Abstract The Brazilian tick fauna currently comprises 77 valid species categorized into two families: Ixodidae (53 species) and Argasidae (24 species). In the state of Alagoas, only six Ixodid ticks have been reported to date, with no previous reports of ticks in the Argasidae family. Here, we assessed 33 White-eared Opossum (Didelphis albiventris Lund, 1840) rescued in the metropolitan region of Maceió and referred to the Wild Animal Screening Center (Cetas) in the city. Upon arrival, the animals were examined for ectoparasites within 24 hours. In total, 10/33 (30%) opossums were found to be infested by 26 larvae of the argasid tick Ornithodoros mimon Kohls, Clifford & Jones, 1969. Morphological identification of ticks was corroborated by generating partial sequences of the mitochondrial 16S rRNA gene from three tick specimens. This study marks the first report of an argasid tick in the state of Alagoas. Future studies should investigate whether populations of both O. mimon ticks and their host, D. albiventris, in the state of Alagoas carry potential zoonotic agents capable of causing tick-borne diseases.

Background and Aim: Studies on ticks of public health concern in equine husbandry are scarce in the Northeastern region of Brazil. This study aimed to investigate the presence of ticks on horses in the State of Alagoas, which is classified as a silent area for human rickettsiosis. Materials and Methods: Ticks infesting horses were collected using anatomical tweezers or a commercial hook and kept in ethanol-labeled tubes for taxonomic identification. Results: A total of 2,238 ticks were found. Ticks were identified as 2,215 (98.89%, 95% CI: 98.41–99.28) Dermacentor nitens, 19 (0.98%, 95% CI: 0.05–1.38) Amblyomma sculptum, and 4 (0.18%; 95% CI: 0.007–0.46) Rhipicephalus microplus. Conclusion: This is the first study to report A. sculptum and D. nitens in the State of Alagoas. The presence of A. sculptum should draw the attention of public health managers once Alagoas State is considered a silent area for rickettsial diseases, which means the absence of local surveillance programs for these pathogens.

Leishmania amazonensis causes a clinical form called diffuse cutaneous leishmaniasis (DCL) with challenges to treatment, like low efficiency and drug toxicity. Therefore, it is necessary to investigate new therapies using less toxic leishmanicidal compounds, such as flavonoids like naringenin, and their combination with conventional drugs, such as miltefosine. Antileishmanial dose/response activity, isobologram, calculation of dose reduction index (DRI), and fractional inhibitory concentration index (FICI) tests were performed on in vitro assays using reference promastigote forms of L. amazonensis (IFLA/BR/67/PH8) to assess the combinatorial effect between naringenin and miltefosine. The in vitro results of isobologram, DRI, and FICI calculations showed that the combination of the compounds had an additive effect and was able to reduce the half maximal inhibitory concentration (IC50) of miltefosine in the promastigote forms of the parasite compared to the treatment of the drug alone. This study demonstrated in vitro the viability of a combination action of the flavonoid with the treatment with miltefosine, opening space for further investigations on the association of natural compounds with the drugs used for the treatment of L. amazonensis.

Veterinarians are commonly exposed to occupational stressors, including excessive workload and financial constraints. These stressors can lead to psychological distress, which typically results in mental health disorders such as depression, anxiety, and burnout and can even culminate in suicide attempts or suicide deaths. Risk factors associated with poor mental health and high rates of suicide in veterinary practitioners include continuous exposure to challenging scenarios, such as interpersonal conflicts, performing euthanasia, and easy access to lethal means of suicide, such as opioids and anesthetics. The previous studies highlight the urgent need for a better understanding of predisposing factors, mental health-related improvements in the professional environment, and the subsequent establishment of primary mental health-related care policies. Effective ways to promote mental health and prevent suicide may include social support, resilience, developing coping skills, promoting a healthy work environment, and discouraging perfectionist behaviors. This review aimed to summarize findings in studies that have investigated mental health and suicide in veterinarians and veterinary students and highlight measures that could be implemented as options for mental health promotion and suicide prevention.

Abstract Glossina species are known to transmit African Trypanosomiasis, one of the most important infectious diseases for both livestock and humans in sub-Saharan Africa. Therefore, the aim of this study was to characterize trapped Glossina spp. from The Gambia using morphological and molecular techniques in relation to the vegetation cover types. A line transect survey was carried out in all the administrative regions of The Gambia. Tsetse fly trapping was carried out for 14 days during each season using line transect. A total of 220 Glossina spp. specimens (117 F and 103 M) were captured, and DNA was extracted from the legs of 100 randomly selected Glossina spp. Further, DNA samples were tested by a conventional PCR assay. A total of 135/220 (61%; 95% CI: 54.6-67.8%) and 85/220 (39%; 95% CI: 32.2-45.4%) flies were identified as Glossina morsitans submorsitans and Glossina palpalis gambiensis, respectively, with most caught during wet season (53.6%) and more females (53.2%) than males. Results of the morphological identification agreed with those of molecular identification. The type of vegetation cover significantly influenced the caught of tsetse flies. Animals and humans at the various trapping sites are at risk of being bitten by tsetse flies.

Brazil is composed of large ecosystems with vast biodiversity in fauna and flora. Agriculture and livestock farming provides an interaction between humans and domestic and wild animals, which favors the dissemination of infectious and parasitic agents. Wild rodents present a wide ecological diversity; therefore, it is necessary to know which species may be carriers of pathogens. This study aims to identify endo and ectoparasites, and hemoparasites in Spix's yellow-toothed cavy (Galea spixii Wangler, 1831), and determine the hematological parameters of this species. The study was carried out with 16 adult specimens from both sexes, captured in the rural areas of Remigio and Areia, Paraiba State, northeastern Brazil. All Spix's yellow-toothed cavies presented ectoparasites, Gliricola quadrisetosa, Gyropus ovalis, Laelaps sp. and Chirodiscoides caviae, and endoparasites identified by the presence of Nematode eggs, Cestode eggs, and coccidian oocysts in the stool samples. To the best of our knowledge, this is the first report of parasitism by Gyropus ovalis, Laelaps sp. and Chirodiscoides cavia in Spix's yellow-toothed cavy. Animals tested negative by PCR for hemotropic Mycoplasma sp., Babesia sp., Ehrlichia sp., and Bartonella sp. For the first time, the hematological profile of these animals was determined. These results can be the basis for future research with the species, which can be used as a sample animal in experiments in clinical settings as an epidemiological guide since this species is routinely used by impoverished populations in the Northeastern region of Brazil as pets, as well as a source of protein.

