Introduction: To provide quality care at the end of life of chronically sick patient’s nurses must have good knowledge and practice about Palliative care because they are the key member of health team who typically has the greatest contact with patient’s problem. When a patient enters the end stage of life threatening disease like Cancer, treatment of pain and other symptoms must be considered to preserve quality of life. Nurse’s expertise is essential in providing physical and emotional care to the patient and family and in organizing the patient’s environment to minimize burden and control of suffering. Nurses provide better care in all health care setting to all groups of peoples according to situation but there is scarce data on the relative field. Aim of the study: The aim of the study was to observe association of social characteristics with knowledge and practice of pain management among nurses. Methods: This descriptive cross-sectional study was conducted at the palliative care units of Dhaka Medical College Hospital (DMCH), National Institute of Cancer Research Hospital (NICRH), Bangabandhu Sheikh Mujib Medical University (BSMMU), and Ahsania Mission Cancer & General Hospital, Dhaka. The study duration was one year, from 1st January to 31st December, 2020. The study was conducted with a total of 112 nurses working at the palliative care Units of the study hospitals. Result: Among the participants, the mean age was 29.79, (±SD 6.075), and majority were from the age group of 21-30 years. 88.4% were female 74.1% were graduates, 47% received training on pain management, 68.8% worked in government hospitals, 74.1% had less than 5 years of working experience. No significant association was observed between knowledge and social characteristics. The practice level of nurses was significantly associated with Age (p=0.03) and Training (p=0.01). A combination of nutritionally inferior diets and improper feeding practices are major contributing factors to the development of childhood malnutrition. Complementary feeding improvement should be of highest priority for nutrition of infant and young children containing all essential nutrients at required amount. The objective of this study was designed to investigate the inclusion of weaning food formulation and evaluating compositional and functional properties of the products. The analysis undertaken in this study was infant food formulation, proximate composition, mineral concentration and sensory properties. The moisture content in this study ranged from 1.49 ± 0.12 to 3.89 ±0.12, crude protein from 15.09 ± 0.05 to 16.79± 0.01, crude fat from 11.11 ± 0.07 to 12.80 ± 0.00, total ash from 2.06 ± 0.03 to 2.21 ± 0.00, carbohydrate from 65.9740 ± 0.24 to 68.2641 ± 0.14 and energy value ranged from 431.68 ± 0.05 to 449.18 ± 0.57. From the result provided, sample-A contained the highest protein, moisture, fat content than sample-B. But carbohydrate and energy are highest in sample-B than sample-A. The two macro minerals mg/100 g of Ca and Zn in the study were ranged from lower 0.60±0.07 to 0.79±0.01 and 1.05±0.01 to 2.44±0.06 respectively. Beta-carotene is varied from 0.90±o.01 to 1.17±0.00. Sensory parameters such as colour, appearance, texture, taste, aroma and overall acceptability is high in sample-A than to sample-B. Finally, from the general trend observed in this study that both the weaning foods prepared from cereal, legume, pulse flour provided better nutritional and functional compositions to meet nutrient dense of infant foods. Background In situations of increased need, such as mass casualty incidents (MCIs) and COVID-19, donated blood products are in shortage across the United States. Medical students are a potential pool for blood donors. The aim of this study was to determine overall attitudes of medical students at a single academic institution toward blood donation during times of increased need. Methods Three anonymous REDCap surveys were administered to all medical students at a rural academic institution. Surveys 1 and 2 were administered preceding and after an institution-wide MCI drill, in September and November 2019, respectively. Survey 3 was administered following a student-organized COVID-19 blood drive in June 2020. Multivariable analysis was performed to determine if factors, ie, experience with MCI drills and emergency medical services (EMS) training, were associated with willingness to donate blood. Furthermore, barriers to donation among those not willing to donate were assessed. Results Overall response rate for MCI surveys (surveys 1 and 2) was 38% (mean age 25.2 years and 50% women). 91% (n = 210) of respondents were willing to donate blood. Previous participation in MCI drills and EMS training was not associated with higher willingness to donate blood. Response rate for survey 3 was 15.6% (59.4% women), and 30 (31.3%) respondents indicated they did not volunteer to donate blood during the COVID-19 drive. Most common reasons for not donating were “other,” medical concerns, and being out-of-town. Conclusions Majority of medical students are willing to donate blood during times of increased need and offer a possible solution to increase blood donor pool.

