Objective: To analyze wound contraction and histomorphometric pattern of lesions in Wistar rats undergoing doxorubicin extravasation. Method: Sixty adult female rats were used, divided into four groups of fifteen animals: Group 1 (Control, without antidote); Group 2 (Hyaluronidase); Group 3 (Photobiomodulation), and Group 4 (Hyaluronidase + Photobiomodulation). Doxorubicin 1mg (0.5 ml) was applied subcutaneously on the animals’ back, inducing the wound. Macroscopic and morphometric evaluation of the lesions was performed every two days for 28 days. On the 30th day, euthanasia was performed and the material was collected for histological evaluation. Results: The animals in the photobiomodulation and photobiomodulation + Hyaluronidase groups presented regeneration tissue with neovascularization and acute inflammation, with improvement in wound healing, which did not occur in the other groups. The contraction rates were better in those treated with photobiomodulation and photobiomodulation + Hyaluronidase, with healing percentages of 76.6% and 72.1%, respectively. Conclusion: The combination of photobiomodulation (660 nm–1 J) with topical hyaluronidase (65 UTR) proved to be effective in the process of wound healing due to extravasation of doxorubicin, and can be incorporated into the practice of clinical oncology. RESUMEN Objetivo: Analizar la contracción de heridas y patrón histomorfométrico de las lesiones en ratones Wistar sometidos a la extravasación de doxorrubicina. Método: Fueron utilizadas 60 ratas adultas, divididas en cuatro grupos de quince animales: Grupo 1 (Control sin antídoto); Grupo 2 (Hialuronidasa); Grupo 3 (Fotobiomodulación) y Grupo 4 (Hialuronidasa + Fotobiomodulación). Se aplicó 1 mg (0,5 ml) de doxorrubicina en el subcutáneo del dorso de los animales, induciendo la herida. Realizada evaluación macroscópica y morfométrica de las lesiones cada dos días por 28 días. En el 30° día se realizó la eutanasia y recolección del material para evaluación histológica. Resultados: Los animales de los grupos fotobiomodulación y fotobiomodulación+ Hialuronidasa presentaron tejido de regeneración con neovascularización e inflamación aguda, con mejoría de la cicatrización de las heridas, lo que no ocurrió en otros grupos. Las tasas de contracción fueron mejores en los tratados con fotobiomodulación y fotobiomodulación + Hialuronidasa, con porcentaje de cicatrización de 76,6% y 72,1%, respectivamente. Conclusión: la combinación de la fotobiomodulación (660nm–1J) con hialuronidasa tópica (65 UTR) demostró ser más eficaz en el proceso de cicatrización de heridas por extravasación de doxorrubicina, lo que posibilitó adoptarla en la práctica de la oncología clínica. Método: Utilizados 60 ratos fêmeas adultos, divididos em quatro grupos de quinze animais: Grupo 1 (Controle, sem antídoto); Grupo 2 (Hialuronidase); Grupo 3 (Fotobiomodulação) e Grupo 4 (Hialuronidase + Fotobiomodulação). Aplicado 1 mg (0,5 ml) de doxorrubicina no subcutâneo do dorso dos animais, induzindo a ferida. Realizada avaliação macroscópica e morfométrica das lesões a cada dois dias por 28 dias. As taxas de contração foram melhores nos tratados com fotobiomodulação e fotobiomodulação + Hialuronidase, com percentual de cicatrização de 76,6% e 72,1%, respectivamente. Lipids are the main forms of energy reserve in most organisms and they perform a variety of cellular functions. Among these, the polyunsaturated fatty acids of the omega-3 series stand out for having beneficial effects on human health. In view of this, this work sought to verify the effect over the brain activity of supplementation with omega-3 in Wistar rats that were exposed to ionizing radiation at a dose of 18 Gy on head and neck regions, the same dose used in radiotherapy treatments. 16 Rattus norvegicus were used, divided into 2 experimental groups, a group of animals supplemented with fish oil (GT) (1ml / OW / day / 100g LW) (n = 8), which is rich in omega-3, and another group who received a solution of 0.9% NaCl (GNT) (1ml / OW / day / 100g LW) (n = 8), both group were evaluated 24 hours before irradiation, in 24 hours and 1 week after irradiation. At the 17th postnatal week, the animals were submitted to a surgical procedure to implant electrodes. On the 15th postoperative day, the animals were irradiated. The ECoG recording of the animals was performed during space exploration of 24 hours pre-exposure, 24 hours post-exposure and 1 week post-exposure during 30 minutes. The ECoG signals were amplified, recorded and segmented. In the analysis of the time series of the ECoG signal, the power spectrum (PS) and the detrended fluctuation analysis (DFA) were used. These mathematical methods were sensitive