Abstract (1) Background: Endophytic bacteria represent an important component of plant wellness. They have been widely studied for their involvement in plant development and enhancement of stress tolerance. In this work, the endophytic communities of roots, stems, and leaves of blackberry (Rubus ulmifolius Schott) were studied in three different niches: natural, riverside, and human-impacted niches. (2) Results: The microbiome composition revealed that Sphingomonadaceae was the most abundant family in all samples, accounting for 9.4–45.8%. In contrast, other families seem to be linked to a specific tissue or niche. Families Microbacteriaceae and Hymenobacteraceae increased their presence in stem and leaf samples, while Burkholderiaceae abundance was important in riverside samples. Alpha and beta diversity analyses showed that root samples were the most diverse, and they gathered together in the same cluster, apart from the rest of the samples. (3) Conclusions: The analysis of the microbiome of R. ulmifolius plants revealed that the composition was essentially the same in different niches; the differences were primarily influenced by plant tissue factors with a core genome dominated by Sphingomonadaceae. Additionally, it was observed that R. ulmifolius can select its own microbiome, and this remains constant in all tissues evaluated regardless the niche of sampling.

Abstract Background Soils affected by salinity are a recurring problem that is continually increasing due to the impact of climate change on weather conditions and ineffective agricultural management practices. The use of plant growth promoting (PGP) Bacteria can alleviate its effects. In this regard, the genus Rhizobium has demonstrated excellent PGP capabilities through various plant growth promotion mechanisms and may therefore be a promising biofortifier under saline conditions. However, little is known about the production of volatile organic compounds (VOCs) by bacteria of this genus and their effects on plant development. Here, we aim to characterize the volatilome (the set of volatile metabolites synthesized by an organism) of Rhizobium for the first time and to further investigate the direct and VOC-mediated interaction between a strain of this genus and lettuce, a crop severely affected by salinity, both under saline and non-saline conditions. Results In this study, it was shown that the use of Rhizobium sp. GPTR29 was able to increase the production of lettuce (Lactuca sativa L.) under normal and saline conditions. We analyzed the Rhizobium volatilome under non-saline (0 mM NaCl) and saline (100 mM NaCl) conditions by HS-SPME-GC‒MS and found a differential composition in response to salinity. We detected 20 different compounds, where 3-methyl-1-butanol, 2-methyl-1-butanol, and α-pinene were the backbone of the Rhizobium volatilome. Exposure to these compounds in bicameral plates under salt stress resulted in increases in plant development of 17.1%, 16.0% and 33.1% in aerial part size, number of leaves and root length, respectively. Under greenhouse conditions and salinity, the inoculation of Rhizobium sp. GPTR29 resulted in an increase of 17.8% and 27.4% in shoot fresh and dry weight, respectively. Phenolic compounds were analyzed by HPLC–DAD-MS, revealing an increase in total flavonoid content under salinity conditions (100 mM NaCl) and apigenin derivative, luteolin 7-O-glucoside and quercetin 3-O-glucuronide individually. Conclusions These results provide new avenues for the study of PGP mechanisms in this bacterial genus, such as VOCs and their effects on plant growth, which play an important role in mediating plant–microorganism interactions. Abstract Background Plant-associated microbial communities play important roles in host nutrition, development and defence. In particular, the microbes living within internal plant tissues can affect plant metabolism in a more intimate way. Understanding the factors that shape plant microbial composition and discovering enriched microbes within endophytic compartments would thus be valuable to gain knowledge on potential plant–microbial coevolutions. However, these interactions are usually studied through reductionist approaches (in vitro models or crop controlled systems). Here, we investigate these ecological factors in wild forest niches using proximally located plants from two distant taxa (blueberry and blackberry) as a model. Results Although the microbial communities were quite similar in both plants, we found that sampling site had a high influence on them; specifically, its impact on the rhizosphere communities was higher than that on the roots. Plant species and sample type (root vs. rhizosphere) affected the bacterial communities more than the fungal communities. For instance, Xanthobacteraceae and Helotiales taxa were more enriched in roots, while the abundance of Gemmatimonadetes was higher in rhizospheres. Acidobacteria abundance within the endosphere of blueberry was similar to that in soil. Several taxa were significantly associated with either blackberry or blueberry samples regardless of the sampling site. For instance, we found a significant endospheric enrichment of Nevskia in blueberry and of Sphingobium, Novosphingobium and Steroidobacter in blackberry. Conclusions There are selective enrichment and exclusion processes in the roots of plants that shapes a differential composition between plant species and sample types (root endosphere—rhizosphere). The special enrichment of some microbial taxa in each plant species might suggest the presence of ancient selection and/or speciation processes and might imply specific symbiosis. The selection of fungi by the host is more pronounced when considering the fungal trait rather than the taxonomy. This work helps to understand plant–microbial interactions in natural ecosystems and the microbiome features of plants.

Abstract Background Ips typographus (European spruce bark beetle) is the most destructive pest of spruce forests in Europe. As for other animals, it has been proposed that the microbiome plays important roles in the biology of bark beetles. About the bacteriome, there still are many uncertainties regarding the taxonomical composition, insect-bacteriome interactions, and their potential roles in the beetle ecology. Here, we aim to deep into the ecological functions and taxonomical composition of I. typographus associated bacteria. Results We assessed the metabolic potential of a collection of isolates obtained from different life stages of I. typographus beetles. All strains showed the capacity to hydrolyse one or more complex polysaccharides into simpler molecules, which may provide an additional carbon source to its host. Also, 83.9% of the strains isolated showed antagonistic effect against one or more entomopathogenic fungi, which could assist the beetle in its fight against this pathogenic threat. Using culture-dependent and -independent techniques, we present a taxonomical analysis of the bacteriome associated with the I. typographus beetle during its different life stages. We have observed an evolution of its bacteriome, which is diverse at the larval phase, substantially diminished in pupae, greater in the teneral adult phase, and similar to that of the larval stage in mature adults. Our results suggest that taxa belonging to the Erwiniaceae family, and the Pseudoxanthomonas and Pseudomonas genera, as well as an undescribed genus within the Enterobactereaceae family, are part of the core microbiome and may perform vital roles in maintaining beetle fitness. Conclusion Our results indicate that isolates within the bacteriome of I. typographus beetle have the metabolic potential to increase beetle fitness by proving additional and assimilable carbon sources for the beetle, and by antagonizing fungi entomopathogens. Furthermore, we observed that isolates from adult beetles are more likely to have these capacities but those obtained from larvae showed strongest antifungal activity. Our taxonomical analysis showed that Erwinia typographi, Pseudomonas bohemica, and Pseudomonas typographi species along with Pseudoxanthomonas genus, and putative new taxa belonging to the Erwiniaceae and Enterobacterales group are repeatedly present within the bacteriome of I. typographus beetles, indicating that these species might be part of the core microbiome. In addition to Pseudomonas and Erwinia group, Staphylococcus, Acinetobacter, Curtobacterium, Streptomyces, and Bacillus genera seem to also have interesting metabolic capacities but are present in a lower frequency. Future studies involving bacterial-insect interactions or analysing other potential roles would provide more insights into the bacteriome capacity to be beneficial to the beetle.

