Abstract Background Urinary tract infection (UTI) is one of the most common infectious diseases at the community level. In order to assess the adequacy of the empirical therapy, the prevalence and the resistance pattern of the main bacteria responsible for UTI in the community (in Aveiro, Portugal) was evaluated throughout a ten-year period. Methods In this retrospective study, all urine samples from patients of the District of Aveiro, in ambulatory regime, collected at the Clinical Analysis Laboratory Avelab during the period 2000–2009 were analysed. Samples with more than 105 CFU/mL bacteria were considered positive and, for these samples, the bacteria were identified and the profile of antibiotic susceptibility was characterized. Results From the 155597 samples analysed, 18797 (12.1%) were positive for bacterial infection. UTI was more frequent in women (78.5%) and its incidence varied with age, affecting more the elderly patients (38.6%). Although E. coli was, as usual, the most common pathogen implicated in UTI, it were observed differences related to the other bacteria more implicated in UTI relatively to previous studies. The bacteria implicated in the UTI varied with the sex of the patient, being P. aeruginosa a more important cause of infection in men than in women. The incidence of the main bacteria changed over the study period (P. aeruginosa, Klebsiella spp and Providencia spp increased and Enterobacter spp decreased). Although E. coli was responsible for more than an half of UTI, its resistance to antibiotics was low when compared with other pathogens implicated in UTI, showing also the lowest percentage of multidrug resistant (MDR) isolates (17%). Bacteria isolated from females were less resistant than those isolated from males and this difference increased with the patient age. Conclusions The differences in sex and age must be taken into account at the moment of empirical prescription of antimicrobials. From the recommended antimicrobials by the European Association of Urology guidelines, the first line drugs (pivmecillinam and nitrofurantoin) and the alternative antibiotic amoxicillin-clavulanic acid (AMX-CLA) are appropriate to treat community-acquired UTI, but the fluoroquinolones should not be suitable to treat male infections and the trimethoprim-sulfamethoxazole (SXT) shall not be used in the treatment of UTI at this level.

Cten is an oncogene promoting EMT in many signaling pathways, namely through Snail. We investigated whether Cten function could be mediated through Src. Cten levels were modulated by forced expression in HCT116 and gene knockdown in SW620 CRC (colorectal cancer) cell lines. In all cell lines, Cten was a positive regulator of Src expression. The functional importance of Src was tested by simultaneous Cten overexpression and Src knockdown. This resulted in abrogation of Cten motility-inducing activity and reduction of colony formation ability together with failure to induce Cten targets. In SW620 ΔCten reduced Src expression increased following restoration of Cten, also leading to increased cell motility and colony formation, which were lost if Src was concomitantly knocked down. By qRT-PCR we showed modulation of Cten had no effect on Src mRNA. However, a CHX pulse chase assay demonstrated stabilization of Src protein by Cten. Finally, expression of Cten and Src was tested in a series of 84 primary CRCs and there was a significant correlation between them (P = 0.001). We conclude that Src is a novel and functionally important target of the Cten signaling pathway and that Cten protein causes post-transcriptional stabilization of Src in promoting EMT and possibly metastasis in CRC., (© 2019 Japanese Society of Pathology and John Wiley & Sons Australia, Ltd.)

