UNLABELLED: As the closest living relatives of animals, choanoflagellates offer insights into the ancestry of animal cell physiology. Here, we report the isolation and characterization of a colonial choanoflagellate from Mono Lake, California. The choanoflagellate forms large spherical colonies that are an order of magnitude larger than those formed by the closely related choanoflagellate Salpingoeca rosetta. In cultures maintained in the laboratory, the lumen of the spherical colony is filled with a branched network of extracellular matrix and colonized by bacteria, including diverse Gammaproteobacteria and Alphaproteobacteria. We propose to erect Barroeca monosierra gen. nov., sp. nov. Hake, Burkhardt, Richter, and King to accommodate this extremophile choanoflagellate. The physical association between bacteria and B. monosierra in culture presents a new experimental model for investigating interactions among bacteria and eukaryotes. Future work will investigate the nature of these interactions in wild populations and the mechanisms underpinning the colonization of B. monosierra spheres by bacteria. IMPORTANCE: The diversity of organisms that live in the extreme environment of Mono Lake (California, USA) is limited. We sought to investigate whether the closest living relatives of animals, the choanoflagellates, exist in Mono Lake, a hypersaline, alkaline, arsenic-rich environment. We repeatedly isolated members of a new species of choanoflagellate, which we have named Barroeca monosierra. Characterization of B. monosierra revealed that it forms large spherical colonies containing diverse co-isolated bacteria, providing an opportunity to investigate mechanisms underlying physical associations between eukaryotes and bacteria.

Upper-extremity injuries are more common than lower-extremity, spine, and pelvis injuries. In this review, we focus on the most common snowboarding injuries of the extremities, spine, and pelvis and provide an overview of their respective rehabilitation and return-to-sport protocols. Despite many of the injuries seen in snowboarding also occurring in other sports, objective data about rehabilitation and return to sport are lacking for many injuries. This provides an opportunity for research in the area with regard to many sports and many different injuries. Recently, the structure and dynamics of microemulsions adjacent to a planar hydrophilic wall have been characterized using grazing incidence small angle neutron scattering (GISANS) and grazing incidence neutron spin echo (GINSES) spectroscopy, respectively. It has been found that a bicontinuous microemulsion develops a lamellar structure, and that the relaxation rate is faster for a membrane close to the interface compared to the bulk. A model developed by Seifert was employed to explain the discovered acceleration for the near-surface lamellar ordered membranes. In this contribution we present how confinement of a flat interface influences the elastic properties of surfactant membranes and discuss the effect of locally introduced flat interfaces by clay platelets. This analysis sheds light onto the undulation mode spectrum of the surfactant membrane.

Legal Metrology’s requirements can be transferred into the IT security domain applying a generic set of standardized rules provided by the Common Criteria (ISO/IEC 15408). We will outline the transfer and cross validation of such an approach. As an example serves the integration of Legal Metrology’s requirements into a recently developed Common Criteria based Protection Profile for a Smart Meter Gateway designed under the leadership of the Germany’s Federal Office for Information Security. The requirements on utility meters laid down in the Measuring Instruments Directive (MID) are incorporated. A verification approach to check for meeting Legal Metrology’s requirements by their interpretation through Common Criteria’s generic requirements is also presented.

After the dynamics of microemulsions adjacent to a planar hydrophilic wall have been characterized using grazing incidence neutron spin echo spectroscopy, the model of Seifert was employed to explain the discovered acceleration for the surface near lamellar ordered membranes. Reflections of hydrodynamic waves by the wall – or the volume conservation between the membrane and the wall – explain faster relaxations and, therefore, a lubrication effect that is important for flow fields in narrow pores. The whole scenery is now spectated by using different scenarios of a bicontinuous microemulsion exposed to clay particles and of a lamellar microemulsion adjacent to a planar wall. The Seifert concept could successfully be transferred to the new problems.

Emissions of C2-C5 alkanes from the U.S. oil and gas sector have changed rapidly over the last decade. We use a nested GEOS-Chem simulation driven by updated 2011NEI emissions with aircraft, surface and column observations to 1) examine spatial patterns in the emissions and observed atmospheric abundances of C2-C5 alkanes over the U.S., and 2) estimate the contribution of emissions from the U.S. oil and gas industry to these patterns. The oil and gas sector in the updated 2011NEI contributes over 80% of the total U.S. emissions of ethane (C2H6) and propane (C3H8), and emissions of these species are largest in the central U.S. Observed mixing ratios of C2-C5 alkanes show enhancements over the central U.S. below 2 km. A nested GEOS-Chem simulation underpredicts observed C3H8 mixing ratios in the boundary layer over several U.S. regions and the relative underprediction is not consistent, suggesting C3H8 emissions should receive more attention moving forward. Our decision to consider only C4-C5 alkane emissions as a single lumped species produces a geographic distribution similar to observations. Due to the increasing importance of oil and gas emissions in the U.S., we recommend continued support of existing long-term measurements of C2-C5 alkanes. We suggest additional monitoring of C2-C5 alkanes downwind of northeastern Colorado, Wyoming and western North Dakota to capture changes in these regions. The atmospheric chemistry modeling community should also evaluate whether chemical mechanisms that lump larger alkanes are sufficient to understand air quality issues in regions with large emissions of these species.