Accurate trajectory tracking is a paramount objective when a mobile robot must perform complicated tasks. In high-speed movements, hardware-induced delays may produce overshoots and even instability when controlling the system. In this case, Smith predictor controllers can be used because they are well suited for delayed processes. This paper addresses the accurate positioning of a mobile robot on a terrain of an unknown slope. This slope produces disturbance torques of unknown amplitudes in the robot actuators that yield a steady-state error in the positioning. Because our actuators are integrating plus time delay plants, the standard Smith predictor cannot remove these disturbances. This paper proposes a modification of this control scheme in order to remove these disturbances yielding a zero steady-state error in the actuators. Our new scheme is compared with other modified SPs existing in the literature by means of simulations. These simulations show the superior performance of our scheme in the sense of removing the steady-state error more efficiently (i.e., faster) than other schemes. Finally, the performance of our control scheme is tested experimentally in a low-cost mobile robot.

ABSTRACT Each Earth ecosystem has unique microbial communities. Pseudomonas bacteria have evolved to occupy a plethora of different ecological niches, including living hosts, such as animals and plants. Many genes necessary for the Pseudomonas-niche interaction and their encoded functions remain unknown. Here, we describe a comparative genomic study of 3,274 genomes with 19,056,667 protein-coding sequences from Pseudomonas strains isolated from diverse environments. We detected functional divergence of Pseudomonas that depends on the niche. Each group of strains from a certain environment harbored a distinctive set of metabolic pathways or functions. The horizontal transfer of genes, which mainly proceeded between closely related taxa, was dependent on the isolation source. Finally, we detected thousands of undescribed proteins and functions associated with each Pseudomonas lifestyle. This research represents an effort to reveal the mechanisms underlying the ecology, pathogenicity, and evolution of Pseudomonas, and it will enable clinical, ecological, and biotechnological advances. IMPORTANCE Microbes play important roles in the health of living beings and in the environment. The knowledge of these functions may be useful for the development of new clinical and biotechnological applications and the restoration and preservation of natural ecosystems. However, most mechanisms implicated in the interaction of microbes with the environment remain poorly understood; thus, this field of research is very important. Here, we try to understand the mechanisms that facilitate the differential adaptation of Pseudomonas—a large and ubiquitous bacterial genus—to the environment. We analyzed more than 3,000 Pseudomonas genomes and searched for genetic patterns that can be related with their coevolution with different hosts (animals, plants, or fungi) and environments. Our results revealed that thousands of genes and genetic features are associated with each niche. Our data may be useful to develop new technical and theoretical advances in the fields of ecology, health, and industry.

(1) Background: Nowadays, special attention has been paid to red and purple fruits, including blueberries and sweet cherries, since they are highly attractive to consumers due to their organoleptic properties, standing out due to their vibrant red and purple colours and sweet flavour, and nutritional value. (2) Methods: The present study evaluated the phenolic profile of phenolic-enriched extracts from blueberries and sweet cherries and explored their antioxidant potential against DPPH, superoxide and nitric oxide radicals, and ferric species, and their potential to inhibit the α-glucosidase enzyme. Furthermore, their antimicrobial activity was also determined by microdilution method against four Gram-positive strains (Enterococcus faecalis ATCC 29212, Bacillus cereus ATCC 11778, Listeria monocytogenes LMG 16779, and Staphylococcus aureus ATCC 25923) and five Gram-negative strains (Salmonella enterica subsp. enterica ATCC 13311 serovar Typhimurium, Klebsiella pneumoniae ATCC 13883, Proteus mirabilis CECT 170, Serratia marcescens CECT 159, and Acinetobacter baumannii LMG 1025). (3) Results: By chromatographic techniques, eight anthocyanins were detected in blueberry coloured fraction and total extract, and five anthocyanins were detected in sweet cherry total extract and coloured fraction, while quercetin aglycone and chlorogenic acids were the dominant non-coloured compounds in blueberries and sweet cherries, respectively. All extracts demonstrated significant antioxidant properties, as well as the ability to inhibit the activity of α-glucosidase enzyme and the development of various microorganisms. (4) Conclusion: The obtained data evidence the promising biological potential of blueberries and sweet cherries, being highly correlated with the presence of phenolic compounds.

