Introduction: NRF2 and NQO1 proteins are very important for cell protection; Therefore, the purpose of this research was investigating the effect of resistance training with coenzyme Q10 supplementation on the content of NRF2 and NQO1 proteins in the hippocampus of male Sprague-Dawley rats. Methods: In this experimental study, thirty-two 2-month-old Sprague-Dawley rats with an average weight of 200±20 gr were selected. Rats were randomly divided into four groups: 1-control, 2-resistance training, 3-resistance training+Q10 supplement and 4-Q10 supplement. The resistance training program consisted of climbing a ladder with 26 steps and a slope of 85 degrees, for 8 weeks and 3 sessions every week. The daily consumption of Q10 supplement was 200 mg/kg of body weight of rats. The content of proteins was measured through Western-Blot laboratory method. Data analysis was done by one-way ANOVA and Tukey's post hoc tests and the effect size of eta squared test using SPSS version 16 and Graphpad Prism software version 10.2.3. The significance level was P≤0.05. Results: Eight weeks of resistance training with Q10 supplementation led to a significant change in NRF2 content (F=4.80, p≤0.03, ƞ2=0.64). Tukey's post hoc test showed this significant change between pairs of resistance training groups and Q10 supplement group (p≤0.04); medium effect size was observed in NRF2 content (ƞ2=0.64). No significant difference was observed in NQO1 content (F=3.01, p≤0.09, ƞ2=0.33). A weak effect size was observed in NQO1 content (ƞ2=0.33). Conclusion: Doing resistance training together with Q10 supplementation alone or in combination cannot significantly change the content of NRF2 and NQO1 proteins in the hippocampus of the brain.

Abstract Telocytes are interstitial cells found in different tissues, including cardiac stem cell niches. The purpose of this study was to investigate the response of the telocytes to the cardiac growth that occurs in response to resistance and endurance exercise trainings using rats distributed into control, endurance, and resistance training groups. Results revealed that the ratio of heart weight to body weight, cardiomycyte number, cardiomyocyte area, thickness of the left ventricular wall were significantly higher in the training groups compared to the control group. We observed increment in the cardiomyocytes surface area and thickness of the left ventricular wall in the resistance-training group than endurance-training group. We conclude that both resistance and endurance exercise trainings will lead to an increased number of cardiac telocytes, consequently, promote activity of the cardiac stem cells, and results in physiological cardiac growth, and this response does not seem to depend on the type of exercise.

Background and Purpose: Pathogenic cardiac hypertrophy is a type of heart disorder that is associated with changes in the structure of the heart, apoptosis and fibrosis of the heart muscle cells, which leads to a decrease in the pumping ability of the heart and may eventually lead to heart failure and sudden death. It has been shown that the signaling pathway of transcription factors NF-κB and NFAT play an important role in the process of pathogenic hypertrophy of the heart, and considering that few studies have investigated the effect of exercise on these factors in the process of pathogenic hypertrophy,the aim of the present study was to identify the effect of eight weeks of aerobic exercise before isoproterenol induction on the expression of NF-κB and NFAT genes in the left ventricular tissue of male Wistar rats.Materials and Methods: 18 male Wistar rats were randomly divided into two groups of endurance training and control. Endurance training was performed for eight weeks, one hour a day and six days a week on a treadmill with a slope of 15 degrees. After eight weeks of training, pathological hypertrophy was induced by subcutaneous injection of three mg / kg isoprenaline for seven days. 24 hours after the last injection session, rats were anesthetized by intraperitoneal injection of 50 mg ketamine and 10 mg xylazine. The rats were then dissected and cardiac tissue was extracted and transferred to -70 ° C. Also, in the present study, Real Time PCR was used to evaluate the expression of NF-κB and NFAT genes. Data analysis was performed using SPSS software version 24 and independent t-test at a significance level of P ≥ 0.05Results: The results of the present study showed that the indices of heart weight, heart weight to body weight ratio (H / W) and left ventricular weight to body weight ratio (V / W) were significantly (P = 0.01) and (P = 0.03): Respectively, increased in the exercise group compared to the control group. Also, the relative expression of NF-κB gene was significantly (P = 0.03) decreased in the exercise group compared to the control group, while the relative expression of NFAT gene was not significantly changed in the study groups (P = 0.40).Conclusion: Eight weeks of aerobic endurance training may play an important role in the prevention of pathological cardiac hypertrophy by reducing the relative expression of the NF-κB gene.

