L’analisi scientifica è da un po’ di tempo entrata nel mondo dei beni culturali, non sono più solo gli archeologi e gli storici dell’arte a occuparsi dello studio dei reperti archeologici e storici, ma anche gli scienziati sono entrati a gamba tesa portando le loro conoscenze al servizio di questa vasta gamma di materiali. Lo studio di reperti botanici e zoologici, le competenze chimiche e mineralogiche, lo studio di fenomeni fisici, ha permesso di porre e rispondere a nuove domande, colmando così lacune sulla storia dell’umanità. Reti commerciali, migrazioni, tecniche produttive, molte sono state le scoperte avvenute grazie anche all’intervento scientifico, oltre a fornire strumenti utili al restauro e alla conservazione dei reperti. Nei miei tre anni di dottorato mi sono occupato dell’applicazione dell’analisi in Fluorescenza a Raggi X (XRF) per analizzare campioni metallici e ceramici. Questa tecnica ha il vantaggio di poter essere applicata in maniera non invasiva e non distruttiva su un reperto per ottenere informazioni sulla sua composizione elementare. In particolare, il nostro obiettivo è quello di ottenere informazioni sui diversi strati che compongono un campione. Spesso, infatti, i manufatti presentano una struttura stratificata causata dal passaggio degli anni o dalla natura stessa dell’oggetto, che presenza una serie di decorazioni superficiali. L’impiego di tecniche non invasive permetterebbe quindi di ottenere informazioni più dettagliate anche su campioni al momento inaccessibili, in quanto troppo fragili o troppo preziosi. Lo studio si è quindi avvalso di tecniche a scansione angolare, per cui il segnale di fluorescenza caratteristica dipende sia dalla posizione dell’analita all’interno del campione, sia dalla struttura e composizione dello stesso, sia dalla geometria di analisi. Per valutare l’applicabilità delle tecniche sono stati analizzati tre casi diversi: campioni metallici preparati ad hoc in laboratorio, un campione ceramico, e un campione di lustro. Per i primi due campioni è stata applicata l’XRF a Risoluzione Angolare (AR-XRF) per cui il campione è stato ruotato con uno step angolare inferiore a un grado, e misurato ad ogni step. Nel caso dei campioni metallici i profili misurati sono stati confrontati direttamente con i profili calcolati usando il metodo dei Parametri Fondamentali. Nel caso del campione ceramico invece, sono stati confrontati i rapporti dei profili, in quanto la geometria del campione era piuttosto complessa. Nello studio sui campioni metallici, composti da una doratura depositata su una lamina di rame, è stato possibile calcolare lo spessore dello strato superficiale. Nel caso del campione di Majolica sono invece state studiate le varie decorazioni, valutando così anche diversi limiti della tecnica, in particolar modo nel caso in cui la composizione della decorazione sia simile alla composizione dello strato sottostante, o nel caso in cui avvengano processi di diffusione a lunghe distanze. In casi in cui invece lo spessore della decorazione era limitato e ben separato dallo strato sottostante, è stato possibile ottenere informazioni sua sulla composizione della decorazione che sul suo spessore. Infine, lo studio delle ceramiche lustrate è stato effettuato presso la linea XRF del Sincrotrone di Elettra, impiegando tecniche di analisi in radenza. In questo caso stato possibile solo evidenziare come il nano-strato di nano-particelle di argento, caratteristico di questa tipologia di campioni, sia effettivamente distinguibile applicando queste tecniche, in quanto il profilo di fluorescenza di tale elemento è molto diverso dagli altri. Rimangono comunque dei problemi, legati soprattutto all’analisi dati e all’allineamento che vanno ancora risolti, rendendo necessari ulteriori studi a riguardo. In the last decades scientific analysis has been deeply employed in the world of cultural heritage, thus, archaeologists and art historians are no more the sole front line workers of this field. Scientists, and science, have joined the team, giving new inputs and tools for the study of historical and archaeological samples, allowing to explore new paths and receive new answers, collecting information otherwise inaccessible on human history and culture. New discoveries have been made on the trade networks, migrations and on the technologies employed; besides, science also gave precious inputs on conservation and restoring procedures, allowing to better preserve fragile and sensitive artifacts. In my three years as a PhD student, I worked on the application of X-Ray Fluorescence (XRF) analysis to analyze ceramic and metal samples. XRF is a non-invasive technique that retrieves the elemental composition of a sample. In particular the aim of my PhD project is to obtain information on the layered structure of an unknown sample, distinguishing and characterizing the different layers. Indeed, artifacts usually concerning the field of Cultural Heritage present a layered structure; sometimes it is due to the presence of alteration layers, other times, instead, the objects are made of different layers from the principle, for example in the case of a glazed ceramic or of a painting. The possibility to get this information in a non-invasive way will give the possibility to analyze objects that are nowadays unattainable, because they cannot be sampled. My project has, thus, focused on the analysis of three kinds of samples employing angular dependent techniques (Angle Resolved-XRF, Grazing Emission-XRF}, Grazing Incidence-XRF}); indeed, the fluorescence signal of an analyte depends on its position inside the sample, on the sample composition and on the geometry of analysis. The chosen specimens allowed to verify the feasibility of this analytical method in an increasing complexity: a gilded laboratory-made sample, a ceramic Majolica sherd, and an Italian renaissance lustered fragment. The first two samples have been analyzed through AR-XRF where the measure is performed while tilting the sample, one spectrum is collected for each tilting angle. In the case of the gilded sample the measured profiles have been compared directly with the calculated profiles employing the Fundamental Parameters method. For the ceramic Majolica sample, instead, we studied the ratio of the profiles, as the sample surface is not flat. In the study of the metallic samples, made of gilded copper plate, we could infer the thickness of the top-layer. While in the case of the Majolica sample, we studied the different decorations, evaluating the limits of the technique, in particular in the case that the top-layer composition is similar to the underling layer, or in the case of a long-range diffusion. Instead, in the case of two well-separate layers we could retrieve information on both the composition and the thickness of the layer. Finally, the study of the lustered ceramic has been carried out at the XRF beamline of the Elettra Synchrotron of Trieste, employing grazing techniques. In this case we could only highlight and distinguish the presence of the silver nanoparticles in the luster nanolayer, which is the peculiar feature of this kind of artifacts. However, there are still many questions left, especially concerning the data analysis and the alignment of the sample, which requires more investigations.

