An extensive growth of serial remote sensing images paves the way for abundant data intended for the sequential spatial pattern determination in several fields like monitoring of agriculture, development of urban areas, and the vegetative area. However, conventional spatial sequential pattern mining is not applied efficiently or directly in the aspect of serial remote sensing images. Therefore, a residue feature analysis with empirical mode decomposition is proposed so as to enhance the spatial sequential pattern mining efficacy from the raster serial remote sensing images. At first, input images are being extracted by means of minima and maxima pattern by computing the mean of envelops and the intrinsic mode function components. If the intrinsic mode function condition is satisfied, then it is being subtracted from the original image; finally, the image is decomposed into many intrinsic mode functions and residue. The experimental outcomes attained indicate that the proposed strategy is proficient of mining spatial sequential pattern from the images of serial remote sensing. Though the support values of the patterns might not be attained accurately, the presented scheme guarantees that the whole patters are being extracted at lower consumption of time.

Photovoltaic technology, a rising renewable energy source, relies heavily on irradiation and temperature for performance. Defects like shading, hot spots, and soiling can disrupt the current–voltage curve, challenging inverters to operate at maximum power point (MPP). Indirect measurement and real-time loss detection methods are essential due to the inaccessibility of PV cell circuits. The conventional methods of performance measurement consider the performance based on standard test conditions. The main focus of the present work is to develop a novel performance ratio, which captures real-time efficiency involved in Solar farm operations using data-driven methodologies. The major contributions of this work are: Monitoring health and performance of solar farms by collecting 8 months of SCADA data using IEC standard 61724; Developing a data-driven model to predict ideal I-V curve and MPP at each transient state in the absence of panel-level data; Developing a new performance ratio which provides insights into the transient operational state of the inverter and the deviation of real-time data from the MPP. The results reveal the interplay of different inverters and the evolving overall performance of solar farm over time. This underscores necessity for maintenance and highlights the potential for enhancing solar farm’s output.

Worker's safety and water quality maintenance are the two predominant process that is followed inside the Mines. Many models had been proposed in the past to enhance the whole process but they failed to do so due to the practical limitations in the deployment in terms of cost. Though technology had improved much, human intervention is still unavoidable in mines. Considering the practical issues, in this article, a novel methodology using the Internet of Things (IoT) is proposed. This study is mainly focused on the change in the geometry/structure of the coal mining field due to pressure and water contamination that occurs in the region during coal extraction. Several sensors are adopted to measure the water quality and computational technique is used to analyze roof pressure variation in the coalfield. The roof pressure experienced by the nascent structure is compared with a structure that is roof bonded with a glass fiber composite layer. The significance of nanofiber technology is analyzed and reported. To ensure water quality, Harmonic Water Optimization (HWO) Algorithm is developed, here 10 different water flow areas are chosen and their corresponding data is gathered using IoT sensors. Harmony Management Preprocessing (HMP) Algorithm is developed to preprocess the redundant and ambiguous data. Heuristic Fusion Clustering (HFC)algorithm is developed for the data fusion, which can cluster and improve the data quality. The analysis of the proposed work reveals that it would greatly improve the sustainable development of mining safety and water quality which is the most vital factor.

GRTH/DDX25 is a testis-specific DEAD-box family of RNA helicase, which plays an essential role in spermatogenesis and male fertility. There are two forms of GRTH, a 56 kDa non-phosphorylated form and a 61 kDa phosphorylated form (pGRTH). GRTH-KO and GRTH Knock-In (KI) mice with R242H mutation (lack pGRTH) are sterile with a spermatogenic arrest at step 8 of spermiogenesis due to failure of round spermatids (RS) to elongate. We performed mRNA-seq and miRNA-seq analysis on RS of WT, KI, and KO to identify crucial microRNAs (miRNAs) and mRNAs during RS development by establishing a miRNA–mRNA network. We identified increased levels of miRNAs such as miR146, miR122a, miR26a, miR27a, miR150, miR196a, and miR328 that are relevant to spermatogenesis. mRNA–miRNA target analysis on these DE-miRNAs and DE-mRNAs revealed miRNA target genes involved in ubiquitination process (Ube2k, Rnf138, Spata3), RS differentiation, and chromatin remodeling/compaction (Tnp1/2, Prm1/2/3, Tssk3/6), reversible protein phosphorylation (Pim1, Hipk1, Csnk1g2, Prkcq, Ppp2r5a), and acrosome stability (Pdzd8). Post-transcriptional and translational regulation of some of these germ-cell-specific mRNAs by miRNA-regulated translation arrest and/or decay may lead to spermatogenic arrest in KO and KI mice. Our studies demonstrate the importance of pGRTH in the chromatin compaction and remodeling process, which mediates the differentiation of RS into elongated spermatids through miRNA–mRNA interactions.

