In today's digitally driven landscape, robust Information Technology (IT) risk assessment practices are essential for safeguarding systems, digital communication, and data. This paper introduces 'AssessITS', an actionable method designed to provide organizations with comprehensive guidelines for conducting IT and cybersecurity risk assessments. Drawing extensively from NIST 800-30 Rev 1, COBIT 5, and ISO 31000, 'AssessITS' bridges the gap between high-level theoretical standards and practical implementation challenges. The paper outlines a step-by-step methodology that organizations can simply adopt to systematically identify, analyze, and mitigate IT risks. By simplifying complex principles into actionable procedures, this framework equips practitioners with the tools needed to perform risk assessments independently, without too much reliance on external vendors. The guidelines are developed to be straightforward, integrating practical evaluation metrics that allow for the precise quantification of asset values, threat levels, vulnerabilities, and impacts on confidentiality, integrity, and availability. This approach ensures that the risk assessment process is not only comprehensive but also accessible, enabling decision-makers to implement effective risk mitigation strategies customized to their unique operational contexts. The Machine learning (ML) is a rapidly evolving field of technology that has the potential to greatly impact society in a variety of ways. However, there are also concerns about the potential negative effects of ML on society, such as job displacement and privacy issues. This research aimed to conduct a comprehensive analysis of the current and future impact of ML on society. The research included a thorough literature review, case studies, and surveys to gather data on the economic impact of ML, ethical and privacy implications, and public perceptions of the technology. The survey was conducted on 150 respondents from different areas. The case studies conducted were on the impact of ML on healthcare, finance, transportation, and manufacturing. The findings of this research revealed that the majority of respondents have a moderate level of familiarity with the concept of ML, believe that it has the potential to benefit society, and think that society should prioritize the development and use of ML. Based on these findings, it was recommended that more research is conducted on the impact of ML on society, stronger regulations and laws to protect the privacy and rights of individuals when it comes to ML should be developed, transparency and accountability in ML decision-making processes should be increased, and public education and awareness about ML should be enhanced., Comment: 12 pages

Protein-protein interactions (PPIs) are critical for various biological processes, and understanding their dynamics is essential for decoding molecular mechanisms and advancing fields such as cancer research and drug discovery. Mutations in PPIs can disrupt protein binding affinity and lead to functional changes and disease. Predicting the impact of mutations on binding affinity is valuable but experimentally challenging. Computational methods, including physics-based and machine learning-based approaches, have been developed to address this challenge. Machine learning-based methods, fueled by extensive PPI datasets such as Ab-Bind, PINT, SKEMPI, and others, have shown promise in predicting binding affinity changes. However, accurate predictions and generalization of these models across different datasets remain challenging. Geometric graph learning has emerged as a powerful approach, combining graph theory and machine learning, to capture structural features of biomolecules. We present GGL-PPI, a novel method that integrates geometric graph learning and machine learning to predict mutation-induced binding free energy changes. GGL-PPI leverages atom-level graph coloring and multi-scale weighted colored geometric subgraphs to extract informative features, demonstrating superior performance on three validation datasets, namely AB-Bind, SKEMPI 1.0, and SKEMPI 2.0 datasets. Evaluation on a blind test set highlights the unbiased predictions of GGL-PPI for both direct and reverse mutations. The findings underscore the potential of GGL-PPI in accurately predicting binding free energy changes, contributing to our understanding of PPIs and aiding drug design efforts.

Understanding and accurately predicting protein-ligand binding affinity are essential in the drug design and discovery process. At present, machine learning-based methodologies are gaining popularity as a means of predicting binding affinity due to their efficiency and accuracy, as well as the increasing availability of structural and binding affinity data for protein-ligand complexes. In biomolecular studies, graph theory has been widely applied since graphs can be used to model molecules or molecular complexes in a natural manner. In the present work, we upgrade the graph-based learners for the study of protein-ligand interactions by integrating extensive atom types such as SYBYL and extended connectivity interactive features (ECIF) into multiscale weighted colored graphs (MWCG). By pairing with the gradient boosting decision tree (GBDT) machine learning algorithm, our approach results in two different methods, namely $^\text{sybyl}\text{GGL}$-Score and $^\text{ecif}\text{GGL}$-Score. Both of our models are extensively validated in their scoring power using three commonly used benchmark datasets in the drug design area, namely CASF-2007, CASF-2013, and CASF-2016. The performance of our best model $^\text{sybyl}\text{GGL}$-Score is compared with other state-of-the-art models in the binding affinity prediction for each benchmark. While both of our models achieve state-of-the-art results, the SYBYL atom-type model $^\text{sybyl}\text{GGL}$-Score outperforms other methods by a wide margin in all benchmarks.

Molecular surface representations have been advertised as a great tool to study protein structure and functions, including protein-ligand binding affinity modeling. However, the conventional surface-area-based methods fail to deliver a competitive performance on the energy scoring tasks. The main reason is the lack of crucial physical and chemical interactions encoded in the molecular surface generations. We present novel molecular surface representations embedded in different scales of the element interactive manifolds featuring the dramatically dimensional reduction and accurately physical and biological properties encoders. Those low-dimensional surface-based descriptors are ready to be paired with any advanced machine learning algorithms to explore the essential structure-activity relationships that give rise to the element interactive surface area-based scoring functions (EISA-score). The newly developed EISA-score has outperformed many state-of-the-art models, including various well-established surface-related representations, in standard PDBbind benchmarks.

Faces and their expressions are one of the potent subjects for digital images. Detecting emotions from images is an ancient task in the field of computer vision; however, performing its reverse -- synthesizing facial expressions from images -- is quite new. Such operations of regenerating images with different facial expressions, or altering an existing expression in an image require the Generative Adversarial Network (GAN). In this paper, we aim to change the facial expression in an image using GAN, where the input image with an initial expression (i.e., happy) is altered to a different expression (i.e., disgusted) for the same person. We used StarGAN techniques on a modified version of the MUG dataset to accomplish this objective. Moreover, we extended our work further by remodeling facial expressions in an image indicated by the emotion from a given text. As a result, we applied a Long Short-Term Memory (LSTM) method to extract emotion from the text and forwarded it to our expression-altering module. As a demonstration of our working pipeline, we also create an application prototype of a blog that regenerates the profile picture with different expressions based on the user's textual emotion., Comment: Accepted in VISAPP2022

A new preconditioner based on a block $LDU$ factorization with algebraic multigrid subsolves for scalability is introduced for the large, structured systems appearing in implicit Runge-Kutta time integration of parabolic partial differential equations. This preconditioner is compared in condition number and eigenvalue distribution, and in numerical experiments with others in the literature: block Jacobi, block Gauss-Seidel, and the optimized block Gauss-Seidel method of Staff, Mardal, and Nilssen [{\em Modeling, Identification and Control}, 27 (2006), pp. 109-123]. Experiments are run on two test problems, a $2D$ heat equation and a model advection-diffusion problem, using implicit Runge-Kutta methods with two to seven stages. We find that the new preconditioner outperforms the others, with the improvement becoming more pronounced as spatial discretization is refined and as temporal order is increased., Comment: 20 pages