Il volume costituisce un parziale ma dettagliato resoconto dell’attività di visiting professorship tenuta presso il Dipartimento di Culture del Progetto dell’Università Iuav di Venezia dall’architetto statunitense Paul O. Robinson durante l’anno accademico 2014–2015. Professore presso la Fakulteta za Arhitekturo v Ljubljani, Robinson ha al suo attivo un’intensa attività sia progettuale che artistica, accompagnata da una profonda riflessione teorica sugli strumenti critici e figurativi del progettista e dell’artista contemporaneo. Durante il suo soggiorno veneziano, Robinson ha tenuto, presso la Sala Gino Valle dello Iuav, una mostra dei suoi lavori (25 maggio – 5 giugno 2015), dal suggestivo titolo “The Cella Was Empty” (La cella era vuota), e un workshop (16 ottobre – 18 dicembre 2015), intitolato programmaticamente “Forms of Absence – Radiographs | Paintings | Reliquaries”. L’attività prevista all’interno del workshop era costituita da una serie di lezioni ex–cathedra e soprattutto da laboratori in cui inediti approcci teorici e applicativi hanno permesso agli studenti di apprendere un processo non convenzionale di lettura e interpretazione critica di oggetti architettonici o artefatti artistici esistenti, attraverso nuove pratiche espressive e proiettive.

InAs nanowires have been grown by molecular beam epitaxy using Mn as growth catalyst. Nanowires (NWs) have been obtained on SiO2 and oxidized GaAs for growth temperatures in the range of 370-410 degrees C. The growth temperature is found to be the same as that of the growth of Au-catalyzed InAs wires. These results suggest that the InAs NWs grow with the Mn nanoparticle in the solid phase and allow some comparison with the existing models for Au-catalyzed nanowires. The morphology and the lattice structure were investigated by electron microscopy techniques. The lattice of the wire body is found to be mainly wurtzite InAs. Mn K-edge x-ray absorption fine structure was used to determine the local environment of the Mn atoms: The authors found that most of the Mn atoms are found in a hexagonal MnAs phase. The present work of thesis is focused on application of X-ray K-edge technique to paintings. This technique allows one to achieve a topographic map of a pigment on the whole surface of the painting. The digital acquisition of radiographic images by using monochromatic X-ray beams allows to take advantage of the sharp rise of X-ray absorption coefficient of the elements, the K-edge discontinuity. Working at different energies, bracketing the K-edge peak, allows recognition of the target element. The K-edge radiography facility installed at Larix Laboratory, at Department of Physics in Ferrara, consists of a quasi-monochromatic X-ray beam obtained via Bragg diffraction on a mosaic crystal from standard X-ray source. In this work the K-edge technique and the facility used are illustrated. A characterization of the monochromatic beams in the energy range 7-40 KeV range and the elemental mapping on pictorial layers obtained are presented. At the end, a transportable facility for digital radiography is presented and some radiographic analysis of works of art performed are shown.