in detecting changes in ECoG, due to the effect of omega-3 and radiation. In animals treated with omega-3, the PS technique showed that there was a significant decrease (p = 0.0184) in the beta rhythm power. While the DFA technique was applied to the different frequency ranges (theta, delta, alpha and beta) it revealed a statistically significant difference only for the theta wave (p = 0.0003). In the evaluation of the radiation effect through the PS, it was observed an increase in the power of the theta rhythm of the ECoG (p = 0.0453), only when they were evaluated one week after the irradiation (Late effect). Regarding the effect of radiation in animals treated with omega-3, it was observed that there was an increase in the power of the theta rhythm of the ECoG (p = 0.0220), only when they were evaluated one week after irradiation. The DFA technique applied to the different frequency bands also showed a statistically significant difference for the theta wave (p = 0.0053) in the group with omega-3 and non-irradiated in relation to the group with omega-3 and irradiated, groups evaluated one week after exposure to ionizing radiation. Difference also was found in the evaluation carried out between the group without omega-3 and with omega-3, after 24 hours (0.0055) and 1 week (p = 0.0252) of the irradiation. As for the beta rhythm, in the evaluation performed 1 week after irradiation, the DFA of the different frequency bands showed an alteration (p = 0.0067) between the group without and with omega-3. Os tratamentos oncológicos, como a radioterapia, produzem profundo impacto nas funções fisiológicas do organismo, causando efeitos diretos e indiretos nas estruturas vivas, podendo afetar o metabolismo de proteínas, lipídios e carboidratos. Os lipídeos são as principais formas de reserva energética na maioria dos organismos e desempenham uma variedade de funções celulares. Dentre esses, os ácidos graxos poliinsaturados da série ômega-3 se destacam por apresentarem efeitos benéficos à saúde humana. Em vista disso, buscou-se verificar o efeito da suplementação com ômega-3 na atividade cerebral em ratos Wistar que foram expostos a radiação ionizante numa dose de 18 Gy na região de cabeça e pescoço, a mesma utilizada em tratamentos radioterápicos. Foram utilizados 16 Rattus norvegicus divididos em 2 grupos experimentais, um grupo de animais suplementados com óleo de peixe (GT) (1ml/ VO/ dia/100g PV) (n = 8), que é rico em ômega-3, e um outro grupo de animais que recebeu uma solução de Na Cl 0,9% (GNT) (1ml/ VO/ dia/ 100g PV) (n = 8), ambos avaliados 24 horas antes da irradiação e 24 horas e 1 semana depois da irradiação. Na 17º semana pós-natal os animais foram submetidos ao procedimento cirúrgico para implante de eletrodos. No 15º dia pós-operatório os animais foram irradiados. O registro do ECoG dos animais foi realizado durante exploração espacial 24 horas pré-exposição, 24 horas pós-exposição e 1 semana pós-exposição durante 30 minutos para cada registro. Os sinais do ECoG foram amplificados, registrados e segmentados. Na análise da série temporal do sinal do ECoG foram utilizados o espectro de potência (EP) e a análise de flutuação Destendenciada (DFA). Esses métodos matemáticos foram sensíveis em detectar alterações no ECoG, tanto em decorrência do efeito do ômega-3 como da radiação. Nos animais tratados com ômega-3, a técnica do EP mostrou que houve uma redução significativa (p=0,0184) na potência do ritmo beta, enquanto, a técnica do DFA quando aplicada às diferentes faixas de frequência (teta, delta, alfa e beta) revelou uma diferença estatisticamente significativa unicamente para a onda teta (p= 0,0003). Na avaliação do efeito da radiação através do EP foi observado que houve um aumento na potência do ritmo teta do ECoG (p= 0,0453), somente quando avaliados uma semana após a irradiação. Quanto ao efeito da radiação em animais tratados com ômega-3 foi observado que houve um aumento na potência do ritmo teta do ECoG (p= 0,0220), somente quando avaliados uma semana após a irradiação (Efeito tardio). A técnica do DFA aplicada às diferentes faixas de frequência também mostrou diferença estatisticamente significativa para a onda teta (p= 0,0053) no grupo com ômega-3 e não irradiado em relação ao grupo com ômega-3 e irradiado, avaliados uma semana após a exposição a radiação ionizante. Diferença também encontrada na onda Teta para a avaliação realizada entre o grupo sem ômega-3 e com ômega-3, após 24 horas (0,0055) e 1 semana (p=0,0252) da irradiação. Quanto ao ritmo beta, na avaliação realizada com 1 semana após a irradiação, o DFA das diferentes faixas de frequência, mostrou alteração (p=0,0067) entre o grupo sem ômega-3 e com ômega-3.

