A metástase é uma consequência comum e a principal causa de mortalidade por esta doença. A identificação de marcadores tumorais pode ser uma ferramenta útil para a determinação do prognóstico pelo câncer de mama. Assim, os objetivos deste estudo foram avaliar a expressão gênica e proteica do ATM, p53 e VEGF na glândula mamária de cadelas e tentar correlacioná-las com a evolução tumoral e possível envolvimento metastático. Com isso foram utilizadas 53 amostras de glândula mamária de cadelas divididas em quatro grupos: G1= mamas normais, G2 = lesões mamárias benignas, G3 = carcinomas mamários não metastáticos e G4 = carcinomas mamários metastáticos. A análise de expressão gênica foi realizada pelo método de RT-qPCR e de expressão proteica pelo método de imuno-histoquímica. Baixa regulação de transcritos do ATM foi observada entre as neoplasias e as mamas normais, além da diminuição de níveis proteicos, sendo esta diminuição maior no grupo 4 quando comparado com o grupo 1. Os transcritos de TP53 e VEGF não apresentaram diferença estatística entre os grupos analisados. Entretanto, verificou-se diminuição proteica da p53 e aumento da proteína VEGF nos carcinomas mamários metastáticos em comparação com a mama normal. As expressões gênica e proteica do ATM apresentaram comportamento semelhante ao observado em mulheres, assim, o processo de desenvolvimento de metástase do carcinoma mamário pode ser semelhante em ambas as espécies. Canine mammary tumor is the major tumor that affects bitches and their biological behavior is similar to what occurs in women, so bitches can be an excellent comparative model for the study of the carcinogenic process in this type of neoplasm. Metastasis is a common consequence and the major cause of death by the tumor. Identification of tumor markers can be a useful tool for determining the prognosis of mammary gland cancer. So, the objectives of this study were to evaluate ATM, p53, and VEGF gene and protein expression in canine mammary tissues and try to correlate them with tumor development and metastatic involvement potential. Therefore 53 bitches mammary gland tissue samples were used divided in four groups: G1 = normal mammary glands, G2: benign mammary lesions, G3 = non-metastatic mammary gland carcinomas and G4 = metastatic mammary gland carcinomas. Gene expression analysis was performed by RT-qPCR and protein expression by immunohistochemistry. Down-regulation of ATM transcripts was observed between tumor tissue and normal mammary gland and protein levels were also reduced, it was mainly observed in group 4 when compared to group 1. TP53 and VEGF markers did not show statistical difference in gene expression among the groups. However, it was observed lower level of p53 protein in malignant tumors and higher level of VEGF protein in tumor tissues. ATM showed similar behavior when compared to women, so tumorigenesis process of breast cancer may be similar in both species. This paper reports the case of a two-year-old, mixed-breed dog diagnosed with hydronephrosis of the left kidney six months after being kicked in the thoraco-abdominal region. The diagnosis was based on radiographic and ultrasonographic examinations. Percutaneous antegrade pyelography was also performed and allowed the visualization of opacification of the left kidney, which confirmed the diagnosis. Because the exams indicated that the patient had normal renal function, nephrectomy was performed. The patient remained in good health during the postoperative period and in subsequent evaluations. Embora existam inúmeros protocolos quimioterápicos descritos para o tratamento do mastocitoma cutâneo canino, os mesmos foram avaliados comparando-se grupos heterogêneos, com diferentes fatores prognósticos clínicos e histopatológicos, dificultando desta forma a escolha do melhor protocolo a ser utilizado. Pretende-se com este relato descrever os efeitos do protocolo neoadjuvante à base de vimblastina e lomustina num caso de mastocitoma grau III/ alto grau na região do períneo, de prognóstico reservado a mau, baseado nos fatores clínicos (localização, tamanho tumoral, velocidade de crescimento), características histopatológicas (grau de Patnaik e Kiupel) e imuno-histoquímica (imunomarcação de Ki-67, c-kit e glicoproteína P) desfavoráveis que respondeu favoravelmente ao tratamento instituído e apresenta até o presente momento uma sobrevida de 110 semanas sem sinais de recidiva da doença. The clinical and biological behavior of canine cutaneous mast cell tumors is highly variable, making