Abstract Chronic kidney disease (CKD) poses a significant global health dilemma, emerging from complex causes. Although our prior research has indicated that a deficiency in Reticulon-3 (RTN3) accelerates renal disease progression, a thorough examination of RTN3 on kidney function and pathology remains underexplored. To address this critical need, we generated Rtn3-null mice to study the consequences of RTN3 protein deficiency on CKD. Single-cell transcriptomic analyses were performed on 47,885 cells from the renal cortex of both healthy and Rtn3-null mice, enabling us to compare spatial architectures and expression profiles across 14 distinct cell types. Our analysis revealed that RTN3 deficiency leads to significant alterations in the spatial organization and gene expression profiles of renal cells, reflecting CKD pathology. Specifically, RTN3 deficiency was associated with Lars2 overexpression, which in turn caused mitochondrial dysfunction and increased reactive oxygen species levels. This shift induced a transition in renal epithelial cells from a functional state to a fibrogenic state, thus promoting renal fibrosis. Additionally, RTN3 deficiency was found to drive the endothelial-to-mesenchymal transition process and disrupt cell-cell communication, further exacerbating renal fibrosis. Immunohistochemistry and Western-Blot techniques were used to validate these observations, reinforcing the critical role of RTN3 in CKD pathogenesis. The deficiency of RTN3 protein in CKD leads to profound changes in cellular architecture and molecular profiles. Our work seeks to elevate the understanding of RTN3’s role in CKD’s narrative and position it as a promising therapeutic contender.

ObjectivesWe investigate the impact of wildfire disasters on cognitive health (i.e., thinking and language skills) in individuals aged 10 years and older using 2014 and 2018 wildfire and cognitive ability survey data from China.MethodsWe distinguished wildfires in each county at different wind directions each day by exogenous wind direction changes, and analyzed the effects of wildfires on cognitive abilities through upwind and non-upwind wildfires.ResultsOur analysis shows that for every 10-unit increase in upwind wildfires compared to non-upwind wildfires, respondents’ scores on word and math tests decrease by 0.235 and 0.236 standard deviations, respectively. Furthermore, we find that the impact of wildfire on cognitive ability is more pronounced in younger individuals, and those with lower defensive expenditures experience more severe impacts. Additionally, wildfires negatively affect individuals’ cognitive abilities by generating air pollution.ConclusionWildfires significantly reduce individuals’ cognitive abilities. Two recommendations are as follows: 1) governments should follow the principle of green development, introduce relevant regulations, and increase investment in adaptive technologies. 2) Individuals should raise awareness of climate hazards preparedness and strengthen defensive protection.

Objective: This study presents a detailed analysis of the clinical and genetic characteristics of uterine leiomyoma associated with Hereditary Leiomyomatosis and Renal Cell Cancer (HLRCC), combined with exploration of family history, pathology, and management procedures, supported by thorough evidence collection. Methods: Blood samples were collected from the proband, and the pathogenic variant was verified using Sanger sequencing. A comprehensive review of family history, FH deficiency pathology, FH and 2SC immunohistochemistry staining was conducted. Functional evidence was derived from clinical and genetic information, supplemented by a literature collection and mutation was reclassified based on ACMG/AMP guidelines. Results: The study successfully identifies a novel missense mutation (c.1240A>G; p.Lys414Glu) in exon 9 of FH, with no prior reports in existing databases. The patient's phenotype and family history, coupled with evidence collected from the literature, contribute to the preliminary determination of the variant as likely pathogenic. We also emphasize that the importance of combining FH-deficient morphology and immunohistochemical staining with 2SC for enhanced sensitivity. Conclusion: This research adds a novel missense mutation to the repertoire of FH gene variants, emphasizing its likely pathogenic nature based on a multidimensional analysis of phenotype, family history, and literature evidence. The findings enhance our understanding of the genetic landscape associated with FH and underscore the importance of thorough characterization for accurate variant classification.

The effect of Heamatoccus pluvialis on astaxanthin accumulation, nutrition, antioxidant activity, and gut health in juvenile crucian carp was investigated based on two ratios of alga powder in the feed (1:1 and 2:1). After 93 days of feeding, various organs and tissues of crucian carp, e.g., serum, scale, skin, muscle, kidney, and gut liver, were isolated and tested for astaxanthin content, fatty acids, amino acids, intestinal microbiota, and physiological and biochemical indicators. The results showed that among the various types of tissues, the scales and guts were revealed with the highest contents of astaxanthin (higher than 45.00 μg/g), and 2.35 μg/g in muscles in the fish. Although the compositions of astaxanthin in the fish tissues were mainly dependent on the contents of astaxanthin in the feed, the different tissues in the fish were selective in their uptake of astaxanthin isomers. The astaxanthin isomerization occurred in the fish, as suggested by the presence of two astaxanthin isomers (i.e., 13-Z astaxanthin and 9-Z astaxanthin) which were not available in the feed. The nutritional value of crucian carp muscles was increased by the elevated contents of essential amino acids and polyunsaturated fatty acids. Compared to the control group, the fish fed with H. pluvialis was revealed with significant changes in biochemical parameters and gut microbial compositions. Both the levels of antioxidant activity, as indicated by the blood total antioxidant capacity and the contents of catalase, malondialdehyde, glutathione peroxidase, and superoxide dismutase, and the antimicrobial activity, as indicated by the content of lysozyme, were significantly increased. In the gut, the beneficial bacteria (e.g., Lactobacillus and Lactococcus) became the dominant species, while the abundance of pathogenic bacteria (e.g., Xanthobacter, Legionella, and Gordonella) was significantly decreased. This study provided strong experimental evidence to support the application of H. pluvialis in the feed for crucian carp as an effective way to improve the nutritional value and immune activity of crucian carp, greatly promoting the healthy farming and food safety of crucian carp.

