We recently reported pheochromocytoma 12 (PC12) cells and rats subjected to intermittent hypoxia (IH), a hallmark manifestation of obstructive sleep apnea (OSA), exhibit reduced histone deacetylase activity and HDAC5 protein. Our study further suggested that posttranslational modifications rather than transcriptional mechanism(s) mediate IH-induced HDAC5 degradation. These observations prompted our current study to investigate the mechanism(s) underlying HDAC5 degradation by IH in PC12 cell cultures. IH-induced HDAC5 degradation was blocked by an antioxidant, and reactive oxygen species (ROS) mimetics decreased HDAC5 protein, suggesting that ROS mediates HDAC5 degradation by IH. NADPH oxidases (NOX) 2 and 4 were identified as sources of ROS that mediate the effects of IH. HDAC5 degradation during IH was associated with dephosphorylation of HDAC5 at serine259, and this response was blocked by a NOX inhibitor, suggesting that ROS-dependent dephosphorylation mediates HDAC5 degradation. IH-induced dephosphorylation of HDCA5 was inhibited by calyculin A, an inhibitor of protein phosphatase (PP)-1 and -2, or by the overexpression of nuclear inhibitor of PP1 (NIPP1). HDAC5 dephosphorylation by IH lead to augmented hypoxia-inducible factor (HIF)-1α protein and an increase in its transcriptional activity. These data suggest that PP1-dependent dephosphorylation of S259 destabilizes HDAC5 protein in response to IH, resulting in HIF-1α stabilization and transcriptional activity. Our findings highlight hither to unexplored role of protein phosphatases, especially PP1 in regulating HDAC5 protein, which is an upstream activator of HIF-1 signaling by IH.

Obstructive sleep apnea (OSA) is characterized by sporadic collapse of the upper airway leading to periodic disruptions in breathing. Upper airway patency governed by genioglossal nerve activity originates from the hypoglossal motor nucleus. Mice with targeted deletion of the gene Hmox2, encoding the carbon monoxide (CO) producing enzyme, heme oxygenase-2 (HO-2), exhibit severe OSA, yet the contribution of central HO-2 dysregulation to the phenomenon is unknown. Using the rhythmic brainstem slice preparation, which contains the preBötzinger complex (preBötC) and the hypoglossal nucleus, we tested the hypothesis that central HO-2 dysregulation weakens hypoglossal motoneuron output. Disrupting HO-2 activity increased transmission failure as determined by the intermittent inability of the preBötC rhythm to trigger output from the hypoglossal nucleus. Failed transmission was associated with a reduced input-output relationship between the preBötC and the motor nucleus. These network phenomena were related to smaller inspiratory drive currents and reduced intrinsic excitability among hypoglossal neurons. In addition to HO-2, hypoglossal neurons also expressed the CO-regulated H2S producing enzyme cystathionine □-lyase (CSE). H2S abundance was higher in hypoglossal neurons of HO-2 null mice than wild-type controls. Disrupting CSE function normalized transmission in HO-2 null mice and an H2S donor mimicked the effects of HO-2 dysregulation. These findings demonstrate a hitherto uncharacterized modulation of hypoglossal activity through the interaction of HO-2 and CSE-derived H2S, and supports the perspective that centrally derived HO-2 activity plays an important role regulating upper airway control.

