Fruits are essential for health but are highly perishable and suffer from post-harvest microbial deterioration. Edible fruit coatings are being considered as an unconventional, eco-friendly and safe alternative to postharvest chemical treatments as they can act as a physiological barrier, affect ethylene regulation and can be antimicrobial/antioxidant. Hence, the present work compared the effect of treatments viz. chitosan coating (CS), which acts as a physiological barrier, TL (Ocimum sanctum) and GT (Camellia sinensis) leaf extract treatment which are antimicrobial as well as an antioxidant in nature. The antibacterial activity of CS, TL and GT was examined and the maximum zones of inhibition 21mm and 18mm at 100µg/mL were examined in gram-positive Bacillus subtilis (MTCC 441) and gram-negative Escherichia coli (MTCC1885) bacteria in TL and GT. The antioxidant behavior of the TL and GT leaf extract was analyzed using a DPPH scavenging assay. The shelf-life of fruit was increased up to 30 days at ambient temperature by using CS (0.5% w/v), TL and GT 10mg/mL aqueous extract without damaging the fruit quality. TL coating was able to minimize disease incidence, and total weight loss and maximize the fruit quality parameters as maximal firmness, TSS, total protein, ascorbic acid, and SOD content even up to 30 days as compared to CS, GT and control. Density functional theory (DFT)-based calculations were performed for 36, 72 and 108 water molecules forming twisted ribbon-like nano-ring structures of diameter 1.54, 3.10 and 4.56 nm, respectively. We explore the electric field evolution of the structure, energetics and stability of the water nano-ring structures that are essentially symmetric and non-polar, or less-symmetric and polar, hydrogen-bonded clusters. The results suggest that for fields applied perpendicular to the ring, larger rings are more susceptible to the field influence, while fields applied parallel to the ring quickly transform the ring structures into arbitrary ones, regardless of the ring size. Infrared and Raman spectral analysis of local modes demonstrate the typical vibrational response of water molecules to various H-bonding environments and applied fields, providing a spectroscopic signature that can be used to identify the manifestation of such H-bonded ring networks. We study the structural, energetic and electronic properties of the structured water chain clusters within the density functional theory. We refer the structured water chains to those water clusters that have specific geometric patterns stretched along one direction. External electric field required to keep the structures open chain, thereby preventing them to form closed structures, is applied along the length of the chain. The structures are essentially periodic with basic repeating unit consisting of the corner- or edge-sharing 4-, 5- or 6-membered ring water clusters. Our calculations underscore the possible existence of such structured water clusters in the electrostatic environments, which we simulate in its simplicity employing a dipolar, uniform and static electric field. Analysis reveals that the 5-membered ring water chain clusters, i.e. the pentamer chain clusters have the lowest average dipole moment per water molecule while the threshold field, that marks the onset of the field-induced closure of the HOMO (highest occupied molecular orbital)-LUMO (lowest unoccupied molecular orbital) energy gap, is highest, followed by that for the tetramer and hexamer chains. Various synthetic routes are utilized to synthesize the tetrafunctionalized A[B.sub.3]-type porphyrin building blocks containing two different types of functional groups with [N.sub.4], [N.sub.3]O, [N.sub.3]S, and [N.sub.2][S.sub.2] porphyrin cores. The results from the studies on porphyrin tetrads and porphyrin pentads revealed that the monomeric porphyrin subunits in tetrads and pentads retained their individual characteristics features and exhibited weak interaction among the porphyrin subunits.

SONET/SDH is historically the dominant telecom transport infrastructure for backbone networks, and it is optimized for reliable delivery of voice and private-line services. Efficient utilization of the existing infrastructure through novel methods and algorithms can lead to higher revenue, which is attractive for network operators. In this study, we examine a network of inter-connected SONET/SDH rings that use dual-node interconnection employing the drop-and-continue facility, which is the de-facto standard for interworking SONET/SDH protection architectures. We develop an efficient algorithm for provisioning in such a network while keeping fragmentation of capacity low and taking into account the various constraints imposed by the underlying physical layer. We examine the case of dual-node interconnected ring networks employing both contiguous and virtual concatenation (a next-generation SONET/SDH architecture), and as expected, we discover that provisioning higher-bandwidth connections with virtual concatenation offers significant improvement. The stringent time-slot alignment and contiguity constraints imposed by contiguous concatenation are further compounded by the constraints imposed by the ring interworking architecture, and virtual concatenation allows capacity to be used more efficiently. We investigate reliable multipath provisioning of traffic in high-capacity backbone mesh networks, e.g., next-generation SONET/SDH networks supporting virtual concatenation (VCAT). VCAT enables a connection to be inversely multiplexed on to multiple paths, a feature that may lead to significantly improved performance over conventional single-path provisioning. Other mesh networks such as those employing optical wavelength-division multiplexing (WDM) and multiprotocol label switching (MPLS) can also benefit from this multipath provisioning approach. We propose effective multipath bandwidth as the metric to provision a connection while satisfying its reliability requirements (measured in terms of availability). We demonstrate that effective multipath bandwidth provides more flexibility and lower blocking probability without the cost and the complexity associated with traditional protection schemes developed for optical WDM and MPLS networks. We also investigate the practical problem of provisioning effective multipath bandwidth with cost constraints. We analyze the tractability of the problem and present a heuristic which results in significantly reduced number of blocked connections due to cost constraints. [ABSTRACT FROM AUTHOR]