Under the influence of multiple uncertain factors at home and abroad, urban amenities, as the underlying support for urban renewal activities, are of great significance in enhancing urban economic resilience. The panel data of Chinese cities from 2011 to 2019 is used in this study. Urban amenity is measured from artificial amenities and climate amenities, respectively. By using a two-way fixed effects model, we empirically test the impact of urban amenities on urban economic resilience. The key findings of this study are as follows. (1) Urban amenities can significantly enhance urban economic resilience. (2) Heterogeneity analysis shows that there are regional differences in the role of urban amenities in promoting urban economic resilience, with cities in the eastern region, strong environmental regulations, and high urbanization rates benefiting more. (3) We further find that urban amenities mainly enhance economic resilience by promoting population agglomeration, attracting labor migration, improving the quality of human capital, and stimulating urban innovation. Our conclusions recommend to rationally allocate and optimize urban amenity resources, strengthen urban planning and construction management, and create a more livable urban environment, thereby enhancing urban economic resilience.

Objective To explore the prognostic value of serum levels of miR-155 and Smad5 in hemodialysis (HD) patients with acute renal failure (ARF). Methods From August 2018 to July 2019, 120 ARF patients on HD were selected as study group while another 116 healthy subjects as control group; the serum expressions of miR-155 and Smad5 mRNA were detected by quantitative reverse transcriptase-mediated polymerase chain reaction (qRT-PCR). Pearson’s method was utilized for examining the correlation between serum expression levels of miR-155 and Smad5 mRNA and the levels of creatinine and blood urea nitrogen (BUN); Kaplan-Meier method was employed for examining the survival of HD patients with ARF; COX regression for analyzing the prognostic factors; receiver operating characteristic curve (ROC) curve for assessing the predictive value of serum miR-155 and Smad5 mRNA levels. Abstract Micro/nanorobots are promising for a wide range of biomedical applications (such as targeted tumor, thrombus, and infection therapies in hard‐to‐reach body sites) because of their tiny size and high maneuverability through the actuation of external fields (e.g., magnetic field, light, ultrasound, electric field, and/or heat). However, fully synthetic micro/nanorobots as foreign objects are susceptible to phagocytosis and clearance by diverse phagocytes. To address this issue, researchers have attempted to develop various cytomembrane‐camouflaged micro/nanorobots by two means: (1) direct coating of micro/nanorobots with cytomembranes derived from living cells and (2) the swallowing of micro/nanorobots by living immunocytes via phagocytosis. The camouflaging with cytomembranes or living immunocytes not only protects micro/nanorobots from phagocytosis, but also endows them with new characteristics or functionalities, such as prolonging propulsion in biofluids, targeting diseased areas, or neutralizing bacterial toxins. In this review, we comprehensively summarize the recent advances and developments of cytomembrane‐camouflaged medical micro/nanorobots. We first discuss how cytomembrane coating nanotechnology has been employed to engineer synthetic nanomaterials, and then we review in detail how cytomembrane camouflage tactic can be exploited to functionalize micro/nanorobots. We aim to bridge the gap between cytomembrane‐cloaked micro/nanorobots and nanomaterials and to provide design guidance for developing cytomembrane‐camouflaged micro/nanorobots.

Jasminum sambac (jasmine flower), a world-renowned plant appreciated for its exceptional flower fragrance, is of cultural and economic importance. However, the genetic basis of its fragrance is largely unknown. Here, we present the first de novo genome assembly of J. sambac with 550.12 Mb (scaffold N50 = 40.10 Mb) assembled into 13 pseudochromosomes. Terpene synthase (TPS) genes associated with flower fragrance are considerably amplified in the form of gene clusters through tandem duplications in the genome. Gene clusters within the salicylic acid/benzoic acid/theobromine (SABATH) and benzylalcohol O-acetyltransferase/anthocyanin O-hydroxycinnamoyltransferases/anthranilate N-hydroxycinnamoyl/benzoyltransferase/deacetylvindoline 4-O-acetyltransferase (BAHD) superfamilies were identified to be related to the biosynthesis of phenylpropanoid/benzenoid compounds. Several key genes involved in jasmonate biosynthesis were duplicated, causing an increase in copy numbers. In addition, multi-omics analyses identified various aromatic compounds and many genes involved in fragrance biosynthesis pathways. Furthermore, the roles of JsTPS3 in β-ocimene biosynthesis, as well as JsAOC1 and JsAOS in jasmonic acid biosynthesis, were functionally validated. The genome assembled in this study for J. sambac offers a basic genetic resource for studying floral scent and jasmonate biosynthesis, and provides a foundation for functional genomic research and variety improvements in Jasminum.

The physics potential of detecting ^8 B solar neutrinos will be exploited at the Jiangmen Underground Neutrino Observatory (JUNO), in a model-independent manner by using three distinct channels of the charged current (CC), neutral current (NC), and elastic scattering (ES) interactions. Due to the largest-ever mass of ^13 C nuclei in the liquid scintillator detectors and the expected low background level, ^8 B solar neutrinos are observable in the CC and NC interactions on ^13 C for the first time. By virtue of optimized event selections and muon veto strategies, backgrounds from the accidental coincidence, muon-induced isotopes, and external backgrounds can be greatly suppressed. Excellent signal-to-background ratios can be achieved in the CC, NC, and ES channels to guarantee the observation of the ^8 B solar neutrinos. From the sensitivity studies performed in this work, we show that JUNO, with 10 yr of data, can reach the 1 σ precision levels of 5%, 8%, and 20% for the ^8 B neutrino flux, ${\sin }^{2}{\theta }_{12}$ , and ${\rm{\Delta }}{m}_{21}^{2}$ , respectively. Probing the details of both solar physics and neutrino physics would be unique and helpful. In addition, when combined with the Sudbury Neutrino Observatory measurement, the world's best precision of 3% is expected for the measurement of the ^8 B neutrino flux.

