Photothermal therapy (PTT) is widely used in wound healing because it produces heat under near‐infrared irradiation, eliminating bacterial infections. However, excessive production of reactive oxygen species (ROS) occurs during PTT, which causes oxidative stress damage. In this study, a nanozyme, PDA‐MnO2 (PM), is synthesized and encapsulated in the needle tip layer of a bi‐layer GelMA microneedle patch (PbMP), which has excellent mechanical properties and can easily penetrate the skin. In a study involving an SD rat wound model infected with S. aureus, the PbMP significantly accelerated the healing process of bacterially challenged wounds, resulting in better blood vessel growth, cell proliferation, and collagen biosynthesis than other wound dressings. The experiment shows that the PM nanoparticles in PbMP improve photothermal conversion efficiency and activate SOD and CAT, resulting in an efficient cascade enzyme reaction system to eliminate ROS during the PTT process. Abstract This study investigated postharvest dopamine treatment efficiency in ameliorating chilling injury of banana fruits during storage at 7 ºC for 21 days. Our results showed that dopamine treatment at 150 µM promoted phenols and flavonoids biosynthesis acquired by higher phenylalanine ammonia-lyase (PAL) expression and activity concurrent with lower polyphenol oxidase (PPO) expression and activity leading to higher DPPH, FRAP, and ABTS radicals scavenging activity. In addition, dopamine treatment at 150 µM promoted endogenous proline biosynthesis by activating pyrroline-5-carboxylate synthetase (P5CS) and ornithine δ-aminotransferase (OAT) expression and activity concurrent with suppressing proline dehydrogenase (ProDH) expression and activity. Furthermore, higher endogenous γ-aminobutyric acid (GABA) biosynthesis in banana fruits by 150 µM dopamine treatment was accompanied by higher glutamate decarboxylase (GAD) and GABA transaminase (GABA-T) expression and activity. Therefore, our results suggest that dopamine treatment at 150 µM might be employed for banana fruits chilling injury amelioration by enhancing phenylpropanoid pathway activity and boosting endogenous proline and GABA biosynthesis.

The CCCH zinc finger proteins play critical roles in a wide variety of growth, development, and stress responses. Currently, limited reports are available about the roles of animal CCCH proteins in plants. In this study, we report the identification of human TTP (hTTP) with functional similarity to PdC3H17 in a hybrid poplar. hTTP and PdC3H17 shared highly similar tandem CCCH zinc-finger RNA-binding domains. The fragments excluding the CCCH domain of both hTTP and PdC3H17 possessed transcriptional activation activities in yeast cells. Compared to the controls, ectopic expression of hTTP in poplar caused dwarfism, and resulted in significant increases in stem xylem vessel number and photosynthetic and ROS-scavenging abilities, thereby enhancing plant tolerance to drought stress. Our results suggest that hTTP may perform a function in poplar through the PdC3H17-mediated system, and provide an example for the application of animal genes in plants.

The stress conditions imposed by the impact of metal and non‐metal oxide nanoparticles over plant systems enhances the synthesis of reactive oxygen species (ROS), resulting in oxidative damage at cellular level. The objective of this study was to synthesise the gold nanoparticles (GNps) from the leaves protein of Nicotiana tabacum L. cv. xanthi, its characterisation, and response on plant physiology and ROS scavenging activity on plants after exposure to different stresses. The authors have treated N. tabacum L. cv. xanthi plants with 100, 200, 300, 400, and 500 ppm biochemically synthesised GNps and examined physiological as well as biochemical changes. Results showed that biochemically synthesised GNps exposure significantly increased the seed germination (P < 0.001), root (P < 0.001), shoot growth (P < 0.001), and antioxidant ability (P < 0.05) of plants depending on bioengineered GNPs concentrations. Low concentrations (200–300 ppm) of GNps boosted growth by ∼50% and significantly increase in photosynthetic parameters such as total chlorophyll content (P < 0.05), membrane ion leakage (P < 0.05) as well as malondialdehyde (P < 0.05) content with respect to untreated plants under stress conditions. The high concentration (400–500 ppm) of GNps affected these parameters in a negative manner. The total antioxidant activity was also elevated in the exposed plants in a dose‐dependent manner. Schisandra rubriflora is a dioecious, underestimated medicinal plant species known from traditional Chinese medicine. The present study was aimed at characterising the polyphenolic profile composition and the related antioxidant capacity of S. rubriflora fruit, stem and leaf and in vitro microshoot culture extracts. Separate analyses of material from female and male specimens were carried out. This study was specifically aimed at detailed characterisation of the contribution of phenolic compounds to overall antioxidant activity using ultra-high-performance liquid chromatography with a photodiode array detector coupled to electrospray ionization ion trap mass spectrometry (UHPLC-DAD-ESI-MS3) and a high-performance liquid chromatography-diode array detector (HPLC-DAD). Using UHPLC-DAD-ESI-MS3, twenty-seven phenolic compounds from among phenolic acids and flavonoids were identified. Concentrations of three phenolic acids (neochlorogenic, chlorogenic and cryptochlorogenic acids) and eight flavonoids (hyperoside, rutoside, isoquercitrin, guaijaverin, trifolin, quercetin, kaempferol, and isorhamnetin) were determined using HPLC-DAD using reference standards. The highest total phenolic content was confirmed for the stem and leaf extracts collected in spring. The contents of phenolic compounds of in vitro biomasses were comparable to that in the fruit extracts. The methanolic extracts from the studied plant materials were evaluated for their antioxidant properties using various in vitro assays, namely free radicals scavenging estimation using 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH), ferric-reducing antioxidant power (FRAP) and cupric-reducing antioxidant capacity (CUPRAC) as well as QUick, Easy, New, CHEap, and Reproducible CUPRAC (QUENCHER-CUPRAC) assays. A close relationship between the content of polyphenolic compounds in S. rubriflora and their antioxidant potential has been documented.