Bacterial serine/threonine kinases modulate a wide number of cellular processes. The serine/threonine kinase PrkC from the human pathogen Staphylococcus aureus was also shown to induce germination of Bacillus subtilis spores, in response to cell wall muropeptides. The presence of muropeptides in the bacterial extracellular milieu is a strong signal that the growing conditions are promising. In the present paper, we report the X-ray structure of the entire extracellular region of PrkC from S. aureus. This structure reveals that the extracellular region of PrkC, EC-PrkC, is a linear modular structure composed of three PASTA (penicillin binding-associated and serine/threonine kinase-associated) domains and an unpredicted C-terminal domain, which presents the typical features of adhesive proteins. Using several solution techniques, we also found that EC-PrkC shows no tendency to dimerize even in the presence of high concentrations of muropeptides. X-ray structural results obtained in the present study provide molecular clues into the mechanism of muropeptide-induced PrkC activation.

The bonding environment of oxygen implanted in GaN is studied using near edge X-ray absorption fine structure spectroscopy at the O-K-edge. The 70 keV oxygen ions form a 200 nm-thick subsurface layer that is highly defective or amorphous depending on the implantation fluence which ranges from 1 x 10(15) cm-2 to 1 x 10(17) cm(-2). The information depth of the fluorescence photons varies from 50 to 63 nm, depending on the angle of incidence. The spectra are simulated using the FEFF8 code and assuming various models, e.g., O interstitial, O substitutional in N sites, Ga and N vacancies, and various polymorphs of Ga2O3. The lattice disorder is modelled by displacing atoms from their equilibrium positions by adding to their Cartesian coordinates random numbers that belong to normal distributions. The simulation results reveal that at the low fluence limit, the O atoms occupy interstitial sites preferentially in the empty channels aligned to the c-axis and in the atomic planes containing the Ga atoms. When the fluence is equal to 1 x 10(16) cm(-2) the O atoms substitute N while at 1 x 10(17) cm(-2) they form mixed GaO(x)N(y) phases with the N/O ratio decreasing with increasing depth, i.e., as we approach the peak of the implanted O profile.

The bonding environment of oxygen implanted in GaN is studied using Near Edge X-ray Absorption Fine Structure (NEXAFS) spectroscopy. The implantation of 70 keV O ions in GaN results in the formation of a 200 nm – thick subsurface layer that is highly defective or amorphous depending on the implantation fluence which ranges from 1 × 1015 to 1 × 1017 cm−2. The NEXAFS spectra are simulated using the FEFF8 code assuming models that account for the formation of point defects (various configurations of O interstitial and O substitutional in N and Ga sites) as well as chemical effects such as the formation of various polymorphs of Ga oxides and oxynitrides. The implantation-induced lattice disorder is modeled by displacing atoms from their equilibrium positions by adding to their Cartesian coordinates random numbers that belong to normal distributions. The simulations reveal that when the fluence is 1 × 1015 cm−2, the O implants occupy interstitial sites preferentially in the empty channels aligned parallel to the c-axis in the plane that contains the Ga atoms and/or in the columns that consist of Ga and N atoms along the c-axis. When the fluence is equal to 1 × 1016 cm−2 the O ions substitute for N while at 1 × 1017 cm−2 they participate in the formation of mixed GaOxNy phases.

In recent years, X-ray CT has become an important tool for investigating all kinds of materials. Due to its non-destructive nature, it is especially suited to investigate rare or expensive samples. CT has been recently introduced in the field of Cultural Heritage diagnostics, where it can be used for the investigation of different works of art, as it preserves the integrity of the object and gives morphological and physical information on its inner structure. This paper wants to point out a methodological approach on the use of the X-ray CT technique to study items belonging to cultural heritages with the aim to obtain information related to their preservation state and therefore, for determining adequate maintenance and restoration procedures. Important applications are for example the study of porosity and of pore size distribution and connectivity of different porous materials and the study of the kinetics of capillary fluid absorption in sedimentary rocks. Other applications are relevant to the possibility to investigate in a non invasive way the presence of defects or fractures inside an object and not last in order of importance, the possibility to study different typologies of woods or waterlogged woods and the presence of microbial deterioration. In this paper are reported the results of some case studies, obtained through the combined use of CT systems with different resolutions. Other expected future developments will be addressed to the integration of CT data with results of compatible non-destructive techniques.

