The pedunculopontine tegmental nucleus of the brainstem (PPTg) has extensive interconnections and neuronal‐behavioural correlates. It is implicated in movement control and sensorimotor integration. We investigated whether single neuron activity in freely moving rats is correlated with components of skilled forelimb movement, and whether individual neurons respond to both motor and sensory events. We found that individual PPTg neurons showed changes in firing rate at different times during the reach. This type of temporally specific modulation is like activity seen elsewhere in voluntary movement control circuits, such as the motor cortex, and suggests that PPTg neural activity is related to different specific events occurring during the reach. In particular, many neuronal modulations were time‐locked to the end of the extension phase of the reach, when fine distal movements related to food grasping occur, indicating strong engagement of PPTg in this phase of skilled individual forelimb movements. In addition, some neurons showed brief periods of apparent oscillatory firing in the theta range at specific phases of the reach‐to‐grasp movement. When movement‐related neurons were tested with tone stimuli, many also responded to this auditory input, allowing for sensorimotor integration at the cellular level. Together, these data extend the concept of the PPTg as an integrative structure in generation of complex movements, by showing that this function extends to the highly coordinated control of the forelimb during skilled reach to grasp movement, and that sensory and motor‐related information converges on single neurons, allowing for direct integration at the cellular level. A majority of stroke patients have persisting upper extremity impairments, with a large number of individuals who do not regain the ability to reach and manipulate everyday objects despite receiving prolonged and intensive therapy. Varied interventions have been employed to improve reaching functions. However, there is a lack of a comprehensive and updated review of all the interventions that can be used to improve upper extremity reaching function in stroke. To review the existing interventions aimed to improve upper extremity reaching function in persons with stroke. Studies with quantitative, qualitative, and mixed methods design which have employed interventions to improve upper extremity reaching function following stroke and have been published from database inception to 25th February 2021 in English. The proposed scoping review will follow the Preferred Reporting Items for Systematic reviews and Meta-Analyses extension for Scoping Reviews (PRISMA-ScR) Checklist and will be conducted as per the methodological framework proposed by Arksey and O'Malley. A database-specific search strategy will be used in four electronic databases (MEDLINE, Scopus, CINAHL, Web of Science) and four grey literature platforms (ProQuest, MedNAR, OpenGrey, New York Academy of Medicine). The screening and extraction of data from eligible studies will be done independently by two reviewers. The extracted data will be presented with the help of tables and visual representation. Previously conducted reviews have enumerated the various types of interventions that have been used to improve upper extremity reaching function, but are incomprehensive and in need of an update. This study investigated how modified control of a virtual hand executing reach-to-grasp affects functional performance and agency (perception of control). The objective of this work was to demonstrate positive relationships between reaching performance and grasping agency and motivate greater consideration of agency in movement rehabilitation. We hypothesized that agency and performance have positive correlation across varying control modes of the virtual hand. In this study, each participant controlled motion of a virtual hand through motion of his or her own hand. Control of the virtual hand was modified according to a specific control mode. Each mode involved the virtual hand moving at a modified speed, having noise, or including a level of automation. These specific modes represent potential control features to adapt for a rehabilitation device such as a prosthetic arm and hand. In this study, significant changes in agency and performance were observed across the control modes. Overall, a significant positive relationship (p < 0.001) was observed between the primary performance metric of reach (tracking a minimum path length trajectory) and an implicit measurement of agency (intentional binding). Intentional binding was assessed through participant perceptions of time-intervals between grasp contact and a sound event. Other notable findings include improved movement efficiency (increased smoothness, reduced acceleration) during expression of higher agency and shift toward greater implicit versus explicit agency with higher control speed. Positively relating performance and agency incentivizes control adaptation of powered movement devices, such as prostheses or exoskeletons, to maximize both user engagement and functional performance. Agency-based approaches may foster user-device integration at a cognitive level and facilitate greater clinical retention of the device. Future work should identify robust and automated methods to adapt device control for increased agency. Decreased dexterity in chronic stroke survivors results in diminished hand use and impacts quality of life. We studied reach-and-grasp coordination and aperture scaling during reach to grasp using different grasp types and object sizes (33-55mm). Chronic stroke survivors with hand paresis and controls grasped cylinders with the whole hand or fingertips. Three stroke subjects with more severe hand paresis had disrupted reach! grasp coordination and used compensatory strategies for arm transport and/or grasping. Nine stroke subjects with less severe paresis scaled aperture to cylinder diameter (p < .001) but had slower movements, used excessive trunk movement, and had prolonged deceleration times. Relative time to maximal grip aperture (TMA) occurred earlier in stroke subjects and in both groups when grasping the small cylinder (p < .002). Despite deficits in reach and grasp, chronic stroke survivors with mild to moderate hand paresis may retain grip aperture scaling to object size for different grasp types. [ABSTRACT FROM AUTHOR]