Bartonella is an emerging group of facultative intracellular bacteria causing circulatory and systemic disorders. Hosts for Bartonella are mostly mammals, specifically rodents, having a growing number of Bartonella species related to their infection. Capybaras (Hydrochoerus hydrochaeris) are abundant native rodents of Brazil, commonly found in urban parks. In the present study, we aimed to perform molecular screening of capybaras for Bartonella spp. Blood samples were collected from 17 free-ranging animals captured in Paraná State, Southern Brazil. None of the collected samples tested positive for the Bartonella-nuoG gene by quantitative polymerase chain reaction (qPCR), although all of them successfully amplified the mammal endogenous glyceraldehyde-3-phosphate dehydrogenase (gapdh) gene. Additionally, all animals were infested exclusively by Amblyomma dubitatum ticks at the time of sampling. This study was part of an active surveillance program, which is critical for monitoring animal health status, particularly in capybaras.

Parasitic infections are important concern to the Wildlife Conservation Biology, particularly in endangered species. Herein, we report a parasitism by Dipetalonema gracile Rudolphi, 1809 (Spirurida, Filarioidea, Onchocercidae), in the peritoneal cavity of a captive Marcgrave's capuchin monkey (Sapajus flavius) that died at the Wild Animal Screening Center (CETAS) of the Brazilian Institute for the Environment and Renewable Natural Resources (IBAMA) in the municipality of Cabedelo, state of Paraíba, northeastern Brazil. The necropsy revealed two filarial worms D. gracile in the abdominal cavity. Exudates, thin fibrin layers and fibrous adhesions were also present in the mesentery and spleen capsule. The mesenteric, mandibular, and tracheobronchial lymph nodes were enlarged. Multiple small nodules were seen in the spleen parenchyma. Microscopic examination of the lymph nodes and spleen revealed markedly and diffuse inflammatory reaction, with edema, plasma cells, eosinophils, histiocytes, lymphocytes and rare multinuclear giant cells, with obliteration of the normal histological architecture of the organ. This is the first report of D. gracile parasitism in Marcgrave's capuchin monkeys, a critically endangered species. Studies of this nature significantly contribute to the knowledge of the parasitic fauna of endangered species, in addition to helping to formulate conservation strategies (in situ and ex situ) and records of new hosts and new areas of occurrence of parasites.

The order Chiroptera is the second largest group of mammals with bats being identified as reservoir of several viral zoonoses, although, little is known about their role in other groups of pathogens, including hemotropic Mycoplasma spp. To date, hemoplasma species have been found infecting several species of bats with high genetic diversity between 16S rRNA gene sequences. On this study, we aimed to identify the occurrence and characterize 16S and 23S rRNA genes of hemoplasma species in four bats species (Artibeus lituratus, Carollia perspicillata, Sturnira lilium and Sturnira tildae) from forest fragments in Paraná State, southern Brazil, using PCR-based assays. Spleen tissue samples were collected, DNA extracted and further screened by a pan hemoplasma PCR assay. All samples consistently amplified the mammal endogenous gapdh gene. One out of 15 (6.66%; 95% CI: 0.2-31%) bats tested positive for hemotropic Mycoplasma sp. by the PCR assay targeting the 16S rRNA gene. Sequencing of the 16S rRNA gene fragment from the hemoplasma-positive bat showed 99.14% identity with hemotropic Mycoplasma sp. detected in Sturnira parvidens from Belize. Sequencing of the 23S rRNA gene fragment from the hemoplasma-positive bat showed 86.17% identity with Candidatus Mycoplasma haemosphiggurus’ detected in orange spined hairy dwarf porcupines (Sphiggurus villosus) from Southern Brazil.

Abstract Hemoplasmas are epierythrocytic bacteria that infect mammals. ‘Candidatus Mycoplasma haemoalbiventris’ was detected in white-eared opossums (Didelphis albiventris) from southern and central-western Brazil. The present study aimed at: i) screening opossums for tick-borne (TBP) pathogens (Piroplasmida and Anaplasmataceae) and ii) detecting and characterizing hemoplasma species infecting opossums from Curitiba and Foz do Iguaçu cities in the Paraná State, southern Brazil. Thirty blood samples from white-eared opossums were evaluated by PCR assays. Animals were not infested by ectoparasites. The mammalian endogenous gapdh gene was consistently amplified in all samples. All opossums tested negative for Theileria/Babesia spp. and Ehrlichia/Anaplasma spp. by PCR based on 18S rRNA and 16S rRNA genes, respectively. A genus-specific PCR assay based on the 16S rRNA gene of hemoplasmas showed that three/13 (23.08%; CI 95%: 8.18-50.26%) opossums from Foz do Iguaçu were positive for hemotropic Mycoplasma sp. All opossums from Curitiba tested negative for hemoplasmas. Sequencing of both the 16S and 23S rRNA genes revealed that the animals were infected by ‘Ca. M. haemoalbiventris’. Although ‘Ca. M. haemoalbiventris’ is prevalent in opossums in Brazil, clinical signs associated with its infection and its putative vectors remain unknown.