Replacing long-lived, rarely disturbed vegetation with short-lived, frequently disturbed vegetation is a widespread phenomenon in the Anthropocene that can influence ecosystem functioning and soil development by reducing the abundance of deep roots. We explore how sources and fate of soil CO2 vary with organic substrate source, abundance of respiring biota (i.e., roots and soil microbes), season, and soil depth. We quantified multiple isotopic signatures of CO2 (δ13C, Δ14C, δ18O) as well as concentrations and δ18O of free O2 in the upper 5 m of soil at sites where root abundances and soil organic C have been previously quantified: in late-successional forests, cultivated fields, and ~ 80 y old regenerating pine forests growing on previously cultivated land. We hypothesized that soil CO2sources would vary across soil depth and land cover, reflecting varying abundances of organic substrates, and seasonally as the dominance of root vs. microbial CO2 production changes through the year. δ13C–CO2 revealed respiration of C4-derived substrates in cultivated fields particularly during the growing season. This effect was not evident in soils of regenerating pine or older hardwood forests, suggesting that ~ 80 y of pine inputs to reforested soils have been sufficient to dominate microbial substrate selection over any remnant, historic agricultural C4 inputs. Δ14C–CO2 diverged by land use at 3 and 5 m, indicating that more recently-produced photosynthate is available for mineralization in forests compared to cultivated plots, and in late-successional forests compared to regenerating pine forests. At 1.5, 3, and 5 m in forested plots we observed evidence of respiratory demands on soil pore space O2. In these soils, we observed declines in [O2] compared to other depths and to the agricultural plots and concurrent increases in δ18O of free O2, consistent with the idea that roots and heterotrophic soil microbes are more active where photosynthate is more available. The δ18O–CO2 values, a likely proxy for δ18O of soil porewater, exhibited 18O enrichment during the winter, when many sampling wells were flooded, compared to growing season values. These data suggest an isotopically-distinct and laterally-flowing source of CO2-laden porewater during winter months. Combined, these datasets document how ~ 80 y of forest regeneration can provide sufficient C inputs to mask any microbial mineralization of decades-old organic inputs, but belowground C inputs still lag those of late successional forests. We also infer that lateral and vertical flows of water can serve as a sink for biotically-generated CO2, and that where deep soil [CO2] is lower due to lower root and microbial activities, production of carbonic acid is also diminished. Where reaction rates are weathering limited, a paucity of deep roots imposed by anthropogenic land cover change thus may limit the production of this agent of soil development and the C sink represented by the silicate weathering it can promote. The data suggest deep and persistent effects of the loss of deeply rooted long-lived vegetation on deep soil C storage and transformations that promote acid-dissolution weathering reactions that help form soil itself. Self-modulation of an electron beam in a plasma has been observed. The propagation of a long (several plasma wavelengths) electron bunch in an overdense plasma resulted in the production of multiple bunches via the self-modulation instability. Using a combination of a radio-frequency deflector and a dipole spectrometer, the time and energy structure of the self-modulated beam was measured. The longitudinal phase space measurement showed the modulation of a long electron bunch into three bunches with an approximately 200  keV/c amplitude momentum modulation. Demonstrating this effect is a breakthrough for proton-driven plasma accelerator schemes aiming to utilize the same physical effect.

Recent measurements over the Northern Hemisphere indicate that the long-term decline in the atmospheric burden of ethane (C2H6) has ended and the abundance increased dramatically between 2010 and 2014. The rise in C2H6 atmospheric abundances has been attributed to oil and natural gas extraction in North America. Existing global C2H6 emission inventories are based on outdated activity maps that do not account for current oil and natural gas exploitation regions. We present an updated global C2H6 emission inventory based on 2010 satellite-derived CH4 fluxes with adjusted C2H6 emissions over the U.S. from the National Emission Inventory (NEI 2011). We contrast our global 2010 C2H6 emission inventory with one developed for 2001. The C2H6 difference between global anthropogenic emissions is subtle (7.9 versus 7.2 Tg yr-1), but the spatial distribution of the emissions is distinct. In the 2010 C2H6 inventory, fossil fuel sources in the Northern Hemisphere represent half of global C2H6 emissions and 95% of global fossil fuel emissions. Over the U.S., unadjusted NEI 2011 C2H6 emissions produce mixing ratios that are 14-50% of those observed by aircraft observations (2008-2014). When the NEI 2011 C2H6 emission totals are scaled by a factor of 1.4, the Goddard Earth Observing System Chem model largely reproduces a regional suite of observations, with the exception of the central U.S., where it continues to underpredict observed mixing ratios in the lower troposphere. We estimate monthly mean contributions of fossil fuel C2H6 emissions to ozone and peroxyacetyl nitrate surface mixing ratios over North America of ~ 1% and ~8%, respectively.

