Kaempferia galanga L. extract contains ethyl p-methoxycinnamate (EPMC) and ethyl cinnamate (EC), which have several pharmacological activities. EPMC and EC have been successfully isolated, but the %yield was low. Therefore, developing an isolation method to increase the %yield result of EPMC and EC is essential. The molecularly imprinted polymers have been applied to separate lot of active compounds from natural products with excellent results. MIP synthesis is usually performed using a single template with high selectivity for the target analyte but only detect single chemical compounds. Hence, this study synthesized multi-template molecularly imprinted polymers (MT-MIPs) for isolating EPMC and EC simultaneously using methacrylic acid as a functional monomer and ethylene glycol dimethacrylate or trimethyl propane trimethacrylate (TRIM) as a crosslinker. The study results indicate that MT-MIP produced with TRIM is more effective in separating EPMC and EC simultaneously in K. galanga L. extracts. However, the yields of EPMC and EC were still low. The yields of EPMC and EC in n-hexane extracts were 1.557 and 1.929%, with purity of 66.330 and 61.510%, respectively. Further research is necessary to determine the ideal functional monomer and its ratio to template molecules to obtain the excellent selectivity of the MT-MIPs used for simultaneously isolating EPMC and EC.

Molecularly Imprinted Microspheres (MIMs) or Microsphere Molecularly Imprinted Polymers represent an innovative design for the selective extraction of active compounds from natural products, showcasing effectiveness and cost-efficiency. MIMs, crosslinked polymers with specific binding sites for template molecules, overcome irregularities observed in traditional Molecularly Imprinted Polymers (MIPs). Their adaptability to the shape and size of target molecules allows for the capture of compounds from complex mixtures. This review article delves into exploring the potential practical applications of MIMs, particularly in the extraction of active compounds from natural products. Additionally, it provides insights into the broader development of MIM technology for the purification of active compounds. The synthesis of MIMs encompasses various methods, including precipitation polymerization, suspension polymerization, Pickering emulsion polymerization, and Controlled/Living Radical Precipitation Polymerization. These methods enable the formation of MIPs with controlled particle sizes suitable for diverse analytical applications. Control over the template-to-monomer ratio, solvent type, reaction temperature, and polymerization time is crucial to ensure the successful synthesis of MIPs effective in isolating active compounds from natural products. MIMs have been utilized to isolate various active compounds from natural products, such as aristolochic acids from Aristolochia manshuriensis and flavonoids from Rhododendron species, among others. Based on the review, suspension polymerization deposition, which is one of the techniques used in creating MIPs, can be classified under the MIM method. This is due to its ability to produce polymers that are more homogeneous and exhibit better selectivity compared to traditional MIP techniques. Additionally, this method can achieve recovery rates ranging from 94.91% to 113.53% and purities between 86.3% and 122%. The suspension polymerization process is relatively straightforward, allowing for the effective control of viscosity and temperature. Statins are cholesterol-lowering drugs with a mechanism of inhibiting 3-hydroxy-3- methylglutaryl-CoA reductase, but long-term use can cause side effects. An example of a plant capable of reducing cholesterol levels is Angelica keiskei (ashitaba). Therefore, this study aimed to obtain suitable compounds with inhibitory activity against the HMG-CoA reductase enzyme from ashitaba through in silico tests. The experiment began with screening and pharmacophore modeling, followed by molecular docking on ashitaba’s compounds, statins groups, and the native ligand was (3R,5R)-7-[4-(benzyl carbamoyl)-2-(4-fluorophenyl)-5-(1-methylethyl)-1H-imidazole1-yl]-3,5-dihydroxyheptanoic acid (4HI). Based on the results of the molecular docking simulations, 15 hit compounds had a small binding energy (∆G). Pitavastatin, as the comparator drug (∆G = −8.24 kcal/mol; Ki = 2.11 µM), had a lower ∆G and inhibition constant (Ki) than the native ligand 4HI (∆G = −7.84 kcal/mol; Ki = 7.96µM). From ashitaba’s compounds, it was found that 4′ -O-geranylnaringenin, luteolin, isobavachalcone, dorsmannin A, and 3′ -carboxymethyl-4,2′ - dihydroxy-4′ -methoxychalcone have low ∆G of below −6 kcal/mol. The lowest ∆G value was found in 3′ -carboxymethyl-4,2′ -dihydroxy-4′ -methoxy chalcone with a ∆G of −6.67 kcal/mol and Ki value of 16.66 µM, which was lower than the ∆G value of the other comparator drugs, atorvastatin (∆G = −5.49 kcal/mol; Ki = 1148.17 µM) and simvastatin (∆G = −6.50 kcal/mol; Ki = 22.34 µM). This compound also binds to the important amino acid residues, including ASN755D, ASP690C, GLU559D, LYS735D, LYS691C, and SER684C, through