In this paper we present several hydrological time series from Argentina that include, evapotranspiration, precipitation, and stream flow. We survey previous results and apply the 0-1 test for chaos to classify the sequences as regular or chaotic. Previous studies have shown evidence of chaos in several observables from hydrology using the traditional phase space reconstruction method and the computation of Lyapunov exponents. The 0-1 test for chaos can be used as a first step to identify the type of time series, that later can be subjected to the more detailed analysis of the phase space reconstruction. Assuming that the systems that generated these time series are deterministic, the 0-1 test for chaos classifies all of them as chaotic

Evapotranspiration is a key variable of the water cycle. Its calculation requires several ground data that frequently are not available. This study contains a detailed method and measurements of meteorological and energy balance variables that can be used to estimate the daily actual evapotranspiration (ETa). A linear generalized model is obtained to calculate the ETa from common variables measured in meteorological stations. The vegetation water status is a crucial variable for modelling of drought impact, vegetation productivity and water fluxes. Methods for spatial estimation of this variable still need to be improved. The integration of remotely sensed data of land surface temperature (LST) and water vegetation indices based on near-infrared (NIR) and short-wave infrared (SWIR) reflectance for estimation of vegetation water content and water available for evapotranspiration require more analysis. This study contains a detailed method and measurements of LST, NIR and SWIR reflectance of soybean, corn and barley taken in field campaigns in central Argentine Pampas and laboratory with a ST PRO Raytek (8–14 µm) and a spectrometer SVC HR-1024i (0.35 and 2.5 µm). Also, relative water content of leaves was measured in laboratory during the dehydration process. This method and dataset could be also used for researching other wavelengths between 0.35 and 2.5 µm as indicator of water vegetation status (e.g. solar-induced chlorophyll fluorescence, photosynthesis). • Procedures useful to measure field spectra of vegetation are presented. • Not only the traditional method to measure leaves spectra in laboratory, but also in field were applied. • The method allows the integration of spectra and thermal data as a proxy of vegetation water status.

Currently, quantifying global solar radiation at surface in Argentina is crucial for the development of projects related to solar energy, calculation of evapotranspiration and eco-sustainability architecture, among other environmental issues. In recent years, several models have been developed to estimate the solar energy resources by means of various techniques, e.g. satellite imaging, kriging, or Artificial Neural Networks. The use of satellite data allows for a better spatial representation, being of great relevance in areas with lack of terrain measurements. In this paper, we use the CERES_SYN1deg to develop a new Global Solar Radiation Atlas for Argentina. In this study, we developed maps of annual and monthly mean daily global solar radiation using CERES_SYN1deg data between 2000 and 2016. In order to validate the global solar radiation data provided by CERES_SYN1deg, they were compared with ground-based measurements in the time overlap of both instruments, in four monitoring sites of the SAVER-Net project and an additional site in Tandil, which belongs to the Remote Sensing Group of IHLLA. The maps show the spatial and temporal variation of global solar radiation in Argentina. Comparisons with ground-based pyranometers reveal relative differences of around 3% at a monthly scale for all sites, while the biases can be neglected. Therefore, it is possible to conclude that the maps could be very useful for different technical and scientific purposes, and the comparison with ground-based data demonstrates CERES_SYN1deg’s reliability. Shallow Pampean lakes are located in the most productive plain of Argentina. They are highly variable in salinity, turbidity and surface area. Laguna Chascomús has been monitored as a representative example of them. We developed a linear model based on satellite images validated against field measurements (2001–2011 period). A vegetation index and Landsat Surface Reflectance (Band 4) produced the best correlations with chlorophyll-a (Chl-a) and Secchi Disk Depth (SDD), respectively. In a second instance, a retrospective analysis (1986–2013) was performed. As a result, significant positive trends were observed for SDD and Chl-a. In addition, both variables displayed trends related to rainfall and site depth. Keywords: Chlorophyll-a, Turbidity, Remote sensing

Botrytis cinerea is a phytopathogenic fungus responsible for economic losses from USD 10 to 100 billion worldwide. It affects more than 1400 plant species, thus becoming one of the main threats to the agriculture systems. The application of fungicides has for years been an efficient way to control this disease. However, fungicides have negative environmental consequences that have changed popular opinion and clarified the need for more sustainable solutions. Biopesticides are products formulated based on microorganisms (bacteria or fungi) with antifungal activity through various mechanisms. This review gathers the most important mechanisms of antifungal activities and the microorganisms that possess them. Among the different modes of action, there are included the production of diffusible molecules, both antimicrobial molecules and siderophores; production of volatile organic compounds; production of hydrolytic enzymes; and other mechanisms, such as the competition and induction of systemic resistance, triggering an interaction at different levels and inhibition based on complex systems for the production of molecules and regulation of crop biology. Such a variety of mechanisms results in a powerful weapon against B. cinerea; some of them have been tested and are already used in the agricultural production with satisfactory results.

Canola (Brassica napus L. var. oleracea) is the third most common oil-producing crop worldwide after palm and soybean. Canola cultivation requires the use of chemical fertilizers, but the amount required can be reduced by applying plant growth-promoting bacteria (PGPB). Among PGPB, endophytic bacteria have certain advantages as biofertilizers, but canola endophytic bacteria have rarely been studied. In this work, we identified a collection of bacterial endophytes isolated from canola roots using MALDI-TOF MS, a technique that is still rarely used for the identification of such bacteria, and rrs gene sequencing, a methodology that is commonly used to identify canola endophytes. The results demonstrated that some bacterial isolates from canola roots belonged to the genera Bacillus, Neobacillus, Peribacillus (Pe.), and Terribacillus, but most isolates belonged to the genera Paenibacillus (P.) and Pseudomonas (Ps.). Inoculation of these isolates indicated that several of them could efficiently promote canola seedling growth in hydroponic conditions. These results were then confirmed in a microcosm experiment using agricultural soil, which demonstrated that several isolates of Pseudomonas thivervalensis, Paenibacillus amylolyticus, Paenibacillus polymyxa, Paenibacillus sp. (Paenibacillus glucanolyticus/Paenibacillus lautus group), and Peribacillus simplex (previously Bacillus simplex) could efficiently promote canola shoot growth under greenhouse conditions. Among them, the isolates of Paenibacillus and Peribacillus were the most promising biofertilizers for canola crops as they are sporulated rods, which is an advantageous trait when formulating biofertilizers.

Pseudomonas is a large and diverse genus broadly distributed in nature. Its species play relevant roles in the biology of earth and living beings. Because of its ubiquity, the number of new species is continuously increasing although its taxonomic organization remains quite difficult to unravel. Nowadays the use of genomics is routinely employed for the analysis of bacterial systematics. In this work, we aimed to investigate the classification of species of the genus Pseudomonas on the basis of the analyses of the type strains whose genomes are currently available. Based on these analyses, we propose the creation of three new genera (Denitrificimonas gen nov. comb. nov., Neopseudomonas gen nov. comb. nov. and Parapseudomonas gen nov. comb. nov) to encompass several species currently included within the genus Pseudomonas and the reclassification of several species of this genus in already described taxa.