Background & objective: Diabetes causes chronic problems in the structure and function of tissues, such as apoptosis and fibrosis in addition to glycemic disorders. In this study the effect of 8 weeks of endurance and resistance training on various signaling pathways of apoptosis and tissue fibrosis of the heart of diabetic rats was investigated. Methods: Thirty Wistar rats, approximately 8-10 weeks old, weight about 210-250 grams, received intraperitoneal injection of diabetic streptozotocin and were randomly divided into three groups: endurance training, resistance training and control group. The rats of the endurance training group were trained on the treadmill for 8 weeks, 5 days a week with intensity of vo2Max 60-80%. The resistance training group was trained on the ladder with a slope of 85 degrees and with a weight equals to 30-100% of their body weight. Forty eight hours after the last training session, blood samples were collected and ventricular tissues of mice were extracted. Glucose, insulin, serum insulin resistance index and Bcl-2, Bax, caspase 8 gene expression levels and Bax to Bcl-2 ratio were evaluated. Masson's trichrome and hematoxylin-eosin staining methods were used for histological examination of diabetic rat's heart structure to detect fibrosis. Results: There was a significant decrease in Bax gene expression and the ratio of Bax to Bcl-2, and also there was a significant increase in Bcl-2 and caspase 8 in the endurance and resistance training groups in comparison with the control group. The rate of cardiomyocyte fiber rupture in the endurance and resistance groups was less than the control group, and the presence of lymphocyte cells was observed in some fibers in the control group. (p≤0.05). Conclusion: The results showed that high-intensity resistance training and moderate-intensity endurance training can prevent tissue fibrosis caused by collagen deposition in diabetes, and these two types of training can reduce the factors involved in apoptosis both in the internal and external pathways. On the other hand, this training intensity can be used as an effective non-pharmacological method to reduce the problems of apoptosis and fibrosis caused by diabetes in the heart tissue.

Background and Aim Adropin is a new hormone plays an important role in improving lipid metabolism and vascular protection. The current study aims to determine the effect of eight weeks of high-intensity interval training (HIIT) on Adropin level and nitric oxide (NO) metabolites in overweight men. Methods & Materials This is a quasi-experimental study with a pre-test/post-test design. Participants were 32 overweight men (Mean±SD age: 30.5±3.07 years, BMI: 27.93±4.2 kg/m2). They were randomly divided into two groups of HIIT (n=16) and control (n=16). The VO2max test was first conducted. Then, the HIIT group performed 8 weeks of HIIT (3 days per week) at 20 sessions of 30 seconds with a 13-s interval between each session. Body composition was measured using a bioelectrical impedance analyzer and the blood sampling was done for measuring Adropin and NO metabolites by ELISA kits in pre-test and post-test phases. Background and objectives: Vascular senescence refers to specific aging-associated changes in the endothelial cells. The purpose of this study was to investigate effects of 12 weeks of resistance training with elastic bands on circulating endothelial cell-derived microRNA-92a and endothelin-1 (ET-1) in elderly women with osteosarcopenic obesity (OSO). Methods: In the present randomized clinical trial, 48 elderly women with OSO were randomly divided into a control group (n=22) and an experimental group (n=26). The subjects in the intervention group performed 12 weeks of resistance training with elastic bands three times a week. The participants became familiar with targeted number of repetitions and OMNI-resistance exercise scale to control exercise intensity. Results: The mean age, body fat percentage, body mass index (BMI), and bone mineral density T-score for the hip and L1-L4 lumbar spine of the subjects were 64.13±3.68 years, 45.4%±6.56, 33.1±3.71 kg/m2, and -1.86±1.42, respectively. After the 12-week resistance training, we observed a significant decrease in serum level of mir-92a in the experimental group compared to the control group (p=0.03). However, there was no significant difference between the study groups in terms of body weight, BMI, body fat percentage, total cholesterol, and ET-1 (p≥0.05). Conclusion: The 12-week resistance training program seems to modulate and decrease serum mir-92a expression in elderly women with OSO. The lack of a significant change in BMI, body fat percentage, and ET-1 levels following the exercise training might be due to the type and intensity of the exercises.