Intermittent hypoxia (IH) is a hallmark manifestation of obstructive sleep apnea (OSA). Rodents treated with IH exhibit hypertension. Hypoxia-inducible factor (HIF)-1-dependent transcriptional activation of NADPH oxidases ( Nox) and the resulting increase in reactive oxygen species (ROS) levels is a major molecular mechanism underlying IH/OSA-induced hypertension. Jumanji C (JmjC)-containing histone lysine demethylases (JmjC-KDMs) are coactivators of HIF-1-dependent transcriptional activation. In the present study, we tested the hypothesis that JmjC-KDMs are required for IH-evoked HIF-1 transcriptional activation of Nox4 and the ensuing hypertension. Studies were performed on pheochromocytoma (PC)12 cells and rats. IH increased KDM6B protein and enzyme activity in PC12 cells in an HIF-1-independent manner as evidenced by unaltered KDM6B activation by IH in HIF-1α shRNA-treated cells. Cells treated with IH showed increased HIF-1-dependent Nox4 transcription as indicated by increased HIF-1α binding to hypoxia-responsive element (HRE) sequence of the Nox4 gene promoter demonstrated by chromatin immunoprecipitation (ChiP) assay. Pharmacological blockade of KDM6B with GSKJ4, a specific KDM6 inhibitor, or genetic silencing of KDM6B with shRNA abolished IH-induced Nox4 transcriptional activation by blocking HIF-1α binding to the promoter of the Nox4 gene. Treating IH-exposed rats with GSKJ4 showed: 1) absence of KDM6B activation and HIF-1-dependent Nox4 transcription in the adrenal medullae, and 2) absence of elevated plasma catecholamines and hypertension. Collectively, these findings indicate that KDM6B functions as a coactivator of HIF-1-mediated Nox4 transactivation and demonstrates a hitherto uncharacterized role for KDMs in IH-induced hypertension by HIF-1.

In Tibetans, noncoding alleles in EPAS1 —whose protein product hypoxia-inducible factor 2α (HIF-2α) drives the response to hypoxia—carry strong signatures of positive selection; however, their functional mechanism has not been systematically examined. Here, we report that high-altitude alleles disrupt the activity of four EPAS1 enhancers in one or more cell types. We further characterize one enhancer (ENH5) whose activity is both allele specific and hypoxia dependent. Deletion of ENH5 results in down-regulation of EPAS1 and HIF-2α targets in acute hypoxia and in a blunting of the transcriptional response to sustained hypoxia. Deletion of ENH5 in mice results in dysregulation of gene expression across multiple tissues. We propose that pleiotropic adaptive effects of the Tibetan alleles in EPAS1 underlie the strong selective signal at this gene.

Chronic intermittent hypoxia (CIH) is a hallmark manifestation of obstructive sleep apnea (OSA), a widespread breathing disorder. CIH-treated rodents exhibit activation of the sympathetic nervous system and hypertension. Heightened carotid body (CB) activity has been implicated in CIH-induced hypertension. CB expresses high abundance of olfactory receptor (Olfr) 78, a G-protein coupled receptor. Olfr 78 null mice exhibit impaired CB sensory nerve response to acute hypoxia. Present study examined whether Olfr78 participates in CB-dependent activation of the sympathetic nervous system and hypertension in CIH-treated mice and in hemeoxygenase (HO)-2 null mice experiencing CIH as a consequence of naturally occurring OSA. CIH-treated wild-type (WT) mice showed hypertension, biomarkers of sympathetic nerve activation, and enhanced CB sensory nerve response to hypoxia and sensory long-term facilitation (sLTF), and these responses were absent in CIH-treated Olfr78 null mice. HO-2 null mice showed higher apnea index (AI) (58 ± 1.2 apneas/h) than WT mice (AI = 8 ± 0.8 apneas/h) and exhibited elevated blood pressure (BP), elevated plasma norepinephrine (NE) levels, and heightened CB sensory nerve response to hypoxia and sLTF. The magnitude of hypertension correlated with AI in HO-2 null mice. In contrast, HO-2/Olfr78 double null mice showed absence of elevated BP and plasma NE levels and augmented CB response to hypoxia and sLTF. These results demonstrate that Olfr78 participates in sympathetic nerve activation and hypertension and heightened CB activity in two murine models of CIH. NEW & NOTEWORTHY Carotid body (CB) sensory nerve activation is essential for sympathetic nerve excitation and hypertension in rodents treated with chronic intermittent hypoxia (CIH) simulating blood O(2) profiles during obstructive sleep apnea (OSA). Here, we report that CIH-treated mice and hemeoxygenase (HO)-2-deficient mice, which show OSA phenotype, exhibit sympathetic excitation, hypertension, and CB activation. These effects are absent in Olfr78 null and Olfr78/HO-2 double null mice.

