4. Effectiveness of Online Capacity Building Programs in Wholistic Development of Faculties: An Empirical Analysis

Abstract

Purpose: Faculties have a vital role to play in ensuring that their graduates are trained with relevant skills. Formally arranged capacity building programs (CBPs) can aid in training and retraining the faculties with relevant contemporary skills so that they in turn can do justice to their students. The purpose of this study is to analyze the efficacy of such public-funded online CBPs in enhancing faculties' research and teaching capabilities. Design/methodology/approach: A scientifically designed questionnaire was sent to faculties who attended 12-day public-funded online capacity building workshop. Binomial logistic regression models were constructed to analyze as to how effective the online workshop was in enhancing teaching and research skills of the faculties involved. Findings: From the research study, the authors were able to infer that although faculties were able to understand theoretical concepts of qualitative nature relatively easily, the authors felt that its value addition in enhancing the research output was rather limited. The study also found that the faculties felt teaching concepts outside their "syllabus" to be counterintuitive. A significant finding of the study was that research had an important role in enhancing their teaching efficacy. Originality/value: Studies which are undertaken to test the efficacy of online capacity building workshops are scarce. This domain is going to gain importance in near future as technology is evolving at a rapid rate and online training of faculties helps in optimizing scarce resources.

Abstract

Autophagy is vital for maintaining cellular homeostasis by breaking down unnecessary organelles and proteins within cells. Its activity varies abnormally in several diseases, including cancer, making it a potential target for therapeutic strategies. The Wnt/β‐catenin signaling pathway significantly impacts cancer by stabilizing β‐catenin protein and promoting the transcription of its target genes. Therefore, we aimed to identify candidate substances targeting this signaling pathway. We designed and tested a thiouracil conjugate, discovering that TTP‐8 had anti‐tumor effects on human breast cancer cell lines MCF‐7 and MDA‐MB231. Our findings showed that TTP‐8 upregulated the expression of LC3 protein, a marker of autophagy in breast cancer cells, suggesting that TTP‐8 might induce autophagy. Further analysis confirmed an increase in autophagy‐related proteins, with consistent results obtained from flow cytometry and confocal microscopy. Interestingly, the induction of LC3 expression by TTP‐8 was even more pronounced in MCF‐7 and MDA‐MB231 cells transfected with β‐catenin siRNA. Thus, our research supports the idea that the Wnt/β‐catenin signaling pathway influences the regulation of autophagy‐related proteins, thereby inducing autophagy. Abstract

Whenever Indian Economy had to tackle significant inflationary pressure, Securities Exchange Board of India (SEBI), which is the apex regulator of capital and commodity markets, has time and again resorted to stop the trade in futures of essential agricultural commodities while allowing trade in futures of essential energy commodities. SEBI has justified the step on the grounds that, doing so prevents volatility in agricultural spot market. Our study tries to analyze the nature of correction in the two segments with the help of vector error correction model in the backdrop of inflationary and non-inflationary periods. In energy segment, among select commodities, the speed of error correction was 1to 2 days more as compared to non-inflationary period. With regards to commercial agricultural segment, the rate of error correction among select commodities was 4 to 7 days more as compared to non-inflationary period. Given the underdeveloped nature of agricultural futures market, SEBI's action seems bit too stringent. Although prior studies have been undertaken about Indian spot and derivative markets, empirical studies which have focused on analyzing economic rationale of SEBI's decision of restricting trade in agricultural futures during inflation are scarce. Copyright © 2018 Mohan, Bharathkumar, Dukanya, Rangappa, Shanmugam. Hepatocellular carcinoma (HCC) is a fatal disease and ranked fifth in cancer related mortality. Persistent activation of NF-?B is responsible for the oncogenesis, metastasis, tumor evasion, anti-apoptosis, angiogenesis and proliferation in HCC. Therefore, designing of chemically novel, biologically potent small molecules that target NF-?B signaling cascade have gained prominent clinical interest. Herein we synthesized a novel class of 4-(substituted)-2H-pyrido[3,2-b][1,4]oxazin-3(4H)-one by reacting 2H-pyrido[3,2-b][1,4]oxazin-3(4H)-one with various alkyl halides by using combustion derived bismuth oxide. We evaluated the antiproliferative efficacy of newly synthesized compounds against HCC cells and identified 4-(4-nitrobenzyl)-2H-pyrido[3,2-b][1,4]oxazin-3(4H)-one (NPO) as lead anticancer agent. In addition, we investigated the effect of NPO on the DNA binding ability of NF-?B and NF-?B regulated luciferase expression in HCC cells. The results demonstrated that NPO can induce significant growth inhibitory effects in HepG2, HCCLM3 and Huh-7 cells in dose and time-dependent manner. Interestingly, NPO induced significant downregulation in p65 DNA binding ability, p65 phosphorylation and subsequent expression of NF-?B dependent luciferase gene expression in diverse HCC cell lines. Further, in silico docking analysis suggested that NPO can show direct physical interaction with NF-?B. Finally, NPO was found to significantly abrogate tumor growth at a dose of 50 mg/kg in an orthotopic mouse model. Thus, we report the potential anticancer effects of NPO as a novel inhibitor of NF-?B signaling pathway in HCC.

Abstract

Aberrant activation of NF-κB is linked with the progression of human malignancies including hepatocellular carcinoma (HCC), and blockade of NF-κB signaling could be a potential target in the treatment of several cancers. Therefore, designing of novel small molecule inhibitors that target NF-κB activation is of prime importance in the treatment of several cancers. In the present work, we report the synthesis of series of 1,3,4-oxadiazoles, investigated their anticancer potential against HCC cells, and identified 2-(3-chlorobenzo[b]thiophen-2-yl)-5-(3-methoxyphenyl)-1,3,4-oxadiazole (CMO) as the lead compound. Further, we examined the effect of CMO on cell cycle distribution (flow cytometry), apoptosis (annexin V-propidium iodide-FITC staining), and phosphorylation of NF-κB signaling pathway proteins (IκB and p65) in HCC cells. We found that CMO induced antiproliferative effect in dose- and time-dependent manner. Also, CMO significantly increased the percentage of sub-G1 cell population and induced apoptosis. Furthermore, CMO found to decrease the phosphorylation of IκB (Ser 32) in the cytoplasmic extract and p65 (Ser 536) in the nuclear extract of HCC cells. It also abrogated the DNA binding ability and transcriptional activity of NF-κB. CMO induced the cleavage of PARP and caspase-3 in a time-dependent manner. In addition, transfection with p65 small interfering RNA blocks CMO-induced caspase-3/7 activation. Molecular docking analysis revealed that CMO interacts with the hydrophobic region of p65 protein. Thus, we are reporting CMO as an inhibitor of NF-κB signaling pathway.
