Background: The retinal changes that develop in the eyes during amblyopia are still unclear. It has been proven that thickness of the nerve fiber layer of the retina is positively correlated with refractive error. The objective of the present study was to measure the Retinal Nerve Fiber Layer Thickness (RNFLT) using Ocular Coherence Tomography (OCT) in patients of amblyopia. Method: A descriptive study was conducted in the Department of LRBT Free Eye Hospital, Lahore for 6 months. After meeting the inclusion criteria, 80 patients (40 amblyopic and 40 normal) were taken and grouped as A and B. Socio-demographic details of all the patients were recorded. They then underwent a complete ophthalmologic evaluation that included BCVA, slit lamp biomicroscopy, OCT examination was carried by TOPCON OCT. Each examination was done using the “fast retinal thickness” protocol for each test eye. Mean RNFLT was calculated for each eye. Results: The mean age of patients was 12.58±4.44 years. There were 20 (50%) males and 20 (50%) females. In amblyopic eyes, the mean RNFLT was 125.82±13.06mm while in normal eyes, the mean RNFLT was 94.82±1.11mm. Capsicum has been an important part of diet since centuries and has been widely used traditionally for ailments. The objective of the present analysis was to determine the diuretic and antidiarrheal potentials of four variations of Capsicum annum L. fruits available in local market of Karachi, Pakistan. It belongs to family Solanaceae and extracts were prepared in 95% ethanol and was given in doses of 200 and 400mg/kg and the activity was correlated with commonly used diuretics and anti-diarrheal further reinforcing its conventional use. Fruits extracts were evaluated for anti-diarrheal properties in castor-oil induced diarrhea to confirm its activity. The diuretic activity of the 95% ethanolic extracts of Capsicum annum L. was assessed based on diuresis of 24 hr. collected through metabolic cage. Urinary excretion of water and electrolytes (Na+ and K+) is significantly increased by the standard diuretic drug furosemide, which is comparable with the extract. The anti-diarrheal effects of 95% ethanolic extracts of Capsicum annum L. were examined by castor oil induced diarrhea and all varieties of Capsicum annum L. significantly inhibited the number of defecations.

Aims: Anemia is prime health issue of the population in developing region of world mostly due to dietary deficiencies and poor sanitation. Supplementary medicines as well as iron rich foods are utilized to combat this problem. Herbal products also have value in such diets. We evaluated role of Foeniculum vulgare incorporated diet on hemoglobin concentration and related indices. Study Design: laboratory centred randomized controlled trial. Place and Duration of Study: Pharmacology Department of University of Karachi, Karachi between June 2018 and September 2018. Methodology: After selecting 30 healthy rabbits of either sex, we placed them in three groups; Control, 2% Foeniculum vulgare and 4% Foeniculum vulgare group. Control group, 2% and 4% Foeniculum vulgare groups were maintained on standard rodent diet, standard rodent diet containing 2% and 4% Foeniculum vulgare crushed seeds, respectively. Complete blood count of all rabbits was done twice after interval of a month. Results: In comparison to control group, hemoglobin levels increased in both the study groups, while leucocyte count (WBC) increase was noted only in 4% Foeniculum vulgare group. Conclusion: Foeniculum vulgare may have some role in maintenance of hemoglobin levels if not improving it.

Aims: Alzheimer disease and epilepsy are two of the central nervous system (CNS) disorders that not only affect the quality of life of patients but also of family members and caretakers. Remedies for these illnesses are available in allopathic medicines but not without side effects. Herbal products are being investigated for these ailments. Protective role of Foeniculum vulgare Mill. was assessed in this study. Study Design: Laboratory based randomized controlled trial. Place and Duration of Study: Conducted in Pharmacology Department of University of Karachi, between March 2018 and April 2018. Methodology: Mice and rats were divided in three groups, control, 2% and 4% F. vulgare groups, each containing 10 rodents. Control group was fed standard rodent diet, whereas, study groups were given 2% and 4% F. vulgare seeds (crushed) incorporated in standard rodent diet. Epilepsy model was made for mice and Alzheimer disease study was done using rats and passive avoidance test. Results: Lower incidence of seizures and mortality in both study groups as compared to control in epilepsy model and memory retaining effect in both treated groups in Alzheimer disease model was recorded with statistical significance. Conclusion: Clinical studies should be conducted to validate the protective role of this herb in these disorders.

Aims: Coagulation profile shows the clotting ability of blood. Biochemical tests indicate health status of vital organs such as liver, heart and kidneys. Herbal products are being assessed for their role in affecting these parameters. Coagulation profile and some biochemical parameters were done after interval of a month, for two months. Results: Platelet count and fibrinogen increased while activated partial thromboplastin time (APTT) levels decreased in both the study groups animals as compared to control, while blood urea nitrogen (BUN), creatinine phosphokinase (CPK) and lactate dehydrogenase (LDH) elevation was noted both Foeniculum vulgare groups but within normal range. Conclusion: Foeniculum vulgare may have some role in affecting coagulation and biochemical profile These parameters, however, need clinical trial to validate reliability.

