Abstract

A study was conducted to show that the reaction of RhoA with redox agents leads to different functional consequences from the Rac1 and Cdc42 due to the presence of an additional cysteine (GXXXCGK(S/T)C) in the RhoA redox-active motif. The findings suggest that in addition to redox agents, platinated-chemotherapeutic agents and arsenic complexes might modulate the activity of GTPases containing the GXXXCGK(S/T)C motif (RhoA and RhoB).

Abstract

Teenage pregnancy is a significant social issue in the United States, resulting in increased levels of poverty. Most public health family planning efforts have traditionally focused on teaching teens the how-to of contraception, with little focus on teaching the why-to. During my time as a nurse practitioner in a public health department family planning clinic, I developed a method to open discussions with patients about the possibilities of a future that includes delayed childbearing. Abstract: The Ras superfamily of small GTPases cycle between inactive GDP-bound and active GTP-bound states to modulate a diverse array of processes involved in cellular growth control. While the basic mechanisms by which GTPase regulatory proteins regulate GTPase substrates have been revealed through numerous studies, detailed studies into the mechanism(s) of free radical-mediated GTPase regulation have only more recently been tackled. This article reviews the mechanism of free radical-mediated GTPase regulation and shows nitric oxide can serve as important regulator of small GTPase proteins (i.e. Ras and RhoA) through protein modifications such as S-nitrosation. ABSTRACTThe ability of the endospore-forming, gram-positive bacterium Bacillus anthracisto survive in activated macrophages is key to its germination and survival. In a previous publication, we discovered that exposure of primary murine macrophages to B. anthracisendospores upregulated NOS 2 concomitant with an ·NO-dependent bactericidal response. Since NOS 2 also generates O2·−, experiments were designed to determine whether NOS 2 formed peroxynitrite (ONOO−) from the reaction of ·NO with O2·−and if so, was ONOO−microbicidal toward B. anthracis. Our findings suggest that ONOO−was formed upon macrophage infection by B. anthracisendospores; however, ONOO−does not appear to exhibit microbicidal activity toward this bacterium. In contrast, the exosporium of B. anthracis, which exhibits arginase activity, protected B. anthracisfrom macrophage-mediated killing by decreasing ·NO levels in the macrophage. Thus, the ability of B. anthracisto subvert ·NO production has important implications in the control of B. anthracis-induced infection.

46. Simulation Design in Nursing Education: The Impact of Mid-Scenario Reflection on Learning Satisfaction and Self-Confidence

Abstract

While high-fidelity simulation (HFS) is used increasingly in nursing education, there is a dearth of quantitative evidence regarding the most effective role of the instructor in simulation. This descriptive correlational study examined the effect of a simulation design incorporating mid-scenario reflection on learning outcomes. The results were obtained by employing descriptive statistical analysis (mean and standard deviation) and correlational statistical analysis (multiple linear regression). A convenience sample of 47 junior nursing students enrolled in a women’s health clinical course was used for this study. Students participated in a HFS obstetrical scenario and cared for a laboring patient who developed an infection. Once the students reached the point when it became necessary to telephone the CNM to request treatment orders, the instructor paused the simulation. The group then adjourned to an adjacent classroom for a five to ten minute instructor-led guided reflection period that included a discussion regarding assessment findings and the diagnoses based on the findings. At that time, the group mutually developed a plan of care and returned to the simulation lab in order to resume the scenario and begin interventions. Following the simulation, participants completed a demographic questionnaire, the Simulation Design Scale, and the Student Satisfaction and Self-Confidence in Learning Scale. Results from the Student Satisfaction and Self-Confidence in Learning Scale indicate that participants were satisfied with the simulation exercise and generally felt self-confident after the HFS. The participants perceived that all five design characteristics included in the Simulation Design Scale were incorporated into the HFS. The correlational analyses revealed that all five simulation design elements were significantly correlated with the learning outcomes of self-confidence and learner satisfaction. A multiple regression analysis indicted that the simulation design elements accounted for over half the variance in learning outcomes. The mid-scenario reflection did not significantly contribute to the level of self-confidence or learner satisfaction. Rather, the design characteristic of Outcomes was the single best predictor for both learning outcomes while Support significantly contributed to the level of learner satisfaction.
