La trypanosomiase humaine africaine ou maladie du sommeil est causée par plusieurs sous-espèces membres de l'espèce Trypanosoma brucei : Trypanosoma brucei rhodesiense (Tbr) et Trypanosoma brucei gambiense groupe 1 (Tbg1). Leur biologie a été étudiée en détail, en particulier en ce qui concerne les protéines impliquées dans les mécanismes de résistance au sérum humain (SH). Il existe un autre groupe également capable d'infecter l'homme nommé Trypanosoma brucei gambiense groupe 2 (Tbg2). Celui-ci se résume à une trentaine de souches répertoriées dans la littérature, ne pose pas de problème de santé publique et par conséquent a été très peu étudié, pour ne pas dire oublié. Après un rappel des mécanismes qui permettent à Tbr et Tbg1 de résister à l'action lytique du SH, nous résumerons les connaissances acquises sur Tbg2. Les phénotypes de résistance au sérum humain sont très variables et seulement une de ces souches a fait l'objet de quelques recherches. La résistance de cette dernière est conditionnelle et implique des mécanismes ayant une action directe sur ApoL-1. Cette revue sera donc l'occasion non seulement de rappeler l'état des connaissances concernant la « course à l'armement » menée entre les trypanosomes et l'homme au cours de l'évolution, mais nous permettra également d'entamer une réflexion sur ce groupe particulier et hétérogène qu'est Tbg2, un trypanosome pathogène pour l'homme mais dont le caractère zoonotique doit être pris en compte dans la perspective du contexte de post-élimination de la maladie du sommeil.

Les trypanosomiases humaines et animales, maladies mortelles affectant les plus pauvres, sont dues à des trypanosomes transmis de manière cyclique par la mouche tsé-tsé, et mécaniquement par d'autres insectes hématophages. Toute action visant à éliminer le vecteur aura donc un impact sur la transmission de ces parasites. Nous revoyons ici les différents contextes, stratégies, et méthodes de lutte existantes appliquées en Afrique, ainsi que quelques perspectives de recherche prometteuses

Population genetics is a convenient tool to study the population biology of non-model and hard to sample species. This is particularly true for parasites and vectors. Heterozygote deficits and/or linkage disequilibrium often occur in such studies and detecting the origin of those (Wahlund effect, reproductive system or amplification problems) is uneasy. We used new tools (correlation between the number of times a locus is found in significant linkage disequilibrium and its genetic diversity, correlations between Wright's F-IS and F-ST, F-IS and number of missing data, F-IT and allele size and standard errors comparisons) for the first time on a real data set of tsetse flies, a vector of dangerous diseases to humans and domestic animals in sub-Saharan Africa. With these new tools, and cleaning data from null allele, temporal heterogeneity and short allele dominance effects, we unveiled the coexistence of two highly divergent cryptic clades in the same sites. These results are in line with other studies suggesting that the biodiversity of many taxa still largely remain undescribed, in particular pathogenic agents and their vectors. Our results also advocate that including individuals from different cohorts tends to bias subdivision measures and that keeping loci with short allele dominance and/or too frequent missing data seriously jeopardize parameter's estimations. Finally, separated analyses of the two clades suggest very small tsetse densities and relatively large dispersal.