Malaysia is a developing country and agro-industries contribute country’s economy. However, it is largely unknown the scenarios of agricultural practice and food supply over the years. To understand the fact, this study was designed to explore the agricultural land use changes and domestic food supply for last 50 years in Malaysia. The result showed that oil palm plantation area and yield, both are increased by folds, conforming “oil palm” mono-culture practice. Rice cultivation area remained almost unchanged but the yield increased. However, both area of plantation and yield in case of rubber, coca and coconut were decreased. When, it comes to domestic food supply, in 1973 food grain (Rice, Wheat, Maize) import was lower than domestic production. However, the import increased more than 3 times than the domestic production in 2013. Domestic production of vegetables and vegetables import was almost 50:50. Fruits import increased almost 3 times of domestic production, though domestic production was increased. In conclusion, it is understood that Malaysian agriculture economy is standing on “oil palm” wearers domestic food supply is import dependent. It is high time to reshuffle the policy for sustainable development and achieving food sovereignty.

INTRODUCTION The opioid epidemic is a growing problem in the United States. There is a high rate of opioid oversupply for treatment of symptomatic nephrolithiasis, partly due to patients being seen by multiple providers. In Pennsylvania, there are efforts to integrate a prescription drug monitoring program (PDMP) within the electronic medical record (EMR). The objectives of this study were to evaluate prescribing practices for opioids for symptomatic nephrolithiasis and the incidence of prescriptions not documented within the EMR.Adults who presented for treatment of symptomatic nephrolithiasis were sequentially evaluated from May - October 2017 at Penn State Milton S. Hershey Medical Center. With IRB approval, we evaluated opioids prescribed in the EMR, which was compared to the PDMP for each stone episode. We calculated daily morphine milligram equivalents (MME) and total MME available to patients.A total of 301 patients were identified (52% male) with a mean age of 50.0 +/- 16.7 years and 249 (83%) of patients were prescribed narcotics with an average of 226.8 +/- 232.2 MME for their stone episode. Of patients that were prescribed narcotics, 19% had additional narcotics prescribed to them that were not entered into the EMR and later identified using PDMP. The average additional opioid prescribed was 371.8 +/- 404.2 total MME.The majority of patients presenting with symptomatic nephrolithiasis were prescribed an opioid. Approximately one-fifth of patients were receiving opioids from other providers that were not documented in the EMR at the time of their opioid prescription. PDMP, or similar resources, should be utilized by providers to minimize opioid use and reduce oversupplying patients.

Background Mesenteric venous thrombosis (MVT) is typically associated with poor prognosis. Although prophylactic antibiotics are sometimes given with the intent of limiting bacterial luminal load and translocation in patients with MVT, this approach has not been universally adopted. The aim of this study is to analyze whether utilizing antibiotics empirically in those with MVT improves patient outcomes and survival when compared to those who do not receive empiric antibiotics. Methods A retrospective review of patients admitted with MVT between 2002 and 2019 at a single academic institution was performed. Demographics and rates of mortality need for bowel resection, readmission, and Clostridium difficile ( C. diff) infection were compared between patients treated with empiric antibiotics and patients not treated with antibiotics. Results Eighty-three patients (mean age 64.5 years and 55.4% male) who were admitted for MVT were included. Empiric antibiotics were utilized in 53% (n = 44) of MVT patients while 47% (n = 39) received supportive treatment without empiric antibiotics. Death occurred in 4 patients treated with antibiotics and 6 patients treated without antibiotics (9.1% vs. 15.3%, P = .50). Readmissions occurred in 12 patients (27.3%) treated with antibiotics and 10 patients (25.6%) not treated with antibiotics (27.3% vs. 25.6%, P = .87). C. diff infection occurred in 6 patients treated with antibiotics and in no patients not treated with antibiotics (13.6% vs. 0%, P = .03). Conclusions Empiric antibiotic usage may not improve rates of mortality or hospital readmission in patients with MVT and may unnecessarily expose patients to an increased risk of C. diff infection.