Background Anecdotal experience and studies have shown that most pediatric patients fail to reach target therapeutic vancomycin trough levels (VTLs) and required higher total daily doses (TDD). This retrospective study aims to evaluate the frequency of hospitalized children who achieved target VTLs with a vancomycin (VNCO) dosing regimen of 40-60 mg/kg/d q6h and to assess the VNCO-TDD required to attain the target and their effects on clinical outcomes in pediatric patients. Methods After ethical approval, patients of 3 month-12 years were evaluated in this chart review study who received ≥ 3 intravenous-VNCO doses and appropriately drawn blood samples of VTLs between October 2019 to June 2020. Data were retrieved for demographic and clinical characteristics, culture reports, VNCO-regimen, subsequent steady-state VTLs, concomitant nephrotoxic medications, and serum creatinine. Clinical pharmacists made interventions in VNCO therapy and higher VNCO-TDD were used. Safety of higher vs standard daily doses and their clinical impact on duration of therapy, hospital stay, and survival were evaluated. Results A total of 89 (39.1%) patients achieved target VTLs (SD-group). The smallest proportion (18.2%) of 2–6 years patients achieved target VTLs and reported the lowest mean value of 10.1 ± 0.2 mg/L which was a significant difference (p p = 0.025) longer in SD-group [18.4 ± 12.2 days] than HD-group [15.1 ± 8.9 days]. Nephrotoxicity and electrolyte imbalance were comparable in groups. Length of hospital stay was significantly (p = 0.011) longer [median 22 (range 8–55) days] in SD-group compared to HD-group [median 16 (range 8–37) days]. Number of patients survived in HD-group were significantly (p = 0.008) higher than SD-group [129 (92.8%) vs 75 (84.3%)]. Conclusion Initial Vancomycin doses of 72 ± 8.9 mg/kg/day q6h are required to achieve therapeutic target in 3 months to 12 years patients. High doses are not associated with higher nephrotoxicity than reported with low doses. In addition, efficient pharmacist intervention for the use of higher VNCO-TDD may improve clinical outcomes in terms of duration of therapy, hospital stay, and survival.

Retinal detachment (RD) describes the separation of neurosensory retina from the underlying pigment epithelium. There are various methods of treating RD but in many cases, an unusual delay between occurrence of retinal detachment and surgery has been observed. This study was conducted to find the extent of factors involved in delay in surgery. This cross sectional study was carried out at LRBT Eye Hospital, Lahore for 6 months. The non-probability, consecutive sampling technique was used. The demographic information was recorded. The patients were asked for causes of delay in retinal detachment surgery and all factors were measured. Data was analyzed by SPSS version 21. The mean age of patients was 52±9.86 years; the male to female ratio was 1.5:1. About 9.3% patients said that they do not know where to go, 30% patients thought that it was not a severe condition,36.4% patients thought that it would self-heal,17.1% patients didn't go to the doctor due to financial constraints whereas 7.1% patients did not have VR ophthalmologist near their residence. Statistically significant difference was found between the factors and education level of the patients i.e. p-value0.05. Our study results concluded that people needed to be educated regarding the importance of retinal detachment and surgical procedures and complications associated with it.

Background Therapeutic drug monitoring (TDM) of Vancomycin (VCM) is required to prevent inappropriate dosage-associated bacterial resistance, therapeutic failure, and toxicities in pediatrics. Anecdotal experience and studies show that many healthcare institutions confront barriers while implementing TDM services, this study aimed to assess a pharmacist-directed VCM–TDM service for optimizing patient care in our institution. Materials and methods Patients aged 1 month–18 years who received intravenous VCM were included in this quasi-experimental study. The pre-implementation phase (March–June 2018) consisted of retrospective assessment of pediatric patients, the interventional phase (July 2018 to February 2020) included educational programs and the post-implementation phase (March–June 2020) evaluated the participants based on pharmacist-directed VCM–TDM services as a collaborative-practice model including clinical and inpatient pharmacists to provide 24/7 TDM services. Outcomes of the study included the mean difference in the number of optimal (i) prescribed initial VCM doses (primary) (ii) dosage adjustments and (iii) VCM-sampling time (secondary). After ethical approval, data were collected retrospectively. Results A hundred patients were there in each phase. The number of cases who were correctly prescribed initial VCM doses was significantly higher in the post-implementation phase, mean difference of 0.22, [95% CI (0.142–0.0.358), p p p = 0.079]. Conclusion This study showed the significant role of pharmacist-directed TDM services to optimize the correct prescribing of initial VCM doses and dose adjustments.