Salinity is one of the main causes of abiotic stress in plants, resulting in negative effects on crop growth and yield, especially in arid and semi-arid regions. The effects of salinity on plant growth mainly generate osmotic stress, ion toxicity, nutrient deficiency, and oxidative stress. Traditional approaches for the development of salt-tolerant crops are expensive and time-consuming, as well as not always being easy to implement. Thus, the use of plant growth-promoting bacteria (PGPB) has been reported as a sustainable and cost-effective alternative to enhance plant tolerance to salt stress. In this sense, this review aims to understand the mechanisms by which PGPB help plants to alleviate saline stress, including: (i) changes in the plant hormonal balance; (ii) release of extracellular compounds acting as chemical signals for the plant or enhancing soil conditions for plant development; (iii) regulation of the internal ionic content of the plant; or iv) aiding in the synthesis of osmoprotectant compounds (which reduce osmotic stress). The potential provided by PGPB is therefore an invaluable resource for improving plant tolerance to salinity, thereby facilitating an increase in global food production and unravelling prospects for sustainable agricultural productivity.

Trifolium rubens L. is a leguminous plant “Preferential Attention”, according to the Catalog of Protected Flora of Castile and Leon (Spain). In this study we aimed to analyze the potential of three bacterial strains of the genus Rhizobium to improve the growth and development of this plant. All three strains produced 3-indoleacetic acid (IAA), but the strain ATCC 14480 produced the most. In addition, all strains produced biofilms and cellulases, although in different quantities. The synthesis of these products has been described as being related to the processes of the adherence of bacteria to the plant root surface and their entrance into the plant, respectively. In addition, in vitro assays and assays conducted under controlled and sterile conditions were performed, showing that the three strains were capable of nodulating T. rubens L. and effectively fixed nitrogen for the plant. These results were corroborated by morphological and histological analysis of nodules. Finally, greenhouse assays determined the effects of the strains under more competitive conditions, and it was concluded that inoculated plants presented greater lengths and weights, both aerial and radicular, and also chlorophyll and nitrogen content compared to the uninoculated controls.

The negative effects on the environment and human health caused by the current farming systems based on the overuse of chemical fertilizers have been reported in many studies. By contrast, bacterial inoculations produce positive effects on yields without causing this type of harm. Hence, during recent years, the commercialization of biofertilizers has been on the increase, and the number of companies and products available are expanding worldwide every year. In addition to the notable enhancement of crop production, many studies have shown how the application of bacteria has positive effects on food quality such as improved vitamin, flavonoid and antioxidant content, among other benefits. This advantage is interesting with respect to food that is consumed raw, such as fruits and many vegetables, as these bioactive molecules are maintained up until the moment the food is consumed. As regards this review focuses on the collection of studies that demonstrate that microorganisms can act as plant probiotics of fruit and horticultural crops, essential types of food that form part of a healthy diet.

The phyllosphere supports a large and complex bacterial community that varies both across plant species and geographical locations. Phyllosphere bacteria can have important effects on plant health. The sweet chestnut (Castanea sativa Mill.) is an economically important tree species affected worldwide by the fungal pathogens Cryphonectria parasitica and Phytophthora cinnamomi. We examined the culturable phyllosphere bacterial community of the sweet chestnut at two nearby locations in Central Spain in order to know its geographical variability and to explore its potential as source of biological control agents against these two pathogenic fungi. The bacterial diversity at strain level was high but it varied significantly between locations; however, phylotype richness and diversity were more comparable. The isolates were affiliated with the phyla Actinobacteria, Firmicutes and Proteobacteria. Most of them were members of recognized bacterial species, with a notable proportion of representative of the genera Dietzia and Lonsdalea, but a small fraction of the strains revealed the existence of several potential novel species or even genera. Antagonism tests showed the occurrence in the chestnut phyllosphere of bacterial strains potentially useful as biological control agents against the two pathogenic fungi, some of which belong to species never before described as fungal antagonists. Chestnut phyllosphere, therefore, contains a great diversity of culturable bacteria and may represent an untapped source of potential biocontrol agents against the fungi causing blight and ink diseases of this tree species.

Optimization of field techniques is crucial to ensure high quality soil moisture data. The aim of the work is to present a sampling method for undisturbed soil and soil water content to calibrated soil moisture probes, in a context of the SMOS (Soil Moisture and Ocean Salinity) mission MIRAS Level 2 soil moisture product validation in Pampean Region of Argentina. The method avoids soil alteration and is recommended to calibrated probes based on soil type under a freely drying process at ambient temperature. A detailed explanation of field and laboratory procedures to obtain reference soil moisture is shown. The calibration results reflected accurate operation for the Delta-T thetaProbe ML2x probes in most of analyzed cases (RMSE and bias ≤ 0.05 m3/m3). Post-calibration results indicated that the accuracy improves significantly applying the adjustments of the calibration based on soil types (RMSE ≤ 0.022 m3/m3, bias ≤ −0.010 m3/m3). • A sampling method that provides high quality data of soil water content for calibration of probes is described. • Importance of calibration based on soil types. • A calibration process for similar soil types could be suitable in practical terms, depending on the required accuracy level.

The legume nodules are a rich source not only of rhizobia but also of endophytic bacteria exhibiting plant growth-promoting mechanisms with potential as plant biostimulants. In this work we analyzed the genomes of Phyllobacterium endophyticum PEPV15 and Rhizobium laguerreae PEPV16 strains, both isolated from Phaseolus vulgaris nodules. In silico analysis showed that the genomes of these two strains contain genes related to N-acyl-homoserine lactone (AHL) and cellulose biosynthesis, involved in quorum sensing and biofilm formation, which are essential for plant colonization. Several genes involved in plant growth promotion such as those related to phosphate solubilization, indole acetic acid production, siderophore biosynthesis and nitrogen fixation were also located in both genomes. When strains PEPV15 and PEPV16 were inoculated in lettuce and carrot in field assays, we found that both significantly increased the yield of lettuce shoots and carrot roots by more than 20% and 10%, respectively. The results of this work confirmed that the genome mining of genes involved in plant colonization and growth promotion is a good strategy for predicting the potential of bacterial strains as crops inoculants, opening new horizons for the selection of bacterial strains with which to design new, effective bacteria-based plant biostimulants.