Infections are most frequent at the extremes of life, especially among newborns, reflecting age-specific differences in immunity. Monocytes maintain tissue-homeostasis and defence-readiness by escaping circulation in the absence of inflammation to become tissue-resident antigen presenting cells in vivo. Despite equivalent circulating levels, neonates demonstrate lower presence of monocytes inside peripheral tissues as compared to adults. To study the ability of monocytes to undergo autonomous transendothelial extravasation under biologically accurate circumstances we engineered a three-dimensional human vascular-interstitial model including collagen, fibronectin, primary endothelial cells and autologous untreated plasma. This microphysiological tissue construct enabled age-specific autonomous extravasation of monocytes through a confluent human endothelium in the absence of exogenous chemokines and activation. Both CD16− and CD16+ newborn monocytes demonstrated lower adherence and extravasation as compared to adults. In contrast, pre-activated tissue constructs were colonized by newborn monocytes at the same frequency than adult monocytes, suggesting that neonatal monocytes are capable of colonizing inflamed tissues. The presence of autologous plasma neither improved newborn homeostatic extravasation nor shaped age-specific differences in endothelial cytokines that could account for this impairment. Newborn monocytes demonstrated significantly lower surface expression of CD31 and CD11b, and mechanistic experiments using blocking antibodies confirmed a functional role for CD31 and CD54 in neonatal homeostatic extravasation. Our data suggests that newborn monocytes are intrinsically impaired in extravasation through quiescent endothelia, a phenomenon that could contribute to the divergent immune responsiveness to vaccines and susceptibility to infection observed during early life.

A novel qNMR method is described for the quantitative determination of total aluminum and phosphate in aluminum phosphate (AlPO4) adjuvanted vaccine samples using solution 27Al and 31P nuclear magnetic resonance (NMR) spectroscopy. External standard calibrations of AlPO4 solutions established excellent linearity in the range of 15-40 × 10-3 M and additional studies determined the level of detection for both nuclei. A commercialized combination vaccine product (Quadracel®), along with several individual adsorbed antigen components used in the vaccine were employed as model systems for method development. The developed method is also capable of quantitating the free phosphate (i.e. the fraction not bound to AlPO4 particles) in adjuvanted vaccines. This study is the first demonstration of a solution NMR method that is suitable for measuring total aluminum, and free and total phosphate concentrations in vaccine formulations consisting of antigen(s) adsorbed to aluminum adjuvant, in a single analytical workflow.

Cruel oil or green gold is a dilemma for the oil palm industry. The oil palm industry (planting and milling) has a devastating impact on the environment and ecosystems. Oil palm plantations and expansion cause deforestation, habitat loss, forest fragmentation, biodiversity loss, food chain disruption, soil property changes, water and air pollution, conversion of wetlands and arable lands, and increased greenhouse gas (GHG) emissions, resulting in annual fires as well as increasing both subsidence and flood risk. Palm oil mill effluents (POME) are toxic compounds that cause eutrophication and acidification, pollute terrestrial and aquatic systems and release greenhouse gases. However, the oil palm industry is a million-dollar industry that ensures food security (oil and fat). There is increasing demand for palm oil due to population growth and for use as a biofuel feedstock. Significant higher production per hectare in comparison to other oil crops is the main advantage of oil palm. The anthropogenic pressure on the environment is increasing to fulfil the demand and increasing susceptibility to natural disasters. Therefore, the sustainability of this industry is an urgent need. This critical review identified gaps and researched ways for the oil palm industry to be sustainable. Maintaining ecological integrity (ecological health, connectivity, resilience); justifying land allocation (ecosystem service mapping); providing awareness, good management practices, no/minimum production gaps, high yield and disease resistant cultivar generation and plantations, supplemental forms of alternative sources, zero-waste milling technology; and locating plantations on suitable land without further deforestation can fulfil the oil palm industry's present and future demands without impairing the ecosystem or environment.