Aim: To evaluate the antiamnesic and antidepressant effect of Foeniculum vulgare whole crushed seeds. Study Design: Laboratory based randomized control study. Place and Duration of Study: Department of Pharmacology, University of Karachi between January 2018 to August 2018. Methodology: Thirty swiss albino mice and wistar rats were divided equally in three groups. Control group was fed on standard rodent diet, group 2 was fed on 2% Foeniculum vulgare diet and group 3 was given 4% Foeniculum vulgare diet. Antidepressant activity was assessed using forced swimming test. Memory enhancement effect was evaluated by stationary rod test, passive avoidance test and water maze test. Results: Increased duration of struggling time was noted in both group 2 and group 3 as compared to control in forced swimming test. Decrease in time to reach platform in both water maze and stationary rod test was recorded in both the study group (group 2 & 3). Increase in step through latency was also seen in group 2 and group 3 as compared to control group. Conclusion: The results showed the memory enhancing and antidepressant actions of Foeniculum.

Natural oils are rich in polyunsaturated fatty acids (PUFs) like omega 3, omega 6 and other nutrients that boost physical and mental health. Traditionally these oils have been used to treat joint pain associated with several inflammatory conditions. In this study, we investigated the antioxidant and analgesic properties of the sesame oil (SO), fish oil (FO) and combination of these two oils (SO+FO). Different concentrations of the SO, FO and SO+FO combination 0.02-4mg/ml were used for assessing the free radical scavenging activity by DPPH method and the IC50 value was calculated. Acetic acid-induced abdominal writhing test, tail immersion and hot plate models were used to determined analgesic effect. Results showed that both oils were well tolerated as no signs of toxicity or death were noticed during the observational study period. SO+FO combination showed the best antioxidant properties as shown by DPPH assay. Similarly in analgesic models, SO and FO significantly reduced the number of abdominal contractions (p0.05) however, SO+FO (1:1) exhibited highly significant results (p0.001) in writhing reflex test. Furthermore, SO and FO both increased the reaction time on a hot plate as well as in tail flick test (p0.05) whereas, SO+FO significantly increased reaction time (p0.001) in hot plate and in tail flick test as compared to SO and FO single treatments. Conclusively, our results suggest that the combination of both oils (SO+FO) exhibited significant antioxidant and analgesic potential that it could be considered as one of the active combinations for relieving pain in adjunctive treatment for joint pain associated with rheumatoid arthritis.

The idea of the local drug delivery system is getting popular nowadays to treat gingivitis and periodontitis. The method of delivering the drug locally is quite easy and requires minimal intervention. This delivery system not only treats the periodontal diseases effectively but also prevents the side effects linked with the use of the drugs which are used orally for longer periods to cure these diseases. Chlorhexidine (CHX) is being widely used to treat these conditions because of its broad spectrum anti-bacterial effect and is found to be more effective in lowering plaque formation. The aim of this study was to appraise the effect of the local drug delivery system by using 1% CHX gel in patients with periodontal diseases. 1% CHX gel was prepared and its physicochemical characteristics were then assessed. Clinical parameters and inflammatory salivary biomarkers were evaluated in two groups of patients. Group I: standard treatment group. Group II: gel treatment group. These parameters were evaluated before treatment and after 4 weeks of treatment. 1% CHX gel was highly effective in reducing gingivitis and periodontitis by using the local drug delivery system which allowed the drug to retain into the periodontal pocket for prolong period of time.

The objective of this study was to evaluate the effect of Foeniculum vulgare (FV) on fertility of mice and on serum sex hormone values of rats and rabbits. We used powdered form of Foeniculum vulgare seed and added it to food of mice, rats and rabbits in 2% and 4% ratio which was provided ad libitum throughout study period. Fertility study on mice was conducted for a period of nine months and 3 generations of mice fed on study drug were observed. Rats and rabbits were given Foeniculum vulgare seed powder containing diet for two month and then serum levels of FSH, LH, estrogen, progesterone and testosterone were checked. The number of pups increased in each succeeding generation and no gross abnormality was observed in any of the pups of all 3 generations. The sex hormone levels increased in both sexes of rats and rabbits with exception of FSH and LH levels which were less than 0.1mIU/mL in all study animals. Data was analysed on SPSS. Fertility and sex hormones increased in both male and female when given special diet containing 2% and 4% Foeniculum vulgare.

We determined anti-Parkinson's activity of M. chamomilla L. tea in chlorpromazine (CPZ) developed investigational animal model. In this research, effects of M. chamomilla L. tea 2.14ml/ kg P.O were studied on cataleptic behavior and its effect on brain histopathological changes and immunohistochemistry (IHC) in rats. The experimental design was developed by administering CPZ (3mg/kg, I/P) for twenty-one days to produce Parkinson's disease-like symptoms to 4 animal groups. We observed that chlorpromazine significantly produced motor dysfunctions (catalepsy) in a time period of twenty-one days. The M. chamomilla L. significantly (P0.005) minimized/shorten/taper down catalepsy in rats just like standard group (Levodopa/carbidopa treated group). The maximum reduction was observed from both treated and standard groups on the 21st day. M. chamomilla L. treated rats mid brain sections showed presence of proliferative blood vessels, increase cellularity with reactive glial cells as compared to CPZ group. Furthermore, immunostaining CD68CD21 of M. chamomilla L. treated rats mid brain region showed few CD68 cellsno polymorphs neutrophils after CD21 staining. Thus, this research work disclosed the neuroprotective effect of M. chamomilla L. tea against Parkinson's disease-like symptoms or anti-Parkinson's activity induced by CPZ.

