Abstract About 50% of all cancers carry a mutation in p53 that impairs its tumor suppressor function. The p53 missense mutation p53R175H (p53R172H in mice) is a hotspot mutation in various cancer types. Therefore, monoclonal antibodies selectively targeting clinically relevant mutations like p53R175H could prove immensely value. We aimed to evaluate the in vitro and in vivo binding properties of two novel anti-p53R175H monoclonal antibodies and to assess their performance as agents for molecular imaging. In vitro, 125I-4H5 and 125I-7B9 demonstrated long shelf life and antigen-specific binding. Our in vivo study design allowed head-to-head comparison of the antibodies in a double tumor model using repeated SPECT/CT imaging, followed by biodistribution and autoradiography. Both tracers performed similarly, with marginally faster blood clearance for 125I-7B9. Repeated molecular imaging demonstrated suitable imaging characteristics for both antibodies, with the best contrast images occurring at 48 h post-injection. Significantly higher uptake was detected in the mut-p53-expressing tumors, confirmed by ex vivo autoradiography. We conclude that molecular imaging with an anti-p53R175H tracer could be a promising approach for cancer diagnostics and could be further applied for patient stratification and treatment response monitoring of mutant p53-targeted therapeutics.

The high spatial resolution data presents a problem when it comes to mapping and identifying distinct tree species based on the characteristics of their canopies. The deep learning Semantic Segmentation approach based on U-Network (U-Net.) artificial intelligence model that we provide here can recognize, and map Azadirachta indica trees canopy cover. This method trains its model by making use of image chips and labels of the item being segmented. The new testing images processed for multiple stages of pixel level of convolution and pooling operations. The sampling methods allow increase to make complete to make the recognized object on the image. The model’s ability to identify items based on canopy shape, structure, and pixel data makes it very useful for mapping and recognizing a single tree species as well as several tree species. The model validation results indicated an accuracy of 84–89 percent, which is regarded to be rather good. Based on ground census data, the overall accuracy of identification is 89 percent, F1 score 0.91–0.94, while the complete tree canopy validation (Intersection to Union) for canopy matching area is 0.79–0.89. The method has the potential to be utilised for identification, mapping of tree canopy. The approach has the potential to be used for important research initiatives i.e tree censuses and the identification and mapping of crop plant identification. The deep learning model used as inferences for automatization of the identification of the tree species helps to resolve identification and mapping based complex problems in agro-forestry allied fields.

Abstract In this work, echo-planar spectroscopic imaging (EPSI) with flyback readout gradient-echo train was implemented in a preclinical MR scanner. The aim of this study is to visualize and quantify the ghost spectral lines produced by two, three and four interleaved echo trains with different amplitudes of the readout gradients, and to investigate the feasibility of the flyback data acquisition in micro-imaging of small animals. Applied multi-slice EPSI sequence utilizes asymmetric gradient-echo train that combines the shortest possible rewind gradients with readout gradients. It simplifies data processing because all echoes are acquired with the same polarity of the readout gradient. The approach with four interleaved gradient-echo trains and with four echoes in each train provides broad spectral bandwidth in combination with narrow receiver bandwidth and a good water-fat signal separation. It improves signal-to-noise ratio without the undesired consequence of water-fat shift artifacts that are eliminated during data processing. Position, number, and intensity of the ghost spectral lines can be controlled by the suitable choice of spectral bandwidth, number of echo train interleaves, and the number of echoes in each interleave. This study demonstrates that high-spatial resolution EPSI with interleaved flyback readout gradient-echo trains is feasible on standard preclinical scanners.