INTRODUCTION: Disease severity of SARS-CoV-2 is correlated with patient age and comorbidities One population initially thought to be at increased risk of infection are patients with Inflammatory Bowel Disease (IBD) Understanding how this population is affected by SARS-CoV-2 is necessary for effective detection and management We present the case of COVID-19 in a young Crohn's disease patient on Infliximab CASE DESCRIPTION/METHODS: A 24-year-old female with Crohn's disease on Infliximab presented febrile with anosmia on 3/11 Five days later she developed myalgias, chest pain, coughing, and sneezing She denied fever, dyspnea and GI upset Presumed to be seasonal allergies, she was instructed to monitor symptoms On 3/27, she developed a fever of 99 5 deg F COVID-19 testing was performed and returned positive on 3/30 She completed 14-day quarantine with symptom resolution on 4/7, unremarkable lab work and chest X-ray with no signs of pneumonia However, on 4/20, she had recurrence of symptoms, including worsening dyspnea and chest pain Telehealth visit the next day revealed resting O2 sat of 98%, 6 minute walking desaturation test with O2 sat of 96% and heart rate of 119 bpm The patient also reported coughing and increasing dyspnea Two minutes later, heart rate normalized to 82 bpm and O2 sat was 97% The patient was followed by a dedicated COVID-19 clinic until complete symptom resolution DISCUSSION: In this case, our patient presented febrile with dyspnea five days after her Infliximab infusion Current literature suggests IBD patients on immunotherapy are not at higher risk of contracting COVID-19 [1] Also, many reports link symptoms of nausea, vomiting, and diarrhea with this infection In a recent case report, an elderly woman with Ulcerative Colitis had initially presented with signs of IBD flare, instead of typical URI symptoms, which unfortunately resulted in misdiagnosis and her fatality [2] Our patient did not exhibit GI distress Lastly, she had symptom resurgence after 12 days of being asymptomatic, which has yet to be described in an IBD patient Considering the complexity of this coronavirus, more information is needed to understand varying manifestations and its implications in IBD patients

This study describes the NMR-based method to determine the limit of quantitation (LOQ) and limit of detection (LOD) of cholesterol, a process-related impurity in the replication-deficient Herpes Simplex Virus (HSV) type 2 candidate vaccine HSV529. Three signature peaks from the 1D 1H NMR of a cholesterol reference spectrum were selected for the identification of cholesterol. The LOQ for a cholesterol working standard was found to be 1 μg/mL, and the LOD was found to be 0.1 μg/mL. The identity of cholesterol, separated from the formulation of growth supplement by thin layer chromatography (TLC), was confirmed by 1D 1H NMR and 2D 1H-13C HSQC NMR. The three signature peaks of cholesterol were detected only in a six-times concentrated sample of HSV529 candidate vaccine sample and not in the single dose HSV529 vaccine sample under similar experimental conditions. Taken together, the results demonstrated that NMR is a direct method that can successfully identify and quantify cholesterol in viral vaccine samples, such as HSV529, and as well as in the growth supplement used during the upstream stages of HSV529 manufacturing. Comfortable life, economic growth, industrialisation, global warming, energy security and sustainable environment are burning issues facing modern civilisation. The availability of adequate renewable energy is in demand. Jatropha is being explored as a potential biofuel crop candidate because of its biodiesel production potential, high oil content, rapid growth, easy propagation, drought tolerant nature, relatively less irrigation and agricultural inputs, insect and pest resistance. However, previous programmes for Jatropha plantation did not satisfy the expectation because of the absence of a good commercial variety, large scale propagation without evaluating the planting material, knowledge gap and consideration as low a impute crop. Lack of systematic breeding programmes, the inexistence of a collaboration between scientists in this field, the unavailability of desired germplasm and more importantly less variability within the species are the constraints for the conventional breeding for a Jatropha improvement programme. Biological techniques have proven records for the improvement of many crops. Jatropha “organogenesis”, which has insignificant contribution to genetic improvement, is studied. Several genomic and transgenic studies have been reported, but it is still far behind in comparison to other crops. It is time to investigate somaclonal variation, in vitro selection and haploid breeding for Jatropha improvement. Resequencing and transcriptom analysis are necessary for high dance linkage map and a good reference genome. Genome wide association studies (GWAS) and genomic selection (GS) are pending. Genetic engineering, particularly to increase female flowers in inflorescence, eliminates the toxic component and increases tolerance to diseases, insects and pests should be given priority.