The Heat Shock Transcription Factor (Hsf) serves as a critical transcription factor in plants, exerting its function by binding to specific promoter regions in response to heat stress. This activation leads to the up-regulation of heat shock proteins. Taraxacum kok-saghyz Rodin (TKS) is a plant species that produces high quality natural rubber in its root latex. During the growth of TKS, various stresses can affect its defense mechanisms and overall growth. However, studies investigating the Hsf transcription factors in TKS remain limited. In this study, we identified and named 31 Hsf transcription factors in TKS and performed a comparative analysis of their core motifs and gene structures. Based on phylogenetic analysis, these transcription factors were classified into three distinct classes. By cis-element analysis of the TKS promoter genes, we gained valuable insights into the evolutionary characteristics of the TKS Hsf gene family. Transcriptome expression profiling data revealed different expression of various genes within the same tissues. Several TKS Hsf genes exhibited responsiveness to various abiotic stresses and hormonal treatments, indicating their involvement in regulating the plant's response to different environmental conditions. Overall, the Hsf gene family in TKS plays a critical role in plant growth, development, and adaptation to abiotic stresses. This study provides a basis for further functional validation of TKS Hsf genes and elucidation of the regulatory mechanisms of target genes.

Tumor-associated macrophages (TAMs) are key components of tumor immune microenvironment and play a dual role in promoting tumor growth and anti-tumor immunity. Therefore, regulating TAMs has become a promising method in cancer immunotherapy. NF- κB pathway is the key regulatory pathway of TAMs. Targeting this pathway has shown the potential to improve tumor immune microenvironment. At present, there are still some controversies and the idea of combined therapy in this field. This article reviews the progress in the field of immunotherapy in improving tumor immune microenvironment by exploring the mechanism of regulating TAMs (including promoting M1 polarization, inhibiting M2 polarization and regulating TAMs infiltration).

Abstract Main goal of the JUNO experiment is to determine the neutrino mass ordering using a 20 kt liquid-scintillator detector. Its key feature is an excellent energy resolution of at least 3% at 1 MeV, for which its instruments need to meet a certain quality and thus have to be fully characterized. More than 20,000 20-inch PMTs have been received and assessed by JUNO after a detailed testing program which began in 2017 and elapsed for about four years. Based on this mass characterization and a set of specific requirements, a good quality of all accepted PMTs could be ascertained. This paper presents the performed testing procedure with the designed testing systems as well as the statistical characteristics of all 20-inch PMTs intended to be used in the JUNO experiment, covering more than fifteen performance parameters including the photocathode uniformity. This constitutes the largest sample of 20-inch PMTs ever produced and studied in detail to date, i.e. 15,000 of the newly developed 20-inch MCP-PMTs from Northern Night Vision Technology Co. (NNVT) and 5000 of dynode PMTs from Hamamatsu Photonics K. K.(HPK).

Abstract The structure of the human vitreous varies considerably because of age-related liquefactions of the vitreous gel. These changes are poorly studied in vivo mainly because their high transparency and mobility make it difficult to obtain reliable and repeatable images of the vitreous. Optical coherence tomography can detect the boundaries between the vitreous gel and vitreous fluid, but it is difficult to obtain high resolution images that can be used to convert the images to three-dimensional (3D) images. Thus, the purpose of this study was to determine the shape and characteristics of the vitreous fluid using machine learning-based 3D modeling in which manually labelled fluid areas were used to train deep convolutional neural network (DCNN). The trained DCNN labelled vitreous fluid automatically and allowed us to obtain 3D vitreous model and to quantify the vitreous fluidic cavities. The mean volume and surface area of posterior vitreous fluidic cavities are 19.6 ± 7.8 mm3 and 104.0 ± 18.9 mm2 in eyes of 17 school children. The results suggested that vitreous fluidic cavities expanded as the cavities connects with each other, and this modeling system provided novel imaging markers for aging and eye diseases.

Abstract We study damping signatures at the Jiangmen Underground Neutrino Observatory (JUNO), a medium-baseline reactor neutrino oscillation experiment. These damping signatures are motivated by various new physics models, including quantum decoherence, ν 3 decay, neutrino absorption, and wave packet decoherence. The phenomenological effects of these models can be characterized by exponential damping factors at the probability level. We assess how well JUNO can constrain these damping parameters and how to disentangle these different damping signatures at JUNO. Compared to current experimental limits, JUNO can significantly improve the limits on τ 3/m 3 in the ν 3 decay model, the width of the neutrino wave packet σ x , and the intrinsic relative dispersion of neutrino momentum σ rel.

Based on 2010-2019 Chinese logistics listed companies as research samples, the paper used the binary Logit model measuring degree of financing constraints. The Kernel density function and Markov chain model are used to forecast China listed companies financing logistics dynamic constraints and business performance growth. Furthermore the stock of knowledge was chosen as a threshold variable to explore the impact of financing constraints on corporate performance growth of listed logistics enterprises. We find that the degree of financing constraints of logistics enterprises in our country has not been significantly eased. Corporate performance has not changed significantly and there are no obvious spatial gap and polarization with the passage of time. The impact of financing constraints on the corporate performance growth of logistics enterprises in China has a double threshold effect of knowledge stock, and has an inhibitory effect that first increases and then decreases. This is because in the short term, the investment of knowledge stock by enterprises can crowd out more corporate liquidity, and in the long run, it is related to the conversion rate of the knowledge stock itself. Because of the uneven regional distribution of resources and differences in the degree of economic development, there is a growing disincentive effect in central China as the stock of knowledge accumulates.