Cten (C-terminal tensin-like) is a member of the tensin protein family found in complex with integrins at focal adhesions. It promotes epithelial-mesenchymal transition (EMT) and cell motility. The precise mechanisms regulating Cten are unknown, although we and others have shown that Cten could be under the regulation of several cytokines and growth factors. Since transforming growth factor beta 1 (TGF-β1) regulates integrin function and promotes EMT/cell motility, we were prompted to investigate whether TGF-β1 induces EMT and cell motility through Cten signalling in colorectal cancer. TGF-β1 signalling was modulated by either stimulation with TGF-β1 or knockdown of TGF-β1 in the CRC cell lines SW620 and HCT116. The effect of this modulation on expression of Cten, EMT markers and on cellular function was tested. The role of Cten as a direct mediator of TGF-β1 signalling was investigated in a CRC cell line in which the Cten gene had been deleted (SW620 ΔCten ). When TGF-β1 was stimulated or inhibited, this resulted in, respectively, upregulation and downregulation of Cten expression and EMT markers (Snail, Rock, N-cadherin, Src). Cell migration and cell invasion were significantly increased following TGF-β1 stimulation and lost by TGF-β1 knockdown. TGF-β1 stimulation of the SW620 ΔCten cell line resulted in selective loss of the effect of TGF-β1 signalling pathway on EMT and cell motility while the stimulatory effect on cell proliferation was retained. These data suggested Cten may play an essential role in mediating TGF-β1-induced EMT and cell motility and may therefore play a role in metastasis in CRC., (© 2019 The Authors. Background: Canine inflammatory mammary cancer (IMC) and its human counterpart, inflammatory breast cancer, are extremely aggressive types of cancer. Our aim was to characterize immunohistochemical expression of C-C chemokine receptor 2, colony stimulating factor 1 receptor and metalloproteinase-9 in canine IMC versus non-IMC and to analyze associations with clinicopathological variables., Materials and Methods: Immunohistochemical staining of CCR2, CSF1R and MMP9 was performed in a series of 25 IMC and 15 non-IMC tumors., Results: No differences in the expression of these biomarkers between IMC and non-IMC were observed. Distinct nuclear subcellular expression of CCR2 was observed in IMC (p<0.001). For IMC, higher CCR2 expression was associated with increased nuclear grade (p=0.037), and higher neoplastic MMP9 expression was associated with fewer mitoses (p=0.022), higher nuclear grade (p=0.047) and increased CSF1R expression (p=0.025)., Conclusion: Expression of CCR2, CSF1R and MMP9 in canine IMC could contribute to increased nuclear pleomorphism, but the biological mechanisms involved warrant further investigation., (Copyright© 2016 International Institute of Anticancer Research (Dr. John G. Clinical and molecular similarities between canine mammary tumours and human breast cancer have been described in recent decades. Clinically, the similarities are very strong: spontaneous tumours, hormonal aetiology, age of onset and an identical course of the disease. The clinical characteristics that have an impact on the clinical outcome are also identical: tumour size, lymph node invasiveness and clinical stage. Nowadays, as far as human medicine is concerned, the goal is to identify prognostic factors, mainly at the molecular level, such as those involved in metastasis, which could be used as therapeutic targets to support a better outcome. Moreover, in this area, canine mammary tumours seem to mimic human breast cancer, as a range of similarities are found at the molecular level concerning the overexpression of steroid receptors, proliferation markers, epidermal growth factor, p53 supressor gene mutations, metalloproteinases, cyclooxygenases, among many others. Clinical and molecular data that support canine mammary tumours as a model to study human breast cancer are analysed in this review. Additionally, it is shown that some recent molecular targets in canine mammary tumours may be seen as indicators for similar research to be performed in the corresponding human disease.

Unlabelled: Iron deficiency is the world most prevalent nutritional disorder., Objective: To determine the prevalence of iron deficiency anemia in a population of infants with nine months of age, and the variation of this prevalence according to infants feeding regimes., Material and Methods: One hundred eighty eight infants with nine months of age were studied for anthropometry, socio-economic factors and feeding regime. Blood samples were taken for blood cell counts, serum ferritin, iron and transferrin. Infants were labelled anaemic if haemoglobin level was less than 110 g/L and with iron deficiency anaemia if, in addition, they had a ferritin value of less than 12 g/L., Results: The mean (standard deviation) values for haemoglobin were 111.8 g/L (9.1 g/L). Seventy-nine infants (42%) were anaemic. The prevalence of iron deficiency anaemia was 20%. Infants with iron deficiency anaemia were fed with iron fortified formula for 2.3 months in average in contrast with 3.8 months of those without iron deficiency anaemia (.005). Cereals fortified with iron were introduced in the nourishment of infants with iron deficiency anaemia at 4.3 months in average in contrast with 3.9 months among those without it (.009). The infants with iron deficiency anaemia were breast fed for 4.8 months in average in contrast with 3.7 months among those without it (.079). Age of the introduction of cow's milk, social class and Graffar, vitamin C supplementation and anthropometric parameters were not significantly associated with iron deficiency anaemia., Conclusions: This study points information about which groups are more at risk of developing iron deficiency anaemia in infants but when this evidence is only based in feeding practices one can not completely separate infants with and without iron deficiency anaemia; therefore a universal screening seems an indispensable tool for identification of iron deficiency anaemia.
