Periodontitis is a chronic multifactorial polymicrobial infection, characterized by profound modifications of the composition and proportion of the subgingival microbiota. Microbiological laboratory tests are sometimes used in periodontal diagnosis and monitoring of treatment, but both conventional cultivation methods and molecular techniques have some major drawbacks. Therefore, other performant bacterial identification methods must be considered. The aim of the current study was to use Matrix-assisted Laser Desorption Ionization–Time of Flight Mass Spectrometry (MALTI-TOF MS) analysis in association with bacterial culture method to evaluate the modifications of the subgingival bacterial composition in periodontitis patients, before and after cause-related subgingival therapy. Subgingival plaque samples were collected from periodontal pockets before and after subgingival mechanical instrumentation and adjunctive local antimicrobial applications and were cultured in aerobic and anaerobic conditions. Microbial colonies were further assessed using MALDI-TOF-MS. A total of 36 bacterial strains were isolated from a group of 16 patients. All species from the orange complex were identified by MALDI-TOF MS. A marked reduction of detection frequency was observed in most bacterial strains, including the orange complex after cause-related periodontal treatment. The results of this study indicate that MALDI-TOF MS could be considered an accurate method for oral microbial identification and the cause-related periodontal treatment is useful for reducing the microbial burden.

(1) Background: This cross-sectional investigation appreciated the role of serum C-reactive protein (CRP), several hematologic-cell markers, and salivary inflammation-related molecules [calprotectin (S100A8/A9), interleukin-1β (IL-1β), kallikrein] to predict periodontitis in patients with atherosclerotic cardiovascular disease (ACVD), arrhythmia, or both. Also, we appreciated the relationship between the inflammatory burden and periodontal destruction with the type of cardiac pathology. (2) Methods: Demographic, behavioral characteristics, periodontal indicators, blood parameters, and saliva samples were collected. (3) Results: All 148 patients exhibited stage II or III/IV periodontitis. Stage III/IV cases exhibited significantly increased S100A8/A9 levels (p = 0.004). A positive correlation between S100A8/A9 and IL-1β [0.35 (<0.001)], kallikrein [0.55 (<0.001)], and CRP [0.28 (<0.001)] was observed. Patients with complex cardiac involvement had a significantly higher number of sites with attachment loss ≥ 5 mm [19 (3–30)] compared to individuals with only arrhythmia [9 (3.25–18)] or ACVD [5 (1–12)] [0.048♦ {0.162/0.496/0.14}]. (4) Conclusions: Severe, extensive attachment loss may be indicative of patients with complex cardiac conditions, which underscores the essential role of periodontal status in relation to systemic diseases. Human papillomavirus (HPV) encompasses a diverse array of viruses, comprising approximately 200 serotypes that affect humans. While the majority of HPV strains are associated with benign skin or mucous membrane growths, a subset is implicated in severe health conditions, such as cervical, anal, vulvar, and vaginal cancers. Despite the established effectiveness of HPV vaccines in preventing cervical and anal carcinomas in particular, their therapeutic potential in addressing cutaneous diseases linked to diverse HPV strains remains an intriguing area of investigation. This narrative review critically examines the existing literature to assess the viability of HPV immunization as a therapeutic intervention for prevalent cutaneous conditions. These include genital and extragenital cutaneous warts, epidermodysplasia verruciformis, and keratinocyte carcinomas. The findings suggest a promising dual role for HPV vaccines in preventing and treating dermatologic conditions while emphasizing future research directions, including the immunization perspective against β-HPVs. Moreover, the presence of conflicting study outcomes underscores the imperative for larger-scale, randomized trials with well-matched control groups to validate the efficacy of HPV immunization in the dermatologic context. (1) Background: Periodontitis is a chronic infectious, inflammatory disease associated with significant local disastrous effects due to alveolar bone destruction and systemic morbidities. Some associations between periodontitis and coronary and cerebrovascular diseases have been highlighted in the literature. The present study investigated a group of patients with atherosclerotic cardiovascular diseases and periodontitis and aimed to as-sess the comprehensive effects of nonsurgical periodontitis treatment. (2) Methods: The present study included 11 patients with atherosclerotic cardiovascular disease and perio-dontitis, that were diagnosed after a complete and a complex periodontal evaluation. These patients received periodontal treatment and periodontal reevaluation after 2 months from initial therapy. (3) Results: A significant improvement of the oral hygiene and gingival bleeding scores was recorded at two months follow-up. For moderate pockets of 5 mm, there was a significant reduction of their median values at two months follow-up moment (p=0.021). (4) Conclusion: The nonsurgical periodontal treatment significantly improves the clinical-related parameters of the periodontal tissues, as revealed by gingival bleeding and pocket closure. Objectives. The aim of this study was to evaluate the prevalence of selected bacterial species from periodontal pockets samples in a group of young adult periodontitis patients and explore the additional value of the Real-Time Polymerase Chain Reaction (RT-PCR) assay. Material and Methods. Patients with periodontitis diagnosis and good general health were included in the study after a complete periodontal examination and following the inclusion/exclusion criteria. Samples from the deepest periodontal pockets were collected, and RT-PCR assay was performed in a private laboratory in order to identify eleven bacterial strains associated with periodontitis. The patient’s report provided the bacterial pathogen load quantified according to the detection threshold and classified as negative, weakly positive, positive, or intensely positive. Results. The