In this work, a bus encoding method is proposed that reduces the effect of crosstalk. The crosstalk usually occurs when the data is in parallel communicated. In planar structures, the crosstalk effect is large due to the usage of parallel communication and wide data patterns. In bus technique, the huge amount of wires is laid in equal over a significant time. One way to reduce crosstalk without changing the parallel communicating data lines is to reduce the wideband data patterns so as to reduce the power utilization. The proposed encoding method can minimize the crosstalk by reducing wide data patterns without degrading the performance. The architecture is implemented on Artix 7 FPGA at a 28nm technology node. The simulation is done using the HDL tool and the results are compared with the existing FPGA architecture. With the proposed method, the wire density and the power consumption are reduced by 57.4% and 50% respectively as compared with existing 45 nm technologies.

Respiration and blood pressure (BP) are regulated to maintain optimal delivery of O2 to every cell in the body. Arterial hypoxemia is sensed by the carotid body (CB), which initiates sympathetic reflex arcs to the diaphragm to increase ventilation, and to the adrenal medulla (AM) to increase catecholamine secretion and thereby increase BP. However, the underlying molecular mechanisms have not been fully delineated. Here, we report that the relative activities of hypoxia-inducible factor-1 (HIF-1) and HIF-2 determine the set point for the CB and AM, with respect to their maintenance of BP and respiration. In Hif2a+/- mice, which are heterozygous for a knockout allele at the locus encoding HIF-2α, expression of HIF-1α and NADPH oxidase 2 was increased in the CB and AM, resulting in an oxidized intracellular redox state with augmented sensitivity to hypoxia, increased BP, and respiratory abnormalities, which were all normalized by treatment with a HIF-1α inhibitor or a superoxide anion scavenger. By contrast, in Hif1a+/- mice, which are heterozygous for a knockout allele at the locus encoding HIF-1α, the expression of HIF-2α and superoxide dismutase 2 was increased in the CB and AM, resulting in a reduced intracellular redox state with impaired CB and ventilatory responses to chronic hypoxia, which were normalized by treatment with a HIF-2α inhibitor. None of the abnormalities that were observed in Hif1a+/- or Hif2a+/- mice were observed in Hif1a+/-; Hif2a+/- double- heterozygous mice. Our results demonstrate that redox balance in the CB and AM, which is determined by mutual antagonism between HIF-α isoforms, establishes the set point for responses of the CB and AM to hypoxia, and is required for the maintenance of normal BP and respiration.