Objective Removing the apocrine sweat glands with nasal endoscope assisted suction cutter is a new technique that we have explored for the treatment of axillary odors. The aim is to introduce this new technique, compare it with traditional treatment and study the efficacy and patients satisfaction it brought about. Method A total of 82 patients (164 sides) with axillary odor were treated, 49 patients (98 sides) were treated with subcutaneou endoscopic assisted suction cutter, and 33 patients (66 sides) were treated with small-cut trimming. The efficacy of postoperative recovery was evaluated according to Tung grading method, and postoperative complications and satisfaction rate were also included in the comparative evaluation. Climate change and land-use change are making landscape fires worse, causing them to grow in intensity and spread in range across Earth’s ecosystems. Extreme landscape fires can be devastating to people, ecosystems, and sociology. However, most research on landscape fires has not considered their potential impact on the economy, particularly with regard to entrepreneurial activity. Entrepreneurial activity includes the entry of new markets and the creation of new products or services, thereby facilitating the creation and expansion of economic activity. This manuscript empirically analyzes the impact of landscape fires on entrepreneurial activity, based on satellite monitoring data of landscape fires in China from 2014 to 2018. Different wind direction models and instrumental variable methods are used for empirical analysis. The results of the analysis show, first, that an increase in landscape fires in a county can significantly reduce local entrepreneurial activity. We further adopt the wind direction approach and instrumental variable approach to deal with potential endogeneity issues, and the regression results are consistent. Second, compared to eastern or high-economic-development areas, central and western or low-economic-development areas are more susceptible to exogenous landscape fires. Third, landscape fires have a negative impact on entrepreneurial activity through increasing air pollution, damaging human health, increasing risk aversion, and reducing the labor supply. It is important for both the government and the public to fully recognize the potential dangers that landscape fires pose to corporate behavior. This awareness can help reduce the impact of natural disasters such as landscape fires, protect the ecological environment, and provide solid support for corporate investment and regional development.

Background: Chronic kidney disease, a global public health problem, results in kidney damage or a gradual decline in the glomerular filtration rate. Alport syndrome is commonly characterized by chronic glomerulonephritis caused by a structural disorder in the glomerular basement membrane. Currently, three disease-causing genes, namely collagen type IV alpha 3–5 (COL4A3, COL4A4, and COL4A5), have been associated with the occurrence of Alport syndrome.Methods: We enrolled a Chinese family where the affected individuals suffered from recurrent hematuria and proteinuria. The proband was selected for whole-exome sequencing to identify the pathogenic mutations in this family.Results: After data filtering, a novel heterozygous COL4A4 variant (NM_000092: c.853G>A/p. G285A) was identified as the putative genetic lesion in the affected individuals. Further co-segregation analysis using Sanger sequencing confirmed that this novel COL4A4 mutation (c.853G>A/p. G285A) exists only in the affected individuals and is absent in other healthy family members as well as in the control cohort of 200 individuals from the same locality. According to American College of Medical Genetics and Genomics guidelines, the mutation was classified as ‘potentially pathogenic’. A bioinformatics-based prediction analysis revealed that this mutation is pathogenic and may disrupt the structure and function of type IV collagen. This variant is located at an evolutionarily conserved site of COL4A4.Conclusion: In this study, we identified a novel heterozygous COL4A4 variant (c.853G>A) in a Chinese AS family and assisted to diagnose this AS proband as autosomal-dominant Alport syndrome (ADAS). Our study expands the spectrum of Alport syndrome mutations and contributes to the genetic counseling and diagnosis of patients with Alport syndrome.

The ability of bioactive peptides to exert biological functions has mainly contributed to their exploitation. The exploitation and utilization of these peptides have grown tremendously over the past two decades. Food-derived peptides from sources such as plant, animal, and marine proteins and their byproducts constitute a more significant portion of the naturally-occurring peptides that have been documented. Due to their high specificity and biocompatibility, these peptides serve as a suitable alternative to pharmacological drugs for treating non-communicable diseases (such as cardiovascular diseases, obesity, and cancer). They are helpful as food preservatives, ingredients in functional foods, and dietary supplements in the food sector. Despite their unique features, the application of these peptides in the clinical and food sector is to some extent hindered by their inherent drawbacks such as toxicity, bitterness, instability, and susceptibility to enzymatic degradation in the gastrointestinal tract. Several strategies have been employed to eliminate or reduce the disadvantages of peptides, thus enhancing the peptide bioactivity and broadening the opportunities for their applications. This review article focuses on the current research status of various bioactive peptides and the strategies that have been implemented to overcome their disadvantages. It will also highlight future perspectives regarding the possible improvements to be made for the development of bioactive peptides with practical uses and their commercialization.