L’interesse principale nell’applicazione delle tecniche di diffrazione di raggi X nell’ambito dei beni culturali risiede nella identificazione qualitativa delle fasi cristalline.Viene data una breve presentazione della teoria della diffrazione dei raggi X, delle tecniche di diffrazione ed in particolare del metodo delle polveri e dell’uso di banche dati cristallografiche. Vengono presentati alcuni esempi applicativi su pitture e manufatti in porcellana, metallo e terracotta.

Per saperne di più riguardo agli effetti biologici delle radiazioni ionizzanti, abbiamo esaminato l’apoptosi indotta dai raggi X nei linfociti umani di sangue periferico (HPBL). I linfociti sono stati esposti ai raggi X in presenza o in assenza di Pifithrin-α (PFT-α), un inibitore reversibile dell’attivita di trans-attivazione trascrizionale della proteina p53, nella fase G0 e nella fase G2 del ciclo cellulare. Gli esprimenti condotti sui linfociti nella fase G2 del ciclo cellulare dimostrano che la PFT-α non altera la cinetica del ciclo cellulare sia nelle cellule irradiate che nei campioni di controllo e suggeriscono l’esistenza di una via apoptotica p53-indipendente nei linfociti umani, in tale fase del ciclo. Per testare la capacità degli HPBL di riparare il danno primario al DNA in presenza o in assenza di PFT-α, è stata utilizzata la metodica del Comet-assay ai tempi di recupero successivi all’irradiazione nella fase G0 del ciclo cellulare (da 0h a 4h). Nei linfociti co-trattati con raggi X e PFT-a abbiamo riscontrato una cinetica di riparazione ritardata rispetto ai linfociti esposti ai soli raggi X. Dall’analisi parallela della morfologia nucleare apoptotica e delle aberrazioni cromosomiche abbiamo riscontrato, una correlazione, tempo dipendente, tra la riduzione di cellule apoptotiche e l’incremento sia nella frequenza dei cromosomi dicentrici che nella quantità di cellule aberrate. I dati ottenuti dagli studi condotti nella fase G0 del ciclo cellulare dimostrano che negli HPBL l’inibizione dell’apoptosi promuove l’aumento delle aberrazioni cromosomiche. Tuttavia la PFT-a oltre al suo effetto diretto sull’apoptosi potrebbe influenzare anche i processi di riparazione del DNA provocando un aumento degli eventi di miss-riparazione del danno indotto in seguito all’esposizione ai raggi X come risulta dall’analisi della cinetica di riparazione studiata con il Comet-assay. To learn more about the biological effects of radiations, we examined radiation-induced apoptosis in response to x-ray irradiations in human peripheral blood lymphocytes (HPBL). HPBL were exposed to x-rays in the presence or absence of Pifithrin-α (PFT-α), a transcriptional trans-activation reversible inhibitor for p53 protein, both in the G0 and in the G2 phase of cell cycle. Experiments with HPBL in the G2 phase demonstrate that PFT-α does not affect cell cycle kinetics both in irradiated cells and in control samples and indicate the existence of a p53-independent apoptotic pathway in human lymphocytes in this phase of the cell cycle. Alkaline Comet-assay was performed at early recovery times (0h to 4h) after irradiation in the G0 phase, to test HPBL DNA repair capability in the presence or absence of PFT-α after irradiation. A delayed repair kinetic was observed in lymphocytes co-treated with x-rays and PFT-α respect to only x-ray-irradiated lymphocytes. The parallel analysis of both nuclear morphology and chromosomal aberrations indicated a time-dependent correlation between decrease in the yield of apoptotic cells and increase in the frequency of both dicentrics and aberrant cells. The overall data demonstrate that the inhibition of p53-mediated apoptosis promotes the enhancement of chromosomal aberrations in G0 x-ray-irradiated HPBL. PFT-α then, beside its direct effect on apoptosis could have an effect also on DNA repair processes increasing the mis-repair of X-rays-induced chromosomal damage. Structure, cation distribution, Mn oxidation states, and conductivity behavior of the Mn-substituted (up to 30% of Ti ions) Li4Ti5O12 have been investigated by the combined use of X-ray powder diffraction, electron paramagnetic resonance (EPR), 7Li MAS NMR, and impedance spectroscopy techniques. The spinel structure of the lithium titanate is preserved and the lattice parameter decreases with increasing the Mn content. Mn2+ ions progressively occupy the tetrahedral site up to an approximately constant value reached for 10% Mn-substituted samples. Mn3+ ions occupy both octahedral and tetrahedral sites, with a constant value on the tetrahedral one, independent of the total Mn amount; Mn4+ ions are not detected. The Mn2+ paramagnetic ions give rise to a through-space interaction with Li+ ions of both cationic sites, as evaluated by the area, proportional to the Mn2+ ions content, of a peak at ∼8 ppm observed in the 7Li NMR spectra for the substituted samples. The obtained cation distribution and the Mn valence states satisfactorily explain the decrease of conductivity observed in the Mn-doped samples with respect to the pure Li4Ti5O12

