1. Cation and voltage dependence of lidocaine inhibition of the hyperpolarization-activated cyclic nucleotide-gated HCN1 channel

Abstract

Abstract Lidocaine is known to inhibit the hyperpolarization-activated mixed cation current (Ih) in cardiac myocytes and neurons, as well in cells transfected with cloned Hyperpolarization-activated Cyclic Nucleotide-gated (HCN) channels. However, the molecular mechanism of Ih inhibition by this drug has been limitedly explored. Here, we show that inhibition of Ih by lidocaine, recorded from Chinese hamster ovary (CHO) cells expressing the HCN1 channel, reached a steady state within one minute and was reversible. Lidocaine inhibition of Ih was greater at less negative voltages and smaller current amplitudes whereas the voltage-dependence of Ih activation was unchanged. Lidocaine inhibition of Ih measured at −130 mV (a voltage at which Ih is fully activated) was reduced, and Ih amplitude was increased, when the concentration of extracellular potassium was raised to 60 mM from 5.4 mM. By contrast, neither Ih inhibition by the drug nor Ih amplitude at +30 mV (following a test voltage-pulse to −130 mV) were affected by this rise in extracellular potassium. Together, these data indicate that lidocaine inhibition of Ih involves a mechanism which is antagonized by hyperpolarizing voltages and current flow.

Abstract

In the area of the construction sector, the primary concern is economic growth. There are many issues and challenges associated with it. The growth and rapid development of this industry put their players, mainly the construction firms feeling the heat by competing with each other. The main challenge is the completion period for a particular project. This study was done in Manila, Philippines, where Design and Build (D&B) method has gained much interest and considered very important today. The rise of new innovative systems saw the emergence of new factors in the construction industry that will become future adoption. This study focuses on factors affecting the successful completion period of the construction industry, whereby it is very crucial for the country's economic and market development. A quantitative research methodology was applied in this study. A total of 967 survey questionnaires sent out, and only 396 respondents fit to be analysed. Respondents involved were contractors, consultants, and developers. It was learned that critical success factors in project management play significant roles in driving projects to examine the determinants of success completion time of Design & Build construction projects. Findings from this study shall be used by the authority in drafting a policy guideline for future construction development. It is hoped that this study provides a better understanding of construction challenges for future undertaking.

Abstract

This study reviews the developments in the onshore and offshore renminbi derivatives markets. The onshore market has seen a rapid build-up in the market infrastructure and price discovery mechanism in the past year, with empirical evidence suggesting that its pricing is increasingly determined by financial fundamentals, such as the covered interest rate parity. However, the growth of the market has been restrained by restrictions on the participant base, limited variations in the RMB/US$ exchange rate, market participants’lack of technical capacity and experience, and inadequate supporting financial market infrastructure. The non-deliverable forward (NDF) market, concentrated in Hong Kong and Singapore, is more developed, but has the drawback that its pricing is not tied to financial fundamentals. The comparison between onshore and offshore markets suggests that two issues are of particular importance for future derivatives market development in China: the balance between regulation and development, and the relationship between onshore and offshore markets.

Abstract

Damping-off and root rot of greenhouse cucumbers grown in rock wool, caused by Pythium aphanidermatum, is a recurring problem for growers throughout the major production areas of Canada. Four commercially formulated biocontrol agents, Streptomyces griseoviridis strain K61 (Mycostop®), Trichoderma harzianum strain T-22 (RootshieldTM Drench), Trichoderma virens strain GL-21 (SoilGardTM 12G), and Gliocladium catenulatum strain J1446 (PrestopTM WP, Prestop mix) were evaluated in growth-chamber and greenhouse trials conducted over 2 years for efficacy against this disease. The agents were applied twice, once at seeding time and again 11 days later, as a drench (or incorporated into the sawdust medium for SoilGard). This was followed by inoculation with a P. aphanidermatum mycelial suspension 12 days after seeding. In growth-chamber trials conducted at 28–30 °C and 90% relative humidity, seedling mortality in the treatment receiving P. aphanidermatum alone was over 80%. 9. The Macroeconomic Impact on Hong Kong of Hypothetical Mainland Shocks