Stroke patients are often left with hemiplegia or hemiparesis of the upper extremities, severely limiting the ability to perform bimanual and functional activities. No studies have investigated how stroke patients adapt their movements to changes in object size in functionally asymmetric bimanual tasks. The influence of object size on intralimb and interlimb coordination during an asymmetrical, functional bimanual task was examined in patients with left cerebral vascular accidents (LCVA) and healthy controls. Fourteen LCVA patients and 13 age-matched controls were instructed to reach to grasp a large and a small jar with the right/affected hand and to open the cap with the other hand. Movement kinematics was analyzed for intralimb coordination (spatial and temporal planning of reaching and grasping) and interlimb coordination (bimanual synchronization and temporal association of the hands). The results demonstrate a spatial adaptation of reaching in the affected hand to the object size and deficits in temporal planning of grasping with the affected hand to object size in the stroke patients. Movement adaptations of the unaffected hand in the stroke patients were similar to those in the healthy adults. Bimanual coordination was independent of object size for both groups. For human beings, monitoring others' errors is essential for efficient goal-directed behavior. Indeed, the mere observation of other individuals' errors provides a rich source of information that can be used to avoid potential errors and improve our performance without direct experience. Recent studies have outlined that vicarious experience of errors influences the observer's overt behavior. This observational posterror slowing (oPES) is supposed to reflect a strategic increase in control aimed at reducing the probability of an error. Because the consequences of error observation have been exclusively investigated by means of arbitrary button-press responses, which limit the investigation to premovement processes, it is unclear whether the observation of an error also influences the online control of goal-directed actions. In the present study, for the first time, we investigated the effect of error observation on the reach-to-grasp movement by means of kinematical analysis. The results revealed that error-observation effects are not confined to premovement processes—they also strategically affect spatial movement trajectories. How patterns of covariance in motor output and neural activity emerge over the course of learning is a topic of ongoing investigation. Vaidya et al. The development of coordinated reach-to-grasp movement has been well studied in infants and children. However, the role of motor cortex during this development is unclear because it is difficult to study in humans. We took the approach of using a brain-machine interface (BMI) paradigm in rhesus macaques with prior therapeutic amputations to examine the emergence of novel, coordinated reach to grasp. Previous research has shown that after amputation, the cortical area previously involved in the control of the lost limb undergoes reorganization, but prior BMI work has largely relied on finding neurons that already encode specific movement-related information. In this study, we taught macaques to cortically control a robotic arm and hand through operant conditioning, using neurons that were not explicitly reach or grasp related. Over the course of training, stereotypical patterns emerged and stabilized in the cross-covariance between the reaching and grasping velocity profiles, between pairs of neurons involved in controlling reach and grasp, and to a comparable, but lesser, extent between other stable neurons in the network. In fact, we found evidence of this structured coordination between pairs composed of all combinations of neurons decoding reach or grasp and other stable neurons in the network. The degree of and participation in coordination was highly correlated across all pair types. In social interactions, the movements performed by others can be used to anticipate their intention. The present paper investigates whether cooperative vs competitive contexts influence the kinematics of object-directed motor actions and whether they modulate the effect of social intention on motor actions. An “Actor” and a “Partner” participated in a task consisting in displacing a wooden dowel under time constraint. Before this Main action , the Actor performed a Preparatory action which consisted in placing the dowel at the center of the table. Information about who would make the forthcoming Main action was provided only to the Actor through headphones. Results demonstrate an exaggeration of spatial and temporal actions’ parameters when acting for the Partner, in cooperative, as well as in competitive context. This finding suggests that the motor manifestation of social intention is largely determined by non-conscious implicit processes that seem little influenced by the context of social interaction. It has been suggested that the brain adopts a simplified strategy to coordinate a large number of degrees of freedom in motor control. Synergies have been proposed as a strategy to produce movements by recruitment of a small number of fixed modular patterns. However, there is no direct support for a synergistic organization of the brain itself. In