Nepal's burgeoning poultry industry which is a key sector in its economy leads to a considerable generation of slaughterhouse waste (SHW), necessitating effective and sustainable disposal methods. This study explores anaerobic digestion as an optimal solution for poultry SHW management, aiming to produce energy-rich biogas while efficiently mitigating pollution in these facilities. Particularly, the feasibility and effectiveness of co-digestion with chicken litter (CL) were investigated to enhance biogas production and waste utilization. Four distinct runs of anaerobic digestion were performed, each utilizing varying substrate compositions with SHW to CL ratios of 1:0, 4:1, 1:1, and 1:4. Throughout the process, essential parameters, including total solids (TS), volatile solids (VS), biological oxygen demand (BOD5), pH, temperature, biogas generation, were meticulously measured. The cumulative biogas production for each run was as follows: 14.87 liters and a biogas yield of 88.73 ml/gVS with a 17.39 % VS reduction for Run 1, 25.98 liters and a biogas yield of 147.21 ml/gVS with a 28.64 % VS reduction for Run 2, 78.32 liters and a biogas yield of 314.69 ml/gVS with a 54.32 % VS reduction for Run 3, and 89.195 liters and a biogas yield of 344.36 ml/gVS with a 63.05 % VS reduction for Run 4. Notably, as the proportion of CL increased in the mixture from 4:1 to 1:1, a considerable VS reduction was observed. Furthermore, when the ratio of SHW to CL reached 1:1 and 1:4, a significant BOD5 reduction of 50 % and 63.13 % was achieved, respectively, surpassing previous runs. The results reveal that the addition of CL in an appropriate ratio effectively manages poultry SHW, with the optimum SHW:CL ratio for significant biogas yield lying between 1:1 and 1:4.

IntroductionDental caries, caused by oral microbial pathogens, are a global health concern, further exacerbated by the presence of methicillin-resistant Staphylococcus aureus (MRSA). Bioactive proteins and peptides (BAPs) exhibit potent antimicrobial properties, targeting multiple cellular mechanisms within pathogens, reducing the likelihood of resistance development. Given the antimicrobial potential of BAPs, this study aimed to compare the efficacy of BAPs extracted from cultivated (Pleurotus ostreatus, PoC) and wild (Pleurotus ostreatus, PoW) mushrooms against pathogens responsible for dental caries.MethodsBAPs were extracted from both PoC and PoW using a TCA-acetone method. Antimicrobial activities were tested against seven bacteria and one fungus using agar well diffusion and MIC determination. Antibiofilm activity was assessed via modified CV assay, while DPPH and erythrocyte lysis tests evaluated free radical scavenging.ResultsPoC showed superior antimicrobial efficacy, with lower MIC and MBC values, and disrupted biofilm integrity at increasing concentrations. PoW exhibited better antioxidant activity with higher DPPH scavenging, though its antimicrobial efficacy was slightly lower than PoC.DiscussionBoth PoC and PoW BAPs inhibited dental pathogens, with PoC showing stronger inhibition against MRSA and nystatin-resistant Candida albicans. This suggests BAPs may target additional cellular mechanisms beyond membranes, PBPs, and ergosterols. Despite PoW’s stronger antioxidant properties, both BAPs had comparable antibiofilm activity. These findings suggest complementary actions of BAPs from PoC and PoW both, in treating dental caries, offering broad-spectrum antimicrobial and antioxidant benefits.

Background: Free radicals, harmful by-products of a cell's natural metabolism, are responsible for various health problems. The search for plant-based supplements or medicines is always in high demand, as is the antioxidant activity that contributes to the therapeutic efficacy of plants. Aim: In the present study, the hydroalcoholic extracts from the aerial parts of Cassia tora were used for in vitro analysis of their antioxidant activity. Methods: Six separate assay methods were used to evaluate the antioxidant activity, i.e., against hydroxyl radical, DPPH, superoxide anions, nitric oxide, and also total flavonoid and phenolic content, were investigated. This was done by standardizing hydroalcoholic extract (70/30 ethanol to water) of Cassia tora and ascorbic acid. Results: Percentage scavenging activity and IC50 value were measured for extract prepared at various concentrations. The results IC50 values were 25.54 µg/ml, 45.04 µg/ml, 36.56 µg/ml, and 97.61 µg/ml for DPPH, superoxide radicals, hydroxyl radicals, and nitric oxide, respectively. Subsequently, the total phenolic and flavonoid content in the extract obtained was 1.927±0.73 mg GAE/gm and 1.018±0.29 mg QE/gm, respectively. Conclusion: The hydroalcoholic extract of Cassia tora contains more phytoconstituents. This suggests it has a wide range of medicinal antioxidant properties that make it helpful in treating many diseases. With the increasing demand for safer herbal treatments, scientific efforts in this field are making significant contributions and advances and supporting innovation.