GRTH/DDX25 is a member of the DEAD-box family of RNA helicases that play an essential role in spermatogenesis. GRTH knock-in (KI) mice with the human mutant GRTH gene (R242H) show loss of the phospho-species from cytoplasm with preservation of the non-phospho form in the cytoplasm and nucleus. GRTH KI mice are sterile and lack elongated spermatids and spermatozoa, with spermatogenic arrest at step 8 of round spermatids which contain chromatoid body (CB) markedly reduced in size. We observed an absence of phospho-GRTH in CB of GRTH KI mice. RNA-Seq analysis of mRNA isolated from CB revealed that 1,421 genes show differential abundance, of which 947 genes showed a decrease in abundance and 474 genes showed an increase in abundance in GRTH KI mice. The transcripts related to spermatid development, differentiation, and chromatin remodeling (Tnp1/2, Prm1/2/3, Spem1/2, Tssk 2/3/6, Grth, tAce, and Upf2) were reduced, and the transcripts encoding for factors involved in RNA transport, regulation, and surveillance and transcriptional and translational regulation (Eef1a1, Ppp1cc, Pabpc1, Ybx3, Tent5b, H2al1m, Dctn2, and Dync1h1) were increased in the CB of KI mice and were further validated by qPCR. In the round spermatids of wild-type mice, mRNAs of Tnp2, Prm2, and Grth were abundantly co-localized with MVH protein in the CB, while in GRTH KI mice these were minimally present. In addition, GRTH binding to Tnp1/2, Prm1/2, Grth, and Tssk6 mRNAs was found to be markedly decreased in KI. These results demonstrate the importance of phospho-GRTH in the maintenance of the structure of CB and its role in the storage and stability of germ cell-specific mRNAs during spermiogenesis.

Abstract Intervertebral disc degeneration is the most significant, and least understood, cause of chronic back pain, affecting almost one in seven individuals at some point of time. Each intervertebral disc has three components; central nucleus pulposus (NP), concentric layers of annulus fibrosus (AF), and a pair of end plate (EP) that connects the disc to the vertebral bodies. Understanding the molecular and cellular basis of intervertebral disc growth, health, and aging will generate significant information for developing therapeutic approaches. Rapid and efficient preparations of homogeneous and pure cells are crucial for meaningful and rigorous downstream analysis at the cellular, molecular, and biochemical level. Cross‐sample contamination may influence the interpretation of the results. In addition to altering gene expression, slow or delayed isolation procedures will also cause the degradation of cells and biomolecules that create a bias in the outcomes of the study. The mouse model system is extensively used to understand the intervertebral disc biology. Here we describe two protocols: (a) for efficient isolation of pure NP, AF, and EP cells from mouse lumbar intervertebral disc. We validated the purity of the NP and AF cells using ShhCre/+; R26mT/mG/+ dual‐fluorescent reporter mice where all NP cells are GPF+ve, and by the sensitive approach of qPCR analysis using TaqMan probes for Shh, and Brachyury as NP‐specific markers, Tenomodulin as AF‐specific marker, and Osteocalcin as bone‐specific marker. (b) For isolation of high‐quality intact RNA with RIN of 9.3 to 10 from disc cells. These methods will be useful for the rigorous analysis of NP and AF cells, and improve our understanding of intervertebral disc biology.

