2. Evaluation of EUCAST rapid antimicrobial susceptibility test directly from positive blood culture for Pseudomonas aeruginosa.

In this study, we evaluated the performance of the EUCAST RAST method on a collection of 154 clinical strains of P. aeruginosa, including strains resistant to ceftazidime and carbapenems. While the test is convenient for routine laboratories, we observed significant rates of VME (ranging from 0.0 to 15.0%) and ME (ranging from 1.3 to 16.3%) after 6 h, particularly for key antibiotics such as ceftazidime, piperacillin/tazobactam, and meropenem. Extending the incubation time to 8 h may improve results (CA ranging from 87.2 to 99%), but caution is required in interpretation due to persistence of VME (ranging from 0.0 to 15.6%) and ME (ranging from 0.0 to 11.7%). 3. Assessing the Effect of Short-Term Interval Training on Acceleration Ability and Anaerobic Power of Novice Sprinters.

- Abstract
Background. Acceleration ability and anaerobic power are critical determinants of sprinting performance, particularly for novice sprinters. Objectives. The purpose of the study was to examine the effects of a four-week interval training program on acceleration ability and anaerobic power among novice sprinters. Materials and methods. A within-subjects quasi-experimental design was used to fulfil the study’s objectives. A total of fifteen participants (age: 20.5 ± 1.4 years) underwent baseline assessments of acceleration ability (50 m dash) and anaerobic power (Running-Based Anaerobic Sprint Test). The adopted progressive interval training protocol consisted of four weekly sessions conducted over a four-week period, focusing on striding, high knees, single-leg hops, curve sprints and double-leg jumps to enhance acceleration ability and anaerobic power. Results. Following the intervention, significant improvements were observed in acceleration ability over a distance of 10 m (pre: 2.14 ± 0.16 s, post: 2.02 ± 0.14 s, p < 0.05, Cohen’s d = 1.02) and 20 m segments (pre: 3.47 ± 0.23 s, post: 3.32 ± 0.20 s, p < 0.05, Cohen’s d = 0.78), indicating moderate to large effect sizes. However, no considerable enhancements were found in anaerobic power parameters (average power, maximum power, minimum power, fatigue index, relative peak power, and anaerobic capacity) at the post-intervention stage (p > 0.05). Conclusions. The study highlights the effectiveness of the interval training protocol in enhancing initial acceleration among novice sprinters, although it suggests a limited impact on anaerobic power within the study period. Further research should investigate the longer-term effects and tailored training approaches to optimize comprehensive sprint performance strategies. [ABSTRACT FROM AUTHOR]

V primerih, ko okolje za plod ni ustrezno, kot npr. kadar se mati neustrezno prehranjuje ali ima pridružene bolezni, lahko pride pri plodu do presnovnih sprememb, ki imajo posledice za vse življenje. Meritve sestave telesa so pomembne za zagotavljanje ustreznih tekočinskih in prehranskih potreb zlasti v skupinah ogroženih novorojenčkov – novorojenčkov rojenih velikih ali majhnih za gestacijsko starost, tistih s prirojeno srčno napako ali z ledvičnimi boleznimi ter nedonošenčkov. Metode merjenja sestave telesa so različne in vsaka od njih ima določene prednosti in slabosti.

The inhibition zones were read at 4, 6, 8 and 16-20 hour of incubation as per European Committee on Antimicrobial Susceptibility Testing (EUCAST) guidelines. The identification of the isolates was confirmed by Vitek-2 compact system. Results: In our study, the area of technical uncertainty (ATU) percentage was initially high at 4 hours but decreased significantly in later incubation periods. At 4 hours, none of the S. aureus isolates showed >90% categorical agreement (CA) for any antimicrobial tested. However, clindamycin achieved the highest CA (100%) at 6 hours and 90% thereafter, with no very major errors (VME) or major error (ME). Cefoxitin required 8 hours to reach >90% CA, with no VME observed at any time point, but up to 75% ME at 8 hours. At 4 hours, most antimicrobials had high (>1.5%) rates of VME among Enterobacteriales. By 6 hours, only Meropenem and Gentamicin had >90% CA, with no VME observed for other antibiotics. Conclusion: The RAST method is relatively easy to implement in clinical microbiology labs, offering cost-effectiveness, simplicity, and rapid results, especially in resource-limited settings. - Abstract
A training model that combines physicality and tactics is really needed in futsal. This study aims to analyse the effect of rotation play training with HIIT Protocol on the anaerobic abilities of amateur futsal players. This research uses an experimental method with a two-group pre-test and post-test design. The sample in this study consisted of 30 amateur futsal players with an age of 19.23 ± 0.57 years, height of 165.60 ± 5.63 cm, body mass of 59.17 ± 5.24 kg, and BMI of 21.60 ± 1.93. Samples were divided randomly into an experimental group (EG) and a control group (CG). Data analysis used a Paired Sample T-test and an Independent T-test with a significance level of .05 and used Cohen's ES to measure the magnitude of differences between groups. This study used the running anaerobic sprint test (RAST). The variables in RAST that will be analysed are Peak Power (W); Mean Power (W); Peak Power per Weight (W.kg-1); Mean Power per Weight (W.kg-1); Fatigue Index (%); Total effort time (s). The paired sample t-test resulted in a significant difference in EG with significance values of Peak Power p = .010, Mean Power p = .007, Peak Power per Weight p = .011, Mean Power per Weight p = .008, Fatigue Index p = .045, and total effort time p = .011, while in CG all variables do not have significant differences. The Independent T-test found that there was a significant difference between EG and CG with a significance value of p < .05. This research concludes that the rotation play training model with HIIT Protocol has a positive effect on the anaerobic endurance of amateur futsal players. [ABSTRACT FROM AUTHOR]

