Abstract

The aim of the present study was to develop extended release formulation of Ibrutinib to maintain constant therapeutic levels of the drug for over 12 hrs. HPMC K100M, HPMC (K4M) and Carbopol 71 G were employed as polymers. All the formulations were passed various physicochemical evaluation parameters and they were found to be within limits. Whereas from the dissolution studies it was evident that the formulation (F6) showed better and desired drug release pattern i.e., 98.85% in 12 hours. It contains the HPMC (K4M) as Extended release material. It followed peppas release kinetics mechanism. Keywords: Ibrutinib, Extended release system.

Abstract

A green and efficient synthesis of 4-heteryl-quinolines ( 9a, 9b, 9c, 9d), ( 10a, 10b, 10c, 10d) and ( 11a, 11b, 11c, 11d) has been described using PEG-600 as a green solvent. Initially, 4-chloro-2-methylquinolines ( 5a, 5b, 5c, 5d) on reaction with aromatic heterocyclic thiols ( 6), ( 7), and ( 8) using PEG-600 at 100°C for 30-40 min resulted in ( 9), ( 10), and ( 11) in good yields. Alternatively, ( 9), ( 10), and ( 11) could also be prepared in dimethylformamide using K2CO3 as base and tetrabutylammonium bromide as phase transfer catalyst at 100°C for 1-2 h. All the compounds were synthesized and characterized by IR, NMR, mass spectroscopy, and 13C NMR analysis. All synthesized compounds were screened for their against clinical strains that include Gram-positive (Bacillus subtilis MTCC 121, staphylococcus aureus MLS-16 MTCC 2940, Micrococcus lutes MTCC 2470, and Staphylococcus aureus MTCC 96) and Gram-negative bacteria (Candida albicans MTCC 3017, Klebsiella planticola MTCC 530, Escherichia coli MTCC 739, and Pseudomonas aeruginosa MTCC 2453). The results revealed that compounds ( 9a, 9d, 10a, 10c, 11b, and 11d) exhibited significant almost equal to the standard drug, that is, Ciprofloxacin. [ABSTRACT FROM AUTHOR]

27. Tetrabutylammonium Acetate: Green and Efficient Synthesis of Dihydropyrano [3, 2-C] Quinolones from 1-Methylquinoline-2, 4(1H, 3H)-Diones

Abstract

An efficient one-pot synthesis of dihydropyrano[3,2-c]quinolones 4a-p by three-component reaction of N-methylquinoline-2,4-dione (1) with malononitrile (3a) or ethylcyanoacetate (3b) and benzaldehydes 2a-h in water in the presence of TBA acetate has been described. The above synthesis was also tried alternatively by the reaction of 1 with 2a-h in refluxing water using TBA acetate for 30 min and yielded 5. Compound 5 on reaction with 3a or 3b did not yield the target compound 4. This was approached alternatively by the reaction of 2a-h with 3a or 3b that gave benzylidine derivatives 6a-h which on further reaction with 1 gave the title compound 4a-p. The methodology described here has the advantages of environment friendliness, higher yields, shorter cost, low reaction times and convenient operation.

Abstract

One-pot, four component syntheses for the preparation of 1-H Pyrazolo[1,2-b]phthalazine- 5,10-diones (5a-5h) and 2H-indazolo[2,1-b]phthalazine-1,6,11(13-H)-triones (7a-7f) have been described from phthaloyl dichloride (1) , hydrazine hydrate (2), benzaldehydes (3) and malononitrile (4a), ethyl cynoacetate (4b) or dimedone (6) in refluxing water for 1-1.5 h by in-situ generation of HCl as a catalyst. These reactions have easy workup, provide excellent yields, and use water as the solvent.

Eco-friendly, one-pot three-component syntheses of 2-(3H-imidazo[4,5-b]pyridine-2-yl)-N-arylbenzamides have been developed by combining phthalic anhydride with anilines and pyridine-2,3-diamine in water without any catalyst. These reactions involves easy workup, provide good yields and use of water as solvent which were the merits of this preparation.