This leads to a scenario with a high wound prevalence, being therefore essencial to improve the protocols for the treatment of skin lesions with pathological disturbances like these. The goal of this research is to describe the healing activity and toxicity of the aqueous extract of Psidium guajava in diabetic rats. The treatments were topical and oral in 4 groups of animals for the test of cicatrization (cutaneous lesions) and toxicity respectively: (I) non diabetic rats treated with the extract (0.2 g/mL); (II) non diabetic rat without treatment; (III) diabetic rats treated with the extract (0.2 g/mL) and (IV) diabetic rats without treatment, for the cicatrization tests. And, (I) rats treated with water; (II) rats treated with 100, 200 and 400 mg/KG, for the toxicity tests. It was performed histological, morphometric, histochemical and immunohistochemical tests for the cicatrization study, and histopathological, histochemical, biochemical and morphometric tests for the toxicity study. It was observed statistically significant keratinization in the animal groups I, II and III only. In the animals treated with the extract it was found the presence of numerous macrophages and giant cells. In the lesions of diabetic animals without treatment the concentration of collagen was smaller, and the rate of healing was higher by the 14 th day for the animals treated with the extract when compared to the other animals. The serum levels of enzymes Alanine ALT, AST, urea, creatinine and albumin were elevated in the animals treated with 400 mg/Kg. In the liver of the animals treated with 400 mg/Kg we evidenced a congestion of the portal vein, centrilobular and sinusoid capillaries. In the kidneys, with the same treatment, we observed the presence of areas with proliferative glomerulonephritis. The liver morphometry revealed a significant reduction of the lobular parenchyma and increase of the non lobular parenchyma in the animals treated with 400 mg/Kg as well as a significant glomerulus increase in diameter and volume , besides the diameter and volume of the Bowman capsules in the kidneys of animals treated with the same dosage. In the biochemical evaluation for glycogen there was a significant reduction in the liver of animals treated with 400 mg/Kg in relation to the others, which represents greater disadvantage with the use of such dosage, fact which does not occur in significant number for the other dosages, allowing therefore the recommendation of the crude extract of P. guajava as a promising phytotherapic asset. O aumento das doenças crônicas e de estimativas que apontam para 300 milhões de pessoas com Diabetes Mellitus, no mundo até o ano de 2025, indicam um cenário com elevada prevalência de feridas, portanto, é imprescindível o melhoramento dos protocolos de tratamento das lesões cutâneas com distúrbios patológicos como esse. O objetivo desta pesquisa foi descrever a atividade cicatrizante a toxicidade do extrato aquoso de Psidium guajava em ratos diabéticos. Os tratamentos foram tópico e oral em 4 grupos de animais para os teste de cicatrização (com lesões cutâneas) e toxicidade respectivamente: (I) ratos não diabéticos tratados com extrato (0,2 g/mL); II) ratos não diabéticos sem tratamento; III) ratos diabéticos tratados com extrato (0,2 g/mL) e IV) ratos diabéticos sem tratamento nos testes de cicatrização. (I) ratos tratados com água; (II) ratos tratados com 100, 200 e 400 mg/Kg para os testes de toxicidade. Foram realizadas as avaliações histológica, morfométrica, histoquímica e imunohistoquímica para o estudo da cicatrização e histopatológica, histoquímica, bioquímica e morfométrica para o de toxicidade. Observou-se queratinização significativa, estatisticamente, nos grupos de animais I, II e III apenas. Nos animais tratados com o extrato foi verificado a presença de numerosos macrófagos e células gigantes. Nas lesões dos animais diabéticos sem tratamento o teor de colágenos foi menor e a taxa de cicatrização, na avaliação ao 14° dia, foi elevada nos animais tratados com o extrato, quando comparados aos outros animais. Os níveis séricos das enzimas alanina ALT, AST, ureia, creatinina e albumina foram elevados nos animais tratados com 400 mg/Kg. No fígado dos animais tratados com 400 mg/Kg foi verificada a presença de congestão de veia porta, centrolobular e nos capilares sinusóides. Já nos rins, com o mesmo tratamento, foram observadas a presença de áreas com glomerulonefrite proliferativa. A morfometria do fígado revelou redução significativa do parênquima lobular e aumento no parênquima não lobular dos animais tratados com 400 mg/kg assim como aumento significativo do diâmetro e volume do glomérulo, além do diâmetro e volume da cápsula de Bowman nos rins dos animais tratados com a mesma dosagem. Na avaliação bioquímica para o glicogênio houve redução significativa no fígado dos animais do grupo tratado com 400 mg/kg em relação aos demais, o que mostra maior prejuízo com uso dessa dosagem, não ocorrendo alterações significativas para as demais e permitindo assim, a indicação do extrato bruto de P. guajava como recurso fitoterápico promissor.

The purpose of this study was to prove that the administration of Jati (Guazuma ulmifolia Lamk) leaves extract improves the lipid profile of dyslipidemic male Wistar rats. Subjects were 20 rats (Rattus norvegicus), male, Wistar strain, dyslipidemia (total cholesterol ≥ 200 mg dl-1), aged 2 months old, weighing 180-200 grams. The control group (10 rats) were given a placebo of 3 ml aquadest (P0) and the treatment group was given extracts of the Jati (Guazuma ulmifolia Lamk) leaves extract of 25 mg kg-1 BW (P1). Before and after treatment for 14 days, total cholesterol, triglyceride, LDL, and HDL levels were examined. To investigate the relationship between prognosis, changes in mitochondrial calcium uptake, and bioenergetic status in the heart during sepsis.In vivo and ex vivo controlled experimental studies.University research laboratory.Male adult Wistar rats.Sepsis was induced by intraperitoneal injection of fecal slurry. Sham-operated animals served as controls. Confocal microscopy was used to study functional and bioenergetic parameters in cardiomyocytes isolated after 24-hour sepsis. Electron microscopy was used to characterize structural changes in mitochondria and sarcoplasmic reticulum. The functional response to dobutamine was assessed in vivo by echocardiography.Peak aortic blood flow velocity measured at 24 hours was a good discriminator for 72-hour survival (area under the receiver operator characteristic, 0.84 ± 0.1; p = 0.03) and was used in ex vivo experiments at 24 hours to identify septic animals with good prognosis. Measurements from animals with good prognostic showed 1) a smaller increase in mitochondrial calcium content and in nicotinamide adenine dinucleotide fluorescence following pacing and 2) increased distance between mitochondria and sarcoplasmic reticulum on electron microscopy, and 3) nicotinamide adenine dinucleotide redox potential and adenosine triphosphate/adenosine diphosphate failed to reach a new steady state following pacing, suggesting impaired matching of energy supply and demand. In vivo, good prognosis animals had a blunted response to dobutamine with respect to stroke volume and kinetic energy.In situations of higher energetic demand decreased mitochondrial calcium uptake may constitute an adaptive cellular response that confers a survival advantage in response to sepsis at a cost of decreased oxidative capacity.

