Abstract

We study a mid-order wavefront sensor (MOWFS) to address fine cophasing errors in exoplanet imaging with future large segmented aperture space telescopes. Observing Earth analogs around Sun-like stars requires contrasts down to $10^{-10}$ in visible light. One promising solution consists of producing a high-contrast dark zone in the image of an observed star. In a space observatory, this dark region will be altered by several effects, and among them, the small misalignments of the telescope mirror segments due to fine thermo-mechanical drifts. To correct for these errors in real time, we investigate a wavefront control loop based on a MOWFS with a Zernike sensor. Such a MOWFS was installed on the high-contrast imager for complex aperture telescopes (HiCAT) testbed in Baltimore in June 2023. The bench uses a 37-segment Iris-AO deformable mirror to mimic telescope segmentation and some wavefront control strategies to produce a dark zone with such an aperture. In this contribution, we first use the MOWFS to characterize the Iris-AO segment discretization steps. For the central segment, we find a minimal step of 125 $\pm$ 31 pm. This result will help us to assess the contribution of the Iris-AO DM on the contrast in HiCAT. We then determine the detection limits of the MOWFS, estimating wavefront error amplitudes of 119 and 102 pm for 10 s and 1 min exposure time with a SNR of 3. These values inform us about the measurement capabilities of our wavefront sensor on the testbed. Abstract

SPIDER is a balloon-borne instrument designed to map the cosmic microwave background at degree-angular scales in the presence of Galactic foregrounds. SPIDER has mapped a large sky area in the Southern Hemisphere using more than 2000 transition-edge sensors (TESs) during two NASA Long Duration Balloon flights above the Antarctic continent. During its first flight in January 2015, SPIDER observed in the 95 GHz and 150 GHz frequency bands, setting constraints on the B-mode signature of primordial gravitational waves. Its second flight in the 2022-23 season added new receivers at 280 GHz, each using an array of TESs coupled to the sky through feedhorns formed from stacks of silicon wafers. These receivers are optimized to produce deep maps of polarized Galactic dust emission over a large sky area, providing a unique data set with lasting value to the field. Abstract

Wavefront sensing and control (WFSC) will play a key role in improving the stability of future large segmented space telescopes while relaxing the thermo-mechanical constraints on the observatory structure. Coupled with a coronagraph to reject the light of an observed bright star, WFSC enables the generation and stabilisation of a dark hole (DH) in the star image to perform planet observations. While WFSC traditionally relies on a single wavefront sensor (WFS) input to measure wavefront errors, the next generation of instruments will require several WFSs to address aberrations with different sets of spatial and temporal frequency contents. The multiple measurements produced in such a way will then have to be combined and converted to commands for deformable mirrors (DMs) to modify the wavefront subsequently. We asynchronously operate a loop controlling the high-order modes digging a DH and a control loop that uses the rejected light by a Lyot coronagraph with a Zernike wavefront sensor to stabilize the low-order aberrations. Using the HiCAT testbed with a segmented telescope aperture, we implement concurrent operations and quantify the expected cross-talk between the two controllers. We then present experiments that alternate high-order and low-order control loops to identify and estimate their respective contributions. We show an efficient combination of the high-order and low-order control loops, keeping a DH contrast better than 5 x 10-8 over a 30 min experiment and stability improvement by a factor of 1.5. In particular, we show a contrast gain of 1.5 at separations close to the DH inner working angle, thanks to the low-order controller contribution. Concurrently digging a DH and using the light rejected by a Lyot coronagraph to stabilize the wavefront is a promising path towards exoplanet imaging and spectroscopy with future large space observatories.

