Nanotechnology is arising as a fast-developing research discipline with many usages areas. The silver nanoparticles synthesis (sm-AgNPs) is accomplished by reduction of silver ions in treatment with aqueous extract of Sideritis montana L. leaves. The colour change from yellow to dark brown confirmed the structures. The spectroscopic studies revealed the desired structure. In the UV-Vis spectrum, the maximum absorption was observed at 480 nm. The diffraction peaks (2θ) at the degrees of 38.14°, 44.29°, 64.48°, and 77.38° can correspond to 111, 200, 220, and 311 facets that indicates the nanostructure to be a face-centered cubic unit structure. The scanning electron microscope (SEM) and dynamic light scattering (DLS) analyses indicated that the synthesized nanoparticles were spherical with an average particle size of 36.42 nm. The zeta potential of sm-AgNPs was found as -35.2 mV which indicated the repulsion among nanoparticles and their stability. The peaks from Fourier transform infrared spectrometer (FTIR) were associated with the phenols, flavonoids, terpenoids, and alkaloids, indicating that the corresponding compounds might act as reducing agents. The photocatalytic effect of sm-AgNPs was examined by degradation of methylene blue and sm-AgNPs were able to degrade the dye by about 67% at 96 h.

This study aims to quantify and identify phenolic compounds in the hydroethanolic extract of Mentha piperita L. harvested in the Algerian Sahara by colorimetric and LC-MS/MS analysis, assess its antioxidant capacity by DPPH and ABTS assays and examine the impact of its inclusion at 2 to 6% rates on sensory and oxidative properties (DPPH, ABTS, TEBARS) and the lipid profile analyzed by GC-FID in steamed yoghurt. The extract contained 41.29 mg GAE/ml total phenolics, 22.45 mg QE/ml total flavonoids and only 0.59 mg CE/ml condensed tannins. LCMS/MS analysis detected 26 phenolic compounds, among which rosmarinic acid was the most abundant. DPPH and ABTS assays showed IC50 values of 3.18 and 2.49 mg freeze-dried extract/ml, indicating the high antioxidant potential of the examined extract. Yogurts containing 2% and 4% of this extract were well accepted by the sensory evaluation panel. Background: Hypericum heterophyllum has been used as a traditional medicine and includes significant bioactive compounds. Objective: To quantitatively determine the phenolic and flavonoid contents of Hypericum heterophyllum flowers and evaluate the antioxidant activity. Also, to present the activity-compound relationship. Method: Flowers of H. heterophyllum Vent., endemic species for Turkey were collected from Yozgat-Turkey. H. heterophyllum flowers were extracted with methanol, ethanol, acetone, and chloroform individually. LC-ESI-MS/MS analysis for phytochemicals was carried out for each extract. Total phenolic and flavonoid content were carried out for each extract. The antioxidant activity was performed using DPPH•, ABTS•+, hydroxyl radical, and superoxide radical scavenging assays. Results: Chlorogenic acid was found as a major compound in all extracts. Among the extracts, methanol extract included the most phenolic compound. The methanol extract and acetone extract consisted of the most flavonoids. Acetone and ethanol extracts revealed the most DPPH and ABTS activity respectively. Acetone extract exhibited the highest hydroxyl radical and superoxide radical scavenging effect Conclusion: Flowers of H. heterophyllum contain bioactive compounds and they revealed good antioxidant activity. Hence, H. heterophyllum flowers could be a promising agent for the pharmaceutical and food industries.