RESUMEN: Evaluar el proceso de reparación alveolar en ratas sometidas a cirugía de simulacro u ovariectomizadas tras el relleno alveolar con coágulo o con biosilicato cristalino. Sesenta ratas Wistar fueron divididas en cuatro grupos (n=15) de acuerdo con el tratamiento: Grupo 1- ratas sometidas a cirugía de simulacro con alveolos rellenados con coágulo; Grupo 2- ratas sometidas a cirugía de simulacro con alveolos rellenados con biosilicato cristalino; Grupo 3- ratas ovariectomizadas con alveolos rellenados con coágulo; Grupo 4- ratas ovariectomizadas con alveolos rellenados con biosilicato cristalino. Después de 7, 14 y 28 días, los animales fueron sacrificados, se tomaron muestras óseas que fueron teñidas con hematoxilina-eosina y analizadas al microscopio para realizar un análisis histomorfométricos. Los mayores porcentajes de formación de hueso se presentaron en los grupos 1 (32 % a los 7 días, 46 % a los 14 días y 83.5 % a los 28 días) y 4 (27,1 % a los 7 días, 41,1 % a los 14 días y 79,7 % a los 28 días). En los alveolos rellenados con coágulo, las ratas sometidas a cirugía de simulacro mostraron los mejores resultados, mientras que, en los alveolos rellenados con biosilicato, las ratas ovariectomizadas tenían porcentajes significativamente mayores. En este estudio, el biosilicato cristalino se comportó como un biomaterial adecuado para la reparación ósea, favoreciendo la osteoconducción. ABSTRACT: The objective of this study was to assess the process of alveolar bone repair in rats subjected to sham surgery or ovariectomized rats, after alveolar filling with clot or with crystalline biosilicate. Sixty Wistar rats were divided into four groups (n = 15) according to the treatment: Group 1 - rats subjected to sham surgery with sockets filled with clot; Group 2- rats submitted to sham surgery with sockets filled with crystalline biosilicate; Group 3 ovariectomized rats with sockets filled with clot; Group 4 ovariectomized rats with sockets filled with crystalline biosilicate. After 7, 14 and 28 days, the animals were sacrificed, bone samples were taken, stained with hematoxylin-eosin and analyzed under a microscope to perform a histomorphometric analysis. The highest percentages of bone formation were presented in groups 1 (32 % at 7 days, 46 % at 14 days and 83.5 % at 28 days) and 4 (27.1 % at 7 days, 41.1 % at 14 days and 79.7 % after 28 days). In the sockets filled with clot, the rats subjected to sham surgery showed the best results, while in the sockets filled with biosilicate, the ovariectomized rats had significantly higher percentages. In this study, the crystalline biosilicate behaved as an adequate biomaterial for bone repair, favoring osteoconduction.

International audience; Quinone reductase 2 (QR2, E.C. 1.10.5.1) is an enzyme with a feature that has attracted attention for several decades: in standard conditions, instead of recognizing NAD(P)H as an electron donor, it recognizes putative metabolites of NADH, such as N-methyl- and N-ribosyl-dihydronicotinamide. QR2 has been particularly associated with reactive oxygen species and memory, strongly suggesting a link among QR2 (as a possible key element in pro-oxidation), autophagy, and neurodegeneration. In molecular and cellular pharmacology, understanding physiopathological associations can be difficult because of a lack of specific and powerful tools. Here, we present a thorough description of the potent, nanomolar inhibitor [2-(2-methoxy-5H-1,4b,9-triaza(indeno[2,1-a]inden-10-yl)ethyl]-2-furamide (S29434 or NMDPEF; IC50 = 5-16 nM) of QR2 at different organizational levels. We provide full detailed syntheses, describe its cocrystallization with and behavior at QR2 on a millisecond timeline, show that it penetrates cell membranes and inhibits QR2-mediated reactive oxygen species (ROS) production within the 100 nM range, and describe its actions in several in vivo models and lack of actions in various ROS-producing systems. The inhibitor is fairly stable in vivo, penetrates cells, specifically inhibits QR2, and shows activities that suggest a key role for this enzyme in different pathologic conditions, including neurodegenerative diseases.