Background: Head and neck squamous cell carcinoma (HNSCC) is a common malignancy of the mucosal epithelium of the oral cavity, pharynx, and larynx. Laryngeal squamous cell carcinoma (LSCC) and oral squamous cell carcinoma are common HNSCC subtypes. Patients with metastatic HNSCC have a poor prognosis. Therefore, identifying molecular markers for the development and progression of HNSCC is essential for improving early diagnosis and predicting patient outcomes.Methods: Gene expression RNA-Seq data and patient clinical traits were obtained from The Cancer Genome Atlas-Head and Neck Squamous Cell Carcinoma (TCGA-HNSC) and Gene Expression Omnibus databases. Differentially expressed gene (DEG) screening was performed using the TCGA-HNSC dataset. Intersection analysis between the DEGs and a list of core matrisome genes obtained from the Matrisome Project was used to identify differentially expressed matrisome genes. A prognostic model was established using univariate and multivariate Cox regression analyses, least absolute shrinkage, and selection operator (LASSO) regression analysis. Immune landscape analysis was performed based on the single-sample gene set enrichment analysis algorithm, Gene Ontology, Kyoto Encyclopedia of Genes and Genomes, prognostic value, receiver operating characteristic curve analysis, and gene mutation analyses. Immunohistochemical results regarding prognostic protein levels were obtained from the Human Protein Atlas. Single-gene RNA-sequencing data were obtained from GSE150321 and GSE172577 datasets. CCK-8 and Transwell assays were used to confirm cell proliferation and migration.Results: A total of 1,779 DEGs, including 939 upregulated and 840 downregulated genes, between tumor and normal samples were identified using the TCGA-HNSC microarray data. Intersection analysis revealed 52 differentially expressed matrisome-related genes. After performing univariate and multivariate Cox regression and LASSO analyses, a novel prognostic model based on six matrisome genes (FN1, LAMB4, LAMB3, DMP1, CHAD, and MMRN1) for HNSCC was established. This risk model can successfully predict HNSCC survival. The high-risk group had worse prognoses and higher enrichment of pathways related to cancer development than the low-risk group. Silencing LAMB4 in HNSCC cell lines promoted cell proliferation and migration.Conclusion: This study provides a novel prognostic model for HNSCC. Thus, FN1, LAMB4, LAMB3, DMP1, CHAD, and MMRN1 may be the promising biomarkers for clinical practice.

Nowadays, great efforts have been spent on addressing concerns over energy and environmental crises. Among these efforts, electrocatalysis is widely recognized and studied for its high efficiency and easy processability. As a class of emerging electrocatalysts, metal aerogels (MAs) stand out in the last decade. In virtue of their three-dimensional conductive pathways, their library of catalytically/optically active sites, and their robust network structures, MAs have unique advantages in electrocatalysis. However, due to the short history of MAs, there is insufficient research on them, leaving significant room for material design and performance optimization. This perspective will mainly focus on electrocatalysis with MAs, aiming to summarize the state-of-the-art progress and to guide the on-target design of efficient MAs-based electrocatalysts towards energy- and environment-related applications.

The rapid development of medical micromotors draws a beautiful blueprint for the noninvasive or minimally invasive diagnosis and therapy. By combining stimuli-sensitive hydrogel materials, micromotors are bestowed with new characteristics such as stimuli-responsive shape transformation/morphing, excellent biocompatibility and biodegradability, and drug loading ability. Actuated by chemical fuels or external fields (e.g., magnetic field, ultrasound, light, and electric field), hydrogel-based stimuli-responsive (HBSR) micromotors can be utilized to load therapeutic agents into the hydrogel networks or directly grip the target cargos (e.g., drug-loaded particles, cells, and thrombus), transport them to sites of interest (e.g., tumor area and diseased tissues), and unload the cargos or execute a specific task (e.g., cell capture, targeted sampling, and removal of blood clots) in response to a stimulus (e.g., change of temperature, pH, ion strength, and chemicals) in the physiological environment. The high flexibility, adaptive capacity, and shape morphing property enable the HBSR micromotors to complete specific medical tasks in complex physiological scenarios, especially in confined, hard-to-reach tissues, and vessels of the body. Herein, this review summarizes the current progress in hydrogel-based medical micromotors with stimuli responsiveness. The thermo-responsive, photothermal-responsive, magnetocaloric-responsive, pH-responsive, ionic-strength-responsive, and chemoresponsive micromotors are discussed in detail. Finally, current challenges and future perspectives for the development of HBSR micromotors in the biomedical field are discussed.

Here the authors describe the role of reductants, unlocking ligand chemistry, and largely expanding the composition space of noble metal aerogels for high-performance electrocatalysis.

Chronic obstructive pulmonary disease (COPD) is associated with morphologic abnormalities of airways with various patterns and severities. However, the way of effectively representing these abnormalities is lacking and whether these abnormalities enable to distinguish COPD from healthy controls is unknown. We propose to use deep convolutional neural network (CNN) to assess 3D lung airway tree from the perspective of computer vision, thereby constructing models of identifying COPD. After extracting airway trees from CT images, snapshots of their 3D visualizations are obtained from ventral, dorsal and isometric views. Using snapshots of each view, one deep CNN model is constructed and further optimized by Bayesian optimization algorithm to indentify COPD. The majority voting of three views presents the final prediction. Finally, the class-discriminative localization maps have been drawn to visually explain the CNNs' decisions. The models trained with single view (ventral, dorsal and isometric) of colorful snapshots present the similar accuracy (ACC) (86.8%, 87.5% and 86.7%) and the model after voting achieves the ACC of 88.2%. The ACC of the final voting model using gray and binary snapshots achieves 88.6% and 86.4%, respectively. Our specially designed CNNs outperform the typical off-the-shelf CNNs and the pre-trained CNNs with fine tuning. The class-discriminative regions of COPD are mainly located at central airways; however, regions in HC are scattering and located at peripheral airways. It is feasible to identify COPD using snapshots of 3D lung airway tree extracted from CT images via deep CNN. The CNNs can represent the abnormalities of airway tree in COPD and make accurate CT-based diagnosis of COPD.

