Abstract

Storm water detention ponds are used to manage the discharge of rainfall runoff from urban areas to nearby streams. Their purpose is to reduce the hydraulic impact and sediment loads of the receiving waters. Detention ponds are currently designed based on static controls: the output flow of a pond is capped at a fixed value. This is not optimal with respect to the current infrastructure capacity and for some detention ponds it might even violate current regulations set by the European Water Framework Directive. We apply formal methods to synthesize (i.e., derive automatically) a safe and optimal active controller. We model the storm water detention pond, including the urban catchment area and the rain forecasts, as a hybrid Markov decision process. Subsequently, we use the tool Uppaal Stratego to synthesize a control strategy minimizing the cost related to pollution (optimality) while guaranteeing no emergency overflow of the detention pond (safety). Simulation results for an existing pond show that Uppaal Stratego can learn optimal strategies that prevent emergency overflows, where the current static control is not always able to prevent it. 24. The semantics of צדד lexemes in the Hebrew psalter

Abstract

Psalms containing lexemes derived from the Hebrew root צרר1 (to bind, be in distress) are studied in order to reveal a previously-unnoticed generic subgroup in the Psalter. Chapter 1 discusses methodology, particularly with respect to the structural and cognitive linguistic principles used throughout the study. Chapter 2 determines which psalms will be the subject of the study. Since the Hebrew Psalter contains both צרר1 lexemes as well as lexemes derived from the homonym צרר2 (to be hostile, be an enemy), chapter 2 investigates translational patterns in ancient versions as well as elements of Hebrew grammar and syntax in order to separate instances of these two roots. The core of the study (chapters 3 through 5) conducts a two-fold investigation of psalms that include צרר1 lexemes, with respect to these psalms’ uniqueness in the Psalter. First, the structuralist concept of co-occurrence restriction is applied, resulting in a set of syntagms that only appear in psalms that include צרר1 lexemes. Second, these syntagms are classified using cognitive semantic principles, resulting in a set of three cognitive domains ([POWERLESSNESS], [PALPABLE THREAT], and [ENTREATY]) which comprise a cognitive profile of distress in the Psalter. Chapters 6 and 7 round out the cognitive profile of distress with an analysis of two more salient domains ([EXTRA-LINGUISTIC CAUSE], and [GUILTY?] which, although being necessary components of the profile, are not unique to distress psalms. Chapter 7 studies צרר1 lexemes with respect to the overall editing of the Hebrew Psalter. Chapter 8 then explores Psalm 107 as a test case and exemplar of the above cognitive profile. The final chapter draws conclusions with respect to the generic identity of distress psalms.

Abstract

Yield improvements in cell factories can potentially be obtained by fine-tuning the regulatory mechanisms for gene candidates. In pursuit of such candidates, we performed RNA-sequencing of two α-amylase producing Bacillus strains and predict hundreds of putative novel non-coding transcribed regions. Surprisingly, we found among hundreds of non-coding and structured RNA candidates that non-coding genomic regions are proportionally undergoing the highest changes in expression during fermentation. Since these classes of RNA are also understudied, we targeted the corresponding genomic regions with CRIPSRi knockdown to test for any potential impact on the yield. From differentially expression analysis, we selected 53 non-coding candidates. Although CRISPRi knockdowns target both the sense and the antisense strand, the CRISPRi experiment cannot link causes for yield changes to the sense or antisense disruption. Nevertheless, we observed on several instances with strong changes in enzyme yield. The knockdown targeting the genomic region for a putative antisense RNA of the 3′ UTR of the skfA-skfH operon led to a 21% increase in yield. In contrast, the knockdown targeting the genomic regions of putative antisense RNAs of the cytochrome c oxidase subunit 1 (ctaD) , the sigma factor sigH , and the uncharacterized gene yhfT decreased yields by 31 to 43%. Abstract

Purpose: To assess long-term outcomes of restrictive versus standard intravenous (IV) fluid therapy in adult intensive care unit (ICU) patients with septic shock included in the European Conservative versus Liberal Approach to Fluid Therapy in Septic Shock in Intensive Care (CLASSIC) trial. Methods: We conducted the pre-planned analyses of mortality, health-related quality of life (HRQoL) using EuroQol (EQ)-5D-5L index values and EQ visual analogue scale (VAS), and cognitive function using Mini Montreal Cognitive Assessment (Mini MoCA) test at 1 year. Deceased patients were assigned numerical zero for HRQoL as a state equal to death and zero for cognitive function outcomes as worst possible score, and we used multiple imputation for missing data on HRQoL and cognitive function. Results: Among 1554 randomized patients, we obtained 1-year data on mortality in 97.9% of patients, HRQoL in 91.3%, and cognitive function in 86.3%. One-year mortality was 385/746 (51.3%) in the restrictive-fluid group versus 383/767 (49.9%) in the standard-fluid group, absolute risk difference 1.5%-points [99% confidence interval (CI) − 4.8 to 7.8]. Mean differences were 0.00 (99% CI − 0.06 to 0.05) for EQ-5D-5L index values, − 0.65 for EQ VAS (− 5.40 to 4.08), and − 0.14 for Mini MoCA (− 1.59 to 1.14) for the restrictive-fluid group versus the standard-fluid group. The results for survivors only were similar in both groups. Conclusions: Among adult ICU patients with septic shock, restrictive versus standard IV fluid therapy resulted in similar survival, HRQoL, and cognitive function at 1 year, but clinically important differences could not be ruled out.

Abstract

På linje med de øvrige lande i EU har Danmark valgt at bruge FAIR (Findable,Accessible,Interoperable,Reusable) som model for data management af offentligt finansierede forskningsdata. Derfor har Uddannelses- og Forskningsministeriet gennem DeiC fået udarbejdet en national strategi for data management baseret på FAIR principper, som blev offentliggjort i 2021. Strategien gælder for offentligt finansierede forskningsdata, forstået som resultatet af forskning finansieret af offentlige midler eller forskning udført af offentlige institutioner, hvor der er inkluderet privat finansiering. Strategien er udformet i overensstemmelse med Direktivet om Åbne Data - artikel 10 og med forslaget til den danske PSI-lov. Som en del af strategien nedsatte Uddannelses- og Forskningsministeriet en følgegruppe med den opgave at følge og koordinere implementeringen af strategien. Strategien indeholder en række principper og forslag til indsatser, der skal understøtte strategiens realisering. Følgegruppen skal særligt fokusere på den del af implementeringen, som kræver en national koordinering på tværs af interessenter (eksempelvis forskningsinstitutioner og DeiC). Følgegruppen har, som en del af sin opgaveportefølje, udarbejdet en handlingsplan for koordinering, opfølgning og prioritering af de indsatser, som kræver en særlig national tilgang. Nærværende rapport er en statusrapportering for handlingsplanen. Forslaget til handlingsplanen lægger desuden op til overvejelser mht. fordele og ulemper ved, at de forskningsfinansierende fonde stiller krav til bevillingsmodtagere om udarbejdelse af FAIR-baserede data management planer. Følgegruppen har nedsat 5 tværinstitutionelle arbejdsgrupper med det opdrag at følge hver deres indsatsområde. Følgegruppen har derudover valgt at fokusere på effektiv kommunikation, dels som redskab mellem arbejdsgruppernes medlemmer, dels i forhold til at skabe en national ramme for let tilgængelig og let forståelig information om data management baseret på FAIR Principperne for universiteterne. Det anses som en absolut nødvendighed, da implementeringen af FAIR må anses for en af de største samlede kultur- og praksisændringer forskningsverdenen har oplevet i nyere tid.
