Abstract

Diabetes is a major health concern that the next-generation of on-demand insulin releasing implants may overcome via personalized therapy. Therein, 3D-printed phenylboronic acid-containing implants with on-demand glucose-triggered drug release abilities are produced using high resolution stereolithography technology. To that end, the methacrylation of phenylboronic acid is targeted following a two-step reaction. The resulting photocurable phenylboronic acid derivative is accordingly incorporated within bioinert polyhydroxyethyl methacrylate-based hydrogels at varying loadings. The end result is a sub-centimeter scaled 3D-printed bioinert implant that can be remotely activated with 1,2-diols and 1,3-diols such as glucose for on-demand drug administration such as insulin. As a proof of concept, varying glucose concentration from hypoglycemic to hyperglycemic levels readily allow the release of pinacol, i.e., a 1,2-diol-containing model molecule, at respectively low and high rates. In addition, the results demonstrated that adjusting the geometry and size of the 3D-printed part is a simple and suitable method for tailoring the release behavior and dosage. 35. Vitrimer-like Polyampholyte Networks: Toward Elevated Creep Resistance and Fast Dynamic Exchanges

Abstract

Vitrimers represent dynamic polymer networks with unique viscoelastic behavior, combining the best attributes of thermosets and thermoplastics. In this work, we report the concept of polyampholyte physical networks that go far beyond the merits of vitrimer-like materials by combining elevated creep resistance and fast dynamic exchanges. By contrast with most vitrimeric systems requiring out-of-stoichiometric approaches or external catalysts, we simply design polyampholyte physical networks of charge-balanced ionic methacrylates via free radical copolymerization combining poly(ethylene glycol) methyl ether methacrylate (PEGMA). The high ion concentration (i.e., from 33 to 67 mol %) enabled the formation of a physical network containing ionic cross-links acting as both sacrificial and permanent bonds mitigated by the presence of neutral PEGMA. In addition to their vitrimer-like behavior demonstrated using thermomechanical and rheological tools, these physical networks show exceptionally short relaxation times and high creep resistance. Conceptually, these polyampholyte physical networks due to extensive dynamic ion exchange open the door to more circular vitrimer-like systems toward more sustainable plastic materials.

46. Balancing Acute and Chronic Occupational Risks: The Use of Nitrile Butadiene Rubber Undergloves by Firefighters to Reduce Exposure to Toxic Contaminants

Abstract

Firefighters are exposed via multi-route exposure to a multitude of chemicals (PAHs, VOCs, flame retardants, dioxins, etc.) that may cause acute and long-term health effects. The dermal absorption of contaminants is a major contributor to the overall exposure and can be reduced by wearing appropriate personal protective equipment. As leather firefighters’ gloves cannot be decontaminated regularly by wet cleaning, many Belgian firefighters wear supplementary undergloves made of nitrile butadiene rubber (NBR) to protect against the accumulation of toxicants. However, the safety of this practice has been questioned. In this commentary, the current practice and risks are outlined for the first time, assessed by an interdisciplinary working group of the Belgian Superior Health Council. As NBR sticks to the skin more at high temperatures, the contact time on removal will be prolonged, posing an additional risk for deeper burns. However, based on the physicochemical properties of NBR and the existing experience of firefighters and burn centers, it is estimated that such incidents occur relatively rarely in practice. On the other hand, the risk of repeated exposure to contaminated gloves if no undergloves are worn is unacceptable. Despite the slightly increased risk for deeper burns, it is concluded that wearing disposable NBR gloves under regular firefighters’ gloves is an appropriate and effective preventive measure against toxic contamination. Biodegradation of polyesters driven by enzymes is considered as one of the most effective way of degradation of these materials, as a way to control plastics accumulation in the environment. In this study, we present two different strategies to tune the enzymatic degradation of PBS films triggered by a lipase from Pseudomonas cepacia. Firstly, the kinetics of enzymatic degradation of PBS films was regulated by applying multilayer coats of polysaccharide alginate and chitosan (Alg/Chi) films. Secondly, self-degradable PBS films were prepared by embedding lipase-filled alginate particles. For comparison purposes, a detailed enzymatic degradation study of neat PBS films exposed to a lipase from P. cepacia in solution was made to determine the main experimental parameters influencing their degradation in solution. The results showed that an increase in enzyme concentration increased the degradation extent and rate of neat PBS films. At a fixed enzyme concentration, stirring of the solution containing the enzyme and the PBS also increased the biodegradation rate. In the case of the PBS films coated with a different number of Alg/Chi layers by spray-assisted LbL and subjected to enzymatic degradation experiments in solution, the extent of degradation was found to be dependent on the number of protective coating layers. Therefore, the Alg/Chi biobased coating constitutes an effective barrier to the diffusion of the lipase, thus proving its effectiveness in modulating the enzymatic activity as a function of coating thickness. In the case of self-degradable PBS containing lipase-embedded alginate beads (employed to protect the enzyme during high-temperature processing), only limited biodegradation was observed as the amount of encapsulated enzyme employed was too small.
