Abstract

Reg protein was first found in pancreatic stones. It was named Pancreatic Stone Protein and later renamed lithostathine, as it was assumed to prevent stone formation. The 144 amino acid protein is O-glycosylated on Thr-5. The glycan chain is variable in length and in charge. Lithostathine 3-D organization is of the C-lectin type, even though it is unlikely to have any functional calcium-binding site. The Arg11-Ile12 bond is readily cleaved by trypsin; the resulting C-terminal polypeptide precipitates at physiological pH and tends to form fibrils. The protein was more recently found in the regenerating endocrine pancreas and it was named Reg (for regenerating) protein. Numerous proteins related to Reg have been identified successively in several mammalian species. They constitute the Reg superfamily. Reg genes show the same organization and are located in the same chromosome region. These genes are therefore likely to derive from a common ancestor gene by duplication. In the course of evolution, they may have diverged in tissue-related expression and function. In the endocrine pancreas, Reg protein stimulates islet beta-cell growth and reduces experimental diabetes via the activation of a high affinity receptor. The role of the protein produced by the exocrine pancreas, however, is controversial. Not only is Reg/lithostathine unlikely to be a physiologically relevant pancreatic stone inhibitor, but it may contribute to stone formation. We suggest that it might help prevent the harmful activation of protease precursors in the pancreatic juice. The protein provides a useful model for examining the conformational changes associated with globular to fibril transformation.

10. Polypeptide effect on Mg2+ hydration inferred from CaCO3 formation: A biomineralization study by counter-diffusion

Abstract

The use of a counter-diffusion system allows the evaluation of diverse parameters involved in a crystallization process. In this study, this tool has been used to infer the hydration status of Mg during CaCO formation experiments in an agarose highly viscous sol entrapping charged polypeptides. The experimental data allow us to infer that the hydration status of Mg is altered by the presence of poly-l-aspartate or poly-l-glutamate. This changes the CaCO polymorphic distribution in favor of Mg-calcite with respect to aragonite, but does not favor the isomorphic substitution of Mg with Ca within the calcite lattice. The latter may exclude the formation of an amorphous transient form, which leads to a high Mg-calcite, as expected when using a counter-diffusion system set up. The presence of poly-l-lysine does not affect the hydration of Mg, but favors the formation of aragonite with respect to calcite. In this case an inhibition of calcite formation and an alteration of the hydration sphere of Ca could be invoked; both effects are able to increase CaCO supersaturation. In conclusion, this study reveals that charged polypeptides can orchestrate CaCO formation by also controlling the hydration status of cations.

Abstract

Dal 2008, in testa all'indice dei cosiddetti “failed states” la Somalia riveste un ruolo di primo piano a livello massmediatico quando si parli di guerre civili, violenza, terrorismo, devastazione, tribalismo, perdita di speranze. La clessidra del tempo contemporaneo somalo ha subito una fortissima scossa nel 1991 in seguito al crollo dell'allora Repubblica Democratica Somala e i conseguenti conflitti che, in molte aree dell'ex-Stato Somalo, perdurano. Per alcuni quella frattura storica collettiva ha segnato anche il tempo del proprio fermarsi, bloccati dal peso della sofferenza in una condizione di riconosciuto disagio mentale. Molti, siano essi rifugiati in paesi terzi o rimasti in Somalia, hanno fatto esperienza di atrocità “letteralmente di là dall’immaginazione per la maggior parte delle persone”(Malkki,1995). Ciononostante non hanno bisogno di attenzione “psichiatrica” oppure non manifestano alcun quadro clinico ascrivibile alle narrative dominanti della memoria traumatica oppure, ancora, non sono inquadrabili in un quadro nosologico internazionalmente riconosciuto. L'attenzione, poca, al tema della salute mentale in Somalia si è concentrata su queste dimensioni essenzializzandolo e riducendolo a un rapporto tra l'individuo e il proprio passato (“traumatico”) o tra il primo e la violenza (intesa come atto di rottura comprensibile a uno sguardo occidentale). Con questo lavoro si mostrerà come sia necessario affrancarsi da approcci medicalizzanti dell'esperienza di guerra (Almedon et al., 2004) per dare senso, attraverso il lavoro etnografico, all'articolazione tra biografie individuali e dinamiche sociali nel quadro delle trasformazioni in corso nella società Somala contemporanea. Portando al centro del discorso l'esperienza di sofferenza delle famiglie e la comprensione locale delle forme di disagio mentale, ci chiederemo inoltre se alcune di queste manifestazioni ci aiutino a comprendere le modalità in cui la violenza muta forma e se ci dicano qualcosa

