Abstract Background Stephanofilaria stilesi is a vector-borne filarioid nematode of cattle in North America that is transmitted via the hematophagous horn fly (Haematobia irritans) intermediate host. Despite being relatively common, little attention has been given to a thorough description of S. stilesi lesions and the potential integration of pathological and molecular diagnostic findings to confirm infection. Methods To characterize the cutaneous lesions caused by S. stilesi in cattle (Bos taurus taurus and Bos taurus indicus), skin of the ventral abdominal midline was collected from 22 animals during postmortem examination. Skin samples were processed for histology, transmission electron microscopy (TEM), DNA extraction, PCR, and Sanger sequencing targeting molecular markers cytochrome oxidase c subunit 1 (cox1), 12S, 18S rDNA, and 28S rDNA. Results Macroscopically, lesions ranged from 5 × 4 cm to 36 × 10 cm, consisting of one large single lesion, or two to four ovoid areas at the ventral abdominal midline, surrounding the umbilicus. Each lesion presented as ulcerative dermatitis with dry, serocellular crusts, or alopecic and lichenified areas. Histologically, eosinophilic, neutrophilic, and ulcerative dermatitis with furunculosis, folliculitis, and epidermal hyperplasia was observed. Cross sections of adult nematodes were identified in ~ 60% of the cases (n = 13) within intact follicles, sebaceous ducts, crusts, and areas of furunculosis. Stephanofilaria first-stage larvae (L1) were observed in five cases within “vitelline membranes” in the superficial dermis and crusts. Ultrastructurally, the L1 cross sections were compounded of smooth multilayered cuticle and somatic cells. The “vitelline membrane” is a tri-layered membrane where L1 are suspended in a matrix. Stephanofilaria stilesi DNA was found in 5 out of the 13 cases in which adults or L1 were histologically observed (38%) and in 1 out of the 9 cases without adults or L1 present (11%). Phylogenetic analyses suggest a closer relationship of the genus Stephanofilaria with Thelazioidea, instead of the family Filariidae (Filarioidea), in which it has been historically allocated. Conclusions Our study improved the characterization of lesions and described ultrastructural findings of S. stilesi and highlights that molecular tools should be utilized in combination with histology for improved diagnostic resolution. Abstract Post-weaning enteropathies in swine caused by pathogenic E. coli, such as post-weaning diarrhea (PWD) or edema disease (ED), remain a significant problem for the swine industry. Reduction in the use of antibiotics over concerns of antibiotic resistance and public health concerns, necessitate the evaluation of effective antibiotic alternatives to prevent significant loss of livestock and/or reductions in swine growth performance. For this purpose, an appropriate piglet model of pathogenic E. coli enteropathy is required. In this study, we attempted to induce clinical signs of post-weaning disease in a piglet model using a one-time acute or lower daily chronic dose of a pathogenic E. coli strain containing genes for both heat stable and labile toxins, as well as Shiga toxin. The induced disease state was monitored by determining fecal shedding and colonization of the challenge strain, animal growth performance, cytokine levels, fecal calprotectin, histology, fecal metabolomics, and fecal microbiome shifts. The most informative analyses were colonization and shedding of the pathogen, serum cytokines, metabolomics, and targeted metagenomics to determine dysbiosis. Histopathological changes of the gastrointestinal (GI) tract and tight junction leakage as measured by fecal calprotectin concentrations were not observed. Chronic dosing was similar to the acute regimen suggesting that a high dose of pathogen, as used in many studies, may not be necessary. The piglet disease model presented here can be used to evaluate alternative PWD treatment options.

A 3-year-old castrated male, American Pit Bull Terrier presented to Texas A&M University due to a 3-week mixed cerebellar and general proprioceptive ataxia, circling, head tilt, and dull mentation. Neurologic examination revealed signs of vestibular and mesencephalic dysfunction. Postmortem examination revealed a 1.1 × 1 × 0.8-cm, soft, dark red, well-circumscribed, left-sided mass, extending from the crus cerebri of the midbrain caudally to the pons. Microscopically, the neoplasm was composed of a spindle-shaped interstitial population of cells interspersed between a prominent capillary network, consistent with the reticular pattern of hemangioblastoma. Interstitial cells had strong, diffuse, intracytoplasmic immunolabeling for neuron-specific enolase (NSE) and were variably positive for intracytoplasmic glial fibrillary acidic protein (GFAP). Vascular endothelial cells had strong diffuse, intracytoplasmic immunolabeling for von Willebrand factor (VWF) glycoprotein. To date, only six cases of hemangioblastoma have been reported in canines, five in the spinal cord, and one in the rostral cerebrum. Our case may represent the first canine hemangioblastoma localized to the brainstem.

ABSTRACT: Adult learning, or andragogy, provides a novel way of appreciating using food analogies as an effective learning tool in veterinary pathology. Facilitation of adult learning requires that new concepts be presented in a way that draws on the learner’s experience. Because veterinary students will have had considerable experience with a plethora of food items prior to enrolling in a pathology course, food analogies can provide an easy conduit for incorporating key learning concepts regarding veterinary pathology. In this paper, many of these analogies are presented, along with the mechanisms responsible for each of the characteristic lesions, in the hopes that their usefulness in the classroom can be highlighted to create a more engaging and facilitated learning environment.

ABSTRACT: The shape is one of the key features of a lesion and a pathologist must be able to identify and interpret these forms in the context of any gross and microscopic changes. One of the principles of adult learning is to engage the learner with previously understood information. If, when presenting material that is new, a connection with something the student already has familiarity with, the learning process is accelerated. As the learners are already familiar with shapes they have encountered throughout their pre-pathology learning, these can be used to hasten the incorporation and understanding of lesions. This paper describes various shapes that are used in describing lesions in veterinary pathology.

Host genetic background is a significant driver of the variability in neurological responses to viral infection. Here, we leverage the genetically diverse Collaborative Cross (CC) mouse resource to better understand how chronic infection by Theiler's Murine Encephalomyelitis Virus (TMEV) elicits diverse clinical and morphologic changes in the central nervous system (CNS). We characterized the TMEV-induced clinical phenotype responses, and associated lesion distributions in the CNS, in six CC mouse strains over a 90 day infection period. We observed varying degrees of motor impairment in these strains, as measured by delayed righting reflex, paresis, paralysis, seizures, limb clasping, ruffling, and encephalitis phenotypes. All strains developed neuroparenchymal necrosis and mineralization in the brain, primarily localized to the hippocampal regions. Two of the six strains presented with axonal degeneration with myelin loss of the nerve roots in the lumbar spinal cord. Moreover, we statistically correlated lesion distribution with overall frequencies of clinical phenotypes and phenotype progression to better understand how and where TMEV targets the CNS, based on genetic background. Specifically, we assessed lesion distribution in relation to the clinical progression of these phenotypes from early to late TMEV disease, finding significant relationships between progression and lesion distribution. Finally, we identified quantitative trait loci associated with frequency of lesions in a particular brain region, revealing several loci of interest for future study: lysosomal trafficking regulator (Lyst) and nidogen 1 (Nid1). Together, these results indicate that the genetic background influences the type and severity of clinical phenotypes, phenotypic resilience to TMEV, and the lesion distribution across strains.

The identification of Parrot bornaviruses (PaBV) in psittacine birds with proventricular dilatation disease (PDD) has not been sufficient to explain the pathogenesis of this fatal disease, since not all infected birds develop clinical signs. Although the most accepted theory indicates that PaBV directly triggers an inflammatory response in this disease, another hypothesis suggests the disease is triggered by autoantibodies targeting neuronal gangliosides, and PDD might therefore resemble Guillain-Barré Syndrome (GBS) in its pathogenesis. Experimental inoculation of pure gangliosides and brain-derived ganglioside extracts were used in two different immunization studies. The first study was performed on 17 healthy chickens (Gallus gallus domesticus): 11 chickens were inoculated with a brain ganglioside extract in Freund’s complete adjuvant (FCA) and six chickens inoculated with phosphate-buffered saline. A second study was performed five healthy quaker parrots (Myiopsitta monachus) that were divided into three groups: Two quaker parrots received purified gangliosides in FCA, two received a crude brain extract in FCA, and one control quaker parrot received FCA alone. One chicken developed difficult in walking. Histologically, only a mild perivascular and perineural lymphocytic infiltrate in the proventriculus. Two quaker parrots (one from each treatment group) had mild lymphoplasmacytic encephalitis and myelitis. However, none of the quaker parrots developed myenteric ganglioneuritis, suggesting that autoantibodies against gangliosides in birds are not associated with a condition resembling PDD.

Parrot bornaviruses (PaBVs) are the causative agents of proventricular dilatation disease, however key aspects of its pathogenesis, such as route of infection, viral spread and distribution, and target cells remain unclear. Our study aimed to track the viral spread and lesion development at 5, 10, 20, 25, 35, 40, 60, 80, 95 and 114 dpi using histopathology, immunohistochemistry, and RT-PCR. After intramuscular inoculation of parrot bornavirus 2 (PaBV-2) in the pectoral muscle of cockatiels, this virus was first detected in macrophages and lymphocytes in the inoculation site and adjacent nerves, then reached the brachial plexus, centripetally spread to the thoracic segment of the spinal cord, and subsequently invaded the other spinal segments and brain. After reaching the central nervous system (CNS), PaBV-2 centrifugally spread out the CNS to the ganglia in the gastrointestinal (GI) system, adrenal gland, heart, and kidneys. At late points of infection, PaBV-2 was not only detected in nerves and ganglia but widespread in the smooth muscle and/or scattered epithelial cells of tissues such as crop, intestines, proventriculus, kidneys, skin, and vessels. Despite the hallmark lesion of PaBVs infection being the dilation of the proventriculus, our results demonstrate PaBV-2 first targets the CNS, before migrating to peripheral tissues such as the GI system.

