Interest and interest in the sport of volleyball becomes a very attractive option in school. Volleyball is not only suitable for all ages but can also be enjoyed by all genders. The interest in volleyball is reflected in the natural inclination of a person interested in this game, which provides opportunities for each individual to explore and develop his skills. This research is quantitative descriptive research that aims to determine the interest and factors in learning volleyball games. The sample in this study was all class XI students at Al-Fityah IT High School Pekanbaru with a total of 58 students. The instrument used in this study is a questionnaire using a Likert scale and questionnaire data can be analyzed using a percentage formula. Based on the results of the research conducted, it turns out that the interest of class XI students of Al-Fityah Pekanbaru IT High School towards volleyball games is quite high, reaching 50% or equivalent to half of the total class XI students of Al Fityah IT High School. In addition, it was found that internal factors influencing interest in the game of volleyball were also at a high level, reaching 56.9%. Meanwhile, student interest based on external factors is also included in the high category, which is 48.3%.

This research aims to (1) to develop design of learning software as a learning source on the Pencak Silat subject, designing appropriate theory to be used in learning media of Pencak Silat, (2) to determine the feasibility of learning media for Pencak Silat. This research can be categorized as a Research and Development. To get validity product, it would be tested by validity test. Validation was conducted by the expert in Pencak Silat theory, expert in media and trial on a group of students to obtain the advisability of the media that would be used. The validity test used questionnaire test. The data was analyzed by using the quantitative descriptive analysis technique that revealed in scores of distribution and category scales of grading that determined. Based on the evaluation results by the experts of in material aspect owned average score is 4.0 and it has good category. Then the assessment by judges of media aspect obtained average score is 4.34, it has good category. The students’ response in small group average is 4.35 and it has good category. In conclusion, media that developed can be used for learning.

Background: While it has been known that the development of chronic kidney disease (CKD) and age-related cognitive impairment involves several mediators, the evidence in clinical practice only reveals nitride oxide synthase (NOS) and klotho. However, the evidence for this topic is conflicted. The aim of this study was to assess the role of NOS and klotho single nucleotide polymorphisms (SNPs) in the pathogenesis of CKD and age-related cognitive impairment. Methods: We performed a meta-analysis during October to December 2019. Paper collection was performed in major scientific websites, and we extracted information of interest from each paper. Data were analyzed using a Z-test with either random or fixed effect model. Results: Our initial assessment identified NOS3 G894T, NOS3 T786C, NOS3 4b/4a, klotho (KL) G395A, and KL C1818T as the gene candidate for our meta-analysis. Our pooled calculation revealed that NOS3 G894T was associated with the risk of both age-related cognitive impairment and CKD. Increased susceptibility to age-related cognitive impairment was observed in the GG genotype, and increased risk of CKD was found in patients with a single T allele and TT genotype for NOS3 nucleotide 894. For NOS3 4b/4a, increased risk of CKD was only found in 4a4a genotype. For NOS3 T786C, we failed to show the association with both CKD and age-related cognitive impairment. Subsequently, for KL G395A, A allele and GA genotype were found to correlate with increased susceptibility to CKD, while its correlation to age-related cognitive impairment was failed to clarify. For KL C1818T, our analysis failed to find the correlation with the risk of CKD. Conclusions: Our results reveal that the NOS3 G894T gene polymorphism has a crucial role in the pathogenesis of both CKD and age-related cognitive impairment.

Entering the 4.0 era, innovation and learning development are needed as well in the world of refereeing, development is intended to make it easier and help to reduce mistakes in leading matches. This study aims to develop lectora inspire which will be used in martial art refinement. This type of research is a research and development (R&D) research. The subjects of this study use a small scale with a total of 10 students from the Department of Physical Education, Health and Recreation, Faculty of Teacher Training and Education, Universitas Riau. Meanwhile, the analysis technique used in this research is descriptive quantitative. The results of the research on the development tools of lectora inspire on martial art refinement are good, this is explained from the results of the validation of the content/material aspects with an average score of 4.29, the learning aspects are categorized as good with an average score of 4.34, the programming aspects and the media aspects have good categories. From the results of small group trials conducted by 10 subjects, data are obtained and it is concluded that the Lectora Inspire application is in good category with an average score of 4.32 and is able to be applied in learning martial art refinement. These results are intended to make it easier and help to reduce errors in the referee's assignment when serving on a small scale. Based on the results of this study, it can be concluded that the development of learning media for refereeing martial art by utilizing lectora inspire is suitable for use as a medium in learning audio-visual based martial art on a small scale.

Based on observation, researchers found front kick ability of pencak silat athletes were low. Factors that influence this finding was physical condition such as explosive power leg mucle and agility. The aim of this research is to find out the correlation between explosive power leg muscle with agility to the ability of front kick on pencak silat PPLP Pekanbaru Riau Province. The instrument of this study were Jump MD for explosive power muscle leg, agility using shuttle run and front kick ability test. We performed total sampling of 10 athletes of Pencak Silat PPLP Pekanbaru Riau Province. We used the product moment correlation analisys test, the level of significance of α = 0.05. Based on research and data analysis then relationship which mean together of muscle leg explosive power and agility with the ability of front kick pencak silat PPLP Pekanbaru Riau Province the results showed Fhitung (4.422) < Ftabel (4.74) and Rhitung (0.747) with stronger category for interpretasi table.