Amino acids are the building blocks of biomacromolecules in organisms, among which isoleucine (Ile) is the precursor of JA-Ile, an active molecule of phytohormone jasmonate (JA). JA is essential for diverse plant defense responses against biotic and abiotic stresses. Botrytis cinerea is a necrotrophic nutritional fungal pathogen that causes the second most severe plant fungal disease worldwide and infects more than 200 kinds of monocot and dicot plant species. In this study, we demonstrated that Ile application enhances plant resistance against B. cinerea in Arabidopsis, which is dependent on the JA receptor COI1 and the jasmonic acid-amido synthetase JAR1. The mutant lib with higher Ile content in leaves exhibits enhanced resistance to B. cinerea infection. Furthermore, we found that the exogenous Ile application moderately enhanced plant resistance to B. cinerea in various horticultural plant species, including lettuce, rose, and strawberry, suggesting a practical and effective strategy to control B. cinerea disease in agriculture. These results together showed that the increase of Ile could positively regulate the resistance of various plants to B. cinerea by enhancing JA signaling, which would offer potential applications for crop protection.

Abstract The cryosphere plays a major role in earth's climate system. Most cryospheric assessments focus on one or more of its components and their response to climate change. However, to date, there has not been a comprehensive evaluation of the entire global cryosphere. We therefore determine such a holistic estimate and quantify changes to the hemispheric and global cryosphere due to climate change, by synthesizing sea ice, snow cover, and frozen ground extents into one global cryospheric extent data set. The 1981–2010 climatology of daily global cryospheric extent ranges from 45.7 ± 0.7 × 106 to 87.2 ± 2.0 × 106 km2 (9.0%–17.1%), from 13.3 ± 0.8 × 106–66.3 ± 1.7 × 106 km2 (5.2%–26.0%) in the Northern Hemisphere (NH), and from 17.9 ± 0.3 × 106 to 33.6 ± 0.4 × 106 km2 (7.0%–13.2%) in the Southern Hemisphere (SH). The monthly maximum cryospheric extent of 85.84 ± 1.91 × 106 km2 occurs in December, whereas minimum occurs in July with 45.92 ± 0.70 × 106 km2. During 1979–2016, global cryospheric area extent lost approximately 87 ± 11 × 103 km2/yr, with a decrease of 102 ± 9.7 × 103 km2/yr in the NH that was partly offset by an increase of 14.6 ± 4.4 × 103 km2/yr in the SH. The first day of cryospheric cover was delayed by 3.6 days at a rate of 0.95 days/decade, and the last day advanced by 5.7 days, at a rate of 1.5 days/decade. The duration and number of cryospheric cover days decreased by 8.7 days and 7.6 days over the study period, respectively. These variations of global cryospheric extent are correlated with rising air temperatures. Our findings highlight the importance of assessing the cryosphere as a whole, and provide a way to quantitatively estimate its overall changes.

Protein content, a vital component determining the nutritional quality of oysters, is unevenly distributed in different parts of oyster. In this study, the spatial distribution (visceral mass, mantle, gill, and adductor) patterns and structural characteristics of proteins, including water–soluble proteins (WSP), salt–soluble proteins (SSP), acid–soluble proteins (ASP) and alkali–soluble proteins (ALSP) of oysters (Crassostrea hongkongensis) were investigated with the amino acid analyzer, circular dichroism spectroscopy (CD), fourier transform infrared spectroscopy (FTIR), and fluorescence spectroscopy. The results showed that oyster proteins were mainly distributed in the visceral mass and mantle. The protein composition was WSP, SSP, ALSP, and ASP in descending order, which conformed to the ideal amino acid pattern. Variations in secondary structure, molecular weight distribution, and thermal denaturation temperatures of the oyster proteins were observed. SSP had wider bands (16–270 kDa) than those of ASP (30–37 kDa) and ALSP (66–270 kDa). Among the four proteins, the SSP of the mantle showed the highest thermal stability (87.4 °C), while ALSP of the adductor muscle had the lowest the lowest the peak denaturation temperature (Tm) (53.8 °C). The proportions of secondary structures in oyster proteins were different, with a higher proportion of solid protein β–folds, and the exposure of aromatic amino acid residues and disulfide bonds and the microenvironment in which they were located were also different.

Abstract Surface air temperatures affect a diverse set of physical and biological systems in many parts of the world. For regional‐scale studies, gridded surface air temperature data sets are frequently used as input variables. Here we evaluate 10 commonly used gridded air temperature products with spatial resolutions ranging from 0.1° × 0.1° to 5.0° × 5.0°, relative to observations from in situ weather stations on the Tibetan Plateau. Gridded temperatures are consistently lower, with mean annual air temperature biases ranging from −4.68 °C to −1.72 °C, and root mean square error (RMSE) from 3.24 °C to 6.11 °C. The mean biases of mean seasonal air temperatures for spring, summer, autumn, and winter are −3.21 °C, −2.98 °C, −2.89 °C, and −2.98 °C, respectively, and RMSEs of 4.81 °C, 4.39 °C, 4.3 °C, and 4.7 °C. Of all 10 products, the China meteorological forcing data set compares best with observations. Annual and season temperatures all increase significantly during 1980–2018 on the Tibetan Plateau, with winter dominating the warming rate.

Myopia is a global health issue, and the prevalence of high myopia has increased significantly in the past five to six decades. The high incidence of myopia and its vision-threatening course emphasize the need for automated methods to screen for high myopia and its serious form, named pathologic myopia (PM). Artificial intelligence (AI)-based applications have been extensively applied in medicine, and these applications have focused on analyzing ophthalmic images to diagnose the disease and to determine prognosis from these images. However, unlike diseases that mainly show pathologic changes in the fundus, high myopia and PM generate even more data because both the ophthalmic information and morphological changes in the retina and choroid need to be analyzed. In this review, we present how AI techniques have been used to diagnose and manage high myopia, PM, and other ocular diseases and discuss the current capacity of AI in assisting in preventing high myopia.

