The crosstalk between clathrin-mediated endocytosis (CME) and the autophagy pathway has been reported in mammals; however, the interconnection of CME with autophagy has not been established in plants. Here, we report that the Arabidopsis CLATHRIN LIGHT CHAIN (CLC) subunit 2 and 3 double mutant, clc2-1 clc3-1, phenocopies Arabidopsis AUTOPHAGY-RELATED GENE (ATG) mutants in both autoimmunity and nutrient sensitivity. Accordingly, the autophagy pathway is significantly compromised in the clc2-1 clc3-1 mutant. Interestingly, multiple assays demonstrate that CLC2 directly interacts with ATG8h/ATG8i in a domain-specific manner. As expected, both GFP-ATG8h/GFP-ATG8i and CLC2-GFP are subjected to autophagic degradation, and degradation of GFP-ATG8h is significantly reduced in the clc2-1 clc3-1 mutant. Notably, simultaneous knockout of ATG8h and ATG8i by CRISPR-Cas9 results in enhanced resistance against Golovinomyces cichoracearum, supporting the functional relevance of the CLC2-ATG8h/8i interactions. In conclusion, our results reveal a link between the function of CLCs and the autophagy pathway in Arabidopsis.

Retinopathy of prematurity (ROP) is one of the leading causes of childhood visual impairment and blindness. However, there are still very few effective pharmacological interventions for ROP. Histone deacetylase 6 (HDAC6)-mediated disassembly of photoreceptor cilia has recently been implicated as an early event in the pathogenesis of ROP. Herein it is shown that enhanced expression of HDAC6 by intravitreal injection of adenoviruses encoding HDAC6 induces the typical pathological changes associated with ROP in mice, including disruption of the membranous disks of photoreceptor outer segments and a decrease in electroretinographic amplitudes. Hdac6 transgenic mice exhibit similar ROP-related defects in retinal structures and functions and disassembly of photoreceptor cilia, whereas Hdac6 knockout mice are resistant to oxygen change-induced retinal defects. It is further shown that blocking HDAC6-mediated cilium disassembly by intravitreal injection of small-molecule compounds protect mice from ROP-associated retinal defects. The findings indicate that pharmacological targeting of the HDAC6-cilium axis may represent a promising strategy for the prevention of ROP.

SrTiO3 is a new type of inorganic non-metallic photocatalyst with unique properties, which has excellent performance in the decomposition of water to produce hydrogen. However, the existing SrTiO3-based photocatalysts still have the problems of low light absorption capacity and low electron and hole separation efficiency, resulting in too low hydrogen production efficiency. In order to solve these two problems, a series of SrTiO3–TiO2 hollow structure nanospheres were prepared by in-situ growth of SrTiO3 nanocube crystals on TiO2 solid by hydrothermal method. By controlling the time of hydrothermal reaction to regulate the morphology of hydrothermal reaction products, the hollow structure similar to lychee shell was prepared, so that the incident light could be reflected multiple times inside. Through the hydrothermal of SrTiO3 on the surface of TiO2, to complete the coverage of SrTiO3 on the surface of TiO2 to construct SrTiO3–TiO2 heterojunction, which improves the separation efficiency of photogenerated electrons and holes. The photocatalytic hydrogen production performance of SrTiO3–TiO2 heterojunction is achieved 374.07 μmol ·g−1 ·h−1, and it is 2.35 times higher than pure sample. This work provides a new simple method for preparing high-yield hydrogen photocatalysts and provides a new solution for the conversion of solar energy to hydrogen energy. [ABSTRACT FROM AUTHOR]