La presente invenzione concerne un’apparecchiatura per la produzione di fasci di elettroni e di raggi X per trattamenti di radio terapia interstiziale ed intra operatoria più comunemente denominata IORT (Intra Operative Radio Therapy). Più particolarmente l'invenzione concerne un'apparecchiatura IORT che utilizza una macchina tipo Plasma Focus, associata ad una opportuna guida per elettroni da convogliare direttamente in prossimità del tessuto da trattare tramite irraggiamento elettronico o fotonico a bassa energia ed altissimo rateo di dose (dose rate).

The bright low-mass X-ray binary Cyg X-2 was in the field of view of the IBIS telescope during the early Cygnus region observations, executed during the INTEGRAL Performance Verification Phase. The data presented are spanning about one week and cover the rising edge of one of the two peaks of the ~82 day cycle of the Cyg X-2 light curve. The IBIS data in the energy range 20-40 keV exhibit flux variation correlated with the RXTE/ASM light curve. Two different main exposures, separated by ~5 days are found to be characterized by sensibly different spectra, with significant softening and higher X-ray luminosity in the second part, coincident with the long-term cycle peak. At high energies, both measured spectra are very steep. The ratio of the 30-45 keV and 20-30 keV detected fluxes is ~0.30, against a value of 0.95 expected for a Crab-like spectrum. No positive detection exists for E>=45 keV, with a flux upper limit (5sigma ) of ~ 1.4*E-10 erg cm-2 s-1 in the 45-100 keV band.

Il tre marzo 1997 alle otto di mattina sotto gli occhi esterefatti dei rari passanti presenti a quell’ora, crolla una parte del portico esterno della Santissima Annunziata di Bologna, un grande complesso conventuale situato subito fuori la Porta S.Mamolo di Bologna. Di proprietà demaniale, era in uso in parte alla parrocchia ed in parte all’amministrazione militare. Prima del progetto di restauro generale e del progetto di ricostruzione della parte crollata redatto dall'arch. N. santopuoli vengono svolti studi e indagini sulle strutture e sulla composizione dei materiali lapidei per verificarne la natura e la resistenza meccanica. Sulla base degli studi eseguiti è stato poi redatto un progetto generale di restauro che ha interessato la totalità del portico: dalle lunette dipinte, al colonnato, dagli elementi decorativi agli intonaci e le coperture. Nella parte iniziale dell’articolo viene introdotto la storia del porticato esterno, risalente al 1488 e attribuito alla Scuola del Formigine e vengono descritte le lunette con pitture murali policrome collocate all’interno del porticato e risalenti al 1619 e raffiguranti la storia della Vergine, attribuite alla Scuola del Carracci e di Giacomo Lippi, detto Giacomone da Buorlo. Dopo la descrizione dei materiali e delle tecniche costruttive che caratterizzano le volte, il colonnato e la copertura vengono illustrate le cause del crollo per arrivare alle motivazioni del progetto di ricostruzione delle parti crollate così pure il progetto di consolidamento statico.

The experimental determination of the scaling of the superconducting critical temperature ${(T}_{c})$ vs the Fermi temperature ${(T}_{f})$ of the holes in the boron \ensuremath{\sigma} subband is presented. The Fermi level has been tuned near the ``shape resonance,'' i.e., the two- to three-dimensional crossover of the Fermi surface of the boron \ensuremath{\sigma} subband by changing the Al/Mg content in ${\mathrm{Al}}_{1\ensuremath{-}x}{\mathrm{Mg}}_{x}{\mathrm{B}}_{2}.$ The product ${k}_{f}{\ensuremath{\xi}}_{0}$ of the Fermi wave vector ${(k}_{f})$ times the superconducting Pippard coherence length $({\ensuremath{\xi}}_{0}),$ that is a measure of the pairing strength, remains constant, ${k}_{f}{\ensuremath{\xi}}_{0}=90$ for $xg0.66.$ This high-${T}_{c}$ phase occurs in the boron superlattice under a tensile microstrain in the range $3%l\ensuremath{\varepsilon}l6%.$