The gold standard therapy for treating the periodontal infections is the removal of bacterial plaque and deposits of calculus from tooth surfaces by scaling and root planning. In order to eliminate these bacterial reservoirs, beside conventional treatment, chemo therapeutic agents are commonly prescribed by periodontologists. To avoid the systemic side effects and development of antibiotics resistance, local drug delivery methods has gained the attention of dentists to treat periodontal infections, along with scaling and root planning. The aim of this study was to evaluate the effectiveness of local drug delivery system in combination with scaling and root planning, by using 1% metronidazole gel and mouthwash. The patients were divided into 3 groups. Group I: conventional treatment group. Group II: patients received treatment with gel. Group III: patients received treatment with mouthwash. All groups received treatment for 30 days. Clinical parameters and salivary concentration of TNF-α, PGE2 and nitric oxide were measured before and after treatment in both groups. All clinical parameters and inflammatory biomarkers significantly reduced in gel and mouthwash group patients (p≤0.001) as compared to patients received conventional treatment. The gel is found to be more efficacious than mouthwash especially in reducing clinical attachment loss (p0.05) and in reducing inflammatory biomarkers (p≤0.001). We strongly suggest the use of metronidazole via local drug delivery system combined with scaling and root planning to treat periodontal diseases.

Herbal products are wealthy in dynamic phytochemicals, such as the terpenoids, various collection of flavonoids, sulfides, lignans constiuents, coumarins concentrates, saponins moities, polyphenolics composite, numerous alkaloids, polyines, furyl mixtures, proteins and related compounds, thiophenes and peptides groups. In this review we discussed pathogeneis, immunity and current herbal treatment strategies of nCOVID-19 to cure this world wide pandemic.

Plants are vital in drug discovery, since many safe and bioactive molecules have been discovered from plants in past, hence this study was designed to evaluate analgesic, anti-inflammatory and toxic effects of Cucumis melo and Citrullus lanatus. Seeds of these plants were selected due to their traditional value for medicinal use. Analgesic activity was determined in mice by Eddy's Hot plate and tail flick method, while anti-inflammatory activity was evaluated by hind paw edema method. Both seed extracts produced highly significant analgesic effects comparable to standard drugs at all three doses by both methods. The extract of C. lanatus showed significant anti-inflammatory activity at 100 mg while showed highly significant activity at 200 mg between 3 to 24 hours as compared to standard drugs. Both extracts did not reveal any mortality up to 1000mg/kg, while there was also no change in normal the gross behavior pattern of the animals at the dose of 50 and 100mg/kg, however there was increase in passivity, sedation and startle response at 200mg/kg. Analgesic and anti-inflammatory effects of extracts may be due to presence of cucurbitacin A, B or E in both seeds which are thought to inhibit COX 2. Results indicate that seeds of C. melo and C. lanatus may be effectively used as adjuvant analgesic and anti-inflammatory agents in situation of chronic pain and inflammation.

The majority of the world population suffers from mental and behavioral disorder. It is the need of the time to find an alternate of presently available medicines in order to decrease the medical expense. Homeopathic remedies are available and prescribed by homeopaths for treatment of anxiety and depression. Unfortunately, no data are available that proves its potential to relieve mental illness. The current study is designed to assess neuro behavioral and antidepressant like effects of homeopathic remedies Staphysagria, Argentum nitricum and Ignatia amara in comparison with standard drug (escitalopram). Different neuro behavioral activities were analyzed. The animals were administered the doses of all homeopathic remedied (60 µl to the rats) and escitalopram (0.042 mg to rats) through the oral route. The activities were observed on day 30th and day 60th. Our result suggests that the swimming time in Staphysagria treated group were significantly improved (p0.001) after day 60th and significance rise was observed (p0.01) in Ignatia amara treated animals, whereas significant decline (p0.05) in struggling time was observed in Argentum nitricum administered animals after the 60th day as compared to 30th day. The central square crossings were improved highly significantly (p0.001) after the 30th day dosing, by all three remedies and peripheral squares crossing were found highly significantly increased (p0.001) after chronic dosing in Staphysagria and Ignatia amara treated groups. It is concluded from the results that all three homeopathic remedies produce comparable effects like standard drug while among all three remedies Staphysagria possess a potent antidepressant activity. To the best of our knowledge the current study reports first time the anti-depressant potential of homeopathic remedies in rodents.