The tandem repeats, conserved short segments of DNA, which are found in all prokaryotic and eukaryotic genomes, are called microsatellites. It is also known as variable number tandem repeats (VNTRs), simple sequence repeats (SSRs) and short tandem repeats (STRs). Microsatellites present in both coding and non-coding regions of a genome. The high polymorphism of microsatellites makes them powerful genetic markers for genome mapping of many organisms. It is also suitable for ancient and forensic DNA studies for population genetics and conservation of biological resources. The major disadvantage of microsatellites is that for the first time they need to be isolated de novo from most species being examined. The task of microsatellite isolation is quite cumbersome involving in terms of effort and time, because it traditionally involves screening of genomic libraries. Cross-species amplification, Mining microsatellites from nucleotide sequenced data and Genomic library- based method are general methods of microsatellite isolation. Cross-species method may not effective for all species, Data mining is not applicable if there is no or limited data of DNA sequence. Genomic library based method is the best choice. Traditional protocol, primer extension protocol, selective hybridization, and Fast Isolation by AFLP of Sequences containing repeats (FIASCO) are the protocols of microsatellite development based on genomic library. The probability of a pregnancy terminating in a full-term, healthy live birth is a powerful indicator of the health status of its women, and of the quality of health care available to them during pregnancy and birth. The present study conducted to find out the maternal outcome of prolonged pregnancy. This study carried out in the department of obstetrics, Sir Salimullah Medical College and Mitford Hospital, Dhaka, between the period of February 2003 and December 2003. Patients admitted in labour ward having the history of post dates but not in labour and some were admitted during first stage of labour. Patients who were sure about their Last Menstrual Period (LMP) and those patients who had regular menstrual cycle were included in the study. Total 139 respondents were included in the study. Among them 114 (82.01%) were in the age group of 18 to 29 years and 25 (17.99%) were in the age group of ?30 years. Among the respondents 67 (48.2%) were primi gravida and 72 (51.8%) were multi gravida. Among the respondents 92 (66.2%) were in the 1st stage of labour, 7 (5.0%) were in the 2nd stage of labour and rest 40 (28.8%) were not in labour. Mode of delivery of highest number of respondents was caesarian section (54.0%) followed by normal vaginal delivery (39.7%). Other mode of delivery were ventouse and forceps and they were 07(05.0%) and 2(1.4%) respectively. Out of 75 respondents under gone caesarian section, indication of C/S was fetal distress in 1st stage of labour, prolong 1st stage with maternal distress, failed induction, cephalopelvic disproportion (CPD) and breech presentation with big baby were 32.0%, 25.3%, 24.0%, 16.0% and 2.7% respectively. Maternal morbidity like PPH, UTI, puerperal sepsis and wound infection were 10.0%, 14.40%, 3.60% and 5.70% respectively. In postdated pregnancy maternal morbidity is common finding. Molecular markers usually do not have any biological effect. They are identifiable DNA sequences, found at specific locations of the genome, and transmitted from one generation to the next. Marker assisted selection (MAS) is a novel technique that can complement traditional breeding methods for rapid genetic gains. Genetic gain through selective breeding is the objective of a breeder to achieve long term improvement in animal and plant genomes; however the pace of improvement is inversely proportional to the Generation Interval. Genetic improvement in livestock, particularly those with long generation intervals, requires decades for tangible results. Successful MAS breeding programmes require gene mapping, marker genotyping, quantitative trait loci (QTL) detection, genetic evaluation and finally MAS. Genomic selection is a form of markerassisted selection. Using markers covering the whole genome could mean potentially that all the genetic variance is explained; and the markers are assumed to be in linkage disequilibrium with the QTL so that the number of effects per QTL to be estimated is small. MAS drastically reduces generation interval and increases selection accuracy. Therefore, a breeding strategy based upon markers making the best use of the two approaches can facilitate rapid genetic gain though selection of markers related to economic traits such as milk and meat production. The incorporation of intrinsically disordered domains enables proteins to engage a wide variety of targets, with phosphorylation often modulating target specificity and affinity. Although phosphorylation can clearly act as a chemical driver of complexation in structured proteins, e.g., by abrogating or permitting new charge-charge interactions, the basis for enhancement of the hydrophobically driven interactions that are typical of disordered protein-target complexation is less clear. To determine how phosphorylation can positively impact target recruitment in disordered domains, we have examined the interaction between the disordered N-terminal transactivation domain (TAD) of p53 and the pleckstrin homology (PH) domain of p62. Using time-resolved electrospray ionization with hydrogen-deuterium exchange, we demonstrate that phosphorylation has little effect on the conformation of the p53 TAD when it is bound to the PH domain but instead increases the degree of conformational disorder in the unbound state. We propose that this increase in the degree of disorder creates a wider free energy gap between the free and bound states, providing a target-independent mechanism for enhanced binding when the phosphorylated and unphosphorylated p53-target complexes have similar free energies.