Objective: Due to the uncertainty of the impact of thyme plant on endurance sport performance, despite its high antioxidant property, this study aims to assess the effect of aerobic exercise combined with thyme extract supplementation on the PGC-1α gene expression in male rats. Methods: This experimental study was conducted on 40 adult male Wistar rats randomly classified into five groups of 8 including control, sham, aerobic exercise, thyme extract (400 mg per kg per day), and aerobic exercise + thyme extract. The rats began their exercise on a treadmill for 10 minutes a day with a speed of 10 m/min and a slope of 10%. Speed and duration of exercise gradually increased over the next two weeks until they reached 27 m/min and one hour per day, respectively. The real-time Polymerase Chain Reaction (PCR) method was used to investigate the PGC-1α gene expression extracted from the soleus muscle of rats. Results: There was a significant difference in PGC-1α gene expression after intervention between the combined group and control and sham groups (Mean=1.01±0.0306; F=4.238, P=0.05). However, the PGC-1α gene expression rate in aerobic exercise group and supplementation group was no significantly different compared to control and sham groups (Mean=1.01±0.0306; F=0.00001, P=0.005). Conclusion: It seems that thyme extract supplementation along with aerobic exercise increases the PGC-1α gene expression.

Purpose: long-term high intensity endurance training might be associated with an increased risk of cardiac fibrosis. The aim of this study was to determine whether long-term and high intensity endurance training can lead to pathological structural changes in the heart of the rats.Methods: Sixteen male Wistar rats (four weeks old) were randomly divided into two groups: control and training. Extreme endurance training was performed on a treadmill (five sessions per week for 16 weeks). After euthanasia, ventricular collagen deposition was quantified by Masson trichrome–stained images method, and PKP2 and TGF-β1 gene and proteins expression were measured by Real Time-PCR and Western blotting, respectively. Data were analyzed by independent t-test at P ≤ 0.05.Results: the rate of collagen deposition and TGF-β1 gene and protein expression in the right ventricles of the rats were significantly increased. On the other hand, PKP2 gene expression in the training group was significantly decreased, but PKP2 protein expression was decreased in the exercise group and was not statistically significant.Conclusion: Based on the results of this animal study, cardiac fibrosis and increased expression of desmosomal proteins were observed after intense and prolonged exercise. This suggests that vigorous and prolonged endurance exercise may possibly cause pathological changes in the right ventricle by disrupting the structure of the desmosomes and forming fibrous tissue.Keywords: Desmosomal proteins, Fibrosis markers, Endurance training