Inflammation of the GI tract leads to compromised epithelial barrier integrity, which increases intestine permeability. A compromised intestinal barrier is a critical event that leads to microbe entry and promotes inflammatory responses. Inflammatory bowel diseases that comprise Crohn's disease (CD) and ulcerative colitis (UC) show an increase in intestinal permeability. Nerolidol (NED), a naturally occurring sesquiterpene alcohol, has potent anti-inflammatory properties in preclinical models of colon inflammation. In this study, we investigated the effect of NED on MAPKs, NF-κB signaling pathways, and intestine epithelial tight junction physiology using in vivo and in vitro models. The effect of NED on proinflammatory cytokine release and MAPK and NF-κB signaling pathways were evaluated using lipopolysaccharides (LPS)-stimulated RAW 264.7 macrophages. Subsequently, the role of NED on MAPKs, NF-κB signaling, and the intestine tight junction integrity were assessed using DSS-induced colitis and LPS-stimulated Caco-2 cell culture models. Our result indicates that NED pre-treatment significantly inhibited proinflammatory cytokine release, expression of proteins involved in MAP kinase, and NF-κB signaling pathways in LPS-stimulated RAW macrophages and DSS-induced colitis. Furthermore, NED treatment significantly decreased FITC-dextran permeability in DSS-induced colitis. NED treatment enhanced tight junction protein expression (claudin-1, 3, 7, and occludin). Time-dependent increases in transepithelial electrical resistance (TEER) measurements reflect the formation of healthy tight junctions in the Caco-2 monolayer. LPS-stimulated Caco-2 showed a significant decrease in TEER. However, NED pre-treatment significantly prevented the fall in TEER measurements, indicating its protective role. In conclusion, NED significantly decreased MAPK and NF-κB signaling pathways and decreased tight junction permeability by enhancing epithelial tight junction protein expression.

The immune system plays a critical role in inflammation by initiating responses to infections or tissue damage. The nuclear factor-κB (NF-κB) pathway plays a key role in inflammation and innate immunity, as well as other cellular activities. Dysregulation of this well-choreographed pathway has been implicated in various diseases, including cancer. CARD11 is a key molecule in the BCL10-MALT1 complex, which is involved in transducing the signal downstream of the NF-κB pathway. This study aims to elucidate how CARD11 overexpression exacerbates the prognosis of colorectal cancer (CRC). To identify the cellular pathways influenced by CARD11, transcriptomic analysis in both CRC cell lines and patients was carried out on CARD11– overexpressed HCT-116 and HT-29 CRC cell lines alongside empty vector-transfected cell lines. Furthermore, a comparison of transcriptomic data from adenoma and carcinoma CRC patients with low- (CARD11–) and high-(CARD11+) CARD11 expression was carried out. Whole transcriptomics and bioinformatics analysis results indicate that CARD11 appears to play a key role in CRC progression. Absolute GSEA (absGSEA) on HCT-116 transcriptomics data revealed that CARD11 overexpression promotes cell growth and tissue remodeling and enhances immune response. Key genes co-expressed with CARD11, such as EP300, KDM5A, HIF1A, NFKBIZ, and DUSP1, were identified as mediators of these processes. In the HT-29 cell line, CARD11 overexpression activated pathways involved in chemotaxis and extracellular matrix (ECM) organization, marked by IL1RN, MDK, SPP1, and chemokines like CXCL1, CXCL3, and CCL22, which were shown to contribute to the more invasive stage of CRC. In patient samples, adenoma patients exhibited increased expression of genes associated with the tumor immune microenvironment, such as IL6ST, collagen family members, and CRC transition markers, such as GLI3 and PIEZO2, in CARD11+ adenoma patients. Carcinoma patients showed a dramatic increase in the expression of MAPK8IP2 in CARD11+ carcinoma patients alongside other cancer-related genes, including EMB, EPHB6, and CPEB4. Macrolide antibiotic azithromycin is widely used in clinical practice to treat respiratory tract infections and inflammatory diseases. However, its mechanism of action is not fully understood. Given the involvement of the CD27 pathway in the pathophysiology of various T-lymphocyte-mediated inflammatory, autoimmune, and lymphoproliferative diseases, we examined the impact of AZM on CD27 regulation and potential consequences on CD4+ and CD8+ T-cell phenotypes. Using cellular immunology approaches on healthy donors' peripheral blood mononuclear cells, we demonstrate AZM-mediated downregulation of surface CD27 expression as well as its extracellular release as soluble CD27. Notably, AZM-exposed CD27high (hi) cells were defective in their ability to expand compared to CD27intermediate (Int) and CD27low (lo) subsets. The defective CD27hi subset expansion was found to be associated with impaired cell proliferation and cell division. At the molecular level, the CD27hi subset exhibited lower mTOR activity than other subsets. Functionally, AZM treatment resulted in marked depletion of helper CD4+ (Th1) and cytotoxic CD8+ T-lymphocyte (Tc1)-associated CXCR3+CD27hi effector cells and inhibition of inflammatory cytokine IFN-g production. These findings provide mechanistic insights on immunomodulatory features of AZM on T-lymphocyte by altering the CD27 pathway. From a clinical perspective, this study also sheds light on potential clinical benefits observed in patients on prophylactic AZM regimens against various respiratory diseases and opens avenues for future adjunct therapy against Th1- and Tc1-dominated inflammatory and autoimmune diseases. This study explored gaps between industry expectations and perceptions of engineering graduates' skill sets in the Middle East and North Africa (MENA) region. This study measured the importance that managers of engineers placed on 36 skills relevant to engineers. Also measured was managers' satisfaction with engineering graduates' skill sets. Importance and satisfaction were used to calculate skill gaps for each skill. A principal components analysis was then performed, consolidating the 36 skills into 8 categories. The means of importance, satisfaction, and skill gaps were ranked to determine the areas in which graduates needed improvement. Results showed significant gaps between managers' expectations of and satisfaction with all 36 skills. The areas in which managers felt that graduates needed most improvement were communication, time management, and continuous learning. Managers reported that recent engineering graduates exhibited low overall preparedness for employment. These findings may help to inform curricular reform in engineering education.