Study question How does viscosity influence the flagellar beating behaviour of free-swimming bull, mouse, and human sperm? Summary answer Sperm flagellar beating behaviour exhibits a transition from an irregular three-dimensional (3D) beating at 5 mPa·s to an organized two-dimensional (2D) waveform at 20 mPa·s. What is known already Sperm migrate in a complex viscoelastic environment through the female reproductive tract. The viscoelastic properties of the oviductal fluid significantly influence the progressive motility of sperm, acting as one of the key guidance mechanisms in vivo. However, the biomechanics of sperm flagellar activity in response to varying viscosity of the oviductal fluid is poorly understood. Understanding sperm flagellar behaviour in physiologically relevant environments is crucial to understanding reproduction and may help to describe unknown causes of infertility. Lack of high-speed high-resolution imaging techniques and automated image-analysis capabilities have been the main barriers to fully describe the flagellar beating behaviour. Study design, size, duration We used a custom-built high-speed high-resolution dark-field microscopy platform to resolve the flagellar dynamics of human, bull, and mouse sperm near surfaces in viscoelastic media ranging in viscosity from 1 to 250 mPa·s. The imaging system includes an automated image analysis algorithm to quantify sperm flagellar waveform and motility characteristics by extracting the flagellar centreline, reconstructing the waveform and calculating tangent-angle profiles. 20 sperm from 3 different bull, mice and humans were analyzed. Participants/materials, setting, methods Bull, mouse, and human sperm were used in this study. In each experiment, a diluted sperm sample in a buffer supplemented with methylcellulose was used and free-swimming sperm were imaged using dark-field microscopy at 200 frames per second. An automated image analysis algorithm was used to extract sperm flagellar centreline and Proper Orthogonal Decomposition (POD) was then used to study sperm flagellar waveform. Statistical analysis was performed using one-way ANOVA. Main results and the role of chance The reconstructed flagellar beating pattern was different for sperm swimming in low and high-viscosity media. Bull sperm exhibited a lower flagellar beating amplitude along the end piece when swimming in a high-viscosity media, a potential energy-efficient strategy to navigate a high viscosity fluid. The first two dominant POD modes (shape modes) describe more than 90% of the beating pattern for all species. Bull sperm exhibited a transition mode with irregular loops in 5 mPa·s buffer, but the flagellar shape cycle created an organised repetitive circular cycle in 1 mPa.s (3D beating) buffer and at viscosities higher than 5 mPa·s (2D beating). Human sperm also indicated a similar behaviour but with the transition happening at higher viscosities. Mouse sperm in high-viscosity media had a lower flagellar beating amplitude across the principal piece and higher beating amplitude across the end piece. The flagellar shape cycle in mouse sperm showed a periodic flagellar beating behaviour at high-viscosities (>20 mPa·s), but a shape cycle with distorted loops at lower viscosities. Our results showed in quantitative detail that increasing viscosity alters sperm flagellar beating pattern, and how sperm migration behaviour in low viscosity media can be distinct from their swimming behaviour in vivo. Limitations, reasons for caution A more comprehensive study of sperm motility parameters such as curvilinear velocity, average path velocity and straight line velocity with a larger sample size is required to fully characterise sperm swimming behaviour as a function of viscosity. Wider implications of the findings The increasing viscosity of the oviductal fluid regulates the sperm flagellar beating behaviour to switch from a 3D swimming behaviour with irregular shape cycles at lower viscosities to a 2D slithering mode with repetitive circular shape cycles at higher viscosities to achieve a more energy-efficient beating pattern for navigation. Naked mole rats (NMRs) exhibit blunted hypoxic (HVR) and hypercapnic ventilatory responses (HCVR). The mechanism(s) underlying these responses are largely unknown. We hypothesized that attenuated carotid body (CB) sensitivity to hypoxia and hypercapnia contributes to the near absence of ventilatory responses to hypoxia and COWe measured ex vivo CB sensory nerve activity, phrenic nerve activity (an estimation of ventilation), and blood gases in urethane-anesthetized NMRs and C57BL/6 mice breathing normoxic, hypoxic, or hypercapnic gases. CB morphology, carbon monoxide, and HRelative to mice, NMRs had blunted CB and HVR. Morphologically, NMRs have larger CBs, which contained more glomus cells than in mice. Furthermore, NMR glomus cells form a dispersed pattern compared to a clustered pattern in mice. Hemeoxygenase (HO)-1 mRNA was elevated in NMR CBs, and an HO inhibitor increased CB sensitivity to hypoxia in NMRs. This increase was blocked by an HConsistent with our hypothesis, impaired CB responses to hypoxia contribute in part to the blunted HVR in NMRs. Conversely, the HCVR and CB are more sensitive to CO

