small set of seed samples (32 in our case) are automatically discovered by analyzing the temporal changes, which are manually labeled to train a segmentation and representation learning module. The segmentation module gathers high dimensional image representations, and the representation learning module transforms image representations into embedding vectors. After that, a scoring module uses the embedding vectors to sample images from a large pool of unlabeled images and generates pseudo-labels for the sampled images. These sampled images with their pseudo-labels are added to the training set to update the segmentation and representation learning modules iteratively. To analyze the effectiveness of our technique, we construct a large geographically marked dataset of temporary slums. This dataset constitutes more than 200 potential temporary slum locations (2.28 square kilometers) found by sieving sixty-eight thousand images from 12 metropolitan cities of Pakistan covering 8000 square kilometers. Furthermore, our proposed method outperforms several competitive semi-supervised semantic segmentation baselines on a similar setting. The code and the dataset will be made publicly available.

Abstract

BackgroundLevels of plasma SARS-CoV-2 nucleocapsid (N) antigen may be an important biomarker in patients with COVID-19 and enhance our understanding of the pathogenesis of COVID-19.ObjectiveTo evaluate whether levels of plasma antigen can predict short-term clinical outcomes and identify clinical and viral factors associated with plasma antigen levels in hospitalized patients with SARS-CoV-2.DesignCross-sectional study of baseline plasma antigen level from 2540 participants enrolled in the TICO (Therapeutics for Inpatients With COVID-19) platform trial from August 2020 to November 2021, with additional data on day 5 outcome and time to discharge.Setting114 centers in 10 countries.ParticipantsAdults hospitalized for acute SARS-CoV-2 infection with 12 days or less of symptoms.MeasurementsBaseline plasma viral N antigen level was measured at a central laboratory. Delta variant status was determined from baseline nasal swabs using reverse transcriptase polymerase chain reaction. Associations between baseline patient characteristics and viral factors and baseline plasma antigen levels were assessed using both unadjusted and multivariable modeling. Association between elevated baseline antigen level of 1000 ng/L or greater and outcomes, including worsening of ordinal pulmonary scale at day 5 and time to hospital discharge, were evaluated using logistic regression and Fine-Gray regression models, respectively.ResultsPlasma antigen was below the level of quantification in 5% of participants at enrollment, and 1000 ng/L or greater in 57%. Baseline pulmonary severity of illness was strongly associated with plasma antigen level, with mean plasma antigen level 3.10-fold higher among those requiring noninvasive ventilation or high-flow nasal cannula compared with room air (95% CI, 2.22 to 4.34). Plasma antigen level was higher in those who lacked antispike antibodies (6.42 fold; CI, 5.37 to 7.66) and in those with the Delta variant (1.73 fold; CI, 1.41 to 2.13). Additional factors associated with higher baseline antigen level included male sex, shorter time since hospital admission, decreased days of remdesivir, and renal impairment. In contrast, race, ethnicity, body mass index, and immunocompromising conditions were not associated with plasma antigen levels. Plasma antigen level of 1000 ng/L or greater was associated with a markedly higher odds of worsened pulmonary status at day 5 (odds ratio, 5.06 [CI, 3.41 to 7.50]) and longer time to hospital discharge (median, 7 vs. 4 days; subhazard ratio, 0.51 [CI, 0.45 to 0.57]), with subhazard ratios similar across all levels of baseline pulmonary severity.LimitationsPlasma samples were drawn at enrollment, not hospital presentation. No point-of-care test to measure plasma antigen is currently available.ConclusionElevated plasma antigen is highly associated with both severity of pulmonary illness and clinically important patient outcomes. Multiple clinical and viral factors are associated with plasma antigen level at presentation. BackgroundEnsovibep (MP0420) is a designed ankyrin repeat protein, a novel class of engineered proteins, under investigation as a treatment of SARS-CoV-2 infection.ObjectiveTo investigate if ensovibep, in addition to remdesivir and other standard care, improves clinical outcomes among patients hospitalized with COVID-19 compared with standard care alone.DesignDouble-blind, randomized, placebo-controlled, clinical trial. 45. Vacuum Stability Constraints on Flavor Mixing Parameters and Their Effect on Gluino Decays