Abstract Background It has been reported that long non-coding RNAs (lncRNAs) play vital roles in diabetic nephropathy (DN). Our study aims to research the function of lncRNA KCNQ1OT1 in DN cells and the molecular mechanism. Methods Human glomerular mesangial cells (HGMCs) and human renal glomerular endothelial cells (HRGECs) were cultured in high glucose (30 mM) condition as models of DN cells. KCNQ1 opposite strand/antisense transcript 1 (KCNQ1OT1) and miR-18b-5p levels were detected by quantitative real-time polymerase chain reaction (qRT-PCR). The mRNA and protein levels of Sorbin and SH3 domain-containing protein 2 (SORBS2), Type IV collagen (Col-4), fibronectin (FN), transcriptional regulatory factor-beta 1 (TGF-β1), Twist, NF-κB and STAT3 were measured by qRT-PCR and western blot. Cell viability was detected by cell counting kit-8 (CCK-8) assay for selecting the proper concentration of glucose treatment. Additionally, 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) and flow cytometry assay were employed to determine cell proliferation and apoptosis, respectively. The targets of KCNQ1OT1 was predicted by online software and confirmed by dual-luciferase reporter assay. Results KCNQ1OT1 and SORBS2 were elevated in DN. Both knockdown of KCNQ1OT1 and silencing of SORBS2 restrained proliferation and fibrosis and induced apoptosis in DN cells. Besides, Overexpression of SORBS2 restored the KCNQ1OT1 knockdown-mediate effects on proliferation, apoptosis and fibrosis in DN cells. In addition, miR-18b-5p served as a target of KCNQ1OT1 as well as targeted SORBS2. KCNQ1OT1 knockdown repressed NF-ĸB pathway. Conclusion KCNQ1OT1 regulated DN cells proliferation, apoptosis and fibrosis via KCNQ1OT1/miR-18b-5p/SORBS2 axis and NF-ĸB pathway.

Graphitic carbon nitride (g-C3N4) is a fresh type of environmentally friendly photocatalytic material with the potential to degrade pollutants in water. In particular, g-C3N4 (CN) has significant characteristics in the treatment of antibiotics, which has attracted extensive attention from researchers. However, due to the less active sites and fast carrier recombination rate resulting from the block morphology, the CN has unsatisfactory photocatalytic performance up to now. Therefore, in this paper, CN nanosheets with more active sites and slower carrier recombination rate were successfully prepared by a combination of P element doping and triple calcination modifications. The modified 1.5% P-CN-3 not only has a 2.2-fold increase in the degradation ability of antibiotic Tetracycline (TC) compared to pure CN, but also has a hydrogen production rate of 2268.3 µmol·g−1·h−1 for water decomposition. A stochastic discrete fractional Cournot duopoly game model with a unique interior Nash equilibrium is developed in this study. Some sufficient criteria of the Lyapunov stability in probability for the proposed model at the interior Nash equilibrium are derived using the Lyapunov theory. The proposed model's finite time stability in probability is then investigated using a nonlinear feedback control approach at the interior Nash equilibrium. The stochastic Bellman theory is also used to explore the locally optimum control problem. Furthermore, bifurcation diagrams, time series, and the 0-1 test are used to investigate the chaotic dynamics of this model. Finally, numerical examples are given to illustrate the obtained results. Sacituzumab govitecan is widely used for the treatment of breast cancer and urothelial carcinoma, but available information regarding adverse events (AEs) is limited. We aim to explore the AE induced by sacituzumab govitecan by mining the FDA Adverse Event Reporting System (FAERS) database. The association between sacituzumab govitecan and AEs was evaluated using the information component. A multivariate logistic regression analysis was conducted for all identified signals to explore the risk factors associated with AEs leading to hospitalization. In total, 1,884 reports related to sacituzumab govitecan were retrieved, and 114 AE signals involving 20 systems were identified. The median time for onset of AEs was ~ 6–7 days after initiating treatment with sacituzumab govitecan, with over 80% of AEs occurring within 30 days. Subgroup analysis revealed that 14 signals were reported in men and 110 in women. There were 58 signals reported in patients under 65 following the use of sacituzumab govitecan, 59 signals in patients over 65, and 31 signals were present in both groups. Multivariable analysis showed that being male and the occurrence of colitis, pneumonitis, febrile neutropenia, pyrexia, sepsis, dehydration, and diarrhea were risk factors leading to hospitalization with an area under the curve (AUC) of 0.89. Additionally, sensitivity analysis revealed that this study had good robustness. This is the first retrospective analysis based on FAERS to review the safety of sacituzumab govitecan. The results highlight the need to closely monitor adverse reactions such as neutropenia, diarrhea, colitis, and sepsis when using sacituzumab govitecan. [ABSTRACT FROM AUTHOR]