OBJECTIVE: To characterize the chemical profile of methanolic crude extract and its fractions (Ethyl acetate, n-butanol and aqueous) using liquid chromatography-mass spectrometry (LC-MS) analysis, to evaluate their biological and pharmacological properties: antioxidant (1, 1-diphenyl-2-pycrylhydrazyl (DPPH), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic) (ABTS(·+)), galvinoxyle free radical scavenging, reducing power, phenanthroline and β carotene-linoleic acid bleaching assays), enzymes inhibitory ability against several enzymes [acetyl-cholinesterase (AChE), buthyrylcholinesterase (BChE), urease and tyrosinase]. METHODS: Secondary metabolites were extracted from Tamarix africana air-dried powdered leaves by maceration, the crude extract was fractionated using different solvents with different polarities (Ethyl acetate, n-butanol and aqueous). The amount of polyphenols, flavonoids and tannins (hydrolysable and condensed) were determined using colorimetric assays. A variety of in vitro biochemical tests were carried out to assess antioxidant and oxygen radical scavenging properties using DPPH, ABTS, galvinoxyle free radical scavenging, reducing power, phenanthroline and β carotene-linoleic acid bleaching methods. Neuroprotective effect was examined against acetylcholinesterase and buthy-rylcholinesterase enzymes. The anti-urease and anti-tyrosinase activities were performed against urease and tyrosinase enzymes respectively. The extract's components were identified using LC-MS and compared to reference substances. RESULTS: The results indicated that Tamarix africana extracts presented a powerful antioxidant activity in all assays and exhibited a potent inhibitory effect against AChE and BChE as well as urease and tyrosinase enzymes. LC-MS analysis identified amount of eight phenolic compounds were revealed in this analysis; Apigenin, Diosmin, Quercetin, Quercetine-3-glycoside, Apigenin 7-O glycoside, Rutin, Neohesperidin and Wogonin in methanolic extract and its different fractions of Tamarix africana from leaves. CONCLUSIONS: Based on these findings, it is reasonable to assume that Tamarix africana could be considered as a potential candidate for pharmaceutical, cosmetics, and food industries to create innovative health-promoting drugs.

Origanum genus is an aromatic and medicinal plant with significant bioactive compounds. Origanum species have been used as a folk medicine for years. In this study, the aerial part of Origanum bilgeri P.H. Davis was extracted with hexane and methanol successively, the methanol extract was used for quantitative analysis of bioactive compounds by High‑Performance Liquid Chromatography. Rosmarinic acid was found as a major product (26.61 mg/g extract). The other major compounds were detected as catechin hydride (5.65 mg/g extract), trans-cinnamic acid (2.65 mg/g extract), and resveratrol (1.33 mg/g extract). Consequently, Origanum bilgeri may be a valuable material for drug development and food., Origanum cinsi önemli biyoaktif bileşikler içeren aromatik ve tıbbi bir bitkidir. Origanum türleri yıllardır halk ilacı olarak kullanılmaktadır. Bu çalışmada, Origanum bilgeri P.H. Davis sırasıyla heksan ve metanol ile ekstrakte edildi, metanol ekstraktı, Yüksek Performanslı Sıvı Kromatografisi ile biyoaktif bileşiklerin kantitatif analizi için kullanıldı. Ana ürün olarak rosmarinik asit bulundu (26.61 mg/g ekstrakt). Diğer ana bileşikler kateşin hidrit (5,65 mg/g ekstrakt), trans-sinnamik asit (2,65 mg/g ekstrakt), resveratrol (1,33 mg/g ekstrakt) olarak tespit edildi. Sonuç olarak, Origanum bilgeri ilaç geliştirme ve gıda için değerli bir materyal olabilme potansiyeline sahiptir.

Background: Natural products have been used commonly in the pharmaceutical industry as well as traditional medicine due to their bioactive contents. Objective: The objective of this study is to isolate the polymethoxy flavones (PMFs) and evaluate the antiproliferative activity. Method: The PMFs were extracted from Mentha dumetorum with boiling hot water and then partitioned with hexane and the extract was subjected to chromatographic techniques such as Sephadex LH-20, silica gel, and preparative TLC, and HPLC to isolate the compounds. The structures of isolated compounds were elucidated by spectroscopic techniques such as 1H-NMR, 13C-NMR, LC-TOF-MS, and UV-Vis. The anti-cancer effects of isolated compounds were evaluated using a real-time cell analyzer–single plate (RTCA-SP) instrument against HeLa and HT29 cell lines. Results: The isolated PMFs were identified as xanthomicrol (1), 5-hydroxy-6,7,8,4'-tetrametoxy flavanone (2), 5-desmetil sinensetin (3), 5-demetil nobiletin (4), gardenin B (5), 5-hydroxy-6,7,8,3',4'-penta methoxy flavanone (6) and 5-hydroxy-6,7,4'-trimethoxy flavone (7). Compounds 2 and 5 were found to be the most active against both cell lines. Conclusion: The isolated compounds as well as the plant extract of Mentha dumetorum could be promising agents for the drug development process, especially drugs for cancer treatment. Moreover, isolation methods were developed for the corresponding compounds.