Today there is an urgent need to find new ways to satisfy the current and growing food demand and to maintain crop protection and food safety. One of the most promising changes is the replacement of chemical fertilizers with biofertilizers, which include plant root-associated beneficial bacteria. This work describes and shows the use of B. halotolerans SCCPVE07 and R. laguerreae PEPV40 strains as efficient biofertilizers for escarole crops, horticultural species that are widely cultivated. An in silico genome study was performed where coding genes related to plant growth promoting (PGP) mechanisms or different enzymes implicated in the metabolism of phenolic compounds were identified. An efficient bacterial root colonization process was also analyzed through fluorescence microscopy. SCCPVE07 and PEPV40 promote plant development under normal conditions and saline stress. Moreover, inoculated escarole plants showed not only an increase in potassium, iron and magnesium content but also a significant improvement in protocatechuic acid, caffeic acid or kaempferol 3-O-glucuronide plant content. Our results show for the first time the beneficial effects in plant development and the food quality of escarole crops and highlight a potential and hopeful change in the current agricultural system even under saline stress, one of the major non-biological stresses.

Symbiosis between microbes and insects has been raised as a promising area for understanding biological implications of microbe–host interactions. Among them, the association between fungi and bark beetles has been generally recognized as essential for the bark beetle ecology. However, many works investigating bark beetle bacterial communities and their functions usually meet in a common finding: Pseudomonas is a broadly represented genus within this holobiont and it may provide beneficial roles to its host. Thus, we aimed to review available research on this microbe–host interaction and point out the probable relevance of Pseudomonas strains for these insects, in order to guide future research toward a deeper analysis of the importance of these bacteria for the beetle’s life cycle.

Pisum sativum L. (pea) is one of the most cultivated grain legumes in European countries due to the high protein content of its seeds. Nevertheless, the rhizobial microsymbionts of this legume have been scarcely studied in these countries. In this work, we analyzed the rhizobial strains nodulating the pea in a region from Northwestern Spain, where this legume is widely cultivated. The isolated strains were genetically diverse, and the phylogenetic analysis of core and symbiotic genes showed that these strains belong to different clusters related to R. laguerreae sv. viciae. Representative strains of these clusters were able to produce cellulose and cellulases, which are two key molecules in the legume infection process. They formed biofilms and produced acyl-homoserine lactones (AHLs), which are involved in the quorum sensing regulation process. They also exhibited several plant growth promotion mechanisms, including phosphate solubilization, siderophore, and indole acetic acid production and symbiotic atmospheric nitrogen fixation. All strains showed high symbiotic efficiency on pea plants, indicating that strains of R. laguerreae sv. viciae are promising candidates for the biofertilization of this legume worldwide.

Rapeseed (Brassica napus L.) is an important crop worldwide, due to its multiple uses, such as a human food, animal feed and a bioenergetic crop. Traditionally, its cultivation is based on the use of chemical fertilizers, known to lead to several negative effects on human health and the environment. Plant growth-promoting bacteria may be used to reduce the need for chemical fertilizers, but efficient bacteria in controlled conditions frequently fail when applied to the fields. Bacterial endophytes, protected from the rhizospheric competitors and extreme environmental conditions, could overcome those problems and successfully promote the crops under field conditions. Here, we present a screening process among rapeseed bacterial endophytes to search for an efficient bacterial strain, which could be developed as an inoculant to biofertilize rapeseed crops. Based on in vitro, in planta, and in silico tests, we selected the strain Pseudomonas brassicacearum CDVBN10 as a promising candidate; this strain produces siderophores, solubilizes P, synthesizes cellulose and promotes plant height in 5 and 15 days-post-inoculation seedlings. The inoculation of strain CDVBN10 in a field trial with no addition of fertilizers showed significant improvements in pod numbers, pod dry weight and shoot dry weight. In addition, metagenome analysis of root endophytic bacterial communities of plants from this field trial indicated no alteration of the plant root bacterial microbiome; considering that the root microbiome plays an important role in plant fitness and development, we suggest this maintenance of the plant and its bacterial microbiome homeostasis as a positive result. Thus, Pseudomonas brassicacearum CDVBN10 seems to be a good biofertilizer to improve canola crops with no addition of chemical fertilizers; this the first study in which a plant growth-promoting (PGP) inoculant specifically designed for rapeseed crops significantly improves this crop’s yields in field conditions.

European Bark Beetle Ips typographus is a secondary pest that affects dead and weakened spruce trees (Picea genus). Under certain environmental conditions, it has massive outbreaks, resulting in the attacks of healthy trees, becoming a forest pest. It has been proposed that the bark beetle’s microbiome plays a key role in the insect’s ecology, providing nutrients, inhibiting pathogens, and degrading tree defense compounds, among other probable traits yet to be discovered. During a study of bacterial associates from I. typographus, we isolated three strains identified as Pseudomonas from different beetle life stages. A polyphasic taxonomical approach showed that they belong to a new species for which the name Pseudomonas typographi sp nov. is proposed. Genome sequences show their potential to hydrolyze wood compounds and synthesize several vitamins; screening for enzymes production was verified using PNP substrates. Assays in Petri dishes confirmed cellulose and xylan hydrolysis. Moreover, the genomes harbor genes encoding chitinases and gene clusters involved in the synthesis of secondary metabolites with antimicrobial potential. In vitro tests confirmed the capability of the three P. typographi strains to inhibit several Ips beetles’ pathogenic fungi. Altogether, these results suggest that P. typographi aids I. typographi nutrition and resistance to fungal pathogens.