Abstract

Combining large segmented space telescopes, coronagraphy and wavefront control methods is a promising solution to produce a dark hole (DH) region in the coronagraphic image of an observed star and study planetary companions. The thermal and mechanical evolution of such a high-contrast facility leads to wavefront drifts that degrade the DH contrast during the observing time, thus limiting the ability to retrieve planetary signals. Lyot-style coronagraphs are starlight suppression systems that remove the central part of the image for an unresolved observed star, the point spread function, with an opaque focal plane mask (FPM). When implemented with a flat mirror containing an etched pinhole, the mask rejects part of the starlight through the pinhole which can be used to retrieve information about low-order aberrations. We propose an active control scheme using a Zernike wavefront sensor (ZWFS) to analyze the light rejected by the FPM, control low-order aberrations, and stabilize the DH contrast. The concept formalism is first presented before characterizing the sensor behavior in simulations and in laboratory. We then perform experimental tests to validate a wavefront control loop using a ZWFS on the HiCAT testbed. By controlling the first 11 Zernike modes, we show a decrease in wavefront error standard deviation by a factor of up to 9 between open- and closed-loop operations using the ZWFS. In the presence of wavefront perturbations, we show the ability of this control loop to stabilize a DH contrast around 7x10^-8 with a standard deviation of 7x10^-9. Active control with a ZWFS proves a promising solution in Lyot coronagraphs with an FPM-filtered beam to control and stabilize low-order wavefront aberrations and DH contrast for exoplanet imaging with future space missions.

Abstract

In this work we describe upgrades to the Spider balloon-borne telescope in preparation for its second flight, currently planned for December 2021. The Spider instrument is optimized to search for a primordial B-mode polarization signature in the cosmic microwave background at degree angular scales. During its first flight in 2015, Spider mapped ~10% of the sky at 95 and 150 GHz. The payload for the second Antarctic flight will incorporate three new 280 GHz receivers alongside three refurbished 95- and 150 GHz receivers from Spider's first flight. In this work we discuss the design and characterization of these new receivers, which employ over 1500 feedhorn-coupled transition-edge sensors. We describe pre-flight laboratory measurements of detector properties, and the optical performance of completed receivers. Abstract

We present a publicly-available toolkit of flight-proven hardware and software to retrieve 5 TB of data or small physical samples from a stratospheric balloon platform. Before launch, a capsule is attached to the balloon, and rises with it. Upon remote command, the capsule is released and descends via parachute, continuously transmitting its location. Software to predict the trajectory can be used to select a safe but accessible landing site. We dropped two such capsules from the SuperBIT telescope, in September 2019. The capsules took ~37 minutes to descend from ~30 km altitude. They drifted 32 km and 19 km horizontally, but landed within 300 m and 600 m of their predicted landing sites. We found them easily, and successfully recovered the data. We welcome interest from other balloon teams for whom the technology would be useful.

Abstract

Balloon-borne telescopes present unique thermal design challenges which are a combination of those present for both space and ground telescopes. At altitudes of 35-40 km, convection effects are minimal and difficult to characterize. Radiation and conduction are the predominant heat transfer mechanisms reducing the thermal design options. For long duration flights payload mass is a function of power consumption making it an important optimization parameter. SuperBIT, or the Super-pressure Balloon-borne Imaging Telescope, aims to study weak lensing using a 0.5m modified Dall-Kirkham telescope capable of achieving 0.02" stability and capturing deep exposures from visible to near UV wavelengths. To achieve the theoretical stratospheric diffraction-limited resolution of 0.25", mirror deformation gradients must be kept to within 20nm. The thermal environment must thus be stable on time scales of an hour and the thermal gradients must be minimized on the telescope. Abstract

We describe the construction and characterization of the 280 GHz bolometric focal plane units (FPUs) to be deployed on the second flight of the balloon-borne SPIDER instrument. These FPUs are vital to SPIDER's primary science goal of detecting or placing an upper limit on the amplitude of the primordial gravitational wave signature in the cosmic microwave background (CMB) by constraining the B-mode contamination in the CMB from Galactic dust emission. Each 280 GHz focal plane contains a 16 x 16 grid of corrugated silicon feedhorns coupled to an array of aluminum-manganese transition-edge sensor (TES) bolometers fabricated on 150 mm diameter substrates. In total, the three 280 GHz FPUs contain 1,530 polarization sensitive bolometers (765 spatial pixels) optimized for the low loading environment in flight and read out by time-division SQUID multiplexing. In this paper we describe the mechanical, thermal, and magnetic shielding architecture of the focal planes and present cryogenic measurements which characterize yield and the uniformity of several bolometer parameters. The assembled FPUs have high yields, with one array as high as 95% including defects from wiring and readout. We demonstrate high uniformity in device parameters, finding the median saturation power for each TES array to be ~3 pW at 300 mK with a less than 6% variation across each array at one standard deviation. These focal planes will be deployed alongside the 95 and 150 GHz telescopes in the SPIDER-2 instrument, slated to fly from McMurdo Station in Antarctica in December 2018.