Objective To evaluate the β-hydroxy-βmethylbutyrate supplementation influence on the morphological and morphometric characteristics of the diaphragm muscle of rats and verify whether there are sex differences. Methods Experimental study with 48 Wistar rats (24 of each sex) divided into 3 groups: Control Group: in which a daily diet with saline solution was offered; Experimental Group: the same amount of food from the Control group consumed in the previous day and 0.3g/kg of β-hydroxy-βmethylbutyrate; Ad libitum Experimental Group: ad libitum feeding with the same dose of the supplement. Background: Whole body vibration training (WBV) is a new training program, which is safe and effective. It can be followed by the public. However, data on the safety and efficacy of vibration on myocardial ischemia reperfusion (IR) injury are lacking. Objective: To examine the effect of WBV on the tolerance of the myocardium to acute IR injury in an experimental rat model. Methods: Twenty-four male Wistar rats were divided into control and vibration groups. Vibration training consisted of vertical sinusoidal whole body vibration for 30 min per day, 6 days per week, for 1 or 3 weeks (WBV1 and WBV3 groups, respectively). All the rats were submitted to myocardial IR injury. Myocardial infarct size and ischemia-induced arrhythmias were assessed. Differences between variables were considered significant when p < 0.05. Results: No differences were observed between the groups regarding the baseline hemodynamic parameters. Infarct size was smaller in the experimental group (control, 47 ± 2%; WBV1, 39 ± 2%; WBV3, 37 ± 2%; p < 0.05, vs. control). Vibration produced a significant decrease in the number and duration of ventricular tachycardia (VT) episodes compared to the control value. All ventricular fibrillation (VF) episodes in the vibration groups were self-limited, while 33% of the rats in the control group died due to irreversible VF (p = 0.02). Conclusion: The data showed that vibration training significantly increased cardiac tolerance to IR injury in rats, as evidenced by reduction in the infarct size and cardiac arrhythmias, and by facilitating spontaneous defibrillation. Resumo Fundamento: O treinamento com vibração de corpo inteiro (WBV) é um novo programa de treinamento seguro e eficaz, e pode ser seguido pelo público. No entanto, dados sobre a segurança e eficácia da vibração na lesão de isquemia e reperfusão (IR) do miocárdio estão em falta. Objetivo: Examinar o efeito da WBV na tolerância do miocárdio à lesão aguda por IR em um modelo experimental em ratos. Métodos: Vinte e quatro ratos Wistar machos foram divididos em 2 grupos: controle e vibração. O treino de vibração consistiu em vibração sinusoidal vertical de corpo inteiro durante 30 min por dia, 6 dias por semana, durante 1 ou 3 semanas (grupos WBV1 e WBV3, respectivamente). Todos os ratos foram submetidos a lesão por IR do miocárdio. O tamanho do infarto do miocárdio e as arritmias induzidas por isquemia foram avaliados. As diferenças entre as variáveis foram consideradas significativas quando p < 0,05. Resultados: Não foram observadas diferenças entre os grupos em relação aos parâmetros hemodinâmicos basais. O tamanho do infarto foi menor no grupo experimental (controle, 47 ± 2%; WBV1, 39 ± 2%; WBV3, 37 ± 2%; p < 0,05, vs. controle). A vibração produziu uma diminuição significativa no número e duração das taquicardia ventriculares (TV) em comparação com o valor de controle. Todos os episódios de fibrilação ventricular (FV) nos grupos de vibração foram autolimitados, enquanto 33% dos ratos do grupo controle morreram devido a FV irreversível (p = 0,02). Conclusão: Os dados mostraram que o treinamento com vibração de corpo inteiro aumentou significativamente a tolerância cardíaca à lesão de IR em ratos, como evidenciado pela redução do tamanho do infarto e arritmias cardíacas, e pela facilitação da desfibrilação espontânea.