We synthesized a series of S-methyl-5-bromosalicylidene-N-alkyl substituted thiosemicarbazones and their cis-dioxomolybdenum(VI) complexes having long alkyl chains (propyl, butyl, pentyl, hexyl, and octyl) on thioamide nitrogen. The compounds were characterized by using analytical and spectroscopic methods. The structure of the complex with pentyl group (complex 3 ) as a representative molecule was determined by X-ray single-crystal diffraction method. The free ligand and their dioxomolybdenum(VI) complexes have been tested for in vitro antioxidant capacity by reduction of copper (II) neocuproine (Cu(II)-Nc) using the CUPRAC (CUPric Reducing Antioxidant Capacity) method. The ligands exhibited more potent in vitro antioxidant capacity than that of the complexes. The obtained trolox equivalent antioxidant capacity (TEAC) value of complex 1 (TEAC CUPRAC = 0.73) was higher than those of other complexes. Furthermore, the catalase activity and reactive oxygen species (ROS) scavenging ability of the free ligands and their complexes were determined, showing that complex 1 had significant scavenging activity for ROS . The condensation of taxifolin with glyoxylic acid was examined, and the properties of the resulting product were compared with those of taxifolin. The structure of the product was determined by NMR spectroscopy. The ability of the polyphenols to scavenge reactive oxygen species (ROS) was estimated by luminol-dependent chemiluminescence. The iron-chelating and iron-reducing activities were studied using absorption spectrophotometry. It was shown that the condensation leads to the formation of a dimer consisting of two taxifolin units linked through a carboxymethine bridge at the C-6 and C-8 positions of the A ring. The dimer exhibited a somewhat higher ROS scavenging activity than taxifolin. The iron-binding capacity of the compounds was proportional to the number of polyphenol units. The iron-reducing ability of the dimer was lower than that of taxifolin. Thus, the dimer possessed a higher antioxidant activity than the parent flavonoid. The data obtained may be useful for a better understanding of processes occurring in foods and beverages and in a search for new active compounds. [ABSTRACT FROM AUTHOR]

The aim of this study is to evaluate the effect of spent coffee ground (SCG) ethanol extract on UVB-induced skin aging in hairless mice. An ethanol extract of SCG (ESCG) was prepared using the residue remaining after extraction of oil from roasted SCG. High performance liquid chromatography (HPLC) analysis showed that the content of caffeine (41.58 ± 0.54 μg/mg) was higher than that of chlorogenic acid isomers (~ 9.17 μg/mg) in ESCG. ESCG significantly decreased the UVB-induced intracellular reactive oxygen species in HaCaT cells. UVB-induced wrinkle formation in mice dorsal skin was effectively reduced by ESCG administration; high dose of ESCG (5 g/L) caused the reduction of wrinkle area by 30% compared with UVB-treated control (UVBC). This result correlated with the ESCG-mediated decrease in epidermis thickness (25%). In addition, ESCG administration significantly reduced transdermal water loss (20%) and erythema formation (35%) derived from UVB exposure. Collagen type I (COL-1) level in dorsal skin was effectively recovered by ESCG administration. These results were supported by down-regulation of collagen-degrading matrix metalloproteinase 2 (MMP2) and 9 (MMP9) expressions. Our results indicate that ESCG protects mouse skin from UVB-induced photoaging by suppressing the expression of matrix metalloproteinases. Our study suggests that ESCG may be anti-photoaging agent. For the last few decades, peroxidase mimicking inorganic nanoparticles are widely used as compared to natural peroxidase due to some basic advantages especially high durability, stability in harsh conditions, easy and cost-effective synthesis, and easy storage. However, most of these peroxidase mimicking nanoparticles are not always effective in a neutral medium, most of the show high activity in an acidic medium which seriously impedes their uses in bioanalytical applications. In the current study, we have developed an inorganic β-cyclodextrin (β-CD) grafted rhombohedral cupric oxide (R-CuO) nanozyme which showed high catalytic activity at neutral conditions (pH = 7) and also exhibited catalytic activity in a wide range of pH with enzymatic substrate TMB (3,3–5,5 tetramethylbenzidine).The current nanozyme has exhibited outstanding peroxidase mimic activity with low Km and very high Vmax value which was calculated by the double reciprocal Lineweaver Burk plot. Considering