Introduction: Dysregulation of the complement system in schizophrenia with increased activation of the classical complement pathway has been observer earlier. The present study was undertaken to find out the effects of dysregulation of complement factors in schizophrenic patients, for confirmation and making an early diagnosis for high-risk patients by analysing of the complement factors. Methodology: All patients aged 18-65 years who presented with schizophrenia were enrolled. A total of 80 blood samples were collected from three study groups. Diagnosis was made using the ICD-10 criteria for schizophrenia. Participants fulfilling the inclusion criteria of ability to understand the nature of test and being drug naïve and willing to participate in the study were recruited after written consents from participants and their first degree relatives at Psychiatric Centre, SMS Medical College, Jaipur, Rajasthan (India). Results: This study observed a significantly lower mean value of C4 and CR1 in schizophrenic patients than control and significant dysregulation of various complement factors was seen with respect to certain sociodemographic profile. No significant dysregulation was observed in first degree relatives. Conclusion: The role of complement dysregulation in etio pathogenesis was found in the present study. This could be helpful in early diagnosis and identification of individuals at high risk of developing schizophrenia.

Abstract Invasive plant species are considered one of the significant drivers of habitat loss, leading to biodiversity loss. They have also been observed to alter the local ecology, resulting in a decline of native flora. The management of invasive species is widely recognised as one of the most severe challenges to biodiversity conservation. The International Union for Conservation of Nature (IUCN) considers Lantana camara, as one of the ten worst weeds. Over time, native and indigenous species may evolve to co-exist or compete with invasive species, reducing invader fitness. It is observed that species competition fluctuates throughout environmental gradients, life phases, and abundances. Hence, competition outcome is very context-dependent. To address this challenge, we conducted a comprehensive study in three phases: we identified native species coexisting with Lantana in their natural habitats in the Doon Valley (Phase I) and documented the phenotypic traits of selected coexisting species using the Landmark BBCH (Biologische Bun-desantalt, Bundessortenamt und Chemische Industrie) scale, revealing the phenological growth patterns of selected co-existing species (Phase II). This was followed by conducting pot (Phase IIIa) and field (Phase IIIb) experiments to study the interactions between them. Notably, Justicia adhatoda, Broussonetia papyrifera, Pongamia pinnata, Urtica dioica and Bauhinia variegata demonstrated promising results in both pot and field conditions. Furthermore, after the mechanical removal of Lantana and prior to the plantation in the field experiments, four native grass species were introduced using the seed ball method. Among these, Pennisetum pedicellatum and Sorghum halpense exhibited prompt regeneration and effectively colonised the field, densely covering the cleared area. The study provides a comprehensive management plan for the restoration of Lantana affected areas through competition using native species. This study utilizes phenological assessment for native plant selection using reclamation from native grasses and proposes a management plan for combating invasive Lantana.

Abstract Considering the major source of plant-derived low/non-calorie steviol glycosides (SGs), comprehensive physiological, biochemical, and deep transcriptional investigations were conducted to explicit deeper insight into multiple abiotic stress responses in Stevia rebaudiana. The physiological indicators including photosynthesis, chlorophyll, relative water content, shoot growth, electrolyte leakage, and SG biosynthesis were negatively impacted under drought (DS), followed by salinity (SS) and waterlogging (WS). Global transcriptional analysis revealed significant upregulated expression of the genes encoding for ROS detoxification (GST, SOD, APX, glutathione peroxidase), osmotic adjustment (alpha-trehalose-phosphate and S-adenosylmethionine decarboxylase), ion transporters (CAX, NHX, CNGS, VPPase, VATPase), water channel (PIP1, TIP) and abiotic stress-responsive candidate genes (LEA, HSPs, and Dehydrins) regulating abiotic stress response in S. rebaudiana. These inferences were complemented with predicted interactome network that revealed regulation of energy metabolism by key stress-responsive genes (GST, HKT1, MAPKs, P5CSs, PIP), transcription factors (HSFA2, DREB1A, DREB2A), and abiotic stress responsive pathways (ABA, ethylene, ion stress). This is the first detailed study to comprehend the molecular regulation of stress response and their interplay under DS, SS, and WS. The key genes and regulators can be functionally validated, and will facilitate targeted gene editing for genetic improvement of crop sustainability under changing environmental conditions in S. rebaudiana.