Este trabalho descreve a colheita adequada de amostras, as técnicas/procedimentos disponíveis para o diagnóstico de influenza A em suínos, assim como os resultados e suas respectivas interpretações, para auxiliar médicos veterinários de campo na identificação dessa doença. Em suínos vivos, as amostras adequadas são: secreção nasal, fluido oral e sangue (soro). Para suínos mortos, colher preferencialmente amostras de pulmão com consolidação cranioventral. Secreção nasal e fragmentos de pulmão refrigerado são utilizados para detectar partícula viral viável (isolamento viral - IV) ou ácido nucleico viral (RT-PCR convencional e RT-PCR em tempo real). As amostras não devem ser congeladas, pois o vírus é inativado a -20°C. A caracterização molecular dos isolados é feita pela análise filogenética obtida pelo sequenciamento de DNA. O soro é utilizado para a detecção de anticorpos (Acs) por meio do teste da inibição da hemaglutinação e ELISA. O fluido oral pode ser utilizado para detecção de anticorpo (ELISA) ou de vírus. Fragmentos de pulmão fixados em formol a 10% são examinados microscopicamente para identificar pneumonia broncointersticial e para detecção de antígeno viral pela imuno-histoquímica (IHQ). Para o sucesso do diagnóstico, as amostras devem ser colhidas de suínos que estão preferencialmente na fase aguda da doença, para aumentar as chances de detecção viral. As melhores opções para o diagnóstico de influenza A em suínos vivos são RT-PCR e isolamento viral de amostras de swab nasal ou fluido oral. Pulmão para análise por RT-PCR, isolamento viral ou IHQ é a amostra de escolha em suínos mortos. Testes sorológicos têm valor diagnóstico limitado e são utilizados apenas para determinar o estado imune do rebanho, não indicando doença clínica, pois os Acs são detectados 7-10 dias pós-infecção (fase subaguda). O diagnóstico de influenza é importante para avaliar o envolvimento desse agente no complexo de doença respiratória suína. Além disso, o isolamento do vírus influenza é essencial para o monitoramento dos principais subtipos circulantes em uma determinada região ou país, assim como para a detecção de novos rearranjos virais, já que influenza é considerada uma zoonose.This article is intended to describe the adequate sample collection, the laboratory procedures/techniques, the expected results and their interpretation for diagnosis of influenza infection in swine, serving as a support for field veterinarians. In live pigs, the samples to be taken are nasal secretions, oral fluids and blood. For dead pigs, preference should be given to samples of cranioventral lung consolidation. Nasal discharge and chilled lung fragments are used for detection of virus (virus isolation - VI) or viral nucleic acids (conventional RT-PCR and real-time RT-PCR). Samples should not be frozen, because the virus is inactivated at -20°C. Molecular characterization of isolates is performed by phylogenetic analysis of gene sequences obtained by DNA sequencing. Serum is used for the detection of antibodies using hemagglutination inhibition (HI) test and ELISA. Oral fluid may be used for either antibody (ELISA) or viral detection. Fragments of lung fixed in 10% formaldehyde are used for histopathological analysis to identify bronchointerstitial pneumonia, and for immunohistochemistry (IHC) for antigens. For a successful diagnosis, sampling should be preferably performed in the acute phase of the disease to improve chances of virus detection. The best options to perform the diagnosis of influenza A in a swine herd are RT-PCR and VI from nasal swabs or oral fluid in live pigs and/or lung tissue for RT-PCR, VI or IHC in dead pigs. Serological tests are of very limited diagnostic value and are useful only to determine the immune status of the herd, not indicating clinical disease, because antibodies are detected after 7-10 days post infection (subacute phase). The diagnosis of influenza is important to evaluate the involvement of this agent in the complex of respiratory diseases in pigs. Furthermore, the isolation of influenza virus is essential for monitoring the main subtypes circulating in a given region or country, as well as for the detection of potential new viral reassortants, because influenza is considered a zoonosis.

Os protocolos de necropsia de 335 eqüinos necropsiados no LPV-UFSM entre 1968-2007 foram revisados com o objetivo de determinar os achados de necropsia relacionados com a causa da morte ou razão de eutanásia. A distribuição desses achados de acordo com os sistemas afetados foi a seguinte: digestivo (79/335 [23,6%]), músculo-esquelético (47/335 ([14,0%]), nervoso (37/335 [11,0%]), respiratório (35/335 [10,4%]), tegumentar (31/335 [9,3%]), hematopoético (24/335 [7,2%]), cardiovascular (13/335 [3,9%]), reprodutor (12/335 [3,5%]), urinário (7/335 [2,1%]) e endócrino (3/335 [0,9%]). Não foi possível determinar a causa da morte em 47 (14,0%) eqüinos necropsiados. As principais afecções do sistema digestivo foram as alterações na posição dos intestinos (17/79 [21,5%]), seguidas pelas obstruções e impactações (14/79 [17,7%]). As torções foram as principais alterações da posição dos intestinos (14/17 [82,4%]). Dentre as torções, as mais prevalentes foram as localizadas no intestino delgado (7/14 [50%]). A maioria dos eqüinos que morreram em razão de fratura óssea tinham idades entre 1-5 anos. As duas doenças mais freqüentemente diagnosticadas no sistema nervoso foram leucoencefalomalacia e tripanossomíase. Depressão respiratória causada por anestesia foi a principal causa de morte relacionada com o sistema respiratório. A anemia infecciosa eqüina foi a doença infecciosa mais diagnosticada e a principal razão para eutanásia observada neste estudo.The necropsy reports of 335 horses necropsied at the LPV-UFSM between 1968-2007 were reviewed in order to determine the necropsy findings related with cause of death or reason for euthanasia. The distribution of these findings by organ system were as follows: digestive (79/335 [23.6%]), striated muscle and skeleton (47/335 [14.0%]), nervous (37/335 [11.0%]), respiratory (35/335 [10.4%]), integument (31/335 [9.3%]), hematopoietic (24/335 [7.2%]), cardiovascular (13/335 [3.9%]), reproductive (12/335 [3.5%]), urinary (7/335 [2.1%]), and endocrine (3/335 [0.9%]). The cause of death was not possible to be determined in 47 (14.0%) necropsied horses. Displacements of the intestines (17/79 [21.5%]) were the main findings in digestive system, followed by obstruction and impactation (14/79 [17.7%]). Torsion were the type of displacement more frequently observed in the intestines (14/17 [82.4%]). Among those the more prevalent affected the small intestine (7/14 [50%]). Most horses dying from fractured bones were 1-5-year-old. The most prevalent diseases in the nervous system were leukoencephalo-malacia and trypanosomiasis, whereas respiratory depression due to anesthesia was the leading cause of death related to the respiratory system. Equine infectious anemia was the most diagnosed infectious disease and the main reason leading to euthanasia.

Foi realizado um levantamento nos arquivos do Laboratório de Patologia Veterinária (LPV) da Universidade Federal de Santa Maria (UFSM) e revisados os laudos de necropsias de bovinos realizadas entre 1990 e 2005. Foram revisados 2.912 casos referentes a necropsias realizadas por membros do LPV ou a materiais de necropsias realizadas por veterinários de campo que enviaram amostras para avaliação histológica no LPV. Em 461 (15,83%) das necropsias, a causa da morte foi atribuída à ingestão de plantas tóxicas. Em ordem decrescente de freqüência, intoxicações pelas seguintes plantas foram diagnosticadas: Senecio spp (56,14%), Pteridium aquilinum (12,06%), Ateleia glazioviana (10,31%), Solanum fastigiatum (5,04%), Baccharis coridifolia (3,29%), Xanthium cavanillesii (3,07%), Senna occidentalis (2,63%), Ramaria flavo-brunnescens (2,41%), Amaranthus spp (2,19%), Vicia villosa (1,54%), Ipomoea batatas, Prunus sellowii e polpa cítrica (0,44% cada), Cestrum parqui, Claviceps paspali, Claviceps purpurea, Brachiaria spp e Lantana sp (0.22% cada). Em um determinado surto o número de bovinos afetados era substancialmente maior que o número de necropsias realizadas. São discutidos os aspectos relacionados à distribuição geográfica, fatores que induziram a ingestão, índices de morbidade, mortalidade e letalidade, sinais clínicos, achados de necropsia e histopatológicos para cada intoxicação. Quando conhecidos, foram incluídos na discussão aspectos relacionados ao princípio ativo e a patogenia da intoxicação.From 1990 to 2005, tissues from 2,912 cattle necropsies were examined at the Laboratory of Veterinary Pathology (LPV) of the Federal University of Santa Maria (UFSM), Brazil. These tissues came from necropsies performed by faculty members of the LPV or were mailed-in samples from necropsy performed by veterinarian practitioners. In 461 (15.83%) of these necropsies the cause of death was attributed to the ingestion of poisonous plants. In decreasing order of frequency poisoning by the following plants were registered: Senecio spp (56.14%), Pteridium aquilinum (12.06%), Ateleia glazioviana (10.31%), Solanum fastigiatum (5.04%), Baccharis coridifolia (3.29%), Xanthium cavanillesii (3.07%), Senna occidentalis (2.63%), Ramaria flavo-brunnescens (2.41%), Amaranthus spp (2.19%), Vicia villosa (1.54%), Ipomoea batatas, Prunus sellowii, cytrus pulp (0.44% each), Cestrum parqui, Claviceps paspali, Claviceps purpurea, Brachiaria spp and Lantana sp (0.22% each). In a given outbreak the number of affected cattle was substantially higher than the number of necropsies performed. The following aspects are discussed for each plant: geographical distribution; factors inducing ingestion; morbidity, mortality and lethality rates, clinical signs, necropsy findings, histopathology. For those plants in which information on the active principle and pathogenesis are available, these aspects are included in the discussion.

