The metabolic fate of tolazamide, 1-(hexahydroazepin-1-yl)-3-p-tolylsulfonylurea (1), was studied in man and in the rat using tritium-labeled 1. The metabolites were isolated in crystalline form from urine for structure determination. The crystal structure and final molecular structure of one of these, 1-(4-hydroxyhexahydroazepin-1-yl)-3-p-tolylsulfonylurea (5), were determined using single-crystal X-ray techniques. Following oral administration of tritiated tolazamide to male humans, 85% of the radioactivity was excreted in urine during a 5-day period. In addition to being excreted in urine unchanged, tolazamide was metabolized to 1-(hexahydroazepin-1-yl)-3-p-(carboxyphenyl)sulfonylurea (2), p-toluenesulfonamide (3), 1-(hexahydroazepin-1-yl)-3-p-(hydroxymethylphenyl)sulfonylurea (4), 1-(4-hydroxyhexahydroazepin-1-yl)-3-p-tolylsulfonylurea (5) and a labile, unidentified metabolite 6 by man. The relative amounts of these materials excreted in 0-24-h urine collections from eight subjects averaged 7, 17, 26, 10, 25, and 15% for 1-6, respectively. In the female rat, 79% of an orally administered dose of tritiated tolazamide was excreted in urine during a 5-day period as 1-4. The relative amounts of these materials excreted during the 24-h period following administration of tolazamide were 10, 5, 5, and 80% for 1-4, respectively.

A spectrophotometric procedure for the determination of steroid oximes is described utilizing 11β-hydroxy-iia-methyl-5β-pregnane -3, 20 -dione dioxime (I) and iia-hydroxy-progesterone dioxime (II) as models. The method is based on (a) acid hydrolysis to release hydroxylamine, (b) oxidation of hydroxylamine to nitrous acid with iodine, (c) diazotization of sulfanilic acid with the nitrous acid, (d) coupling of the diazonium compound with N-(i-naphthyl)-ethylenediamine dihydrochloride, and (e) measurement of the azo dye at 550 nm. A stoichiometric yield of hydroxylamine is obtained. Analyses of I and II have given typical recoveries (mean ± S. D.) of 99.5 ± 1.1% and 97.8 + 3.5%, respectively. The method is sensitive to ≥ 0.3 × 10−5 M dioxime.

The effective use of tritiated compounds in biological systems often requires determination of the extent to which the tritium label has been removed from the compound and equilibrated with biological fluids. (1) This usually is accomplished by first determining the total tritium content of a water-containing sample and then determining either the tritiated water separated by evaporation or the tritium content of the dried sample. The former method gives a direct measure of tritiated water but involves isolating the tritiated water. In the latter method, the amount of tritium in the water is calculated as the difference between the tritium contents of the water-containing and dried samples. Although this indirect method is less accurate than the direct method, it does not require isolating the tritiated water. In both cases, it is important that very mild conditions be used for separating water from the aqueous sample in order to prevent further removal of tritium from the compound. This is accomplished conveniently by a freeze-drying procedure. Although this method is old and likely to be in common use, a convenient apparatus for processing large numbers of samples has not been described in the literature. The apparatus described in this report has been used routinely in several of our laboratories for many years to separate and collect tritiated water from biological samples, usually urine.

Minoxidil (2,4-diamino-6-piperidinopyrimidine 3-oxide), a potent new hypotensive agent, was 14C-labeled in the 2-position of the pyrimidine ring in 17 percent yield from Ba14CO3. This material was used to study the absorption, distribution, and excretion of the drug in rats, dogs, and monkeys. Following oral administration of a single dose, the drug was rapidly and well absorbed and rapidly eliminated by each species as judged by plasma levels and urinary excretion of unchanged drug and total drug-related materials. Chronic oral administration of the drug at a high level (10 mg/kg) for 30 days slightly increased the rate of clearance of minoxidil and minoxidil-related material from circulation. Water diuresis, resulting from water loading of dogs, caused an even greater increase in the rates of disappearance of the drug and drug-related material. Whole-body autoradiography studies in rats showed that minoxidil was rapidly distributed following its oral and intravenous administration. It was subsequently concentrated, primarily in the excretory system. Minoxidil-related material was detected in aorta walls, but not in the CNS, following both routes of drug administration.

Colestipol hydrochloride, a polymeric, ion-exchange type, hypocholesterolemic agent, acting by sequestering bile acids, was labeled with carbon-14. The disposition of the labeled material was studied in the human, dog and rat. The extent of absorption from the gastrointestinal tract, as judged by urinary excretion of radioactivity, was very small and correlated well with the contents of water-soluble and dialyzable materials in the colestipol hydrochloride. Results were consistent with the dialyzable material in the drug being the absorbable species.

Serum drug levels following oral administration of large single doses of chlorphenesin carbamate and methocarbamol were fitted to a one-compartment open model. The appearance of each drug was described by a short lag time followed by a rapid first-order absorption and attainment of maximum levels. No statistically significant differences were found between estimates of mean lag times (t0, 0.5 and 0.2 hr.), half-lives for absorption (t1/2, 0.4 and 0.6 hr), and times for attainment of maximum serum concentrations (tm, 1.9 and 1.4 hr.) for chlorphenesin carbamate and methocarbamol, respectively. Chlorphenesin carbamate was distributed throughout a significantly larger relative volume (1.27 versus 0.48 1./kg.) and yielded a correspondingly smaller maximum serum concentration (15.3 versus 29.8 mcg./ml., 2-g. dose). The mean biological half-life for chlorphenesin carbamate, 3.14 hr., was significantly greater than the value of 1.20 hr. obtained for methocarbamol.