this study, we recorded neural activities from the dorsal premotor cortex (PMd) of monkeys trained to reach and grasp differently shaped objects (grasping task) or the same object in different positions (reaching task). Non-negative matrix factorization (NNMF) was applied to the neural data to extract neural synergies, whose functional roles were verified in several ways. We found that motor cortex used similar neural synergies for grasping different objects; combining only a few of the synergies accounted for most of the variance in the original data. When used for single-trial task decoding, the synergy coefficients performed as well and robustly as the original data in both tasks. The synergy amplitudes for each unit were significantly correlated with the corresponding neuron’s firing rate. In addition, we also observed synergies shared between tasks and task-specific synergies, as shown before for muscle synergies. Altogether, we demonstrated that neural synergies are effective in describing neural population activity during reach to grasp movements and provide a new tool for interpreting neural data for movement control. Our understanding of reach-tograsp movements has evolved from the original formulation of the movement as two semi-independent visuomotor channels to one of interdependence. Despite a number of important contributions involving perturbations of the reach or the grasp, some of the features of the movement, such as the presence or absence of coordination between the digits during the pincer grasp and the extent of spatio-temporal interdependence between the transport and the grasp, are still unclear. In this study, we physically perturbed the index finger into extension during grasping closure on a minority of trials to test whether modifying the movement of one digit would affect the movement of the opposite digit, suggestive of an overarching coordinative process. Furthermore, we tested whether disruption of the grasp results in the modification of kinematic parameters of the transport. Our results showed that a continuous perturbation to the index finger affected wrist velocity but not lateral displacement. Moreover, we found that the typical flexion of the thumb observed in nonperturbed trials was delayed until the index finger counteracted the extension force. These results suggest that physically perturbing the grasp modifies the kinematics of the transport component, indicating a two-way interdependence of the reach and the grasp. Furthermore, a perturbation to one digit affects the kinematics of the other, supporting a model of grasping in which the digits are coordinated by a higher-level process rather than being independently controlled. NEW & NOTEWORTHY A current debate concerning the neural control of prehension centers on the question of whether the digits in a pincer grasp are controlled individually or together. Employing a novel approach that perturbs mechanically the grasp component during a natural reach-to-grasp movement, this work is the first to test a key hypothesis: whether perturbing one of the digits during the movement affects the other. When a monkey selects a piece of food lying on the ground from among other viable objects in the near vicinity, only the desired item governs the particular pattern and direction of the animal's reaching action. It would seem then that selection is an important component controlling the animal's action. But, we may ask, is the selection process in such cases impervious to the presence of other objects that could constitute potential obstacles to or constraints on movement execution? And if it is, in fact, pervious to other objects, do they have a direct influence on the organization of the response? The kinematics of macaques' reaching movements were examined by the current study that analysed some exemplars as they selectively reached to grasp a food item in the absence as well as in the presence of potential obstacles (i.e., stones) that could affect the arm trajectory. Changes in movement parameterization were noted in temporal measures, such as movement time, as well as in spatial ones, such as paths of trajectory. Generally speaking, the presence of stones in the vicinity of the acting hand stalled the reaching movement and affected the arm trajectory as the hand veered away from the stone even when it was not a physical obstacle. We concluded that nearby objects evoke a motor response in macaques, and the attentional mechanisms that allow for a successful action selection are revealed in the reaching path. The data outlined here concur with human studies indicating that potential obstacles are internally represented, a finding implying basic cognitive operations allowing for action selection in macaques. Coordinated reach-to-grasp movements require precise spatiotemporal synchrony between proximal forelimb muscles (shoulder, elbow) that transport the hand toward a target during reach, and distal muscles (wrist, digit) that simultaneously preshape and orient the hand for grasp. The precise mechanisms through which the redundant neuromuscular circuitry coordinates reach with grasp, however, remain unclear. Recently, Geed and Van Kan (2016) demonstrated, using exploratory factor analysis (EFA), that limited numbers of global, template-like transport/preshape- and grasp-related muscle components underlie the complexity and variability of intramuscular electromyograms (EMGs) of up to 21 distal and proximal muscles recorded while monkeys performed reach-to-grasp tasks. Importantly, transport/preshape- and grasp-related muscle components showed invariant spatiotemporal coupling, which provides a potential mechanism for coordinating forelimb muscles during reach-to-grasp movements. In the present study, we tested whether ensemble discharges of forelimb neurons in the cerebellar nucleus interpositus (NI) and its