A meningoencefalite por herpesvírus bovino-5 (BoHV-5) é uma doença infecto-contagiosa, aguda ou subaguda, geralmente fatal e que afeta principalmente bovinos jovens submetidos a situações de estresse. A doença tem sido freqüentemente diagnosticada em várias regiões do Brasil e em outras partes do mundo. BoHV-5 é um vírus da família Herpesviridae e subfamília Alphaherpesvirinae e possui como genoma uma molécula de DNA fita dupla. Esses vírus são caracterizados por rápida replicação em cultivo, que resulta em lise das células infectadas, e afetam várias espécies de hospedeiros, estabelecendo latência principalmente em neurônios de gânglios sensoriais. A transmissão de BoHV-5 ocorre principalmente por contato direto ou indireto entre bovinos. Após a replicação primária nas mucosas oral, nasal, ocular e orofaríngea, o vírus invade as terminações nervosas e é transportado até os neurônios de gânglios sensoriais, onde replica ativamente e estabelece latência. A invasão viral do encéfalo pode resultar em replicação viral massiva e produção de doença neurológica. A maioria dos bovinos que desenvolvem doença neurológica morre em decorrência de meningo-encefalite, porém alguns podem desenvolver infecção subclínica e, após recuperação, permanecerem portadores da infecção latente. A disseminação viral nos rebanhos é facilitada em situações de grande concentração de animais, introdução de bovinos e desmame de lotes de bezerros em idade que coincide com o decréscimo da imunidade passiva. Certas condições, naturais ou induzidas, podem reativar o vírus do estado latente e propiciar condições para sua transmissão e disseminação a outros indivíduos. A doença pode ocorrer na forma de surtos ou em casos isolados, com coeficientes de morbidade que podem variar de 0,05%-5%; a letalidade é quase sempre de 100%. Os sinais clínicos incluem depressão, descarga nasal e ocular, ranger de dentes, andar em círculos, cegueira, febre, movimentos de pedalagem, disfagia, dor abdominal, nistagmo, tremores, sialorréia, incoordenação, opistótono, pressão da cabeça contra objetos, quedas e convulsões. A evolução do quadro clínico pode variar de 1 a 15 dias. Achados de necropsia podem estar ausentes, mas normalmente se observa tumefação das porções rostrais do córtex telencefálico e achatamento das circunvoluções, com segmentos amarelados e amolecidos (malacia). Com a evolução da doença, essas áreas se tornam gelatinosas e acinzentadas, e em casos avançados ocorre o desaparecimento segmentar do córtex telencefálico frontal (lesão residual). Em muitos casos podem ser observados focos de malacia na substância cinzenta dos núcleos basais e do tálamo. Histologicamente observa-se meningoencefalite não-supurativa necrosante, principalmente no córtex telencefálico frontal, associada a inclusões intranucleares eosinofílicas em astrócitos e neurônios, embora a freqüência dessas inclusões seja irregular. O diagnóstico de meningoencefalite por BoHV-5 deve ser feito com base nos achados epidemiológicos, clínicos, de necropsia e histopatológicos, associados com o isolamento do vírus em cultivo celular células ou com detecção de antígenos virais em seções do encéfalo ou em células descamadas presentes nas secreções nasais. A identificação e caracterização de BoHV-5 pode ser realizada por meio de testes com anticorpos mono-clonais, reação em cadeia de polimerase (PCR) e por análise de restrição genômica. Não há tratamento específico para a meningoencefalite por BoHV-5. Como o BoHV-1 e o BoHV-5 são antigenicamente muito semelhantes, recomenda-se a vacinação com vacinas para BoHV-1 como forma de reduzir as perdas causadas por BoHV-5, principalmente durante surtos de doença neurológica. Adicionalmente, outras medidas podem ser adotadas para prevenir ou reduzir os prejuízos ocasionados pela enfermidade, como testar sorologicamente os bovinos a serem introduzidos nos rebanhos, minimizar situações de estresse, sobretudo no desmame, e isolamento dos bovinos afetados.Meningoencephalitis caused by bovine herpesvirus-5 (BoHV-5) is an often fatal, acute or subacute infectious disease that affects mainly young cattle under stressing conditions. The disease has been recognized in several Brazilian regions and in other parts of the world. BoHV-5 is a double stranded DNA virus member of the Herpesviridae family and subfamily Alphaherpesvirinae. The virus is characterized by rapid and lytic replication in cell cultures and by the ability to establish lifelong latent infection in sensory nerve ganglia of the host. BoHV-5 is transmitted mainly by direct and indirect contact and replicates acutely in the oral, nasal, oropharingeal or ocular mucosae. After primary replication, the virus invades nerve endings and is transported to the neuron cell bodies of the sensory ganglia where it replicates actively and/or establishes latency. Viral invasion of the brain may result in massive virus replication and production of neurological disease. Virtually all cattle developing neurological disease die of meningoencephalitis; yet the infection may be subclinical in some animals. These animals recover and become latently infected. Viral dissemination within a herd is facilitated by conditions such as crowding, introduction of cattle from other herds and weaning of calves in ages that coincide with decrease of passive immunity. Certain natural or induced conditions may reactivate the latent virus and favor its transmission and dissemination to other susceptible individuals. The disease may occur as outbreaks or as sporadic cases, with morbidity rates ranging of 0.05%-5%; lethality is almost always 100%. Clinical signs include depression, nasal and ocular discharge, grinding of teeth, circling, blindness, fever, paddling movements, disphagia, abdominal pain, nystagmus, muscle tremors, drooling, incoordinated gait, opisthotonus, head pressing, falls and convulsions. Clinical course is usually 1-15 days. Necropsy findings may be absent but often there is swollen of the rostral portions of the cerebral cortex and flattening of gyri, with softening and segmental yellow discoloration (malacia). As the disease progresses the affected areas become gelatinous and grey and, in advanced cases, there is segmental loss of the cerebral cortex of the frontal lobe of the brain (residual lesion). In several cases there is malacia of the basal nuclei and of the thalamus. Histologically, there is necrotizing non-suppurative meningoencephalitis affecting mainly the cerebral cortex of the frontal lobe associated with eosinophilic intranuclear inclusion bodies in neurons and astrocytes, although the frequency of the inclusion bodies is inconsistent. The diagnosis of meningoencephalitis by BoHV-5 should be based on epidemiology, clinical signs, necropsy and histological findings. The diagnosis should be confirmed by viral isolation in cell culture and/or by detection of viral antigens in brain sections or in exfoliated cells from nasal secretions. The identification and characterization of BoHV-5 can be done by the use of mono clonal antibodies, polymerase chain reaction (PCR) and/or by restriction enzyme analysis of the viral genome. There is no specific treatment for the disease. As BoHV-1 and BoHV-5 are antigenically related, vaccination using BoHV-1 vaccines may be recommended as a means of reducing the losses caused by BoHV-5 infection, mainly during outbreaks of neurologic disease. Additionally, measures such as serologic testing of new additions to the herd; and management practices to prevent stress and to reduce conditions for virus dissemination among animals may help in reducing the incidence and the consequences of BoHV-5 infection and disease.