The black-eared opossum (Didelphis aurita) is a South American synanthropic marsupial. The presence of opossums in domestic spaces is relevant in the One-Health context since they are hosts of pathogens and ectoparasites that may affect the health of domestic animals and humans. In this study, we aim to determine the occurrence of hemoplasmas and selected tick-borne pathogens in free-ranging black-eared opossums, along with their molecular characterization, hematological and biochemical evaluation and factors associated with infection, in the municipality of Viçosa, State of Minas Gerais, southeastern Brazil. Thirty black-eared opossums were trapped between March 2021 and June 2022. Ectoparasites were collected. Hematological and biochemical analyses were performed. DNA from EDTA-blood samples were analyzed by PCR and qPCR assays. By molecular analyses, ‘Candidatus Mycoplasma haemoalbiventris’ was the most prevalent hemoparasite (73.3%), followed by Hepatozoon sp. (22.2%). Significant differences were observed in the number of platelets, and in the concentration of protein and globulins in the animals infected by ‘Ca. M. haemoalbiventris’ when compared with the negative group. This is the first report of ‘Ca. M. haemoalbiventris’ infection in D. aurita.

This is the first report of Babesia vogeli molecular detection in dogs from the state of Acre, northern Brazil. This study aimed to perform the molecular detection of Babesia vogeli in dogs in the municipality of Rio Branco, Acre. Blood samples were collected from 47 dogs presenting with clinical signs comparable to hemoparasitosis. These were dogs which were attended in veterinary clinics from Rio Branco municipality, Acre. Physical examinations, packed cell volume (PCV) determination, platelet number estimation, hemoparasite investigation in the blood (collected from the pinna and peripheral blood), and polymerase chain reaction (PCR) for piroplasm based on the 18S rRNA gene, were performed. One dog (1/47, 2.1%; CI 95%: 0.1-11.3%) tested positive to Babesia vogeli in the polymerase chain reaction (PCR) assay for piroplasms and the resulting sequence showed 100% identity with Babesia vogeli isolates deposited in GenBank®. Co-infection with Ehrlichia spp. was also observed by direct examination (via blood smear). The clinical and hematological alterations observed in the positive animal were anorexia, dehydration, white mucous membranes, anemia and thrombocytopenia.

Health monitoring programs in animals used as experimental models are essential, since only disease-free subjects are considered suitable for research purposes. In laboratory-kept animals, hemoplasmas have been described as an important confounding variable. Different hemoplasma species have been detected infecting non-human primates (NHP) from Brazil. However, the occurrence of hemoplasma species in laboratory-kept NHP in Brazil has not-yet been assessed. Accordingly, this study aimed (i) to screen laboratory-kept rhesus monkeys for hemoplasmas, (ii) to verify if any of the hemoplasma-positive animals demonstrate hematological abnormalities, and (iii) to assess the genotype diversity of hemoplasma species in NHP from Brazil. Five out of eight (62.5%; 95% CI: 3.05–8.63) rhesus monkeys tested positive for hemotropic Mycoplasma spp. by PCR. Sequencing, phylogenetic, distance, and genotype diversity analyses of partial 16S rRNA gene demonstrate that rhesus monkeys were infected by ‘Candidatus Mycoplasma haematomacacae’ (formerly ‘Candidatus Mycoplasma haemomacaque’). Assessments of partial 16S rRNA diversity of hemoplasma species in NHP suggest that at least four genetically diverse groups may occur in Brazil. Although no hematological abnormalities were demonstrated in rhesus monkeys evaluated herein, future studies are needed to elucidate the influence of ‘Ca. M. haematomacacae’ as a confounding variable on research studies.

Abstract Although anemia has been historically linked to Haemonchus contortus infection, other infectious agents, such as hemotropic mycoplasmas and tick-borne disease pathogens, may also lead to anemic crisis in sheep. This study has aimed to investigate infections related to anemia in a sheep herd from Bandeirantes City, Paraná State, southern Brazil. Seven out of forty-two (16.6%; 95% CI: 8.32–30.6%) sheep were positive for hemoplasmas by a PCR targeting the 16S rRNA gene and all tested negative for A. marginale/A. ovis and Babesia/Theileria spp. by PCR based on msp4 and 18S rRNA genes, respectively. Two (4.7%; 95% CI: 1.32–15.79%) animals were infested with Rhipicephalus microplus ticks. Fecal egg counting was performed in 38 sheep and 24 (63.15%; 95% CI: 47.2–76.6%) presented > 500 eggs per gram. Phylogenetic analysis of partial sequences of the detected hemotropic Mycoplasma sp. 16S and 23S rRNA genes confirmed that the animals were infected with Mycoplasma ovis. Polymorphism analysis of partial 16S rRNA sequences showed three different genotypes of M. ovis infecting sheep assessed in the present study. Mycoplasma ovis and gastrointestinal nematodes occurs in sheep from the northern region of Paraná State.

Abstract Anaplasma marginale and piroplasm species are widespread among Brazilian cattle herds. Both of these tick-borne pathogens hamper livestock production and cause a significant economic impact. Although buffaloes have demonstrated a high level of adaptability, data on tick-borne pathogens are scarcely reported in Brazil. Thus, the aim of this study was to screen water buffaloes from the state of Maranhão for piroplasm and A. marginale occurrence using PCR assays. All samples were negative for A. marginale. One of the 287 (0.35%) water buffaloes tested was positive for Theileria sp. Sequencing of the 18S rDNA fragment (356 bp) showed that the Theileria sp. identified was closely related to the T. buffeli /orientalis group. Future studies on the clinical signs of infection and the main vector in this country are needed.