Tsetse flies (Glossina), are African hematophagous Diptera, main vectors of animal and human trypanosomoses in Africa. The Sterile Insect Technique (SIT), has been success-ful to control tsetse when conducted on isolated populations. A new approach, called boosted SIT, combining SIT with the contamination of wild females by sterile males coated with pyriproxyfen (PP) has been proposed for large-scale control of vector pop-ulations. The aim of the study was to determine in laboratory conditions the transfer dynamics of PP from a sterile male to a female during mating, as well as the impact of PP on female fertility. To do this, we developed a method of treating sterile males by impregnating them with a powder containing 40 % of PP. The results showed that the PP had no impact on the survival of sterile males. With respect to persistence, PP per-sisted on sterile males up to 10 days at a dose of 100 ng. In addition, we showed a hor-izontal transfer of PP from a sterile male treated to a female. During mating, females received an average of 50 ng PP and by simple contact without mating more than 10 ng PP. Ongoing experiments will verify the impact of the transferred PP on female fertility. These initial results must be confirmed under field conditions. Eventually, the large-scale application of the boosted SIT approach against tsetse flies could be implement-ed in Africa. Six partenaires le CIRAD, le CNRS, l'EID Méditerranée, l'INRA, l'IRD, l'UM (Université de Montpellier), collaborent autour des problématiques liées aux arthropodes vecteurs de pathogènes responsables de maladies infectieuses humaines ou animales et aux ravageurs de cultures. Vectopole Sud est un réseau unique en Europe, regroupant à Montpellier de très nombreuses compétences autour de ses problématiques avec un dispositif d'environ 400 agents dont plus de 110 cadres de recherche permanents. Les Unités Mixtes de Recherche (UMR) ASTRE, DGIMI, MIVEGEC, INTERTRYP et l'EID-Méditerranée intégrées à ce réseau mènent des activités de recherche et de formation se situant à de multiples interfaces entre biologie et santé, écologie et environnement, évolution et médecine. La démarche du réseau Vectopole Sud est d'associer santé publique humaine, vétérinaire et végétale en 'Une seule Santé' ('One Health'). A cette fin, Vectopole Sud regroupe 4 plateformes d'expérimentation, insectariums confinés et laboratoires associés, sur les arthropodes vecteurs et ravageurs, réparties sur 4 sites de l'agglomération Montpelliéraine : - La plateforme Vectopole du site de la Valette (Campus de l'IRD – UMR Mivegec, IRD/UM/CNRS), disposant de compétences en matière de recherche sur les vecteurs et les pathogènes d'importance médicale ; -la plateforme du site de Baillarguet (Campus du CIRAD – UMR ASTRE, CIRAD/INRA), disposant de compétences en matière de recherche sur les vecteurs et les pathogènes d'importance vétérinaire ; -la plateforme du site de Triolet (PIQ – Plateforme d'Insectes de Quarantaine, campus de l'UM – UMR DGIMI, UM/INRA), disposant de compétences en matière de recherche sur les insectes d'intérêt agronomique ; -la plateforme de l'EID Méditerranée, opérateur public de démoustication, développant des méthodes et outils de lutte innovants contre les moustiques nuisants et vecteurs potentiels de maladies infectieuses.

Background Important control efforts have led to a significant reduction of the prevalence of human African trypanosomiasis (HAT) in Côte d’Ivoire, but the disease is still present in several foci. The existence of an animal reservoir of Trypanosoma brucei gambiense may explain disease persistence in these foci where animal breeding is an important source of income but where the prevalence of animal African trypanosomiasis (AAT) is unknown. The aim of this study was to identify the trypanosome species circulating in domestic animals in both Bonon and Sinfra HAT endemic foci. Methodology/Principal findings 552 domestic animals (goats, pigs, cattle and sheep) were included. Blood samples were tested for trypanosomes by microscopic observation, species-specific PCR for T. brucei sl, T. congolense, T. vivax and subspecies-specific PCR for T. b. gambiense and T. b. gambiense immune trypanolysis (TL). Infection rates varied significantly between animal species and were by far the highest in pigs (30%). T. brucei s.l was the most prevalent trypanosome species (13.7%) followed by T. congolense. No T. b. gambiense was identified by PCR while high TL positivity rates were observed using T. b. gambiense specific variants (up to 27.6% for pigs in the Bonon focus). Conclusion This study shows that domestic animals are highly infected by trypanosomes in the studied foci. This was particularly true for pigs, possibly due to a higher exposure of these animals to tsetse flies. Whereas T. brucei s.l. was the most prevalent species, discordant results were obtained between PCR and TL regarding T. b. gambiense identification. It is therefore crucial to develop better tools to study the epidemiological role of potential animal reservoir for T. b. gambiense. Our study illustrates the importance of “one health” approaches to reach HAT elimination and contribute to AAT control in the studied foci., Author summary In Africa, significant efforts to control human African trypanosomiasis (HAT) over the past three decades have drastically reduced the prevalence of the disease and elimination seems today an achievable goal. However, potential animal reservoirs of Trypanosoma brucei gambiense may compromise this ambitious objective. In the Bonon and Sinfra HAT endemic foci in Côte d’Ivoire, no recent data are available about the prevalence of animal African trypanosomiasis (AAT). The aim of this study was to identify trypanosomes circulating in domestic animals in these two HAT foci using serological, parasitological and molecular tools. We showed that T. brucei s.l. and T. congolense were the most prevalent trypanosome species and that pigs and cattle were the most infected animals. Discordant results were observed between the T. b. gambiense specific molecular and serological tools and the presence of an animal reservoir for T. b. gambiense remains unclear. Nevertheless, improved control strategies can be proposed based on this study to reach HAT elimination and contribute to AAT control in the study areas.