hydrogen bonds. Ashitaba or also known as Angelica keiskei is the plant used in our community service activities. This plant is an herbal plant originating from Japan which has many properties as traditional medicines. Ashitaba leaves are now cultivated well in Indonesia. Ashitaba is known to be very potential because it contains many active compounds such as chalcone and phenol which has activity to inhibit tyrosinase enzymes and antioxidants so that it can be used as a lightening agent. In addition, ashitaba is also known to have benefits for cosmetics, especially for skin health. Lemon is known to have a specific odor derived from volatile compounds and phenols. Lemon extract has anti-acne activity. The community service activities regarding the use of a combination of lemon and ashitaba as raw materials for cosmetic preparations are important in order to take advantage of the potential of plants in Indonesia. Vitamin K can reduce warfarin's anticoagulant action, causing a variance in response among individuals taking warfarin. Vitamin K comes in two forms, namely Vitamin K1 (phylloquinone) and K2 (menaquinones). Menaquinone-4 (MK-4) is a kind of Vitamin K2 found in meat and dairy products. Analysis of MK-4 levels in human plasma is very useful for patients who receive warfarin therapy. High-performance liquid chromatography (HPLC) can be used for warfarin's bioanalysis, and it must be validated. The purpose of this study was to validate the bioanalytical method for quantification of Vitamin K2 (MK-4) in human plasma according to the 2019 European Medicines Agency (EMA) guideline. Vitamin K2 (MK-4) was extracted using acetonitrile. HPLC with an ultraviolet detector at 245 nm, using a T3 column set at 30°C and an isocratic mobile phase containing methanol: phosphate buffer (95:5) at pH 3, a flow rate of 1 mL/min was used in this study. The warfarin concentration of 0.5–3 μg/mL was used. About 5.50%–17.42% and 6.18%–8.74%, respectively, were the average ranges of percentage coefficient of variation and percentage difference. There was no response at the analyte's retention time in the six blank plasmas and at the analyte's retention time in the blank after the injection of upper limit of quantification, indicates that the procedure was very selective and did not result in any carryover. This bioanalytical method fulfills the parameters of selectivity, accuracy, precision, and carryover based on the 2019 EMA guidelines. [ABSTRACT FROM AUTHOR]

Diazepam is a benzodiazepine that has the potency to be misused because it is effective, easily obtained, and inexpensive. The misuse of diazepam is to replace illegal drugs and be a sedative. Separation of diazepam is needed to detect possible drug abuse and to monitor drug levels in blood to ensure the effectiveness of the drug. This study was conducted to obtain a molecularly imprinted solid-phase extraction (MI-SPE) sorbent to separate diazepam from serum samples. This work started at the synthesis stage with the bulk polymerization method, using methyl methacrylate and acrylamide as functional monomers, diazepam as a template, and ethylene glycol dimethacrylate as a crosslinker. The polymer obtained was identified by its adsorption capacity and packaged into a solid-phase extraction (SPE) cartridge, and the extraction conditions were optimized. The optimization results were then used to extract diazepam from the serum sample. The test results showed that the adsorption ability of the molecularly imprinted polymer (MIP) with the functional monomer, methyl methacrylate, was 63.98 ± 0.1%, which is higher than that of the acrylamide MIP monomer, with a value of 43.27 ± 0.1%. The MIP sorbent of methyl methacrylate was applied to the SPE with 200 mg of polymer in a 3 mL cartridge. Diazepam added to serum samples were then passed through the MIP-SPE producing a percent recovery value of 95.31 ± 1.1% for MIP and 60.83 ± 0.3% for nonimprinted polymer (NIP). The results showed that the MI-SPE sorbent made from the monomer methyl methacrylate gave higher extraction recovery results than acrylamide, and it could be used for extracting diazepam from serum samples with or without other substances. Objective: Atenolol is one of beta-blocker are prohibited as doping based on World Anti-Doping Agency (WADA). The purpose of this study was to the synthesis of molecular imprinted polymer (MIP) for extraction of atenolol from the sample. Method: This research compared the two of the method, bulk and precipitation polymerization. The MIP was successfully prepared from methacrylic acid as a functional monomer, ethyleneglycoldimethacrylate as a crosslinker, benzoyl peroxide as an initiator, butanol as a porogenic solvent with atenolol as a template molecule. Result: The result showed that the bulk polymerization method produces sorbents that have good adsorption capacity and small particle compare to the precipitation polymerization. Both methods were selective for atenolol. Conclusion: Generally, the MIP solid phase extraction is an alternative method for extraction atenolol from the sample. [ABSTRACT FROM AUTHOR]