Abstract

As the Hong Kong economy became increasingly integrated with that of Mainland China, it has benefited tremendously from the strong performance of the Mainland economy. At the same time, skilful macroeconomic management and sustained structural reforms on the Mainland have engendered a virtuous cycle, which reduces risks to Hong Kong from any potential adverse developments on the Mainland. Nevertheless, questions have been raised how Hong Kong will fare in the unlikely event that the Mainland is hit by economic shocks, which could cause the economy to deviate from its envisaged robust medium-term growth path. Shocks could be transmitted from the Mainland to Hong Kong through two main channels. First, Hong Kong¡¦s exports and imports are likely to be affected by sharp fluctuations in Mainland macroeconomic variables. Reduced export earnings and changes in terms of trade will then spill over to the Hong Kong domestic economy. Second, monetary and financial conditions in Hong Kong may be altered by changes in investor confidence and in fund flows as a result of Mainland shocks. The resulting change in Hong Kong dollar interest rates will impact asset prices and domestic demand. We use an econometric model to quantify the impact on Hong Kong of a range of Mainland macroeconomic shocks. Seven hypothetical scenarios are considered: external shocks include a large renminbi revaluation, a significant US economic slowdown, a trade war, and an oil price hike; domestic shocks include an investment retrenchment, a credit crunch, and financial instability. The magnitudes of the shocks are deliberately set to be large¡Xtypically taken as two standard deviations of the shock variable based on historical observations over a ten year period. 10. New process monitor for reticles and wafers: the MEEF meter

Abstract

In this paper, we present experimental results with a prototype design of a 'MEEF Meter,' and investigate its sensitivity to defocus and exposure changes. We find that the results of the MEEF meter complement the results obtained through conventional CD metrology, with conventional CDs being a good indicator of exposure changes, while the MEEF meter is a good indicator of defocus changes. We also investigate two kinds of MEEF meter, a dense MEEF meter and 'Process' MEEF meter design, and find the results for MEEF similar, but that the 'Process' MEEF meter design is far more susceptible to noise and bridging.

Abstract

Three 4X test reticles for 0.15 - 0.18 micrometer lithography are characterized using a critical-dimension scanning electron microscope (CD-SEM). The reticles, which contain metrology cells of isolated and nested lines, are manufactured with an i-line (365 nm) laser patterning tool and wet etch (reticle A), an e-beam patterning tool and dry etch (reticle B), and an e-beam patterning tool and wet etch (reticle C). We investigate the nature of pattern density-induced CD variations by studying groups of metrology cells with systematically varying linewidths and line spacings. The CD bias between nested and isolated lines is a smoothly increasing function of pattern density on reticles B and C. The CD signatures on reticle B exhibit strong microloading and loading signatures related to the plasma etch process. The bias characteristic on reticle A demonstrates a threshold effect, increasing sharply as the space between adjacent nested lines becomes less than approximately 600 nm. Matching images taken with secondary and backscattered electron spectra support the conclusion that the laser write process is resolution-limited at this pattern density. The SEM data is also compared to data from broad-band optical metrology tools to show that the optical response for dense features becomes unreliable when the feature spacing is less than approximately 600 nm.

Abstract

Anti‐CD19 Chimeric Antigen Receptor (CAR)‐T cell therapies have shown promise for treating B cell malignancies, but the clinical outcome is influenced by both the CAR‐T product and the patient's immune system. The role of γδ T cells in the context of CAR‐T cell therapy remains poorly understood. This study investigates the transcriptional heterogeneity, clonal expansion and dynamics of γδ T cells in patients undergoing anti‐CD19 CAR‐T cell therapy. Longitudinal single cell multi‐omics analysis was performed on γδ T cells from four patients receiving anti‐CD19 CAR‐T cell therapy. Single cell RNA‐seq, antibody‐based protein profiling (AbSeq) and full‐length TCRγδ sequences revealed clonally expanded populations displaying plasticity in T cell differentiation, and temporal dynamics of large clones, suggesting ongoing expansion and differentiation. Clonally expanded γδ T cells had heterogeneous gene expression profiles, occupying seven transcriptionally distinct clusters. Analysis of chemokine markers indicated cluster‐specific homing tendencies of circulating γδ T cells to peripheral tissues. We found unexpectedly high frequencies of Vδ1 and Vδ3 cells in the blood with distinct gene and protein expression profiles. This analysis provides insights into the dynamic and heterogeneous nature of γδ T cells following anti‐CD19 CAR‐T cell therapy, contributing valuable information for optimizing CAR‐T cell therapies in B cell malignancies.