Abstract

Background: The EUROCARE-5 study revealed disparities in childhood cancer survival among European countries, giving rise to important initiatives across Europe to reduce the gap. Extending its representativeness through increased coverage of eastern European countries, the EUROCARE-6 study aimed to update survival progress across countries and years of diagnosis and provide new analytical perspectives on estimates of long-term survival and the cured fraction of patients with childhood cancer. Methods: In this population-based study, we analysed 135 847 children (aged 0–14 years) diagnosed during 2000–13 and followed up to the end of 2014, recruited from 80 population-based cancer registries in 31 European countries. We calculated age-adjusted 5-year survival differences by country and over time using period analysis, for all cancers combined and for major cancer types. We applied a variant of standard mixture cure models for survival data to estimate the cure fraction of patients by childhood cancer and to estimate projected 15-year survival. Findings: 5-year survival for all childhood cancer combined in Europe in 2010–14 was 81% (95% CI 81–82), showing an increase of three percentage points compared with 2004–06. Significant progress over time was observed for almost all cancers. Survival remained stable for osteosarcomas, Ewing sarcoma, Burkitt lymphoma, non-Hodgkin lymphomas, and rhabdomyoscarcomas. For all cancers combined, inequalities still persisted among European countries (with age-adjusted 5-year survival ranging from 71% [95% CI 60–79] to 87% [77–93]). The 15-year survival projection for all patients with childhood cancer diagnosed in 2010–13 was 78%. We estimated the yearly long-term mortality rate due to causes other than the diagnosed cancer to be around 2 per 1000 patients for all childhood cancer combined, but to approach zero for retinoblastoma. 22. Looking at and with Images: Crowds in Joker, Joker in the Crowd

Abstract

In this article, I provide a ‘symptomatic’ reading of the film, Joker, by analyzing the complex dynamics behind the emergence of this supervillain narrative at this particular moment in history, focusing on the images and discourses of social movements to which popular culture cannot remain indifferent to and quickly embodies in different forms. I focus on the ways in which the widespread political restlessness across geographies and the increased visibility of inequalities and reactions to it generate visual and discursive patterns. These patterns in turn become parts of popular cultural narratives, from Beyoncé’s pose on a sinking police car and her dancing the white western canon away in Louvre museum, to Childish Gambino’s This is America clip and the Pepsi advertisement relying on a mixture of Occupy Wall Street and Black Lives Matter movements’ visual repertoire. Situating Joker in this larger picture allows exploring how the narrative engages with social reality and how it informs/is informed by it, without being limited to appropriation theories that would claim that popular cultural products deprive radical social content of its meaning and function. Within this theoretical framework, I focus particularly on two questions on the construction and circulation of Joker’s narrative: How is Joker’s embodiment of social reality symptomatic of an uneasy oscillation between potentially revolutionary class struggle-oriented leftist/anarchist mobilizations on the one hand and alt-right outbursts of anger centering around vulnerable white souls on the other? And what happens to the meaning and function of the narrative when the film spills out of the theater and is reclaimed in the streets, as in the case of people painting their faces as Jokers in Lebanon or Chile at the moment, while struggling against class society, neoliberalism and oppression?

Item does not contain fulltext, Horizontal gene transfer from viruses to eukaryotic cells is a pervasive phenomenon. Somatic viral integrations are linked to persistent viral infection whereas integrations into germline cells are maintained in host genomes by vertical transmission and may be co-opted for host functions. In the arboviral vector Aedes aegypti, an endogenous viral element from a nonretroviral RNA virus (nrEVE) was shown to produce PIWI-interacting RNAs (piRNAs) to limit infection with a cognate virus. Thus, nrEVEs may constitute a heritable, sequence-specific mechanism for antiviral immunity, analogous to piRNA-mediated silencing of transposable elements. Here, we combine population genomics and evolutionary approaches to analyse the genomic architecture of nrEVEs in A. aegypti. We conducted a genome-wide screen for adaptive nrEVEs and searched for novel population-specific nrEVEs in the genomes of 80 individual wild-caught mosquitoes from five geographical populations. We show a dynamic landscape of nrEVEs in mosquito genomes and identified five novel nrEVEs derived from two currently circulating viruses, providing evidence of the environmental-dependent modification of a piRNA cluster. Overall, our results show that virus endogenization events are complex with only a few nrEVEs contributing to adaptive evolution in A. aegypti.