Lesões hepáticas causadas pela intoxicação por Senecio spp em bovinos são progressivas e mortes podem ocorrer vários meses após a ingestão da planta. Testes laboratoriais da função hepática nem sempre são indicadores confiáveis de animais subclinicamente afetados. A biópsia hepática pode ser indicada para identificar bovinos com lesões hepáticas, mas sem sinais clínicos, e pode ter também valor prognóstico, uma vez que se acredita que as lesões hepáticas evoluam para causar insuficiência hepática e morte. Tais bovinos poderiam ser identificados pela biópsia hepática e enviados para o abate antes de desenvolverem os sinais clínicos, minimizando assim os prejuízos. Este estudo visou avaliar a biópsia hepática como um método de diagnóstico e prognóstico em casos de intoxi-cação por Senecio spp. em bovinos. Um surto de intoxicação por Senecio brasiliensis foi diagnosticado em bezerros de leite que haviam ingerido feno contaminado por 5-10% dessa planta tóxica. Biópsia hepática com agulha de Menghini por abordagem transtorácica foi realizada em 135 bezerros que ingeriram feno contaminado. Os bezerros biopsiados foram acompanhados por um período de 26 meses após a biópsia. Dezessete bezerros biopsiados tinham lesões típicas da intoxicação por Senecio spp (bezerros positivos) e 118 tinham fígados histologicamente normais (bezerros negativos). As lesões hepáticas dos bovinos positivos incluíam fibrose, hepatomegalocitose e hiperplasia de ductos biliares. Quinze dos 17 bezerros positivos morreram com sinais clínicos típicos de intoxicação por Senecio spp. 17-149 dias após a biópsia; 13 desses foram necropsiados e apresentavam lesões macro e microscópicas típicas de intoxicação por Senecio spp. Dois bovinos positivos mantinham-se clinicamente normais ao final do período de observação pós-biópsia. O valor prognóstico (sensibilidae) do teste foi considerado alto, uma vez que 88,23% dos bezerros positivos morreram. A especificidade do teste foi considerada bastante alta (99.16%), uma vez que apenas um dos 118 bovinos negativos na biópsia hepática morreu durante o período de observação pós-biópsia. Em nenhum bovino foi observado qualquer efeito negativo relacionado à técnica da biópsia hepática.Liver lesions caused by Senecio spp poisoning in cattle are progressive and deaths may occur many months after the plant is ingested. Laboratory tests of liver function are not always reliable indicators of subclinical affected animals. Liver biopsy could be useful to identify cattle with hepatic lesions but without clinical signs and would have also a prognostic value since it is generally believed that hepatic lesions will eventually cause liver failure and death. Such animals could be picked out by liver biopsy before clinical signs develop and be sent to slaughter, minimizing economic losses. This study was aimed to evaluate the liver biopsy as a diagnostic and prognostic tool in cases of Senecio spp. poisoning in cattle. An outbreak of Senecio brasiliensis was diagnosed in dairy calves which ingested hay contaminated by 5-10% of this Senecio species. Liver biopsy using a Menghini needle by right transthoracic approach was carried out in 135 calves that ingested the contaminated hay. Biopsed calves were followed up for 26 months after the biopsy. Seventeen biopsied calves had typical lesions of Senecio spp poisoning (positive calves) and 118 had histologically normal livers (negative calves). Hepatic lesions of positive calves included fibrosis, hepatomeglocytosis, and biliary hyperplasia. Fifteen out of the 17 positive calves died with typical clinical signs of Senecio spp poisoning within 17-149 days after the biopsy; 13 of those were necropsied and had typical gross and histopathological lesions of Senecio spp poisoning. Two positive calves were clinically normal at the end of the post-biopsy observation period. The prognostic value (sensibility) of the test was considered high since 88.23% of the positive calves died. The specificity of the test was considered very high (99.16%) since only one of the 118 negative calves died in the observation period. In none of the biopsed calves a negative effect related to the biopsy technique was observed.

Dezenove casos de intoxicação por Solanum fastigiatum são descritos em bovinos de três municípios do Rio Grande do Sul no período de 2003-2005. Os coeficientes de morbidade e mortalidade foram respectivamente de 6,7% e 3,4%. Os bovinos afetados tinham idade média de 5 anos (2-8 anos) e a evolução clínica variou de 3-18 meses. Os sinais clínicos observados em todos os bovinos foram de disfunções cerebelares caracterizadas por hipermetria, incoordenação, quedas, tremores musculares, convulsões esporádicas e permanência em estação com os membros em base ampla. Em um bovino foi observada hemorragia traumática subdural no encéfalo e em outro, atrofia macroscópica do cerebelo. As lesões histológicas eram confinadas ao cerebelo e consistiram de vacuolização parcial ou total do pericário dos neurônios de Purkinje com ocasionais esferóides axonais na camada de células granulares e na substância branca cerebelar. Nos casos avançados, havia acentuada perda dos neurônios de Pukinje e proliferação dos astrócitos de Bergmann. O estudo morfométrico do número de neurônios de Purkinje e da espessura da camada molecular revelou um decréscimo no número de células de Purkinje que se correlacionou com conseqüente diminuição da espessura da camada molecular.

Foram estudados 30 bovinos com neoplasias no trato alimentar superior (TAS) associadas ao consumo espontâneo de samambaia (Pteridium aquilinum) provenientes de 27 propriedades rurais, sendo 23 no município de Jaguari e quatro em Nova Esperança do Sul, Rio Grande do Sul. A população bovina total das 27 propriedades em que ocorreram os casos era de 1.090 bovinos e havia quantidade abundante de samambaia nas áreas de pastoreio dos animais. Vinte e seis bovinos eram vacas e 4 eram machos castrados. A idade variou de 3 a 13 anos, sendo o maior número de casos entre 7 e 8 anos (46,6%). Os sinais clínicos observados incluíram emagrecimento progressivo, atonia ruminal, tosse, disfagia, regurgitação, halitose, diarréia e timpanismo. Outros sinais clínicos menos freqüentes foram apetite seletivo, dispnéia e salivação. Dois bovinos tiveram morte espontânea e 28 foram submetidos à eutanásia em estágios avançados da doença e necropsiados. Os principais achados macroscópicos e histológicos observados nos 30 bovinos localizavam-se nos mesmos locais do TAS e consistiram de papilomas, papilomas em transformação para carcinomas de células escamosas (CCEs) e CCEs. Metástases de CCEs para linfonodos regionais e outros órgãos (como fígado e pulmões) foram também observadas (18/30). Vinte e nove bovinos tinham papilomas de diversos tamanhos, sendo a quantidade variável entre leve (45%), moderada (38%) e acentuada (17%). Nos papilomas examinados microscopicamente, foram observadas três fases de desenvolvimento: a) fase de inicial de crescimento, b) fase de desenvolvimento, e c) fase de regressão. Em 16 bovinos, observou-se a transformação maligna de papilomas em CCEs. Os CCEs eram únicos (12/30) ou múltiplos (18/30) e variaram quanto ao grau de diferenciação celular entre bem, moderadamente ou pouco diferenciados. Quando a distribuição dos CCEs de maior extensão foi agrupada em regiões cranial (base da língua, faringe/orofaringe, epiglote), média (terços cranial, médio e caudal do esôfago) e caudal (entrada do rúmen e rúmen) do TAS, observou-se que a localização era cranial em 39% dos casos, média em 16%, e caudal em 45%. Utilizando-se esse mesmo critério de agrupamento, porém considerando o número total de vezes em que CCEs (de tamanhos variados) foram diagnosticados nas regiões cranial, média e caudal, os números alteraram-se para 34, 26 e 40%, respectivamente. As evidências epidemiológicas e histomorfológicas relatadas neste estudo reforçam as observações de uma estreita correlação entre a infecção pelo papiloma-vírus bovino tipo 4, causador da papilomatose digestiva, e a co-carcinogênese química dos princípios tóxicos da samambaia na patogênese dos CCEs do TAS de bovinos. Entretanto, a presença de alterações pré-neoplásicas (áreas de displasia), áreas de CCE in situ ou CCEs em estágios iniciais de desenvolvimento, independentemente da presença de papilomas no local, mostram claramente ser possível o desenvolvimento de CCEs diretamente do epitélio normal, possivelmente por uma ação direta dos carcinó-genos químicos da samambaia.Thirty bovine with neoplasms of the upper digestive tract (UDT) associated with spontaneous ingestion of bracken fern (Pteridium aquilinum) were studied. They were from 27 farms, located in the municipalities of Jaguari (23) and Nova Esperança do Sul (4), Rio Grande do Sul, Brazil. The total cattle population in those farms was 1,090 and large amounts of bracken fern were found in the pastures. Twenty-six of the affected cattle were cows and four were castrated males, 3-13 years of age; most of them were 7-8 years old (46,6%). Clinical signs observed in the affected animals were progressive weight loss, absence of ruminal movements, cough, dysphagia, regurgitation, halitosis, diarrhea, and bloat. Less frequent signs were selective appetite, dyspnea, and salivation. Two bovine died and 28 were submitted to euthanasia in advanced stage of disease and necropsied. The main gross and microscopic alterations were found in identical areas of the UDT. They consisted of papillomas, transforming papillomas, and squamous cell carcinomas (SCCs). Metastases of SCCs to regional lymph nodes and other organs, such as liver and lungs, were also observed (18/30). Twenty-nine bovine had papillomas of various sizes in several areas of the UDT. The digestive papillomatosis ranged from mild (45%), to moderate (38%), to severe (17%). Three developing phases were observed microscopically in the examined papillomas: an early growing phase, a developing phase, and a regressing phase. In 16 cases, there was malignant transformation of papillomas into SCCs. The SCCs were solitary (12/30) or multiple (18/30) and were histologically well, moderately, or poorly differentiated. Grouping the distribution of SCCs of larger extension in the UDT into cranial region (base of the tongue, pharynx/oropharynx, and epiglottis), medial region (esophagus), and caudal region (cardia and rumen), the distribution was cranial in 39%, middle in 16%, and caudal in 45% of the cases. By the same grouping criteria, but considering the total number of times SCCs of varied extensions were diagnosed in the cranial, middle, and caudal regions, the percentages changed to 34%, 26%, and 40%, respectively. The epidemiological and histomorphological evidences found in this study are in agreement with the observations that point out the co-carcinogenesis between bovine papillomavirus type 4 infection and chemicals of bracken fern in the pathogenesis of the SCCs in the UDT of cattle. However, the presence of pre-neoplastic changes and SCCs in situ or in early stages of development, independently of the presence of papillomas, clearly indicates the possibility of development of SCCs from normal epithelium, probably due to the direct action of the chemical carcinogens contained in bracken fern.