the establishment of prognosis and choose of the best treatment to be instituted a challenge to veterinary oncologists. While there are many chemotherapeutic protocols outlined for treatment of canine cutaneous mast cell tumors, they were evaluated by comparing heterogeneous groups, with different clinical and histopathological prognostic factors, thus making it difficult to select the best protocol to use. The intention of this report is describe the effects of a neoadjuvant protocol with vinblastine and lomustine in a case of grade III / high degree mast cell tumour in perineum, with worse prognosis, based on unfavorable clinical factors (location, tumor size, growth rate ), histopathological characteristics (degree of Patnaik and Kiupel) and immunohistochemistry (immunostaining of Ki-67, kit and P-glycoprotein ) who responded favorably to treatment and has to date a survival of 110 weeks without signs of recurrence disease. Cancer is one of the main causes of death in canines and felines, and this fact is probably related to the increase in the longevity of these species. The longer the animals live, the higher the exposure to carcinogenic agents will be. With the high incidence of cancer in companion animals, new studies are currently being performed with the aim of finding therapeutic options which make the complete inhibition of the development of neoplasms in animals possible in the future. The correlation of cyclooxygenase-2 (COX-2) whith the development of cancer opens the way for the use of new therapeutic approaches. This relationship has been suggested based on various studies which established an association between the chronic use of nonsteroidal anti-inflammatory drugs (NSAID) and a decrease in the incidence of colon carcinoma. As cancer progresses, COX-2 participates in the arachidonic acid metabolism by synthesizing prostaglandins which can mediate various mechanisms related to cancer development such as: increase in angiogenesis, inhibition of apoptosis, suppression of the immune response, acquisition of greater invasion capacity and metastasis. Accordingly, overexpression of this enzyme in tumors has been associated with the most aggressive, poorprognosis cancer types, especially carcinomas. Therefore, treatments which use COX-2 inhibitors such as coxibs, whether administered as single agents or in combination with conventional antineoplastic chemotherapy, are an alternative for extending the survival of our cancer patients. CAPES - Coordenação de Aperfeiçoamento de Pessoal de Nível Superior FAPEMIG - Fundação de Amparo a Pesquisa do Estado de Minas Gerais O mastocitoma é dos tumores cutâneos mais comuns em cães e caracteriza-se pela proliferação de mastócitos neoplásicos, normalmente em derme e epiderme. Seu comportamento é variável, dificultando o estabelecimento do tratamento e tornando imprescindível a investigação de fatores prognóstico que possam auxiliar na previsão do comportamento destes tumores. Objetivou-se avaliar: alterações histomorfológicas em mastocitomas caninos, a expressão imuno-histoquímica de c-KIT e Ki-67, a marcação para AgNOR, a correlação entre estas variáveis e sua influência na evolução clínica de cães com mastocitoma, assim como características epidemiológicas e clínicas do tumor e sua relação com a morte do animal. Observou-se relação entre arquitetura e grau segundo Kiupel (p= 2,907-6), mitose e grau segundo Patnaik (p=0,003). Animais SRD apresentaram mais neoplasias grau II (p=0,045), e a morte dos animais esteve relacionada com ulceração do tumor (p=0,006) e idade do animal (p=0,001). Houve associação entre quantidade de eosinófilos e extensão tumoral (p=0,221) e arquitetura (0,338); arquitetura e quantidade de mitoses (0,339); hemorragia e graduação segundo Patnaik (p=0,190). The laparoscopic lymphadenectomy is routinely performed in medicine, but not in veterinary medicine. This report describes a case of female canine with tumors in the left mammary chain (M4 and M5), which was subjected to intra-dermal application of sterile methylene blue, around the largest tumor (M5) which tried to demarcate the lymphatic vessels and regional lymph nodes. After 15 minutes, laparoscopic lymphadenectomy involving the abdominal in the left pelvic region started, followed by laparoscopic ovariohysterctomy (OVH) using three portals access. It was also performed an unilateral left total mastectomy. For histology, two abdominal lymph nodes were free of tumor cells. The patient had no recurrence in 60 days. [ABSTRACT FROM AUTHOR]