Wnt signaling, named after the secreted proteins that bind to cell surface receptors to activate the pathway, plays critical roles both in embryonic development and the maintenance of homeostasis in many adult tissues. Two particularly important cellular programs orchestrated by Wnt signaling are proliferation and stem cell self-renewal. Constitutive activation of the Wnt pathway resulting from mutation or improper modulation of pathway components contributes to cancer development in various tissues. Colon cancers frequently bear inactivating mutations of the adenomatous polyposis coli (APC) gene, whose product is an important component of the destruction complex that regulates β-catenin levels. Stabilization and nuclear localization of β-catenin result in the expression of a panel of Wnt target genes. We previously showed that Mule/Huwe1/Arf-BP1 (Mule) controls murine intestinal stem and progenitor cell proliferation by modulating the Wnt pathway via c-Myc. Here we extend our investigation of Mule's influence on oncogenesis by showing that Mule interacts directly with β-catenin and targets it for degradation under conditions of hyperactive Wnt signaling. Our findings suggest that Mule uses various mechanisms to fine-tune the Wnt pathway and provides multiple safeguards against tumorigenesis.

In 2018, anonymous online provocative comments were submitted to student leaders of a Syrian Refugee Initiative (SRI) at the Penn State College of Medicine. This triggered a series of actions with students and medical school leaders aimed at identifying the person who submitted the comments, trying to understand mutual perspectives, and managing the impact of the event on the student body.We describe the history of our college's commitment to humanism and how the SRI was a direct outgrowth of that culture. Voices of the student leaders who were directly impacted by the provocative comments and educational leaders who worked to resolve the crisis are presented. We also describe a collaborative process that involved engaging cybersecurity experts to identify the perpetrator, and share how the students and educational leaders were able to develop trust despite initial skepticism by students over the leadership's avowed commitment to taking the hate speech seriously.While the perpetrator was never identified and opportunities for improvement were identified along the way, by including student leaders in the process, students and administrators were able to develop trust and reach reasonable closure on this disturbing event. Take home messages are presented to guide other institutions in navigating instances of provocative comments or speech.

