Abstract

ABSTRACT Objective Methods Results Conclusions This non‐interventional, prospective, single‐center study aimed to develop a technique to measure ruxolitinib (RUX) concentrations and provide preliminary data on the distribution of plasma drug levels in patients with steroid refractory (SR) GvHD.Between April 2023 and May 2024, we analyzed 48 blood samples from 29 patients with SR‐GvHD.Median individual plasma concentrations varied across different RUX doses and largely overlapped: 39.2 ng/mL at 10 mg b.i.d (range: 0–73), 13.1 ng/mL at 10–5 mg (range, 6.1–35.6), and 31.6 ng/mL at 5 mg b.i.d (range: 0.7–99.9). Samples taken under non‐optimal temperature conditions showed a lower median concentration of 0.77 ng/mL (range: 0–7.4 ng/mL). The four patients who did not respond at days +28 and +180 after RUX initiation (3 with lower gastrointestinal aGvHD, and 1 with ocular, hepatic, and pulmonary cGvHD) showed a median concentration of only (7.4 ng/mL (range, 0–29) ng/mL) with full dosing.The introduction and validation of a liquid chromatography–tandem mass spectrometry method for quantifying plasma RUX concentrations was feasible in our center. Administering predetermined and fixed doses of RUX in patients with SR‐GvHD showed highly variable and overlapping plasma drug concentrations. Abstract

Objective: This retrospective single center study aims to describe changes in the severity and organ‐specific distribution of GvHD, by comparing the outcomes of 3 distinct GvHD prophylaxis approaches. Methods: Between January 2012 and June 2022, 226 patients underwent allogeneic hematopoietic stem cell transplantation from HLA‐matched or 1‐allele mismatched related or unrelated donors. Fifty‐eight (26%) received prophylaxis with calcineurin inhibitor in combination with mycophenolate mofetil or a short course of methotrexate (Cohort‐1), 87 (38%) tacrolimus plus sirolimus (Cohort‐2), and 81 (36%) post‐transplant cyclophosphamide (PTCy) plus tacrolimus (Cohort‐3). Results: The incidence of grade II‐IV aGvHD was 69% vs. 41.4% vs. 27.2%; p <.01. The most significant reduction with PTCy was observed in both stage 3–4 skin and lower gastrointestinal (GI) involvement (p <.01). The incidence of moderate‐to‐severe cGvHD at 12 months was 34.5% vs. 34.5% vs. 6.2%; p <.01. Moderate‐to‐severe skin and GI cGvHD was less common after PTCy (p <.01). The 1‐year GvHD‐free/relapse‐free survival was higher with PTCy (p <.01). Conclusions: Our study indicates that PTCy‐based GvHD prophylaxis reduces the frequency and severity of both acute and chronic GvHD, with a notable decrease in severe GI and cutaneous manifestations. The higher GRFS may result in lower GvHD‐related mortality, leading to an improved quality of life among survivors. [ABSTRACT FROM AUTHOR]

Abstract

The effects of the climate change are expected to affect direct and indirect on livestock physiology and health by increasing exposure to environmental stressors like heat stress or water deprivation, mainly in arid and semi-arid regions, especially in the south edge of the Mediterranean basin. The aim of this study is to identify candidate genes or genomic regions associated with ovine stress biomarkers through genome-wide association analysis. Two-hundred and one ewes from the Rasa Aragonesa breed were challenged with total water restriction for 5 days. Temperature and percentage of humidity was also measured, as well as intake, live weight, and body condition score daily during the 5 days restriction. Blood samples were collected just prior to the challenge (0d) and at the end of the challenge (5d), obtaining haematological (white, red blood cells and platelet traits) and metabolite (total protein, glucose, non-esterified fatty acids (NEFA), cortisol, DHEA and DHEAS concentration phenotypes) measurements. Wool samples were also collected at day 0 and 4 weeks later (28d), and cortisol, DHEA and DHEAS were also measured. In addition to these measurable traits for each blood/wool sample, variation of traits between blood or wool samples collected at different time points were also calculated for each animal. Genotyping was performed using the Ovine680kBeadChip. AIREMLF90 package from the BLUPF90 family programs was used to estimate variance components for each trait using the average information-restricted maximum likelihood (AI-REML). GCTA software (Yang et al., 2011) was used for GWAS analysis. Mixed model statistical analysis indicated that all haematological and metabolite levels showed consistent changes throughout water restriction period with the exception of plasma cortisol and MCH. Abstract

Treatment of chronic lymphocytic leukemia (CLL) has dramatically evolved over the last decades thanks to the introduction of targeted therapies. We aimed to describe retrospectively the evolution in the frontline prescription in the CLL patients from our institution. As a secondary objective, the impact of frontline therapy on the time-to-next-treatment (TTNT) and overall survival (OS). After a median of 6.4 years (0.1–36.4) of follow-up from diagnosis, 323 of 780 CLL patients (41.4%) required therapy. Alkylating agents in monotherapy (chlorambucil) were the most used until 2012, and from then, chemoimmunotherapy. Since 2018, targeted therapies were the most common therapeutic strategy (74.1%). Patients who received targeted therapies had significantly longer TTNT compared to other regimens. In the multivariable analyses, mutated IGHV genes targeted therapies and chemoimmunotherapy regimens were related to longer TTNT, and sex female, age younger than 65, and mutated IGHV genes were associated with better OS. Abstract