β-carotene is obtained from both plants and animals and has been the subject of intense research because of its provitamin-A, antioxidant, and anticancer effects. Its limited absorption and oxidative degradation significantly reduce its antitumor efficacy when taken orally. In our study, we utilize a central composite design to develop “bio-safe and highly bio-compatible” solid lipid nanoparticles (SLNs) by using only the combination of palmitic acid and poloxamer-407, a block co-polymer as a surfactant. The current research aim to develop and characterize SLNs loaded with β-carotene to improve their bioavailability and therapeutic efficacy. In addition, the improved cytotoxicity of solid lipid nanoparticles loaded with β-carotene was screened in-vitro in human breast cancer cell lines (MCF-7). The nanoparticles exhibits good stability, as indicated by their mean zeta potential of −26.3 ± 1.3 mV. The particles demonstrated high drug loading and entrapment capabilities. The fabricated nanoparticle's prolonged release potential was shown by the in-vitro release kinetics, which showed a first-order release pattern that adhered to the Higuchi model and showed a slow, linear, and steady release over 48 h. Moreover, a diffusion-type release mechanism was used to liberate β-carotene from the nanoparticles. For six months, the nanoparticles also showed a notable degree of physical stability. Lastly, using the MTT assay, the anti-cancer properties of β-carotene-loaded solid lipid nanoparticles were compared with intact β-carotene on MCF-7 cell lines. The cytotoxicity tests have shown that the encapsulation of β-carotene in the lipid bilayers of the optimized formulation does not interfere with the anti-cancer activity of the drug. When compared to standard β-carotene, β-carotene loaded SLNs showed enhanced anticancer efficacy and it is a plausible therapeutic candidate for enhancing the solubility of water-insoluble and degradation-sensitive biotherapeutics like β-carotene.

Healers of Jamu- a Traditional Medicine in Solo City, Central Java, Indonesia apply several therapies for treatment of different diseases, including joint pain. The aim of the ethnomedicinal study is the identification of herbal formulations/ medicinal plants/method of preparation and their mode of administration for joint pain therapy in Jamu Medicine. An ethnomedicinal survey was conducted to collect data from traditional healers of Jamu Medicine regarding the practice of joint pain therapy. Data was collected from 25 healers through interview by snowball technique. The data obtained from the study revealed that, 25 herbal formulations /potions consisting of 32 types of medicinal plants from 19 families have been registered by traditional healers for joint pain therapy. Most commonly used plants for joint pain therapy have been identified as Piper retrofractum Vahl, Zingiber officinale Roscoe, Curcuma xanthorrhiza D.Dietr., Kaempferia galanga L., and Curcuma longa L. Traditional herbal formulations are prepared by different processing methods like washing, peeling, crushing, pounding then squeezing with warm water (infusion)/ boiling in water (decoction) to get their extract. These herbal formulations are administered orally or applied locally as massage on the affected joint. Through this study we will preserve the traditional knowledge of Jamu Medicine because the younger generation of traditional healers is no more interested in continuing it and their number is gradually decreasing. This valuable data will help the scientist to make new medicinal breakthroughs in the future especially to treat joint pain.