The self-modulation instability of long particle beams was proposed as a new mechanism to produce driver beams for proton driven plasma wakefield acceleration (PWFA). The PWFA experiment at the Photo Injector Test facility at DESY, Zeuthen site (PITZ) was launched to experimentally demonstrate and study the self-modulation of long electron beams in plasma. Key aspects for the experiment are the very flexible photocathode laser system, a plasma cell and well-developed beam diagnostics. In this contribution we report about the plasma cell design, preparatory experiments and the results of the first PWFA experiment at PITZ.

We have developed semi-independent methods for determining CH2O scavenging efficiencies (SEs) during strong midlatitude convection over the western, south-central Great Plains, and southeastern regions of the United States during the 2012 Deep Convective Clouds and Chemistry (DC3) Study. The Weather Research and Forecasting model coupled with chemistry (WRF-Chem) was employed to simulate one DC3 case to provide an independent approach of estimating SEs and the opportunity to study CH2O retention in icewhen liquid drops freeze. Measurements of CH2O instorminflow and outflow were acquired on board the NASA DC-8 and the NSF/National Center for Atmospheric Research Gulfstream V (GV) aircraft employing cross-calibrated infrared absorption spectrometers. This study also relied heavily on the nonreactive tracers i-/n-butane and i-/n-pentane measured on both aircraft in determining lateral entrainment rates during convection as well as their ratios to ensure that inflow and outflow air masses did not have different origins. Of the five storm cases studied, the various tracermeasurements showed that the inflow and outflow from four storms were coherently related. The combined average of the various approaches from these storms yield remarkably consistent CH2O scavenging efficiency percentages of: 54%± 3% for 29 May; 54% ± 6% for 6 June; 58%± 13% for 11 June; and 41 ± 4% for 22 June. The WRF-Chem SE result of 53% for 29 May was achieved only when assuming complete CH2O degassing from ice. Further analysis indicated that proper selection of corresponding inflow and outflow time segments is more important than the particular mixing model employed.

The volatile organic compound (VOC) distribution in the Mexico City Metropolitan Area (MCMA) and its evolution as it is uplifted and transported out of the MCMA basin was studied during the 2006 MILAGRO/MIRAGE-Mex field campaign. The results show that in the morning hours in the city center, the VOC distribution is dominated by non-methane hydrocarbons (NMHCs) but with a substantial contribution from oxygenated volatile organic compounds (OVOCs), predominantly from primary emissions. Alkanes account for a large part of the NMHC distribution in terms of mixing ratios. In terms of reactivity, NMHCs also dominate overall, especially in the morning hours. However, in the afternoon, as the boundary layer lifts and air is mixed and aged within the basin, the distribution changes as secondary products are formed. The WRF-Chem (Weather Research and Forecasting with Chemistry) model and MOZART (Model for Ozone and Related chemical Tracers) were able to approximate the observed MCMA daytime patterns and absolute values of the VOC OH reactivity. The MOZART model is also in agreement with observations showing that NMHCs dominate the reactivity distribution except in the afternoon hours. The WRF-Chem and MOZART models showed higher reactivity than the experimental data during the nighttime cycle, perhaps indicating problems with the modeled nighttime boundary layer height. A northeast transport event was studied in which air originating in the MCMA was intercepted aloft with the Department of Energy (DOE) G1 on 18 March and downwind with the National Center for Atmospheric Research (NCAR) C130 one day later on 19 March. A number of identical species measured aboard each aircraft gave insight into the chemical evolution of the plume as it aged and was transported as far as 1000 km downwind; ozone was shown to be photochemically produced in the plume. The WRF-Chem and MOZART models were used to examine the spatial extent and temporal evolution of the plume and to help interpret the observed OH reactivity. The model results generally showed good agreement with experimental results for the total VOC OH reactivity downwind and gave insight into the distributions of VOC chemical classes. A box model with detailed gas phase chemistry (NCAR Master Mechanism), initialized with concentrations observed at one of the ground sites in the MCMA, was used to examine the expected evolution of specific VOCs over a 1-2 day period. The models clearly supported the experimental evidence for NMHC oxidation leading to the formation of OVOCs downwind, which then become the primary fuel for ozone production far away from the MCMA.
