There is increased emphasis on understanding cumulative risk from the combined effects of chemical and non-chemical stressors as it relates to public health. Recent animal studies have identified pulmonary inflammation as a possible modifier and risk factor for chemical toxicity in the lung after exposure to inhaled pollutants; however, little is known about specific interactions and potential mechanisms of action. In this study, primary human bronchial epithelial cells (HBEC) cultured in 3D at the air-liquid interface (ALI) are utilized as a physiologically relevant model to evaluate the effects of inflammation on toxicity of polycyclic aromatic hydrocarbons (PAHs), a class of contaminants generated from incomplete combustion of fossil fuels. Normal HBEC were differentiated in the presence of IL-13 for 14 days to induce a profibrotic phenotype similar to asthma. Fully differentiated normal and IL-13 phenotype HBEC were treated with benzo[a]pyrene (BAP; 1–40 μg/mL) or 1% DMSO/PBS vehicle at the ALI for 48 h. Cells were evaluated for cytotoxicity, barrier integrity, and transcriptional biomarkers of chemical metabolism and inflammation by quantitative PCR. Cells with the IL-13 phenotype treated with BAP result in significantly (p < 0.05) decreased barrier integrity, less than 50% compared to normal cells. The effect of BAP in the IL-13 phenotype was more apparent when evaluating transcriptional biomarkers of barrier integrity in addition to markers of mucus production, goblet cell hyperplasia, type 2 asthmatic inflammation and chemical metabolism, which all resulted in dose-dependent changes (p < 0.05) in the presence of BAP. Additionally, RNA sequencing data showed that the HBEC with the IL-13 phenotype may have increased potential for uncontrolled proliferation and decreased capacity for immune response after BAP exposure compared to normal phenotype HBEC. These data are the first to evaluate the role of combined environmental factors associated with inflammation from pre-existing disease and PAH exposure on pulmonary toxicity in a physiologically relevant human in vitro model.

The ectomycorrhizal fungal symbiont Cenococcum geophilum is of high interest as it is globally distributed, associates with many plant species, and has resistance to multiple environmental stressors. C. geophilum is only known from asexual states but is often considered a cryptic species complex, since extreme phylogenetic divergence is often observed within nearly morphologically identical strains. Alternatively, C. geophilum may represent a highly diverse single species, which would suggest cryptic but frequent recombination. Here we describe a new isolate collection of 229 C. geophilum isolates from soils under Populus trichocarpa at 123 collection sites spanning a ~283 mile north-south transect in Western Washington and Oregon, USA (PNW). To further understanding of the phylogenetic relationships within C. geophilum, we performed maximum likelihood and Bayesian phylogenetic analyses to assess divergence within the PNW isolate collection, as well as a global phylogenetic analysis of 789 isolates with publicly available data from the United States, Japan, and European countries. Phylogenetic analyses of the PNW isolates revealed three distinct phylogenetic groups, with 15 clades that strongly resolved at >80% bootstrap support based on a GAPDH phylogeny and one clade segregating strongly in two principle component analyses. The abundance and representation of PNW isolate clades varied greatly across the North-South range, including a monophyletic group of isolates that spanned nearly the entire gradient at ~250 miles. A direct comparison between the GAPDH and ITS rRNA gene region phylogenies, combined with additional analyses revealed stark incongruence between the ITS and GAPDH gene regions, consistent with intra-species recombination between PNW isolates. In the global isolate collection phylogeny, 34 clades were strongly resolved using Maximum Likelihood and Bayesian approaches (at >80% MLBS and >0.90 BPP respectively), with some clades having intra- and intercontinental distributions. Together these data are highly suggestive of divergence within multiple cryptic species, however additional analyses such as higher resolution genotype-by-sequencing approaches are needed to distinguish potential species boundaries and the mode and tempo of recombination patterns.