Lettuce (Lactuca sativa L.) is a widely consumed horticultural species. Its significance lies in a high polyphenolic compound content, including phenolic acids and flavonols. In this work, we have probed the ability of Rhizobium laguerreae HUTR05 to promote lettuce growth, under in vitro and greenhouse conditions (both non-saline and saline conditions). This strain has shown several in vitro plant growth promotion mechanisms, as well as capacity to colonize lettuce seedlings roots. We have analyzed the effect of the rhizobacterium inoculation on mineral and bioactive compounds in lettuce, under greenhouse conditions, and found a rise in the content of certain phenolic acids and flavonoids, such as derivatives of caffeoyl acid and quercetin. The genome analysis of the strain has shown the presence of genes related to plant growth-promoting rhizobacteria (PGPR) mechanisms, defense from saline stress, and phenolic compound metabolism (such as naringenin-chalcone synthase or phenylalanine aminotransferase).

Antimicrobial resistance is a worldwide problem that threatens the effectiveness of treatments for microbial infection. Consequently, it is essential to study unexplored niches that can serve for the isolation of new microbial strains able to produce antimicrobial compounds to develop new drugs. Bark beetles live in phloem of host trees and establish symbioses with microorganisms that provide them with nutrients. In addition, some of their associated bacteria play a role in the beetle protection by producing substances that inhibit antagonists. In this study the capacity of several bacterial strains, isolated from the bark beetles Ips acuminatus, Pityophthorus pityographus Cryphalus piceae, and Pityogenes bidentatus, to produce antimicrobial compounds was analyzed. Several isolates exhibited the capacity to inhibit Gram-positive and Gram-negative bacteria, as well as fungi. The genome sequence analysis of three Pseudomonas isolates predicted the presence of several gene clusters implicated in the production of already described antimicrobials and moreover, the low similarity of some of these clusters with those previously described, suggests that they encode new undescribed substances, which may be useful for developing new antimicrobial agents. Moreover, these bacteria appear to have genetic machinery for producing antitumoral and antiviral substances. Finally, the strain IA19T showed to represent a new species of the genus Pseudomonas. The 16S rRNA gene sequence analysis showed that its most closely related species include Pseudomonas lutea, Pseudomonas graminis, Pseudomonas abietaniphila and Pseudomonas alkylphenolica, with 98.6, 98.5 98.4, and 98.4% identity, respectively. MLSA of the housekeeping genes gyrB, rpoB, and rpoD confirmed that strain IA19T clearly separates from its closest related species. Average nucleotide identity between strains IA19T and P. abietaniphila ATCC 700689T, P. graminis DSM 11363T, P. alkylphenolica KL28T and P. lutea DSM 17257T were 85.3, 80.2, 79.0, and 72.1%, respectively. Growth occurs at 4-37°C and pH 6.5-8. Optimal growth occurs at 28°C, pH 7–8 and up to 2.5% NaCl. Respiratory ubiquinones are Q9 (97%) and Q8 (3%). C16:0 and in summed feature 3 are the main fatty acids. Based on genotypic, phenotypic and chemotaxonomic characteristics, the description of Pseudomonas bohemica sp. nov. has been proposed. The type strain is IA19T (=CECT 9403T = LMG 30182T).

Many rhizospheric bacterial strains possess plant growth-promoting mechanisms. These bacteria can be applied as biofertilizers in agriculture and forestry, enhancing crop yields. Bacterial biofertilizers can improve plant growth through several different mechanisms: (i) the synthesis of plant nutrients or phytohormones, which can be absorbed by plants, (ii) the mobilization of soil compounds, making them available for the plant to be used as nutrients, (iii) the protection of plants under stressful conditions, thereby counteracting the negative impacts of stress, or (iv) defense against plant pathogens, reducing plant diseases or death. Several plant growth-promoting rhizobacteria (PGPR) have been used worldwide for many years as biofertilizers, contributing to increasing crop yields and soil fertility and hence having the potential to contribute to more sustainable agriculture and forestry. The technologies for the production and application of bacterial inocula are under constant development and improvement and the bacterial-based biofertilizer market is growing steadily. Nevertheless, the production and application of these products is heterogeneous among the different countries in the world. This review summarizes the main bacterial mechanisms for improving crop yields, reviews the existing technologies for the manufacture and application of beneficial bacteria in the field, and recapitulates the status of the microbe-based inoculants in World Markets.

Nowadays In this paper, the design of a robust PI controller for effective level control in the second poolof the irrigation main canal Güira de Melena is developed. A mathematical model that adequately describesthe dynamic behavior of the plant under study is obtained from system identification procedure.This model is characterized by its simplicity (first order with time delay). The robustness analysis of thedesigned PI controller vs parametric variations of the obtained mathematical model showed satisfactoryresults, and therefore the implementation of this controller is validated.

El objetivo de este trabajo fue analizar la separabilidad espectral entre cultivos teniendo en cuenta el efecto de las precipitaciones y las características del suelo sobre el desarrollo y crecimiento de los cultivos. El trabajo se realizó en la provincia de La Pampa, Argentina con datos de muestras de girasol, maíz y soja de las campañas agrícolas 2000-2003. Las diferentes campañas fueron seguidas con sensores remotos a partir de la información captada por los sensores MODIS (Moderate Resolution Imaging Spectroradiometer) mediante análisis multitemporal de imágenes de satélite MODIS. Los datos fueron tratados como mediciones repetidas en el tiempo. Los resultados indican que la separabilidad es significativa entre cultivos de soja, respecto de maíz y girasol. Al considerar el efecto debido al suelo se comprueba separabilidad entre girasol y maíz. Por otra parte, las precipitaciones ejercen efecto en el desarrollo de los cultivos contribuyendo a mejorar la separabilidad espectral.