Clinical and hospital pharmacy services are not just medical and pharmaceutical sciences but also occupy significant placement in healthcare system. Pakistan is a developing state with a huge prerequisite for changes in the general wellbeing framework, specially hospital and clinical aspect of pharmaceutical services. The principal intention of this study is to analyze the services offered by different pharmacies in hospitals of Karachi in terms of infrastructure and personnel service qualities. The study was conducted in a cross sectional way that included stratified sampling technique. Reactions were broken down utilizing descriptive and inferential insights of measurements. The fundamental result procedures incorporated the scope of hospital pharmacy services, the general recruitment of clinical drug specialists (pharmacist), the product and equipment used in hospital pharmacy services, the background of staff (educational), acquisition of proficient training mode, practical involvement and experience. The clinical pharmacy facilities coverage mutually on the departmental scale (median =22.43%) and patient scale (median =17.25%) do not comply the 100% coverage that is obligatory for standard practices. In addition, 48.65% of the pooled hospitals data has shown absence of distinct administration rules for hospital and clinical pharmacists, and 45.33% lacks the use of rational drug software. It is concluded that important parameters like drug monitoring, medication records keeping; appropriate drug information software's and quality assurance in hospitals still need attention for better patient outcomes.

Natural oils are enriched with polyunsaturated fatty acids (PUFAs) which are important for our health. Recent experimental data explained that PUFAs might have a beneficial effect on various brain functions such as anxiety, dementia, epileptic seizures, depression or bipolar and other neurobehavioral diseases. The objective of the current research work was to evaluate the effect of sesame oil, fish oil and mixture of both oils (1:1) on neurobehavioral changes and cognition. For this purpose shark fish oil and sesame oil were extracted out and there poly unsaturated and saturated fatty acids were analyzed by using GCFID that exposed the presence of different PUFs in shark fish oil, sesame oil and mixture of both oils. Neurobehavioral changes were seen after 5ml/kg/day sesame oil, 5ml/kg/day shark fish oil and 1:1 combination of both oil 5ml/kg/day administration on open field, cage crossing, light and dark, stationary rod, forced swimming induced depression test and water maze test. Our GCFID results showed sesame and fish oil enriched with higher amount of PUFs and showed significant anxiolytic and antidepressant like effect after 30 days of treatment (P0.05) however combination of these both oils exhibited greater efficacy (P0.01) in reducing anxiety and depression as imipramine standard drug. Results showed that combination of both oils (sesame oil and fish oil) could be a better option to treat neurobehavioral problems as compared to alone.

Camel milk is reported as anti-diabetic, hepato-protective, anticancer, antioxidant, antiviral and neuroprotectant in numerous studies. Based on its neuroprotective profile, camel milk is investigated for its possible beneficial effect in treating anxiety and depression and its effect on brain biogenic amines in the present study. Head dip, cage crossing, stationary rod, elevated plus-maze, open field, lightdark box and forced swim tests were used to measure change in rodents' behavior after camel milk administration. Any possible change in brain biogenic amines level after camel milk treatment was evaluated using High Performance Liquid Chromatography (HPLC) technique. Camel milk administration resulted in significant increase (p0.001) in exploratory and locomotor activity and showing anxiolytic behavior in rodents. In depression-like model, rats showed significant increase (p0.001) in struggling time after 30-days administration of camel milk. HPLC detection of brain biogenic amines revealed significant increase (p0.001) in norepinephrine, insignificant increase in 5-hydroxytryptamine and significant decrease (p0.001) in dopamine, 3,4-dihydroxyphenylacetic acid, homovanillic acid and 5-hydroxyindoleacetic acid in camel milk treated group. Based on above findings, camel milk is suggested as anxiolytic and antidepressant in the administered doses. However, further experimental and clinical investigations are required to authenticate the same at different doses.

Simvastatin is an anti-hyperlipidemic drug which reduces the cholesterol synthesis and also has anti-inflammatory, immunomodulatory and anti-microbial action against the bacteria. This develops the interest of periodontologist to use it in combination with conventional treatment to treat periodontal diseases. The objective of the study was to develop the gel and mouthwash of simvastatin and use it locally to treat gingivitis and periodontitis as an adjunct to scaling and root planning. The patients were randomly allocated into three groups that were standard treatment group, gel treatment group and mouthwash treatment group. Results indicated that simvastatin gel and mouthwash in 1% preparation showed favorable results by significantly reducing periodontal parameters and inflammatory biomarkers (p0.001) as compared to standard treatment. Thus, we strongly suggest the use of simvastatin by local drug delivery system as an adjunct treatment of scaling and root planning.

The herbal agents rich in flavonoids are progressively becoming popular these days as they are considered to have anti-oxidant effects and also lack bothersome effects. That's why existing research was encircled around the anti-coagulant effect of Nelumbo nucifera fruit (NNF) as it is also a rich source of flavonoids and cultivated in abundance especially in tropical regions of Asia but its usefulness as anti-coagulant agent was never determined pharmacologically. Anticoagulant assessment was done in thirty five male Wister rats which were separated equally in 5 groups. Results of the current study revealed that NNF 200mg/kg significantly prolonged prothrombin time and thrombin time, whereas fibrinogen level was highly significantly reduced as compared to control. Fibrinogen level was also reduced highly significantly with NNF 100 mg/kg as compared to control without affecting other parameters of coagulation i.e. activated partial thromboplastin time, prothrombin time and thrombin time. NNF exhibited strong anti-coagulant activity which may be due to the inhibitory effects on platelet activation, adhesion and aggregation along with inhibitory effects on thromboxane A2 formation. Presence of alkaloid i.e. neferine and flavonoids in it may be a reason of its anti-coagulant activity but more pre-clinical and clinical evaluation needs to be conducted to establish these findings.