Background: Chronic kidney disease affects more than 500 million people worldwide. In this context, the uremic toxins present are related to worsening in tissue healing. Aim: Evaluate on healing of colonic anastomosis in uremic rats, serum and anatomopathological indicators, which may be related to the change tissue repair process. Methods: Twenty Wistar rats, were randomly separated into two groups. In the sham group they were submitted to 5/6 nephrectomy simulation in left kidney, simulation right nephrectomy, median laparotomy, colotomy and colorraphy. Cilj: Cilj ovog diplomskog je pokazati potencijalni pozitivni ili negativni učinak taninske kiseline na srčani mišić hiperglikemičnih štakora i ekspresiju koneksina 43 upotrebom koncentracije koja je već pokazala korisne učinke. Materijal i metode: U ovom je istraživanju dvanaest muških Wistar štakora podijeljeno u tri skupine na temelju napitaka koje su konzumirali. U svakoj skupini bila su po četiri štakora, a ispitivanje je trajalo dvadeset tjedana. Prva skupina, odnosno kontrolna skupina konzumirala je čistu vodu za piće, a druga skupina 30%-tnu vodenu otopinu saharoze. Treća skupina konzumirala je 30%-tnu vodenu otopinu saharoze i 0,1% taninsku kiselinu. Mjerena im je težina, a koncentracije glukoze u plazmi određivane su s glukometrom. Životinje su eutanazirane te njihova srca su uklonjena. Tkivo je pripremljeno i zatim obojeno trikromnim bojanjem po Mallory-ju. Material and Methods: In this trial twelve male Wistar rats were divided into three groups based on beverages they were consuming. There were four rats in each group and Trial lasted twenty weeks. Dietary fat sources may differentially affect the development of inflammation in insulin-sensitive tissues during chronic overfeeding. Considering the anti-inflammatory properties of ω-3 fatty acids, this study aimed to compare the effects of chronic high-fish oil and high-lard diets on obesity-related inflammation by evaluating serum and tissue adipokine levels and histological features in insulin-sensitive tissues (white adipose tissue, skeletal muscle and liver). As expected, a high-lard diet induced systemic and peripheral inflammation and insulin resistance. Conversely, compared with a high-lard diet, a high-fish oil diet resulted in a lower degree of systemic inflammation and insulin resistance that were associated with a lower adipocyte diameter as well as lower immunoreactivity for transforming growth factor β 1 (TGFβ1) in white adipose tissue. A high-fish oil diet also resulted in a lower ectopic lipid depot, inflammation degree and insulin resistance in the skeletal muscle and liver. Moreover, a high-fish oil diet attenuated hepatic stellate cell activation and fibrogenesis in the liver, as indicated by the smooth muscle α-actin (α-SMA) and TGFβ1 levels. The replacement of lard (saturated fatty acids) with fish oil (ω-3 fatty acids) in chronic high-fat feeding attenuated the development of systemic and tissue inflammation.

Background Early revascularization of ischemic organs is key to improving outcomes, yet consequent reperfusion injury may be harmful. Reperfusion injury is largely attributed to excess mitochondrial production of reactive oxygen species (ROS). Sulfide inhibits mitochondria and reduces ROS production. Ammonium tetrathiomolybdate (ATTM), a copper chelator, releases sulfide in a controlled and novel manner, and may offer potential therapeutic utility. Methods and findings In vitro, ATTM releases sulfide in a time-, pH-, temperature-, and thiol-dependent manner. Controlled sulfide release from ATTM reduces metabolism (measured as oxygen consumption) both in vivo in awake rats and ex vivo in skeletal muscle tissue, with a superior safety profile compared to standard sulfide generators. Given intravenously at reperfusion/resuscitation to rats, ATTM significantly reduced infarct size following either myocardial or cerebral ischemia, and conferred survival benefit following severe hemorrhage. Mechanistic studies (in vitro anoxia/reoxygenation) demonstrated a mitochondrial site of action (decreased MitoSOX fluorescence), where the majority of damaging ROS is produced. Conclusions The inorganic thiometallate ATTM represents a new class of sulfide-releasing drugs. What did the researchers do and find? We found that ammonium tetrathiomolybdate (ATTM) releases sulfide in a controlled manner and potentially fits the desired efficacy and safety profile. The chemical nature of sulfide release from ATTM differs from that of other sulfide-releasing drug classes. Le but de ce travail repose sur l'évaluation in vivo de l'effet de l'amphotericine B-desoxycholate liée aux lipoprotéines légères " LDL " et - lipoprotéines lourdes " HDL sur quelques paramètres sériques et urinaires chez le rat Wistar. En effet il est admis que, l'amphotericine B agit sur les cellules eucaryotiques et procaryotiques en perturbant l'organisation de leurs membranes plasmiques entraînant ainsi une altération de structure et aussi de la fonction cellulaire. Comme tous les antifongiques polyéniques, I'amphotericine B de large utilisation clinique induit d'importants effets toxiques ; C'est pourquoi beaucoup de stratégies ont été développées pour essayer de réduire la toxicité de la formulation commerciale (Amphotericine B-desoxycholate Fungizone " à savoir la forme complexée aux lipoprotéines endogènes stabilisant ainsi l'activité antifongale. Dans le cadre de l'élahoration d'une stratégie de développement en vu d'une thérapeutique nouvelle, nous avons étudié l'effet de l'amphoterieine Bdesoxycholate associée aux lipoproreines sériques sur quelques pararnetres - sériques et urinaires chez le rat Wistar. Dans notre travail, nous nous sommes basés d'une part sur la nature des lipoprotéines utilisées notamment les lipoprotéines (L DL, IIDL) qui ont été précipitées par la technique de (BURSTEIN et al, 1970), d'autre part l'effet de concentrations croissantes d'amphotericine B desoxycholate sur la fonction globale du rein fl ressort de cette étude que • La perfusion de Vamp B-desoxycholate à différentes concentrations allant de 2,5 g/ml - 5 ig/ml - 7,5 jig/ml - 10 j.tg/ml chez le rat Wistar à une courte durée induit des différentes perturbations - Hématologiques (hémoglobine, globule rouge) - Troubles électrolytiques