Enteric fermentation is one of the largest contributors of methane release to the environment from the livestock sector. Plant bioactive compounds can modulate rumen fermentation for reduced methanogenesis and fatty acid biohydrogenation. The present study investigates the effects of tannin extract from Ficus glomerata (FG) leaves on the rumen fermentation, methanogenesis, feed digestibility and fatty acid biohydrogenation of a total mixed ration with the aim of developing a feed supplement for enhanced livestock production and product quality with lower methane emission. The tannin extract (70% aqueous acetone extract) of FG leaves in the total mixed ration (oat hay/concentrate mixture; 1:1) was studied at four graded dose regimens (0.0 (control), 0.25 mL (FG-0.25), 0.50 mL (FG-0.50) and 1.0 mL (FG-1.0) per 60 mL of buffered rumen fluid) in three replicates for each treatment in a radio-frequency-based automatic gas production system (ANKOM-RF) at 39 °C for 24 h following the standard in vitro gas production protocol. The total gas production (mL or mL/g incubated dry matter (DM)) was gradually reduced (p < 0.01) at dose levels of FG-0.50 and FG-1.0; however, it remained intermediary and comparable (p > 0.05) for FG-0.25 with the control and FG-0.50. Compared to the control, the methane concentration (%) in the head space gas, as well as the total methane production (mL or mL/g DM incubated, or mL/g DM digested), were found to be gradually reduced (p < 0.01) with increasing doses (0.25–1.0 mL) of FG extract. The reduced (p < 0.05) feed degradability at higher levels (0.50–1.0 mL) of FG extract supplementation and the comparative (p > 0.05) effects with the control at a lower level of supplementation (FG-0.25) are suggestive of the dose-responsive detrimental effects of tannins on fibrolytic microbes in the rumen. However, the ammonia concentration decreased (p < 0.05) in all of the incubations compared to the control. Among the volatile fatty acids, acetate remained comparable (p > 0.05) with enhanced (p < 0.05) propionate at a lower dose (FG-0.25); however, a dose-dependent reduction was evident at higher dose levels (FG-0.50 and FG-1.0). The production of stearic acid (C18:0), which is a product of the rumen biohydrogenation process, was reduced (p < 0.05), irrespective of the concentration of the FG extract. Compared to the control, the concentration of t-vaccenic acid (C18:1), which is a precursor of conjugated linoleic acid (CLA) in animal products, was increased in all the FG-extract-supplemented groups. It may be concluded that Ficus glomerata leaf tannins can modulate rumen fermentation for reduced methanogenesis and fatty acid biohydrogenation in a total mixed ration. As a higher level of inclusion negatively affects feed digestibility, a lower dose (0.25 mL FG extract per 60 mL fermentation fluid or 4.17 mL FG extract per L of fermentation fluid) is suggested to achieve desirable effects on methane abatement (30%) and an improvement in fatty acid profiles in animal products.

Abstract Introduction Good nutrition knowledge and nutrient intake have been regarded as significant determinants in enhancing athletes’ performance and overall health status. This study aimed to assess knowledge, attitude, and practices of nutrition and dietary intake among athletes. Methods A cross-sectional study was conducted from January to April 2022 among national athletes from two sports clubs in Kathmandu Metropolitan City, Nepal. A semi-structured questionnaire was used to collect the data. Anthropometric measurements and dietary intake were recorded. Bivariate and multivariate binary logistic regression was used to estimate the crude odds ratios (cOR), and adjusted odds ratios (aOR) with 95% confidence intervals (CIs). Results A total of 270 players were included in this study (mean age, 25 years; 49.6% male, 50.4% female). Almost half of the athletes had good nutrition knowledge [54.1% (146/270)], attitude [56.7% (153/270)], and practices [50% (135/270)] scores. The mean energy, carbohydrate, protein, and fat intake were 35.0 kcal/kg/day, 5.6, 0.9, and 0.9 g/kg/day, respectively. Likewise, mean calcium and iron intake were 370, and 12.5 mg, respectively. In the multivariate model, families with monthly household income less than 50,000 Nepalese rupees ($400) (adjusted Odds Ratio/aOR = 2.58; 95% CI: 1.12 to 5.96), and those who did not receive diet plan (aOR = 3.14; 95% CI: 1.25 to 7.84) were more likely to have poor nutrition knowledge. Players who did not check food labelling (aOR = 1.44; 95% CI: 0.78 to 2.63) were more likely to have negative attitude towards nutrition. Players who did not ever attend nutrition class (aOR = 3.54; 95% CI: 1.46 to 8.54) and those who did not consume different food during off and on the season of sports (aOR = 2.36; 95% CI: 1.39 to 4.01) were more likely to have poor nutrition practice. Conclusions Half of the athletes’ nutritional knowledge, attitudes, and practices score were satisfactory. Nutrient intake among athletes was suboptimal. Nutrition intervention programs are critical to improve nutritional knowledge, attitude and practice related to dietary intake among national athletes in Nepal.

