Abstract

Inflammation-related pathologies remain a serious health problem with high costs for the community. Citrus flavonoids are known to possess important pharmacological properties, including anti-inflammatory activity. In this study we evaluated the effects of a flavonoid-rich extract of orange juice (OJe) in an experimental model of enteritis induced by Vibrio anguillarum in adult zebrafish (Danio rerio). Administration of V. anguillarum through live feed (Artemia nauplii) for three consecutive days caused evident signs of enteritis in zebrafish. Three days of treatment with OJe before the pathogenic insult resulted in a remarkable reduction of tissue inflammatory events as well as a molecular down-regulation of the inflammatory genes such as IL-1β, IL-6 and TNFα. Our data suggest that OJe counteracts the inflammation of zebrafish intestinal mucosa, indicating that the pool of flavonoids present in orange juice could be useful for the prevention of enteritis. Abstract

Larval nutrition plays a key role in the development of a sustainable aquaculture where fish development, health and wellness are of prime importance. For some species, satisfactory growth and survival rates are met providing exclusively enriched rotifers and Artemia; however, feeding on copepods during the larval period has been shown to improve growth in both larval and juvenile fish, including seahorses. For the first time, the effects of different diets (Artemia and copepods) on the early development of juvenile seahorses (H. guttulatus and H. reidi) development were analysed by combining biometry, traditional histology and FPA-FTIR Imaging spectroscopy. Survival and growth and biochemical composition on the liver in seahorse were significantly affected by the type of diet offered. The results achieved were related to differences in the digestion of the two types of live preys, mainly dependent on their biochemical composition and permeability of the exoskeleton.

Abstract

Due to the undesirable consequences associated with antibiotics use, researchers and food producers have studied alternative feeding, for the control of fish diseases and animal welfare improvement. However, the beneficial properties of RNA yeasts extract in aquaculture have been barely considered. The present study investigated the beneficial properties of RNA yeast extract from Kluyveromyces fragilis on survival rate, weight and length as well as on molecular pathways involved in growth, immuno-system and oxidative stress using zebrafish (Danio rerio) as an experimental model. The yeast extract has been administrated to zebrafish at three different concentrations (60-180-300 ppm) via zooplankton (Artemia salina) for 21 days. Results highlighted yeast extract RNA capability to enhance growth and to improve larvae survival rate in a dose-dependent manner. In fact, gene expression data showed the ability of the RNA yeast extract to up-regulate genes involved in growth and to restore stress-related condition due to the early larval development coupled with the RNA yeast extract administration. In addition, gene expression showed that RNA yeast extract acts as inflammatory-reducer, but did not enhance the immune response. Histological analysis showed that all three treated groups displayed a thicker gut epithelium, higher intestinal crypts coupled with higher enterocytes lengths compared to control group. In conclusion, these findings provide a large gene network through which yeast RNA extract acts and, by inducing transcriptional changes, modulate the physiological control of growth and inflammatory response coupled with the increase in length and dry weight, together with a higher survival ratio and an intestinal architecture amelioration. AbstractDue to minimal environmental impact, compared to most conventional feed commodities, insects deserve a growing attention as candidate ingredients for aquafeeds. This study tested, for the first time during zebrafish larval rearing, the effects of an increasing replacement (0%–25%–50%) of fish meal by black soldier fly (BSF) full-fat prepupae meal. All diets were formulated to be isonitrogenous and isolipidic. A multidisciplinary approach, including biometrics, histology, gas chromatography–mass spectrometry, and molecular analyses, was applied to better understand the biological responses of larval zebrafish to the different partial inclusions of BSF in the feed. Generally, results are promising, but a 50% of BSF meal inclusion in the diet affected both lipid composition and accumulation in the larvae.

Abstract

Simple Summary: Aquaculture accounts for 52% of the world's production of fish for human consumption and its growth relies on finding new alternative protein sources to fishmeal to use in aquafeeds. To make fish feed more sustainable, insects have emerged as valuable alternatives due to their high nutritional value. In this context, black soldier fly (BSF) meals have been used to replace, partially or totally, fishmeal in aquafeeds. However, due to relevant nutritional peculiarities of BSF developmental stages, it is crucial to understand how they can affect fish growth performance and health status. In this study, we evaluated the effects of total dietary replacement of fishmeal with defatted BSF larvae and prepupae meals on the growth performance of zebrafish from larva, through adult stages, in a 60-day feeding trial. Moreover, the effects of the two BSF meals on the expression of genes involved in growth, stress- and immune- responses, as well as hydrolysis of chitin were investigated. Overall, our results showed the beneficial role of defatted BSF meals in zebrafish diet, especially regarding prepupae meal, suggesting that black soldier fly meals could totally replace fishmeal without negatively affecting growth and wellness of zebrafish. The black soldier fly (BSF) Hermetia illucens is receiving increasing attention as a sustainable fishmeal alternative protein source for aquaculture. To date, no studies have explored the effects of fishmeal replacement with BSF V instar larvae or prepupae meals due to their peculiar nutritional properties on fish performances. This study investigated the effects of 100% replacement of fishmeal (control diet) with defatted BSF meals (V instar larvae and prepupae meals, treatments) on growth performance and welfare of zebrafish (Danio rerio), from larvae to adults, in a 60-day feeding trial. Following the inclusion of BSF meals, the expression of key genes involved in growth (igf1, igf2, mstnb, myod1, myog, myf5), hydrolysis of chitin (chia.2, chia.3, chia.5), immune- (il1b, il6, tnfα), and stress- (hsp70 and nr3c1) responses, as assessed by qPCR, was modulated in all of the molecular pathways, except for the stress response. Overall, our findings showed that both BSF meals can totally replace fishmeal without adverse impacts on adult zebrafish growth parameters (final total and standard length, final body weight, weight gain, daily growth rate, specific growth rate) and welfare, with BSF prepupae meal inducing the most beneficial effects, thus suggesting their potential application to meet fish requirements in aquaculture. Abstract

To evaluate the effects of feeding a Hermetia illucens (HI) larvae meal on the different intestinal traits of hens, and to determine the toxic elements&rsquo, concentration in the insect meal and diets, 162 hens were randomly allotted to three groups. The control received a corn-soybean meal-based diet (SBM), the HI25 and HI50 groups received two diets in which the 25% and 50% of the dietary protein were replaced by the HI protein, respectively. The duodenal and jejunal villi height and villi/crypt were higher (p &lt, 0.01) in the SBM than in the HI groups. The ileal villi height was higher (p &lt, 0.05) in the SBM and HI25 groups than the HI50. The HI50 group exhibited a lower duodenal maltase activity. The intestinal alkaline phosphatase (IAP) activity linearly decreased in the duodenum and jejunum as the dietary insect meal inclusion increased. The HI50 group had a higher acetate and butyrate level than the SBM. The levels of cadmium (Cd), lead (Pb), mercury (Hg), and arsenic (As) in the diets and insect meal were lower than the maximum values established by the EU Commission. The 25% soybean protein replacement with Hermetia illucens larvae meal in the diet of laying hens was more suitable and closer to the optimal level than 50%.