O complexo heterogêneo de tecidos formado pelo gânglio de Gasser, rete mirabile carotídea e hipófise (GRH) de 199 bovinos foram macro e microscopicamente avaliados. Vinte e um GRH eram de casos confirmados de raiva, 19 com diagnóstico de meningoencefalite por herpesvírus bovino-5 (BoHV-5), 11 casos de febre catarral maligna (FCM), 7 tinham abscesso de pituitária, 17 apresentaram lesões de encefalopatia hepática (status spongiosus) atribuído à fibrose crônica por intoxicação por Senecio sp; e os 124 GRH restantes eram provenientes de bovinos que não apresentaram sinais neurológicos e que morreram ou foram eutanasiados por causas diversas. Nenhuma alteração histológica significativa foi observada no GRH dos 124 bovinos que não tinham sinais neurológicos, nos 17 bovinos com encefalopatia hepática, nem nos 19 bovinos afetados por meningoencefalite por BoHV-5. Alterações inflamatórias foram encontradas em 20 dos 21 casos de raiva e consistiam de proliferação de células satélites, nódulos gliais e infiltrado linfo-plasmocitário; alterações degenerativas incluíam cromatólise central, vacuolização neuronal e necrose neuronal com neuronofagia. Corpúsculos de inclusão eosinofílicos intracitoplasmáticos (de Negri) foram encontrados em 19 dos 21 casos de raiva; neurite do nervo trigêmeo em 11 casos e neuroipofisite em 4 casos. O complexo GRH de 9 de 11 casos de FCM apresentava arterite necrosante, que afetava as artérias da rete mirabile carotídea. Em 7 dos 199 GRH examinados havia abscessos de pituitária caracterizados por agregados de neutrófilos e detritos celulares circundados por infiltrado mononuclear e cápsula fibrosa. Vários achados incidentais foram observados nos 199 GRH examinados histologicamente e não foram correlacionados com alterações ligadas a doenças. Estes incluíam cistos na cavidade hipofisária, agregados de fibrina e neutrófilos no seio cavernoso da rete mirabile carotídea, leve aumento do número de células satélites ao redor dos neurônios do gânglio de Gasser e ocasional vacuolização e lipofuscinose neuronal. O exame histológico do complexo GRH é uma ferramenta importante e confiável no diagnóstico das principais encefalites em bovinos no Brasil.The complex of heterogenous tissues formed by the Gasserian ganglia, carotid rete mirabile and pituitary gland (GRH) from 199 cattle were grossly and histologically evaluated. Twenty one of the GRH were from comfirmed cases of rabies, 19 were diagnosed as cases of meningoencephalitis caused by bovine herpesvirus-5 (BoHV-5), 11 were confirmed cases of malignant catarrhal fever (MCF), 7 had pituitary abscesses, 17 had lesions of hepatic encephalopathy (status spongiosus) attributable to chronic liver failure due to Senecio sp poisoning; and the remaining 124 GRH were from cattle which did not present neurological signs and died or were euthanatized due to various causes. No significant histologic changes were observed in the GRH from the 124 cattle which did not present neurological signs, in the GRH of the 17 cattle that had hepatic encephalopathy neither in the GRH of the 19 cattle diagnosed with BoHV-5 meningoencephalitis. Inflammatory changes were found in the Gasserian ganglia of 20 out of 21 cases of rabies and consisted of proliferation of sattelite cells, glial nodules and lymphoplasmacytic infiltrate; degenerative changes included central chromatolysis, neuronal vacuolation, and neuronal death with neuronophagia. Characteristic eosinophilic intracytoplasmatic inclusion (Negri) bodies were found in 19 out of 21 cases of rabies, neuritis of the fifth nerve in 11, and neurohypophysitis in 4 cases. The GRH from nine of the eleven cases of MCF had characteristic necrotizing arteritis affecting the arteries of the carotid rete mirabile. In seven cases pituitary abscesses were found; they were characterized by large aggregates of neutrophils admixed with cellular debris and surrounded by mononuclear cells and were walled by a fibrous capsule. Several incidental findings were observed equally in the 199 histologically examined GRH and consequently were non specific for any disease entity. These include cysts in the pituitary cleft, fibrin and neutrophilic aggregates in the cavernous sinus of the carotid rete mirabile, mildly increased numbers of sattelite cells around neurons of the Gasserian ganglia and occasional neuronal vacuolation and lipofuscinosis. It was concluded that the histological examination of the GRH is an important and reliable tool in the diagnosis of the main forms of encephalitis occurring in cattle in Brazil.

São descritos sete surtos e um caso isolado de meningoencefalite por herpesvírus bovino-5 (BoHV-5) em bovinos no Rio Grande do Sul entre 2002 e 2004. Foram afetados bovinos de 1-18 meses, de diversas raças e ambos os sexos. A maior freqüência foi observada em bovinos recém-desmamados e submetidos a outros fatores de estresse. Nesses surtos, de uma população total sob risco de 1.359 bovinos, 54 foram afetados, quatro se recuperaram e 50 morreram espontaneamente ou foram submetidos à eutanásia quando moribundos. Os índices gerais de morbidade, mortalidade e letalidade foram, respectivamente, de 3,97%, 3,67% e 92,59%. A evolução clínica variou de 3-10 dias e os sinais eram caracterizados por depressão, corrimento nasal ou ocular, ranger de dentes, andar em círculos, cegueira, febre, nistagmo, tremores, anorexia, disfagia, sialorréia, incoordenação, pressão da cabeça contra objetos, pêlos arrepiados, taquicardia, taquipnéia, dor abdominal, melena, quedas, decúbito, opistótono, convulsões e movimentos de pedalagem. Dezenove bezerros foram necropsiados. Achados de necropsia foram caracterizados por hiperemia das leptomeninges, tumefação das porções rostrais do telencéfalo, com achatamento das circunvoluções dos lobos frontais; nessas áreas havia focos marrom-amarelados e amolecidos (malacia). Nos casos de evolução clínica mais longa era observada acentuada tumefação, amolecimento e extensas áreas de hemorragia nos lobos frontais telencefálicos. Microscopicamente, todos os bovinos afetados apresentaram meningoencefalite não-supurativa e necrosante, que variou quanto à localização e intensidade nos 19 casos examinados e nas seções de encéfalo de um mesmo caso. A intensidade dessas lesões foi mais acentuada, em ordem decrescente, no córtex telencefálico frontal, nos núcleos da base, tálamo, tronco encefálico, córtex parietal, córtex occipital e cerebelo. O infiltrado inflamatório perivascular era constituído predominantemente por linfócitos e plasmócitos e, menos freqüentemente, neutrófilos. Outros achados incluíam variados graus de gliose, edema, necrose neuronal no córtex telencefálico, caracterizada por encarquilhamento e eosinofilia do citoplasma e picnose nuclear (neurônio vermelho), corpúsculos de inclusão basofílicos intranucleares em astrócitos e neurônios (21,05% dos casos), satelitose e neuronofagia. As áreas de amolecimento do parênquima eram caracterizadas por necrose do componente neuroectodérmico e manutenção das estruturas mesenquimais (vasos e micróglia), com infiltrado de células Gitter e, em casos mais graves, áreas de hemorragia. Nos casos crônicos apenas estruturas vasculares e poucas células Gitter permaneciam, formando uma cavidade entre a substância branca e as leptomeninges (lesão residual).Seven outbreaks and an isolated case of meningoencephalitis caused by bovine herpesvirus-5 (BoHV-5) in cattle in Rio Grande do Sul, Brazil, occurring in 2002-2004, are described. From a total population at risk of 1,359 cattle, 54 1-18-month-old calves from both sexes and several breeds were affected and 50 died spontaneously or were euthanatized while moribund. The highest frequency of cases was in recently weaned calves or calves submitted to other stressing factors. General rates of morbidity, mortality and lethality were respectively 3.97, 3.67 and 92.59%. Clinical courses varied from 3-10 days and included depression, nasal and ocular discharge, grinding of teeth, circling, blindness, fever, nistagmus, trembling, anorexia, dysphagia, drooling, incoordination, head pressing, rough hair coat, tachycardia, tachypnea, abdominal pain, melena, falls, recumbency, opisthotonus, convulsions and paddling. Nineteen calves were necropsied. Necropsy findings were characterized by hyperemia of leptomeninges, swollen rostral portions of the telencephalon, and flattening of frontal lobes gyri; frequently in these frontal areas there were segmental brown-yellow discoloration and softening (malacia) of the cortex. In cases with more protracted clinical courses there were extensive swelling, softening and hemorrhaging of the telencephalic frontal lobes. Microscopically, all affected cattle had a necrotizing non-suppurative meningoencephalitis with variable distribution among the 19 cases and among the various telencephalic regions of the same case. The severity of these changes were more marked, in decreasing order of intensity, in the telencephalic frontal cortex, basal ganglia (nuclei), thalamus, brain stem, parietal telencephalic cortex, occipital telencephalic cortex and cerebellum. Perivascular inflammatory infiltrate consisted predominantly of lymphocytes, plasm cells, and less frequently of neutrophils. Additional microscopic findings included variable degrees of gliosis, edema, neuronal necrosis in the telencephalic cortex characterized by shrinking and eosinophilia of perikaria and nuclear picnosis (red neuron); basophilic intranuclear inclusion bodies in astrocytes and neurons (21.05% of the cases); sattelitosis; and neuronophagia. The areas of softening in the cortical substance consisted of necrosis of the neuroctodermal elements with maintenance of mesenchymal structures (vessels and microglia), infiltrate of Gitter cells, and, in more severe cases, extensive hemorrhages. In chronic cases, only vascular structures and a few Gitter cells remained in the cortical area leaving a cavity between white matter and leptomeninges (residual lesion).