Water scarcity and soil nutrient depletion significantly constrain global maize productivity amidst recurrent droughts due to climate change. So, this study was designed in a two-factorial pot culture, comprising twelve treatments and four replications in a completely randomized design, to examine the synergistic impacts of nitrogen (N) fertilizer sources and soil moisture levels on maize growth and nitrogen uptake. The treatments included nitrogen fertilizer sources (100% urea, 100% nanourea, 50% urea + 50% nanourea, control) and distinct soil moisture regimes (75% FC, 50% FC, and 35% FC). Elevated soil moisture positively correlated with increased plant height, leaf area, and Normalized Difference Vegetation Index (NDVI), while dry root biomass displayed opposing trends. Notably, the 50% Urea + 50% Nano Urea treatment exhibited superior aboveground biomass (350 gm, 314 gm, and 236 gm) and N uptake (2.15%, 1.89%, and 1.99%) across all moisture regimes (75% FC, 50% FC, and 25% FC, respectively). Moreover, plants at 50% FC demonstrated enhanced water use efficiency, surpassing 35% FC and 50% FC by 52% and 63%, respectively. Overall, the combined application of 50% urea and 50% nanourea under optimal soil moisture (75% FC) notably improved growth, nitrogen uptake, and photosynthetic activity. However, their efficacy declined under water stress conditions, with singular nanourea exhibiting a relatively milder susceptibility to water stress.

Abstract Chronic disease can cause tissue and organ damage constituting the largest obstacle to therapy which, in turn, reduces patients’ quality-adjusted life-year. Degenerative diseases such as osteoporosis, Alzheimer’s disease, Parkinson’s disease, and infectious conditions such as hepatitis, cause physical injury to organs. Moreover, damage resulting from chronic conditions such as diabetes can also culminate in the loss of organ function. In these cases, organ transplantation constitutes the therapy of choice, despite the associated problems of immunological rejection, potential disease transmission, and high morbidity rates. Tissue regeneration has the potential to heal or replace tissues and organs damaged by age, disease, or trauma, as well as to treat disabilities. Stem cell use represents an unprecedented strategy for these therapies. However, product availability and mass production remain challenges. A novel therapeutic alternative involving amniotic mesenchymal stem cell metabolite products (AMSC-MP) has been developed using metabolites from stem cells which contain cytokines and growth factors. Its potential role in regenerative therapy has recently been explored, enabling broad pharmacological applications including various gastrointestinal, lung, bladder and renal conditions, as well as the treatment of bone wounds, regeneration and skin aging due to its low immunogenicity and anti-inflammatory effects. The various kinds of growth factors present in AMSC-MP, namely bFGF, VEGF, TGF-β, EGF and KGF, have their respective functions and activities. Each growth factor is formed by different proteins resulting in molecules with various physicochemical properties and levels of stability. This knowledge will assist in the manufacture and application of AMSC-MP as a therapeutic agent.

The present study was carried out to determine the effect of feeding different levels of rice dried distiller grains (rDDGS) on haemato-biochemical profile of Barbari goats. Twenty-four Barbari goats of 1-2 years of age were randomly divided into four groups (Control, T1, T2 and T3) having six animals in each group. Barbari goats in control group were fed with basal diet comprising of wheat straw, chaffed green maize fodder and compounded concentrate mixture in a ratio of 20:30:50. The animals in T1, T2 and T3 groups were fed with basal diet supplemented with 10, 20 and 30% of rDDGS on dry matter basis, respectively for the period of 90 days. Results revealed no significant differences on blood profile except WBC (×103/µl), neutrophil (%) and lymphocytes (%) in the groups supplemented with rDDGS. Total protein was found highly significant in T2 group followed by T1 and T3 groups. Total immunoglobulin, catalase, TBARS and ALT in rDDGS supplemented groups were also found significantly different. It was concluded that rDDGS can be incorporated in Barbari goat ration up to level of 20% without having any detrimental effect on health of goats.

The increasing prevalence of food allergies worldwide and the subsequent life-threatening anaphylactic reactions often have sparse treatment options, providing only symptomatic relief. Great strides have been made in research and in clinics in recent years to offer novel therapies for the treatment of allergic disorders. However, current allergen immunotherapy has its own shortcomings in terms of long-term efficacy and safety, due to the local side effects and the possibility of anaphylaxis. Allergen-specific immunotherapy is an established therapy in treating allergic asthma, allergic rhinitis, and allergic conjunctivitis. It acts through the downregulation of T cell, and IgE-mediated reactions, as well as desensitization, a process of food tolerance without any allergic events. This would result in a protective reaction that lasts for approximately 3 years, even after the withdrawal of therapy. Furthermore, allergen-specific immunotherapy also exploits several routes such as oral, sublingual, and epicutaneous immunotherapy. As the safety and efficacy of allergen immunotherapy are still under research, the exploration of newer routes such as intra-lymphatic immunotherapy would address unfulfilled needs. In addition, the existence of nanoparticles can be exploited immensely in allergen immunotherapy, which would lead to safer and efficacious therapy. This manuscript highlights a novel drug delivery method for allergen-specific immunotherapy that involves the administration of specific allergens to the patients in gradual increasing doses, to induce desensitization and tolerance, as well as emphasizing different routes of administration, mechanism, and the application of nanoparticles in allergen-specific immunotherapy.

