2. Endothelial struts enable the generation of large lumenized blood vessels de novo.

Abstract

De novo blood vessel formation occurs through coalescence of endothelial cells (ECs) into a cord-like structure, followed by lumenization either through cell-1-3 or cord-hollowing4-7. Vessels generated in this manner are restricted in diameter to one or two ECs, and these models fail to explain how vasculogenesis can form large-diameter vessels. Here, we describe a model for large vessel formation that does not require a cord-like structure or a hollowing step. In this model, ECs coalesce into a network of struts in the future lumen of the vessel, a process dependent upon bone morphogenetic protein signalling. The vessel wall forms around this network and consists initially of only a few patches of ECs. To withstand external forces and to maintain the shape of the vessel, strut formation traps erythrocytes into compartments to form a rigid structure. Struts gradually prune and ECs from struts migrate into and become part of the vessel wall. Experimental severing of struts resulted in vessel collapse, disturbed blood flow and remodelling defects, demonstrating that struts enable the patency of large vessels during their formation.

16. Implementing norm-governed multi-agent systems

Abstract

The actions and interactions of independently acting agents in a multi-agent system must be managed if the agents are to function effectively in their shared environment. Norms, which define the obligatory, prohibited and permitted actions for an agent to perform, have been suggested as a possible method for regulating the actions of agents. Norms are local rules designed to govern the actions of individual agents whilst also allowing the agents to achieve a coherent global behaviour. However, there appear to be very few instances of norm-governed multi-agent systems beyond theoretical examples. We describe an implementation strategy for allowing autonomous agents to take a set of norms into account when determining their actions. These norms are implemented using directives, which are local rules specifying actions for an agent to perform depending on its current state. Agents using directives are implemented in a simulation test bed, called Sinatra. Using Sinatra, we investigate the ability of directives to manage agent actions. We begin with directives to manage agent interactions. We find that when agents rely on only local rules they will encounter situations where the local rules are unable to achieve the desired global behaviour. We show how a centralised control mechanism can be used to manage agent interactions that are not successfully handled by directives. Controllers, with a global view of the interaction, instruct the individual agents how to act. We also investigate the use of an existing planning tool to implement the resolution mechanism of a controller. We investigate the ability of directives to coordinate the actions of agents in order to achieve a global objective more effectively. Finally, we present a case study of how directives can be used to determine the actions of autonomous mobile robots.

27. Understanding heterochromatin redistribution in mouse retina by designing and implementing a novel reporter cell-line in retinal organoids

Abstract

In eukaryotic cells, DNA is packed in the form of chromatin distributed three-dimensionally. This allows for the control of gene expression based on the location of chromatin. Euchromatin is transcriptionally active and located at the nuclear interior, while heterochromatin is transcriptionally inactive and found at the nuclear periphery. This is known to be the conventional organization and is found across vertebrates. However, mature nocturnal rod photoreceptors in the retina have the opposite nuclear organization. Hence, euchromatin is located at the nuclear periphery and heterochromatin at the nuclear interior. The purpose of this project is to optimize and develop tools to study the rod-specific nuclear organization. The Kind-lab developed the scDam&T approach, which allows for the simultaneous measurement of genomic positioning and transcription in single cells. Combining this method with an mESC-based in vitro organoid system would allow us to study and perturb the inversion. However, preliminary data showed that the Dam-LaminB1 F1ES line could not differentiate into retinal organoids. First, I optimized the mouse retinal organoid protocol and tested multiple cell lines. Next, we generated an inducible IB10-based Dam-LaminB1 cell line. From this we were able to grow organoids which showed all the hallmarks of retinal organoids.

28. Reference frame modernization in Canada

Abstract

The United States National Geodetic Survey (NGS) will adopt a new geometric reference frame in 2025 (NATRF2022). NATRF2022 will be based on ITRF2020 and separated from NAD83, the currently adopted geometric frame in both countries, by up to 1.5 meters at the Canada-U.S. border. The Canadian Geodetic Survey (CGS) will adopt NATRF2022 in parallel with the US, and is collaborating with NGS to define and realise NATRF2022 to ensure compatibility in both countries. CGS is also leading an effort to adopt NATRF2022 as a unified reference frame across provincial and other jurisdictions, which currently use different versions of NAD83(CSRS) as they have the authority to regulate reference system adoption. In this paper, we describe Canadian considerations for the definition and realisation of NATRF2022, and outline efforts and challenges in migrating to and maintaining NATRF2022 as a unified reference system throughout all jurisdictions in Canada. Abstract

International audience; In 2016, the European Hematology Association (EHA) published the EHA Roadmap for European Hematology Research1 aiming to highlight achievements in the diagnostics and treatment of blood disorders, and to better inform European policy makers and other stakeholders about the urgent clinical and scientific needs and priorities in the field of hematology. Each section was coordinated by 1–2 section editors who were leading international experts in the field. In the 5 years that have followed, advances in the field of hematology have been plentiful. As such, EHA is pleased to present an updated Research Roadmap, now including 11 sections, each of which will be published separately. The updated EHA Research Roadmap identifies the most urgent priorities in hematology research and clinical science, therefore supporting a more informed, focused, and ideally a more funded future for European hematology research. The 11 EHA Research Roadmap sections include Normal Hematopoiesis; Malignant Lymphoid Diseases; Malignant Myeloid Diseases; Anemias and Related Diseases; Platelet Disorders; Blood Coagulation and Hemostatic Disorders; Transfusion Medicine; Infections in Hematology; Hematopoietic Stem Cell Transplantation; CAR-T and Other Cell-based Immune Therapies; and Gene Therapy.