Abstract Objective Despite malaria epidemiology has been extensively studied in primates, few studies were conducted in ungulates. After half a century without descriptions of Plasmodium spp. in deer since its first identification, recent research has rediscovered Plasmodium on ungulates in Africa, Asia, North America and South America, including Central Brazil. Here, a captive herd was evaluated in southern Brazil using light microscopy and PCR. DNA samples were tested for fragment amplification of two Plasmodium spp. genes: mitochondrial cytochrome b and small subunit ribosomal RNA. Results All analyses were negative. However, the tests were performed on samples that were collected at a single time point, and parasitemia may fluctuate over the parasite’s life cycle. Thus, the possibility of occult infection cannot be ruled out. Despite the negative results of all of the methods applied, it cannot be categorically stated that these animals are free from Plasmodium sp. infection. Further monitoring and/or multiple sequential sampling may improve the success rate of detecting parasites. Moreover, although this survey of Plasmodium represents the first molecular study on ungulate malaria parasites from Southern Brazil, further analysis of samples from different ungulate species is important for characterizing the epidemiology of Plasmodium of these mammals in this region.

Neighborhood dogs may act as reservoirs and disseminators of vector-borne diseases in urban areas. Accordingly, the aim of this study was to ascertain the health status and the vector-borne pathogens infecting dogs living in public areas with high levels of human movement in the city of Curitiba, southern Brazil. Blood samples from 21 neighborhood dogs that were found in nine of 22 bus stations and two public parks were subjected to a complete blood cell (CBC) count, serum biochemical profiling, a commercial rapid ELISA test and a commercial real-time PCR panel of vector-borne diseases. The CBC count and serum biochemical profiling were within the normal range for dogs and only 1/21 (4.7%) of the dogs was seroreactive for Borrelia burgdorferi sensu stricto. The commercial real-time PCR panel showed that 7/21 (33.3%) of the dogs had Mycoplasma haemocanis infection, 9/21 (42.8%) had ‘Candidatus Mycoplasma haematoparvum’ and 4/21 (19.0%) had both. No statistical association between infected by the agents found here and abnormalities in physical examinations, laboratory tests or ectoparasite presence was found (p > 0.05). In conclusion, neighborhood dogs showed low prevalence of vector-borne diseases and satisfactory wellbeing, and dogs can be used as sentinels for disease exposure.

Porcine virus type 2 (PCV2) is an emergent virus found in commercial pig farms and may cause clinical or subclinical infection. Wild pigs such as collared peccary (Tayassu tajacu) and white-lipped peccary (Tayassu pecari) may also be infected by PCV2. Accordingly, the aim of this study was to molecularly identify PCV2 in whole blood samples of captive peccaries (16 collared and 6 white-lipped) by conventional and quantitative PCR (qPCR). Although the housekeeping gene (?-actin) DNA was successfully amplified, all 22 peccaries tested negative for PCV2 by both molecular methods. Previous studies have shown no antibodies against PCV2 in 49 free-range collared peccaries of Southern Brazil but PCV2 DNA was detected by PCR in tissue samples of 9/10 (90.0%) collared and 3/3 (100%) white-lipped free-ranging peccaries from Central and Northern Brazil. In conclusion, although PCV2 may be endemic in free ranging wild pigs of Central and Northern Brazil, lack of serological and molecular PCV2 evidence (in whole blood) of both captive and free-range wild pigs may indicate low risk of disease in Southern Brazil.

Theileria equi is the causative agent of worldwide piroplasmosis, an important tick-borne disease of equids associated to a lifetime carrier state of infected horses. Since rapid, accessible and reliable tests have been posted as a challenge for clinicians, the aim of the present study was to verify the agreement between an in-house immunofluorescence antibody assay (IFA) and a commercial competitive inhibition enzyme-linked immunosorbent assay (cELISA) in a population of 198 carthorses from Southern Brazil. The horse seroreactivity for T. equi revealed 152/198 (76.8%) positive samples by IFA and 155/198 (78.3%) by cELISA. Using cELISA as gold standard, IFA has shown a sensitivity of 91.6% (95% CI = 86.18-95.03%) and specificity of 76.7% (95% CI = 62.26-86.85%), with a substantial degree of agreement (k = 0.8445). In conclusion, the in-house IFA may be used as a screening test for diagnosis of equine piroplasmosis.

Equine infectious anemia (EIA) is an infectious viral disease caused by a Lentivirus, which affects equids worldwide. The disease has no currently treatment and euthanasia of infected animals is mandatory by the Brazilian Ministry of Agriculture, Livestock and Supply (MAPA) as basis for disease control. Carthorses are used to move daily throughout the cities with their owners to collect recycling materials. Considering the socio-economic importance of this group of horses, the aim of this study was to determine the infection rate of EIA virus in carthorses from urban areas of Curitiba and surroundings. The detection of anti-EIA virus antibodies was performed by the agar gel immunodiffusion test (AGID). One out of 97 (1.03%) horse was positive for EIA. Active surveillance programs are crucial for monitoring, prevention and control of infectious diseases, particularly in carthorses, which may act as disseminators of pathogens.

SUMMARY The aims of this study were to determine the prevalence of hemoplasmas in a rural Brazilian settlement's population of human beings, their dogs and horses, highly exposed to tick bites; to identify the tick species parasitizing dogs and horses, and analyze factors associated with their infection. Blood samples from 132 dogs, 16 horses and 100 humans were screened using a pan-hemoplasma SYBR green real-time PCR assay followed by a species-specific TaqMan real-time PCR. A total of 59/132 (44.7%) dog samples were positive for hemoplasmas (21 Mycoplasma haemocanisalone, 12 ' Candidatus Mycoplasma haematoparvum' alone and 21 both). Only 1/100 (1.0%) human sample was positive by qPCR SYBR green, with no successful amplification of 16S rRNA or 23 rRNA genes despite multiple attempts. All horse samples were negative. Dogs >1 year of age were more likely to be positive for hemoplasmas ( p= 0.0014). In conclusion, although canine hemoplasma infection was highly prevalent, cross-species hemoplasma transmission was not observed, and therefore may not frequently occur despite overexposure of agents and vectors.