Abstract Background Focal segmental glomerulosclerosis (FSGS), as the frequent primary glomerular diseases in adults, accounts for symptomless proteinuria or nephrotic syndrome with or without renal insufficiency. As the crucial lesion of chronic kidney disease (CKD), accumulating evidence from recent studies show that mutations in Collagen‐related genes may be responsible for FSGS. The aim of this study was to identify the genetic lesion of a Chinese family with FSGS and CKD. Methods In this study, we recruited a Han‐Chinese family with unexplained high serum creatinine, hematuria, and proteinuria. Further renal biopsy and renal pathology indicated the diagnosis of FSGS in the proband. Whole‐exome sequencing and Sanger sequencing were employed to explore the pathogenic mutation of this family. Results A novel heterozygous mutation (NM_000092 c.2030G>A, p.G677D) of the collagen type IV alpha‐4 gene (COL4A4) was detected. Co‐segregation analysis revealed that the novel mutation was carried by all the five affected individuals and absent in other healthy members as well as in our 200 local control cohorts. Bioinformatics predication indicated that this novel mutation was pathogenic and may disrupt the structure and function of type IV collagen. Simultaneously, this variant is located in an evolutionarily conserved site of COL4A4 protein. Conclusion Here, we identified a novel mutation of COL4A4 in a family with FSGS and CKD. Our study expanded the variants spectrum of the COL4A4 gene and contributed to the genetic counseling and prenatal genetic diagnosis of the family. In addition, we also recommended the new classification of collagen IV nephropathies, which may be a benefit to the diagnosis, target drug treatment, and management of patients with COL4A3/COL4A4 mutations.

Arteriosclerotic cardiovascular disease (ASCVD) is one of the major causes of death worldwide and most commonly develops as a result of atherosclerosis (AS). As we all know, dyslipidemia is a leading pathogenic risk factor for ASCVD, which leads to cardiac ischemic injury and myocardial infarction. Dyslipidemias include hypercholesterolemia, hypertriglyceridemia, increased low-density lipoprotein cholesterol (LDL-c) and decreased high density lipoproteins cholesterol (HDL-c). Mutations of dyslipidemia related genes have been proved to be the crucial contributor to the development of AS and ASCVD. In this study, a Han-Chinese family with ASCVD was enrolled and the lipid testing discovered an obvious reduced levels of HDL-c in the affected members. We then performed whole exome sequencing to detect the candidate genes of the family. After data filtering, a novel heterozygous nonsense mutation (NM_007168: c.3460C>T; p.R1154X) of ABCA8 was detected and validated to be co-separated in the family members by Sanger sequencing. Previous studies have proved that deleterious heterozygous ABCA8 variants may disrupt cholesterol efflux and reduce HDL-c levels in humans and mice. This study may be the second report related to ABCA8 mutations in patients with reduced levels of HDL-c. Our study not only contributed to the genetic counseling and prenatal genetic diagnosis of patients with ASCVD caused by reduced HDL-c levels, but also provided a new sight among ABCA8, cholesterol efflux and HDL-c levels.

Synthetic rock crystals often show a typical infrared (IR) absorption band at 3585 cm−1. However, the authors recently found this band in a natural rock crystal with blue coating. The origin of this IR band is controversial as yet. In this paper, the infrared spectra of several natural and synthetic rock crystal samples which were heated to 673 K and 1073 K were measured after these samples returned to room temperature. Comparing the infrared spectra of samples before and after heating, we found the absorption band at 3585 cm−1 was induced by the thermal process, which indicates that this band cannot be used as diagnostic evidence for synthetic rock crystal alone. In addition, the LiOH bands decreased while AlOH bands increased upon thermal processing. And the negative correlation between the LiOH bands and the 3585 cm−1 band was also distinct. The above results reveal that the thermal process destroyed the LiOH defects, leading to the formation of a new AlLi defect. And the isolated OH− defect inside dislocations generated upon thermal processing is considered to be the exact cause of the 3585 cm−1 band.

Abstract Background Myopic foveoschisis (MF) is among the leading causes of visual loss in high myopia. However, it remains controversial whether internal limiting membrane (ILM) peeling or gas tamponade is necessary treatment option for MF. Methods PubMed, EMBASE, CBM, CNKI, WANFANG DATA and VIP databases were systematically reviewed. Outcome indicators were myopic foveoschisis resolution rate, visual acuity improvement and postoperative complications. Results Nine studies that included 239 eyes were selected. The proportion of resolution of foveoschisis was higher in ILM peeling group than non-ILM peeling group (OR = 2.15, 95% CI: 1.06–4.35; P = 0.03). The proportion of postoperative complications was higher in Tamponade group than non-Tamponade group (OR = 10.81, 95% CI: 1.26–93.02; P = 0.03). However, the proportion of visual acuity improvement (OR = 1.63, 95% CI: 0.56–4.80; P = 0.37) between ILM peeling group and non-ILM peeling group and the proportion of resolution of foveoschisis (OR = 1.80, 95% CI: 0.76–4.28; P = 0.18) between Tamponade group and non-Tamponade group were similar. Conclusions Vitrectomy with internal limiting membrane peeling could contribute to better resolution of myopic foveoschisis than non-peeling, however it does not significantly influence the proportion of visual acuity improvement and postoperative complications. Vitrectomy with gas tamponade is associated with more complications than non-tamponade and does not significantly influence the proportion of visual acuity improvement and resolution of myopic foveoschisis.