plasmatiques, urinaires - Troubles de la fonction rénale. • L'addition des lipoprotéines sériques, macromolécules renfermant du cholestérol un site de fixation à l'antifongique, montrent des effets différents sur la toxicité de l'amp B suivant le type de lipoprotéines sériques. A des concentrations d'ainp B-desoxy de l'ordre de 10 ig/ml après 2 heures de perfusion, les lipoprotéines lourdes " HDL " à 7,88 g!t induisent un effet protecteur important qu'avec les lipoprotéines légères ' 1.1)1. Ce travail s’intéresse à l’évolution de quelques paramètres biochimiques sériques chez les rats wistar soumis à une injection intra-péritonéale de différentes doses d’extrait chloroformique des graines de la coloquinte Citrullus colocynthis ‘Handal’ famille des cucurbitacées. Cette plante est utilisée comme remède traditionnelle pour le traitement du diabète sucré dans la région de Maghreb et du Moyen-Orient. Mais, elle devient très toxique voir mortelle à des doses élevées. La présente étude est divisée en deux parties essentielles. La première est l’étude phytochimique de l’extrait chloroformique des graines de Citrullus colocynthis qui a révélé la présence de quelques groupes chimiques (les tanins, les stérols, les triterpènes et les composés réducteurs). En revanche, la deuxième partie consiste de l’évolution de la glycémie, le cholestérol total, les triglycérides, les transaminases (TGO, TGP, ALP) et la créatinine avant et 21 jours après l’injection intra-péritonéale de différentes doses (250mg/kg, 150mg/kg, 75 mg/kg, 50mg/kg). Les résultats obtenus montrent que notre extrait présente un effet toxique à une dose de 75 mg/kg p.c où nous avons marqué une toxicité hépatique et rénale chez les rats soumis à la même dose. La suralimentation durant la croissance a des effets néfastes sur la santé de l’individu en question, accompagnée d’une programmation de ces altérations tout au long de sa vie. La prédisposition à l’obésité et les troubles métaboliques associés persistent jusqu’à l’âge adulte. L’objectif de notre travail est d’évaluer certains paramètres biochimiques des rates nourries au régime hypergras (hyperlipidique et hypercalorique) après sevrage, et le suivi de ces altérations jusqu’à l’âge adulte. L’obésité chez les rates en phase de croissance est induite par le régime hypergras, arriver avec ce même régime jusqu’à l’âge adulte, ces rates obèses présente une hyperphagie suivi d’une augmentation du poids corporel, associée à une hyperglycémie, une hypercholestérolémie et une hypertriglycéridémie. La consommation du régime hypergras par les rates en durant phase de croissance accentue les modifications métaboliques à l’âge adulte. We used 80 submandibular glands of adult Wistar rats and divided into three groups: group I, control (n = 14); group II (n = 34) submitted the intraglandular injection of bovine gelatin (0.25 mg); and group III (n = 32) treated with intraglandular injection of botulinum toxin type A (2.5 U). The groups II and III were divided into 3 subgroups according to the day of analysis of glands (7, 15 and 28 days after treatment), when they were removed, heavy and measures immediately for morphometric analysis. Next, were fixed in paraformaldehyde 4 % and processed conventionally for light microscopy, stained in H.E., P.A.S., alcian blue, Gomori trichrome and picrosirius red. Morphometric differences were not observed between subgroups of group III, however in group II there was a significant difference between the three subgroups in the longitudinal length (p = 0.030) and transverse length (p = 0.025), having the gland has shown greater at day 28th after treatment. Histometrically, serous and mucous acini group III, were presented with a smaller diameter compared with the acinithe subgroups treated with bovine gelatina (group II). The wall thickness of the striated duct Group III was lower than in other. Histologically, there was a greater amount of cytoplasmic vacuolization in group III. addition to the greater spacing between the acini. Thus, we conclude that botulinum toxin type A interferes with the structural organization of the submandibular glands, promoting a reduction in the diameter of serous acini and in wall thickness striated ducts, macroscopically but this difference was not significant. O objetivo deste trabalho foi observar os aspectos morfométricos, histométricos e histológicos das glândulas submandibulares de ratas tratadas com toxina botulínica tipo A (BTX-A) e com gelatina bovina. Foram utilizadas 80 glândulas submandibulares de ratas Wistar adultas e divididas em três grupos: grupo I controle (n = 14); grupo II (n = 34) submetidas a injeção intraglandular de gelatina bovina (0,25 mg); e grupo III (n = 32) tratadas com injeção intraglandular de toxina botulínica tipo A (2,5 U). Os grupos II e III foram divididos em 3 subgrupos de acordo com o dia de análise das glândulas (7, 15 e 28 dias após o tratamento), quando foram removidas, imediatamente pesadas e medidas para análise morfométrica. A seguir, foram fixadas em paraformaldeído 4% e processadas convencionalmente para microscopia de luz, coradas em H.E, P.A.S., alcian blue, tricrômico de Gomori e picrossirius red. Testes estatísticos dos resultados morfométricos e histométricos obtiveram o nível de significância de 0,05 (5%). Morfometricamente não foram observadas diferenças entre os subgrupos do grupo III, entretanto no grupo II houve diferença significativa entre os três subgrupos no comprimento longitudinal (p = 0,030) e comprimento transversal (p = 0,025), tendo a glândula se apresentado maior no 28º dia após o tratamento. Histometricamente, os ácinos serosos e mucosos do grupo III, apresentaram-se com diâmetro menor quando comparado com o ácinos dos subgrupos tratados com gelatina bovina (grupo II). A espessura da parede do ducto estriado do grupo III foi menor que nos demais. Histologicamente, observou-se a maior quantidade de vacuolizações citoplasmáticas no grupo III, além do maior espaçamento entre os ácinos. Dessa forma, concluímos que a toxina botulínica tipo A interfere na organização estrutural das glândulas submandibulares, promovendo uma diminuição no diâmetro dos ácinos serosos e na espessura da parede dos ductos estriados, mas que macroscopicamente tal diferença não se mostrou significante.