Background and Aims: The expression of adhesion molecule such as the intercellular adhesion molecule- I (ICAM-I) and vascular adhesion molecule- I (VCAM-I) in obese people may increase atherosclerosis phenomena and cardiovascular diseases. The purpose of the current study was to investigate the effects of 10-week high-intensity and moderate-intensity aerobic trainings on plasma levels of ICAM-I and VCAM-I in obese boys. Methods: The statistical population of the present study was obese boys of Baharestan city. The sample included 36 obese student boys aged 13.4±0.69 years old, height: 163.06±7.88 cm; weight; 85.72±8.42 kg) who were randomly assigned to three groups: high-intensity training (n=‌12), moderate-intensity training (n=‌12), and control (n=12). The training was performed at 50%-65% maximal heart rate (moderate-intensity training group) and 70%-85% maximal heart rate (high-intensity training group), three days per week for 10 weeks. The blood sample was collected 48 h before and 48 h after the last session of exercise training. The ICAM-I and VCAM-I were measured via ELISA. Data were analyzed using two-way repeated ANOVA in SPPS, version 18.Results: The results indicated that body weight and BMI decreased significantly after moderate- and high-intensity training (p < 0.05). VO2max value significantly increased following 10 weeks of exercise training in both groups. No significant different was observed between exercise groups for plasma levels of ICAM-I. Moreover, the plasma levels of VCAM-I decreased significantly in the high-intensity training compared to the moderate-intensity training and control groups (p < 0.05). Conclusion: It seems that intensity of training may decrease plasma levels of ICAM-I and VCAM-I through decrease of body weight and BMI and increase of VO2max.

Background: The aim of this study was to determine the effect of purslane supplementation on oxidative stress, inflammation and muscle damage biomarkers after completing high-intensity interval exercise (HIIE). Methods: Nine healthy female runners completed a HIIE treadmill protocol comprising 1 min at 100% of the peak treadmill velocity attained in a preliminary exhaustive incremental treadmill test, interspersed by 1 min of passive recovery, until they had run for 2.5 km. Athletes repeated this test following 10 days of supplementation with 1000 mg per day of purslane. Results: At baseline (i.e., absence of purslane supplementation) the HIIE protocol increased 9-HODE (p < 0.001), 13-HODE (p < 0.001), IL-17 (p < 0.001), TNF-α (p < 0.001), and LDH (p < 0.001). After purslane consumption, resting 9-HODE, 13-HODE, IL-17, TNF-α, and LDH were lowered (p < 0.001) and IL-10 was increased (p < 0.001) compared to before purslane supplementation. After purslane consumption and following the HIIE protocol, the responses of these biomarkers were lower compared to baseline (p < 0.001). Conclusion: Ten days of purslane supplementation blunted the perturbation to biomarkers of oxidative stress, inflammation and muscle damage in female runners completing a HIIE protocol.

Purpose: Adipolin is a novel anti-inflammatory adipokine with insulin-sensitizing effects that ameliorates glucose intolerance and insulin resistance. Since physical activity and exercise are one of the effective and safe therapeutic interventions in improving insulin resistance; this study investigated effect of endurance training of two different intensities on adipolin and furin levels and insulin resistance index (HOMA-IR) in sedentary men. Methods: 38 sedentary overweight men (BMI>25 Kg/m2, aged 35-50 years) randomly assigned in to two training groups of low (n=13) and high intensity (n=12) and control (n=13) groups. Subjects in moderate intensity training group run on treadmill at intensity of 50-70 % of maximal heart rate and high intensity training group run at 70-90% of maximal heart rate, 30-45 minutes per session and three sessions per week for 10 weeks. Serum levels of adipolin, furin, insulin and fasting glucose and anthropometric indices measured before and 48 hours after last training session. Statistical analysis was done by one-way ANOVA and p < 0.05 considered significant. Results: changes in adipolin levels were significantly different between group of moderate intensity training and high intensity training one. Moreover, there were significant differences between two groups of moderate and high intensity training and control group for changes in weight, body mass index, body fat percentage and waist to hip ratio. Conclusion: it seems that intensity of endurance training in addition to effecting anthropometric indices, is the important factor to determine changes of adipolin levels in sedentary overweight men; however, it seems more studies are necessary to identify the mediated mechanism.

