1. Physiological Response of Local and Saanen x Local Cross Goats during the Late Rainy Season in North-eastern Brazil

Abstract

In order to compare in vivo performance and environmental adaptation of two genetic types of goats raised in north-eastern Brazil, 20 females (10 local and 10 Saanen x local) were used in this experiment. During the late rainy season the goats were monitored for the following parameters: body weight, body (BT) and skin temperature (ST), respiratory (RR) and heart rate (HR) as well as the air temperature (AT) and humidity (HU) in the pens in the morning (MR) and afternoon (AR). From May to June the crossbred goats showed a significant gain (P< 0.05); the live weight of F1 was superior to the local goats (P< 0.05) at the end of the experiment. The physiological response during the AR was significantly superior to MR (P< 0.05) for all parameters. The local goats achieved the highest values of BT and ST during the experiment. On average, the physiological rhythms (RR and HR) were higher in crossbred than local goats. In conclusion, crossbred goats showed to be less at risk to environmental stress condition in north-eastern Brazil.

The incidence of DM1 is increasing worldwide, and there is currently no way to prevent or delay the onset or to cure the disease. Most diseases, including diabetes, stem from abnormalities in the functioning of proteins, and some studies have reported the expression of protein variation to be involved in the development of DM1. Thus, the aim of this study was to investigate the differential expression of serum proteins in patients with DM1. Materials and methods: Serum of patients with DM1 (n=30) and healthy controls (n=30) was collected. A proteomic approach was used with depletion of albumin and immunoglobulin G chromatography on serum samples followed by data-independent, label-free mass spectrometric analysis. Results: A total of eight serum proteins were identified as being differentially expressed and involved in the immune system, lipid metabolism, and pathways of coagulation. DM1 was associated with the upregulation of six proteins: alpha-2-macroglobulin, apolipoprotein A-II, β2 glycoprotein I, Ig alpha-2 chain C region, alpha-1-microglobulin, and prothrombin. A total of two proteins were downregulated, including pregnancy zone protein and complement C4. Conclusion: To the best of our knowledge, these findings show differential expression of proteins revealing new proteins that may be involved in the development and progression of diabetes. Abstract

Summary: The present study aimed to use an in vitro follicle culture (IVFC) biotechnique as a tool to evaluate the influence of whole flaxseed as a feed supplementation in the diet on the in vitro development of caprine early antral follicles (EAFs) and further embryo production. In total, 18 adult goats were homogeneously allocated into two diet groups: Control and Flaxseed. EAFs from both experimental groups (300–400 µm) were isolated and cultured in vitro for 18 days. After IVFC, recovered cumulus–oocyte complexes were submitted to in vitro maturation, and subsequently to IVF and in vitro embryo culture. The endpoints evaluated were follicular growth and morphology, oocyte recovery rate and diameter, sperm penetration, pronuclei formation, embryo development, and estradiol production. The addition of the whole flaxseed in the diet did not affect (P > 0.05) follicular growth and diameter. A higher (P < 0.05) percentage of oocytes ≥ 110 µm was recovered from the flaxseed treatment. However, the sperm penetration rate was higher (P < 0.05) in the control treatment when compared with the flaxseed treatment, but no differences were found regarding the rate of fertilization nor cleaved embryos. In conclusion, dietary flaxseed increased the recovery rate of fully grown oocytes, but it did negatively affect the sperm penetration rate, even though there was no further effect on the cleavage rate. Abstract

A expressão de RNAm para leptina, receptor de leptina (obRb), adiponectina, receptor de adiponectina (AdipoR1) e resistina foi avaliada por meio da técnica de PCR em tempo real, em tecidos ovariano, hipofisário, adiposo do omento e da região perirrenal, em ovelhas alimentadas sem farelo de mamona ou com farelo de mamona detoxificada durante 14 meses. O tipo de dieta não afetou os níveis de RNAm para leptina, obRb, adiponectina, AdipoR1 e resistina nos diferentes tecidos avaliados (P>0,05). Nos tecidos ovariano e hipofisário, não foi verificada a expressão da adiponecina e da resistina, respectivamente. Como consequência, pode-se concluir que o farelo de mamona detoxificada pode ser utilizado como fonte proteica na dieta de ovelhas, sem afetar a expressão do gene resistina e dos genes leptina e adiponectina, bem como de seus receptores. The expression of leptin, leptin receptor (obRb), adiponectin, adiponectin receptor (AdipoR1) and resistin was assessed by real-time PCR technique in ovarian, pituitary, and the omental adipose perirenal tissue in sheep feed without castor meal or with detoxified castor meal. The type of diet did not affect mRNA levels for leptin, obRb, adiponectin, resistin AdipoR1 evaluated in different tissues (P>0.05). However, in pituitary and ovarian tissues there was no expression of resistin and adiponectin, respectively. The detoxified castor meal can be used in sheep diets as alternative food protein without affecting the expression of leptin and adponectin as well as their receptors and resistin.

