Abstract

Konfirmasi Mikroba Endofit Penyebab Kontaminasi pada Kultur Jaringan Kangkung (Ipomoea aquatica Forssk.) Tanaman kangkung secara alami bersimbiosis dengan mikroba endofit, yang berpotensi menjadi kontaminan pada kultur jaringan kangkung karena berada di dalam jaringan dan sulit dijangkau saat proses sterilisasi eksplan. Tujuan penelitian ini adalah mengonfirmasi mikroba endofit penyebab kontaminasi pada kultur jaringan kangkung ‘Tetraploid’ sehingga dapat menjadi informasi awal untuk metode sterilisasi yang efektif. Sebanyak 14 sampel kontaminan pada media tanam kultur jaringan kangkung diisolasi dan diidentifikasi secara molekuler berdasarkan gen 16S rDNA untuk bakteri, daerah D1/D2 dari gen LSU rRNA untuk khamir, dan daerah ITS dari gen rDNA untuk jamur. Keragaman jenis mikroba yang teridentifikasi dibandingkan dengan keragaman jenis mikroba endofit dari jaringan tanaman kangkung yang ditanam pada media kultur jaringan tidak terkontaminasi mikroba, media kultur jaringan terkontaminasi mikroba, media tanam campuran tanah steril dan tidak steril, serta kangkung yang didapatkan dari pasar. Hasil penelitian ini mengonfirmasi bahwa khamir endofit dari kelompok Ustilaginaceae (basidiomycetous yeast) yang berasal dari jaringan tanaman kangkung sama dengan jenis kontaminan yang mengontaminasi media kultur jaringan kangkung ‘Tetraploid’. Khamir dari kelompok Ustilaginaceae (basidiomycetous yeast) tersebut juga merupakan mikroba paling dominan yang mengontaminasi media tanam kultur jaringan kangkung ‘Tetraploid’. Abstract

Lutein is a carotenoid compound classified as xanthophyll found in certain microalgae and vegetable plants. This compound is also found in the crown of the sunflower (Helianthus annuus). The main function of lutein in the body is an antioxidant that is responsible for macular eye health. This research aimed to extract lutein compounds from sunflowers, analyze phytochemical content, test its activity as an antibacterial and antioxidant. Sunflower crown powder was macerated with n-hexane solvent, then performed digestion using isopropanol and saponified with a 50% NaOH solution. The solution was evaporated, and the extract was washed with a mixture of water and alcohol (1:1) to obtain crude lutein. Furthermore, the potential of lutein as an antibacterial agent was tested using the disc diffusion method, against Bacillus subtilis, Escherichia coli, Salmonella typhi, and Staphylococcus aureus at concentrations of 1.000, 5.000, 10.000, 25.000 and 50.000 ppm. The test results showed the existence of inhibition zones of 11,12 mm to 20,52 mm. The testing of crude lutein's antioxidant activity was carried out using ABTS solution (2,2'-azinobis acid(3- ethylbenzatiazolin)-6sulfonate) showing IC50 of 80,64 µg/mL. [ABSTRACT FROM AUTHOR]

12. Basidiospores attach to the seed of Shorea leprosula in lowland tropical dipterocarp forest and form functional ectomycorrhiza on seed germination.

Abstract

This research aimed to study the ectomycorrhiza formed by basidiospores attached to the outer surface of Shorea leprosula (Dipterocarpaceae) seed collected from a lowland tropical dipterocarp forest. Two groups of seeds were collected: control seeds collected from plastic net hanging 2 m above the ground and forest floor seeds collected on the forest floor. Before planting, 15 seeds from each group were observed for basidiospores attached to the seed. Ten of the 3-week-old S. leprosula seedlings of each group were individually grown in 1.5 kg of sterile zeolite for 8 months in a greenhouse. Pots were fertilized with MMN solution containing half the strength of phosphate. Fungal identity, ectomycorrhizal root tip colonization and anatomy, plant growth, and phosphate uptake were measured. The control seeds did not have basidiospores attached, whereas the forest floor seeds had 2 × 10 basidiospores of Tomentella. Bioassay test results indicate that the seedlings from the control seeds did not form ectomycorrhiza, whereas those seedlings from the forest floor seeds formed 3 morphotypes of ectomycorrhizae. Based on ITS1, 5.8S, and ITS2 rDNA region analyses, the 3 morphotypes belonged to Tomentella sp. HBT2, Tomentella sp. HBT4, and Scleroderma sp. HBS3. Root colonization percentage was above 70% for all three morphotypes. Root colonization in general increased plant growth and phosphate uptake. This is the first report of Tomentella basidiospores attached on the seed surface as a functional inoculum and of Tomentella ectomycorrhiza from dipterocarps lowland tropical forest.