Breath-hold diving is explained as an activity that requires enduring muscle asphyxia and acidosis, high anaerobic capacity, and the tactic of the dive. Therefore, this study aimed to construct and validate tests that will mimic anaerobic processes in the specific media of freedivers. The sample of participants included 34 Croatian freedivers (average age: 26.85 ± 4.0 years, competitive age: 3.82 ± 1.92 years, their body height: 180.14 ± 8.93 cm, and their body mass: 76.82 ± 12.41 kg). The sample of variables consists of anthropometric indices, competitive efficiency (maximal length of a dive (DYN)), and specific anaerobic capacities (100 m and 2 min tests). Newly developed tests included the swimming anaerobic sprint test (SAST) and diving anaerobic sprint test (DAST). DAST and SAST variables included the total time of the test (DAST/SAST) and the fastest interval (DAST
max /SASTmax ). The results showed good reliability of the tests with high Cronbach alpha coefficients (DAST: 0.98, DASTmax : 0.97, SAST: 0.99, SASTmax : 0.91). Furthermore, pragmatic validity shows a high correlation among all variables and DAST (DYN: −0.70, 100 m: 0.66, 2 min: −0.68). High relation is also found between 100 m (0.96), 2 min (−0.94), and a moderate result for DYN (−0.43) and the SAST test. A factor analysis extracted one significant factor. The factor analysis involved DAST, SAST, DYN, 100 m, and 2 min tests regarding factor 1. After the examination of all variables, the total time of the DAST test showed the best predictive values for the performance of divers. Energijsko neravnovesje privede do motenj v delovanju več organskih sistemov. Pri otrocih in mladostnikih so posledice lahko dolgoročne in se kažejo kot neugoden vpliv na rast, pubertetni razvoj in zdravje kosti. Zgodnji prepoznava in obravnava sta tako v tej starostni skupini ključni. Zaradi pomanjkljive ozaveščenosti med mladimi športniki, njihovimi starši, trenerji in zdravstvenim osebjem ter nespecifične klinične slike bolezen pogosto spregledamo.

17. Influence of Feed Restriction and Zinc Oxide Nanoparticles Supplementation on Growth Performance, Blood Biochemistry, Intestinal Morphology and Cecal Fermentation Parameters of Growing Rabbits.