target, the magnocellular red nucleus (RNm), a source of rubrospinal fibers, function as neuronal correlates of the transport/preshape- and grasp-related muscle components we identified. EFA applied to single-unit discharges of populations of NI and RNm neurons recorded while the same monkeys that were used previously performed the same reach-tograsp tasks, revealed neuronal components in the ensemble discharges of both NI and RNm neuronal populations with characteristics broadly similar to muscle components. Subsets of NI and RNm neuronal components were strongly and significantly cross-correlated with subsets of muscle components, suggesting that similar functional units of reach-to-grasp behavior are expressed by NI and RNm neuronal populations and forelimb muscles. Importantly, like transport/preshape- and grasp-related muscle components, their NI and RNm neuronal correlates showed invariant spatiotemporal coupling. Clinical and lesion studies have reported disruption of coupling between reach and grasp following cerebellar damage; the present results expand on those studies by identifying a neuronal mechanism that may underlie cerebellar contributions to spatiotemporal coordination of distal and proximal limb muscles during reaching to grasp. We conclude that finding similar functional units of behavior expressed at multiple levels of information processing along interposito-rubrospinal pathways and forelimb muscles supports the hypothesis that functionally related populations of NI and RNm neurons act synergistically in the control of complex coordinated motor behaviors. Classically, it has been hypothesized that reach-to-grasp movements arise from two discrete parietofrontal cortical networks. As part of these networks, the dorsal premotor cortex has been implicated in the control of reaching movements of the arm, while the ventral premotor cortex has been associated with the control of grasping movements of the hand. Recent studies have shown that such a strict delineation of function along anatomical boundaries is unlikely partly because reaching to different locations can alter distal hand kinematics and grasping different objects can affect kinematics of the proximal arm; Here, we used chronically implanted multi-electrode arrays to record unit spiking activity in both PMd and PMv simultaneously while rhesus macaques engaged in a reach-to-grasp task. Generalized linear models were used to predict the spiking activity of cells in both areas as a function of different kinematic parameters, as well as spike history. To account for the influence of reaching on hand kinematics and vice-versa, we applied demixed principal component analysis to define kinematics synergies that maximized variance across either different object locations or grip types. We found that single cells in both PMd and PMv encode the kinematics of both reaching and grasping synergies suggesting that this classical division of reach and grasp in PMd and PMv, respectively, does not accurately reflect the encoding preferences of cells in those areas. In the conventional view of sensorimotor control, the premotor cortex (PM) plans actions that are executed by the primary motor cortex (M1). This notion arises in part from many experiments that have imposed a preparatory "planning" period, during which PM becomes active without M1. But during many natural movements, PM and M1 are co-activated, making it difficult to distinguish their functional roles. We leverage coupled dynamical systems models (cDSMs) to uncover interactions between PM and M1 during movements performed with no preparatory period. We build cDSMs using neural and behavioral data recorded from two non-human primates as they performed a reach-grasp-manipulate task. PM and M1 interact dynamically throughout these movements. Whereas PM drives the M1 in some situations, in other situations, M1 drives PM activity, contrary to the conventional assumption. Our DSM framework provides additional predictions differentiating the roles of PM and M1 in controlling movement. Recording of neural activity during grasping actions in macaques showed that grasp-related sensorimotor transformations are accomplished in a circuit constituted by the anterior part of the intraparietal sulcus (AIP), the ventral (F5) and the dorsal (F2) region of the premotor area. In humans, neuroimaging studies have revealed the existence of a similar circuit, involving the putative homolog of macaque areas AIP, F5 and F2. These studies have mainly considered grasping movements performed with the right dominant hand and only a few studies have measured brain activity associated with a movement performed with the left non-dominant hand. As a consequence of this gap, how the brain controls for grasping movement performed with the dominant and the non-dominant hand still represents an open question. A functional resonance imaging experiment (fMRI) has been conducted, and effective connectivity (Dynamic Causal Modelling, DCM) was used to assess how connectivity among grasping-related areas is modulated by hand (i.e., left and right) during the execution of grasping movements towards a small object requiring precision grasping. Results underlined boosted inter-hemispheric couplings between dorsal premotor cortices during the execution of movements performed with the left rather than the right dominant hand. More specifically, they suggest that the dorsal premotor cortices may play a fundamental role in monitoring the configuration of fingers when grasping movements are performed by either the right and the left hand. This role becomes particularly evident when the hand less-skilled (i.e., the left hand) to perform such action is utilized. The results are discussed in light of recent theories put forward to explain how parieto-frontal connectivity is modulated by the execution of prehensile movements.

