Abstract

The small giant clam, Tridacna maxima, is distributed from the Red Sea and East African coast to French Polynesia. Across this widespread Indo‐Pacific range, T. maxima shows strong population structure, in agreement with its limited dispersal abilities. Peripheral populations may have smaller effective population sizes, increasing their vulnerability under any environmental changes. Understanding evolutionary processes at play in such regions located at the edges of T. maxima distribution is a prerequisite in the context of transfers and restocking programmes. In this study, giant clams were sampled from 14 atolls and islands within four archipelagos in the peripheral region of French Polynesia, in 2001–2002 and/or in 2012–2013, then genotyped at the COI gene and at nine microsatellite loci. Mitochondrial lineages of T. maxima from French Polynesia diverged from those sampled in Micronesia, Melanesia, the Coral Triangle and the Red Sea by 6.6–7.3%. Within French Polynesia, significant genetic structure was found, indicating restricted gene flow, and it was stable through time. Most of the genetic variation at microsatellite loci was between archipelagos. The most differentiated archipelago was the most geographically isolated (the Austral Islands). The current patterns of genetic structuring of T. maxima in French Polynesia probably result from long‐term genetic isolation with limited dispersal ability. In addition, these results underlined that sufficiently large populations of T. maxima have persisted in the Central Pacific during the last sea‐level regression. Strategies to optimize transfers and restocking programmes should be designed to preserve the genetic diversity and structure observed here, to avoid the risks of altering the genetic structure, allele loss and/or introduction of maladapted alleles in the receiving populations.

Abstract

Oxidative stress contributes to impairment of skin health, the wound healing process, and pathologies such as psoriasis or skin cancer. Five Polynesian medicinal plants, among the most traditionally used for skin care (pimples, wounds, burns, dermatoses) are studied herein for their antioxidant properties: Calophyllum inophyllum, Gardenia taitensis, Curcuma longa, Cordia subcordata, and Ficus prolixa. Plant extracts were submitted to in vitro bioassays related to antioxidant properties and their bioactive constituents were identified by a metabolomic analytical approach. High performance liquid chromatography with tandem mass spectrometry (HPLC-MS/MS) analysis was performed leading to the characterization of 61 metabolites. Compounds annotated for F. prolixa and C. subcordata extracts were reported for the first time. Antioxidant properties were evaluated by total phenolic content (TPC), free radical scavenging DPPH (1,1-diphenyl-2-picryl-hydrazyl), and Ferric Reducing Antioxidant Power activity (FRAP) assays. F. prolixa extract was the most active one and showed antioxidant intracellular activity on keratinocytes by Anti Oxydant Power 1 assay. Online HPLC-DPPH allowed the identification of phenolic bioactive compounds such as quercetin-O-rhamnoside, rosmarinic acid, chlorogenic acid, procyanidins, epicatechin, 5-O-caffeoylshikimic acid, and curcumin as being responsible for the scavenging properties of these plant extracts. These results highlight the potential of F. prolixa aerial roots as a source of antioxidants for skin care applications

Abstract

Calophyllum inophyllum L. (Calophyllaceae), locally called “tamanu” in French Polynesia, is an evergreen pantropical tree growing mostly along the seashores. Its barks, leaves, and fruits are still used in traditional medicine. The oil expressed from the nuts has been also traditionally used. Tamanu oil is topically applied on skins as well as mucous membrane lesions. This oil is especially recommended to heal all kinds of skin ailments. Bioassays and different assessments of Tamanu oil revealed numerous biological activities (antioxidant, anti-inflammatory, antibacterial, wound healing…), so bringing scientific evidence of beneficial effects of this oil on human skin healing. Such biological properties may explain the use of tamanu oil as an active cosmetic ingredient recorded as “Calophyllum inophyllum seed oil” by the INCI (International Nomenclature of Cosmetic Ingredients). Most of the bioactive properties of tamanu oil are attributed to oil composition including the presence of resinous compounds in tamanu oil beside common fatty acids, which constitutes a unique characteristic of this healing oil. Actually, resinous part of tamanu oil is known to contain bioactive secondary metabolites mostly constituted by neoflavonoids including pyranocoumarin derivatives. Herein, chemical constituents and biological properties of tamanu oil are presented with a focus of its traditional use inspiring modern valuations related to cosmetic field.