Nanotechnology has gained great interest due to its widespread application. Anticancer activity of Origanum majorana extract and silver nanoparticles was investigated by MTT assay using human pancreatic adenocarcinoma cell lines (Capan-1), mouse normal fibroblast cell lines (L929), and human colon adenocarcinoma cell line (Caco-2). AgNPs@Om exhibited the excellent cytotoxic effect on Capan-1 cell lines with the cell viability of 29.17% and 25.70%, 22.49%, 21.16% at 1.0, 0.5, 0.25, 0.125 µg/mL respectively. However, the extract was determined to show moderate activity. A considerable fall in the viable cell number in Caco-2 cell lines was observed after the treatment of AgNPs@Om. The viable cells of Caco-2 cell lines were detected as 32.0% at 1.0 µg/mL with the treatment of nanoparticles. Yet, the viable Caco-2 cells were determined as 48.2% with the treatment of O. majorana extract. Both extract and nanoparticles had no impact on the viability of non-tumor cells (L929). Moreover, nanoparticles induced apoptosis. Consequently, AgNPs@Om may be a promising anticancer drug candidate.

The aim of the present study is to evaluate the total phenolic contents, antioxidant and anti-proliferative activities of the n-BuOH extract of Centaurea maroccana (BECM). The total phenolic and flavonoid of the butanolic extract of the plant were assessed by using Folin-Ciocalteau and aluminium chloride colorimetric assays, respectively, and the quantitative estimation of total flavonoids and phenols revealed the richness of the extract in these compounds. Antioxidant activity was evaluated using standard lab colorimetric methods, while the anti-proliferative activity was evaluated using sulforhodamine B (SRB) assay against C6 (Rattus norvegicus brain glioma) and HT29 (Homo sapiens colorectal adenocarcinoma) cell lines. The n-butanolic extract of Centaurea maroccana showed a strong antioxidant activity through DPPH, ABTS•+, DMSO alkalin, Reducing power, β-Carotene-linoleic acid and CUPRAC assays. Furthermore, the anti-proliferative activity against C6 and HT29 of Centaurea maroccana exhibited a high effect by the decrease in viability of both cancer cell lines. Our results suggest a possible use of Centaurea maroccana as a source of natural antioxidant and chemo-preventive agents against cancer.

Objective: Ruta tuberculata forssk. (Rutaceae) is an aromatic plant widely used in traditional Algerian medicine due to its pharmaceutical virtues against various disorders. This study aims to determine the phenolic profile of aqueous (RAE) and methanol (RME) extracts of R. tuberculata aerial parts and to investigate their acute oral toxicity, as well as their possible antiproliferative and hepatoprotective effects. Methods: Polyphenols were identified by quantitative LC-MS/MS analysis. Oral acute toxicity was performed according to OCDE guidelines. The hepatoprotective activity was evaluated by paracetamol-induced hepatotoxicity and supported by biochemical and histological analysis of liver and kidneys. The antiproliferative activity against human colorectal HT-29 and ovarian OV2008 cancer cell lines was determined using SRB assay. Results: LC-MS/MS analysis revealed that RME has higher phenols and flavonoids content than RAE; however, its major identified flavonoids are Kaempferol, rutin, and naringenin. R. tuberculata seems mildly toxic at several doses, with oral LD50 greater than 5000 mg/kg. The significant increase in hepatic markers enzymes activities as well as cholesterol, triglycerides, and glycemic levels, caused by PCM-administration, was potentially reduced following the co-treatments with vitamin C and RME, respectively, compared to RAE. Moreover, RME-treatment markedly prevented all histological changes. Compared to RAE, RME (100 μg/mL) exhibited excellent antiproliferative activity against both tested cancer lines (% inhibition ≥ 80%). Conclusion: Both R. tuberculata extracts (200 mg/kg/daily) were non-toxic and exerted a potential hepatoprotective effect against PCM-induced hepatotoxicity. Accordingly, RME may be considered a good candidate for the development of new therapies against colorectal and ovarian cancers.

Onosma species have been used commonly for traditional medicine for years due to their bioactive compounds content. Onosma bourgaei aerial part was extracted with hexane and methanol successively. The methanol extract was subjected to chromatographic techniques to isolate allantoin (1), 3,4-dihydroxybenzaldehyde (2), luteolin-7-O-glucoside (3), apigenin-7-O-β-glucoside (4), diosmetin-7-O-β-glucoside (5), rosmarinic acid (6), globoidnan A (7). The structure of isolated compounds were identified by spectroscopic techniques such as 1D-NMR, 2D-NMR, FTIR, and LC-TOF/MS/MS. Antiproliferative activity of extract and natural compounds were carried out using HeLa (human epithelial cervix adenocarcinoma, ATCC® CCL-2™), HT-29 (human colorectal adenocarcinoma, ATCC® HTB38™), MCF-7 (human mammary gland adenocarcinoma, ATCC® HTB22™) and A549 (human lung carcinoma, ATCC® CCL185™) cancerous cells and normal cells, FL (human epithelial amnion cell, ATCC® CCL62™). Lactate dehydrogenase (LDH) was performed for cytotoxicity. The compounds, 4, 6, and 7 displayed the strong antiproliferative activity against corresponding cell lines. Apigenin-7-O-β-glucoside (4) revealed the excellent activity on HeLa, HT29, A549, MCF6 cancer cell lines with the values of (IC50, µM) 167.3, 196.8 181.1, 203.5 respectively compared standard compound, cisplatin.