Aim: To evaluate and compare the efficacy of two 5% sodium fluoride varnishes containing amorphous calcium phosphate and casein phosphopeptide amorphous calcium phosphate respectively on the plaque pH, salivary fluoride ion release and patient acceptability. Methods: Seventy children were selected from a residential school for the study. Baseline plaque pH, salivary fluoride ion levels were determined. Varnish application was made, and the plaque pH and salivary fluoride ion release were determined at 24 hours, 3 months’ and 6 months’ post varnish application. Patient acceptability was determined immediately after varnish application. Results: No difference in the plaque pH and salivary fluoride ion levels were obtained among the two study groups at all tested time intervals. However, there was a difference in the salivary fluoride ion levels at baseline and at all the three post interventional time periods for both the groups, i.e., at 24 hours (for Group 1 p= 0.00 for Group 2 p=0.00), at 3 months (for Group 1 p=0.00 for Group 2 p=0.00) and at 6 months (for Group 1 p= 0.00 for Group 2 p=0.00). There was no statistically significant difference was observed in the patient acceptability between the two groups (p=0.709). The role of hypoxia-inducible factor (HIF)-1 in pancreatic β-cell response to intermittent hypoxia (IH) was examined. Studies were performed on adult wild-type (WT), HIF-1α heterozygous (HET), β-cell-specific HIF-1−/− mice and mouse insulinoma (MIN6) cells exposed to IH patterned after blood O2 profiles during obstructive sleep apnea. WT mice treated with IH showed insulin resistance, and pancreatic β-cell dysfunction manifested as augmented basal insulin secretion, and impaired glucose-stimulated insulin secretion and these effects were absent in HIF-1α HET mice. IH increased HIF-1α expression and elevated reactive oxygen species (ROS) levels in β-cells of WT mice. The elevated ROS levels were due to transcriptional upregulation of NADPH oxidase (NOX)-4 mRNA, protein and enzymatic activity, and these responses were absent in HIF-1α HET mice as well as in β-HIF-1−/− mice. IH-evoked β-cell responses were absent in adult WT mice treated with digoxin, an inhibitor of HIF-1α. MIN6 cells treated with in vitro IH showed enhanced basal insulin release and elevated HIF-1α protein expression, and these effects were abolished with genetic silencing of HIF-1α. IH increased NOX4 mRNA, protein, and enzyme activity in MIN6 cells and disruption of NOX4 function by siRNA or scavenging H2O2 with polyethylene glycol catalase blocked IH-evoked enhanced basal insulin secretion. These results demonstrate that HIF-1-mediated transcriptional activation of NOX4 and the ensuing increase in H2O2 contribute to IH-induced pancreatic β-cell dysfunction.

Aim and Objective: Antebrachium fractures are one of the common fractures in paediatric population. Fractures of forearm comprises of about 40% in the fractures of paediatric population [1]. These fractures usually have a high degree of remodelling potential and are usually managed non operatively by closed reduction and maintaining it in a plaster cast. Nowadays the trend for management of these fractures changes towards operative management because of increasing complications by nonoperative methods. The aim of this study is to evaluate the functional outcomes of unstable both bone forearm fractures treated operatively using titanium elastic nailing. Materials and Methods: Study was conducted in the department of orthopaedics at Rajah Muthiah Medical College and Hospital. Total of 20 patients aged were treated using TENS. Closed reduction with internal fixation under c-arm guidance was done in 13 cases and open reduction with internal fixation was done in 7 cases.Results: Patients were followed up for a period of 6 months. Results were analysed with respect to union of fracture, symptoms with activities, range of motion of joints adjacent to the fracture using Price et al. criteria and Anderson et al. criteria. In all cases fractures united at an average of 8 - 10 weeks without any malunion. All 20 cases had excellent outcomes. 3 patient had nail prominence on the ulnar entry side leading to superficial pin site infection.Conclusion: Fixation of both bone fractures of forearm using titanium elastic nailing systems showed many advantages over the conventional plating methods, in terms of smaller incision, minimal soft tissue meddling during fracture fixation, prompt osseous healing, maximum range of motion at the earliest, decrease in complication rate with excellent clinical and radiological results. Thus, titanium elastic nailing used intramedullary is an effective treatment option for the treatment of unstable both bone forearm fractures in paediatric population.