Abstract

Tamanu oil, obtained from the nuts of Calophyllum inophyllum L. (Calophyllaceae), was traditionally used to cure various skin problems and ailments in French Polynesia. Nuts and containing oil are also used for skin care. Since the drying of nuts is an important stage for oil preparation, the objective of our study was to develop an analytical method to evaluate oil quality during nuts drying process following different drying parameters. Using a metabolomic approach combining LC-MS/MS analysis and molecular networking, the obtained data revealed differences in the metabolites chemical classes occurring in resulting oils. For the first time, our study provides new findings regarding the occurrence and evolution of the metabolites in tamanu nuts during the drying process. The developed method provided a powerful analytical tool aiming a better identification of bioactive components formed in tamanu nuts during drying process and will be helpful to produce high quality oil for a natural active cosmetic ingredient. An efficient metabolomic approach was implemented to identify markers inducing variability in chemical composition during drying process of nuts. Thus, this set-up analytical method successfully applied in tamanu could be used to the study of metabolites of a wide range of plants., L'huile de tamanu, obtenue à partir des amandes de Calophyllum inophyllum L. (Calophyllaceae), est traditionnellement utilisée pour soigner divers problèmes et affections cutanées en Polynésie française. Les amandes et l'huile qu'elles contiennent sont également utilisées pour les soins de la peau. Elles sont connues pour traiter différents types d'affections cutanées et utilisées comme ingrédient cosmétique naturel. Le séchage des amandes étant une étape importante pour la préparation de l'huile, l'objectif de notre étude était de développer une méthode analytique pour évaluer la qualité de l'huile (composition chimique et rendement en huile) pendant le processus de séchage des amandes et en fonction de différents paramètres de séchage (facteurs abiotiques). En utilisant une approche métabolomique combinant l'analyse LC-MS/MS et la mise en réseau moléculaire, les données obtenues ont révélé des différences au sein des classes chimiques de métabolites présents dans les huiles. Des marqueurs induisant la variabilité de la composition chimique pendant le processus de séchage des amandes de tamanu ont pu être identifiés. La méthode développée fournit un bon outil analytique qui apporte une meilleure identification des composés bioactifs formés dans les amandes de tamanu pendant le processus de séchage et sera utile pour produire une huile de qualité en tant qu’ingrédient cosmétique naturel. De plus, cette approche métabolomique mise en place dans le cas du C. inophyllum pourra être utilisée pour l'étude des métabolites provenant d’une large gamme de plantes.

Abstract

The term cosmetopoeia refers to the use of plants in folks’ cosmetics. The aerial parts of Bidens pilosa L., the leaves of Calophyllum inophyllum L. and the fruits of Fagraea berteroana A.Gray ex Benth are traditionally used in French Polynesia for hair and skin care. During the hair cycle, dermal papilla cells and their interaction with epithelial cells are essential to promote hair follicle elongation. The aim of our investigations was the identification of metabolites from these three plants and chemical families responsible for their hair growth activity. A bioactivity-based molecular network was produced by mapping the correlation between features obtained from LC-MS/MS data and dermal papilla cell proliferation, using the Pearson correlation coefficient. The analyses pointed out glycosylated flavonols and phenolic acids from B. pilosa and C. inophyllum, along with C-flavonoids, iridoids and secoiridoids from F. berteroana, as potential bioactive molecules involved in the proliferation of hair follicle dermal papilla cells. Our results highlight the metabolites of the plant species potentially involved in the induction of hair follicle growth and support the traditional uses of these plants in hair care

22. Polynesian medicine used to treat diarrhea and ciguatera: An ethnobotanical survey in six islands from French Polynesia

Abstract

Ethnopharmacological relevance In French Polynesia, many pathologies common or endemic to the territory cause diarrhea. This is the case for rotavirus gastroenteritis, salmonella food poisoning, ingestion of water contaminated by bacteria, and ciguatera. To treat these ailments, the population may employ traditional medicine for cultural reasons, geographical isolation, and poor health coverage. Polynesian remedies are often used without medical consultation and there is no data on their benefit-risk balance. A few ethnobotanical studies have been carried out in order to identify the traditional remedies used for various ailments, but few studies have focused on gastrointestinal pathologies. In this context, an ethnobotanical survey was carried out to identify treatments used for diarrhea and ciguatera, inventory the plants used, better understand the local representation of these remedies, and provide efficacy and safety data on these uses. Materials and methods From February to April 2021, a semi-structured survey was conducted on six islands in French Polynesia, including one island in the Windward Islands archipelago (Tahiti), three islands in the Marquesas archipelago (Hiva Oa, Nuku Hiva, Tahuata), and two islands in the Leeward Islands archipelago (Raiatea, Tahaa). A total of 133 people was interviewed including 34 specialists (of which 29 experts in herbalism). Results These people mentioned the use of 27 plants for the treatment of diarrhea, and 24 for the treatment of ciguatera. Citrus aurantiifolia, Psidium guajava and Cordyline fruticosa were the three most cited plant species used for treating diarrhea, while Cocos nucifera, Punica granatum and Barringtonia asiatica were the most cited for ciguatera. A large majority of plants are widespread and introduced plants, which is congruent with the history of Polynesian people. 27. In Vitro Hair Dermal Papilla Cells Induction by Fagraea berteroana, a Tree of the Marquesan Cosmetopoeia (French Polynesia)