In this paper, the authors presented a study on the discrimination of handlebar grip samples, to provide effective forensic science service for hit and run traffic cases. 50 bicycle handlebar grip samples, 49 electric bike handlebar grip samples, and 96 motorcycle handlebar grip samples have been randomly collected by the local police in Beijing (China). Fourier transform infrared microspectroscopy (FTIR) was utilized as analytical technology. Then, target absorption selection, data pretreatment, and discrimination of linked samples and unlinked samples were chosen as three steps to improve the discrimination of FTIR spectrums collected from different handlebar grip samples. Principal component analysis and receiver operating characteristic curve were utilized to evaluate different data selection methods and different data pretreatment methods, respectively. It is possible to explore the evidential value of handlebar grip residue evidence through instrumental analysis and statistical treatments. It will provide a universal discrimination method for other forensic science samples as well.

Purpose. To determine the effect of osthole on the retina in a chronic cerebral hypoperfusion (CCH) rat model and to investigate its therapeutic activity. Methods. Seventy-two rats were randomly allocated into 6 groups. CCH was induced by permanent bilateral common carotid artery occlusion (BCCAO) in five groups. Sham surgery was performed without occlusion of the artery in the sixth group (control group). Animals were administered with saline (model group), osthole (osthole-IG group), aspirin (aspirin group), or ginaton (ginaton group); the osthole-PI group was performed with peribulbar injection of osthole. Four rats in each group were sacrificed every 5 days after drug administration, and histopathology along with morphology of retina were observed. Fundus fluorescein angiography was performed before the animals were sacrificed at day 15. Retinal Akt, NF-κB, Bax, and Bcl-2 levels were assessed using immunohistochemistry, immunofluorescence, and reverse-transcription PCR; retinal injury was assessed using TUNEL in situ; retinal levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were measured. Results. Fundus fluorescein angiography revealed the retinal vascular diameter in the osthole-IG group rats to be wider than that in the model, osthole-PI, aspirin, or ginaton group rats. Histological analysis of retinal tissue revealed an increase in retinal thickness in all treatment groups, and significant improvement was noticed in the osthole-IG group. TUNEL staining revealed fewer apoptotic cells in the osthole-IG and osthole-PI groups than in the other groups. For immunohistochemistry results, in the osthole-IG group, levels of NF-κB and Akt were lower than those in the other treated groups, while levels of the ratio Bcl-2/Bax were higher. Levels of MDA were lower and levels of SOD were higher in the osthole-IG group than in the other groups. Conclusions. Osthole protects the retina from ischemia injury secondary to CCH induced by BCCAO, mainly through anti-inflammatory, antioxidant, and antiapoptotic effects.

The cellulolytic and ethanologenic bacterial community is a promising candidate for the production of bioethanol from lignocellulose. In this study, by artificially changing the ratio of Clostridium thermocellum in the cellulolytic consortium H, ethanol production was increased by 72.7%. Metatranscriptomic analysis was used to elucidate the contribution of Clostridium thermocellum to ethanol production. A comprehensive analysis of genes mapped to the Clostridium thermocellum ATCC 27405 genome was performed; the identified gene expression differences related to cellulosic ethanol pathways were carefully studied. The results indicated that the majority of genes involved in lignocellulose degradation, sugar transport, cellodextrin breakdown, glycolysis, and ethanol synthesis were up-regulated in C. thermocellum when added to H (HCt). More than 18 cellulosome-related genes had 15-fold or greater increased expression. The results illustrate the role of C. thermocellum in the cellulolytic consortium H and HCt and provided useful information for identifying genes and preferred pathways. These results will aid in the metabolic and genetic engineering of bacterial strains for more efficient biofuel production.

The complete molecule of the title compound, C10H8N6, is generated by crystallographic inversion symmetry; the dihedral angle between the planes of the benzene and triazole rings is 16.7 (2)°. In the crystal, inversion dimers linked by pairs of weak C—H...N hydrogen bonds generate R22(6) loops. Weak aromatic π–π stacking interactions [centroid–centroid separation = 3.809 (1) Å] are also observed.

Ethanol production from NaOH-Pretreated solid state fermented sweet sorghum bagasse with an engineered strain of Z. mobilis TSH-ZM-01 was optimized. Results showed that: (1) residual solid removal during ethanol fermentation was unnecessary and 24 h fermentation duration was optimal for ethanol production; (2) ethanol yield of 179.20 g/kg of solid state fermented sweet sorghum bagasse achieved under the optimized process conditions of cellulase loading of 0.04 g/g-glucan, xylanase loading of 0.01 g/g-xylan, liquid to solid ratio of 9:1 and pre-hydrolysis duration for 72 h.

The rising demand for bioethanol, the most common alternative to petroleum-derived fuel used worldwide, has encouraged a feedstock shift to non-food crops to reduce the competition for resources between food and energy production. Sweet sorghum has become one of the most promising non-food energy crops because of its high output and strong adaptive ability. However, the means by which sweet sorghum stalks can be cost-effectively utilized for ethanol fermentation in large-scale industrial production and commercialization remains unclear. In this study, we identified a novel Saccharomyces cerevisiae strain, TSH1, from the soil in which sweet sorghum stalks were stored. This strain exhibited excellent ethanol fermentative capacity and ability to withstand stressful solid-state fermentation conditions. Furthermore, we gradually scaled up from a 500-mL flask to a 127-m3 rotary-drum fermenter and eventually constructed a 550-m3 rotary-drum fermentation system to establish an efficient industrial fermentation platform based on TSH1. The batch fermentations were completed in less than 20 hours, with up to 96 tons of crushed sweet sorghum stalks in the 550-m3 fermenter reaching 88% of relative theoretical ethanol yield (RTEY). These results collectively demonstrate that ethanol solid-state fermentation technology can be a highly efficient and low-cost solution for utilizing sweet sorghum, providing a feasible and economical means of developing non-food bioethanol.