Mycoplasma sp. are hemotropic bacteria of red blood cells and the causative agent of hemoplasmosis. Diagnosis of infection is reached by polymerase chain reaction (PCR) and real-time PCR (qPCR) of the 16S RNA gene of this bacteria. The aim of this study was to screen horses for Mycoplasma sp. Infection using a pan-hemoplasma SYBR green qPCR assay. A total of 198 blood samples from horses were collected, DNA extracted and pan-hemoplasma qPCR performed. All samples were negative by qPCR. To verify the presence of amplifiable DNA, PCR for glyceraldehyde-3-phosphate dehydrogenase gene was performed on all samples. This study was part of an active surveillance program, which is critical for monitoring animal health status, particularly in horses.

In the present study, we evaluated a cocktail of recombinant plasmids encoding the major surface proteins (rMSP1a, rMSP1b, rMSP4, rMSP5, VirB9 and VirB10) to improve the humoral and cellular immune response triggered in calves. Ten calves were divided in three groups: Four calves were inoculated with the plasmids/rMSPs (G1); 2 calves were inoculated with empty vector (G2); and 4 calves received saline (G3). SUMMARY The aims of this study were to determine the seroprevalence of Ehrlichia spp. and risk factors for exposure in a restricted population of dogs, horses, and humans highly exposed to tick bites in a Brazilian rural settlement using a commercial ELISA rapid test and two indirect immunofluorescent assays (IFA) with E. canis and E. chaffeensis crude antigens. Serum samples from 132 dogs, 16 horses and 100 humans were used. Fifty-six out of 132 (42.4%) dogs were seropositive for E. canis. Dogs > one year were more likely to be seropositive for E. canis than dogs ≤ one year (p = 0.0051). Ten/16 (62.5%) and 8/16 (50%) horses were seropositive by the commercial ELISA and IFA, respectively. Five out of 100 (5%) humans were seropositive for E. canis and E. chaffeensis. Rhipicephalus sanguineus (n = 291, 97.98%) on dogs and Amblyomma cajennense (n = 25, 96.15%) on horses were the most common ticks found. In conclusion, anti-Ehrlichia spp. antibodies were found in horses; however, the lack of a molecular characterization precludes any conclusion regarding the agent involved. Additionally, the higher seroprevalence of E. canis in dogs and the evidence of anti-Ehrlichia spp. antibodies in humans suggest that human cases of ehrlichiosis in Brazil might be caused by E. canis, or other closely related species.

Considering the zoonotic potential of tick-borne disease (TBD) agents and the fact that dogs may act as sentinels for human infection, the aim of the present study was to determine the seroprevalence of TBD agents and risk factors for exposure in two different canine populations from Parana State, Southern Brazil. A total of 138 dog serum samples from urban (UA) (n=68) and rural (RA) (n=70) areas were tested with commercial ELISA rapid test for Anaplasma phagocytophilum, Ehrlichia canis and Borrelia burgdorferi antibodies and indirect immunofluorescence assay (IFAT) for Babesia vogeli. An overall of 92∕138 (66.7%) dogs, being 62∕68 (91.2%) from UA and 30∕70 (42.9%) from RA, were seropositive for at least one TBD agent. From the total number of dogs, sixty-two were positive for E. canis (44.9%), 19 (13.8%) for A. phagocytophilum, and 64 (46.4%) for B. vogeli. Anti-B. burgdorferi antibodies were not detected. Dogs from UA showed a higher percentage of tick infestation (p = 0.0135) and were highly associated with seropositivity to E. canis (p = 0.000005), A. phagocytophilum (p = 0.0001), and B. vogeli (p = 0.0012). In summary, the findings indicate that dogs from urban areas present higher potential risk exposure to TBD pathogens than those from rural areas.Considerando o potencial zoonótico das doenças transmitidas por carrapatos (DTCs) e que os cães podem atuar como sentinelas para infecções em humanos, os objetivos deste estudo foram determinar a soroprevalência de agentes das DTCs e fatores de risco para a exposição em duas diferentes populações caninas do Estado do Paraná, região Sul do Brasil. Um total de 138 amostras de soro de cães de área urbana (AU) (n = 68) e rural (AR) (n = 70) foram testadas utilizando um teste de ELISA comercial rápido para detecção de anticorpos contra Anaplasma phagocytophilum, Ehrlichia canis e Borrelia burgdorferi e imunofluorescência indireta (IFI) para Babesia vogeli. Um total de 92∕138 (66,7%) cães, sendo 62∕68 (91,2%) da AU e 30∕70 (42,9%) da AR, foram soropositivos para pelo menos um agente. Do número total de amostras, sessenta e duas (44,9%) foram positivas para E. canis, 19 (13,8%) para A. phagocytophilum e 64 (46,4%) para B. canis vogeli. Anticorpos anti-B. burgdorferi não foram detectados. Os cães da AU apresentaram o maior percentual de infestação por carrapatos (p = 0,0135) e foram altamente associados com a positividade para E. canis (p = 0,000005), A. phagocytophilum (p = 0,0001) e B. vogeli (p = 0,0012). Em resumo, nossos achados indicam que cães de áreas urbanas têm um maior risco potencial de exposição a agentes patogênicos das DTCs comparados aos das áreas rurais.