Objective: Vascular aging and osteosarcopenic obesity (OSO) refers to age-related endothelial cells and body composition disorders. The purpose of this study was to investigate the effect of 12 weeks resistance training with elastic bands on vascular aging-related serum miR-146 expression level and atherosclerosis risk factors in elderly women with OSO. Materials and Methods: In the present single-blind randomized clinical trial, 48 elderly women with OSO (according to Dual energy X-ray absorptiometry (DEXA) method results), were divided randomly into two groups of control (n=22) and experimental (n=26). The experimental group performed resistance training with elastic bands for 12 weeks and three sessions per week for all major muscle groups. Statistical analyses were carried out using SPSS statistical software version 22. Results: Between-group comparisons showed a significant decrease in serum miR-146 expression (P-value= 0.005) and total cholesterol (P-value= 0.034) in the experimental group compared to the control group. There was no significant difference in body weight (P-value= 0.440), BMI (P-value= 0.553), total fat (P-value= 0.093), BMC (P-value= 0.862), BMD (P-value= 0.564), Hs-CRP (P-value= 0.065), HDL-C (P-value= 0.515) and LDL-C (P-value= 0.889). Conclusion: Resistance trainings decrease miR-146 expression in elderly women with OSO, which was associated with decreased LDL levels and increased HDL levels, though BMI, body fat percentage, total cholesterol and HS-CRP were not significantly different probably due to the type and intensity of the exercises that require further investigation.

Background and Objectives: Changes in the expression of pre-inflammatory cytokines play a peculiar role in the acceleration of brain aging. The consumption of some traditional herbs with antioxidant and anti-inflammatory properties(such as rosemary) and regular aerobic exercise are effective factors in the control of premature aging. The present study aimed to assess the effects of aerobic exercise and consumption of rosemary extract on Tumor necrosis factor-alpha (TNF-α) and Interleukin 6 (IL-6) levels in the cerebellum tissue of old male rats. Methods: A total of 40 male Wistar rats (18 months) weighing about 350–450 g were selected. They were randomly assigned to five groups: control, sham (treadmill off and distilled water gavage), practice (12 weeks, 5 days a week), supplement or extract (12 weeks of daily 100 mg/ kg gavage rosemary extract), and practice-supplement(extract). To evaluate the level of cytokines, the cerebellum tissue was quickly removed 24 h after the last practice session according to ethical principles. After homogenization, the levels of TNF-α and IL-6 were evaluated using enzyme-linked immunosorbent assay (ELISA technique). Background: Exercise training and L-arginine are believed to have a major role in managing the hypertension and also their combination may cause further improvement in blood pressure. Therefore, the present study aimed to determine the effect of combined training with or without L-arginine ingestion on the circulation of adropin, VEGFR-2, and nitric oxide (NO) in postmenopausal women with hypertension. Methods: The present study was a randomized double-blind placebo-L-arginine-controlled trial registered in Iranian Registry of Clinical Trials with the code of IRCT20210627051723N1. This work was conducted in the summer and autumn of 2021. We recruited 48 post-menopausal women with hypertension and assigned them randomly (choosing random numbers by participants) in four equal groups, namely training+Larginine (TL), training (T), L-arginine (L), placebo (P) groups. The study intervention (combined training, L-arginine, or both) continued for eight weeks. Purpose: Bax and BCL-2 genes are important genes that involve in the intrinsic pathway of apoptosis. Although the effects of interval and intensive exercises on the mechanisms involved in the gaining processes are less considered. So, the aim of the present study was to investigate the effect of high-intensity interval training on the expression of pro and anti-apoptotic genes (Bax and BCL-2) in myocard in aged male mice.Methods: This experimental study was performed on twenty 24- months old C57BL/6 mice. Animals were randomly assigned to the control (n=10) and experimental groups (n=10). The experimental group performed high interval intensity training for four weeks (five sessions per week). At first, the interval training started at the intensity equal to 85 percent of maximum speed and in the continuation of interval training, 5 percent was added weekly and the last two weeks ended with 95 percent rate. Forty-eight hours after the last training session, myocardial muscle was removed. Few studies have been conducted on the effectiveness of psychological skills training on hypothalamic pituitary adrenal (HPA) axis response and sport performance. The aim of this study was to examine the impact of 8-week psychological skills training on competitive anxiety and salivary cortisol in elite sprinters. This study was a quasi-experimental study by pretest-posttest design with a control group. Statistical group comprised all men sprinters with an age range of 18-29 years in Khuzestan Province. To do this test, 30 male elite sprinters were randomly allocated into experimental and control groups(15 in each). We used salivary cortisol kit (Germany IBL company model RE-52611 model) to and Competitive Anxiety Inventory-2 (CSAI-2). The results were analyzed with SPSS 20. The findings showed significant reduction in salivary cortisol and significant reduction in Competitive Anxiety in experimental group to comparison with the control group after 8 weeks of psychological skills training. The results suggest that psychological skills training can be uses as a promised strategy in reduction of competitive anxiety and salivary cortisol in athletes.