This study aims to investigate the chemical composition, antioxidant, and antimicrobial activity of two essential oils (EOs) from Algerian propolis. The volatile constituents were analyzed by gas chromatography-mass spectrometry. Fifty components were identified from the oils. The major components were found to be: cedrol (17.0%), β-eudesmol (7.7%), and α-eudesmol (6.7%) in EO of propolis from Oum El Bouaghi (EOPO) whilst α-pinene (56.1%), cis-verbenol (6.0%), and cyclohexene,3-acetoxy-4-(1-hydroxy-1-methylethyl)-1-methyl (4.4%) in EO of propolis from Batna (EOPB). The antioxidant properties of EOPO and EOPB were determined using 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonate) (ABTS•+) and cupric reducing antioxidant capacity (CUPRAC assays), respectively. Both EOs had more cupric ion reducing ability than scavenging ABTS•+ radicals. The antimicrobial potential of the two EOs against eight pathogens was assayed by the agar diffusion method and the mode of action was determined by microdilution assay. The results revealed that EOPB was bactericidal for all tested pathogenic bacteria and fungicidal for Candida albicans ATCC 10231, whereas, EOPO showed bacteriostatic effect against Escherichia coli O157:H7 and Pseudomonas aeruginosa ATCC27853 and fungistatic effect against C. albicans ATCC 10231. Thus, the obtained results suggest the important use of propolis EOs as preservative agents.

Nanostructures have distinctive chemical and physical features owing to their surface area and nanoscale size. In this study, silver nanoparticles were synthesized using curcumin, a medicinally valuable natural product. The structure of curcumin-mediated silver nanoparticles (c-AgNPs) was identified by extensive spectroscopic techniques. The maximum absorption was observed at 430 nm in UV–Vis spectrum. The crystal structure of c-AgNPs was identified by XRD. The morphology of the structure was determined by SEM image. The particle size was found as 51.13 nm. The functional groups of curcumin and c-AgNPs were established by FTIR spectroscopy. Cytotoxic activity of c-AgNPs was carried out using A549, DLD-1, and L929 with MTT assay. c-AgNPs revealed excellent activity on DLD-1 cell lines and A549 cell lines at 1.0 mg/mL concentration with the lethal effect of 80%. However, nanoparticles did not show the considerable effect on L929. Moreover, they induced apoptosis. Consequently, c-AgNPs are a promising material for anticancer drugs candidate.

Background:Plant in Rhododendron genus that contains phenolic compounds has been used in traditional medicine and revealed considerable biological activities.Objective:Isolation and identification of antioxidant natural products from Rhododendron ungernii.Methods:Rhododendron ungernii Trautv. flowers were collected and dried in shade. The dried flowers were extracted with methanol for 3 days. The solvent was removed by reduced pressure to yield the extract which was subjected to column chromatography (Sephadex LH-20, C18 reversed phase column) to isolate catechin-7-O-glucoside (1), quercetin-3-O-β-galactoside (2), quercetin-3-O- β-xyloside (3), farrerol (4), myricetin (5), and quercetin (6). The structures of isolated compounds were elucidated by spectroscopic methods such as 1D-NMR, 2D-NMR, and LC-TOF/MS. DPPH scavenging effect, ABTS+ scavenging activity, and reducing power (FRAP) were performed for antioxidant assays of isolated natural compounds.Results:Isolated flavonoids displayed the outstanding antioxidant activities. Catechin-7-O-glucoside (1) and quercetin-3-O-β-galactoside (2) (IC50, 3.66 µg/mL) had the most DPPH• scavenging effect among the compounds. The highest ABTS•+ scavenging activity (IC50, 1.41 µg/mL) and reducing power effect (6.05 mmol TE/g comp) were observed for myricetin (5).Conclusion:R. ungernii extract and isolated compounds could be a promising antioxidant for food and pharmaceutical industries.