Cisticercose bovina é uma doença endêmica em vários países com um importante potencial zoonótico e uma crescente importância na saúde pública. A inspecção da carne nos matadouros é fundamental para estabelecer a prevalência e prevenção de doenças zoonóticas, tais como a cisticercose. No entanto, no Brasil, não há estudo que tenha investigado a dinâmica da cisticercose em bovinos abatidos em todo o país. Assim, o presente estudo teve como objetivo usar Sistema de Informação Geográfica (GIS) como uma ferramenta para analisar a prevalência de cisticercose bovina em bovinos, usando os dados de matadouros inspecionados pelo Serviço de Inspeção Federal do Ministério da Agricultura, Pecuária e Abastecimento (SIF/MAPA), no período de janeiro de 2007 a abril de 2010. As informações de bovinos abatidos foram usadas para gerar um banco de dados sobre cisticercose bovina para todos os estados e municípios do Brasil, a fim de analisar e criar mapas epidemiológicos usando o software GIS. Durante o período analisado, 75.983.590 bovinos foram abatidos e a prevalência de cisticercose bovina no Brasil foi de 1,05%, com a região Sudeste apresentando a maioria dos casos. A maior quantidade de casos cisticercose bovina ocorreram no estado de São Paulo (400.834), seguido por Mato Grosso do Sul (151.735) e Paraná (94.046), enquanto não houve observação de ocorrência em nove estados. Os dados fornecidos por matadouros inspecionados pelo SIF/MAPA podem ser usados como uma fonte informativa para animais e agências de saúde pública para a prevenção do complexo teníase/cisticercose. Este estudo elucida a importância da inspecção da carne associado aos mapas epidemiológicos, visando o controle da doença nos rebanhos bovinos e a vigilância ativa para infecção zoonótica potencial para humano. Os dados deste estudo sugerem que a cisticercose bovina é endêmica em vários estados brasileiros, e o risco de infecção está diretamente relacionado com as principais áreas de criação de bovinos, independentemente da demografia da população humana.

Ehrlichiosis is a disease caused by rickettsial organisms belonging to the genus Ehrlichia. In Brazil, molecular and serological studies have evaluated the occurrence of Ehrlichia species in dogs, cats, wild animals and humans. Ehrlichia canis is the main species found in dogs in Brazil, although E. ewingii infection has been recently suspected in five dogs. Ehrlichia chaffeensis DNA has been detected and characterized in mash deer, whereas E. muris and E. ruminantium have not yet been identified in Brazil. Canine monocytic ehrlichiosis caused by E. canis appears to be highly endemic in several regions of Brazil, however prevalence data are not available for several regions. Ehrlichia canis DNA also has been detected and molecularly characterized in three domestic cats, and antibodies against E. canis were detected in free-ranging Neotropical felids. There is serological evidence suggesting the occurrence of human ehrlichiosis in Brazil but its etiologic agent has not yet been established. Improved molecular diagnostic resources for laboratory testing will allow better identification and characterization of ehrlichial organisms associated with human ehrlichiosis in Brazil.Erliquiose é uma doença causada por rickettsias pertencentes ao gênero Ehrlichia. No Brasil, estudos sorológicos e moleculares têm avaliado a ocorrência de espécies de Ehrlichia em cães, gatos, animais selvagens e seres humanos. Ehrlichia canis é a principal espécie em cães no Brasil, embora a infecção por E. ewingii tenha, recentemente, despertado suspeita em cinco cães. O DNA de E. chaffeensis foi detectado e caracterizado em cervo-do-pantanal, enquanto que E. muris e E. ruminantium ainda não foram identificadas no Brasil. A erliquiose monocítica canina causada pela E. canis parece ser altamente endêmica em muitas regiões do Brasil, embora dados de prevalência não estejam disponíveis em muitas delas. O DNA de E. canis também foi detectado e caracterizado em três gatos domésticos, enquanto anticorpos contra E. canis foram detectados em felídeos neotropicais de vida livre. Evidências sorológicas sugerem a ocorrência de erliquiose humana no Brasil, entretanto, o agente etiológico ainda não foi identificado. A melhoria do diagnóstico molecular promoverá a identificação e caracterização de espécies associadas à erliquiose humana no Brasil.

Introduction: The aim of this study was to evaluate the serological cross-reactivity between Leishmania sp. and other canine pathogens. Methods: Positive serum samples for Ehrlichia canis, Babesia canis, Toxoplasma gondii, Neospora caninum and Trypanosoma cruzi were tested using three serological methods enzyme linked immunosorbent assay (ELISA), indirect immunofluorescent antibody test (IFAT) and Kalazar Detect™, for canine visceral leishmaniasis. Results: Of the 57 dog samples tested, 24 (42.1%) tested positive using one of the three serological methods: 10/57 (17.5%) for ELISA, 11/57 (19.3%) for IFAT and 3/57 (5.3%) for Kalazar Detect™. Conclusions: Our results demonstrated that the presence of other infectious agents may lead to cross-reactivity on leishmaniasis serological tests.

Recent studies have been conducted in Brazil using molecular techniques for the detection of hemotrophic mycoplasmas in several mammals. In domestic cats, Mycoplasma haemofelis, 'Candidatus M. haemominutum', and 'Candidatus M. turicensis' infections have been identified. These species have also been found in free-ranging and captive neotropical felid species. Two canine hemoplasmas, Mycoplasma haemocanis and 'Candidatus Mycoplasma haematoparvum', have been identified in dogs. In commercial swine populations, Mycoplasma suis was found to be highly prevalent, especially in sows. Moreover, novel mycoplasma species have been identified in Brazilian commercial pigs and domestic dogs. A hemoplasma infection in a human patient infected with the human immunodeficiency virus (HIV) was also recently documented. In conclusion, hemoplasma species are common and important infectious agents in Brazil. Further studies should be conducted to better understand their impact on pets, production animals, and wildlife fauna, as well as their role as zoonotic agents, particularly in immunocompromised patients.Estudos recentes utilizando técnicas moleculares para a detecção de micoplasmas hemotróficos em diferentes mamíferos têm sido conduzidos no Brasil. Em gatos domésticos, infecções por Mycoplasma haemofelis, 'Candidatus M. haemominutum' e 'Candidatus M. turicensis' foram identificadas. Estas espécies também foram encontradas em felídeos neotropicais de vida livre e de cativeiro. Dois hemoplasmas caninos, Mycoplasma haemocanis e 'Candidatus Mycoplasma haematoparvum', foram identificados em cães domésticos. Em populações comerciais de suínos, Mycoplasma suis possui alta prevalência, especialmente em porcas. Além disso, novas espécies de hemoplasmas foram detectadas em suínos comercias e cães. Infecção por um hemoplasma em um paciente humano infectado com o vírus da imunodeficiência humana (HIV) foi recentemente documentada. Em conclusão, espécies de hemoplasmas são comuns e importantes agentes de infecções no Brasil. Estudos futuros devem ser conduzidos para melhor entender seu impacto em cães e gatos, animais de produção e na fauna silvestre, e também para determinar o seu papel como agentes zoonóticos, particularmente em pacientes imunocomprometidos.