Introduction: Appropriate movement patterns are important for both injury prevention and athletic performance. Therefore, the use of Functional Movement Screen (FMS), which has been shown to be effective in predicting sport injuries, is recommended by many sports specialists in designing training programs. On the other hand, questions remain regarding its utility as a means to evaluate the effectiveness of training. The purpose of the present study was to investigate whether a functional training program can affect the fundamental movement patterns. Materials and Methods: The study participants consisted of 27 youth male soccer players in the age range of 14 to 16 years and free from pain and previous injury in different layers. The subjects were allocated to experimental and control groups. The experimental group performed functional training for 10 weeks. The total time devoted to training was the same in both groups. Before and after the functional training, FMS was evaluated in both groups. Independent t-test and ANCOVA were used for statistical analysis at the significance level of P < 0.05. Results: A significant difference was observed in FMS score between experimental group (18.07 ± 1.49) and control group (16.07 ± 1.11) (P < 0.001; Effect size = 0.71). Conclusion: This study demonstrated that fundamental movement patterns improved as a result of a functional training intervention in youth male soccer players. Therefore, coaches and sports specialists can be recommended to pay special attention to functional training. The investigation of the effect of functional training on other injury risk factors in future studies seems necessary.

Background & aims: Obesity as a pandemic disease is the high accumulation of adipose tissue which secrets different hormones such as apelin. Apelin as an adipocytokine increases in obesity. Aerobic training induced apelin responses are not clarify well. So, we aimed to determine the effect of eight weeks aerobic training on Apelin-13 and insulin resistance in overweight men. Methods: Current study was quasi-experiment design. Twenty-six overweight men with BMI between 27-30 kg/m2 were randomly enrolled in the present study following public call announcement and match to inclusion criteria. Training group performed 15 (bouts) × 4 (min) exercise training for 60 min with 85 to 90% of VO2max used for four sessions/week (for 3 weeks). Then, blood (for ELISA analyzing) and tissue sampling was performed from rat’s hippocampus and they were evaluated by using a Real-Time PCR method. Also, independent and paired t tests were used to define within and between group differences. Results: Findings showed that gene translations of Lipocalin-2 and Interleukin1-β were significantly decreased in rat’s hippocampus (p=0.0001 and p=0.0001, respectively). Also, similar decreases were shown in serum levels of Lipocalin-2 and Interleukin1-β in rats (p=0.017 and p=0.003, respectively). Conclusion: It seems that significant decrease of Lipocalin-2 subsequent of short-time intensive interval training is due to decreasing and significant changes in Interleukin1-β. Also, current training protocol on treadmill can significantly decrease levels of Lipocalin-2 and Interleukin1-β in rat serum and hippocampus.