Dois surtos de intoxicação pelo cogumelo Ramaria flavo-brunnescens são relatados em bovinos de duas fazendas localizadas nos municípios de Santa Maria e São Gabriel, no Rio Grande do Sul, no período de abril-maio de 2005. De um total de 180 bovinos de sobreano que tiveram acesso a bosques de eucaliptos, 19 adoeceram e 10 morreram. A evolução clínica foi de 8-15 dias e os sinais clínicos incluíam depressão, perda de peso, desidratação, salivação excessiva, afrouxamento e perda dos pêlos longos da cauda, alisamento da superfície dorsal da língua com ocasional ulceração, afrouxamento do estojo córneo dos chifres, fezes em forma de cíbalos e recobertas por película de muco, hipópion, hifema e opacidade da córnea. Dois novilhos tinham leucocitose devido a leve desvio regenerativo à esquerda. Os achados de necropsia confirmaram as observações clínicas e adicionalmente incluíam esofagite fibrinonecrótica, principalmente no terço distal do esôfago. Alterações histopatológicas na pele da cauda incluíam hiperqueratose ortoqueratótica, folículos pilosos com contornos irregulares, espessamento da camada de queratina tricolemal e formação ocasional de tampões de queratina; degeneração e necrose da bainha radicular externa também era observada. Nos cascos havia hemorragia, fibrina e infiltrado neutrofílico nas lâminas dérmicas, hiperplasia do topo das lâminas epidérmicas com queratinização irregular e retenção dos núcleos; várias lâminas epidérmicas estavam encurtadas e fundidas. Na mucosa da língua o epitélio de revestimento estava adelgaçado, com atrofia e perda das papilas filiformes, áreas multifocais de disqueratose e espongiose das células da camada basal. Em algumas porções havia perda do epitélio e a superfície da língua era formada por tecido de granulação e infiltrado inflamatório misto. A mucosa esofágica de seis novilhos apresentava vários graus de necrose epitelial e inflamação. A perda do epitélio de revestimento revelava uma área subjacente de tecido de granulação com marcado infiltrado inflamatório predominantemente neutrofílico e macrofágico. Em seis novilhos, o bulbo, na altura do óbex, apresentava áreas focais bilaterais e simétricas de malacia que afetava a substância branca e partes do núcleo dorsal do vago e do núcleo hipoglosso. São discutidos a epidemiologia, os sinais clínicos, a patologia e a patogênese da intoxicação por R. flavo-brunnescens em bovinos.Two oubreaks of poisoning by the mushroom Ramaria flavo-brunnescens are reported in cattle from two farms located in the municipalities of Santa Maria and São Gabriel, state of Rio Grande do Sul, during April-May 2005. Out of a total of 180 yearling calves that had access to a pasture with eucalyptus woods, 19 were affected and 10 died. The clinical courses were 8-15 days and clinical signs included depression, weight loss, dehydration, drooling, loosening and loss of the long hairs of the tip of the tail, smoothening of the dorsal surface of the tongue with occasional ulceration, loosening of the corneal encasement of horns, hard and ball-shaped feces covered with a film of mucous, hypopion, hyphema and corneal opacity. Two calves had leucocytosis due to mild regenerative left shift. Nine calves were necropsied. Necropsy findings confirmed the clinical observation and additionaly included fibrinonecrotic esophagitis, mainly in the distal third of the esophageal mucosa. Histopathological changes in the skin of the tail included orthokeratotic hyperkeratosis; hair folicles with irregular contours, thickening of the tricolemmal keratin layer with occasional formation of keratin plugs, and degeneration and necrosis of the outer root sheath. At the laminar region of the hooves, there was hyperplasia of the top of epidermal laminae with irregular keratinization and retention of nuclei; several epidermal laminae were shortened and fused. There was hemorrhage, fibrin and neutrophilic infiltrate in the dermal laminae. In the mucosa of the tongue there was thinning of the covering epithelium, atrophy and loss of filiform papillae, multifocal areas of dyskeratosis, and spongiosis of the basal cell layer. In some parts the epithelium was lost and the surface consisted of granulation tissue and mixed inflammatory cell infiltrate. The esophageal mucosae of six calves had varying degree of epithelial necrosis and inflammation. The loss of the covering epithelium revealed an underneath area of granulation tissue with heavy inflammatory infiltrate composed predominantly of neutrophils and macrophages. In six calves there were focal symmetric bilateral areas of malacia observed at the level of obex in the medulla oblongata and affecting the white matter and parts of the dorsal nucleus of the vagus and of the hypoglossal nucleus. The epidemiology, clinical signs, pathology, and pathogenesis of poisoning by R. flavo-brunnescens in cattle are discussed.

Em um rebanho de 100 caprinos, foram observados casos de uma doença neurológica em três animais adultos jovens (identificados como A-C) entre outubro e dezembro de 2004. Os sinais clínicos incluíam desvio da cabeça, torcicolo, nistagmo, andar desequilibrado e quedas que acabavam em decúbito, durante o qual o caprino executava movimentos de pedalagem com os membros rígidos. Tratamento com antibióticos no Caprino C resultou em remissão temporária dos sinais. A evolução clínica foi de 5, 10 e 30 dias, respectivamente, para os Caprinos A, B e C. O Caprino A morreu espontaneamente e os outros dois foram eutanasiados na fase terminal da doença. Não foram observadas alterações macroscópicas. Lesões histológicas eram predominantemente unilaterais e consistiam de alterações inflamatórias e degenerativas que se estendiam do bulbo ao tálamo. Em todos os casos havia manguitos perivasculares de um ou mais tipos de células mononucleares (linfócitos, plasmócitos, macrófagos ativados) e ocasionalmente neutrófilos associados a áreas de malacia onde células Gitter preenchiam os espaços de perda do parênquima. Adicionalmente, o Caprino B tinha microabscessos no bulbo, ponte, e mesencéfalo e infiltrado neutrofílico e linfocítico multifocal nos fascículos do nervo trigêmeo, próximo ao gânglio de Gasser, e nas leptomeninges do cerebelo. No tronco encefálico do Caprino C, a reação era multifocal e com característica granulomatosa e incluía células epitelióides e, ocasionalmente, células gigantes multinucleadas. Listeria sp foi detectada por imunoisto-química em cortes de mesencéfalo rotineiramente processado dos Caprinos A e C e de ponte do Caprino B.