The present study investigated the response of growing rabbits in terms of growth performance, serum biochemical, intestinal morphology, and caecal fermentation parameters to feed restriction and zinc oxide nanoparticles (ZnO-NPs) supplementation. A total of 60 New Zealand male rabbits were randomly distributed into 6 groups: AL-0 (fed ad libitum + fresh water as control); AL-15 and AL-30 (ad libitum + water supplemented with ZnO-NPs in water, 15 and 30 mg/L, respectively); and R-0, R-15 and R-30 were the same as the first 3 groups but with restricted feeding regime. Rabbits fed ad libitum and supplemented with ZnO-NPs (15 mg/L) showed the highest body weight with no significant difference from AL- fed groups or R-0. Feed conversion ratio (FCR) showed no difference among the different experimental groups (P > 0.05). ZnONPs supplementation reduced the serum lipid profile parameters, catalase enzyme in R-30, superoxide dismutase in AL-15 and AL-30 while increased serum malondialdehyde (MDA) in both ad libitum and restricted rabbits. ZnO-NPs administration resulted in lower caecal ammonia in AL-30 compared to its control (AL-0) as well as the content of individual volatile fatty acids (VFAs) (acetate, butyrate and propionate) (P < 0.05). Ileum morphological parameters (mucosal length, villi length, and goblet cell number) were modified in response to the feed restriction and ZnO-NPs addition. In conclusion, feed restriction program applied in this experiment altered rabbit growth performance (final body weight and weight gain with no differences in FCR), improved ileum morphology while had no significant effect on caecal fermentation (VFAs profile) or microbiological parameters. ZnO-NPs supplementation in both levels (15 and 30 mg/L) differently modulated serum lipid profile, antioxidant enzymes and MDA, VFAs profile in cecum and ileal morphology with no differences in rabbit growth performance. Background & Aims: Neural activation is associated with increasing energy requirements, and increasing peripheral Lactate (LA) concentration in response to severe exercise, enhances LA supply as an energy substrate to meet acute neurological needs. The acute speed and football exercises may affect cognitive function by increasing LA concentration and nervous activation. Increasing the neurocognitive performance of growing adolescent boy players an important role in their physical performance. Therefore, the aim of the present study was to investigate the effect of speed and plyometric training activity on the concentrations of nerve proteins in blood circulation. Although speed interval and plyometric training in football players may lead to progressive fatigue and activate the central nervous system and subsequent cognitive disorders, it may also positively increase neurotrophines such as BDNF or IGF-1 and affect brain neutrophils and neurological function. According to this evidence, we assumed that BDNF, BNG and IGF-1 increase in response to a football training session, which may be associated with increasing neurological function of adolescents. Therefore, the purpose of the present study was to investigate the effect of an acute speed and plyometric football-based exercise on plasma concentrations of BDNF, BNG and IGF-1 neural proteins, which may play an important role in enhancing cognitive abilities. And the effect of speed and plyometric training activity on the concentrations of nerve proteins in blood circulation. Methods: The present study is a semi-experimental type with a pre-test and post-test design, which was conducted in two sessions, including an intervention and control session. After calling in Tehran's schools, 20 adolescents 14-15 years of age were selected and accessible as a sample, and randomly participated in 2 sessions of speed and plyometric exercises and control sessions. All subjects are familiar with how to perform speed exercises (RAST Test) during a familiarization period (two sessions). On the day of the main test, after 12 hours of fasting, the test was referred to the test site and the first blood sample (5 cc) was taken from the antiquiobital vein. After the standard breakfast of the subjects, we were wearing sportswear and after ten minutes of general warming up the speed training included six times a maximum of 35 meters with a 10-second recovery between each run. After the test was performed, the plyometric exercise was executed, including a pair jump and one -foot barriers, jump on barriers with 180 degrees rotation, zigzag jump and one foot forward, jump on obstacles with both legs and legs Single-tofoot jumping on the barriers of single -foot and throwing a medicine ball, on the sides, with 12 repetitions. The active rest between turn was considered two to three minutes (29). The subject's heart rate was performed during the controlled exercise (Polar heart rate monitor) and the exercise in the range of 80-90 % of the heart rate. The second blood sample was taken immediately after the training session was completed. All sampling steps for each subject were performed under the same conditions and the BDNF and NGF levels were measured through the USA-made Kit. The IGF-I levels were measured through the ELISA methods and IGF-I Kit. Cognitive neurological function was also measured through the Stroop test (ST) before and after the training session. In this study, the Polish paper version of ST was used. ST consisted of 71 words written with colored ink. The task of the participants was to name the font color regardless of the written word. For statistical analysis, the research data are first described using the mean and standard deviation, then the changes before and after all the variables of the two sessions were calculated and analyzed using the dependent stroop test. A significant level was considered to be P≤0.05 and SPSS 16 was used to perform statistical calculations. Results: The mean and standard deviation of the somatic specifications of the research subjects was as follow: Weight (59.7±3.2), Body mass index (21.4±1.3), Fat percentage (21.7±2.8), and Fat free mase (41.10±4.1) The results of the present study showed that BDNF data changes were significantly different in training and control session (t19=12/89, p<0/001). In the exercise session, the BDNF rate increased by approximately 2.5 times compared with control session. NGF data also showed significant differences in both sessions before and after acute speed and Plyometric training (t19=4.89, p<0/001). Regarding the plasma levels of IGF-1 also the results showed a quite significant difference in comparison of two sessions (t19 =2.89, p=0.019). The time of implementation of the stroop test at two sessions of training and control in the preceding and later times also showed significant differences that in the exercise session the test was significantly reduced than the control session (t19 =3.89, p=0/009). The test time in the exercise session decreased from 59.3s to 53.1s. There was no significant change in the control session at the time of stroop performance (58.6 s and 59.3 s). These findings show that acute speeds and plyometric exercise are useful for improving post exercise cognitive performance in adolescent. Skeletal muscle contraction during high-intensity exercise increases BDNF levels in the brain, and the speed and plyeometric exercise protocol is a more effective and preferred intervention to raise BDNF levels than traditional exercise mode, with moderate intensity. Acute speed and plyometric exercises increase human cognitive function. Improvement of cognitive function may be the synthesis or release of neurological proteins that are adjusted by high blood LA after exercise. Conclusion: In summary, the current findings suggest that acute plyometric and speed training enhances human cognitive performance. The improvement in cognitive performance may be from the synthesis or release of neuroprotective proteins modulated by high blood LA concentrations after exercise. [ABSTRACT FROM AUTHOR]

