The Piauí laterite (NE Brazil) was initially evaluated for Ni but also contains economic concentrations of Co. Our investigations aimed to characterise the Co enrichment within the deposit; by understanding the mineralogy we can better design mineral processing to target Co recovery. The laterite is heterogeneous on the mineralogical and lithological scale differing from the classic schematic profiles of nickel laterites, and while there is a clear transition from saprolite to more ferruginous units, the deposit also contains lateral and vertical variations that are associated with both the original intrusive complex and also the nature of fluid flow, redox cycling and fluctuating groundwater tables. The deposit is well described by the following six mineralogical and geochemical units: SAPFE, a clay bearing ferruginous saprolite; SAPSILFE, a silica dominated ferruginous saprolite; SAPMG, a green magnesium rich chlorite dominated saprolite; SAPAL, a white-green high aluminium, low magnesium saprolite; saprock, a serpentine and chlorite dominated saprolite and the serpentinite protolith. Not all of these units are ‘ore bearing’. Ni is concentrated in a range of nickeliferous phyllosilicates (0.1–25 wt%) including serpentines, talc and pimelite, goethite (up to 9 wt%), magnetite (2.8–14 wt%) and Mn oxy-hydroxides (0.35–19 wt%). Lower levels of Ni are present in ilmenites, chromites, chlorite and distinct small horizons of nickeliferous silica (up to 3 wt% Ni). With respect to Co, the only significant chemical correlation is with Mn, and Mn oxy-hydroxides contain up to 14 wt% Co. Cobalt is only present in goethite when Mn is also present, and these goethite grains contain an average of 0.19 wt% Co (up to a maximum of 0.65 wt%). The other main Co bearing minerals are magnetite (0.41–1.89 wt%), chlorite (up to 0.45 wt%) and ilmenite (up to 0.35 wt%). Chemically there are three types of Mn oxy-hydroxide, asbolane, asbolane-lithiophorite intermediates and romanechite. Spatially resolved X-ray absorption spectroscopy analysis suggests that the Co is present primarily as octahedrally bound Co3+ substituted directly into the MnO6 layers of the asbolane-lithiophorite intermediates. However significant levels of Co2+ are evident within the asbolane-lithiophorite intermediates, structurally bound along with Ni in the interlayer between successive MnO6 layers. The laterite microbial community contains prokaryotes and few fungi, with the highest abundance and diversity closest to ground level. Microorganisms capable of metal redox cycling were identified to be present, but microcosm experiments of different horizons within the deposit demonstrated that stimulated biogeochemical cycling did not contribute to Co mobilisation. Correlations between Co and Mn are likely to be a relic of parent rock weathering rather than due to biogeochemical processes; a conclusion that agrees well with the mineralogical associations.

Cancer is a disease of cellular evolution where single base changes in the genetic code can have significant impact on the translation of proteins and their activity. Thus, in cancer research there is significant interest in methods that can determine mutations and identify the significant binding sites (epitopes) of antibodies to proteins in order to develop novel therapies. Nano molecularly imprinted polymers (nanoMIPs) provide an alternative to antibodies as reagents capable of specifically capturing target molecules depending on their structure. In this study, we used nanoMIPs to capture KRAS, a critical oncogene, to identify mutations which when present are indicative of oncological progress. Herein, coupling nanoMIPs (capture) and liquid chromatography-mass spectrometry (detection), LC-MS has allowed us to investigate mutational assignment and epitope discovery. Specifically, we have shown epitope discovery by generating nanoMIPs to a recombinant KRAS protein and identifying three regions of the protein which have been previously assigned as epitopes using much more time-consuming protocols. The mutation status of the released tryptic peptide was identified by LC-MS following capture of the conserved region of KRAS using nanoMIPS, which were tryptically digested, thus releasing the sequence of a non-conserved (mutated) region. This approach was tested in cell lines where we showed the effective genotyping of a KRAS cell line and in the plasma of cancer patients, thus demonstrating its ability to diagnose precisely the mutational status of a patient. This work provides a clear line-of-sight for the use of nanoMIPs to its translation from research into diagnostic and clinical utility., We show using Molecular imprinted Polymers (MIPs) and LC-MS/SRM that we can identify the KRAS mutation in cancer patients plasma as well as carry out epitope discovery for drug target evaluation.

Purpose The purpose of this article is to provide a general review of the health-care needs in Kenya which focuses on the role of community engagement in facilitating access and diminishing barriers to quality care services. Health-care concerns throughout Kenya and the culture of Kenyan’s health-care practices care are considered. Design/methodology/approach A comprehensive review covered studies of community engagement from 2000 till present. Studies are collected using Google Scholar, PubMed, EBSCOhost and JSTOR and from government and nongovernment agency websites. The approach focuses on why various populations seek health care and how they seek health care, and on some current health-care delivery models. Findings Suggestions for community engagement, including defining the community, are proposed. A model for improved health-care delivery introduces community health workers (CHWs), mHealth technologies and the use of mobile clinics to engage the community and improve health and quality of care in low-income settings. Practical implications The results emphasize the importance of community engagement in building a sustainable health-care delivery model. This model highlights the importance of defining the community, setting goals for the community and integrating CHWs and mobile clinics to improve health status and decrease long-term health-care costs. The implementation of these strategies contributes to an environment that promotes health and wellness for all. Originality/value This paper evaluates health-care quality and access issues in Kenya and provides sustainable solutions that are linked to effective community engagement. In addition, this paper adds to the limited number of studies that explore health-care quality and access alongside community engagement in low-income settings.