The performance of patients with vascular parkinsonism (VPD) on a reach-to-grasp task was compared with that of patients affected by idiopathic Parkinson’s disease (IPD) and age- matched control subjects. The aim of the study was to determine how patients with VPD and IPD compare at the level of the kinematic organization of prehensile actions. We examined how subjects concurrently executed the transport and grasp components of reach-to-grasp movements when grasping differently sized objects. When comparing both VPD and IPD groups to control subjects, all patients showed longer movement duration and smaller hand opening, reflecting bradykinesia and hypometria, respectively. Furthermore, for all patients the onset of the manipulation component was delayed with respect to the onset of the transport component. However, for patients with VPD this delay was significantly smaller than that found for the IPD group. It is proposed that this reflects a deficit - which is moderate for VPD as compared to IPD patients - in the simultaneous (or sequential) implementation of different segments of a complex movement. Altogether these findings suggest that kinematic analysis of reach-to-grasp movement has the ability to provide potential instruments to characterize different forms of parkinsonism.

Hand shaping during prehension involves intricate coordination of a complex system of bones, joints, and muscles. It is widely hypothesized that the motor system uses strategies to reduce the degrees of independent control. Both biomechanical constraints that result in coupling of the fingers and joints and neural synergies act to simplify the control problem. Synergies in hand shaping are typically defined using principal component-like analyses to define orthogonal patterns of movement. Although much less examined, joint angle velocities are also important parameters governing prehension. The primary goal of this study was to evaluate joint angles and joint angle velocities during prehension in monkeys. Fourteen joint angles and angular velocities were measured as monkeys reached to and grasped a set of objects designed to systematically vary hand shapes. Hand shaping patterns in joint angles and velocities were examined using singular value decomposition (SVD). Highly correlated patterns of movements were observed in both joint angles and joint angle velocities, but there was little correlation between the two, suggesting that velocities are controlled separately. Joint angles and velocities can be defined by a small number of eigenvectors by SVD. The unresolved question of the functional relevance of higher-order eigenvectors was also evaluated. This study explores the effects of visual condition and target size during four reach-to-grasp tasks between autistic children and healthy controls. Twenty children with autism and 20 healthy controls participated in the study. Qualisys motion capture system and kinematic measures were used to record movement. Autistic group showed significantly longer movement time, larger normalized jerk score, more movement unit than controls, especially in non-visual feedback and small target blocks. Autistic group also showed significantly larger maximal grip aperture and normalized maximal grip aperture in visual feedback condition than controls. Autistic children demonstrate motor coordination problems and also depend on more visual cuing in high accuracy tasks. Autistic children develop other compensatory skills while performing tasks. Abstract: The purpose of this experiment is to investigate the fine motor performance of young and older adults on a reach to grasp task in a desktop virtual environment with increasing precision requirements. Aging brings about potential loss of an individual's function due to disease, injury, or the degenerative nature of aging itself. Three-dimensional virtual environments have been identified as systems with good potential to ameliorate such problems in older individuals, and precise fine motor skills represent an important class of functional skills. Two groups of participants (Young, n=10, mean age 21.3 years, range 20–24, senior, n=10, mean age 70.7 years, range 60–85) performed a reach to grasp in a desktop virtual environment with simple, low contrast graphics. Results indicate that visual feedback of the hand for sensory guidance of movement did not improve motor performance for either group, and that as precision requirements of the task increased, age group differences in movement time and peak grasp aperture also increased. These findings extend the literature on age group differences in human motor control across the lifespan and differ from previous studies which showed presence of visual feedback of the hand improved motor performance in young adults. Differences in luminance contrast levels in past studies and the current one suggest that control over this feature of the visual scene is an important design consideration for all end-users and warrants additional investigation. Additional recommendations for age-specific design of three dimensional user interfaces include usage of tangibles that are sufficient in size to limit detrimental effects for older adults.