GRTH/DDX25 is a testicular RNA helicase expressed in germ cells that plays a crucial role in completion of spermatogenesis. Previously, we demonstrated a missense mutation (R242H) of GRTH gene in Japanese infertile patients (5.8%) with non-obstructive azoospermia. This mutation upon expression in COS-1 cells revealed absence of the 61 kDa phosphorylated GRTH in cytoplasm and the presence of the 56 kDa non-phosphorylated GRTH in the nucleus. GRTH knock-in (KI) mice carrying the human GRTH (R242H) mutation, lack phosphorylated GRTH, and sperm due to failure of round spermatid elongation during spermiogenesis. To determine the impact of phosphorylated GRTH on molecular events/pathways participating in spermatid development during spermiogenesis, we analyzed transcriptome profiles obtained from RNA-Seq of germ cells from KI and WT mice. RNA-Seq analysis of 2624 differentially expressed genes revealed 1404 down-regulated and 1220 up-regulated genes in KI mice. Genes relevant to spermatogenesis, spermatid development and spermatid differentiation were significantly down-regulated. KEGG enrichment analysis showed genes related to ubiquitin-mediated proteolysis and protein processing in endoplasmic reticulum pathway genes were significantly down-regulated while the up-regulated genes were found to be involved in Focal adhesion and ECM-receptor interaction pathways. Real-Time PCR analysis confirmed considerable reduction in transcripts of ubiquitination related genes Ube2j1, Ube2k, Ube2w, Rnf8, Rnf133, Rnf138, Cul3 and increased expression of Ccnd2, Col1a, Lamb1, Cav1, Igf1, Itga9 mRNA’s in KI mice compared to WT. Also, marked reduction in protein expression of UBE2J1, RNF8, RNF138 (ubiquitination network), MOF (histone acetyltransferase), their modified Histone substrates (H2AUb, H2BUb) and H4Ac, H4K16Ac were observed in KI mice. GRTH-IP mRNA binding studies revealed that Rnf8 and Ube2J1 mRNAs from WT mice associated with GRTH protein and the binding is greatly impaired in the KI mice. Immunohistochemistry analysis showed significantly reduced expression of RNF8, MOF, H4Ac and H4K16Ac in round spermatids of KI mice. Absence of phosphorylated GRTH impairs UBE2J1, RNF8 and MOF-dependent histone ubiquitination and acetylation essential for histone replacement, chromatin condensation and spermatid elongation during spermiogenesis.

Post-transcriptional and translational control of specialized genes play a critical role in the progression of spermatogenesis. During the early stages, mRNAs are actively transcribed and stored, temporarily bound to RNA binding proteins in chromatoid bodies (CBs). CBs are membrane-less dynamic organelles which serve as storehouses and processing centers of mRNAs awaiting translation during later stages of spermatogenesis. These CBs can also regulate the stability of mRNAs to secure the correct timing of protein expression at different stages of sperm formation. Gonadotropin-regulated testicular RNA helicase (GRTH/DDX25) is an essential regulator of spermatogenesis. GRTH transports mRNAs from the nucleus to the cytoplasm and phospho-GRTH transports mRNAs from the cytoplasm to the CBs. During spermiogenesis, there is precise control of mRNAs transported by GRTH from and to the CBs, directing the timing of translation of critical proteins which are involved in spermatid elongation and acrosomal development, resulting in functional sperm formation. This chapter presents our current knowledge on the role of GRTH, phospho-GRTH and CBs in the control of spermiogenesis. In addition, it covers the components of CBs compared to those of stress granules and P-bodies.

Vibrio fluvialis (V. fluvialis) is commonly found in coastal environments and is an emerging pathogen encountered in diarrhoeal outbreaks and sporadic extraintestinal cases. V. fluvialis infections usually manifest as watery bloody diarrhoea, nausea, vomiting, gastroenteritis and peritonitis. In this case report, an unusual presentation of Multidrug Resistant (MDR) V. fluvialis along with Klebsiella pneumoniae co-infection from a case of Walled Off Necrosis (WON)- A complication of Acute Necrotising Pancreatitis (ANP) is discussed. The patient presented with the acute abdomen and had peripancreatic collection for which Ultrasound (USG) guided 12F pigtail drain placed over the right lower quadrant of the abdomen and conservatively managed. Routine cultures were done and the growth on Sheep Blood Agar (SBA) plate and MacConkey (MAC) agar plates were subjected for automated phenotypic identification by Matrix Assisted Laser Desorption Ionisation Time of Flight-Mass Spectrometry (MALDITOF-MS) and Vitek-2 systems and was identified as V. fluvialis. Also, molecular tests like 16S rRNA Polymerase Chain Reaction (PCR) followed by Sanger’s sequencing was done and the identification was further confirmed. Phenotypic RESIST-5 TRURAPID O.K.N.V.I. commercial lateral flow immunochromatographic assay for detection of clinically relevant and common carbapenemase genes like NDM, OXA-48, VIM, IMP and KPC was done which revealed this V. fluvialis isolate as New Delhi Metallo betalactamase (NDM) type of Carbapenemase producer. Patient was managed with levofloxacin and doxycycline and patient condition improved and was discharged.