A contribution to a special issue on Hormones and Human Competition. Previous research and theory suggest testosterone is an important hormone for modulating aggression and self-regulation. We propose that self-construal, a culturally-relevant difference in how individuals define the self in relation to others, may be an important moderator of the relationship between testosterone and behaviors linked to aggression. Within two studies (Study 1 N=80; Study 2 N=237) and an integrated data analysis, we find evidence suggesting that acute testosterone changes in men are positively associated with aggressive behavior for those with more independent self-construals, whereas basal testosterone is negatively associated with aggression when individuals have more interdependent self-construals. Although preliminary, these findings suggest that self-construal moderates the association between testosterone and aggression, thereby paving the way toward future work examining the potential cultural moderation of the behavioral effects of testosterone.

Rationale There is a considerable clinical demand to determine key mutations in genes involved with cancer which necessitates the deployment of highly specific and robust analytical methods. Multiplex liquid chromatography with selected reaction monitoring (LC/SRM) assays offer the ability to achieve quantitation down to levels expected to be present in clinical samples. Ion mobility mass spectrometry (IMS/MS) assays can provide increased peak capacity and hence separation in an extremely short time frame, and in addition provide physicochemical data regarding the collision cross-section of an analyte which can be used in conjunction with the m/z value of an ion to increase detection specificity. Methods For LC/SRM, unlabelled peptides and corresponding stable-isotope-labelled standards were spiked into digested human plasma and analysed using ultrahigh-performance liquid chromatography (UHPLC) coupled to a triple quadrupole mass spectrometer to enable the generation of analyte-specific calibration lines. Synthetic unlabelled peptides were infused into a Synapt G2 mass spectrometer for travelling wave ion mobility separation and TW CCSN2 values were derived from comparison with previously generated TW CCSN2 calibration values. Results Linear calibration lines (0.125 to 25 fmol/μL) were established for each of the KRAS peptides. UHPLC separated the peptides and hence enabled them to be split into different retention time functions/windows. This separation enabled detection of three or four transitions for each light and heavy peptide with at least 10 points per peak for accurate quantitation. All six KRAS G12 peptides were separated using IMS/MS, enabling precise TW CCSN2 values to be determined. Although some of the G12 peptides chromatographically co-eluted, all the peptides were distinguished by m/z, retention time and/or drift time. Conclusions This study advocates that LC/SRM and IMS/MS could both be used to identify single amino acid substitutions in KRAS as an alternative to commonly used methods such as circulating tumour DNA analysis.

Previous research demonstrates that prenatal testosterone exposure increases aggression, possibly through its effects on the structure and function of neural circuits supporting threat detection and emotion regulation. Here we examined associations between regional gray matter volume, trait aggression, and the ratio of the second and fourth digit of the hand (2D:4D ratio) as a putative index of prenatal testosterone exposure in 464 healthy young adult volunteers. Our analyses revealed a significant positive correlation between 2D:4D ratio and gray matter volume of the dorsal anterior cingulate cortex (dACC), a brain region supporting, emotion regulation, conflict monitoring, and behavioral inhibition. Subsequent analyses demonstrated that reduced (i.e., masculinized) gray matter volume in the dACC mediated the relationship between 2D:4D ratio and aggression in women, but not men. Expanding on this gender-specific mediation, additional analyses demonstrated that the shared variance between 2D:4D ratio, dACC gray matter volume, and aggression in women reflected the tendency to engage in cognitive reappraisal of emotionally provocative stimuli. Our results provide novel evidence that 2D:4D ratio is associated with masculinization of dACC gray matter volume, and that this neural phenotype mediates, in part, the expression of trait aggression in women.

Testosterone (T) concentrations change rapidly in the context of human competition, and these changes in neuroendocrine function may serve to modulate future aggressive behavior. However, an increase in T during competition does not translate into aggressive behavior among all individuals. Here, we examined the extent to which individual differences in trait anxiety moderate the relationship between T responses to competition and aggressive behavior. Across two studies, we found that T responses to competition were positively correlated with subsequent aggression, but only among men scoring relatively low in trait anxiety. Trait anxiety did not moderate the relationship between T reactivity and aggression in women. These findings highlight the importance of considering individual difference in trait anxiety when examining the neuroendocrine correlates of human aggression.

Investigations into the formation of new ettringite-type phases with a range of trivalent and tetravalent cations were carried out to further study the potential this structure type has to incorporate cations covering a range of ionic radii (0.53–0.69 A). We report the synthesis and structural characterisation of a new ettringite-type phase, Ca 6 [Ga(OH) 6 ·12H 2 O] 2 (SO 4 ) 3 ·2H 2 O, which was indexed in space group P 31 c with the unit cell parameters a = 11.202(2) A, c = 21.797(3) A and two new thaumasite-type phases Ca 6 [M(OH) 6 ·12H 2 O] 2 (SO 4 ) 2 (CO 3 ) 2 , M = Mn, Sn which were indexed in space group P 6 3 with the unit cell parameters a = 11.071(5) A, c = 21.156(8) A and a = 11.066(1) A, c = 22.420(1) A respectively. These new phases show the versatility of the ettringite family of structures to tolerate a large range of cation sizes on the octahedral M site and highlights the preference of tetravalent cations to crystallise with the thaumasite structure over the ettringite structure.

The new title phases along with ettringite, Ca6 [Al(OH)6·12H2O] 2(SO4)3·2H2O, and its known iron analogue are prepared from solutions of M2(SO4)3 (M: Al, Fe, Ga) or MSO4 (M: Mn, Sn) and CaO mixed with a 10% aq.
