Today youth employment is the biggest concern of the nation. According to the Centre for the Monitoring Indian Economy (CMIE) the unemployment rate in India has been increased by 6.9% in February'2021, as compared to January'2021 i.e., 6.53%. Now again due to the second wave of Covid, the unemployment rate is reaching at its worst and the local lockdown scenario in many states have witnessed a huge impact over the jobs, almost 70 lakhs jobs have been suffered. This also indicates that situation may get even more worse if the lockdown will continue to happen and things don't get under control soon. As per the article published in "The India Express" May 11' 2021, the employability of engineering graduates is registered 47.38% only, which is highest among all other domains, on the one hand where country is facing a continuous increasing challenge of employment, at the same time there is a new wave of online courses have arrived apart from the regular degree/diploma programs. This paper tries to analyse the market potential for these online courses and tries to investigate whether the candidate gets the real benefit after completing these online courses which claims to be specialised in certain area or domain in terms of providing employment. India is recognized, all over the world, as a country which presents great diversity in terms of geography, history, culture and population demography to underline a few. This diversity makes it extremely difficult to suitably categorize the country on economic, political, religious or demographical grounds. Postindependence growth has led to overall growth of the country as a whole but it has also divided it into two distinct segments thus providing a suitable basis of categorization in the form of rural India and urban India. As per the census 2001 only 31.16% population of the country lives in urban segment while the rest are still residing in the inherently characteristic rural India. The situation however has changed significantly since independence when around 82% of the total population of the country was dependent for their livelihood on rural sector. The rural segment is distinct in respect of various features such as purchasing power, economic and infrastructural development, social system and many more. These distinctions relate directly to the kind of divergent demand patterns that the rural sector has in various product segments especially when it comes to financial services. The sector presents a real challenge given its technological backwardness and mass illiteracy as people are still caught in traditional financial systems that are both exploitative and futile. Financial inclusion is the biggest problem in front of the financial system today in rural India and infrastructural bottlenecks are worsening it even further with each passing day. This paper is an attempt to comprehend the significance of Financial Inclusion in the context of a developing country like India wherein a large proportion of population is deprived of the financial services which are very much essential for the overall growth of a country. The paper tries to address the issues relating to access to finance by the poor and vulnerable groups as a prerequisite for poverty reduction, social cohesion and promoting prosperity - reflecting and promoting the true growth story of economically strong India. Background and Objective: Concurrent usage of proton pump inhibitors and their effect on survival and medication termination has been found in individuals receiving protein kinase inhibitor chemotherapy. To investigate the drug-drug interaction mechanism between CDK inhibitors and proton pump inhibitors, the in-silico docking approach was designed by applying computer simulation modules to predict the binding and inhibitory potential., Methods: The interaction potential of proton pump inhibitors and CDK inhibitors was predicted utilising molecular docking techniques that employed Schrödinger algorithms to capture the dynamics of the CYP450 enzyme-inhibitor interaction between proton pump inhibitors and CDK inhibitors. Additionally, the human liver microsomes assay was used to determine the in vitro half-maximal inhibitory concentration (IC 50 ) of proton pump inhibitors and the inactivation of CDK inhibitors via CYP3A4., Results: Proton pump inhibitors alter the conformation of the CYP3A4 and CYP2C19 enzymes and interact with the heme prosthetic group, as determined by docking studies. It may result in the suppression of CDK inhibitors' metabolism via competitive inhibition at the binding site of an enzyme. Omeprazole and rabeprazole both significantly block midazolam's 1'-hydroxylation by CYP3A4 in vitro, with IC 50 values of 9.86μM and 9.71μM, respectively. When omeprazole and rabeprazole are co-incubated in human liver microsomes at a 30μM concentration equivalent to the C max of omeprazole and rabeprazole, rabeprazole significantly prolongs the metabolic clearance of palbociclib, whereas omeprazole affects the ribociclib CYP3A4-mediated metabolism., Conclusion: Using dynamic models, we determined that proton pump inhibitors such as rabeprazole and omeprazole indeed have the potential to cause clinically significant drug-drug interactions with CDK inhibitors in the treatment of estrogen receptor (ER) positive and HER2-positive breast cancer.