En el presente trabajo se desarrolla el diseño de un controlador robusto de orden fraccional -el cual se denota como FPI, debido a que constituye una generalización del controlador PI convencional- para el control efectivo de la distribución de agua en canales principales de riego caracterizados por presentar una amplia variación en sus parámetros dinámicos. El interés en esta clase de controladores se justifica por hecho de que la dinámica de los canales principales de riego puede cambiar de forma drástica con las variaciones de sus regímenes de descargas. El controlador FPI desarrollado se programó en un PLC SIMATIC S-7 300 y se implementó en tiempo real en el primer tramo del canal principal Imperial de Aragón, perteneciente a la Confederación Hidrográfica del Ebro, España. Se realizaron experimentos comparativos de funcionamiento en tiempo real del controlador FPI diseñado con otro controlador PI equivalente, en el sentido de exhibir ambos el mismo comportamiento dinámico en lazo cerrado para las especificaciones temporales de la planta nominal. Los resultados obtenidos de estos experimentos demuestran que cuando los parámetros dinámicos del tramo del canal presentan una amplia variación, el controlador FPI exhibe una mayor robustez, lo cual posibilita lograr una mayor velocidad de respuesta, satisfacer de forma operativa las demandas de los usuarios y minimizar las pérdidas por concepto de operación de los recursos hidráulicos disponibles.

Plants harbor a diversity of microorganisms constituting the plant microbiome. Many bioinoculants for agricultural crops have been isolated from plants. Nevertheless, plants are an underexplored niche for the isolation of microorganisms with other biotechnological applications. As a part of a collection of canola endophytes, we isolated strain CDVBN77T. Its genome sequence shows not only plant growth-promoting (PGP) mechanisms, but also genetic machinery to produce secondary metabolites, with potential applications in the pharmaceutical industry, and to synthesize hydrolytic enzymes, with potential applications in biomass degradation industries. Phylogenetic analysis of the 16S rRNA gene of strain CDVBN77T shows that it belongs to the genus Microvirga, its closest related species being M. aerophila DSM 21344T (97.64% similarity) and M. flavescens c27j1T (97.50% similarity). It contains ubiquinone 10 as the predominant quinone, C19:0 cycloω8c and summed feature 8 as the major fatty acids, and phosphatidylcholine and phosphatidylethanolamine as the most abundant polar lipids. Its genomic DNA G+C content is 62.3 (mol %). Based on phylogenetic, chemotaxonomic, and phenotypic analyses, we suggest the classification of strain CDVBN77T within a new species of the genus Microvirga and propose the name Microvirga brassicacearum sp. nov. (type strain CDVBN77T = CECT 9905T = LMG 31419T).

El conocimiento a escala diaria de la radiación neta (Rn) permite cuantificar la energía que es utilizada en los diferentes procesos que ocurren a nivel de la superficie, como la evapotranspiración. En este estudio se aplica un Modelo de Regresión Lineal Múltiple (MRLM) para la estimación de la Rn en una zona subhúmeda-húmeda de Argentina. En el modelo se utilizaron datos meteorológicos de radiación solar global o total, temperatura, humedad relativa del aire, radiación neta (medida con un radiómetro neto Kipp & Zonen) y el valor del inverso de la distancia relativa tierra-sol o factor de excentricidad. Como resultado, se obtuvieron ocho ecuaciones de estimación de la Rn. Los MRLM se evaluaron a partir de los estadísticos desviación media del error (MBE) y raíz cuadrada del cuadrado medio del error (RMSE). Los resultados mostraron un buen ajuste y un bajo error a escala diaria, destacándose los modelos que involucraron la radiación solar, temperatura, humedad relativa del aire e inverso de la distancia tierra-sol, permitiendo cálculos de la Rn con errores inferiores a 19 W·m-2.

Se propone una modificación de la estructura del predictor de Smith mediante un modelo interno que posibilita aumentar su rechazo al efecto de las per-turbaciones externas no medibles en comparación con la estructura clásica del predictor de Smith. Los resultados obtenidos se aplican en el diseño de un sistema de control del proceso de variación de la temperatura del jugo en los calentadores de una fábrica de azúcar. Los resultados de la simulación del sistema diseñado mostraron su efectividad y robustez en cuanto al rechazo de perturbaciones externas no medibles. The increasing interest in the preservation of the environment and the health of consumers is changing production methods and food consumption habits. Functional foods are increasingly demanded by consumers because they contain bioactive compounds involved in health protection. In this sense biofertilization using plant probiotics is a reliable alternative to the use of chemical fertilizers, but there are few studies about the effects of plant probiotics on the yield of functional fruits and, especially, on the content of bioactive compounds. In the present work we reported that a strain of genus Phyllobacterium able to produce biofilms and to colonize strawberry roots is able to increase the yield and quality of strawberry plants. In addition, the fruits from plants inoculated with this strain have significantly higher content in vitamin C, one of the most interesting bioactive compounds in strawberries. Therefore the use of selected plant probiotics benefits the environment and human health without agronomical losses, allowing the production of highly functional foods.

Bacteria belonging to the family Rhizobiaceae may establish beneficial or harmful relationships with plants. The legume endosymbionts contain nod and nif genes responsible for nodule formation and nitrogen fixation, respectively, whereas the pathogenic strains carry vir genes responsible for the formation of tumors or hairy roots. The symbiotic and pathogenic strains currently belong to different species of the genus Rhizobium and, until now, no strains able to establish symbiosis with legumes and also to induce tumors or hairy roots in plants have been reported. Here, we report for the first time the occurrence of two rhizobial strains (163C and ATCC11325T) belonging to Rhizobium rhizogenes able to induce hairy roots or tumors in plants and also to nodulate Phaseolus vulgaris under natural environmental conditions. Symbiotic plasmids (pSym) containing nod and nif genes and pTi- or pRi-type plasmids containing vir genes were found in these strains. The nodD and nifH genes of the strains from this study are phylogenetically related to those of Sinorhizobium strains nodulating P. vulgaris. The virA and virB4 genes from strain 163C are phylogenetically related to those of R. tumefaciens C58, whereas the same genes from strain ATCC 11325T are related to those of hairy root-inducing strains. These findings may be of high relevance for the better understanding of plant-microbe interactions and knowledge of rhizobial phylogenetic history.