RT-PCR assay detected periodontal pathogens, and their prevalence could be calculated based on the test report. The red complex bacteria were detected in all patients with the highest counts. Alongside the orange complex bacteria were also detected in periodontitis patients. A. actinomycetemcomitans remained undetected in our study. Conclusions. The RT-PCR technique proved to be a useful and easy approach for identifying and quantifying subgingival periodontal pathogens in periodontitis patients. Periodontitis is a major public health concern because of its high prevalence and due to the significant impact on the masticatory function and aesthetics. Periodontitis can cause social inequality and greatly reduce quality of life. If a proper periodontal treatment is implemented, effective personal plaque management, and a thorough supportive phase, the majority of patients with periodontitis can preserve their natural teeth for an extended period of time. The subgingival mechanical instrumentation is the gold standard of periodontitis treatment and it determines a significant change in the subgingival microbiota. Due to the fact that in some clinical situations subgingival instrumentation is not totally efficient different locally delivered antibiotics can be used as adjunctive therapies to periodontitis treatment. Introduction. This study aimed to evaluate the periodontal status of a group of Romanian systemic sclerosis (SSc) patients and to investigate the relationships between periodontitis and SSc subtypes. Materials and methods. This observational study included patients diagnosed with limited SSc (lcSSc) and diffuse SSc (dcSSc). Demographic data were collected from medical records. Each participant underwent a full-mouth periodontal examination including Bleeding on Probing (BoP) index, Oral Hygiene (OH) index, Probing Depth (PD), Gingival Recession (GR), and Clinical Attachment Loss (CAL). The periodontal status was defined according to presently recognised case definition system. Results. The study included 30 patients with lcSSc and 30 patients with dcSSc with a mean age of 52.45±11.75 years. The overall periodontitis frequence in our SSc group was 95%. The frequency of stage III/IV periodontitis was higher in the dcSSc group (90%) than in the lcSSc group (60%). Within the group of SSc patients, significant positive correlations were observed between age, BoP index, OH index, the number of missing teeth, mean PD, mean CAL on one side and periodontitis diagnosis on the other side (r=0.588, p=0.001; r=0.399, p=0.002; r=0.388, p=0.002; r=0.574, p=0.001; r=0.444, p=0.001; r=0.571, p=0.001). A significant positive correlation existed between the diagnostic of periodontitis and SSc subtypes (r=0.327, p <0.001). Conclusions. Periodontitis was highly prevalent in both lcSSc and dcSSc groups. More stage III/IV periodontitis cases were detected dcSSc group of patients. [ABSTRACT FROM AUTHOR]

Objectives. The objective of the study was to investigate the clinical performance of two different resin-based materials 6 months after their placement in dental non-carious cervical lesions (NCCL). This study also aimed to evaluate the effects of the materials on periodontal tissues in terms of clinical changes of several periodontal parameters. Material and methods. NCCLs characteristics (localization, morphology, dimensions) were preoperatively recorded. The clinical behaviour of the restorations based on the modified United States Public Health Service (USPHS) criteria and periodontal parameters (plaque index, bleeding index, probing depth, and attachment loss) were assessed at baseline and after 1, 3 and 6 months. Results. No significant modifications were recorded for the modified USPHS criteria 6 months after the placement of NCCL restorations excepting postoperative hypersensitivity which was associated with 4 restorations after 6 months. Significant improvements of plaque and bleeding indices were recorded, while no significant modifications were noted for the other periodontal parameters. Conclusions. Considering the clinical success related to the good clinical behaviour and the positive effect of the restorations on the periodontal status, the experimental conventional composites and giomers could be considered as a good therapeutic option for the restoration of NCCLs. Objectives. The aim of this in vitro study was to observe through scanning electron microscopy (SEM) the surface morphology of root samples treated with different mechanical instrumentation methods and the additional application of a nanocolloidal silver-based antiseptic solution. Material and methods. Root samples were prepared from extracted molars and divided in four groups: group 1 of samples was instrumented with Gracey curette (7/8), group 2 of samples was instrumented with ultrasonic tip 1S, group 3 of samples was instrumented with ultrasonic tips 1S plus H4R/L, group 4 of samples was treated with ultrasonic tips 1S plus H4R/L and application of the silver-based antiseptic solution. All samples were then subjected to SEM examination. Outcomes. Group 1 of samples presented a smoother surface compared to the samples in the other groups. Group 2, 3 and 4 displayed superficial grooves parallel to the direction of action of the ultrasonic tip. In group 3 and 4, root planning with tip H4R/L did not ameliorate the smoothness of the surface. The smear layer was present, regardless the instrumentation method. In group 4, the additional application of the antiseptic solution did not influence the surface morphology or the amount of smear layer. Conclusions. Gracey curettes created a smoother radicular surface compared to ultrasonic tips. Smear layer was obvious on the radicular surfaces, no matter the instrumentation method. After tooth extraction due different clinical situations, there are some physiological changes that include volumetric resorption of the bone, leading to changes in the dimensions and contours of the alveolar ridge that can negatively impact the aesthetic outcome of an implant or of a conventional prosthetic rehabilitation. In order to prevent and minimize adverse consequences from such dimensional changes, the present review aims to present some fundamental basis regarding alveolar ridge preservation and biomaterials used to prevent bone resorption.