Abstract

Lithostathine (pancreatic stone protein, Reg protein) is, in addition to albumin, the major nonenzymatic protein of the pancreatic juice. It has been assumed to inhibit calcium carbonate precipitation and therefore to prevent stone formation in the pancreatic ducts. This function is, however, debatable. The assumption is based on the inhibition of in vitro crystal nucleation and growth by lithostathine. Considering that these phenomena occur only under certain critical conditions, we re-examined the question using a protein preparation where the purity and folding have been tested by mass spectroscopy and NMR in the absence of nonprotein contaminants. Under these conditions, we showed conclusively that lithostathine does not inhibit calcium carbonate nucleation and crystal growth. We demonstrated that previous findings on the alleged inhibition can be attributed to the uncontrolled presence of salts in the protein preparation used. Moreover, the affinity of lithostathine to calcite crystals, expressed as the half-life of bound iodinated protein in the presence of unlabeled competitor, was significantly lower than that of bovine serum albumin (8.8 and 11.2 h, respectively). Using glass microspheres instead of crystals did not significantly change the half-life of bound lithostathine (8.0 h). These findings are incompatible with the hypothesis of a specific interaction of lithostathine with calcium carbonate crystals. In conclusion, considering that components of pancreatic juice such as NaCl and phosphate ions are powerful inhibitors of calcium carbonate crystal growth, the mechanism of stone formation in pancreatic ducts must be reconsidered. The presence in normal pancreatic juice of small amounts of the 133-residue isoform of lithostathine (PSP-S1), which precipitates at physiological pH, should be noted, and the possibility should be considered that they form micro-precipitates that aggregate and are progressively calcified.

41. Detecting Complications of Direct Arterial Surgery: The Role of Intraoperative Arteriography

Abstract

• Intraoperative arteriography was carried out using a specially constructed operating table and long x-ray film changer that permitted rapid serial exposure of the arterial reconstruction and the distal arteries. During a ten-year period, this method was applied in 1,810 arterial reconstructions, with technical problems being detected in 82 (4.6%) of these patients. The incidence of problems was highest in patients undergoing endarterectomy, where 16.7% of the patients were found to have either residual debris in the cleared segment or areas of stenosis. When bypass grafts were used, technical problems were found in 2.2% of patients and included embolism or thrombosis, graft torsion in vein grafts, stenosis of the distal anastomosis, and the creation of an intimal flap. In the entire series, five patients needed reoperation in the early postoperative period. In three of these, the operative arteriograms had been initially misread.(Arch Surg 112:1115-1118, 1977)

Abstract

Adult individuals of the Symphyla Hanseniella ivorensis had their antennae cut off at the beginning of the first adult stage; this had the effect of triggering the following molt, which occurred 7.5 days after the treatment. We determined, by a radioimmunoassay method, that ecdysteroids were present in the Symphyla throughout the period studied. They were composed mainly of β-ecdysone but small amounts of α-ecdysone and of high polar products were also detected. During the artificially-induced molting process, the ecdysteroid level showed a smooth peak which took place 5 days after the amputation of the antennae. This indicates that ecdysones probably control the molt in the Symphyla as they do in insects or in crustacea. Abstract

The diagnosis of near total occlusion of the internal carotid artery was made on a group of 122 patients (90 symptomatic - 32 asymptomatic) by Doppler examination on one hand and carotid phonoangiography and oculoplethysmography, used together, on the other hand. The angiographic examination, including the study of the extra cranial arteries and the intra cerebral vessels, was performed. It has shown that a stenosis (greater than or equal to 90% of an internal carotid artery was the only lesion on 65 patients (53.3%) and that in 57 cases (46.7%) there were multiple lesions. 96 patients (76 symptomatic - 20 asymptomatic) were operated on. 4 patients (4.2%) died but the mortality was none in the group of 20 asymptomatic patients. 19 patients refused the operation and we have noted 7 occlusions of the internal carotid artery and 2 patients died (10.2%). On 7 patients with a surgical contra indication we have noted 1 secondary occlusion. We have to consider a near total occlusion of the internal carotid artery as an emergency. Its diagnosis by non invasive techniques must be followed by heparin therapy and angiographic examination. The carotid endarterectomy will respect the external carotid artery patency.

Bursicon active hemolymph of Tenebrio, injected into host pupae deprived in bursicon, induces a 2-fold increase of cyclic AMP in the epidermis of the hosts 5 min after the injection. No increase is observed by injecting bursicon inactive hemolymph or insect Ringer. From these experiments it can be concluded that cyclic AMP is a second messenger of bursicon. Abstract

Ecdysone haemolymph levels have been analyzed in neck- or thorax-ligated Pieris larvae in order to explain the physiological significance of critical periods. It appeared that head critical period corresponds to an incomplete activation of prothoracic glands, while the thoracic critical period is related to the secretion of a minimal amount of ecdysone necessary for moulting. RIA of ecdysones are made on nymphal extracts of Pisaura mirabilis during the intermoulting cycle. The hormone level is low during exuviation and the greatest part of the intermoulting period but increases notably before and after ecdysis.