Se determinó el perfil de sensibilización a aeroalérgenos más comunes en la población de Portoviejo, durante el periodo noviembre 2017 - noviembre 2018. Se llevó a cabo un estudio observacional, descriptivo, de carácter retrospectivo y corte transversal, cuyos resultados se obtuvieron de la base de datos del Laboratorio Gamma de la ciudad de Portoviejo. La muestra poblacional estuvo conformada por 112 pacientes que se realizaron el test de RAST, RIDA qLine® Allergy de alérgenos inhalantes. Los resultados obtenidos muestran que el 63,4 % presentó algún tipo de sensibilización. La mayoría de estos casos se produjeron durante el verano (96 %), en total fueron 19 aeroalérgenos que produjeron dicha sensibilización, siendo los de mayor frecuencia, Dermatophagoides pteranyssinus y Dermatophagoides farinae, seguido por polen, donde se destacaron el centeno, roble y mezclas de hierbas, con animales y hongos. Se demostró que en adultos y tercera edad existe una mayor frecuencia de sensibilización al polen de centeno comparados con los ácaros domésticos, mientras que en rango de edad infantil a adulto joven se encontró un predominio de sensibilización a ácaros de la familia Dermatophagoides.

Se estudiaron 30 pacientes con reacciones alérgicas a J3 - Lactámicos claramente demostrados por las manifestaciones clínicas. El objetivo de este trabajo fue determinar la importancia del núcleo (3 -Lactúmico y las cadenas laterales en la inducción de IgE específica para BPO, Ax y Anip por medio de las pruebas cutáneas y el RA ST para estos determinantes. Este grupo se dividió previamente por historia clínica en: 1 - Acelerados (n: 19); 2 - Inmediatos (n:11). Las pruebas del Prick se realizaron con BPO-PL, Ax-PL, Amp-PL, MDM de BR MDM de Ax y MDM de Amp. Presentando en el grupo acelerado: 2 (-1-) a 13P0,4(+) Ax/Amp, 13(+) a todos los reactivos (BPO- Ax - Amp - MDM BP/Ax/Amp). Los pacientes con reacción inmediata: ¡Ocasos (+) a MDM -BPy l(-1-) MDM -Amp. Estos pacientes fueron estudiados con la técnica del RAST para BPO - PL, Ax-PL, Amp PL y PL. Se consideró como línea de corte positiva al nivel ~ a 1524 cpm, tomado tic la curva estandar de Pharniacia Los pacientes con reacción acelerada presentaron en 13/19 casos RAST (+) a BPO- PL, 1/19 (+) BPO- Ax/PL, 3 /19(4) Ax-PL. 1/19 (4-) Am/PL y 1 negativo para todos los reactivos estudiados. Los pacientes con reación inmediata (n: II) en todos los casos lueron negativos para todos los reactivos estudiados. El grupo control (1):20) presentó en 1/20 casos positividad por Prick a Ax- PL y 19 casos negatividad total a todos los reactivos. Los RAST a todos los reactivos fueron negativos en todos los individuos estudiados. Estos resultados indican que la bencilpenicilina (BPO) es el más importante determinante y las cadenas laterales (Ax y Amp) son otros determinantes antigéntcos en -los (3-Lactúmicos, siendo cada uno tIc ellos inductores de un tipo de IgL específica y que raramente aparece una respuesta ¡gE a dos determinantes diferentes en un mismo sujeto. Las pruebas cutáneas son el método de elección para el estudio de alergia a penicilina ya que con el método in vitro se cubre un espectro menor (le reactivos que los que producen reacción acelerada (MDM).

Antimicrobial treatment of patients with bloodstream infections (BSI) is time-sensitive. In an era of increasing antimicrobial resistance, rapid detection and identification of bacteria with antimicrobial susceptibility are critical for targeted therapy early in the disease course. This study describes the performance of a rapid method for identifying and testing antimicrobial susceptibility of Gram-negative bacteria performed directly from blood culture bottles in a routine microbiology laboratory. A total of 284, 120, and 24 samples were analyzed by rapid identification (Rid), rapid susceptibility testing (RAST), and rapid broth microdilution for polymyxin B (rMIC), respectively, and compared with standard methods. Our protocol was able to identify 93% of isolates at the species level using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). We obtained 100% agreement for RAST compared to the standard method and 96% agreement for rMIC. Our protocol has proven to be an excellent tool for rapid identification of Gram-negative bacilli causing BSIs. It can also be used in microbiology laboratory routine along with RAST and faster polymyxin microdilution, especially for carbapenemase-producing bacteria, allowing for rapid, simple, accurate, and cost-effective diagnosis.