Opioids and non-opioids have long been used as analgesic, anti-inflammatory and antipyretic. Long-term use of these drugs may lead to severe toxicities. Therefore natural remedies are now being explored to avoid risk of adverse effects associated with the use of these conventional medicines. Bioactive components from milk of different species have been identified as nutraceuticals, but no experimental or clinical study is conducted so far to explore the analgesic and anti-inflammatory potential of camel milk. In this study we evaluated camel milk for its possible analgesic and antiinflammatory activity. The anti-inflammatory effects of camel milk was studied in rats using paw edema method (induced by acetic acid) while tail-flick method was used to evaluate its analgesic effect in mice. Significantly increased tail-flick latency was shown after camel milk (33ml/kg) treatment when compared with acetylsalicylic acid at all time intervals. Anti-inflammatory activity of camel milk was significant (p0.001) at 4th hour of treatment as shown by maximum percentage inhibition in edema volume (46.84%) in comparison to control. Results of our present study suggested possible use of camel milk as adjuvant therapy in treating various chronic pain and inflammatory ailments. Camel milk could further be investigated in future for recognition of biochemical constituents responsible for its antiinflammatory and pain relieving activities.

This novel study was conducted with objective to evaluate the anxiolytic potential of whole-crushed seeds of Foeniculum vulgare (FV) which were incorporated in diet. Albino mice were divided in three groups: Control-group, Study-group 2% FV and Study-group 4% FV, each having 10 mice. Special dietary pellets containing whole-crushed Foeniculum vulgare seeds were prepared in 2% and 4% ratio, respectively and were fed to respective Study groups whereas Control-group was given regular rodent diet for 2 months. Animal behaviour was assessed using Home Cage Activity test, Head Dip test, Light and Dark Box test and Open Field test at intervals of 15 days for a period of 2 months. The results of this study showed, decrease in Cage Crossing activity, more number of Head Dips, increased time spent in Light box and increase in number of transitions between Light and Dark Box, increased number of Central Squares Crossed and increased time spent in Central Squares of Open Field arena for both study groups in comparison with control group. Foeniculum vulgare whole-crushed seeds diet of 2% and 4% was found to have anxiolytic effect.

Cocos nucifera (coconut) water is a traditionally used beverage and sportsdrink around the world. The recommendations on the use of plants for different conditions often lack proper scientific research. This study is first of its kind to explore, compare and document scientific evidence of the effect of different stages cocos nucifera (coconut) water on hematological and coagulation parameters in rabbits. For this study, we selected healthy rabbits and used their blood samples for our target analysis. 4ml/100g of young and mature cocos nucifera (coconut) water was administered orally once a day to respective groups for 60 days. Control group was administered distilled water orally. Results showed positive effect of coconut water on key hematological parameters including red cells and hemoglobin. Coagulation profile revealed significant increase in platelets and fibrinogen level with decrease in bleeding time and increase in clotting time by coconut water. This study revealed slightly dominating profile of young coconut water over mature coconut water which may be attributed to the unique composition differences between young and mature coconut water including micronutrients, carbohydrates and phenolic components. Outcomes of this research lead to the speculation about possible effect of cocos nucifera (coconut) water in anemia and bleeding disorders.

To determine the effects of Salvia hispanica on activities related to memory, anxiety and depression. Albino rats as well as albino mice were utilized in the current study. Two groups of animals were made including 10 animals in each group. One was a control group and another was treated group. Neuropharmacological parameters were assessed using Light and Dark box test, Stationary rod activity, Water maze test, Open field activity and Home cage activity. The control group was maintained on water and treated group was fed with approximately 106 mg/kg extract of Salvia hispanica for 30 days. The observations were recorded on 1st day, 15th day and 30th day. The results of current study showed an increased time spent in the light box of Light and Dark box model, reduction in elapsed time utilized by animal to reach platform in Stationary rod and water maze model, reduced number of peripheral square and central square crosses in the open field and decreased number of cage crosses in the home cage activity. Salvia hispanica shows memory enhancement and also shows an antidepressant activity on chronic administration.

Methylphenidate is a psychostimulant used for the treatment of (ADHD) attention deficit hyperactivity syndrome in children and adults. After chronic administration it is known to produce behavioral disorders including anxiety. Previous studies demonstrated that co-administration of buspirone can reduce behavioral and cognitive adverse effects produced by methylphenidate. The aim of the present study is to measure the levels vanillylmandelic acid (VMA) excretion in urine following prolong administration of methylphenidate, buspirone and their combination. Samples of urine for the estimation of the urinary VMA excretion were collected from treated and control male Wistar rats. We found significant (P0.01) raised urinary VMA excretion in methylphenidate group however significant (P0.01) reduction in VMA levels were seen after buspirone co-administration. Excretion of VMA in urine would allow the monitoring of sympatho-adrenomedullary system activity. This study could be helpful to increase the clinical use of methylphenidate in the treatment of different disoders.