the R-CuO NPs as a heterogeneous catalyst, a one-pot simple colorimetric system has been developed for dopamine detection, as dopamine turns off the catalytic activity of the R-CuO NPs-TMB peroxidase system. The sensitivity of this current probe to dopamine was promising with a linear range of 2– 24 µM and had a low detection limit of 1.91 µM. More surprisingly, the R-CuO-TMB /dopamine system undergoes recovery of peroxidase activity upon introduction of Cr (+6) (dichromate).The method displays a good selectivity towards Cr (+6) (dichromate) over all the other metal ions with a wide linear relationship in the range of 2 μM–10 μM, and a low detection limit of 0.47 μM. Moreover, the present Nano system was not only applicable for toxic metal and bioactive molecules detection they also used as a ROS scavenger. Our Nano system exhibited scavenging activity for OH radical, O 2 –. species and metal chelation activity which was investigated calorimetrically. Context: Searching for polymorphonuclear neutrophils (PMNs) respiratory burst inhibitors is an important topic in the treatment of human diseases associated with inflammation. Objective: To investigate the inhibitory effects of phenolics isolated from Artocarpus styracifolius Pierre (Moraceae) on respiratory burst induced by phorbol myristate acetate (PMA). Materials and methods: The anti-respiratory burst activities of eight phenolics (20 µM) were assessed by determining luminol-dependent chemiluminiscence in rat PMNs. Cytotoxicity of active compounds (1-1000 µM) was assayed by Trypan blue dye exclusion method. Cell-free models were employed to evaluate scavenging capacity of active compounds (20 µM) against reactive oxygen species. Results: The PMA-induced respiratory burst was significantly inhibited ( p < 0.05) by six isoprenylated phenolics (AS 1- 6) at the concentration of 20 µM (below the toxic concentration) with the inhibition rate ranging from 25.0 to 99.6%. The inhibitory potency estimated by IC50 was in the order of AS 1 (3.1 µM) >AS 6 (5.9 µM) >AS 2 (9.1 µM) >AS 3 (10.0 µM) >AS 5 (29.7 µM) >AS 4 (57.7 µM). AS 1- 4, four isoprenylated flavones, potently quenched superoxide anion, hydroxyl radical, and hydrogen peroxide at the concentration of 20 µM with their scavenging rates in the range of 30.1-78.1%, 35.4-69.7%, and 65.5-86.3%, respectively. In contrast, AS 5- 6, two isoprenylated 2-arylbenzofurans, showed less effect than that exhibited by AS 1- 4. Conclusion and discussion: The isoprenylated phenolics from A. styracifolius can potently inhibit PMA-induced respiratory burst in rat neutrophils without showing cytotoxicity. Abstract: Trimetazidine, the known anti-anginal and anti-ischemic drug, was modified by pyrroline and tetrahydropyridine nitroxides and their hydroxylamine and sterically hindered secondary amine precursors. The synthesized new compounds proved to be better superoxide scavenger molecules compared to the parent Trimetazidine in an in vitro experiment. This reactive oxygen species (ROS) scavenging activity was further supported by ischemia/reperfusion (I/R) studies on Langendorff-perfused rat hearts pretreated with Trimetazidine and with the modified Trimetazidine derivatives before ischemia. Two of the investigated compounds, containing 2,2,5,5-tetramethyl-2,5-dihydro-1H-pyrrole and 4-phenyl-2,2,5,5-tetramethyl-2,5-dihydro-1H-pyrrole substituents on the piperazine ring, provided significant protection from the cardiac dysfunction caused by I/R. Calcium carbonate (CaCO3)-based materials have received notable attention for biomedical applications owing to their safety and beneficial characteristics, such as pH sensitivity, carbon dioxide (CO2) gas generation, and antacid properties. Herein, to additionally incorporate antioxidant and anti-inflammatory functions, we prepared tannylated CaCO3 (TA-CaCO3) materials using a simple reaction between tannic acid (TA), calcium (Ca2+), and carbonate (CO32−) ions. TA-CaCO3 synthesized at a molar ratio of 1:75 (TA:calcium chloride (CaCl2)/sodium carbonate (Na2CO3)) showed 3–6 μm particles, comprising small nanoparticles in a size range of 17–41 nm. The TA-CaCO3 materials could efficiently neutralize the acid solution and scavenge free radicals. In addition, these materials could significantly reduce the mRNA levels of pro-inflammatory factors and intracellular reactive oxygen species, and protect chondrocytes from toxic hydrogen peroxide conditions. Thus, in addition to their antacid property, the prepared TA-CaCO3 materials exert excellent antioxidant and anti-inflammatory effects through the introduction of TA molecules.