The biofertilization of crops with plant-growth-promoting microorganisms is currently considered as a healthy alternative to chemical fertilization. However, only microorganisms safe for humans can be used as biofertilizers, particularly in vegetables that are raw consumed, in order to avoid sanitary problems derived from the presence of pathogenic bacteria in the final products. In the present work we showed that Rhizobium strains colonize the roots of tomato and pepper plants promoting their growth in different production stages increasing yield and quality of seedlings and fruits. Our results confirmed those obtained in cereals and alimentary oil producing plants extending the number of non-legumes susceptible to be biofertilized with rhizobia to those whose fruits are raw consumed. This is a relevant conclusion since safety of rhizobia for human health has been demonstrated after several decades of legume inoculation ensuring that they are optimal bacteria for biofertilization.

Tres métodos de estimación de la evapotranspiración (ET) se analizan, Priestley y Taylor (PT), Turc (T) y Hargreaves (HG), siendo el objetivo evaluar la evapotranspiración estimada con estas metodologías en relación al método de referencia FAO 56 Penman Monteith (FAO 56 PM). La evaluación se realiza a partir de un análisis estadístico de valores de ET para estaciones ubicadas en zonas de clima húmedo, semiárido y árido de Argentina. Los resultados muestran que las fórmulas con mejor performance en la zona húmeda son las de PT y T. En cambio, en las zonas semiárida y árida la ecuación de HG logra una mejor aproximación al método FAO 56 PM. Se concluye que la metodología de HG proporciona valores confiables de ET para zonas de clima semiárido y árido a lo que se suma que dicha metodología es de sencilla aplicación y requiere como datos de entrada de la ecuación valores de temperatura máxima y mínima. Respecto a la zona de clima húmedo, la ecuación de PT y en segundo lugar la de T, presentan una mejor performance, destacando que ambas son metodologías prácticas y de menor complejidad.

Family Rhizobiaceae includes fast growing bacteria currently arranged into three genera, Rhizobium, Ensifer and Shinella, that contain pathogenic, symbiotic and saprophytic species. The identification of these species is not possible on the basis of physiological or biochemical traits and should be based on sequencing of several genes. Therefore alternative methods are necessary for rapid and reliable identification of members from family Rhizobiaceae. In this work we evaluated the suitability of Matrix-Assisted Laser Desorption Ionization-Time-of-Flight Mass Spectrometry (MALDI-TOF MS) for this purpose. Firstly, we evaluated the capability of this methodology to differentiate among species of family Rhizobiaceae including those closely related and then we extended the database of MALDI Biotyper 2.0 including the type strains of 56 species from genera Rhizobium, Ensifer and Shinella. Secondly, we evaluated the identification potential of this methodology by using several strains isolated from different sources previously identified on the basis of their rrs, recA and atpD gene sequences. The 100% of these strains were correctly identified showing that MALDI-TOF MS is an excellent tool for identification of fast growing rhizobia applicable to large populations of isolates in ecological and taxonomic studies.

En la denominada Pampa Deprimida el relieve es sumamente llano y abundan pequeñas depresiones que, dadas las características de subhúmeda a húmeda de la llanura, están colmadas de agua gran parte del año y marcan en el paisaje un sinnúmero de cuerpos de agua de variadas dimensiones y de profundidades inferiores a los 2 metros. Estas depresiones juegan un papel aparentemente muy importante aunque aún no dilucidado en el balance hídrico del acuífero freático. Para analizar el funcionamiento hidrológico de estas depresiones se ha seleccionado un cuerpo de agua en el que se registran su nivel y el de las aguas subterráneas, el flujo a través del fondo de la depresión, los contenidos iónicos de las aguas y las variables climáticas. Los niveles de la laguna están casi continuamente por encima del nivel freático. Esto es especialmente notable cuando los niveles permanecen relativamente bajos. Cuando se producen importantes precipitaciones los niveles se elevan significativamente, alcanzando la laguna en varias oportunidades su nivel de descarga. Es en esos cortos períodos en que el nivel freático puede superar a la laguna. El análisis a partir de los valores del flujo a través de los sedimentos del fondo del cuerpo de agua permitió obtener un valor de conductividad hidráulica de dichos sedimentos de 1,6*10-4 m/día. También se han obtenido valores de los exponentes de las ecuaciones de agotamiento de la laguna y del agua subterránea.

En varias regiones del mundo existen paisajes de escaso relieve superficial, que poseen características hidrológicas poco convencionales. Aunque existe una estricta dependencia del clima, las llanuras que se desarrollan bajo condiciones semi-húmedas son las que ofrecen los mayores enigmas en cuanto al comportamiento de sus recursos hídricos. Este trabajo presenta las conclusiones de estudios llevados a cabo en la llanura central de la Provincia de Buenos Aires, específicamente la correspondiente a la cuenca del arroyo del Azul. Se presentan evidencias de la regionalización lograda a partir del uso combinado de SIG y del análisis multivariado. Por métodos independientes se demuestra que las formas simples de cálculo de la evapotranspiración real no se ajustan a la realidad. De igual modo, se demuestra el rol fundamental de la zona no saturada en cuanto a su incidencia en la recarga (sujeta a eventos especiales de lluvia y de estado de humedad antecedente, y a la existencia de macroporos). Finalmente, se revela las relaciones hidrológicas entre una laguna y las aguas subterránea donde, a diferencia de lo esperable, existe mayormente un aporte de las aguas superficiales al acuífero.

En el presente trabajo se muestran las características hidrogeológicas generales del partido de Salliqueló, provincia de Buenos Aires, Argentina. Se analizan particularmente las condiciones del flujo del agua subterránea en los ámbitos urbano y periurbano, en las que se detecta una alteración en la calidad natural de las aguas subterráneas en la zona más antigua del ámbito urbano y en los alrededores del basurero municipal, en las cercanías de la ciudad. Por otra parte, se realiza una regionalización del partido desde el punto de vista ecológico atendiendo a variables hidrológicas, edáficas, topológicas y biológicas, utilizando para ello los Sistemas de Información Geográfica. Las regiones se obtuvieron por medio del llamado Método de Evaluación Multicriterio, que es una combinación lineal ponderada de las variables analizadas, a la que se llegó mediante una comparación de a pares de las variables en el marco de un proceso de construcción de decisiones denominado Proceso Analítico Jerárquico. La aplicación de este procedimiento resultó en la obtención de cuatro regiones homogéneas.
