Abstract: Seafloor seepage is a widespread phenomenon within salt‐influenced basins as the deformation provides pathways for hydrocarbons to reach the seafloor. However, only minor attention has been given to the distal parts of such systems where the impact of salt tectonic deformation is relatively unpronounced. The stress put on the sedimentary column by moving salt on a continental margin may influence fluid flow systems even outside of the salt province. This stress may lead to overpressure formation within reservoirs and determine the orientation of overpressure‐induced fractures. Seepage in the Congo Fan has been discovered in such a distal position at the Regab pockmark, about 35 km west of the salt front, and its geology and biology have been studied extensively in recent years. We present high‐resolution multichannel seismic data from the Regab pockmark that reveal the underlying migration pathways from a buried channel flank 300 m below seafloor to the seafloor via hydraulic fractures in the sealing overburden. Local doming of the reservoir and the remobilization and uplift of sedimentary strata along the migration pathways are interpreted as the result of overpressure within the reservoir. The orientation of the hydraulic fractures is WSW‐ENE, and the fracture outline corresponds to the area of most intense seepage activity within the seafloor pockmark. Along with a similar orientation of other fractures in the vicinity, we propose that this alignment is due to the stress imposed on the sedimentary column in the fan by the seaward moving salt and rafting sedimentary packages of the salt province further east. The periplasmic (NAP) and membrane-associated (Nar) nitrate reductases of Paracoccus denitrificans are responsible for nitrate reduction under aerobic and anaerobic conditions, respectively. Expression of NAP is elevated in cells grown on a relatively reduced carbon and energy source (such as butyrate); it is believed that NAP contributes to redox homeostasis by coupling nitrate reduction to the disposal of excess reducing equivalents. Here, we show that deletion of either dksA1 (one of two dksA homologs in the P. denitrificans genome) or relA / spoT (encoding a bifunctional ppGpp synthetase and hydrolase) eliminates the butyrate-dependent increase in nap promoter and NAP enzyme activity. We conclude that ppGpp likely signals growth on a reduced substrate and, together with DksA1, mediates increased expression of the genes encoding NAP. Support for this model comes from the observation that nap promoter activity is increased in cultures exposed to a protein synthesis inhibitor that is known to trigger ppGpp synthesis in other organisms. We also show that, under anaerobic growth conditions, the redox-sensing RegAB two-component pair acts as a negative regulator of NAP expression and as a positive regulator of expression of the membrane-associated nitrate reductase Nar. The dksA1 and relA / spoT genes are conditionally synthetically lethal; the double mutant has a null phenotype for growth on butyrate and other reduced substrates while growing normally on succinate and citrate. We also show that the second dksA homolog ( dksA2 ) and relA / spoT have roles in regulation of expression of the flavohemoglobin Hmp and in biofilm formation. IMPORTANCE Paracoccus denitrificans is a metabolically versatile Gram-negative bacterium that is used as a model for studies of respiratory metabolism. The organism can utilize nitrate as an electron acceptor for anaerobic respiration, reducing it to dinitrogen via nitrite, nitric oxide, and nitrous oxide. This pathway (known as denitrification) is important as a route for loss of fixed nitrogen from soil and as a source of the greenhouse gas nitrous oxide. Thus, it is important to understand those environmental and genetic factors that govern flux through the denitrification pathway. Here, we identify four proteins and a small molecule (ppGpp) which function as previously unknown regulators of expression of enzymes that reduce nitrate and oxidize nitric oxide.