A training model that combines physicality and tactics is really needed in futsal. This study aims to analyse the effect of rotation play training with HIIT Protocol on the anaerobic abilities of amateur futsal players. This research uses an experimental method with a two-group pre-test and post-test design. The sample in this study consisted of 30 amateur futsal players with an age of 19.23 ± 0.57 years, height of 165.60 ± 5.63 cm, body mass of 59.17 ± 5.24 kg, and BMI of 21.60 ± 1.93. Samples were divided randomly into an experimental group (EG) and a control group (CG). Data analysis used a Paired Sample T-test and an Independent T-test with a significance level of .05 and used Cohen's ES to measure the magnitude of differences between groups. This study used the running anaerobic sprint test (RAST). The variables in RAST that will be analysed are Peak Power (W); Mean Power (W); Peak Power per Weight (W.kg-1); Mean Power per Weight (W.kg-1); Fatigue Index (%); Total effort time (s). The paired sample t-test resulted in a significant difference in EG with significance values of Peak Power p = .010, Mean Power p = .007, Peak Power per Weight p = .011, Mean Power per Weight p = .008, Fatigue Index p = .045, and total effort time p = .011, while in CG all variables do not have significant differences. The Independent T-test found that there was a significant difference between EG and CG with a significance value of p < .05. This research concludes that the rotation play training model with HIIT Protocol has a positive effect on the anaerobic endurance of amateur futsal players.

31. Snabb resistensbestämning på urinprov med hjälp av flödescytometern UF-5000 och validering av dess sköljprogram

Urinvägsinfektion (UVI) är en av de vanligaste bakteriella infektionerna bland människor. Urinodling är standardmetoden för att identifiera bakterier och kvantifiera mängden av dem i urinen. Flödescytometri ger möjligheten att snabbare analysera urinprover för att utesluta urinvägsinfektion och därmed minska den onödiga antibiotikabehandlingen. Den omfattande användningen av antibiotika utgör en stor orsak till antimikrobiell resistens (AMR) och har resulterat i en stor efterfrågan av metoder för att genomföra snabbare tester för antibiotikaresistens. Målet med projektet blev därför att utvärdera om flödescytometern UF-5000 kan användas för att snabbare diagnostisera urinvägsinfektioner. Urinprover kördes tre gånger genom UF-5000 med olika sköljprogram tillsammans med tre olika efterföljande sterila koksaltlösningsprover och utifrån en formel beräknades carryover rate (överföringsgrad). Sterila koksaltlösningaproverna odlades sedan för att se hur stor korskontaminationen blev. Dessutom genomfördes standard resistensbestämning (AST) och direkt resistensbestämning (RAST) på både kontrollstammar och patientprover. Carryover rate av bakterier blev mindre när sköljprogram med fler antal sköljningar användes. Sköljprogrammet [0-0-1-3-4] översteg aldrig gränserna för tillåten carryover rate. Resultat gick att avläsa för de flesta proverna efter RAST på sex timmar men inte på fyra timmar. Det var skillnader mellan resultaten erhållna efter AST och RAST. Vi kom fram till att det mest tidseffektiva sköljprogrammet som inte resulterade i någon betydlig påverkan på efterföljande prover var sköljprogrammet [0-0-1-3-4] i UF-5000. Vi kom även fram till att resistensbestämning på fyra timmar var svår att göra men att efter sex timmar skulle man kunna utesluta vilka antibiotika man inte ska använda för behandling., Urinary tract infection (UTI) is one of the most common bacterial infections among humans. Urine culture is the standard method for identifying and quantifying the amount of bacteria in the urine. Flow cytometry offers the possibility to analyze urine samples more quickly to rule out urinary tract infection, thereby reducing unnecessary antibiotic treatment. The extensive use of antibiotics is a major cause of antimicrobial resistance (AMR) and has resulted in a high demand for methods to perform faster tests for antibiotic resistance. The aim of the project was therefore to evaluate whether the flow cytometer UF-5000 can be used to diagnose urinary tract infections more quickly. Urine samples were run three times through the UF-5000 with different rinse programs, along with three different subsequent sterile saline samples, and the carryover rate was calculated using a formula. The sterile saline samples were then cultured to determine the extent of cross-contamination. Additionally, antimicrobial susceptibility testing (AST) and rapid antimicrobial susceptibility testing (RAST) were performed on both control strains and patient samples. The carryover rate of bacteria was lower when rinse programs with a higher number of rinses were used. The rinse program [0-0-1-3-4] never exceeded the limits for the allowed carryover rate. Results could be read for most samples after RAST at six hours but not at four hours. There were differences between the results obtained after AST and RAST. We concluded that the most time-efficient rinse program that did not result in any significant impact on subsequent samples was the rinse program [0-0-1-3-4] in the UF-5000. We also found that resistance determination at four hours was difficult to perform, but after six hours, it would be possible to rule out which antibiotics should not be used for treatment.