Mycoplasma suis is a hemotropic bacteria of red blood cells and the causative agent of swine eperythrozoonosis. Diagnosis of infection may be reached by direct examination of blood smears; however, the use of polymerase chain reaction (PCR) of the 16S RNA gene of M. suis improves the sensitivity and specificity of detection. The aim of this study was to screen peccaries (Tayassu tajacu and T. pecari) for M. suis infection using a specific conventional PCR. A total of 28 blood samples from captive collared and white-lipped peccaries were collected, DNA extracted and a specific M. suis PCR assay performed. All samples were negatives by both blood smear examination and PCR testing. To verify the presence of amplifiable DNA, PCR for beta-actin gene was performed in all samples. This study was part of an active surveillance program, which is crucial for monitoring animal health status, particularly in wildlife species.Mycoplasma suis é uma bactéria hemotrópica dos eritrócitos e é o agente causador da eperitrozoonose suína. O diagnóstico da infecção pode ser realizado pelo exame direto de esfregaços sanguíneos; entretanto, o uso da reação em cadeia da polimerase (PCR) baseada no gene 16S RNA de M. suis aumenta a sensibilidade e especificidade da detecção. O objetivo deste estudo foi avaliar catetos e queixadas (Tayassu tajacu e T. pecari) para a infecção por M. suis, utilizando PCR convencional específico. Um total de 28 amostras de sangue de catetos e queixadas de cativeiro foram coletadas, o DNA foi extraído e a PCR específica para a detecção de M. suis realizada. Todas as amostras foram negativas pelo esfregaço sanguíneo e PCR. Para verificar a presença de DNA amplificável, PCR para o gene da beta actina foi realizada em todas as amostras. Este estudo foi parte de um programa de vigilância ativa, o qual é crucial para o monitoramento do estado de saúde animal, particularmente em espécies selvagens.

A cinomose é uma enfermidade contagiosa, grave e multissistêmica, causada por um vírus do gênero Morbillivirus, de disseminação mundial com alta taxa de letalidade e que afeta principalmente os cães. O seu diagnóstico é baseado nos sinais clínicos associados a achados hematológicos. A observação do Corpúsculo de Lentz (CL) em eritrócitos ou leucócitos confere o diagnóstico definitivo para esta enfermidade. Objetivou-se testar a hipótese de que o quadro hematológico de cães com cinomose difere de acordo com o tipo de célula sanguínea que apresenta o CL. Para tal foi avaliado o perfil hematológico de 25 cães positivos para a doença atendidos no Hospital Veterinário “Luis Quintiliano de Oliveira” da Universidade Estadual Paulista (UNESP), campus de Araçatuba. O diagnóstico da doença foi realizado após a visibilização do CL em esfregaços sanguíneos. Do total de casos, 64% apresentaram anemia, 16% leucopenia e 12% leucocitose. A linfopenia ocorreu em 76% dos cães. As inclusões virais foram observadas em neutrófi los (32%), linfócitos (28%) e eritrócitos (12%). Observação concomitante ocorreu em linfócitos e eritrócitos (4%), em linfócitos e neutrófilos (12%), em neutrófilos e monócitos (4%) e em neutrófilos e eritrócitos (4%). Em um único caso o CL foi observado simultaneamente em neutrófilos, monócitos e linfócitos. O quadro hematológico não está associado à presença da inclusão viral em um determinado tipo celular.

The aims of this study were to determine the occurrence ofAnaplasma platys and Ehrlichia canisinfection in dogs in Porto Alegre, Southern Brazil; and to investigate their association with hematological abnormalities. Serum samples from 196 dogs were first tested using dot-ELISA for antibodies against Anaplasmaspp. and Ehrlichia canis. Peripheral blood samples from 199 dogs were subjected to 16S rRNA nested PCR (nPCR) for A. platysand E. canis, followed by DNA sequencing to ensure pathogen identity. A total of 19/196 samples (9.69%) were positive forAnaplasma spp. using ELISA and 28/199 (14.07%) samples were positive for A. platys by nested PCR. All the dog samples were negative for E. canis, both in anti-E. canisantibody tests and in nested PCR. There were no significant differences in hematological parameters between A. platys-PCR positive and negative dogs and Anaplasma spp. serologically positive dogs, except for basophil counts, which were higher in nPCR-positive dogs. This is the first report showing A. platys presence in dogs in Southern Brazil. In conclusion, hematological parameters may not be sufficient to diagnose A. platys infection in dogs in Southern Brazil, probably due either to low pathogenicity or to chronic infection. On the other hand, E. canis may either have very low occurrence or be absent in dogs in Porto Alegre.

Abstract The aims of the present study were to serosurvey dogs, horses, and humans highly exposed to tick bites for anti-Borrelia burgdorferi s.l. antibodies, identify tick species present, and determine risk factors associated with seropositivity in a rural settlement of Paraná State, southern Brazil. Eighty-seven residents were sampled, along with their 83 dogs and 18 horses, and individual questionnaires were administered. Immunofluorescence antibody test (IFAT) was performed on serum samples and positive samples were subjected to western blot (WB) analysis. Anti-B. burgdorferi antibodies were found in 4/87 (4.6%) humans, 26/83 (31.3%) dogs, and 7/18 (38.9%) horses by IFAT, with 4/4 humans also positive by WB. Ticks identified were mostly from dogs and included 45/67 Rhipicephalus sanguineus, 21/67 Amblyomma ovale, and 1/67 A. cajennense sensu lato. All (34/34) horse ticks were identified as A. cajennense s.l.. No significant association was found when age, gender, or presence of ticks was correlated to seropositivity to Borrelia sp. In conclusion, although anti-Borrelia antibodies have been found in dogs, horses and their owners from the rural settlement, the lack of isolation, molecular characterization, absence of competent vectors and the low specificity of the commercial WB kit used herein may have impaired risk factor analysis.

