Abstract

Understanding disease burden and transmission dynamics in resource-limited, low-income countries like Nepal are often challenging due to inadequate surveillance systems. These issues are exacerbated by limited access to diagnostic and research facilities throughout the country. Nepal has one of the highest COVID-19 case rates (915 cases per 100,000 people) in South Asia, with densely-populated Kathmandu experiencing the highest number of cases. Swiftly identifying case clusters (hotspots) and introducing effective intervention programs is crucial to mounting an effective containment strategy. The rapid identification of circulating SARS-CoV-2 variants can also provide important information on viral evolution and epidemiology. Genomic-based environmental surveillance can help in the early detection of outbreaks before clinical cases are recognized and identify viral micro-diversity that can be used for designing real-time risk-based interventions. This research aimed to develop a genomic-based environmental surveillance system by detecting and characterizing SARS-CoV-2 in sewage samples of Kathmandu using portable next-generation DNA sequencing devices. Out of 22 sites in the Kathmandu Valley from June to August 2020, sewage samples from 16 (80%) sites had detectable SARS-CoV-2. A heatmap was created to visualize the presence of SARS-CoV-2 infection in the community based on viral load intensity and corresponding geospatial data. Further, 47 mutations were observed in the SARS-CoV-2 genome. Some detected mutations (n = 9, 22%) were novel at the time of data analysis and yet to be reported in the global database, with one indicating a frameshift deletion in the spike gene. SNP analysis revealed possibility of assessing circulating major/minor variant diversity on environmental samples based on key mutations. Our study demonstrated the feasibility of rapidly obtaining vital information on community transmission and disease dynamics of SARS-CoV-2 using genomic-based environmental surveillance.

Abstract

Understanding the lip movement and inferring the speech from it is notoriously difficult for the common person. The task of accurate lip-reading gets help from various cues of the speaker and its contextual or environmental setting. Every speaker has a different accent and speaking style, which can be inferred from their visual and speech features. This work aims to understand the correlation/mapping between speech and the sequence of lip movement of individual speakers in an unconstrained and large vocabulary. We model the frame sequence as a prior to the transformer in an auto-encoder setting and learned a joint embedding that exploits temporal properties of both audio and video. We learn temporal synchronization using deep metric learning, which guides the decoder to generate speech in sync with input lip movements. The predictive posterior thus gives us the generated speech in speaker speaking style. We have trained our model on the Grid and Lip2Wav Chemistry lecture dataset to evaluate single speaker natural speech generation tasks from lip movement in an unconstrained natural setting. Extensive evaluation using various qualitative and quantitative metrics with human evaluation also shows that our method outperforms the Lip2Wav Chemistry dataset(large vocabulary in an unconstrained setting) by a good margin across almost all evaluation metrics and marginally outperforms the state-of-the-art on GRID dataset., Comment: Accepted at ICPR 2022

Abstract

Abstract Management of vehicle traffic is a challenging task as it is non-deterministic by nature. Vehicular Cyber-Physical Systems (VCPS) is the emerging field of dynamics of vehicle management. Vehicle localization is considered an important task in VCPS. Many researchers proposed methodologies for this based on the Global Positioning System (GPS) which poses few location identification errors. Also, there are more vulnerabilities to the existing vehicular positioning system due to Zig-Zag attacks and bad-mouth attacks. In this work, an error-free and secure environment for communication between dynamically moving vehicle models has been proposed. In our proposed model a localization technique based on mathematical geometry which is capable of GPS outages and encompasses the dynamism of vehicle and on-road trajectory has been developed. The proposed model includes Extended Kalman filter-based routing to predict the neighbouring vehicle position. To avoid vulnerabilities created by the malicious nodes, a trust-based computation is performed by each node on its neighbours perceiving the authenticity of received messages. To validate the methodology, NS2 tool has been used to simulate the VCPS and to test the efficiency with different scenarios such as erroneous location, GPS outage, and malicious attack. The result shows that the proposed approach is more optimal and secure than the existing methodologies.

Abstract

Abstract This study reports that most patients with NSCLC had a significant increase in the nAb response to the currently circulating Omicron variants after bivalent booster vaccination and had Ab titers comparable to healthy participants. Interestingly, though the durability of the nAb response persisted in most of the healthy participants, patients with NSCLC had significantly reduced nAb titers after 4–6 months of vaccination. Our data highlight the importance of COVID-19 bivalent booster vaccination as the standard of care for patients with NSCLC given the evolution of new variants of concern.