Bacterial bloodstream infections represent a severe and potentially life-threatening condition. Diagnostic tools, providing rapid species identification and antimicrobial susceptibility testing have a great impact on disease progression and treatment optimization. Typically, antimicrobial susceptibility testing involves a time-consuming process, and faster methods are likely to contribute in clinical settings. This study aimed to assess the reliability of the European Committee on Antimicrobial Susceptibility Testing (EUCAST) rapid antimicrobial susceptibility testing (RAST) with a 16-20-hour incubation period for implementation in a clinical microbiology laboratory. This method was conducted alongside the standardized disk diffusion method from EUCAST, encompassing all bacterial species compatible with 16-20-hour RAST. The method was executed 80 times, and over 450 readings were performed. It confirms EUCAST's method validation, affirming 16-20-hour RAST as a swift and precise method for blood bacterial resistance determination. All inhibition zones were readable, with 89% of antibiotics tested falling within 1 mm of the set target value. Notable deficiencies were observed in antibiotic-species combinations, such as trimethoprim/sulfamethoxazole and gentamicin against E. coli, and erythromycin against S. pneumoniae. Accurate interpretation of inhibition zones demands precise reading practices as some zones were ambiguous. The study also underscores the importance of overnight cultures in antimicrobial susceptibility testing via standardized disk diffusion, accentuating 16-20-hour RAST's pivotal role in potentially accelerating resistance determination by up to a day. Given its commendable performance, 16-20-hour RAST is ready to be incorporated into the standard procedures at clinical microbiology laboratories, as demonstrated by its successful adoption at Unilabs, Skövde.

33. Fritidslärarens roll på rasten- : En kvalitativ studie om rasten ur lärarnas perspektiv

- Abstract
Bakgrund: Fritidslärare har oftast ett komplext uppdrag genom att vara tilldelade olika ansvarsområden under en skoldag. Ett av dessa områden kan vara att ha ansvaret under rasten. Historiskt sett har rastens syfte och innehåll utvecklats en hel del där det tidigare inte skett något lärande. Idag visar tidigare forskning att rasten är en viktig del i elevernas hela skoldag för deras utveckling och lärande men är även en plats där det sker mycket mobbning och kränkningar. Därför är det viktigt att skolorna utformar en meningsfull och trygg rast för alla elever genom att ta vara på lärarnas kompetens. Det finns även fördelar med att använda fritidslärarnas kompetenser under rasterna för att erbjuda eleverna en meningsfull fritid. Däremot kan detta arbetssätt skilja sig åt i verkligheten mellan skolor där både syfte och utformning kan se olika ut på grund av att alla skolor har olika förutsättningar. Syfte: Syftet med följande arbete är att undersöka de deltagande fritidslärarnas uppfattningar om deras roll på rasten. Utifrån syftet har vi även formulerat fyra frågeställningar: -Hur ser de deltagande fritidslärarna på sitt uppdrag kring rasten? -Hur ser de deltagande fritidslärarnas samsyn ut angående rasten och arbetssättet? -Hur använder de deltagande fritidslärarna sina pedagogiska idéer under rasten? -Vad ser de deltagande fritidslärarna för möjligheter och utmaningar kring arbetet med rasten? Metod: För att besvara studiens syfte och frågeställningar användes kvalitativa metoder där semistrukturerade observationer och intervjuer genomfördes. Totalt observerades och intervjuades sex utbildade fritidslärare på två lokala skolor som oftast är ute och har ansvaret under rasten. Resultat: Genom analysen av datainsamlingen framkommer det att de deltagande fritidslärarna ser rasten som viktig för alla elever under skoldagen samt att fritidslärarnas roll är betydelsefull för eleverna, både för deras sociala, kognitiva och fysiska utveckling. Rasten är den stund på dagen då elevernas sociala förmågor ständigt prövas och de förväntas interagera med andra på skolgården. Länge har rasten varit den enda delen av skoldagen som inte varit planerad och uppstyrd. Nu har det blivit alltmer vanligt att skolor har organiserad rastverksamhet. Studiens syfte var att undersöka lärare i fritidshems uppfattningar om organiserad rastverksamhet utifrån sociala relationer och förebyggande konfliktarbete. I studien får vi ta del av hur sju pedagoger beskriver arbetet med organiserad rastverksamhet på rasterna. Det empiriska materialet har samlats in genom semistrukturerade intervjuer och har analyserats med hjälp av relationellt pedagogiskt perspektiv samt Cohens konfliktpyramid. I resultatet visades det att organiserad rastverksamhet kan vara ett pedagogiskt verktyg där pedagoger kan vägleda elever i det sociala samspelet med andra under rasten. Vidare framkom vikten av att pedagogerna bygger upp relationer till eleverna för att sedan kunna stötta dem i deras relationsarbete. Samtliga pedagoger var övertygade om att rastverksamhet på olika sätt har positiva effekter på det förebyggande konfliktarbetet. Objectives: To evaluate the possible impact of RAST on optimal antimicrobial therapy via de-escalation or escalation, and to determine the reduction in antibiotic susceptibility reporting time with RAST., Methods: In this single-center, prospective descriptive study, RAST was performed on clinical blood cultures containing E. coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Acinetobacter baumannii isolates. Very major error, major error, and categorical agreements with VITEK 2 were analyzed., Results: One hundred and three isolates were included in the study, out of which 29.1 % were carbapenem-resistant and 36.9 % were multidrug-resistant according to VITEK 2. Categorical agreement of the RAST method with standard antimicrobial susceptibility test (AST) was > 90 % at 6 h, except for piperacillin/tazobactam. Antibiotic revision could be carried out in 79.6 % of the patients either by de-escalation (61.2 %) or escalation (18.4 %) for optimal therapy based on the RAST 6 h result. RAST could provide carbapenem-sparing therapy in 24 % of patients. Reduction in antibiotic susceptibility reporting time was 41.5 h (38.8 to 63.2, median (IQR))., Conclusions: RAST can provide early antibiotic revision in a majority of patients with significantly reduced antibiotic susceptibility reporting time. Six hours is the shortest optimal time for antibiotic revision with RAST. 36. Validation of EUCAST rapid antimicrobial susceptibility testing directly from positive blood cultures in a non-automated lab setting.