Despite our current knowledge of the immunology, pathology, and genetics of Anaplasma marginale, prevention in cattle is currently based on old standbys, including live attenuated vaccines, antibiotic treatment, and maintaining enzootic stability in cattle herds. In the present study, we evaluated the use of an immunostimulant complex (ISCOMATRIX) adjuvant, associated with a pool of recombinant major surface proteins (rMSP1a, rMSP1b, rMSP4 and rMSP5) to improve the humoral immune response triggered in calves mainly by IgG2. Ten calves were divided in three groups: 4 calves were inoculated with the ISCOMATRIX/rMSPs (G1); 2 calves were inoculated with ISCOMATRIX adjuvant (G2); and 4 calves received saline (G3). Three inoculations were administered at 21-day intervals. In G1, the calves showed significant increases in total IgG, IgG1 and IgG2 levels 21 days after the second inoculation, compared to the control group (p < 0.05), and G1 calves remained above the cut-off value 28 days after the third inoculation (p < 0.05). The post-immunized sera from calves in G1 reacted specifically for each of the rMSPs used. In conclusion, the ISCOMATRIX/rMSPs induced antigen-specific seroconversion in calves. Therefore, additional testing to explore the protection induced by rMSPs, both alone and in conjunction with proteins previously identified as subdominant epitopes, is warranted.

Abstract Toxoplasmosis is a zoonotic disease of global distribution that affects all warm-blooded animals. The purpose of this investigation was to determine the prevalence of T. gondii infection and identify the risk factors associated with its occurrence in domestic ruminants raised on the island of Fernando de Noronha, Brazil, and to confirm that cattle and sheep raised in Fernando de Noronha Island present statistically different T. gondii prevalence rates. Serum samples were collected from sheep (n=240) and cattle (n=140) for the detection of antibodies by indirect immunofluorescence. Samples were collected from all the animals on all the farms. Risk factors were analyzed by univariate analysis and logistic regression. The prevalence rate of positive sheep was 85.0% while that of cattle was 10.7%. A multivariate analysis revealed that the site of contact of sheep with felines was a risk factor. For cattle, the risk factors identified in this study were: extensive farming system, water source, more than three cats per farm, and the presence of rats in feed storage locations. The findings revealed a significant difference in the prevalence rates in sheep and cattle raised in this insular environment.

Abstract INTRODUCTION: Brazilian native species are reemerging as increasingly free-ranging populations. METHODS: Sera from 31 capybaras (Hydrochoerus hydrochaeris) and 28 peccaries (Pecari tajacu and Tayassu pecari) were tested for anti-Leptospira and anti-Toxoplasma gondii antibodies using microscopic seroagglutination test. RESULTS: Nineteen percent of free-ranging and 10.0% of captive capybaras, along with 31.8% of collared peccaries, were seropositive for T. gondii. None was seropositive for Leptospira sp. CONCLUSIONS: The present findings indicated low risk of disease, particularly among capybaras and white-lipped peccaries; however, active surveillance programs are important for monitoring wildlife health and public health once they are in public parks around cities.

Abstract Neighborhood dogs may act as reservoirs for several zoonotic protozoan infections, particularly in urban areas, thus constituting a potential public health threat. Accordingly, the aim of the present study was to evaluate the exposure of neighborhood dogs to four protozoan pathogens in public areas with high levels of human movement in Curitiba, southern Brazil. Blood samples from 26 neighborhood dogs were screened by means of the indirect immunofluorescent antibody test (IFAT) for Leishmania spp., Toxoplasma gondii, Trypanosoma cruzi and Neospora caninum, and a questionnaire was answered by the respective keeper. A total of 8/26 dogs (30.7%) seroreactive to T. gondii, 3/26 (11.5%) to N. caninum and 2/26 (7.7%) to both were identified. All the samples were seronegative for T. cruzi and Leishmania spp. Pathogen seroreactivity was not associated with the daily human movements or other epidemiological variables investigated (p > 0.05). In conclusion, the low seroprevalence for T. gondii and N. caninum indicated low environmental and food risk for animal infection and the seronegativity for Leishmania spp. and T. cruzi may reflect the absence of these pathogens in urban areas of Curitiba. Moreover, neighborhood dogs may be used as environmental sentinels for the presence of protozoan pathogens and their vectors.

Abstract Opossums are marsupials from the New World of the genus Didelphis and known as synanthropic animals due to their proximity with human beings. To date, ‘Candidatus Mycoplasma haemodidelphis’ has been solely found infecting the North American opossum (Didelphis virginiana). Accordingly, the aim of this study was to screen eight white-eared opossums (Didelphis albiventris) from a public park in Maringa city, Paraná State, southern Brazil, for hemoplasma infection. Blood samples were taken from caudal venipuncture, and DNA was extracted and further screened by a pan-hemoplasma PCR assay. Seven out of eight (87.50%; CI 95%: 47.35-99.68%) white-eared opossums were positive for Mycoplasma spp. Sequencing of the 16S rRNA fragment showed 98,97% identity with ‘Ca. M. haemodidelphis’ detected in the USA. Three out of eight (37.50%; CI 95%: 8.52-75.51%) white-eared opossums were infested by Amblyomma dubitatum ticks. This is the first report on detection of a potentially novel hemotropic Mycoplasma sp. infecting opossums from South America.