Abstract

In this article, we study the two body gluino decays $\tilde{g} \rightarrow \tilde{u}_i \bar{u}_g$, $\tilde{g} \rightarrow \tilde{d}_i \bar{d}_g$ with $i=1,2,...6$ and $g=1,2,3$ in the Minimal Supersymmetric Standard Model (MSSM) with quark flavour violation (QFV). At the outset, constraints on QFV parameters $\delta _{ij}^{U/D(LR)}$ and $\delta _{ij}^{U/D(RL)}$ with $i,j=1,2,3$ and $(i\neq j) $ are calculated for a specific set of MSSM parameters using charge and color breaking minima (CCB) and unbounded from below minima (UFB) conditions. These constraints are more stringent compared to the constraints coming from B-physics observables (BPO), that are already available in literature. In the second step, we re-calculate the partial decay widths of $\tilde{g} \rightarrow \tilde{u}_i \bar{u}_g$, $\tilde{g} \rightarrow \tilde{d}_i \bar{d}_g$ for the allowed range of QFV parameters. The partial decay widths can reach upto 120 GeV in the RR sector of the squarks mass matrices while in the LL sector it can be $\approx 90$ GeV. The QFV in the LR/RL sector is usually ignored due to stringent CCB andd UFB constraints, however, our analysis reveals that this mixing can contribute upto $\approx 12 $ GeV for some parameter points and should not be ignored. We hope that these results will prove helpful for the experimental searches of gluinos at the current and future colliders.

48. Excited lepton triplet contribution to electroweak observables at one loop level

Abstract

In this paper, we present the one-loop radiative corrections to the electroweak precision observable $\Delta \rho$ coming from the $I_W=1$ multiplet excited leptons. We have calculated the couplings of the exotic lepton triplet to the vector bosons and ordinary leptons using effective Lagrangian approach. These couplings are then used to estimate the excited lepton triplet contribution to the $\Delta \rho$ parameter. The mass degenerate excited lepton contribution to $\Delta \rho $ is small and can be neglected. However, if the excited leptons are non-degenerate, their contribution can be large which can result in more stringent constraints on the excited fermion parameter space compared to the constraints from present experimental searches and perturbative unitarity condition.

Abstract

Relative drifts between different species or particle populations are characteristic to solar plasma outflows, e.g., in the fast streams of the solar winds, coronal mass ejections and interplanetary shocks. This paper characterizes the dispersion and stability of the low-beta alpha/proton drifts in the absence of any intrinsic thermal anisotropies, which are usually invoked in order to stimulate various instabilities. The dispersion relations derived here describe the full spectrum of instabilities and their variations with the angle of propagation and plasma parameters. The results unveil a potential competition between instabilities of the electromagnetic proton cyclotron and alpha cyclotron modes. For conditions specific to a low-beta solar wind, e.g., at low heliocentric distances in the outer corona, the instability operates on the alpha cyclotron branch. The growth rates of the alpha cyclotron mode are systematically stimulated by the (parallel) plasma beta and/or the alpha-proton temperature ratio. One can therefore expect that this instability develops even in the absence of temperature anisotropies, with potential to contribute to a self-consistent regulation of the observed drift of alpha particles., Comment: Accepted for publication at Astrophysics and Space Science

Abstract

The emission of muonium ($\mu^+e^-$) atoms into vacuum from silica aerogel with laser ablation on its surface was studied with various ablation structures at room temperature using the subsurface muon beams at TRIUMF and Japan Proton Accelerator Research Complex (J-PARC). Laser ablation was applied to produce holes or grooves with typical dimensions of a few hundred $\mu$m to a few mm, except for some extreme conditions. The measured emission rate tends to be higher for larger fractions of ablation opening and for shallower depths. More than a few ablation structures reach the emission rates similar to the highest achieved in the past measurements. The emission rate is found to be stable at least for a couple of days. Measurements of spin precession amplitudes for the produced muonium atoms and remaining muons in a magnetic field determine a muonium formation fraction of $(65.5 \pm 1.8)$%. The precession of the polarized muonium atoms is also observed clearly in vacuum. A projection of the emission rates measured at TRIUMF to the corresponding rates at J-PARC is demonstrated taking the different beam condition into account reasonably., Comment: 34 pages, 13 figures, submitted to PTEP