Relata-se um surto espontâneo de intoxicação em suínos pela ingestão de sementes de Aeschynomene indica e a reprodução da doença nessa espécie animal. O surto espontâneo ocorreu numa propriedade de criação de suínos localizada na região central do Rio Grande do Sul. Nessa propriedade havia 100 suínos (20 matrizes e 80 suínos jovens de várias categorias). Os suínos eram alimentados com uma ração feita na propriedade pela mistura de 50% farelo de milho, 25% de farelo de soja, 5% de um suplemento vitamínico-mineral de origem comercial e 20% quirera de arroz contaminada por 40% de sementes de A. indica. Embora aparentemente todos os suínos tenham recebido a mesma ração, apenas os suínos de 45 dias de idade foram afetados; as taxas de morbidade, mortalidade e letalidade foram respectivamente 25%-40%, 8,5%-20% e 25%-66%. Os sinais clínicos apareceram cerca de 24 horas após o início da administração da ração contendo sementes de A. indica e incluíam vários graus de incoordenação no andar, quedas, decúbito esternal com membros pélvicos posicionados afastados entre si, decúbito lateral e morte. Não foi possível determinar quantos suínos se recuperaram e quanto tempo levou a recuperação. Um suíno foi submetido à eutanásia e necropsiado na propriedade. A doença foi reproduzida em 5 suínos jovens (A-E) alimentados com uma ração contendo 10% (Suíno A), 15% (Suíno B) e 20% (Suínos C-E) de sementes de A. indica e em um suíno mais velho (Suíno F) que recebeu uma ração com 16,5% de sementes de A. indica. Os sinais clínicos foram semelhantes aos observados nos suínos do surto espontâneo. Os Suínos A, B e F foram submetidos à eutanásia e os Suínos C-E morreram de uma doença aguda respectivamente 16, 21 e 24 horas após o início do experimento. Os achados de necropsia incluíam acentuada hiperemia das leptomeninges em todos os suínos, grandes quantidades de sementes de A. indica no estômago e avermelhamento transmural da parede do intestino e conteúdo intestinal sanguinolento nos Suínos C-E. Um hematoma foi observado no pulmão do Suíno C. Os achados histopatológicos no encéfalo dos suínos alimentados com as maiores concentrações (20%) de sementes de A. indica (C-E) consistiram em áreas focais e simétricas de congestão, edema, hemorragia e tumefação do endotélio vascular em diversos núcleos e no córtex telencefálico. Nos Suínos A e B, que receberam menores concentrações das sementes de A. indica, e no Suíno F, caso espontâneo da doença, as alterações histológicas no encéfalo consistiam de áreas bem definidas de malacia focal simétrica; nessas áreas a neurópila normal era obliterada por numerosos macrófagos espumosos dispostos em estreita aposição, astrocitose e capilares com endotélios tumefeitos. Os focos de malacia focal simétrica em suínos intoxicados com sementes de A. indica afetavam os núcleos cerebelares e vestibulares, a substância negra, o putâmen e os núcleos mesencefálicos, oculomotor e núcleo vermelho. Esses dados indicam que a ingestão de sementes de A. indica é responsável por essa condição neurológica, que a doença pode ser fatal e que parece afetar igualmente suínos jovens e adultos. O desenlace clínico e as alterações patológicas são dependentes da dose e as lesões encefálicas progridem de danos vasculares a edema vasogênico, hemorragia e malacia.A spontaneous outbreak of a neurological disease in swine caused by the ingestion of Aeschynomene indica seeds and the reproduction of the disease in the same animal species are reported. The natural outbreak occurred in a pig-raising facility in the central region of the state of Rio Grande do Sul, Brazil. On the premises there were 100 pigs (20 breeding sows and 80 young weaned pigs from several categories) that were fed a ration made by mixing 50% of corn bran, 25% of soybean bran, 5% of a commercial mix of vitamins and minerals, and 20% of broken rice contaminated with 40% of A. indica seeds. Although all pigs apparently ingested the same ration, only 45-day-old pigs were affected; morbidity, mortality and lethality rates were respectively 25%-40%, 8.5%-20%; and 25%-66%. Clinical signs appeared 24 hours after the beginning of feeding of A. indica seeds contaminated ration and included variable degrees of incoordinated gait, falls, sternal recumbency with the hind limbs in a wide base stance, lateral recumbency and death. It was not possible to ascertain how many pigs recovered nor the time frame of recovery. One pig was euthanatized and necropsied in the premises. The poisoning was reproduced in 5 young pigs (A-E) which were fed a ration containing 10% (Pig A), 15% (Pig B) and 20% (Pigs C-E) of A. indica seeds, and in one older pig (Pig F) which was fed a ration with 16.5% of A. indica seeds. Pigs A, B and F were euthanatized and Pigs C-E died of an acute disease respectively 16, 21 and 24 hours after the beginning of the experiment. Clinical signs were similar to those observed in pigs of the spontaneous outbreak. Necropsy findings included marked hyperemia of the encephalic leptomeninges of all pigs; there were large amounts of A. indica seeds in the stomach and reddening of the intestinal wall and bloody intestinal content in Pigs C-E. A hematoma was observed in the lungs of Pig C. The histopathological findings in the brain of pigs fed rations with larger concentrations (20%) of A. indica seeds (C-E) included congestion, edema and hemorrhage and swollen vascular endothelia with focal symmetrical distribution in several brain nuclei and in the telencephalic cortex. In Pigs A and B, and in Pig F, the case which received the lower dosage of the seeds of A. indica, and in the pig from the spontaneous outbreak, histopathological changes in the brain consisted of discrete focal symmetrical areas of malacia in which closely packed Gitter cells and astrocytosis, and capillaries with swollen endothelium obliterated the normal neuropil. The symmetrical malacic foci caused by the ingestion of A. indica seeds in swine affected cerebellar and vestibular nuclei, putamen, and the mesencephalic oculomotor and red nuclei. This indicates that the A. indica seeds ingestion was responsible for the neurological condition, that it may be fatal and seems to affect equally young and older swine. The clinical outcome and pathological changes were dose-dependent, and the brain lesions progressed from damaged blood vessels to vasogenic edema, hemorrhage and malacia.

São descritos a epidemiologia, os sinais clínicos e os achados de necropsia e histopatológicos observados em casos de febre catarral maligna (FCM), que ocorreram de 1973 a 2003 em bovinos de 15 fazendas no Rio Grande do Sul. Em nove ocasiões (60%), a doença ocorreu de forma esporádica, afetando 1-3 bovinos por rebanho, enquanto, em seis fazendas (40%), a FCM ocorreu de forma epizoótica, afetando vários bovinos em cada rebanho. As taxas de morbidade variaram de 2,4% a 20%, e as taxas de letalidade foram de 83,3% a 100%. Bovinos de todas as idades e ambos os sexos foram afetados. Nos casos em que foi possível obter essa informação (9 fazendas), sempre havia ovinos em contato com os bovinos afetados, e o maior número de casos de FCM ocorreu na primavera e verão. A doença clínica era aguda ou subaguda, e os sinais clínicos incluíam febre, corrimento nasal e ocular, conjuntivite, salivação excessiva, hematúria, necrose das papilas bucais que se tornavam rombas, linfadenopatia, diarréia e distúrbios neurológicos. Os achados de necropsia incluíam opacidade da córnea, erosões e ulcerações em várias membranas mucosas do trato alimentar, respiratório, urogenital e conjuntiva; aumento de volume e hemorragia em linfonodos e múltiplos focos brancos nos córtices renais e nas tríades portais hepáticas. Dermatite crostosa foi observada em alguns casos. Os principais achados histopatológicos incluíam vasculite, necrose dos epitélios de revestimento e acúmulos de células inflamatórias em vários órgãos. A vasculite era associada com necrose fibrinóide da túnica média das artérias e acompanhada de infiltrado inflamatório constituído por linfoblastos, linfócitos, plasmócitos e macrófagos.The epidemiology, clinical signs, necropsy and histopathological findings in cases of malignant catarrhal fever (MCF) occurring in cattle from 15 farms in Rio Grande do Sul, Brazil, from 1973 to 2003, are described. In 9 instances (60%) the disease occurred as sporadic cases affecting 1-3 cattle whereas in six farms (40%) MCF occurred as epizootics involving several cattle in each affected herd. Morbidity rates ranged from 2.4% to 20% and lethality rates were 83.3% and 100%. Cattle of all ages and both sexes were affected. Where the information was available (9 farms) sheep were in contact with affected cattle and cases of MCF occurred more frequently in spring and summer. Clinical courses were acute or subacute and clinical signs included fever, nasal and ocular discharges, conjunctivitis, drooling, hematuria, necrosis and blunting of buccal papillae, enlargement of lymph nodes, diarrhea and neurological disturbances. Necropsy findings included opaque corneas, reddening, erosions and ulcerations in several mucous membranes of the alimentary, respiratory and urogenital tracts, and the conjunctiva; enlargement and haemorrhage of lymph nodes and multiple white foci in the renal cortices and in the hepatic portal triads. Crustous dermatitis was observed in some cases. Main histopathological findings included vasculitis, necrosis of the surface epithelia and accumulation of inflammatory cells in several organs. Vasculitis were associated with fibrinoid necrosis of the medial layer of arteries and inflammatory cells included lymphoblasts, lymphocytes, plasma cells and macrophages.

São descritos dois surtos de intoxicação por Baccharis coridifolia em bovinos no Sul do Brasil. Os casos ocorreram quando bovinos estressados, com fome e sede, provenientes de pastagens livres de B. coridifolia, foram introduzidos em locais altamente infestados por essa planta tóxica. A morbidade nos dois surtos foi de 21,73% e 22,51%, e a letalidade foi virtualmente de 100%. Os sinais clínicos incluíam discreto timpanismo, instabilidade dos membros pélvicos, tremores musculares, focinho seco, fezes secas ou diarréicas, polidipsia e inquietação. Alterações presentes em todas as necropsias incluíam desidratação, grande quantidade de conteúdo líquido no rúmen, avermelhamento e erosões da mucosa dos pré-estômagos. As principais alterações histopatológicas encontradas foram degeneração e necrose do revestimento epitelial dos pré-estômagos e do tecido linfóide.Two outbreaks of Baccharis coridifolia poisoning in cattle in southern Brazil are described. Cases occurred when stressed, hungry and thirsty cattle brought from pastures free of B. coridifolia were placed into pastures heavily infested by this poisonous plant. In the two outbreaks morbidity was 21.73% and 22.51% and lethality was virtually 100%. Clinical signs included mild bloat, instability of hind limbs, muscle tremors, dry muzzle, dry feces or diarrhea, polydipsia and restlessness. Consistent necropsy findings included dehydration, large amounts of ruminal fluid, reddening and erosions of the mucosae of the forestomachs. Degeneration and necrosis of the lining epithelium of the forestomachs and of lymphoid tissue were the main histopathological changes encountered.