The Regab pockmark is a large cold seep area located 10 km north of the Congo deep sea channel at about 3160 m water depth. The associated ecosystem hosts abundant fauna, dominated by chemosynthetic species such as the mussel Bathymodiolus aff. boomerang, vestimentiferan tubeworm Escarpia southwardae, and vesicomyid clams Laubiericoncha chuni and Christineconcha regab. The pockmark was visited during the West African Cold Seeps (WACS) cruise with RV Pourquoi Pas? in February 2011, and a 14,000-m2 high-resolution videomosaic was constructed to map the most populated area and to describe the distribution of the dominant megafauna (mussels, tubeworms and clams). The results are compared with previous published works, which also included a videomosaic in the same area of the pockmark, based on images of the BIOZAIRE cruise in 2001. The 10-year variation of the faunal distribution is described and reveals that the visible abundance and distribution of the dominant megafaunal populations at Regab have not changed significantly, suggesting that the overall methane and sulfide fluxes that reach the faunal communities have been stable. Nevertheless, small and localized distribution changes in the clam community indicate that it is exposed to more transient fluxes than the other communities. Observations suggest that the main megafaunal aggregations at Regab are distributed around focused zones of high flux of methane-enriched fluids likely related to distinct smaller pockmark structures that compose the larger Regab pockmark. Although most results are consistent with the existing successional models for seep communities, some observations in the distribution of the Regab mussel population do not entirely fit into these models. This is likely due to the high heterogeneity of this site formed by the coalescence of several pockmarks. We hypothesize that the mussel distribution at Regab could also be controlled by the occurrence of zones of both intense methane fluxes and reduced efficiency of the anaerobic oxidation of methane possibly limiting tubeworm colonization. Abstract: Thirteen new species of gastropods are described from the Zairov 1-2 and Biozaire 1-3 cruises to the methane seeps off the Congo River: Patellogastropoda: Paralepetopsis sasakii sp. nov. (Neolepetopsidae); Cocculiniformia: Pyropelta oluae sp. nov. and P. sibuetae sp. nov. (Pyropeltidae); Tentaoculus granulatus sp. nov. (Pseudococculinidae); Neomphalina: Leptogyra costellata sp. nov. (Family uncertain); Vetigastropoda: Puncturella similis sp. nov. (Fissurellidae); Lepetodrilus shannonae sp. nov. (Lepetodrilidae); Caenogastropoda: Provanna reticulata sp. nov. and P. chevalieri sp. nov., Cordesia provannoides gen. et sp. nov. (Provannidae); Phymorhynchus coseli sp. nov. and P. cingulata sp. nov. (Conidae); Heterobranchia: Hyalogyrina rissoella sp. nov. (Hyalogyrinidae). All species except T. granulatus (from a settlement trap) belong to groups known from cold seeps and the entire seep fauna here is new to science. Biogeographical affinity of this gastropod fauna is to the West Atlantic seeps, not to the Mediterranean seeps or Mid-Atlantic vents. Fragments of the autecology of the species are presented. The evolution of the seep gastropod fauna is briefly discussed and a continuous immigration of taxa is supported. The oldest verified occurrences of modern taxa in the seeps date back to Cenomanian (Cretaceous) time, while some taxa seem not to appear until very late Tertiary. [Copyright &y& Elsevier]

Abstract: A giant 800-m-diameter pockmark named REGAB was discovered on the Gabon continental margin actively emitting methane at a water depth of 3200m. The microbial diversity in sediments from four different assemblages of chemosynthetic organisms, Mytilidae, Vesicomyidae, Siboglinidae and a bacterial mat, was investigated using comparative 16S rRNA gene sequence analysis. Aggregates of anaerobic methanotrophic archaea (ANME-2) and bacteria of the Desulfosarcina/Desulfococcus cluster were found in all four chemosynthetic habitats. Fluorescence in situ hybridization targeting the ANME-2/Desulfosarcina/Desulfococcus aggregates showed their presence few centimeters (3–5cm) below the surface of sediment. 16S rRNA gene sequences from all known marine ANME groups were detected in the pockmark sediments, as well as from both known bacterial partners. The archaeal diversity was limited to the ANME cluster for all investigated samples. The bacterial diversity included members of the Proteobacteria, Bacilliales, Cytophaga/Flavobacteria, Verrucomicrobia, JS1 and Actinobacteria clusters. Bacterial 16S rRNA gene sequences related to those of known sulphide-oxidizing symbionts were recovered from tissues of several invertebrates including vesicomyid clams and siboglinid tubeworms of REGAB.