Background: Rift Valley Fever (RVF) is a mosquito-borne viral zoonosis that was first isolated and characterized in 1931 in Kenya. RVF outbreaks have resulted in significant losses through human illness and deaths, high livestock abortions and deaths. This report provides an overview on epidemiology of RVF including ecology, molecular diversity spatiotemporal analysis, and predictive risk modeling. Methodology: Using the Preferred Reporting Items for Systematic reviews and Meta-Analyses (PRISMA) guidelines, we systematically searched for relevant RVF publications in repositories of the World Health Organization Library and Information Networks for Knowledge (WHOLIS), U.S Centers for Disease Control and Prevention (CDC), and Food and Agricultural Organization (FAO). Detailed searches were performed in Google Scholar, SpringerLink, and PubMed databases and included conference proceedings and books published from 1931 up to 31st January 2015. Results and discussion: A total of 84 studies were included in this review; majority (50%) reported on common human and animal risk factors that included consumption of animal products, contact with infected animals and residing in low altitude areas associated with favorable climatic and ecological conditions for vector emergence. A total of 14 (16%) of the publications described RVF progressive spatial and temporal distribution and the use of risk modeling for timely prediction of imminent outbreaks. Using distribution maps, we illustrated the gradual spread and geographical extent of disease; we also estimated the disease burden using aggregate human mortalities and cumulative outbreak periods for endemic regions. Conclusion: This review outlines common risk factors for RVF infections over wider geographical areas; it also emphasizes the role of spatial models in predicting RVF enzootics. It, therefore, explains RVF epidemiological status that may be used for design of targeted surveillance and control programs in endemic countries.

The Facility for Antiproton and Ion Research (FAIR), under construction at Darmstadt will provide intense relativistic beams of exotic nuclei at its Superconducting-FRagment Separator. High-resolution in-beam γ-ray spectroscopy will be performed in the HISPEC experiment, using the European Advanced GAmma-ray Tracking Array (AGATA). The PreSPEC-AGATA campaign is the predecessor of HISPEC and runs from 2012 to 2014 at GSI Helmholtzzentrum für Schwerionenforschung GmbH. Up to19 AGATA modules were used at GSI's F Ragment Separator in 2012. We report on the status of the experiment including preliminary results from performance commissioning.

Cosmetic outcomes of breast-conserving treatment (BCT) and radiation therapy (RT) are important for survivorship. One important factor is breast volume loss. Smoking during RT has been associated with increased risk of toxicity, but it is unknown whether smoking history affects volume loss. It was hypothesized that smoking would be associated with increased volume loss, via increased susceptibility to radiation toxicity. This study aimed to explore any associations. Breast volume changes were calculated for 112 patients (113 breasts) following T1 tumor lumpectomies. From preoperative baseline to seven years post-radiation, volumes were calculated from mammograms using π/3* height*radius2. Mann-Whitney U-Tests compared volume changes at each time point between patients actively smoking (n = 1) or with a smoking history (n = 29), and patients who never smoked (n = 83). Multivariate analysis assessed volume change as a function of patient age, pack-years smoked, diabetes, radiation dosage, fractions, technique, boost dose, whole/partial breast RT, treatment length, chemotherapy (hormonal/cytotoxic), baseline breast volume, and months since treatment. Smoking group average age was 62.6 (SD = 6.8) years, median pack-years smoked was 5 (IQR = 19.9), median RT dose was 4272 cGy (IQR = 749) over 16 (IQR = 4.8) fractions for median duration 3.2 (IQR = 1.0) weeks. N = 24 (80%) received endocrine therapy. Non-smoking group average age was 57.6 (SD = 8.8) years, median RT dose was 5005 cGy (IQR = 749) over 20 (IQR = 4) fractions for median duration 3.6 (IQR = 1.3) weeks. N = 64 (77.1%) received endocrine therapy. Treatment details are summarized in Table 1. Patients with a smoking history experienced significantly greater volume loss during the first two years after treatment. By year one, 26.5% of baseline breast volume was lost compared to 13.4% among non-smoking patients (P = 0.02). By year two, smoking group volume loss was 23.9%, compared to 16.3% for non-smoking patients (P = 0.04). Larger baseline breast volume and increased pack-years smoked were significant predictors of increased volume loss during years one, two and three (P <0.05). For year three a diabetes diagnosis was also significant (P = 0.03). Findings suggest prior smoking may contribute to increased breast shrinkage within the first two years following BCT/RT. Increased pack-years and larger baseline breast volume may predict greater volume loss. Anticipating these changes is important for patient education and planning future interventions.