Pain is a very common symptom and pain management is most important challenge to alleviate the suffering and for improving the quality of life of those living with chronic pain. Beet root is a vegetable consumed as salad and present study is designed to evaluate the analgesic activity of its lyophilized extract. The study was conducted in 2016 (March) on albino mice of both sexes weighing 18-25 gm, divided into groups comprising of 10 animals each. Group I was taken as control and administered 0.1ml distilled water orally. Group II was labeled as Treated and administered lyophilized beet root in the dose of 1000mg/kg. Group III was taken as Standard and was given acetyl salicylic acid 300mg/60kg. Analgesic activity was evaluated using Hot plate apparatus, Tail Flick method and Writhing method. Results showed significant (p0.001) analgesic effect by beet root as compared to control. Beet root also showed significant (p0.001) effect as compared to standard but the effect was not seen from the beginning indicating its role in the second phase of analgesia. This study suggests that beet root powder possesses analgesic potential and can be used for central as well as for peripheral analgesia.

The study is conducted to observe and investigate the effects of oral dosing of methanolic extracts of Cuminum nigrum (L) and Centratherum anthelminticum (L) on neuropharmacological activities of mice. Methanolic extracts of Cuminum nigrum (L) and Centratherum anthelminticum (L) were soluble in Dimethyl sulphoxide (DMSO) i.e. an organic solvent, so it is used in this study. Screening for anxiolytic and antidepressant effects were performed using open field test, head dip test, stationary rod test, cage crossing test, light and dark box and swimming- induced depression test. Thirty animals were divided into three groups of 10 animals each and numbered as 1 (control, on DMSO), 2(on methanolic extract of Cuminum nigrum (L), 3 (on methanolic extract of Centratherum anthelminticum (L). The extracts and DMSO were administered orally for 60 days. Any possible change in animal behavior was evaluated on day 15, 30 and 60 of dosing. The groups 2 and 3 showed significant increase (p0.001, p0.01) in open field activity and light and dark box test respectively, while significantly decreased activity was observed in head dip and cage crossing activity (p0.01) after 60 days of dosing. Based on above findings, it is suggested that the extracts of Centratherum anthelminticum (L) and Cuminum nigrum (L) have antidepressant and anxiolytic potential with sedative effects.

This retrospective study reports impact of diabetes on incidence rate of dose limiting symptoms of neurological toxicity and chemotherapy induced peripheral neuropathy (CIPN). Post-surgical colorectal cancer (CRC) patients with metastatic disease, treated with four different schedules of FOLFOX were included in this study. Neurological adverse effects were assessed by CTC v2.0. The incidence rate of adverse neurological symptoms in CRC patients, clinically diagnosed with diabetes (n=6) were compared with non-diabetic CRC patients (n=32). The results show that the difference in the incidence rate of paresthesia is significant (p=0.043) between diabetic and non-diabetic patients. The difference in the incidence rates of hypoesthesia (p=0.445), peripheral neuropathy (p=0.889), dizziness (p=0.445), insomnia (p=0.690), taste disturbances (p=0.258), and headache (p=0.498) in diabetic and non-diabetic CRC patients was not significant. The findings indicate that risk of frequent, distal and transient paresthesia within the first few minutes of Oxaliplatin infusion is higher in diabetic CRC patients.

In Pakistani population the prevalence of Calcium and vitamin D deficiency is at alarming rate. Previous studies show that globally vertebral osteoporosis is most commonly recognized site causing deterioration to personal life satisfaction. It is very unfortunate that in Pakistan ample amount of research work has not been done in the area, consequently, information on rate of vertebral osteoporosisfracture are rare in Pakistan. There is no reduction in T-score on supplementation with calcium and vitamin D3 administration. There is reduction in T-score on supplementation with calcium and vitamin D3 administration. The prime objective of the current work was to determine vertebral spine osteoporosis treatment efficacy in local population. This is an intervention experimental study with no control. The study population was selected from the local community; consisting of individuals with vertebral spine osteoporosis, further they were followed for up to 6 months. Data was analyzed by SPSS-22. Tabs Chewable: Calcium: 1250 mg, Cholecalciferol: 125 IU, BD/Day was advised. The mean T-score before and after treatment were recorded as; Mean ±S.D: 2.890 ±1.7217 and Mean ±S.D: -2.456±0.8064 respectively. The findings of the current work do not provide support for routine supplementation with calcium and vitamin D3 orally for osteoporosis.