Background: Although less is known about the molecular mechanisms responsible for its genetic compatibility, regular training is identified as a non-pharmacological treatment for obesity and type-II diabetes. This study aimed to determine the effect of a 3 months aerobic training on pancreatic TCF7L2 expression and glycemic profile in type II diabetic rats. Materials and Methods: In this experimental study, type II diabetes was induced in male Wistar rats (n=16, weight: 220±30 g) by intraperitoneal injection of streptozotocin- nicotinamide. Animals were randomly divided into Exercise (n=8) and Control (n=8) groups. Exercise group, but not Control group, was completed a 3 month aerobic training (3 sessions/week). Forty-eight hours after the last exercise session, the relative expression of pancreatic TCF7L2, fasting glucose and serum insulin were measured in two groups. Results: Compared to Control rats, exercise resulted in a significant decrease in fasting glucose in Exercise group (P=0.001). Serum insulin was increased significantly by aerobic training in Exercise group compared to Control one (P=0.014). However, pancreatic TCF7L2 expression did not change by aerobic training (P=0.876). Conclusion: Based on these data, while we concluded that a long-term aerobic training effectively improves the glycemic profile and insulin concentration of type II diabetic rats, such improvements cannot be attributed to TCF7L2 expression in pancreatic tissue.

Background and aims: Lipocalin-2 (LCN-2) with Interleukin1-β (IL1-β) are inflammatory proteins that induced the inflammatory reactions due to insufficient of physical activity. The purpose of the current study was to determine the effect of high intensity interval training on Lipocalin-2 and Interleukin1-β gene expressions in fast- and slow muscles of rats. Methods: In the current study 20 Wistar rats were divided into HIIT and control groups. HIIT protocol was performed 60 min in each session for four sessions in each week. After warm-up, HIIT group was carried out 15 × 4 min bouts of HIIT with 85 to 90% of VO2max that continued with three min recovery (between each bout of HIIT) with 70% of VO2max. Abstract Background: Protein tyrosine phosphatase 1B (PTP1B) is a key enzyme in dephosphorylation of the insulin receptor (IR) and it is a central factor to induce the insulin resistance. The purpose of this study was to evaluate the effect of 12-week aerobic training on protein tyrosine phosphatase 1B gene expression and insulin resistance in diabetic rats. Materials and Methods: In this study, 16 Wistar rats were divided into aerobic training and control groups. After inducing diabetes intra protaneally, aerobic training group performed training protocol for 12 weeks and 5 session/week. The duration and speed of each session increased progressively as 18 to 26 m/min and 10 to 55 min, respectively. Then, blood and tissue (from gastrocnemius) sampling were carried out in diabetic rats. Insulin resistance markers and PTP1B gene expression were evaluated by commercial kits and Real-Time PCR method, respectively. Results: Findings showed that PTP1B significantly was decreased in diabetic rats of aerobic training group (p=0.0001). Also, glucose and insulin resistance significantly was decreased in aerobic training groups (p=0.02 and p=0.006, respectively). However, insulin in control rats was significantly increased (p=0.015). Conclusion: It seems that, current aerobic training protocol has capability to decrease PTP1B and insulin resistance in diabetic rats. Furthermore, the direct correlation between PTP1B and insulin illustrated that any changes in insulin resistance due to exercise training associated with diminution of negative regulation of insulin signaling pathway.

Abstract Background: Irisin is a new myokine that exerts its positive effects via increasing energy expenditure. The purpose of this study was to determine the effect of submaximal aerobic training on serum irisin level and irisin-related insulin-resistance change in obese men. Materials and Methods: In this study, 26 obese men were selected as statistical sample and they were divided into experimental groups. Former group (13 men) consisted of subjects who carried out exercise training with 50-60 percent of maximum max heart rate for 10 continuous weeks (3 sessions per week for 30 to 45 minutes in each session). Later group was consisted of subjects who do not carried out exercise training. After the interventions of study, irisin serum levels, insulin resistance indices, body composition and maximum oxygen consumption were determined. Finally, data were analyzed by Stata software. Results: Statistical data analysis illustrated that the amounts of irisin serum were significantly increased in exercise training group than control group (14.75±6.22; p=0.022) after the intervention. Also, only body fat percentage (1.242±0.57) and max oxygen consumption (1.509±0.328) of subjects were significantly increased (p=0.034 and p=0.0001, respectively). However, there were not statistically significant changes in irisin- related insulin-resistance indices (p>0.05). Conclusion: Current study presented that serum irisin was increased after 30 submaximal exercise training sessions regardless of insulin resistance changes in obese men.