L’objectif principal de cette étude était d’évaluer l’impact d’un régime hyperlipidique hypercalorique administré pour une période de 8 semaines chez le rat male Wistar (que nous avons utilisé comme modèle expérimental d’obésité) sur le métabolisme lipidique. Ainsi, nous avons déterminé les teneurs en CT, TG et protéines totals au niveau sérique, lipoprotéique et des organes (foie, tissu adipeux, muscle, intestin, cerveau), ainsi que les activités de la lipoprotéine lipase (LPL) au niveau des organes, et de la lipase hormonosensible (LHS) au niveau de tissu adipeux. Les résultats obtenus montrent qu’un régime riche en graisses et en calories engendre une accumulation accrue des lipides au niveau de tissu adipeux et conduit à une obésité viscérale fortement associée à la résistance à l’insuline et à une accumulation ectopique des lipides dans d’autre organes telque le foie et entrainant des désordres métaboliques pouvant évoluer vers un évènement cardiovasculaire. De plus, nos résultats montrent une perturbation des enzymes des voies de stockage (LPL) et de mobilisation des lipides (LHS). L’obésité apparaitrait ainsi comme une réponse normale de nos gènes à un environnement inadéquat.

OBJETIVO: Avaliar a hiperalgesia e as alterações histológicas induzidas pelo contato do gânglio da raiz dorsal com o núcleo pulposo. MÉTODOS: Foram utilizados 20 ratos Wistar, divididos em dois grupos experimentais. Em um dos grupos um fragmento do NP autólogo retirado da região sacrococcígea foi colocado sobre os gânglios da raiz dorsal de L5, e no outro grupo (controle) um fragmento de tecido adiposo foi colocado sobre o gânglio da raiz dorsal de L5. A hiperalgesia mecânica e térmica foi avaliada no terceiro dia, na primeira, terceira, quinta e sétima semanas de pós-operatório. O gânglio da raiz dorsal de L5 foi retirado na primeira, terceira, quinta e sétima semanas de pós-operatório para estudo histológico por meio da coloração com HE e estudo histoquímico (marcação específica para iNOS). RESULTADOS: Foi observada hiperalgesia térmica e mecânica de maior intensidade no grupo de animais em que o NP foi colocado em contato com o GRD, e nesse grupo o estudo histológico evidenciou alterações dos tecidos do gânglio da raiz dorsal, caracterizadas por processo inflamatório e degeneração axonal. As alterações histopatológicas dos tecidos do gânglio da raiz dorsal apresentaram intensidade crescente com o aumento do período de observação, apresentando correlação com a manutenção da hiperalgesia observada na avaliação comportamental. A imunohistoquímica com marcação específica para iNOS demonstrou, no grupo de animais em que o núcleo pulposo foi colocado em contato com o gânglio da raiz dorsal, aumento da expressão dessa enzima nos núcleos das células inflamatórias (células da glia) ao redor dos neurônios. CONCLUSÃO: O contato do núcleo pulposo com o gânglio da raiz dorsal de L5 induziu a hiperalgesia mecânica e térmica e provocou alterações histológicas dos componentes do gânglio da raiz dorsal. As alterações foram caracterizadas por processo inflamatório e degenerativo das estruturas do gânglio da raiz dorsal e apresentaram aumento da intensidade nos períodos mais longos de observação. OBJECTIVE: To evaluate hyperalgesia and histological changes of dorsal root ganglia induced by nucleus pulposus (NP) contact. METHODS: Twenty Wistar rats were used, divided into two experimental groups. In one of the groups, a fragment of the autologous NP was removed from the sacroccocigeal region and deposited on the L5 dorsal root ganglia. In the control group, the NP was removed from the sacrococcygeal region, L5 dorsal root ganglia were exposed and covered by a piece of adipous fat tissue. Hyperalgesia was evaluated by the von Frey electronic test and Hargreaves test, and histological changes of the dorsal root ganglia by HE staining and immunohistochemistry using iNOS. The evaluation of hyperalgesia and histological changes of the dorsal root ganglia were performed on the third postoperative day and after 1, 3, 5, and 7 weeks. RESULTS: NP induced higher intensity mechanical and thermal hyperalgesia. Dorsal root ganglia in contact with nucleus pulposus presented histological changes and the intensity of these changes were proportional to the length of time in contact. The expression of iNOS was higher in the glial cells in contact with the nucleus pulposus. CONCLUSION: The contact of nucleus pulposus with dorsal root ganglia induced histological changes and mechanical and thermal hyperalgesia. These changes were more intense after longer period of evaluation.