Methylphenidate is commonly use for the treatment of attention deficit hyperactivity disorder (ADHD), but its long term use was found to produce hepatic necrosis in mice. Purpose of this study was to investigate that co-administration of buspirone (drug which attenuates methylphenidate induced sensitization) may attenuate methylphenidate-induced hepatotoxic effects and to determine the effect of challenge dose of haloperidol (D2 antagonist that blocks the effects of methylphenidate in case of intoxication) on SGPT and SGOT levels in methylphenidate treated rats. Estimation of SGPT and SGOT were performed using kit method. Prolong oral administration of methylphenidate at a dose of 2.0 mg/kg/day, buspirone at a dose of 10 mg/kg/day, their co-administration and challenge dose of haloperidol (1 mg/kg i.p.) in rats increased SGPT concentration and decreased SGOT concentration, effect is more pronounced in methylphenidate treated rats and potentiate with administration of haloperidol challenge dose. In conclusion our analysis showed that methylphenidate and challenge dose of haloperidol is associated with elevation of SGPT in rats, which is attenuate in co-administration of methylphenidate buspirone treated rats. To quantify the risk of methylphenidate-induced hepatic injury and role of buspirone to reduce the injury further pharmacoepidemiological investigations are required.

Parkinsonism is characterized by rest tremor, inflexibility, balance debilitation, slow motion and dementia. It is known to be caused by the deficiency of dopaminergic neurons in nigrostriatal pathway. Different studies propose that oxidative burden may be included in the apoptotic process in parkisnons disease. Zamzam water being alkaline in composition may diminish the oxidative stress and hence relieve the symptoms. Therefore, the purpose of this study was to explore the neuroprotective effect of zamzam water in chlorpromazine induced animal model of Parkinsonism. Results revealed that zamzam water did not show significant anticataleptic effect after 21 days as compared to chlorpromazine treated group. However, after 30 days of giving zamzam water showed highly significant decrease (p0.001) in cataleptic score as compared to chlorpromazine treated group that is negative control. After 30 days of dosing, cataleptic scores by zamzam water were closer to standard drug but standard drug (levodopa/carbidopa) still showed better results than zamzam water. Results from histopathological study of rat's brain also revealed regenerative changes by zamzam treated water when compared with negative control. This regenerative change after zamzam water treatment might play a positive role in future if administered continuously. These results also suggest that zamzam water can be used in combination with standard drug to produce synergistic effect in the management of parkinsons disease.

Kidney cells damage and subsequent renal adverse effects with oxaliplatin are less reported phenomena, whereas cisplatin (CDDP, first generation platinum compound) has therapeutic limitations due to renal toxicity. This experimental study reports oxaliplatin (third generation platinum compound) induced direct damage in rat kidney tissues and alterations in renal biochemical profile. Oxaliplatin was administered in albino wistar rats with 5-FU (5 Fluorouracil) to mimic as model of FOLFOX, the mainstay chemotherapeutic regimen in colorectal cancer (CRC). This study reports changes in renal biochemical profile (serum creatinine and urea) in rats treated in different treatment groups with cisplatin, oxaliplatin, 5-FU, cisplatin+5-FU and oxaliplatin+5-FU which are compared with group of rats treated with normal saline (control group). Subjective renal toxicity in tissues was compared among rats treated with oxaliplatin alone and cisplatin, with and without 5-FU by light microscopy. Cast formation, medial hypertrophy of the vessel wall, vacuolization and necrosis was seen in kidney tissues of oxaliplatin treated rat. Changes in serum creatinine well-above diagnostic risk levels were noted. Apparent tubular degenerative sequence associated with vacuolization and cast formation was observed in 5-FU treated rats. Kidney damage ensued after treatment with 5-FU and oxaliplatin are slightly comparable to massive tubular damages, hemorrhage, casts and vacuolization along with multiple foci of alterations induced by cisplatin.

Parkinson's disease (PD) is a long-lasting neurodegenerative brain disease. It is characterized by a gradual decline in motor and non motor symptoms especially postural instability, tremors and memory impairment with localized loss of neurons mainly in the Substantia nigra. In the current research we evaluated the effects of Non-steroidal anti inflammatory drugs (NSAIDs) on motor coordination and memory in chlorpromazine (CPZ) induced Parkinson's experimental model. Intraperitoneal (i.p.) injection of CPZ (3 mg/kg) was given to all rats for 21 days to induce Parkinson like symptoms; ibuprofen (40mg/kg/day) and celecoxib (20mg/kg) were administered 30 minutes after CPZ injection. Behavioral parameters like Catalepsy, muscle strength (wire hanging test), locomotor activity (open field test) were observed. Moreover, its effect on memory was explored by the use of water maze and passive avoidance test. Our results showed CPZ significantly induced motor fluctuation and cognitive impairment in a period of 21 days. Celecoxib and ibuprofen significantly improved cataleptic scores (P0.01), locomotion and muscular coordination in open field (P0.01) and in wire hanging test (P0.01). Significant improvement in memory was observed with celecoxib (P0.01) and ibuprofen (P0.05) in water maze test as well as in passive avoidance test. Therefore, the present study showed neuroprotective and memory enhancing effect of ibuprofen and celecoxib against CPZ induced Parkinson's model.