Introduction: To speed up antimicrobial susceptibility testing (AST), the European Committee on Antimicrobial Susceptibility Testing (EUCAST) proposed rapid AST (RAST), a disk diffusion method to be read after 4, 6 and 8 hours of incubation. We investigated the feasibility of implementation of RAST in a non-automated lab setting., Materials & Methods: To this end, reference strains as well as a variety of clinical and resistant strains were used to spike sterile hemocultures (BioMérieux BACT/ALERT 3D® and Becton Dickinson BACTEC FX® systems), followed by RAST in comparison to classical long-incubation AST., Results & Conclusion: Our results with reference strains show that reading RAST after 4 hours is frequently too soon to obtain clinical results, and that Streptococcus pneumoniae reference strain did yield readable inhibition zones in RAST when harvested from BioMérieux BACT/ALERT 3D® bottle cultures. In a wider panel of strains, Gram positives RAST results were very similar to standard AST, while with Gram negative species errors were more frequently observed, limiting clinical implementation.

The purpose of the research is to reveal in the diachronic aspect some principles of development of the mugham genre tasnif. The research aims also to study some features of the musical language of tasnifs and investigate the role of singers and performers of folk instruments in the development of the mugham of Azerbaijan. The research methodology is aimed at using the methods of historical musicology and the application of certain methodological principles of ethnomusicology in the study of the musical language of Tasnif. The scientific novelty of the research. The article presents some new facts related to the creation and implementation of tariffs. Conclusions. Tasnifs are vocal-instrumental musical works with a certain point basis, melodic and formal structure. Outstanding Azerbaijani singers played an important role in the development of this genre. In the Soviet period, tasnifs created by singers were presented under the name of folk songs.