Meningoencefalite granulomatosa foi observada em 7 de 8 vacas leiteiras adultas afetadas por doença granulomatosa sistêmica associada ao consumo de ervilhaca, embora nenhum dos bovinos afetados apresentasse sinais de distúrbios nervosos. Os infiltrados inflamatórios localizavam-se nas leptomeninges e como manguitos perivasculares no interior do encéfalo; consistiam de macrófagos epitelióides, linfócitos, plasmócitos e eosinófilos. Essas alterações inflamatórias variavam de leves a acentuadas de animal para animal e entre diferentes regiões do encéfalo de um mesmo animal. Geralmente, os manguitos perivasculares eram mais acentuados que os infiltrados nas leptomeninges. As regiões do encéfalo afetadas, em ordem decrescente de intensidade, diencéfalo através da massa intermedia; mesencéfalo na altura dos colículos rostrais; ponte e pedúnculos cerebelares, bulbo na altura do óbex, lobo frontal na altura do joelho do corpo caloso e cerebelo. O tipo e a distribuição das alterações inflamatórias são enfatizados em relação ao diagnóstico diferencial de outras doenças e lesões do sistema nervoso central de bovinos no contexto do programa brasileiro de vigilância para a encefalopatia espongiforme bovina.Granulomatous meningoencephalitis was observed in 7 out of 8 adult dairy cows affected by vetch-associated systemic granulomatous disease, although there was no neurological signs associated with the condition. The cellular inflammatory infiltrates were located in the leptomeninges and as perivascular cuffings within the brain and consisted of epithelioid macrophages, lymphocytes, plasm cells and eosinophils. These inflammatory changes varied from mild to severe from animal to animal and among different brain regions of the same animal. Perivascular cuffings were usually more marked than the leptomeningeal infiltrates. Affected brain regions, in decreasing order of intensity, included diencephalon through the level of massa intermedia, mesencephalon at the level of rostral colliculi, pons and cerebellar peduncles, medulla at the level of obex, frontal lobe at the level of the genu of the corpus callosum, and cerebellum. The character and the distribution of the inflammatory changes are emphasized regarding the differential diagnosis with other diseases and lesions of the bovine central nervous system in the context of the Brazilian surveillance program for bovine spongiform encephalopath.

São relatados dois surtos de febre catarral maligna (FCM) em bovinos de duas propriedades rurais (A e B) do município de Santiago, Rio Grande do Sul (RS), a transmissão da doença a bovinos suscetíveis e a detecção de DNA viral de herpesvírus bovino-2 (OvHV-2) em tecidos de bovinos afetados. Os dois surtos ocorreram de novembro de 2001 a fevereiro de 2002 (Propriedade A) e de janeiro a fevereiro de 2003 (Propriedade B). O número de bovinos sob risco, as taxas de morbidade e de letalidade foram, respectivamente, 170, 10,59% e 83,33% na Propriedade A e 500, 2,4% e 100% na Propriedade B. Em ambas as propriedades havia contato de ovinos com os bovinos afetados, mas somente na Propriedade A havia ovelhas em parição. Nos bovinos afetados nas duas propriedades, a duração do curso clínico, os achados de necropsia e a histopatologia foram semelhantes. A maioria dos bovinos afetados morreu ou foi submetida à eutanásia in extremis após um curso clínico de 2 a 8 dias. Os sinais clínicos incluíam febre (40,5 e 41,5°C), corrimento nasal e ocular, opacidade da córnea, conjuntivite, salivação, erosões e ulcerações em mucosas, diarréia, hematúria e distúrbios neurológicos. Foram realizadas onze necropsias (nove na Propriedade A e duas na Propriedade B). Lesões macroscópicas incluíam erosões e úlceras nas mucosas dos cornetos nasais, cavidade oral e tratos gastrintestinal e urogenital; hemorragia e necrose da ponta das papilas bucais, aumento de volume dos linfonodos, múltiplos focos brancos no córtex renal e hiperemia das leptomeninges. Microscopicamente, havia arterite e degeneração fibrinóide em artérias de médio e pequeno calibre e em arteríolas de múltiplos órgãos e tecidos, necrose e inflamação em várias superfícies mucosas, ceratite, conjuntivite, uveíte, nefrite intersticial e encefalite. A transmissão experimental foi tentada em cinco bezerros (E1-E5) através da inoculação de cada um deles, por via intravenosa, com 500 ml de sangue total heparinizado oriundo de bovino afetado por FCM. A transmissão foi conseguida em pelo menos três (E1-E3) dos bezerros experimentais que adoeceram após um período de incubação de 15 a 27 dias. Quatro dos bezerros do experimento morreram ou foram submetidos à eutanásia in extremis após um curso clínico que durou de 3 dias a 8 semanas. O bezerro experimental remanescente (E5) recuperou-se após uma doença branda e foi submetido à eutanásia 14 semanas após a inoculação. Os cinco bezerros foram necropsiados. Sinais clínicos, achados de necropsia e histopatologia de três bezerros (E1-E3) eram característicos de FCM. O DNA viral de OvHV-2 foi detectado pela técnica de reação em cadeia de polimerase (PCR) em tecidos emblocados em parafina de sete dos 11 bovinos espontaneamente afetados por FCM e em tecidos emblocados em parafina de três bezerros experimentais (E1-E3). A técnica de PCR resultou negativa nos restantes quatro dos 11 bovinos testados nos casos espontâneos de FCM e em dois (E4-E5) dos cinco bezerros usados nos experimentos de transmissão. Testes de imunoistoquímica realizados em cortes de tecido linfóide do bezerro E4 resultaram negativos para antígeno do vírus da diarréia viral bovina. A transmissão experimental de FCM de bovino para bovino e a caracterização do agente etiológico da doença em bovinos como OvHV-2 foi conseguida pela primeira vez no Brasil.Two oubreaks of malignant catarrhal fever (MCF) occurring in cattle on two farms (A and B) in the municipality of Santiago, state of Rio Grande do Sul (RS), Brazil, and the transmission of the disease to susceptible calves as well as the detection of ovine herpesvirus-2 (OvHV-2) in tissues of affected cattle are reported. The two epizootics occurred from November 2001 to February 2002 (Farm A) and in January-February 2003 (Farm B). Numbers of cattle at risk, morbidity and letality rates were respectively 170, 10.59% and 83.33% for Farm A and 500, 2.4% and 100% for Farm B. Contact between affected cattle and sheep was detected in both farms, but lambing ewes were present only in farm A. Duration of clinical courses, gross findings and histopathology were the same for the affected cattle in both farms. Most affected cattle died or were euthanatized in extremis after a clinical course of 2-8 days. Clinical signs included fever (40.5 and 41.5°C), nasal and ocular discharge, corneal opacity, conjunctivitis, drooling, erosions and ulcerations of the mucosae, diarrhea, hematuria, and neurological disturbances. Eleven necropsies (9 on Farm A, 2 on Farm B) were performed. Gross lesions included erosions and ulcers affecting the mucosae of nasal turbinates, oral cavity, gastrointestinal and urogenital tracts; hemorrhage and necrosis of the tip of the buccal papillae, lymph node enlargement, multifocal white foci in renal cortex, and hyperemia of leptomeninges. Microscopically, there were arteritis and fibrinoid degeneration in medium and small arteries and arterioles of multiple organs and tissues, necrosis and inflammation in several mucosal surfaces, keratitis, conjunctivitis, uveitis, intersticial nephritis, and encephalitis. Transmission experiments were attempted in five calves (E1-E5) by inocculating each of them intravenously with 500 ml of whole heparinized blood from a MCF affected cow. The transmission was suscessful in at least three (E1-E3) of the experimental calves which became sick after an incubation period of 15-27 days. Four experimental calves either died or were euthanatized in extremis after a clinical course which varied from 3 days to 8 weeks. The remaining experimental calf (E5) recovered from a mild disease and was euthanatized 14 weeks after inocculation. Necropsies were performed in all five calves. Clinical signs, necropsy and histopathological findings of three calves (E1-E3) were characteristic of MCF. OvHV-2 viral DNA was detected by the polimerase chain reaction (PCR) test in paraffin embedded tissues from seven cattle out of the 11 spontaneous MCF cases and from three experimental calves (E1-E3). PCR tests resulted negative in the remaining four of the 11 spontaneous MCF cases tested and in two (E4,E5) of the five experimental calves. Immunohistochemistry performed in sections of lymphoid tissue from calf E4 failed to detect BVD virus antigen. The experimental transmission of MCF and the characterization of the etiological agent as OvHV-2 were successfully attempted in cattle for the first time in Brazil.

Dois surtos de uma doença associada ao pastoreio de duas espécies de ervilhaca (predominantemente Vicia villosa e, em menor grau, V. sativa) foram observados em agosto-setembro de 2001, em vacas Holandesas adultas de duas propriedades rurais do Rio Grande do Sul. Foram afetadas, em uma das propriedades, quatro de 42 vacas (9,5%) e, na outra, uma de oito vacas (12,5%). Os sinais clínicos incluíam, embora não em todos os casos, febre, prurido, espessamento e enrugamento da pele com placas multifocais de alopecia, conjuntivite, corrimento nasal seroso, perda de peso, acentuada queda na produção de leite e diarréia. O curso clínico foi de aproximadamente duas semanas. Todas as vacas afetadas clinicamente morreram, uma foi sacrificada; três foram necropsiadas. Em cada um desses animais havia um padrão de lesões sistêmicas que consistiam de nódulos multifocais ou coalescentes, macios ou moderadamente firmes e branco-acinzentados, que infiltravam vários órgãos, mas eram particularmente proeminentes no miocárdio, nos linfonodos, no baço, na glândula adrenal e no córtex renal. Essas lesões resultavam em aumento de volume e alteração na arquitetura do órgão invadido. Microscopicamente, as lesões consis-tiam de extensa infiltração celular composta de proporções variáveis de macrófagos epitelióides, linfócitos, plasmócitos, células gigantes multinucleadas e eosinófilos. Os números de eosinófilos eram geralmente altos. Essa infiltração granulomatosa causava degeneração e perda de células parenquimatosas no órgão afetado. A intensidade das lesões variou entre as três vacas e entre os vários órgãos de cada animal. Essa é a primeira documentação no Brasil de doença granulomatosa sistêmica em bovinos associada ao pastoreio de ervilhaca.