A simple, rapid, precise and cost effective method has been developed and validated for determination of Vildagliptin in pharmaceutical tablet dosage form. The chromatographic separation was carried out with Shimpack VP-ODS, 150 × 4.6 mm, 5?m analytical column and mobile phase containing 0.02M phosphate buffer (pH 4.6) and acetonitrile at the ratio (80:20% v/v). pH of the buffer solution was adjusted with orthophosphoric acid. The instrumental settings include flow rate 0.7 ml/min, column temperature at 25ºC and detector wavelength of 210nm using a photodiode array detector. An efficient protocol was established for in vitro plant regeneration of Paederia foetada L . (Family. Rubiaceae), a widely used medicinal vine of Bangladesh through callus culture in using nodal segment. Yellowish green nodular callus was observed from nodal segments on MS basal medium supplemented with 1.5 mg/L BAP + 0.5 mg/L NAA within three weeks. Large number of shoots (14.4±1.29) were obtained when the callus was sub cultured on MS medium with 0.5 mg/L BAP. In vitro raised shoots rooted on half strength MS medium with 0.5 mg/L IBA. An efficient protocol was developed for in vitro plant regeneration of a popularly used anti-diabetic medicinal plant, Coccinea cordifolia (Linn.) Cogn.(Cucurbitaceae) through direct organogenesis using shoot tip and nodal explants. Best shoot induction was observed on MS basal medium supplemented with 0.5 mg/l BAP, in which 88.2% of nodal explants responded to produce maximum number (6.2 ± 0.58) of shoots per culture. (Acanthaceae) - a rare medicinal shrub of Bangladesh, through in vitro culture using shoot tip and nodal explants. A protocol was established for mass propagation of the valuable medicinal plant Ficus religiosa L. (Moraceae) through in vitro culture using apical and axillary buds of young sprouts from selected plants. Best shoot induction was observed on MS basal medium supplemented with 0.5 mg/l BAP + 0.1 mg/l IAA, in which 78 per cent of the explants produced 16 shoots per culture. Repeated subcultures in the same medium, resulted rapid shoot multiplication with 24 shoots per culture. The present study describes a protocol for high frequency plant regeneration of Physalis minima . Shoots were induced by culturing nodal segments and shoot tips from 15 day old seedlings. About 29 and 32 shoots were found to be induced from nodal segment and shoot tip explants, respectively, cultured on MS medium supplemented with 1.0 mg/l BAP. When shoots were subcultured on the fresh medium with same component as mentioned above, the shoots were elongated. Shoots rooted well when they were excised individually and implanted on half-strength MS medium with 0.3 mg/l NAA, where 98% shoots rooted within 12-15 days. In vitro grown plantlets with strong root system were successfully established in normal room temperature for seven days before transplanting in pots where they were reared for three weeks through successive acclimatization. The regenerated plants were successfully transferred to the soil with 90% survival rate. Ubiquitin-specific protease 7 (USP7) is a deubiquitinating enzyme found in all eukaryotes that catalyzes the removal of ubiquitin from specific target proteins. Here, we report that UbE2E1, an E2 ubiquitin conjugation enzyme with a unique N-terminal extension, is a novel USP7-interacting protein. USP7 forms a complex with UbE2E1 in vitro and in vivo through the ASTS USP7 binding motif within its N-terminal extension in an identical manner with other known USP7 binding proteins. We show that USP7 attenuates UbE2E1-mediated ubiquitination, an effect that requires the N-terminal ASTS sequence of UbE2E1 as well as the catalytic activity of USP7. Additionally, USP7 is critical in maintaining the steady state levels of UbE2E1 in cells. This study reveals a new cellular mechanism that couples the opposing activities of the ubiquitination machinery and a deubiquitinating enzyme to maintain and modulate the dynamic balance of the ubiquitin-proteasome system.