The pathetic malignant mesothelioma (MM) is a extremely uncommon and confrontational tumor that evolves in the mesothelium layer of the pleural cavities (inner lining- visceral pleura and outer lining- parietal pleura), peritoneum, pericardium, and tunica vaginalis and is highly resistant to standard treatments. In mesothelioma, the predominant pattern of lesions is a loss of genes that limit tumour growth. Despite the worldwide ban on the manufacture and supply of asbestos, the prevalence of mesothelioma continues to increase. Mesothelioma presents and behaves in a variety of ways, making diagnosis challenging. Most treatments available today for MM are ineffective, and the median life expectancy is between 10 and 12 months. However, in recent years, considerable progress has already been made in understanding the genetics and molecular pathophysiology of mesothelioma by addressing hippo signaling pathway. The development and progression of MM are related to many important genetic alterations. This is related to NF2 and/or LATS2 mutations that activate the transcriptional coactivator YAP. The X-rays, CT scans, MRIs, and PET scans are used to diagnose the MM. The MM are treated with surgery, chemotherapy, first-line combination chemotherapy, second-line treatment, radiation therapy, adoptive T-cell treatment, targeted therapy, and cancer vaccines. Recent clinical trials investigating the function of surgery have led to the development of innovative approaches to the treatment of associated pleural effusions as well as the introduction of targeted medications. An interdisciplinary collaborative approach is needed for the effective care of persons who have mesothelioma because of the rising intricacy of mesothelioma treatment. This article highlights the key findings in the molecular pathogenesis of mesothelioma, diagnosis with special emphasis on the management of mesothelioma.

Lung cancer is a fatal disease caused by an abnormal proliferation of cells in the lungs. Similarly, chronic kidney disorders affect people worldwide and can lead to renal failure and impaired kidney function. Cyst development, kidney stones, and tumors are frequent diseases impairing kidney function. Since these conditions are generally asymptomatic, early, and accurate identification of lung cancer and renal conditions is necessary to prevent serious complications. Artificial Intelligence plays a vital role in the early detection of lethal diseases. In this paper, we proposed a modified Xception deep neural network-based computer-aided diagnosis model, consisting of transfer learning based image net weights of Xception model and a fine-tuned network for automatic lung and kidney computed tomography multi-class image classification. The proposed model obtained 99.39% accuracy, 99.33% precision, 98% recall, and 98.67% F1-score for lung cancer multi-class classification. Whereas, it attained 100% accuracy, F1 score, recall and precision for kidney disease multi-class classification. Also, the proposed modified Xception model outperformed the original Xception model and the existing methods. Hence, it can serve as a support tool to the radiologists and nephrologists for early detection of lung cancer and chronic kidney disease, respectively.

Breeding for aluminium (Al) tolerance is a vital approach for enhancing the productivity of field crops in acidic soil regions where Al toxicity seems to be the most restraining factor for crop performance. Sunflower is generally considered extremely sensitive to Al toxicity; although no comprehensive information on the evaluation of sunflower genotypes for Al tolerance is available. In this study, 50 sunflower genotypes (set-I and set-II) were evaluated for Al tolerance at the seedling stage under hydroponic conditions. Substantial genetic variability in Al tolerance was observed among the studied genotypes. High estimates of heritability were obtained for both the total root length (TRL) and root regrowth (RRG), together with high estimates of genetic advance. A cluster analysis separated the genotypes into five different groups among the studied germplasm, the genotypes; NDLR-06 and EC-601861 were observed to be highly Al tolerant in terms of root regrowth under Al stress. In conclusion, the findings lreveal the complex mechanisms of Al tolerance in sunflower and may help to find new genetic resource for the improvement of Al tolerance in sunflower breeding.