Background: Hereditary hemorrhagic telangiectasia (HHT) is a rare genetic disorder usually known to manifest with epistaxis, mucocutaneous telangiectasia, and visceral arteriovenous malformations (AVMs). Clinical Description: A 12-year-old girl presented with altered sensorium on hand ventilation, referred as a case of an acute cerebrovascular event due to cerebral AVM. She was afebrile with labored breathing and profound hypoxia. General and systemic examinations were unremarkable, except for clubbing in all digits. Management and Outcome: Investigations were largely normal, except for elevated hematocrit. Chest-X-ray showed mild infiltrates, while echocardiography was unremarkable. After stabilization and improvement of the sensorium, the child continued to have refractory hypoxemia, even with high ventilatory settings. The presence of digital clubbing and elevated hematocrit, in the background of cerebral AVM, led to suspicion of pulmonary AVM, the latter being detected by bubble contrast echocardiography and digital subtraction angiography (DSA). Genetic analysis confirmed a likely pathogenic variant in the ENG gene. Coil embolization of the pulmonary AVMs was done in stages, following which saturations improved and the child became asymptomatic. Conclusion: Persistent hypoxemia, in a child with cerebral AVMs, should raise suspicion of pulmonary AVMs, as a part of the disorder of HHT. Appropriate investigations can identify the AVMs and prompt intravascular intervention can be successful. Genetic analysis confirms the underlying HHT.

Transfusion therapy in an alloimmunized liver transplantation (LT) recipient still remains a major challenge due to the unpredictable blood unit requirements and difficulty in finding compatible antigen-negative units. We present the case report of a 30-year-old male patient with multiple red blood cell alloantibodies admitted for LT. Immunohematological tests by column agglutination method (gel) were employed for testing for antibodies. The antibodies were confirmed to be anti-E, anti-S, and anti-Fyb, and over a period of 10 days, nearly ninety units were crossmatched and phenotyped matched units were reserved for surgery. During the intraoperative phase, the patient could be supported with all three antigen-negative blood units. However, in the postoperative period, transfusions were switched over to two antigen-negative compatible units (E-Fyb−) under close monitoring. Direct antiglobulin test remained negative with no evidence of hemolysis throughout the perioperative period. This case delineates the approach to a case with multiple red blood cell antibodies and accentuates the role of well-equipped immunohematology laboratory for timely detection of alloantibodies and transfusion support for patients. An effective strategy for such patients needs to be devised well ahead to prevent the possible risk of any hemolytic reactions during the perioperative course. This case also re-emphasizes on the need of adequate inventory, advance assays like monocyte monolayer assay, and the active national rare donor registry to aid in the management of patients with single/multiple antibodies.

Aim: The aim of this study was to determine the fall risk amongst the elderly and to assess the effect of preventive rehabilitation. Study Design: This was a prospective pre–post study. Materials and Methods: Sixty patients attending the outpatient department were selected according to the inclusion and exclusion criteria. Timed up and go (TUG) test and functional reach test (FRT) and modified FRT were done to identify the risk groups. Appropriate preventive rehabilitation measures such as strengthening and balance exercises, footwear modifications and assistive aids along with treatment of the disease for which they approached the clinic were done. The study population was assessed after 2 weeks for fall risk using the same TUG and FRT. Results: A statistically significant reduction in fall risk was seen after appropriate rehabilitative strategies. Fall risk can be detected early by simple clinical tools such as TUG test and FRT. Conclusion: Simple clinical tools such as TUG test and FRT can be used for the early identification of fall risk amongst the elderly. This study establishes that a comprehensive preventive rehabilitation to the geriatric community with risk of fall can reduce the “fall risk” in the future and can also improve their functioning.
