27. GC-MS profiling and evaluation of anti-tyrosinase and anti-proliferative activities of Curcuma amada leaf essential oil: An integrated experimental and in silico approach.

Abstract

Curcuma amada Roxb. (Zingiberacae) is a highly valued rhizomatous perennial herb widely recognized for its culinary and medicinal properties. However, the anti-tyrosinase and anti-proliferative properties of C. amada leaf essential oil (CALEO) remain unexplored Therefore, the present study aimed to characterize the chemical composition and bioactivities of CALEO with an in silico analysis of its major constituents. A total of 46 compounds, accounting for 90.56% of the total oil, were identified by gas chromatography-mass spectrometry (GC-MS) analysis. CALEO was characterized by a high content of monoterpene hydrocarbons with camphor (13.48%) being the predominant constituent. CALEO showed significant antiproliferative activity against PC3, A375 and HepG2 cell lines with IC50 values of 26.67, 39.14 and 30.89 μg/mL, respectively. Additionally, CALEO exhibited potent tyrosinase inhibitory activity (IC50: 95.05 μg/mL) when compared to kojic acid (IC50: 82.84 μg/mL). Curdione, β-caryophyllene, germacrone and ar-turmerone showed strong binding affinities (< −5.5 kcal/mol) against mushroom tyrosinase, Bcl-2 and EGFR proteins. PASS prediction results highlighted the dermatological, antimetastatic and antineoplastic potential of spathulenol and β-caryophyllene. The cell-line cytotoxicity predictor (CLC) results predicted that all the major constituents have strong anticancer activity against various cancerous cell lines. The present results suggest that CALEO could be used as a biologically active natural product with promising antiproliferative and dermatoprotective effects. [ABSTRACT FROM AUTHOR]

Abstract

Piper nigrum (Black pepper), popularly called the "King of Spices" is one of nature's most ancient and miraculous traditional plants to treat several deadly maladies. The essential oil of P. nigrum is highly considered to have illustrious therapeutic significance in domestic and industrial applications. To validate the traditional claim of its essential oil's efficacy, the proposed study was designed to analyze the volatile profile and assess the antimicrobial and antiproliferative effect of P. nigrum fruit essential oil. This is the first study on the analysis of essential oil of P. nigrum collected from Eastern India. The Gas chromatography-mass spectrometry analysis of P. nigrum essential oil revealed β-caryophyllene (25.6%), limonene (15.7%), sabinene (11.6%), β-pinene (11.1%) and δ-carene (8.6%) as its major constituents. In antibacterial assays, P. nigrum essential exhibited significant activity against multiple microbial pathogens like Salmonella enterica serovar Typhi (ZOI-19±0.4 mm; MIC-0.781 μg/ml), Escherichia coli (ZOI-18±0.4 mm; MIC-1.562 μg/ml) and Staphylococcus saprophyticus (ZOI-18±0.4 mm; MIC-1.562μg/ml). MTT assay demonstrated selective cytotoxicity against cancerous cells such as HepG-2 (IC50-71.2±1.3 μg/ml); PC-3 (IC50-68.38±1.2 μg/ml) as compared to noncancerous 3T3L-1 cells (IC50- 235.77±1.6 μg/ml).To the best of our knowledge, this is the first-ever study from India on the cytotoxic potential of P. nigrum essential oil against the above two robust cancerous cell lines. The present findings revealed that P. nigrum essential oil exhibited remarkable therapeutic potential, which could enhance its medicinal worth and commercial significance in current scenario and in future perspectives for designing antimicrobial and anticancer drugs. Abstract

Herein, a low RESET current 1T1R mushroom‐cell phase‐change memory (PCM) device that uses fiber‐textured homostructure GeSbTe (GST) grown on highly oriented TiTe2 seed layer is reported. The homostructure device outperformed the industry standard device, that uses doped polycrystalline GST, on most figures of merit. The homostructure devices are also benchmarked against superlattice (SL) PCM devices with 10 periods of 5/5 nm GST/Sb2Te3 grown on the TiTe2 seed layer, and are found to have same low RESET current. It is also observed by transmission electron microscopy that the alternating layers of GST/Sb2Te3 and TiTe2/Sb2Te3 in SL devices are intermixed in the switched region after the devices are cycled with RESET/SET pulses. Additionally, when the SL device is left in the SET state, the intermixed switched region crystallinity is textured and exhibits van der Waals gaps. The SL PCM devices require a precise layered structure that is hard to yield on a full wafer scale. In contrast, fiber‐textured homostructure PCM cells reported here are easily manufacturable, while providing similarly low RESET current and low‐resistance drift, which makes this device suitable for analog artificial intelligence computation.