Background:In view of the recent upsurge in the phenomenon of therapeutic failure, drug resistance in Leishmania, developed under natural field conditions, has become a great concern yet little understood. Accordingly, the study of determinants of antimony resistance is urgently warranted. Efflux transporters have been reported in Leishmania but their role in clinical resistance is still unknown. The present study was designed to elucidate the mechanism of natural antimony resistance in L. donovani field isolates by analyzing the functionality of efflux pump(s) and expression profiles of known genes involved in transport and thiol based redox metabolism Methodology/Principal Findings:We selected 7 clinical isolates (2 sensitive and 5 resistant) in addition to laboratory sensitive reference and SbIII resistant mutant strains for the present study. Functional characterization using flow cytometry identified efflux pumps that transported substrates of both P-gp and MRPA and were inhibited by the calmodulin antagonist trifluoperazine. For the first time, verapamil sensitive efflux pumps for rhodamine 123 were observed in L. donovani that were differentially active in resistant isolates. RT-PCR confirmed the over-expression of MRPA in isolates with high resistance index only. Resistant isolates also exhibited consistent down regulation of AQP1 and elevated intracellular thiol levels which were accompanied with increased expression of ODC and TR genes. Interestingly, γ-GCS is not implicated in clinical resistance in L. donovani isolates. Conclusions/Significance:Here we demonstrate for the first time, the role of P-gp type plasma membrane efflux transporter(s) in antimony resistance in L. donovani field isolates. We study the problem of reliably provisioning traffic in high-capacity backbone mesh networks supporting virtual concatenation (VCAT). Traditional approaches handled reliability requirements using full protection, which offers high assurance but can be costly. We take a less expensive approach by offering an expected bandwidth, rather than an absolute amount, but at lower costs than full-protection approaches. We propose a new routing algorithm that uses minimum-cost flow to find efficient collections of paths that satisfy traffic requests for expected bandwidth. We find that in most realistic network settings, paths are reliable enough so that only few additional network resources are required to compensate for the bandwidth loss incurred from possible network failures. We compared our approach to a previous algorithm under both a uniform setting, with symmetric traffic distribution and equal edge capacities, and in a more realistic setting with asymmetric traffic and differing edge capacities. Results show that our algorithm is an attractive approach in both settings, and much more effective than the previous scheme. We also compare our approach to a full-protection approach. Results show that by using fewer network resources, our algorithm is able to satisfy significantly more traffic requests than full protection. Nanoparticles (NPs) exhibit fascinating size-dependent chemical and physical characteristics that make them useful for a variety of applications. The present paper reports the green synthesis of CuO NPs and B-doped CuO NPs (B-CuO NPs) from Livistona chinensis leaf extract. Not much work has been reported on the use of the plant extract for the fabrication of NPs, particularly those of Cu and its doped counterparts. Various spectroscopic techniques were used to characterize the synthesized NPs. In the FT-IR spectra, peaks obtained at 504 cm -1 to 600 cm -1 were due to Cu-O vibrations. The energy dispersive X-ray analysis (EDX) spectra confirmed the CuO NPs' composition and B's presence inside the NPs. The peak pattern in the X-ray diffraction (XRD) spectrum confirmed the crystalline and monoclinic phases of the NPs. The average crystalline size of CuO NPs and B-CuO NPs was 19.56 nm and 17.30 nm respectively. The CuO and B-CuO NPs were tested against three Gram-positive bacterial strains namely Bacillus subtilis, Micrococcus luteus, and Staphylococcus aureus and three Gram-negative Escherichia coli, Salmonella abony, and Pseudomonas aeruginosa through Agar well diffusion method and it was found that CuO NPs showed higher activity than B-CuO NPs. Gentamicin was used as the positive control. Clinical resistance to pentavalent antimonial compounds has long been recognized as a major problem in the treatment of visceral leishmaniasis in India. However, mechanisms of natural resistance are unclear. In this study, we observed that Leishmania donovani clinical isolates not responsive to sodium stibogluconate showed resistance to antimony treatment in both in vitro and in vivo laboratory conditions. The resistant isolates have increased levels of intracellular thiols. This increase in thiol levels was not mediated by the amplification of gamma-glutamylcysteine synthetase, but was accompanied by amplification of trypanothione reductase and an intracellular ATP-binding cassette transporter gene MRPA. The resistance of parasites to antimony could be reversed by the glutathione biosynthesis-specific inhibitor, buthionine sulfoximine, which resulted in increased drug susceptibility. These results suggest the possible role of thiols and MRPA in antimony resistance in field isolates.