Abstract

In order to verify the oestrus activity and to observe the body weight of Saanen goats, when reared in intensive condition of Northeast Brazil during four different seasons, ten adult Saanen goats were used. In this experiment four climatic seasons were considered; early rainy (ER), late rainy (LR), early dry (ED) and late dry (LD). Oestrus behaviour was monitored twice a day (8:00 a.m. and 4:00 p.m.) during all the experiment, using two vasectomised bucks. The oestrus cycles were quantified within each season and classified as short (< 17 days), normal (17-25 days) or long (> 25 days). The goats were weighed every seven days. During the late rainy season, a significant fall of the mean body weight of the animals was observed when compared to early rainy season. A larger percentage of goats in oestrus were observed during the late rainy season. Oestrus duration was longer in the late rainy and early dry season than in early rainy and late dry season. During the experiment, there were 12.0% of short cycles, 50.3% of normal cycles and 37.7% of long cycles. The percentages of abnormal cycles (short and long cycles), in relation to total cycles monitored in each season were 52, 58, 38 and 50% for early rainy, late rainy, early dry and late dry, respectively. From the results of this study it can be concluded that pen-fed Saanen goats do not show seasonality for oestrus behaviour. Rainfall appeared to be an extero-receptive factor influencing the quality of sexual activity and performance in these goats. Made available in DSpace on 2017-09-10T00:26:20Z (GMT). Abstract

The aim of this study was to evaluate the efficiency of 1 M dimethylsulphoxide (DMSO), ethylene glycol (EG), propylene glycol (PROH) and glycerol (GLY) to cryopreserve primordial follicles. The first evaluation was performed soon after cryopreservation and the second evaluation after 4 days of in vitro culture, using the cryoprotectants that allowed the higher results (higher follicular survival rate) after cryopreservation. The results after follicular isolation (control) and cryopreservation using 1 M DMSO, EG, PROH and GLY showed that the mean number (+/- SEM) of live follicles per millilitre was 3204 (100%) +/- 319.27, 2798 (87%) +/- 239.14, 2492 (78%) +/- 345.8, 448 (14%) +/- 46.3 and 208 (7%) +/- 75.26, respectively. Higher follicular survival was reported when DMSO and EG were used. Control follicles and follicles cryopreserved with these two cryoprotectants were cultured and the percentage of follicular survival was 55% (control), 42% (EG) and 34% (DMSO). Similar results were found between control and follicles cryopreserved with EG. In conclusion, 1 M EG is the most effective cryoprotectant to preserve primordial follicles isolated from ovaries of sheep.

Abstract

The aim of this work was to evaluate the influence of season of birth on the development of the preantral follicular population in Massese lambs. The ovaries of 30 lambs (15-30 days old) were collected at a local slaughterhouse from May to June (M-J) (n = 15) and from October to November (O-N) (n = 15). The research was conducted in Tuscany (Italy), a region characterized by a Mediterranean climate, with a nutritional and photoperiodic peak in the spring (March-June) and a decrease during summer-autumn (August-November). The lamb ovaries were submitted to quantitative and qualitative hystological analyses. Results in the M-J group showed an increase of primary follicles (P < 0.05), and a lower number of primordial populations, compared to the O-N (P < 0.05) group. In the O-N group, qualitative analysis showed a significant (P < 0.05) reduction in follicle size, oocyte size and number of granulosa cells, in the primary and secondary follicle classes, when compared to the M-J group. It was concluded that the autumn season of birth negatively affected the preantral follicular population in Massese lambs. © 2004 Elsevier B.V. All rights reserved.