Background: Preeclampsia is a multisystem disorder of unknown etiology characterized by development of hyperten­sion to the extent of 140/90 mm of Hg or more with proteinuria after the 20th gestational week in a previously normoten­sive and non protein uric women. According to the National High blood presure Working group (NHBPEP) and Ameri­can college of obstetricans and Gynecologiests (ACOG) hypertension in pregnancy is defined as a diastolic blood pressure of 90 mm Hg or higher after 20 weeks of gestation in a woman with previously normal blood pressure (NHBPEP, 2000; ACOG, 2002). If the disease is allowed to progress to the HELLP syndrome or eclampsia, maternal morbidity and mortality increases. The majority of perinatal losses are related to placental insufficiency, which causes intrauterine growth retardation, prematurity associated with preterm delivery, or abruptio placentae. Objectives: This study tried to explore the effect of serum C reactive protein concentration in preeclamptic women and its effect on pregnancy outcome.Methods: This case control study included 60 third trimester pregnant women (30 normotensive and 30 preeclamptic) who attended Department of Obstetrics and Gynaecology, BIRDEM and DMCH, during July 2009 and June 2010. Estimation of serum C reactive protein (CRP) concentrations was done by liquid phase immunoprecipitation assay and turbulometry at DMC.Results: Mean (±SD) age showed no significant difference between groups; however, BMI, SBP, DBP and CRP were significantly (PConclusion: CRP concentration was high in preeclamptics resulting in adverse pregnancy outcome.

The Neurosporacrassa protein kinase C (NPKC) is reported to be a regulator of light responsive genes. It phosphorylates the light receptor WC-1 and regulates the levels of the circadian clock protein FRQ and transcription of the light-induced albino-2 gene. In mammals, the conventional and novel isoforms of PKC are activated by diacylglycerol (DAG), which induces PKC translocation from the cytoplasm to membranes. To investigate the interaction of NPKC and DAG in Neurospora, we constructed a strain that expresses a PKC-GFP fusion protein. We found that NPKC localizes to growing tips and sub-apical plasma membrane in actively growing hyphae, and actively participates in septum development. NPKC is activated by exogenous DAG and phorbol esters, and translocates to the plasma membrane from the cytoplasm. We have previously reported that choline depletion of the chol-1 mutant of Neurospora increases DAG levels and lengthens the period of the circadian rhythm of conidiation. We have found that the activity of NPKC is rhythmic, and that NPKC levels are increased on choline depletion. However, over-expression of NPKC did not lengthen the conidiation period, indicating that PKC in Neurospora may not be responsible for the lengthened period in low choline cultures.

An efficient method was developed for regeneration of Asparagus racemosus Wild, from axillary explants. MS media supplemented with different concentrations of cytokinin (BAP) alone or in combination of different concentrations of auxin (NAA or IBA) was tested for their efficiency in multiple shoot induction. High frequency of multiple shoot regeneration was achieved on MS medium supplemented with BAP (0.1mg/l) and NAA (0.05 mg/l). When shoots were well developed they were dissected and subcultured on the same medium to promote more multiple shoots. Eighteen to twenty shoots were obtained on this medium. The shoots were rooted best on half MS medium supplemented with 0.05 mg/l BAP and 1.0 mg/l IBA. Among the five levels of pH tested, 5.7 was the best for multiple shoot proliferation. The result presented here proved to be suitable for the rapid propagation system of A. racemosus. A protocol was developed for in vitro clonal propagation of Rauvolfia serpentina through direct regeneration from shoot tip explants. Multiple shoots (eight shoots per explant) induction were obtained on MS medium supplemented with 4.0 mg/l BAP and 0.5 mg/l NAA within 10-15 days. The elongated shoots rooted well in half strength MS medium with 0.5 mg/l NAA. The in vitro raised plantlets were acclimatized in glass house and successfully transplanted to field condition with 80 % survival.