ABSTRACT

The SARS-CoV-2 Receptor Binding Domain (RBD) is both the principal target of neutralizing antibodies, and one of the most rapidly evolving domains, which can result in the emergence of immune escape mutations limiting the effectiveness of vaccines and antibody therapeutics. To facilitate surveillance, we developed a rapid, high-throughput, multiplex assay able to assess the inhibitory response of antibodies to 24 RBD natural variants simultaneously. We demonstrate that immune escape can occur through two mechanisms, antibodies that fail to recognize mutations, along with antibodies that have reduced inhibitory capacity due to enhanced variant RBD-ACE2 affinity. A competitive approach where antibodies simultaneously compete with ACE2 for binding to the RBD may therefore more accurately reflect the physiological dynamics of infection. We describe the enhanced affinity of RBD variants N439K, S477N, Q493L, S494P and N501Y to the ACE2 receptor, and demonstrate the ability of this assay to bridge a major gap for SARS-CoV-2 research; informing selection of complementary monoclonal antibody candidates and the rapid identification of immune escape to emerging RBD variants following vaccination or natural infection.

Abstract

BACKGROUND: As vaccines against SARS-CoV-2 are now being rolled out, a better understanding of immunity to the virus, whether from infection, or passive or active immunisation, and the durability of this protection is required. This will benefit from the ability to measure antibody-based protection to SARS-CoV-2, ideally with rapid turnaround and without the need for laboratory-based testing. METHODS: We have developed a lateral flow POC test that can measure levels of RBD-ACE2 neutralising antibody (NAb) from whole blood, with a result that can be determined by eye or quantitatively on a small instrument. We compared our lateral flow test with the gold-standard microneutralisation assay, using samples from convalescent and vaccinated donors, as well as immunised macaques. FINDINGS: We show a high correlation between our lateral flow test with conventional neutralisation and that this test is applicable with animal samples. We also show that this assay is readily adaptable to test for protection to newly emerging SARS-CoV-2 variants, including the beta variant which revealed a marked reduction in NAb activity. Lastly, using a cohort of vaccinated humans, we demonstrate that our whole-blood test correlates closely with microneutralisation assay data (specificity 100% and sensitivity 96% at a microneutralisation cutoff of 1:40) and that fingerprick whole blood samples are sufficient for this test. INTERPRETATION: Taken together, the COVID-19 NAb-testTM device described here provides a rapid readout of NAb based protection to SARS-CoV-2 at the point of care. Abstract

In this work we describe upgrades to the Spider balloon-borne telescope in preparation for its second flight, currently planned for December 2021. The Spider instrument is optimized to search for a primordial B-mode polarization signature in the cosmic microwave background at degree angular scales. During its first flight in 2015, Spider mapped ~10% of the sky at 95 and 150 GHz. The payload for the second Antarctic flight will incorporate three new 280 GHz receivers alongside three refurbished 95- and 150 GHz receivers from Spider's first flight. In this work we discuss the design and characterization of these new receivers, which employ over 1500 feedhorn-coupled transition-edge sensors. We describe pre-flight laboratory measurements of detector properties, and the optical performance of completed receivers. These receivers will map a wide area of the sky at 280 GHz, providing new information on polarized Galactic dust emission that will help to separate it from the cosmological signal.

Abstract

SUMMARY Necrosis of infected macrophages constitutes a critical pathogenetic event in tuberculosis releasing mycobacteria into the extracellular environment where they can grow unrestricted. In zebrafish infected with Mycobacterium marinum, a close relative of Mycobacterium tuberculosis, excess Tumor Necrosis Factor triggers programmed necrosis of infected macrophages through the production of mitochondrial reactive oxygen species (ROS) and the participation of cyclophilin D, a component of the mitochondrial permeability transition pore. Here we show that this necrosis pathway is not mitochondrion-intrinsic but rather results from an interorganellar circuit initiating and culminating in the mitochondrion. Mitochondrial ROS induce production of lysosomal ceramide which ultimately activates the cytosolic protein BAX. BAX promotes calcium flow from the endoplasmic reticulum into the mitochondrion through ryanodine receptors. The resultant mitochondrial calcium overload triggers cyclophilin D-mediated necrosis. We identify ryanodine receptors and plasma membrane L-Type calcium channels as specific druggable targets to intercept mitochondrial calcium overload so as to inhibit macrophage necrosis.