The integrin ITGB3 has been previously described to play an essential role in breast cancer metastasis, but the precise mechanisms remain undefined. We have now uncovered essential and thus far unknown roles of ITGB3 in vesicle uptake. The functional requirement for ITGB3 derives from its interactions with heparan sulfate proteoglycans (HSPGs) and the process of integrin endocytosis, allowing the capture of extracellular vesicles and their endocytosis-mediated internalization. Key for the function of ITGB3 is the interaction and activation of focal adhesion kinase (FAK), which is required for endocytosis of these vesicles. Thus, ITGB3 has a central role in intracellular communication via extracellular vesicles, proposed to be critical for cancer metastasis. Abstract

NRAS-mutated melanoma lacks a specific line of treatment. Metabolic reprogramming is considered a novel target to control cancer; however, NRAS-oncogene contribution to this cancer hallmark is mostly unknown. Here, we show that NRASQ61-mutated melanomas specific metabolic settings mediate cell sensitivity to sorafenib upon metabolic stress. Mechanistically, these cells are dependent on glucose metabolism, in which glucose deprivation promotes a switch from CRAF to BRAF signaling. This scenario contributes to cell survival and sustains glucose metabolism through BRAF-mediated phosphorylation of 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase-2/3 (PFKFB2/PFKFB3). In turn, this favors the allosteric activation of phosphofructokinase-1 (PFK1), generating a feedback loop that couples glycolytic flux and the RAS signaling pathway. An in vivo treatment of NRASQ61 mutant melanomas, including patient-derived xenografts, with 2-deoxy-D-glucose (2-DG) and sorafenib effectively inhibits tumor growth. Thus, we provide evidence for NRAS-oncogene contributions to metabolic rewiring and a proof-of-principle for the treatment of NRASQ61-mutated melanoma combining metabolic stress (glycolysis inhibitors) and previously approved drugs, such as sorafenib. Targeted therapeutic options for NRAS-mutant melanoma are limited. Here, the authors show that under metabolic stress NRAS-mutant melanoma cells activate a BRAF-dependent glycolysis pathway for survival, leading to improve efficacy of sorafenib when combined with glycolysis inhibitors. Abstract

Background: The general picture of human genetic variation has been vastly depicted in the last years, yet many populations remain broadly understudied. In this work, we analyze for the first time the Merchero population, a Spanish minority ethnic group that has been scarcely studied and historically persecuted. Mercheros have been roughly characterised by an itinerant history, common traditional occupations, and the usage of their own language. Results: Here, we examine the demographic history and genetic scenario of Mercheros, by using genome-wide array data, whole mitochondrial sequences, and Y chromosome STR markers from 25 individuals. These samples have been complemented with a wide-range of present-day populations from Western Eurasia and North Africa. Our results show that the genetic diversity of Mercheros is explained within the context of the Iberian Peninsula, evidencing a modest signal of Roma admixture. In addition, Mercheros present low genetic isolation and intrapopulation heterogeneity. Conclusions: This study represents the first genetic characterisation of the Merchero population, depicting their fine-scale ancestry components and genetic scenario within the Iberian Peninsula. Since ethnicity is not only influenced by genetic ancestry but also cultural factors, other studies from multiple disciplines are needed to further explore the Merchero population. As with Mercheros, there is a considerable gap of underrepresented populations and ethnic groups in publicly available genetic data. Thus, we encourage the consideration of more ethnically diverse population panels in human genetic studies, as an attempt to improve the representation of human populations and better reconstruct their fine-scale history.

Abstract

Trabajo fin de máster en Bioinformática y Biología Computacional, El complejo de P. aeruginosa es un conjunto de bacterias pertenecientes al orden Pseudomonadales, unas γ-proteobacterias gram negativas que se encuentran en distintos nichos ecológicos y que está compuesto por una gran variedad de especies. A lo largo de los años, las especies se han definido en base a su morfología y sus actividades metabólicas. Después mediante la secuenciación de relojes filogenéticos como el 16S y por hibridación DNA-DNA para comparaciones de especies o cepas dos a dos, pero estas técnicas se están sustituyendo por métodos de caracterización genotípica utilizando una serie de genes marcadores que se encuentran en todas las especies. En esto se basan los Análisis Multilocus de Secuencia (MSLA en inglés), aunque existen otros métodos basados en el estudio del genoma completo para obtener filogenias más precisas que las que se obtienen mediante MLSA, tales como la identidad media de nucleótidos (Average Nucleotide Identity, ANI), ANIb (ANI como una identidad media de todas las coincidencias de blastn) y ANIm (ANI con el software MUMmer). En este trabajo se estudiaron las relaciones filogenéticas de las especies pertenecientes al complejo de P.aeruginosa utilizando MLSA y ANIb. Al mismo tiempo, también se definió el genoma central del complejo, el genoma principal de cada grupo, el genoma específico de cada especie, el genoma específico de cada grupo y el pangenoma gracias al estudio de los ortólogos, así como las distintas características funcionales que definan a cada grupo de P. aeruginosa.