40. Comparison of Anaerobic Performance with Laboratory and Field Tests in Trained Children.

Anaerobic performance is considered an indicator of performance for short-term muscle activities of high intensity. It is important to determine whether different anaerobic field tests performed to measure anaerobic performance can be used as an alternative to WAnT performed in the laboratory. The study aimed to compare the anaerobic performance with laboratory and field tests in trained children. One-hundred four athletes between the ages of 10 and 16, minimum age of training of one year in different sports voluntarily participated in the study. Wingate Anaerobic Test (WAnT) was performed as the reference test. Besides, Runningbased Anaerobic Sprint Test (RAST) and Pediatric RAST (PRAST) were performed to determine anaerobic performance. Peak power (PP), average power (AP), minimum power (MP), fatigue index (FI) and total exercise time (TED) were determined for each test. All variables of WAnT, RAST, and PRAST were significantly different (p<0.01). According to test-retest results of all tests, ICC [95% CI] values have a high-reliability coefficient for all variables. It was found there is a high correlation significantly between WAnT and RAST for all variables (p<0.01). Besides, there were also high correlations significantly between WAnT-PRAST and RAST-PRAST excluding fatigue index (p<0.01). As a result of this study, it was determined all tests have high reliability. Considering that WAnT requires complex, expensive device and tools, trained staff and is performed in the form of cycling in the laboratory, RAST and PRAST performed with body weight in field conditions can be used to determine anaerobic performance in trained children. The objective of this study was to provide the clinic with rapid and accurate results of antimicrobial susceptibility testing for the treatment of patients with bloodstream infections. To achieve this, we applied the Clinical and Laboratory Standards Institute (CLSI) blood culture direct rapid antimicrobial susceptibility test (rAST) to assess the susceptibility of the most common Enterobacterales found in blood cultures. In this study, we utilized the CLSI blood culture direct rapid antimicrobial susceptibility test to assess the susceptibility (rAST) of the most common Enterobacterales present in blood cultures. We chose this method for its simplicity in analysis, and our aim was to predict minimum inhibitory concentrations (MICs) using the rAST. As a benchmark, we assumed that Broth Macrodilution method (BMD) results were 100% accurate. For data evaluation, we employed the terms categorical agreement (CA), very major errors (VME), and major errors (ME). Our findings demonstrate that the CLSI rAST method is reliable for rapidly determining the in vitro susceptibility of Enterobacterales to common antimicrobial drugs in bloodstream infections. We achieved a concordance rate of 90% in classification within a 10-hour timeframe. We identified a total of 112 carbapenem-resistant Enterobacterales (CRE) strains, and there was no significant difference in the detection rate of CRE at 6, 10, and 16 hours. This suggests that CRE can be identified as early as 6 hours. The CLSI rAST is a valuable tool that can be utilized in clinical practice to quickly determine the susceptibility of Enterobacterales to antimicrobial drugs within 10 hours. The aim of this study was to replace fish meal (FM) with soy protein concentrate (SPC) in carp diets. During a carp feeding trial, the replacement of FM with SPC in four diets 100% replacement (SPC100); 50% replacement (SPC50); 25% replacement (SPC25), and; 0% replacement (SPC0) had no negative effects on the feed conversion ratio or the feed efficiency ratio of the live carp. However, significant differences in the specific growth rate and condition factor were found. The data obtained showed the four different carp diets led to differences in the chemical composition of the resultant carp meat. Between dietary treatments, significant differences were seen in the level of saturated fatty acids (FAs) in carp meat (P < 0.05). The levels of monounsaturated FAs and polyunsaturated FAs in carp meat differed significantly between dietary treatments (P = 0.05). Pearson's correlation coefficient indicates a statistically significant correlation between the FA composition of the diet and the resultant carp meat. It is possible to replace up to 25% of the FM with SPC. Diets SPC25 and SPC0 had no significant FA composition and had amino acid balances that, more than the other diets studied, closely met the requirements of the carp. [ABSTRACT FROM AUTHOR]

Passion fruit is valued for its intense flavoured juice used in juice mixes. Though cultivation is prominent in many tropical regions, it is a new crop in southwest Nigeria regardless of the favourable ecology. New crop establishment depends on meeting the nutrient requirements, among other growth resources. Two experiments were conducted at Abeokuta to evaluate the optimum rate(s) of Sunshine Organic Fertilizer® (SOF) required for the growth of the seedlings. The SOF was applied at 0, 0.5, 1.0, 1.5, 2.0, and 2.5 t/ha in experiments arranged in Completely Randomised Design (CRD) replicated thrice. The application of SOF improved the growth of seedlings, but a rate above 2.0 t/ha resulted in depressed growth. In experiment 1, the application of 0.5 t/ha SOF gave the highest values of 152.7 cm, 19 mm and 19.0 for vine length, vine diameter and the number of leaves. In experiment 2, the number of leaves was the highest at 1.0 t/ha though similar to 1.5 and 2.0 t/ha. The application of SOF had a significant influence on the seedling leaf area (LA) in the experiments. The largest LA occurred in seedlings treated with SOF at 2.0 and 2.5 t/ha. Dry weight and foliar nutrient uptakes in both experiments were significantly influenced by SOF application. The application of SOF at 0.5 t/ha in experiment 1 resulted in higher N uptake, but the reverse was the trend in experiment 2, with higher nutrient uptakes at 1.5-2.5 t/ha SOF. Conclusively, an optimal rate of 2.0 t/ha had positive effects on seedling response. [ABSTRACT FROM AUTHOR]