Background: Pathological hypertrophy is one of the negative consequences of cardiac sympathetic hyperactivity. Recent studies have shown that YAP1 plays a critical role in cardiomyocytes hypertrophy. Considering the preventive role of exercise training in cardiovascular diseases, the present study was conducted to examine the effect of aerobic exercise training on YAP1 gene expression and its upstream components. Methods: Eighteen male Wistar rats were randomly divided into aerobic training and control groups. Aerobic training was performed one hour/day, five days per week, for eight weeks, on a treadmill at 65-75% VO2max. Pathological hypertrophy was induced by injecting 3 mg/kg-1 of isoproterenol for seven days. Monobenzyltin Schiff base complex, [N-(3,5-dichloro-2-oxidobenzylidene)-4-chlorobenzyhydrazidato](o-methylbenzyl)aquatin(IV) chloride, C1, is an organotin non-platinum metal-based agent. The present study was conducted to investigate its effects on MCF-7 cells with respect to the induction of apoptosis and its inhibitory effect against MCF-7 breast cancer stem cells. As determined in a previous study, compound C1 revealed strong antiproliferative activity on MCF-7 cells with an IC50 value of 2.5 μg/mL. Annexin V/propidium iodide staining coupled with flow cytometry indicated the induction of apoptosis in treated cells. Compound C1 induced apoptosis in MCF-7 cells and was mediated through the intrinsic pathway with a reduction in mitochondrial membrane potential and mitochondrial cytochrome c release to cytosol. Complex C1 activated caspase 9 as a result of cytochrome c release. Subsequently, western blot and real time PCR revealed a significant increase in Bax and Bad expression and a significant decrease in the expression levels of Bcl2 and HSP70. Furthermore, a flow cytometric analysis showed that treatment with compound C1 caused a significant arrest of MCF-7 cells in G0/G1 phase. The inhibitory analysis of compound C1 against derived MCF-7 stem cells showed a significant reduction in the aldehyde dehydrogenase-positive cell population and a significant reduction in the population of MCF-7 cancer stem cells in primary, secondary, and tertiary mammospheres. Moreover, treatment with C1 down-regulated the Wnt/β-catenin self-renewal pathway. These findings indicate that complex C1 is a suppressive agent of MCF-7 cells that functions through the induction of apoptosis, cell cycle arrest, and the targeting of MCF-7-derived cancer stem cells. This work may lead to a better treatment strategy for the reduction of breast cancer recurrence.

The changes in knee laxity and relaxin receptor expression at different phases of rodent estrous cycle are not known. Here, changes in the parameter were investigated in rats at different phases of the estrous cycle. Estrous cycle phases of intact female rats were determined by cytological examination of the vaginal smear. Following phase identification, blood was collected for serum hormone analyses. Knee passive range of motion (ROM) was determined by using a digital miniature goniometer. The animals were then sacrificed and patellar tendon, collateral ligaments and hamstring muscles were harvested for relaxin/insulin-like family peptide receptor 1 and 2 (RXFP1/RXFP2) analyses. Knee passive ROM was the highest at proestrus followed by diestrus and the lowest at estrus. Estrogen level was the highest at proestrus while progesterone and relaxin levels were the highest at diestrus. A strong correlation was observed between relaxin and progesterone levels. At proestrus, expression of RXFP1 and RXFP2 proteins and mRNAs were the highest at proestrus followed by diestrus and estrus. The finding shows that higher level of progesterone and relaxin in diestrus might be responsible for higher laxity of knee joint in rats.