Abstract

Fagraea berteroana is a tree used in traditional medicine in various islands of the South Pacific. Here, we studied its hair growth-inducing properties as suggested by one of its Marquesan ethno-uses in haircare. The ethyl acetate extract of the fruits of F. berteroana (FEAE) and four resulting fractions (FEAE-F0, FEAE-F1, FEAE-F2, and FEAE-F3) were tested on hair follicle dermal papilla cells to determine their cell proliferative activity. Furthermore, RT-qPCR analysis enabled gene modulation analysis, while immunostaining of the β-catenin protein was used to follow protein regulation. We found that the plant extracts induced a controlled, dose-dependent cell proliferation. FEAE-F0 simultaneously down-regulated Bone Morphogenetic Protein 2 (BMP2) mRNA expression and upregulated Cyclin-D1 (CCND1) gene expression, which suggests an involvement in the regulation of the Wnt and Transforming Growth Factor beta (TGFβ) pathways that control the hair cycle. FEAE-F0 exhibited a 1.34-fold increase of nuclear β-catenin protein. This is indicative of an active hair growth state. Thus, we conclude that FEAE-F0 could be an innovative candidate in hair care, which opens interesting leads to promote the Marquesan cosmetopoeia.

Abstract

Tamanu oil from Calophyllum inophyllum L. has long been used in traditional medicine. Ethanol extraction was found the best strategy for recovering bioactive compounds from the resin part of Tamanu oil, yielding two neutral and acidic resins fractions with high phenolics, flavonoids and pyranocoumarins concentrations. A further cascade of LPLC/HPLC separations of neutral and acidic resin fractions allowed identifying fifteen metabolites, and among them, calanolide D and 12-oxocalanolide A (both in neutral fraction) were first identified from a natural source. All these extracts, subfractions and isolated metabolites demonstrated increased free radical scavenging, antioxidant, anti-inflammatory, antimicrobial and antimycobacterial activity compared to Tamanu oil and its de-resinated lipid phase. Overall, these results could promote resinous ethanol-soluble Tamanu oil extracts as a useful multifaceted and renewable medicinal resource

Abstract

The first chemotaxonomic study based on volatile components of Porella viridissima (Mitt.) Grolle is reported. The GC-MS analysis of ether extract was performed; ten santalane and five pinguisane-type sesquiterpenes were identified together with perrottetianal A as major diterpene. Most of detected santalane-type sesquiterpenes are reported for the first time in liverwort. P. viridissima was found to belong to the chemotype III (pinguisane/sacculatane) and shared chemical similarities with P. navicularis. Perrotettianal A was isolated and has shown strong cytotoxicity against ovarian cancer.

Abstract

The aim of our work is to develop a functionalized patch for wound dressing application using natural and endemic plant extract from Ethiopia. Acokanthera schimperi (Apocynaceae) "Yemerz Enchet" is one an Ethiopian based plant in which its extract is used for treatment of chronic wounds by the traditional doctors.[1] Therefore, it has been selected to be used in this study. As it is well known, electrospun nanofibers patches are an ideal candidate which satisfies the properties of efficient wound healing applications such as mimicking extracellular matrix structure. Moreover, an efficient bacterial barrier, an appropriate water vapor transmission rate and provision of adequate gaseous exchange is also provided by such kind of patches.[2] The aim of this presentation is unrolled along the different steps that have been followed to functionalized the nano-filament patches with the selected plant "Yemerz Enchet". The challenge to incorporate "Yemerz Enchet" extract into the polymeric nanoweb is also discussed. The polymer that has been chosen, regarding the literature recommendation is the PCL. The production parameters have been selected and adapted from literature issues thank to different trials. Uniform and bead free nanofibrous mats loaded with 1,2 and 3% extract were produced. The fabricated nanofibrous is characterized using SEM and FTIR in order to characterize the modification that occurs during the electro-spun process. FTIR analysis indicates that the chemical nature of extract and polymer polymer are not affected by the electrospinning process. The biologic test, that is currently running, will characterize the anti-bacterial properties of the selected extracts and of the final products.

Abstract

Hair loss is becoming increasingly prevalent as dietary and living habits change. The search for natural products to limit hair loss has led to tapping into traditional cosmetic knowledge. We studied three plants of the Polynesian cosmetopoeia, Bidens pilosa, Calophyllum inophyllum and Fagraea berteroana, to determine their ability to promote hair growth. Their chemical content was characterized by liquid chromatography coupled to mass spectrometry (LC-MS). Their proliferative activity on dermal papilla cells (DPCs) was assessed via MTT assay and molecular targets were evaluated by RT-qPCR analysis of seven factors involved in the modulation of the hair cycle, CCND1, LEF1, DKK1, WNT5A PPARD, TGFΒ1, PPARD and RSPO2. Our results show that our extracts significantly increased proliferation of dermal papilla cells. Furthermore, LC-MS/MS analysis revealed a diversity of molecules, flavonoids, iridoids and organic acids, some known for hair-inducing properties. Finally, specific extracts and fractions of all three plants either upregulated CCND1, LEF1 and PPARD involved in stimulating hair follicle proliferation and/or lowered the gene expression levels of hair growth inhibiting factors, DKK1 and TGFB1. Our findings suggest that extracts from B. pilosa, C. inophyllum and F. berteroana are interesting candidates to stimulate hair growth
